Breast cancer (BC) is one of the frequent tumors that seriously endanger the physical and mental well-being in women with strong heterogeneity, and its pathogenesis involves multiple risk factors. Depending on the type of BC, hormonal therapy, targeted therapy, and immunotherapy are the current systemic treatment options along with conventional chemotherapy. Despite significant progress in understanding BC pathogenesis and therapeutic options, there is still a need to identify new therapeutic targets and develop more effective treatments. According to recent sequencing and profiling studies, non-coding (nc) RNAs genes are deregulated in human cancers via deletion, amplification, abnormal epigenetic, or transcriptional regulation, and similarly, the expression of many ncRNAs is altered in breast cancer cell lines and tissues. The ability of single ncRNAs to regulate the expression of multiple downstream gene targets and related pathways provides a theoretical basis for studying them for cancer therapeutic drug development and targeted delivery. Therefore, it is far-reaching to explore the role of ncRNAs in tumor development and their potential as therapeutic targets. Here, our review outlines the potential of two major ncRNAs, long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) as diagnostic and prognostic biomarkers as well as targets for new therapeutic strategies in breast cancer. Introduction: Matrine (MT) is a potential resistance reversal agent. However, it remains unclear whether MT can reverse the resistance of Haemophilus parasuis (H. parasuis) to β-lactams, and, if so, by what mechanism MT works. Methods: We screened one cefaclor (CEC)-resistant strain (clinical strain C7) from eight clinical (H. parasuis) strains and determined the underlying resistance mechanism. Then, we investigated the reversal effect of MTon the resistance of this strain to CEC. Results and Discussion: The production of β-lactamase, overexpression of AcrABTolC system, and formation of biofilm might not be responsible for the resistance of clinical strain C7 to CEC. Fourteen mutation sites were found in four PBP genes (ftsI, pbp1B, mrcA, and prcS) of clinical strain C7, among which the mutation sites located in ftsI (Y103D and L517R) and mrcA (A639V) genes triggered the resistance to CEC. The minimum inhibitory concentration (MIC) of CEC against clinical strain C7 was reduced by two to eight folds after MT treatment, accompanied by the significant down-regulated expression of mutated ftsI and mrcA genes. Based on such results, we believed that MT could reverse the resistance of H. parasuis to CEC by inhibiting the mutations in ftsI and mrcA genes. Our research would provide useful information for restoring the antimicrobial activity of β-lactams and improving the therapeutic efficacy of Glässer's disease. [ABSTRACT FROM AUTHOR]

Heat shock proteins (HSPs) act as molecular chaperones and are widely distributed in all kinds of organisms. In this study, the small heat shock protein HSP19.5 gene was cloned from Bombyx mandarina (Bmm). Its open reading frame (ORF) is 504 bp in length, encoding 167 amino acids with the HSP20 characteristic domain. The relative molecular weight of the encoding protein was 19.5 kD, and the isoelectric point (pI) is 6.54. Multiple comparisons and phylogenetic analyses showed that the insect small heat shock proteins (sHSPs) are highly conservative. BmmHSP19.5 has the closest relationship with BmHSP19.5 of Bombyx mori. The results of real-time quantitative PCR showed that the BmmHSP19.5 was highly expressed in the testis and ovary of the 5th instar larvae. The expression level of BmmHSP19.5 gene was increased in all tested tissues under short-term high/low temperature stress. BmNPV can induce the expression of the BmmHSP19.5 gene in hemolymph and midgut. The expression of BmmHSP19.5 gene was up-regulated in the midgut of larvae under deltamethrin stress. Larval RNA interference of BmmHSP19.5 caused 36.1% insect mortality and reduced the oviposition amount by 34.7%; while the whole cocoon weight, cocoon shell rate, and unfertilized rate showed no significant differences. Inter- and intra-specific variations in phenotype are common and can be associated with genomic mutations as well as epigenomic variation. Profiling both genomic and epigenomic variants is at the core of dissecting phenotypic variation. However, an efficient targeted genotyping and epigenotyping system is lacking. We describe a new multiplex targeted genotyping and epigenotyping system called improved bulked-PCR sequencing (iBP-seq). We employed iBP-seq for the detection of genotypes and methylation levels of dozens of target regions in mixed DNA samples. iBP-seq can be adapted for the construction of linkage maps, fine mapping of quantitative-trait loci, and detection of genome editing mutations at a cost as low as $0.016 per site per sample. We developed an automated bioinformatics pipeline, including primer design, a series of bioinformatic analyses for genotyping and epigenotyping, and visualization of results. iBP-seq and its bioinformatics pipeline, available at http://zeasystemsbio.hzau.edu.cn/tools/ibp/, can be adapted to a wide variety of species. Background: The high incidence of post-stroke depression (PSD) during rehabilitation exerts a negative effect on the treatment and functional recovery of patients with stroke and increases the risk of mortality. It is necessary to screen PSD in the rehabilitation stage and thus provide effective intervention strategies. However, existing measurements used to assess PSD in the rehabilitation stage in patients with stroke lack specificity. This study aimed to develop a clinical measure to assess symptoms of PSD in the rehabilitation stage. Methods: The research team created the initial items through a literature review and semi-structured interviews of patients with stroke. Then, the symptom-related items were estimated by three panels: healthcare professionals (N = 41), Delphi experts (N = 15), and patients with stroke in the rehabilitation stage (N = 30). Results: The literature review and semi-structured interview produced 51 symptom-related items including six domains, and the items were reduced to 47 by the healthcare professionals. The symptom-related items were further reduced to 33 items by a two-round Delphi consultation. The initiative coefficients of the two Delphi rounds were 71.4 and 100%, the expert authority coefficients were both 0.85, Kendall's W were 0.152 and 0.408 (p < 0.01), and the coefficient of variation (CV) were 0.05-0.32 and 0.00-0.18, respectively. The item-level content validity index (I-CVI) was 0.53-1.00, the scale-level CVI/universal agreement (S-CVI/UA) was 0.26, and the S-CVI/average (S - CVI/Ave) was 0.85 for the first found Delphi consultation; the I-CVI was 0.67-1.00, the S-CVI/UA was 0.61, and the S-CVI/Ave was 0.97 for the second round Delphi consultation. All content validity indicators have been significantly improved compared with the first round. Using mean ≥ 4 and full score ≥ 0.5, combined with CV ≤ 0.16 as the item criteria, a clinical measure of PSD with 33 items and 6 dimensions (cognition, sleep, behavior, emotion, body, and guilt) was finally formed after two rounds. The patients with stroke made no further revisions after evaluation. Advances in the technology of wearable electronic devices have necessitated much research to meet their requirements, such as stretchability, sustainability, and maintenance-free functioning. In this study, we developed an ultrathin all-fiber triboelectric nanogenerator (TENG)-based electronic skin (TE-skin) with high stretchability, using electrospinning and spraying, whereby the silver nanowire (Ag NW) electrode layer is deposited between two electrospinning thermoplastic polyurethane (TPU) fibrous layers. Due to its extraordinary stretchability and prominent Ag NW conductive networks, the TE-skin exhibits a high sensitivity of 0.1539 kPa1 in terms of pressure, superior mechanical property with a low-resistance electrode of 257.3 W at a strain of 150%, great deformation recovery ability, and exceptional working stability with no obvious fluctuation in electrical output before and after stretching. Based on the outstanding performances of the TE-skin, an intelligent electronic glove was fabricated to detect multifarious hand gestures. Moreover, the TE-skin has the potential to record human motion for real-time physiological signal monitoring, which provides promising applications in the fields of flexible robots, human-machine interaction, and multidimensional sports monitoring in next-generation electronics. Objective: Weaning is an important stage in the life of young mammals, which is associated with intestinal inflammation, gut microbiota disorders, and even death. ß-Carotene displays anti-inflammatory and antioxidant activities, which can prevent the development of inflammatory diseases. However, whether ß-carotene can affect intestinal microbiota remains unclear. Methods: Twenty-four piglets were distributed into four groups: the normal suckling group (Con), the weaning group (WG), the weaning+ß-carotene (40 mg/kg) group (LCBC), and the weaning+ß-carotene (80 mg/kg) group (HCBC). The serum, jejunum, colon, and faeces were collected separately from each group. The effects of ß-carotene on the phenotype, overall structure, and composition of gut microbiota were assessed in weaning piglets. Results: The results showed that ß-carotene improved the growth performance, intestinal morphology and relieved inflammation. Furthermore, ß-carotene significantly decreased the species from phyla Bacteroidetes and the genus Prevotella, and Blautia, and increased the species from the phyla Firmicutes and the genera p-75-a5, and Parabacteroides compared to the WG group. Spearman's correlation analysis showed that Prevotella and Blautia were positively correlated, and Parabacteroides and Synergistes were negatively correlated with the levels of interleukin-1ß (IL-1ß), IL-6, and tumour necrosis factor-a (TNF-a), while p-75-a5 showed negative correlation with IL-6 in serum samples from piglets. Conclusion: These findings indicate that ß-carotene could alleviate weaning-induced intestinal inflammation by modulating gut microbiota in piglets. It is conjectured that every edge-colored complete graph G on n vertices satisfying Δmon (G) ≤ n -3k +1 contains k vertex-disjoint properly edge-colored cycles. We confirm this conjecture for k = 2, prove several additional weaker results for general k, and we establish structural properties of possible minimum counterexamples to the conjecture. We also reveal a close relationship between properly edge-colored cycles in edge-colored complete graphs and directed cycles in multipartite tournaments. Using this relationship and our results on edge-colored complete graphs, we obtain several partial solutions to a conjecture on disjoint cycles in directed graphs due to Bermond and Thomassen. Pollution of the marine environment by ship garbage is an urgent problem to be solved at home and abroad. The ship kitchen garbage vacuum collection system is a new environmental protection scheme for garbage disposal. It has many advantages, such as using a pipeline instead of manual operation, creating high-level sanitary conditions, realising completely closed garbage collection and transportation, eliminating cross-pollution, saving space and so on. In this paper, the system is modelled, and the calculation of pipeline pressure loss, the vacuum degree of the vacuum tank and the energy consumption of the system are briefly introduced. In order to reduce the energy consumption, an algorithm for the emptying and discharging port is presented. In order to solve the problem of optimising relevant parameters, the vacuum transport mechanism of garbage is studied based on an optimisation model of the pipe network, and the experimental platform of a simulation device is set up. ß-carotene is a robust modulator of mucosal barriers, and it can amplify the immunoglobulin A (IgA) response via the retinoic acid (RA)-mediated pathway. We investigated the influence of ß-carotene on intestinal barriers in layer-type cockerels. In this study, ß-carotene has a positive influence on growth performance and intestinal morphology. ß-carotene remarkably enhanced serum secretory immunoglobulin A (sIgA) levels, jejunal mucosal sIgA, and IgA concentrations. ß-Carotene significantly enhanced mRNA expression levels of IgA, CC chemokine receptor-9 (CCR9), polymeric immunoglobulin receptor (pIgR), and retinoic acid receptor a (RARa) in the ileal tissues and pIgR in the jejunal tissues. ß-Carotene improves mRNA expression of intestinal barrier-related proteins including: mucin-2 (MUC-2), zonula occludens-2 (ZO- 2), occludins (OCLN), and zonula occludens-1 (ZO-1) in the ileal tissues. Moreover, ß-carotene decreased the levels of Escherichia coli and elevates the levels of Lactobacillus. The results indicate that ß-carotene can promote growth performance and contribute to the gradual development of intestinal barriers in Hyline Brown chicks. Amajor challenge in transforming development to inclusive, sustainable pathways is the pervasive and persistent trade-off between provisioning services (e.g., agricultural production) on the one hand and regulating services (e.g., water purification, flood control) and biodiversity conservation on the other. We report on an application of China's new Ecological Development Strategy, now being formally tested and refined for subsequent scaling nationwide, which aims to mitigate and even eliminate these trade-offs. Our focus is the Ecosystem Function Conservation Area of Hainan Island, a rural, tropical region where expansion of rubber plantations has driven extensive loss of natural forest and its vital benefits to people. We explored both the biophysical and the socioeconomic options for achieving simultaneous improvements in product provision, regulating services, biodiversity, and livelihoods. We quantified historic trade-offs between rubber production and vital regulating services, finding that, over the past 20 y (1998-2017), there was a 72.2% increase in rubber plantation area, leading to decreases in soil retention (17.8%), water purification [reduced retention of nitrogen (56.3%) and phosphorus (27.4%)], flood mitigation (21.9%), carbon sequestration (1.7%), and habitat for biodiversity (6.9%). Using scenario analyses, we identified a two-pronged strategy that would significantly reduce these trade-offs, enhancing regulating services and biodiversity, while simultaneously diversifying and increasing product provision and improving livelihoods. This general approach to analyzing product provision, regulating services, biodiversity, and livelihoods has applicability in rural landscapes across China, South and Southeast Asia, and beyond. On the basis of introducing the nutrient composition and biogas fertilizer, the effects of biogas fertilizer on soil, crops and environment are summarized. Biogas fertilizer can improve soil structure, increase soil organic matter, available nutrient contents and enzyme activity, increase crop yield, quality and resistance, and relieve the non-point source pollution effectively. The harmlessness, application technology and risk are analyzed. Some suggestions are put forward to control the source poUution, strengthen the reseach of fermentation technology, define the standard of bioga fertilizer, and cary out large and medium-sized biogas engineering. In order to classify high spatial resolution images more accurately, in this research, a hierarchical rule-based objectbased classification framework was developed based on a high-resolution image with airborne Light Detection and Ranging (LiDAR) data. The eCognition® software is employed to conduct the whole process. In detail, firstly, the FBSP optimizer (Fuzzy-based Segmentation Parameter) is used to obtain the optimal scale parameters for different land cover types. Then, using the segmented regions as basic units, the classification rules for various land cover types are established according to the spectral, morphological and texture features extracted from the optical images, and the height feature from LiDAR respectively. Thirdly, the object classification results are evaluated by using the confusion matrix, overall accuracy and Kappa coefficients. Objective: Recent studies have challenged the traditional paradigm that growth hormone receptor (GHR) displays physiological functions only in the cell membrane. It has been demonstrated that GHR localizes to the cell nucleus and still exhibits important physiological roles. The phenomenon of nuclear localization of growth hormone (GH)-induced GHR has previously been described in vitro. However, until recently, whether GH could induce nuclear localization of GHR in vivo was unclear. Methods: In the present study, we used pig as an animal model, and porcine growth hormone (pGH) or saline was injected into the inferior vena cava. We subsequently observed the localization of porcine growth hormone receptor (pGHR) using multiple techniques, including, immunoprecipitation and Western-blotting, indirect immunofluorescence assay and electronmicroscopy. Results: The results showed that pGH could induce nuclear localization of pGHR. Taken together, the results of the present study provided the first demonstration that pGHR was translocated to cell nuclei under pGH stimulation in vivo. Conclusion: Nuclear localization of pGHR induced by the in vivo pGH treatment suggests new functions and/or novel roles of nuclear pGHR, which deserve further study. [ABSTRACT FROM AUTHOR]

The internalization and subcellular distribution of growth hormone (GH) from humans and rats have been extensively reported. However, to date, there are no descriptions of the cell behaviour of porcine GH in domestic animals. Thus, freshly isolated porcine hepatocytes were used as a somatic cell model to study the internalization and subcellular localization of porcine growth hormone (pGH), and the results showed that pGH could be internalized into porcine hepatocytes in a time-dependent manner. The results also showed that pGH could be translocated into cell nuclei in a time-dependent manner. In addition, the subcellular localization, according to immunoelectron microscopy, showed that pGH was localized in cytoplasmic organelles, e.g., the mitochondria and Golgi apparatus. The present study shows the first exploration of the behaviour of pGH in a somatic cell model of a domestic animal (pig), providing a basis for further studies on the physiological function of pGH based on its subcellular localization.
