Image-based deformation estimation is an important tool used in a variety of engineering problems, including crack propagation, fracture, and fatigue failure. These tools have been important in biomechanics research where measuring in vitro and in vivo tissue deformations are important for evaluating tissue health and disease progression. However, accurately measuring tissue deformation in vivo is particularly challenging due to limited image signal-to-noise ratio. Therefore, we created a novel deep-learning approach for measuring deformation from a sequence of images collected in vivo called StrainNet. Utilizing a training dataset that incorporates image artifacts, StrainNet was designed to maximize performance in challenging, in vivo settings. Artificially generated image sequences of human flexor tendons undergoing known deformations were used to compare benchmark StrainNet against two conventional image-based strain measurement techniques. StrainNet outperformed the traditional techniques by nearly 90%. High-frequency ultrasound imaging was then used to acquire images of the flexor tendons engaged during contraction. Only StrainNet was able to track tissue deformations under the in vivo test conditions. Findings revealed strong correlations between tendon deformation and applied forces, highlighting the potential for StrainNet to be a valuable tool for assessing rehabilitation strategies or disease progression. Additionally, by using real-world data to train our model, StrainNet was able to generalize and reveal important relationships between the effort exerted by the participant and tendon mechanics. Background: A precipitous decline in health status among people recently released from prison is common. In Victoria, Australia, opioid agonist treatment (OAT) in the community involves frequent contact with primary care, potentially facilitating broader use of primary healthcare services. Among a cohort of men who injected drugs regularly pre-imprisonment, we estimated differences in rates of primary healthcare use and medication dispensation between people who did and did not receive OAT post-release. Methods: Data came from the Prison and Transition Health Cohort Study. Three-month post-release follow-up interviews were linked with primary care and medication dispensation records. Generalised linear models were fit with one exposure (OAT: none/partial/complete) for 13 outcomes relating to primary healthcare use, pathology testing, and medication dispensation, adjusted for other covariates. Coefficients were reported as adjusted incidence rate ratios (AIRR). Results: Analyses included 255 participants. Compared to no OAT use, both partial and complete OAT use were associated with increased rates of standard (AIRR: 3.02, 95%CI: 1.88–4.86; AIRR: 3.66, 95%CI: 2.57–5.23), extended (AIRR: 2.56, 95%CI: 1.41–4.67; AIRR: 2.55, 95%CI: 1.60–4.07) and mental health-related (AIRR: 2.71, 95%CI: 1.42–5.20; AIRR: 2.27, 95%CI: 1.33–3.87) general practitioner (GP) consultations, total medication (AIRR: 1.88, 95%CI: 1.19–2.98; AIRR: 2.40, 95%CI: 1.71–3.37), benzodiazepine (AIRR: 4.99, 95%CI: 2.81–8.85; AIRR: 8.30, 95%CI: 5.28–13.04) and gabapentinoid (AIRR: 6.78, 95%CI: 3.34–13.77; AIRR: 4.34, 95%CI: 2.37–7.94) dispensations, respectively. Partial OAT use was also associated with increased after-hours GP consultations (AIRR: 4.61, 95%CI: 2.24–9.48) and complete OAT use? with increased pathology utilisation (e.g. haematological, chemical, microbiological or immunological tissue/sample testing; AIRR: 2.30, 95%CI: 1.52–3.48). Conclusion: We observed higher rates of primary healthcare use and medication dispensation among people who reported partial and complete OAT use post-release. Findings suggest that access to OAT post-release may have a collateral benefit in supporting broader health service utilisation, underscoring the importance of retention in OAT after release from prison. The influence of resource availability on ecosystem function varies spatially and temporally, among and within ecosystems. Dramatic shifts in moisture‐driven resources can drive bottom‐up effects on animal behaviours and distributions. Further, complexity arises when landscapes are influenced by large mammalian grazers and predator‐induced trophic cascades, such as those mediated by the dingo (Canis familiaris (Dingo)) in the eastern arid Strzelecki Desert in Australia. During the driest two‐year period on record for this region, we investigated the persistence of avian communities associated with structurally distinct dunes and swale habitats, and across two different land management regimes (pastoral land with livestock and dingoes, and Sturt National Park managed for conservation without these animals). We grouped all birds into dietary functional groups to infer patterns of habitat use associated with available resources. We also compared incidental observations of the 'winter' bird community in part of the study region between the extended dry period of 2018/2019 and wet period of 2020/2021. Despite habitat partitioning, the avian community did not differ between land management regimes except in species richness during the dry period, likely driven by the low numbers of birds present during the surveys. Incidental observations indicated that insectivorous and omnivorous species dominated the bird community in the dry period, with granivorous species forming a greater proportion of the bird community during wet times. Birds with completely or partially insectivorous diets dominated avian species composition on surveys in the dry period, but there were distinct structural vegetation associations among functional groups, indicating that heterogeneity in vegetation structure was likely important for the conservation of refuges, which enable the persistence of avifauna during extended dry periods. Distinct habitat type, structure and available resources shaped avian communities in this landscape, during the extremely resource‐limited extended dry period, with implications for conservation and management, particularly given the increasing drying effects of climate change. Eyrean Grasswrens Amytornis goyderi have generally been considered habitat specialists associated with Sandhill Canegrass Zygochloa paradoxa on the dunes of inland Australian deserts in the Lake Eyre Basin. Following above-average rainfall in 2020-2022 and an associated vegetation response in the Strzelecki Desert, Eyrean Grasswrens were observed at 39 locations south-east of their known distribution, with sites up to 11 km inside New South Wales, well beyond the extent of previous easterly records on the New South Wales-South Australia border. Further, two sites were north of the New South Wales-Queensland border, confirming the species 380 km south of previous Queensland records. Nine sites were within feral-free exclosures at the Wild Deserts project site in Sturt National Park, New South Wales. Repeated observations between May 2021 and August 2022 suggest persistence for at least 16 months in ephemeral vegetation on sand dunes supporting no Sandhill Canegrass. Two instances of breeding were recorded, with juvenile birds and feeding by an adult male observed. We explore possible drivers for this apparent range expansion and future scenarios around persistence inside landscape-scale exclosures, within which theorised threats from feral predators and overabundant herbivores are absent, presenting an opportunity to assess the relevancy of these drivers on the ecology of Eyrean Grasswrens. Background: Australian needle and syringe distribution occurs via a mix of modalities, including syringe dispensing machines (SDMs). SDMs are electronic vending machines providing (often) 24-h access to needles/syringes and may attract greater numbers of people who are younger, female, and/or have limited connection to health care services compared to individuals accessing fixed-site needle and syringe programs (NSPs). However, validating the demographic characteristics of SDM clients has proven difficult in previous research. Methods: In this paper, we analyse SDM order and client demographic data from four SDMs located in South-East Melbourne, Australia, and compare this against the managing fixed-site NSP between May 2017 and December 2020. SDM data were collected via a novel 0–9 numeric keypad input tool. Via the tool, SDM clients were requested to input their categorised age, gender and postcode. Given the novelty of the tool, we evaluate the feasibility of the data collection method. We analysed data according to: (1) total SDM orders made, (2) estimated 'unique SDM presentations' and (3) describing the demographics of unique SDM clients. Importantly, we noted substantial invalid demographic data, and consequently, severely restricted data for analysis. Results: There were 180,989 SDM orders made across the four SDMs to an estimated 90,488 unique SDM presentations. There was little variation in unique presentations across days of the week, but 69% occurred out of NSP operating hours. Across the study period, the SDMs distributed 66% of the number of syringes distributed by the fixed-site NSP. Due to invalid demographic data, our restriction method provided only 10,914 (6% of all data) unique presentations for analysis. There were some demographic differences between SDM and NSP client, but these should be treated with caution. Conclusions: The data collection tool provides a novel means of comparing SDM and fixed-site presentations, demonstrating the substantial expansion of service via the SDMs. However, the validity of the demographic data was highly questionable and requires significant data coding, meaning it is not feasible for community NSPs. While we recommend the inclusion of automatically collected SDM order data, the use of a 0–9 numeric keypad to collect demographic data—while an innovation—requires alteration to support NSP data. Aims: To estimate incidence of post‐release injecting drug use (IDU) among men who injected drugs before imprisonment and determine factors associated with post‐release IDU frequency. Design, setting, participants: Prospective cohort study of men reporting monthly IDU before a period of sentenced imprisonment in Victoria, Australia, recruited between September 2014 and May 2016 (n = 195). Measurements: Any post‐release IDU and IDU frequency was measured via self‐report at 3‐month follow‐up interview. IDU frequency, measured over the preceding month, was categorised as no IDU, irregular IDU (1–4 days IDU) and regular IDU (≥5 days IDU). Incidence of any IDU was calculated at 3 months post‐release. Factors associated with IDU frequency were estimated using ordinal logistic regression. Findings Most (83%) participants reported post‐release IDU (265 per 100 person‐years, 95% CI, 227–309); with half (48%) reporting regular IDU, 23% irregular IDU and 29% no IDU in the month preceding follow‐up. Poorer psychological well‐being at follow‐up (General Health Questionnaire [GHQ‐12] score; adjusted odds ratio [AOR], 1.18; 95% CI, 1.07–1.29) and post‐release unemployment (AOR, 4.57; 95% CI, 1.67–12.49) were associated with increased IDU frequency. Retention in opioid agonist treatment (AOR, 0.49; 95% CI, 0.24–0.98) was associated with reduced IDU frequency. Non‐linear (inverted‐u) associations between IDU frequency and age (age: AOR, 1.51; 95% CI, 1.17–1.96; age‐squared: AOR, 0.99; 95% CI, 0.99–0.99) and pre‐imprisonment IDU frequency (pre‐imprisonment IDU frequency: AOR, 1.36; 95% CI, 1.15–1.61; pre‐imprisonment IDU frequency‐squared: AOR, 0.99; 95% CI, 0.99–0.99) were found, with odds peaking at age 39 and 19 days IDU, respectively. Longer baseline sentence length was associated with reduced odds of irregular and regular IDU (AOR, 0.99; 95% CI, 0.99–0.99). Conclusion: Among Australian men who inject drugs before imprisonment, resumption of injecting drug use after release from prison appears to be common, with imprisonment seeming to have little impact on reducing injecting drug use behaviour. Background: Australian harm reduction services are provided via a mix of modalities, including fixed-site needle and syringe programmes (NSP) and syringe-dispensing machines (SDMs). SDMs are cost-effective and provide 24-h anonymous access to needles/syringes, often to underserved geographic areas, and can attract clientele who may choose not to use NSPs. The introduction of COVID-19 control measures saw disruptions and adaptations to the provision of harm reduction services. It is possible that SDMs filled the gap in otherwise disrupted harm reduction services in Melbourne. In this paper, we use data from four SDMs and an NSP to explore changes to harm reduction usage during periods of COVID-19 lockdowns in Melbourne, Australia, in 2020. Methods: Our data span September 2017–December 2020. We analysed daily counts of SDM use and monthly counts of NSP use, according to unique presentations to both. Auto-regressive integrated moving average (ARIMA) time-series models were fitted to the data with the effects of lockdowns estimated via a step function. Results: Across the study period, we estimated 85,851 SDM presentations and 29,051 NSP presentations. Usage across both the SDMs and the NSP declined during the COVID-19 lockdowns, but only the decline in SDM usage was significant in ARIMA analysis. Conclusions: The slight, but significant decline in SDM use suggests barriers to access, though this may have been mitigated by SDM users acquiring needles/syringes from other sources. The decline, however, may be a concern if it led to lowered needle/syringe coverage and a subsequent increase in injecting risk. Further work is needed to properly explore potential changes in preference for needle/syringe acquisition site and associated barriers. Mechanical testing is a valuable tool for assessing intervertebral disc health, but the wide range of testing protocols makes it difficult to compare results from different studies. Normalizing mechanical properties by disc geometry allows for such comparisons, but there is little consistency in the methods by which disc geometry is measured. As such, we hypothesized that methods used to measure disc geometry would impact reported mechanical properties. Disc height and area were measured using computed tomography (CT), digital calipers, and ImageJ to yield three different measurements for disc height and six for disc area. Disc heights measured by digital calipers ex situ were >30% less than disc heights measured in situ by CT, and disc areas measured ex situ using ImageJ were >30% larger than those measured by CT. This significantly affected reported mechanical properties, leading to a 65% reduction in normalized compressive stiffness in the most extreme case. Though we cannot quantitatively correct between methods, results presented in this study suggest that disc geometry measurement methods have a significant impact on normalized mechanical properties and should be accounted for when comparing results. Illicit drug use and mental illness are common among people in prison and are associated with higher rates of reoffending and reimprisonment. We conducted a systematic review, searching MEDLINE, Embase, and PsycINFO to January 10, 2022, for studies reporting criminal justice involvement following exposure to community mental health services among people released from jail or prison who use illicit drugs and have mental illness. Our search identified 6954 studies; 13 were eligible for inclusion in this review. Studies were separated into three broad categories based on community mental health service type. Eleven of 13 studies reported a reduction in criminal justice involvement among participants exposed to community mental health services compared to a comparison group. Findings indicate a need to expand and improve integration and referral mechanisms linking people to community mental health services after jail or prison release, alongside a need for tailored programs for individuals with complex illicit drug use and mental health morbidities. Loot boxes are a popular mechanic within many video games, but it remains unclear if some forms of loot boxes can be seen of as gambling. However, the perspectives of players are often neglected, such as whether they see them as 'fair' game elements and how closely they feel this aligns with gambling. In this paper, we synthesise a conceptualisation for loot boxes through players' actual experience and explore if there are any parallels with gambling. Twenty-one participants who played video games took part in the research through either an interview or online survey. Thematic analysis suggested that six themes were core to exploring loot boxes: Random Chance Effects, Attitudes Towards Content, Implementation, Parallels with Gambling, Game Design, and The Player. The results suggested both indirect and direct parallels with gambling from the players experiences. Implications of game design and classifying loot boxes as gambling are discussed in relation to game design and risk factors of gambling and purchasing behaviour. [ABSTRACT FROM AUTHOR]

Dryland ecosystems have limited and patchily distributed water – a vital resource for species in these landscapes. Degradation of these ecosystems is increasing due to climate change and understanding species' responses to this drying is critical for effective ecosystem management. During an extended dry period, we surveyed drinking visits of an avian community at an artificial waterpoint in semi-arid Australia. Opportunistically, a small rainfall event punctuated the survey, enabling comparisons of avian visitations, before and after the event, as a proxy for increased water availability in the landscape. Visitations of drinking birds to the waterpoint before the rainfall event (17 species) were significantly higher than after (3 species). Permanent waterpoints, such as the single site surveyed in this study, can sustain avifauna during extended dry periods in drylands, affecting spatio-temporal and potentially functional avian community dynamics. Periodic reliance of dryland birds on permanent water may increase with prolonged droughts under climate change, and this reliance must be considered alongside the negative ecological consequences of permanent waterpoints in the management of these ecosystems. Arid rangelands are degraded worldwide, suffering vegetation transformation, soil erosion, introductions, and extinctions. Wild deserts is restoring a desert ecosystem in Sturt National Park, New South Wales, Australia (35,000 ha), eradicating or controlling introduced animals, managing native herbivores, and reintroducing regionally extinct mammals. We describe a Strategic Adaptive Management Plan for restoration of this desert ecosystem, including a vision, model of ecosystem processes, stakeholder input, a hierarchy of objectives linked to triggers and their management actions, producing outcomes and outputs. Our management treatments included two "no restoration" areas and three "restoration" areas. The latter include two exclosures (each 2,000 ha), free of introduced animals (foxes, cats, rabbits), with previously abundant kangaroos removed and regionally extinct mammals to be reintroduced. The third management treatment is a Wild Training Zone (10,400 ha), with introduced animals and kangaroos managed at low levels, using innovative methods, improving survivorship and avoidance behavior of reintroduced mammals to introduced predators. These measures will allow populations of threatened animals to establish, initially in the exclosures, then the Wild Training Zone and potentially more widely. Our strategic adaptive management planning approach is generic and implementable for any natural resource management project, providing explicit steps and processes that track and report transparently on outcomes, fostering learning by doing. Wild Deserts is an ecosystem restoration project in semiarid Sturt National Park, Australia, funded for at least decade. It involves eradicating or controlling invasive animal species, managing native herbivores and reintroducing regionally extinct mammals. The coronavirus disease 2019 (covid-19) pandemic caused by the novel severe acute respiratory syndrome coronavirus 2 has necessitated changes to the way patients with chronic diseases are managed. Given that patients with multiple myeloma are at increased risk of covid-19 infection and related complications, national bodies and experts around the globe have made recommendations for risk mitigation strategies for those vulnerable patients. Understandably, because of the novelty of the virus, many of the proposed risk mitigation strategies have thus far been reactionary and cannot be supported by strong evidence. In this editorial, we highlight some of the risk mitigation strategies implemented at our institutions across Canada during the first wave of covid-19, and we discuss the considerations that should be made when managing patients during the second wave and beyond. Tetrabenzo[5.7]fulvalene, one of the first recognized stable members of mixed fulvalenes, has attracted widespread interest for its remarkable structure. However, little has been known about its photoactivity, most likely owing to its very weak luminescence in the solution state. Here we show for the first time that this compound exhibits aggregation-induced emission (AIE) properties. Species reintroductions into predator-controlled areas are an increasingly used conservation tool. Typically, ecological outcomes of such projects (e.g. recruitment, predation) are the focus but seldom necessary legislative, policy, social and institutional processes required for establishing large projects. This particularly applies to protected areas, managed by governments for conservation. Reintroductions are recommended for a third of threatened Australian vertebrates, with the New South Wales Government boldly embarking on a 2013 initiative to return locally extinct mammals to three protected areas. We detail the legislative, policy, social and institutional processes required for one of these sites, Wild Deserts, in Sturt National Park. Seven locally extinct species, including the greater bilby (Macrotis lagotis), western barred bandicoot (Perameles bougainville), golden bandicoot (Isoodon auratus), greater stick-nest rat (Leporillus conditor), crest-tailed mulgara (Dasycercus cristicauda), western quoll (Dasyurus geoffroii) and burrowing bettong (Bettongia lesueur) will be reintroduced into two exclosures. The scale of the project required identification of an appropriate location, satisfaction of statutory requirements for major infrastructure in a national park, assessments of potential ecological costs and benefits, engagement of the national park agency and other stakeholders (Traditional Owners, other government agencies, neighbours), and staff training to meet government agency requirements. We outline the resourcing, costs and benefits of such a project on government-managed land along with lessons learnt for similar large-scale restoration and reintroduction projects. Future projects would benefit from understanding legislative and policy frameworks and the need for transparency, while maximising efficiencies. We outline the challenges and benefits associated with the establishment phase of a mammal reintroduction project on government-managed land. Research regarding the ergogenic effects of caffeine (CAF) in anaerobic activity remains inconclusive. However, some researchers have found significant improvements in anaerobic performance when testing specifically trained athletes. A double-blind, placebo-controlled, counterbalanced, cross-over design was implemented to assess the impact of CAF on a 30-second Wingate Anaerobic Test (WAnT) in experienced cyclists. Nine experienced cyclists volunteered to participate in this study (men, n = 7 and women, n = 2). The subjects completed 2 separate experimental trials consisting of a 30-second WAnT at a resistance of 9% body mass. In a random order, 1 hour before each WAnT, subjects ingested either a CAF (~280 mg) or placebo (PLAC) coffee. For each trial, heart rate (HR) and blood lactate (BL) values were recorded at rest, pre-WAnT, post-WAnT, and 5 minutes post-WAnT. After each trial, the subjects recorded their perception of which treatment they received. Heart rate and BL responses were not significantly different between the CAF and PLAC conditions. The ingestion of CAF did not significantly improve peak anaerobic power, mean anaerobic power, nor fatigue index. In at least 1 of the 2 trials, 44% of the subjects incorrectly guessed which substance they had ingested. The findings of this study do not show a significant correlation between CAF ingestion and improved anaerobic performance in experienced cyclists. However, performance enhancements may depend on varying individual responses to CAF. Context. Trapping of small vertebrates during their active hot summer periods is vital for conservation and impact assessment studies. Animal Ethics Committees (AECs) protect wildlife by enforcing arbitrary but restrictive temperature limits for trapping. Aims. Empirical data were gathered on the temperatures reached in different trap configurations to inform pragmatic ethical guidelines. Methods. Temperature was measured inside small vertebrate traps at two Australian arid zone sites to generate data on the thermal consequences of: (1) trap design and external shading; (2) provision of protective refuge substrates; and (3) timing of trap clearing. Key results. Shading and increased trap depth significantly reduced temperatures within pitfall traps. A conservative stressful upper temperature limit of 36°C was never exceeded inside deep, shaded, narrow pitfall traps at one study site and only between 1100 and 1300 hours on 3 days at the hotter site, despite ambient temperatures reaching over 42°C. By contrast, potentially lethal upper temperatures were reached in wider, shallower bucket pit traps on most days at both sites, even when optimal shading and refuge substrates were employed. Deployment of surface traps under vegetation and with additional shading significantly reduced maximum temperatures experienced. Temperatures inside shaded Elliott and funnel traps generally tracked ambient air temperatures and thus typically exceeded conservative threshold temperatures between 0700 and 1900 hours when ambient temperatures exceeded 36°C. Conclusions. Temperatures experienced in optimal deep, shaded traps when ambient temperatures exceeded 40°C were 31°C lower than surface temperatures and similar to temperatures recorded at 20 cm below the soil surface, where many species would typically take refuge at these times. Implications. Data suggest that deep (60 cm), narrow pitfall traps with elevated lids for shade and shelter substrate inside should enable trapping to be conducted safely in the study region during summer (December to February). This is even the case in extremely hot weather, as long as trapped animals are removed within 4 h of sunrise. Ecophysiological studies of thermal tolerance within optimum trap arrangements revealed by the present study will allow field ecologists and AECs to develop informed site-specific trapping protocols. Despite progressive treatments with tandem stem-cell transplantation, patients with incurable myeloma eventually succumb to relapsed or refractory disease if left untreated. Promising agents such as proteasome inhibitors and immunomodulating imide drugs (imids), including the newer-generation agent pomalidomide, in combination with lower-dose dexamethasone, have been shown to be effective and to significantly improve and prolong survival in pretreated patients. Although the incidence of pomalidomide hypersensitivity reaction (HSR) in this class of drugs is not as well known, we have documented cutaneous toxicity (grade 3 by the Common Terminology Criteria for Adverse Events, version 4) in 2 separate cases (not yet published). Because the imids are chemically, structurally, and pharmacologically similar, it is not unreasonable to consider possible cross-reactivity in pomalidomide recipients who developed hsr when receiving previous lines of imids. As a patient's advocate, it is only prudent to provide a responsible, and yet practical, means to better address cross-sensitivity for patients. Intervention with the use of a rapid desensitization program (RDP) as a preventive measure should be introduced before initiating pomalidomide. Such a proactive measure for the patient's safety will ensure a smooth transition into pomalidomide treatment. A hsr can be either related or non-related to immunoglobulin E. As imids become an essential treatment backbone for myeloma and other plasma-cell diseases, an increasing number of patients could experience skin and other life-threatening toxicities, resulting in unnecessary discontinuation of these life-prolonging agents. The development of efficient methods for the metathesis and phosphorylation reactions of disulfide compounds is of widespread interest due to their important synthetic utility in polymer, biological, medicinal and agricultural chemistry. Herein, we demonstrate the use of N-heterocyclic carbenes (NHCs) as versatile organocatalysts to promote these challenging reactions under mild conditions. This metal- and oxidant-free protocol is operationally simple with very short reaction times. The first three primary members of the non-benzenoid carbocyclic aromatic ion family, namely cyclopropenium, cyclopentadienide, and cycloheptatrienium (tropylium) ions, have planar cyclic structures with (4 n+2)π electrons in fully conjugated systems. They fulfill Hückel's rule for aromaticity and hence possess extraordinary stability. Since the historic discovery of tropylium bromide in the late 19th Century, these non-benzenoid aromatic ions have attracted a lot of attention because of their unique combination of stability and reactivity. The charge on the aromatic ions makes them more prone to nucleophilic/electrophilic reactions than the neutral benzenoid counterparts. Within the last seven years, there has been a large number of investigations in utilizing aromatic ions to mediate organic reactions. This manuscript summarizes the recommendations that emerged from the first Roundtable on Clinical Research in Immunoglobulin Light-chain Amyloidosis (AL), a meeting sponsored by the Amyloidosis Foundation (Clarkston, MI, USA) to develop a consensus of experts on a modern framework for clinical trial design and drug development in AL. Recent diagnostic and technical advances in AL, and updated consensus guidelines for assessing hematologic and organ responses, enable us to define study populations, appropriate end points, and other criteria for all phases of clinical research. This manuscript provides a framework for the design and conduct of systematic collaborative clinical research in AL to encourage more rapid testing of therapies and to expedite new drug development and approval. N-Heterocyclic olefins (NHOs) have very recently emerged as efficient promoters for several chemical reactions due to their strong Brønsted/Lewis basicities. Here we report the novel application of NHOs as efficient phase-transfer organocatalysts for synthetically important alkylation reactions on a wide range of substrates, further demonstrating the great potential of NHOs in organic chemistry. Parental care strategies have been widely investigated in shorebirds that undertake long-distance regular migrations. In contrast, virtually nothing is known of the parental care of nomadic, opportunistically breeding shorebirds, although the irregular and short-lived nature of their breeding potentially accentuates the trade-offs between investment in successive clutches and between the sexes. We investigated the incubation behaviour of the nomadic, opportunistically breeding Banded Stilt (Cladorhynchus leucocephalus). Seven nests, filmed continuously with day—night cameras over 2–7 days, revealed that both sexes contributed to incubation, with males apparently the sole carer during hatching and early brood-rearing; this is a possible adaptation, which enables females to quickly produce a second clutch if favourable conditions persist. All incubator changeovers occurred after dark; incubation shifts averaged 44.8 ± 10.9 (s.e.m.) h (n = 11, 17.5-139.6 h), the longest recorded for any shorebird. Incubation constancy averaged 96.5%; this high value is possibly an adaptation to high predation and the need for rapid embryonic development in the face of ephemeral resources for breeding. Long incubation shifts may be explained by extended foraging trips to distant areas of the partially inundated salt-lake surface, where food resources had been concentrated by wind-driven water movement. The Flock Bronzewing Phaps histrionica has declined significantly and undergone a northward range contraction in inland Australia since European settlement. In the southern parts of its range, including central northern South Australia, it is uncommonly recorded during times of significant ephemeral vegetation response, triggered by sustained above-average rainfall. In late 2013, while core habitats in Queensland were in extended drought and much of arid South Australia received below-average rainfall, an area of ~10 000 km2 between Roxby Downs and Lake Eyre South supported tens of thousands of Flock Bronzewings, with evidence of widespread successful breeding during September and October and contemporary range expansions of 150 km to the south and west. Through a network of local pastoralists, mining workers and biologists we collated >80 field observations from >40 observers across this region. Direct evidence of predation by Cats Felis catus was detected at a nest-site and in the stomach of a cat shot in the area. Seeds of annual shrubs Trichodesma and Phyllanthus species dominated the crop contents of two dead fledgling Bronzewings that were collected opportunistically. Although the Flock Bronzewing is well known for its irruptive nature, the magnitude and southerly extent of this sustained irruption are unprecedented in the literature and in the living memory of local observers. This event exemplifies the remarkable ability of arid-adapted birds to locate and exploit localised productive habitat within vast dynamic and stochastic landscapes, in this case recruiting new individuals to the population despite widespread unfavourable conditions within their core range. Background The aim was to investigate the percentage of asymptomatic patients presenting for routine optometric eye examinations that have pathology or pathology-related risk factors warranting referral for ophthalmological consultation. Methods This was a retrospective, cohort case study and the inclusion criteria for participants included: (i) the patient presented for routine optometric eye care during a specified period of time; (ii) the patient was found to have pathology (or showed enough risk of pathology) resulting in referral to an ophthalmologist; and (iii) a referral report was received from the consulting ophthalmologist stating the diagnosis and the treatment plan. The data set was further reviewed to indicate presenting symptoms and patient age. Adult patients, ages 20 to 64 years, were reviewed separately; this age group is not covered by provincial health services for routine eye care in Nova Scotia. Files were obtained from two clinics through an electronic charting program. A database was created that included date of referral, clinical reasons for the referral, diagnosis and treatment plan. Clinical reasons for referral were extracted from the referral letters and reports and sorted into six disease categories: age-related macular degeneration, cataract, glaucoma, diabetic retinopathy, retinopathy and 'other'. Results The overall referral rate for the combined data set was nine per cent for all ages; 2.4 per cent of the overall patients were asymptomatic. There was a similar number of asymptomatic patients referred in the adult (20 to 64 years) age group compared to all ages (2.5 per cent). Conclusion A significant number of patients that present for routine eye examinations without any symptoms indicative of ocular disease are subsequently found to have a degree of pathology or risk thereof requiring referral for ophthalmological consultation. Cyclophosphamide, bortezomib and dexamethasone (CyBorD) is a highly active three-drug induction regimen for untreated transplant-eligible multiple myeloma patients. Although CyBorD has been evaluated only in the phase 2 setting in a limited number of patients, its high efficacy and ease of administration have led to its widespread use. Given that clinical trial efficacy can overestimate real-life effectiveness, we reviewed our institutional experience with 109 newly diagnosed patients who were treated with CyBorD in a non-clinical trial setting. After a median of four cycles, overall response rate (ORR) and very good partial response rate or better (⩾VGPR) were 95 and 66%, respectively, comparable to phase 2 studies of CyBorD and other three/four-drug induction regimens. All patients subsequently underwent successful stem cell collection and upgraded responses to ORR 98% and ⩾VGPR 79% post transplant. At a median follow-up of 19.8 months after diagnosis, the 2-year OS probability was 95.3% (95%CI: 89-98). The presence of concurrent plasmacytoma at diagnosis was the only prognostic factor predicting poorer survival (HR=5.56; 95%CI: 0.92-33.74; P=0.03). CyBorD was well-tolerated, with no severe peripheral neuropathy and minimal hematologic toxicity. Multiple myeloma (MM) is incurable in virtually all patients due to the presence of innate and emergent drug-resistance. To identify potential drug resistance mechanisms in MM we used iTRAQ (isobaric tags for relative and absolute quantitation) mass spectrometry to compare protein expression profiles of drug-resistant (RPMI 8226-R5) and sensitive (RPMI 8226-S) isogenic cell lines. We identified selective overexpression of myristoylated alanine-rich C-kinase substrate (MARCKS) in drug-resistant R5 cells. MARCKS overexpression was also observed in several drug-resistant human myeloma cell lines (HMCLs) and in drug-resistant primary MM samples. Functionally, inhibition of MARCKS phosphorylation by enzastaurin or knockdown of the gene by RNAi significantly enhanced the sensitivity of resistant HMCLs and primary MM samples to bortezomib and to other anti-myeloma drugs, providing evidence that MARCKS can modulate drug response. Mechanistically, pMARCKS (phosphorylated form of MARCKS) was found to function as an E2F-1 cofactor to regulate SKP2 transcription. pMARCKS promoted cell-cycle progression by facilitating SKP2 expression, suppressing p27Kip1 and potentially counteracting drug-induced cell-cycle arrest by promoting Cyclin E/CDK2 activity. Importantly, MARCKS knockdown in combination with bortezomib treatment overcame bortezomib resistance, significantly inhibited tumor growth and prolonged host survival in a MM xenograft model. The role of auto-SCT in the treatment of multiple myeloma (MM) in the era of novel agents continues to evolve. It is now clear that the depth of response and clinical outcomes have significantly improved as a result of the combination of these strategies. However, not all patients with MM who undergo auto-SCT are able to sustain a meaningful response and 20% of patients relapse shortly after auto-SCT. In this study, we aimed to assess the impact of early relapse (ER) after auto-SCT on OS for MM patients undergoing single auto-SCT who had received novel agent-based induction regimens. All consecutive patients with MM undergoing single auto-SCT from January 2002 to September 2012 who had novel induction therapy were evaluated. A total of 184 patients were identified. The median OS and PFS for the group of transplanted patients were 93 and 25.4 months, respectively. Median time to relapse was 17.2 months with 40% having relapsed at the time of analysis. ER (<12 months post auto-SCT) was seen in 27 (36%) out of 75 patients who had relapsed, and median OS was significantly shorter than in those with non-ER. Multivariate analysis showed ER as the major independent prognostic factor for OS. On the basis of these findings, we conclude that not only attainment of a good response, but sustainability of it, appears to be a major prognostic variable in MM in the era of novel therapy. Patients with ER post auto-SCT should biologically be characterized in prospective studies to better understand the mechanisms of resistance associated with this particular entity. Numerous studies have reported the feasibility and safety of autologous SCT (ASCT) in patients with multiple myeloma (MM) and mild to moderate renal impairment, but there are limited data in dialysis-dependent patients. In this retrospective study, we reviewed the toxicities and efficacy outcomes of 33 MM patients with dialysis-dependent renal failure who underwent ASCT at our institution from 1998 to 2012. The most common grade 3 non-hematologic toxicities were mucositis (49%), infection (15%) and bleeding (6%). Atrial dysrhythmias (24%) and delirium (30%) of all grades were also common. Hematologic toxicities included febrile neutropenia (88%); and RBC and platelet transfusions were required by 71 and 100% of patients, respectively. Transplant-related mortality (TRM) was high at 15%, predominantly caused by septic shock. Response to ASCT was at least VGPR (very good PR) in 50%, PR in 46.2% and stable disease (SD) in 3.8%. Median OS was 5.6 years, comparable to our overall institutional data. Overall, seven patients became dialysis independent. We conclude that ASCT can be an effective treatment for dialysis-dependent MM patients, with high response rates and survival. ABSTRACT The electrical impedance behavior of gellan gum (GG), GG-carbon nanotube, and GG-carbon nanofiber hydrogel composites is reported. It is demonstrated that the impedance behavior of these gels can be modeled using a Warburg element in series with a resistor. Sonolysis (required to disperse the carbon fillers) does not affect GG hydrogel electrical conductivity (1.2 ± 0.1 mS/cm), but has a detrimental effect on the gel's mechanical characteristics. It was found that the electrical conductivity (evaluated using impedance analysis) increases with increasing volume fraction of the carbon fillers and decreasing water content. For example, carbon nanotube containing hydrogels exhibited a six- to sevenfold increase in electrical conductivity (to 7 ± 2 mS/cm) at water content of 82%. It is demonstrated that at water content of 95 ± 2% the electrical behavior of multiwalled nanotube containing hydrogels transitions (percolates) from transport dominated by ions (owing to GG) to transport dominated by electrons (owing to the carbon nanotube network). © 2014 Wiley Periodicals, Inc. There is no standard therapy for multiple myeloma relapsing after an autotransplant. We compared the outcomes of a second autotransplant (N=137) with those of an allotransplant (N=152) after non-myeloablative or reduced-intensity conditioning (NST/RIC) in 289 subjects reported to the CIBMTR from 1995 to 2008. NST/RIC recipients were younger (median age 53 vs 56 years; P<0.001) and had a shorter time to progression after their first autotransplant. Non-relapse mortality at 1-year post transplant was higher in the NST/RIC cohort, 13% (95% confidence interval (CI), 8-19) vs 2% (95% CI, 1-5, P0.001). Three-year PFS and OS for the NST/RIC cohort were 6% (95% CI, 3-10%) and 20% (95% CI, 14-27%). Similar outcomes for the autotransplant cohort were 12% (95% CI, 7-19%, P=0.038) and 46% (95% CI, 37-55%, P=0.001). In multivariate analyses, risk of death was higher in NST/RIC recipients (hazard ratio (HR) 2.38 (95% CI, 1.79-3.16), P<0.001), those with Karnofsky performance score<90 (HR 1.96 (95% CI, 1.47-2.62), P<0.001) and transplant before 2004 (HR 1.77 (95% CI, 1.34-2.35) P0.001). In conclusion, NST/RIC was associated with higher TRM and lower survival than an autotransplant. The Eastern Grass Owl, Tyto longimembris, has been recorded infrequently from arid zone springs, boredrains and wetlands in South Australia since first reported in the far north-east of South Australia in 1975. During the 2010-2012 La Niña event, rainfall in inland South Australia was at its highest in decades, resulting in irruptions of small native mammal populations, particularly the Long-haired Rat, Rattus villosissimus. We present seven new records of Grass Owls from five locations, including the first records from the western Lake Eyre Basin. Habitat and food resources are discussed in context with the sightings and we suggest methods for future surveys targeted at Grass Owls, along with reference to the management of the artificial bore drain habitats often used by the species. The article explores the importance of the Arcoona Lakes complex to waterbirds in arid northern South Australia from January 2007 to April 2010. It documents the 46 waterbird species, such as Freckled Duck, Banded Stilt, and Black Swans, using the complex and compares species composition and their success in breeding with the available records following the filling in 1989. It examines the relevance of the complex due to the prevalent drought conditions in the area. It also suggests that the complex be listed to Ramsar Convention on Wetlands 2005 for conservation.
