A poly(methacrylic acid-co-ethylene glycol dimethacrylate)-based magnetic sorbent was used for the rapid and sensitive determination of tricyclic antidepressants and their main active metabolites in human urine. This material was characterized by magnetism measurements, zeta potential, scanning electron microscopy, nitrogen adsorption–desorption isotherms, and thermogravimetric analysis. The proposed analytical method is based on stir bar sorptive-dispersive microextraction (SBSDME) followed by liquid chromatography–tandem mass spectrometry. The main parameters involved in the extraction step were optimized by using the response surface methodology as a multivariate optimization method, whereas a univariate approach was employed to study the desorption parameters. Under the optimized conditions, the proposed method was properly validated showing good linearity (at least up to 50 ng mL−1) and enrichment factors (13–22), limits of detection and quantification in the low ng L−1 range (1.4–7.0 ng L−1), and good intra- and inter-day repeatability (relative standard deviations below 15%). Matrix effects were observed for the direct analysis of urine samples, but they were negligible when a 1:1 v/v dilution with deionized water was performed. Finally, the method was successfully applied to human urine samples from three volunteers, one of them consuming a prescribed drug for depression that tested positive for clomipramine and its main active metabolite. A rapid and simple analytical method based on liquid chromatography – ultraviolet detection has been developed and validated for the simultaneous determination of two high added-value cosmetic ingredients, honokiol and magnolol, in commercial cosmetic samples. These compounds from the Magnolia plant present demonstrated benefits for the skin, such as antioxidative and anti-inflammatory properties, besides of antimicrobial and antifungal capacity, which may lead to their possible use as alternative preservatives in cosmetic products. The proposed method has been successfully validated showing good linearity, limits of detection and quantification in the low µg mL−1 level (0.06 to 0.07 µg mL−1 and 0.19 to 0.24 µg mL−1, respectively) and good repeatability (relative standard deviation below 9%). The proposed method has been evaluated by analyzing a laboratory-made cosmetic sample containing known concentrations of the analytes, providing low relative errors (-6% for honokiol and −4% for magnolol) and quantitative recovery values (96 to 111%). Finally, the proposed analytical method has been applied to the determination of both analytes in eight commercial cosmetic samples with different properties, obtaining quantitative recovery values (93 to 115%) and thus showing that matrix effects are negligible. In summary, the results show that this firstly proposed method for honokiol and magnolol determination in cosmetic products is accurate and useful for the routinely quality control of these consumer products containing both ingredients. The world population grows with the tendency to concentrate in urban areas. Having food for everyone and correct information on nutrients and diet for everyone is included in the global scope of the United Nations Millennium Sustainable Development Goals (SDGs). Science and technology play a key role. Recently the term 'Ultra-processed' has become fashionable in certain circles related to nutrition. This term is generating a great deal of confusion in certain consumer groups and in the sector of food production, since its interpretation is controversial. This document analyses the reason for this confusion. From a legal point of view, the use of the expression or concept 'ultra-processed' by the political or administrative authorities could be sanctioned. In this context, both the European Commission and national governments could take measures to avoid the use of this expression, the proliferation of which confuses the consumer, influencing their purchasing decisions and legal security. Nor can it be overlooked that those companies, whose products are disparaged to potential buyers by this label, may take legal measures to compensate for the damages and loss caused. An analytical method for the simultaneous determination of bisphenol A, 4-t-octylphenol, 4-n-octylphenol, and 4-n-nonylphenol in cosmetic samples has been developed. These compounds have toxic effects on human health as they have shown to produce endocrine disrupting properties. Therefore, their presence in cosmetics should be avoided according to the current European Regulations on cosmetic products. The method is based on high-performance liquid chromatography coupled to mass spectrometry (HPLC-MS) detection. Standard addition calibration was used to avoid matrix effects. The limits of detection values ranged between 7 and 15 ng mL−1(threefold of the residual standard deviation of regression lines). The proposed method was validated, and good recovery (90–106%) and repeatability values (2.7–8.2%) were obtained. Finally, the method was successfully applied to ten commercially available cosmetic samples. Vortex-assisted emulsification semimicroextraction is proposed as a one-step solution-extraction procedure for sample preparation in cosmetic products. The procedure allows rapid preparation based on dispersion of the sample in a mixture of 1 mL of n-hexane and 0.5 mL of ethanol, followed by the addition of 0.5 mL of water and centrifugation to obtain two separated phases. This procedure provides good sample clean-up with minimum dilution and is very useful for the determination of ingredients with restricted concentrations, such as bronopol. The procedure was applied to the determination of bronopol by liquid chromatography with UV detection. The best chromatographic separation was obtained by using a C18 column set at 40 °C and performing a stepwise elution with a mixture of ethanol/aqueous 1 % acetic acid solution as mobile phase pumped at 0.5 mL min. The detection wavelength was set at 250 nm and the total run time required was 12 min. The method was successfully applied to 18 commercial cosmetic samples including creams, shampoos, and bath gels. Good recoveries and repeatability were obtained, with a limit of detection of 0.9 μg mL, which makes the method suitable for the analytical control of cosmetic products. Background The Small Hive Beetle (SHB) is considered one of the major threats to the long-term sustainability and economic success of honey bee colonies in Europe. The risk of introduction into the EU had been reported as moderate to high. Indeed, it has been recently reported an outbreak in the south of Italy. Here, the presence of Aethina tumida in beekeeping farms in Spain was evaluated using a previously described qPCR protocol. Findings When hive debris from 398 colonies (collected in 2010 and 2011) was analysed, grouped by region, SHB were not detected in any of the samples, making it unnecessary to analyse the samples individually. The microorganisms Ascosphaera apis, Paenibacillus larvae and Melissococcus plutonius are the three most important pathogens that affect honeybee brood. The aim of the present study was to evaluate the prevalence of these pathogens in honeybee colonies and to elucidate their role in the honeybee colony losses in Spain. In order to get it, a multiplex polymerase chain reaction ( PCR) assay was developed to simultaneously amplify the16 S ribosomal ribonucleic acid ( rRNA) gene of P. larvae and M. plutonius, and the 5.8 S rRNA gene of A. apis. The multiplex PCR assay provides a quick and specific tool that successfully detected the three infectious pathogens ( P. larvae, M. plutonius and A. apis) in brood and adult honeybee samples without the need for microbiological culture. This technique was then used to evaluate the prevalence of these pathogens in Spanish honeybee colonies in 2006 and 2007, revealing our results a low prevalence of these pathogens in most of the geographic areas studied. Vitamin A (retinol) and some of its derivatives are a group of fat-soluble compounds used in cosmetic products as bioactive ingredients. Therefore, it is necessary to perform the quality control of final product to ensure their efficacy and safety. A simple and rapid method to determine retinol, retinal, retinyl acetate, retinyl propionate and retinyl palmitate in cosmetics is presented here. The method is based on vortex and/or ultrasound-assisted leaching of the analytes in ethanol followed by liquid chromatography with ultraviolet detection. The analytical performance of the method was evaluated. It has shown high levels of linearity, at least up to 100 µg mL−1; high precision with RSD values below 14% and high sensitivity with low MLODs ranging between 0.3 × 10−4 and 5.9 × 10−4 % w/w, which are enough to monitor these compounds in cosmetic products. The proposed method was successfully applied to seven commercial cosmetic samples to detect and quantify the target analytes, showing the method is suitable for its employment for quality control in cosmetic industries. Cosmetic samples were spiked at two levels of concentration and recovery values around 100% were obtained, showing no significant matrix effects and, therefore, external calibration was adequate for this determination. A growing awareness of the risks associated with skin exposure to ultraviolet (UV) radiation over the past decades has led to increased use of sunscreen cosmetic products leading the introduction of new chemical compounds in the marine environment. Although coastal tourism and recreation are the largest and most rapidly growing activities in the world, the evaluation of sunscreen as source of chemicals to the coastal marine system has not been addressed. Concentrations of chemical UV filters included in the formulation of sunscreens, such as benzophehone 3 (BZ-3), 4-methylbenzylidene camphor (4-MBC), TiO2 and ZnO, are detected in nearshore waters with variable concentrations along the day and mainly concentrated in the surface microlayer (i.e. 53.6–577.5 ng L-1 BZ-3; 51.4–113.4 ng L-1 4-MBC; 6.9–37.6 µg L-1 Ti; 1.0–3.3 µg L-1 Zn). The presence of these compounds in seawater suggests relevant effects on phytoplankton. Indeed, we provide evidences of the negative effect of sunblocks on the growth of the commonly found marine diatom Chaetoceros gracilis (mean EC50 = 125±71 mg L-1). Dissolution of sunscreens in seawater also releases inorganic nutrients (N, P and Si forms) that can fuel algal growth. In particular, PO43− is released by these products in notable amounts (up to 17 µmol PO43− g−1). We conservatively estimate an increase of up to 100% background PO43− concentrations (0.12 µmol L-1 over a background level of 0.06 µmol L-1) in nearshore waters during low water renewal conditions in a populated beach in Majorca island. Our results show that sunscreen products are a significant source of organic and inorganic chemicals that reach the sea with potential ecological consequences on the coastal marine ecosystem. Nosema ceranae has been suggested to be replacing Nosema apis in some populations of Apis mellifera honeybees. However, this replacement from one to the other is not supported when studying the distribution and prevalence of both microsporidia in professional apiaries in Spanish territories (transverse study), their seasonal pattern in experimental hives with co-infection or their prevalence at individual level (either in worker bees or drones). Nevertheless, N. ceranae has shown to present a higher prevalence at all the studied levels that could indicate any advantage for its development over N. apis or that it is more adapted to Spanish conditions. Also, both microsporidia show a different pattern of preference for its development according to the prevalence in the different Spanish bioclimatic belts studied. Finally, the fact that all analyses were carried out using an Internal PCR Control (IPC) newly developed guarantees the confidence of the data extracted from the PCR analyses. Nosema ceranae is a relatively new and widespread parasite of the western honeybee Apis mellifera that provokes a new form of nosemosis. In comparison to Nosema apis, which has been infecting the honeybee for much longer, N. ceranae seems to have co-evolved less with this host, causing a more virulent disease. Given that N. apis and N. ceranae are obligate intracellular microsporidian parasites, needing host energy to reproduce, energetic stress may be an important factor contributing to the increased virulence observed. Through feeding experiments on caged bees, we show that both mortality and sugar syrup consumption were higher in N. ceranae-infected bees than in N. apis-infected and control bees. The mortality and sugar syrup consumption are also higher in N. apis-infected bees than in controls, but are less than in N. ceranae-infected bees. With both microsporidia, mortality and sugar syrup consumption increased in function of the increasing spore counts administered for infection. The differences in energetic requirements between both Nosema spp. confirm that their metabolic patterns are not the same, which may depend critically on host-parasite interactions and, ultimately, on host pathology. The repercussions of this increased energetic stress may even explain the changes in host behavior due to starvation, lack of thermoregulatory capacity, or higher rates of trophallaxis, which might enhance transmission and bee death. novel and simple device for membrane-assisted liquid-phase microextraction is used for the first time in a three-phase system. The device consists of a glass vial containing the aqueous acceptor phase, whose septum of its screw stopper has been replaced by a sized piece of polytetrafluoroethylene membrane impregnated with n-decane. The vial is assembled to a volumetric flask containing the aqueous donor phase, and the membrane comes in contact alternatively with both donor and acceptor aqueous phases by orbital agitation. The device has been tested for the determination of nitrite in tap water samples, which is extensively carried out in routine analysis, as model analytical application. Experimental variables, such as the organic solvent used to form the supported liquid membrane, the volumes of both donor and acceptor phases, the orbital agitation rate, and the extraction time were studied and optimized in terms of enrichment factor. Under the selected working conditions, the analytical figures of merit for nitrite determination were a linearity range up to 50 ng mL, limits of detection and quantification of 0.15 and 0.50 ng mL, respectively, and a good repeatability (RSD < 10%). The method has been applied to four tap water samples of different origins, and accurate and precise results were achieved. 2-Hydroxy-4-methoxybenzophenone (HMB), which is one of the most commonly used UV filters in sunscreen cosmetics to protect skin from the deleterious effects of the sun, can be percutaneously absorbed, further metabolized, and finally excreted or bioaccumulated. An analytical method for the sensitive determination of HMB and its three metabolites in both human urine and semen is developed. The presented analytical method is based on a solid-phase extraction (SPE) procedure to clean-up and preconcentrate the target analytes from the urine and semen samples followed by liquid chromatography–tandem mass spectrometry (LC-MS/MS) detection. The methodology was fully validated and the standard addition calibration method was used to quantify the target analytes in order to correct the matrix effects observed. Considering this approach, the accuracy of the method was evaluated and the recoveries ranged from 98% to 115% and from 86% to 111% in urine and semen samples, respectively, depending on the analyte. For urine samples, the limits of detection ranged between 0.027 and 0.103 ng mL and the repeatability of the method, expressed as relative standard deviation, was in the range of 7.2–9.2%, depending on the analyte. In the case of semen samples, the limits of detection ranged between 1 and 3 ng mL whereas the repeatability was in the range of 2.2–6.4%, depending on the analyte. The described SPE-LC-MS/MS method was satisfactorily applied to both urine and semen samples from a male volunteer who applied a sunscreen cosmetic product containing HMB. Summary In recent years, a worldwide decline in the Apis mellifera populations has been detected in many regions, including Spain. This decline is thought to be related to the effects of pathogens or pesticides, although to what extent these factors are implicated is still not clear. In this study, we estimated the prevalence of honey bee colony depopulation symptoms in a random selected sample ( n = 61) and we explored the implication of different pathogens, pesticides and the flora visited in the area under study. The prevalence of colony depopulation symptoms in the professional apiaries studied was 67.2% [95% confidence interval (CI) = 54.6-79.8; P < 0.0001]. The most prevalent pathogen found in the worker honey bee samples was Nosema ceranae[65.6%; 95% CI = 52.8-78.3; P < 0.0001], followed by Varroa destructor[32.7%; 95% CI = 20.2-45.4; P < 0.0001] and 97.5% of the colonies infected by N. ceranae were unhealthy (depopulated). Co-infection by V. destructor and N. ceranae was evident in 22.9% (95% CI = 11.6-34.3; P < 0.0001) of the samples and only in unhealthy colonies. Of the 40 pesticides studied, only nine were detected in 49% of the stored pollen samples analysed. Fipronil was detected in only three of 61 stored pollen samples and imidacloprid was not detected in any. Acaricides like fluvalinate, and chlorfenvinphos used to control Varroa mite were the most predominant residues in the stored pollen, probably as a result of their application in homemade formulae. None of the pesticides identified were statistically associated to colony depopulated. This preliminary study of epidemiological factors suggests that N. ceranae is a key factor in the colony losses detected over recent years in Spain. Nowadays, 2-ethylhexyl 4-( N,N-dimethylamino)benzoate (EDP) is one of the most widely used UV filters in sunscreen cosmetics and other cosmetic products. However, undesirable processes such as percutaneous absorption and biological activity have been attributed to this compound. The in vitro metabolism of EDP was elucidated in the present work. First of all, the phase I biotransformation was studied in rat liver microsomes and two metabolites, N,N-dimethyl- p-aminobenzoic acid (DMP) and N-monomethyl- p-aminobenzoic acid (MMP), were identified by GC-MS analysis. Secondly, the phase II metabolism was investigated by means of LC-MS. The investigated reactions were acetylation and glucuronidation working with rat liver cytosol and with both human and rat liver microsomes, respectively. Analogue studies with p-aminobenzoic acid (PABA) were carried out in order to compare the well established metabolic pathway of PABA with the unknown biotransformation of EDP. In addition, a method for the determination of EDP and its two phase I metabolites in human urine was developed. The methodology requires a solid-phase extraction prior to LC-MS analysis. The method is based on standard addition quantification and has been fully validated. The repeatability of the method, expressed as relative standard deviation, was in the range 3.4–7.4% and the limit of detection for all quantified analytes was in the low ng mL−1 range. The potential use of oxidative stress products as disease markers and progression is an important aspect of biomedical research. In the present study, the quantification of urine 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxo-dG) concentration has been used to express the oxidation status of hypertensive subjects. 8-oxo-dG has been simultaneously isolated and assayed in nuclear (nDNA) and mitochondrial DNA (mtDNA). In addition, oxidative stress of mononuclear cells has been estimated by means of GSH and GSSG levels and GSSG/GSH ratio in hypertensive subjects before and after antihypertensive treatment. It is shown that oxidative stress decreases significantly in hypertensive patients after treatment the effect being accompanied by reduction of their blood pressure. A significant correlation is observed comparing the yield of urine 8-oxo-dG and that isolated from mitochondria DNA. Moreover, urinary excretion of 8-oxo-dG also correlates with the GSSG/GSH ratio of cells. Conclusion: urine 8-oxo-dG assay is a good marker for monitoring oxidative stress changes in hypertensives. A sequential injection method is proposed for spectrophotometric determination of p-aminobenzoic acid (PABA) in cosmetic formulations. The method is based on diazotization of the analyte, coupling with 8-hydroxyquinoline, and the subsequent formation of a colored product. The experimental conditions used (coupling reagent, sandwich arrangement, volumes aspirated, propulsion flow rate, reaction coil length) were studied. Response of the sequential injection method were linearly dependent on concentrations up to 25 µg mL–1 and the detection limit was 2 µg mL–1. Throughput was 51 measurements per hour and a complete cycle, including three measurement per sample and a washing step, resulted in a sample throughput of 13.8 h–1. Sunscreen formulations containing PABA were analyzed by the proposed method and validated by use of an HPLC procedure. The two methods gave comparable results. Yields ranged between 97–103% and good precision (RSD between 2 and 6%) was obtained. Simultaneous determination of six internationally authorised organic UV-filters in sunscreen formulations was performed by HPLC with UV spectrophotometric detection. The filters determined were: sulisobenzone, oxybenzone, octyl dimethyl PABA, octyl methoxycinnamate, octyl salicylate and homosalate. A C18 stationary phase and a mobile phase of ethanol water acetic acid (70 : 29.5 : 0.5) were used with a flow rate of 0.5 mL/min. UV measurements were carried out at 313 nm. The time required for the analysis was 25 min and the limits of detection were between 0.2 and 2 mg/L, except for sulisobenzone, which gave a limit of detection of 20 mg/L.
