Neuroblastoma (NB) is the most commonly diagnosed extracranial solid tumor in children, accounting for 15% of all childhood cancer deaths. Although the 5-year survival rate of patients with a high-risk disease has increased in recent decades, NB remains a challenge in pediatric oncology, and the identification of novel potential therapeutic targets and agents is an urgent clinical need. The RNA-binding protein LIN28B has been identified as an oncogene in NB and is associated with a poor prognosis. Given that LIN28B acts by negatively regulating the biogenesis of the tumor suppressor let-7 miRNAs, we reasoned that selective interference with the LIN28B/let-7 miRNA interaction would increase let-7 miRNA levels, ultimately leading to reduced NB aggressiveness. Here, we selected (−)-epigallocatechin 3-gallate (EGCG) out of 4959 molecules screened as the molecule with the best inhibitory activity on LIN28B/let-7 miRNA interaction and showed that treatment with PLC/PLGA-PEG nanoparticles containing EGCG (EGCG-NPs) led to an increase in mature let-7 miRNAs and a consequent inhibition of NB cell growth. In addition, EGCG-NP pretreatment reduced the tumorigenic potential of NB cells in vivo. These experiments suggest that the LIN28B/let-7 miRNA axis is a good therapeutic target in NB and that EGCG, which can interfere with this interaction, deserves further preclinical evaluation. Natural α-glucosidase inhibitors from plant-based foods such as catechins offer an attractive strategy for their potential anti-diabetic effects. In this study, infusions of three different tea types (green, white, and oolong) were investigated for their total phenolic (TPC) and catechins (EGCG, ECG, EGC, and EC) content, and for their α-glucosidase inhibitory activities. We observed that the level of TPC in white tea was significantly higher compared to oolong and green tea, which suggests higher content of EGCG and ECG catechins in fresh young leaves. Our findings showed that the higher content of such catechins in the infusion of white tea well correlated with a strong inhibition of α-glucosidase, and such inhibition was demonstrated to be more effective than the FDA-approved drug acarbose. Then, we computationally explored the molecular requirements for enzyme inhibition, especially for the most active catechins EGCG and ECG, as well as their disposition/stability within the active site. Nanoparticles are widely studied for applications in medical science. In recent years, they have been developed for agronomical purposes to target microbial pest such as bacteria, fungi, and viruses. Nanoparticles are also proposed to limit the use of pesticides, whose abuse is causing environmental impact and human health concerns. In this study, nanoparticles were obtained by using poly-(ε-caprolactone), a polyester chosen for its biocompatibility and biodegradability properties. Poly-(ε-caprolactone) nanoparticles were formulated by using poly(vinyl alcohol) or Pluronic® F127 as non-ionic surfactants, and then loaded with benzophenone or valerophenone thiosemicarbazone, two compounds that inhibit aflatoxin production by Aspergillus flavus. The different types of nanoparticles were compared in terms of size, polydispersity index, morphology, and drug loading capacity. Finally, their effects were investigated on growth, development, and aflatoxin production in the aflatoxigenic species Aspergillus flavus, a ubiquitous contaminant of maize, cereal crops, and derived commodities. Aflatoxin production was inhibited to various extents, but the best inhibitory effect was obtained with respect to sclerotia production that was most effectively suppressed by both benzophenone and valerophenone thiosemicarbazone-loaded nanoparticles. These data support the idea that it is possible to use such nanoparticles as an alternate to pesticides for the control of mycotoxigenic sclerotia-forming fungi. [ABSTRACT FROM AUTHOR]

In this study, the feasibility of frontal polymerization (FP) as an alternative and convenient technique for the preparation of semi-interpenetrating polymer networks made of methyl cellulose (MC) and cross-linked polyacrylamide (PAAm) is demonstrated. FP was performed in water and glycerol, as largely available, nontoxic solvents. Although FP occurred in both media, differences were found by comparing the samples made in the two solvents. In particular, those prepared in water are characterized by larger inhomogeneity and less reproducibility, thus accounting for the boiling effects that influence propagating polymerization fronts when water was used. The effects of the ratio among MC and PAAm, the amount of cross-linker and solvent medium were studied in terms of influence on temperature and velocity of FP fronts, glass transition temperature (dried samples), swelling behavior, dynamic-mechanical properties (gels swollen in both water or glycerol), and tensile behavior. ABSTRACT Polydiolcitrates are an emerging class of biocompatible polyesters with a great potential in the field of biomedicine and packaging for food and drug materials. In this work, a new type of (co-)polydiolcitrates made of citric acid (CA) and ethylene glycol (EG) and/or poly(ethylene glycol) (PEG) is investigated. By varying both the EG/PEG and the CA/diol molar ratios, materials exhibiting very different swelling behavior, mechanical and thermal properties are obtained. In particular, the substitution of EG segments with longer and flexible PEG ones results in an increase in crosslinking density, with remarkable effects on swelling capacity, glass transition temperature, and Young modulus. Moreover, polyesters with CA/diol molar ratio equal to 1:1 exhibit shape memory properties, with full capacity of keeping the temporary shape and high capacity of recovering the original shape. The HIV-1 ribonuclease H (RNase H) function of the reverse transcriptase (RT) enzyme catalyzes the selective hydrolysis of the RNA strand of the RNA:DNA heteroduplex replication intermediate, and represents a suitable target for drug development. A particularly attractive approach is constituted by the interference with the RNase H metal-dependent catalytic activity, which resides in the active site located at the C-terminus p66 subunit of RT. Herein, we report results of an in-house screening campaign that allowed us to identify 4-[4-(aryl)-1H-1,2,3-triazol-1-yl]benzenesulfonamides, prepared by the "click chemistry" approach, as novel potential HIV-1 RNase H inhibitors. Three compounds (9d, 10c, and 10d) demonstrated a selective inhibitory activity against the HIV-1 RNase H enzyme at micromolar concentrations. Drug-likeness, predicted by the calculation of a panel of physicochemical and ADME properties, putative binding modes for the active compounds, assessed by computational molecular docking, as well as a mechanistic hypothesis for this novel chemotype are reported. ABSTRACT Frontal polymerization was successfully used to synthesize copolymer hydrogels of poly( N-vinylcaprolactam- co-itaconic acid). All materials were characterized by response to stimuli (pH and/or temperature), depending on the itaconic acid content. Namely, relatively low amounts of this latter were found to be crucial for determining the degree of swelling. In particular, hydrogels behave differently if swollen at pH values that are higher or lower of 7-8, and exhibit temperature response as well (lower critical solution temperature at ca. 30 °C), which makes these materials potentially interesting for biomedical applications. © 2016 Wiley Periodicals, Inc. Polymeric nanocomposites were prepared by inserting graphene and/or Ag, Au and ZnO nanoparticles in a TEGDA (tetraethyleneglycol diacrylate) polymer matrix. The polymeric films were characterized in terms of their dielectric properties by electrochemical impedance spectroscopy. The impedance data were fitted by generalized relaxation functions in order to determine conductivity, dielectric response and molecular relaxation time of the nanocomposite films. Chemoprevention of human cancer(s) is a viable option for cancer control, especially when chemopreventive intervention is involved during the early stages of the carcinogenesis process. Naturally occurring bioactive food components, such as dietary polyphenols, have shown good antioxidant activity and other beneficial activities. In addition, compounds belonging to the polyphenolic chemical class may play promising roles in cancer prevention. Among them, the phytoalexin resveratrol has demonstrated antiproliferative effects, as well as the ability to inhibit initiation and promotion of induced cancer progression in a wide variety of tumor models. However, resveratrol, like other natural polyphenols, is an extremely photosensitive compound with low chemical stability and limited bioavailibility, which limit the therapeutic application of its beneficial effects. In this context, the development of innovative formulation strategies able to overcome physicochemical and pharmacokinetic limitations of this compound could be beneficial. This may be achieved via nanotechnology approaches utilizing suitable carriers that allow slow, sustained, and controlled release of the encapsulated agent. Beer is a complex mixture of bioactive polyphenols with positive health effects, and its composition is influenced by some technological variables. In this study, the total polyphenol content (TPC) and the antioxidant activity of some Italian craft beers produced by ageing in wood barrel that previously contained red or white wines, and by addition of sapa, a cooked must from wine grapes, were investigated. TPC greatly vary among different beer types. Craft beers stored in wood barrels that contained red wines, such as those added of sapa from red wine grapes, showed a TPC up to 2.5 times higher (1000 GAE mg L-1) compared with other craft beers, and 3.6 times higher than the industrial beer. The antioxidant activity is remarkably different depending on beer type and strictly correlated with TPC. Results indicate that antioxidant properties of the craft beers could be efficiently improved by the explored technological processes. [ABSTRACT FROM AUTHOR]

ABSTRACT A new series of linear and crosslinked copolymers, obtained from 3-octyl-1-vinylimidazolium bromide (VImBr) and N-isopropylacrylamide (NIPAAm), were prepared by radical polymerization. Namely, VImBr was synthesized from 1-bromooctane and an ionic liquid such as 1-vinylimidazole. NIPAAm was used because it gives raise to well known thermoresponsive (co-)polymers. The copolymers were thoroughly characterized by means of 1H NMR and 13C NMR spectroscopies. Besides, differential scanning calorimetry, Fourier transform infrared spectroscopy, and scanning electron microscopy were also used. Moreover, the swelling behavior and the thermoresponsive properties of the corresponding hydrogels were studied. It was found that the VImBr incorporation into the copolymers does have a dramatic influence on both the thermal properties of the dried materials and the lower critical solution temperature of the corresponding hydrogels. In detail, the glass transition temperature was dependent on the monomer ratios, and ranged from 5 to 155 °C. Analogously, the lower critical solution temperature of the resulting hydrogels ranged from less than 10 up to 38 °C, thus including the physiological temperature. NMR spectroscopies, which were performed on the linear polymers, indicated that the monomers exhibit an alternating tendency resulting in a microstructure in which blocks are not present, at least when the two monomers are in equimolar amounts. © 2014 Wiley Periodicals, Inc. In the course of our work aimed at developing novel heterocycles of pharmaceutical interest, we designed and synthesized several polycyclic templates as potential substrates to be used in drug design. We obtained a set of condensed ring systems as versatile structural platforms to generate potential DNA-interactive agents and/or reversible inhibitors of enzymes such as topoisomerases, poly(ADP-ribose) polymerase-1 (PARP-1), telomerase, and, in particular, cyclin dependent kinases. A specific and sensitive high-performance liquid chromatographic method was developed for the determination of metoclopramide in canine plasma. The procedure involves fast liquid-liquid extraction and analysis on an octadecyl silane (ODS) column. A preliminary pharmacokinetic study was performed by applying the developed method to a single oral administration of metoclopramide (MCP) to a dog. The validation method yielded good results regarding linearity, precision, accuracy, and specificity. The procedure is suitable for separation and quantification of MCP in canine plasma, enough to quantify 0.2 ng/ml when 0.5 ml of plasma is used. Solid lipid microparticles (SLM) were used as carriers of juniper oil and proposed for the topical treatment of acne vulgare . The formulations were obtained by the o/w emulsification method. Compritol and Precirol were employed as lipidic materials. Emulsions containing 1.5% (w/w) of lipophilic phase (lipid and oil) and two different lipid to oil ratios (1:1 and 2:1) were prepared. Blank particles were also prepared, as a comparison. The SLM were characterized in terms of encapsulation efficiency, size, and morphology. The particle size stability in aqueous dispersions was monitored over one month. Evaporation of volatile compounds of oil from microparticles by weight loss was investigated. The qualitative composition of Juniper oil before and after the encapsulation process was determined by gas chromatography (GC) and gas chromatography/mass spectrum (GC/MS) analyses. The antimicrobial activity of the oil encapsulated into the lipid microparticles against P. acnes was studied as contact time assay and compared to the activity of the oil not encapsulated. The emulsification method here described was a good technique for the encapsulation of essential oils. Percentage yields of production and encapsulation efficiencies were higher for Compritol preparations than for these prepared using Precirol. All preparations were characterized by similar particle size distributions (dvs about 3–4 µm) regardless of lipid type and lipid to oil ratios. Microscopy observations showed that the microparticles in aqueous dispersions had almost spherical shape, independently from their composition. The scanning electron microscopy (SEM) analyses showed that when the particles were dried, they had an irregular shape and a rough surface. The SLM dispersions based on Compritol revealed particle size stability over the investigated period of 30 days. In contrast, an increase of the mean dimensions in the preparations containing Precirol was observed. A low loss of volatile oil compounds owing to evaporation from dry particles was found in all preparations. This indicated that the microparticles were able to substantially maintain the oil loaded inside their lipidic structure, reducing its volatility. Some modifications of composition were found in the oil encapsulated in SLM with respect to the juniper oil raw material, but these modifications did not decrease the antibacterial activity of the oil. Rumen-stable devices ensure a protection of active ingredients against chemical degradation and bacterial fermentation processes that occur in the rumen. These systems should also provide postruminal bioavailability and controlled release of the active ingredient. The objective of this study was the preparation of bilayer tablets as rumen-stable delivery systems, designed for the oral administration of active ingredients (folic acid) to ruminants. The tablets are composed of two layers: layer A (“high-density layer”) constituted by poly (ℇ-caprolactone) mixed with iron powder and characterized by sufficient density to avoid rumination; layer B (“release layer”), containing folic acid (25 mg), poly (ℇ-caprolactone) or polymethylmethacrylates (Eudragit RS and RL) designed to be rumen-stable and to target a controlled release of folic acid in the intestinal tract. In vitro rumen-protection tests were performed in buffer systems at pH 5.5 and pH 2.0, simulating a ruminal and abomasal environment, to verify the stability of bilayer tablets at these conditions. In vitro release tests were carried out in phosphate buffer at pH 7.4, to study the release behavior of the dosage forms in the intestinal environment. A preliminary in vivo test was carried out with radiographic images made after administration of the tablets to sheep, to evaluate their capacity to be retained in the reticulum–rumen. The amount of iron powder used provides a density of about 2.3 g/cm3 to the whole tablet. The tablets having layer B constituted by poly (ℇ-caprolactone) or Eudragit RS do not disintegrate in buffer media at pH 5.5 and pH 2.0, and they are characterized by a sustained release at pH 7.4. The purpose of this research was to investigate the effects of processing conditions on the characteristics of solid lipid microparticles (SLM) with a potential application as carriers for pulmonary administration. Compritol (5.0% wt/wt) SLM dispersions were prepared by rotor-stator homogenization, at different surfactant concentrations and emulsification times. The SLM were characterized, in terms of morphology and size, after lyophilization and sterilization by autoclaving process. In vivo assessment was carried out in rats by intratracheal instillation of either placebo or SLM dispersion, and by bronchoalveolar lavage for cytological analysis. Mean particle size of 4 to 5 μm was achieved using 0.3% and 0.4% (wt/wt) of emulsifier (Poloxamer 188) and emulsification times of 2 and 5 minutes. The particles showed spherical shape and smooth surface. The morphology of microparticles, the size, and the size distribution were not substantially modified after lyophilization and sterilization. Total cell counts showed no significant differences between placebo and SLM 0.5% or 2.5% groups. Regarding cytology, percentage of polymorphonuclear neutrophils and macrophages did not significantly differ between groups. These results suggest that a single intratracheal administration of the SLMs does not induce a significant inflammatory airway response in rats and that the SLMs might be a potential carrier for encapsulated drug via the pulmonary route. The aim of the study was the development of mucoadhesive vaginal tablets designed for the local controlled release of acriflavine, an antimicrobial drug used as a model. The tablets were prepared using drug-loaded chitosan microspheres and additional excipients (methylcellulose, sodium alginate, sodium carboxymethylcellulose, or. Carbopol 974). The microspheres were prepared by a spray-drying method, using the drug to polymer weight ratios 1∶1 and 1∶2 and were characterized in terms of morphology, encapsulation efficiency, and in vitro release behavior, as MIC (Minimum Inhibitory Concentration), MBC (Minimum Bacterial Concentration), and killing time (KT). The tablets were prepared by direct compression, characterized by in vitro drug release and in vitro mucoadhesive tests. The microparticles have sizes of 4 to 12 μm; the mean encapsulation yields are about 90%. Acriflavine, encapsulated into the polymer, maintains its antibacterial activity; killing time of the encapsulated drug is similar to that of the free drug. In vitro release profiles of tablets show differences depending on the excipient used. In particular Carbopol 974, which is highly cross-linked, is able to determine a drug-controlled release from the matrix tablets for more than 8 hours. The in vitro adhesion tests, carried out on the same formulation, show a good adhesive behavior. The formulation containing microspheres with drug to polymer weight ratios of 1∶1 and Carbopol 974 is characterized by the best release behavior and shows good mucoadhesive properties. Earlier we introduced the concept of 'nanochemoprevention' i.e. the use of nanotechnology to improve the outcome of cancer chemoprevention. Here, we extended our work and developed polymeric EGCG-encapsulated nanoparticles (NPs) targeted with small molecular entities, able to bind to prostate specific membrane antigen (PSMA), a transmembrane protein that is overexpressed in prostate cancer (PCa), and evaluated their efficacy in preclinical studies. First, we performed a molecular recognition of DCL- and AG-PEGylation on ligand binding on PSMA active site. Next, the biocompatible polymers PLGA-PEG-A were synthesized and used as base to conjugate DCL or AG to obtain the respective copolymers, needed for the preparation of targeted NPs. The resulting EGCG encapsulating NPs led to an enhanced anti-proliferative activity in PCa cell lines compared to the free EGCG. The behavior of EGCG encapsulated in NPs in modulating apoptosis and cell-cycle, was also determined. Then, in vivo experiments, in mouse xenograft model of prostatic tumor, using EGCG-loaded NPs, with a model of targeted nanosystems, were conducted. The obtained data supported our hypothesis of target-specific enhanced bioavailability and limited unwanted toxicity, thus leading to a significant potential for probable clinical outcome.
