The variola virus, the causative agent of smallpox, has claimed more human lives than all other infectious diseases combined. Consequently, any epidemiological event involving related viruses (orthopoxviruses) triggers significant concern, as was the case with the pandemic rise of monkeypox virus (MPXV), the causative agent of monkeypox in 2022−2023. Smallpox vaccines, based on vaccinia virus antigens, are expected to provide cross‐protection against MPXV. Studying the general seroprevalence of orthopoxviruses in populations worldwide is crucial for estimating susceptibility, risks, and the need for vaccination campaigns. In this study, we aimed to evaluate the seroprevalence of antibodies capable of recognizing MPXV in a Brazilian population and assess the potential for cross‐protection from previous smallpox vaccinations. We analyzed demographic data, vaccination records, and serum levels of anti‐MPXV IgG measured by enzyme‐linked immunosorbent assay (ELISA) in 319 volunteers from Barreiras, Bahia, Brazil. Our main findings show that the numbers of volunteers with serum samples recognizing MPXV in ELISA, as well as their serum levels of anti‐MPXV IgG, did not differ significantly according to gender or address type. However, both male and female elderly volunteers exhibited significantly higher concentrations of anti‐MPXV IgG than adults, youth, and children. These results suggest that the elderly may benefit from a cross‐reactive immune response due to previous smallpox vaccinations, whereas adults, youth, and children appear susceptible to MPXV. Further investigations involving other populations are necessary to assess the necessity of vaccinating potentially vulnerable groups. [ABSTRACT FROM AUTHOR]

Background and objective: To analyze the association between different domains and intensities of physical activity (PA) in adults. Methods: 269 participants were randomly selected in a community-dwelling representative sampling process. The habitual PA practice was assessed in different domains (occupational, sports practice, and leisure time/commuting) using Baecke's questionnaire and in different intensities (light, moderate, vigorous, and very vigorous) by accelerometry. Linear regression models analyzed the association between variables. Results: Moderate and moderate-to-vigorous PA was positively associated with the three PA domain scores. Vigorous PA was associated with sports practice and leisure time/commuting PA. Only sports practice was associated with very vigorous-intensity PA (β = 1.32; 95% CI = 0.29; 2.35). Different associations were observed in stratified analyses according to sex and age groups. Men and younger participants showed an association of domain scores with higher intensity PA than their counterparts. Conclusion: The findings suggest that higher intensities of PA were observed primarily in leisure time/commuting PA and sports practice, and this association varied according to sex and age. The study aimed to optimize qPCR reactions using oligonucleotides from the first Brazilian molecular diagnostic kit for leprosy on a portable platform (Q3-Plus). In addition, we sought to develop a simplified protocol for DNA extraction that met point-of-care criteria. During optimization on the Q3-Plus, optical parameters, thresholds, and cutoffs for the 16S rRNA and RLEP targets of M. leprae were established using synthetic DNA, purified DNA from M. leprae, and pre-characterized clinical samples. For the simplified extraction protocol, different lysis solutions were evaluated using chaotropic agents, and purification was carried out by transferring the lysed material to FTA cards. The complete protocol (simplified extraction + qPCR on the portable platform) was then evaluated with pre-characterized clinical skin biopsy samples and compared with standard equipment (QuantStudio-5). LOD95% for the optimized reactions was 113.31 genome-equivalents/μL for 16S rRNA and 17.70 genome-equivalents/μL for RLEP. Among the lysis solutions, the best-performing was composed of urea (2 M), which provided good dissolution of the skin fragment and a lower Ct value, indicating higher concentrations of DNA. The complete technological solution showed a sensitivity of 52% in reactions. Our results highlight the need for additional optimization to deal with paucibacillary samples, but also demonstrate the feasibility of the portable platform for the qPCR detection of M. leprae DNA in low infrastructure settings. Author summary: The diagnosis of tropical or neglected infectious diseases, such as leprosy, is highly dependent on techniques that are not very sensitive or very laborious, such as optical microscopy or culture. More sensitive and specific molecular tests are only available in urban centers, where there is a specialized workforce and adequate infrastructure. However, a large portion of the population does not have access to such medical services, either due to financial or distance issues. Thus, performing diagnostic tests at the point of care (POC) provides agility in diagnosis, avoiding the need for patients to travel to medical centers or transport biological samples to clinical analysis centers, ensuring that this population has access to diagnostic tests, even in remote areas or with poor infrastructure. In this work, we optimize and validate, on a laboratory scale, simplified versions of all the procedures necessary to perform molecular tests, from collection and DNA extraction to the detection of genomic targets using portable qPCR equipment. The results presented here are the basis of a novel tool with potential to increase a leprosy screening test in remote areas, facilitating the active search of positive cases and monitoring by health authorities responsible for underserved populations. Introduction: Calcium silicate-based sealers are an alternative to be used into root canal, mainly to their biological properties. However, some biological parameters need to be determined in an in vivo animal research model. So, the aim of the present study was to evaluate in vivo the tissue biocompatibility of a calcium silicate-based sealer (EndoSequence BC Sealer) and an epoxy resin-based sealer (AH-Plus). Materials and Methods: Polyethylene tubes were filled with freshly mixed sealers and implanted in connective subcutaneous tissue of 25 rats (5/euthanasia day) (Rattus norvegicus albinus). Empty tubes were used as controls and no tubes as sham. Histopathological (hematoxylin eosin) and histochemical (Picrosirius red) examinations were conducted at 3, 7, 15, 30 and 60 days (five rats/day) after the implantation procedure (n=5/group). The type/intensity of inflammation and collagenesis was analyzed statistically with Friedman or Kruskal-Wallis/Dunn tests (P<0.05). Results: The profile of inflammation induced by AH-Plus (Median=2, Range=2-3) was significantly greater than that by Endosequence BC Sealer (Median=1, Range=1-1) during the 15-day experimentation period (P=0.018). After 30 days, both materials produced similar tissue reaction (P>0.05). AH-Plus and Endosequence BC Sealer (Median=2, Range=1-2) induced a high level of fibrosis after 60-day than control (Median=1, Range=1-1) and sham (Median=0, Range=0-0) groups (P<0.001) of fibrosis based in type I collagen increase (P=0.025 and P=0.021, respectively). Tissue necrosis was not observed and the bioceramic sealer showed significant signs of endocytosed (Median=1, Range=1-1) material after 7 days than other groups (Median=0, Range=0-0) (P<0.05). The calcium silicate-based sealer induced tissue repair faster than the epoxy resin-based sealer tested. However, both materials showed adequate biocompatibility and tolerance by subcutaneous tissues, with few differences in inflammatory profiles, formation of granulation tissue, and collagenesis. Conclusions: It may be concluded that calcium silicate-based sealer (EndoSequence BC Sealer) and an epoxy resin-based sealer (AH-Plus) presented suitable biocompatibility. [ABSTRACT FROM AUTHOR]

In addition to pain, specific low back pain is frequently accompanied by restricted range of motion (ROM) during gait. To compare the behavior of kinematic and spatiotemporal gait parameters, pain, functional status, and self-efficacy, in patients with a diagnosis of herniated disk or lumbar stenosis undergoing surgery, in the pre- and postoperative periods of 1 and 6 months (PO6). Seven participants and 11 control subjects were assessed. A kinematics system comprising 10 optoelectronic cameras was used to assess gait. The Roland-Morris questionnaire, pain intensity, and self-efficacy, over three periods, were used. The ROM of the pelvis, hip, and knee of the hernia group presented an increase after surgery and the stenosis group presented a reduction of values in the hip. During the stance phase, the pelvis and hip ROM of both groups remained smaller than the control group. There was improvement in pain in individuals with hernia and stenosis (effect size = 0.6 and 0.8, respectively) in the three analyzed moments; for functional status there was improvement in the first postoperative period (ES = 0.4) compared to the preoperative in those individuals with hernia; and those with stenosis had improvement at PO6 when compared to the time before the surgery (ES = 0.2). Surgical intervention modifies the spatiotemporal parameters, the ROM of the pelvis, hip, and knee in the total gait cycle, primarily in the sagittal plane, and causes alterations, particularly in the hip joint, in these individuals during the support phase. Background: Lifestyle and habits, cardiovascular risk factors (CRF), bone and mental health, dietary habits, physical activity, among others are developed in childhood and adolescence. Family environment has shown to play an important role in these outcomes. However, whether the parent-child relationship lifestyle habits and health parameters can be influenced by physical activity patterns still unclear. The objective of this study will be to monitor and investigate the associations between lifestyle habits between parents and their children longitudinally, as well as verify whether in more active parents, the possible associations with lifestyle habits are different from those of parents considered less active. Methods: The sample will consist of parents (father, mother, or both) and their children /adolescents. The participants will be recruited through public call by flyers spread across all the regions of the city and also through social media. The health parameters will include cardiovascular (cardiac autonomic modulation, blood pressure and resting heart rate), bone mineral density, anthropometric indices, handgrip strength, mental health (quality of life, anxiety and depression symptoms and stress), self-reported morbidities and musculoskeletal pain. Lifestyle habits will include physical activity levels, sedentary behavior, sleep parameters, eating patterns, smoking and alcohol consumption. Sociodemographic variables of age, sex, ethnicity and socioeconomic status will be considered as covariates. The follow-up visits of data collection will be scheduled after a period of 12 months from the baseline assessment during every twelve months. Discussion: The family environment has great potential to determine lifestyle habits in children and adolescents. In the fruit processing industry, a large volume of residue is generated, which is usually discarded. However, fruit residues may have appeal as functional ingredients due to the presence of bioactive compound. In this study, the drying of acerola residue in a medium‐sized conventional spouted bed was investigated. The residue was characterized; vitamin C and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) were determined to evaluate the effect of drying on bioactive compounds. To overcome the difficulty of material fluidization, a mixture composed of in natura acerola residue and acerola residue previously dried in an oven as an inert material was used. The effect of air temperature (40 and 60°C) and velocity (28 and 36 m/s) and amount of in natura residue in the mixture (20% and 30%) was evaluated during drying. As expected, higher temperatures and air velocities and lower percentages of in natura residue favored the reduction of moisture. For a mixture containing 20% in natura residue, air velocity of 36 m/s, and temperature of 60°C, it was possible to achieve a final moisture content suitable for storage (14.23% w.b.) and the highest percentage of DPPH free radical capture activity (85.31%). Moreover, a mixture containing 30% in natura residue, velocity of 28 m/s, and temperature of 40°C conserved more vitamin C (48.30 mg of ascorbic acid/100 g). Finally, spouted bed can be considered a good technique for drying acerola residue as it was able to reduce a high amount of moisture content preserving bioactive compounds during short drying periods at low temperatures. Practical Applications: The acerola processing generates a residue that consists basically of the seeds, bagasse, and peels, which is rich in ascorbic acid, phenolic compounds, flavonoids, anthocyanins, and organic acids. This residue can be used to generate new food products, which reduces the environmental problem of its disposal and the loss of raw material in fruit processing. To extend the residue shelf life, it must be subjected to a drying process. In this paper, the drying of acerola residue was investigated in a medium‐sized spouted bed. The spouted bed proved effectiveness in drying the acerola residue and in conserving bioactive compounds when operating for short periods and with low temperature. Thus, it can be considered as a promising technique for drying food products. Also, the data provided in this study can contribute to the solution of the scale‐up problem in spouted beds. In Latin America, rabies virus has persisted in a cycle between Desmodus rotundus vampire bats and cattle, potentially enhanced by deforestation. We modeled bovine rabies virus outbreaks in Costa Rica relative to land-use indicators and found spatial-temporal relationships among rabies virus outbreaks with deforestation as a predictor. Background: Practicing sports during childhood and adolescence provides benefits to cardiac autonomic modulation (CAM) at these stages of life. However, it is not known whether these benefits to CAM persist into adulthood. Therefore, the objective of this study was to analyze the association of early sports practice (sports practice in childhood and/or adolescence) with CAM in adult life, regardless of habitual moderate-to-vigorous PA. Methods: The sample of the present study consisted of 242 adults (141 women and 101 men; age: 41.99 ± 16.24). The assessment of CAM was performed using heart rate variability indices. Sports practice in childhood and adolescence was assessed using a questionnaire. The intensity of physical activity was assessed using accelerometry. To analyze the association between previous sports practice (childhood and/or adolescence) and CAM, the Generalized Linear Model was adopted, considering CAM indices as continuous variables and early sports practice as a 3-fold factor (no sports practice; sports practice in childhood or adolescence; and sports practice in both childhood and adolescence) adjusted by sex, age, socioeconomic condition, and moderate to vigorous PA. Results: Sports practice in childhood was associated with the average standard deviation of all normal RR intervals expressed in milliseconds (SDNN): β = 5.89; 95%CI: 0.25;11.52, and the standard deviation of the long-term intervals between consecutive heartbeats (SD2): β = 7.63; 95%CI:1.04; 14.23 indices. Sports practice in adolescence was associated in adulthood with the SD2 index: β = 7.37; 95%CI: 0.71;14.04. Sports practice in at least one of the periods (childhood or adolescence) was significantly associated with the square root of the mean square of the differences between adjacent normal RR intervals for a period of time expressed in milliseconds (RMSSD) (β = 8.86; 95%CI = 0.71;17.01), and the standard deviation of the instantaneous beat to beat variability (SD1) (β = 6.21; 95%CI = 0.45;11.97). Sports practice at both stages of life was significantly associated with better SDNN (β = 7.70; 95%CI = 1.16;14.23) and SD2 (β = 10.18; 95%CI = 2.51;17.85). Conclusion: Early sports practice was associated with better CAM in adulthood, independently of the current physical activity level. Based on these findings, sports practice is encouraged from childhood and adolescence, for benefits to CAM in adult life. Key Points: Few studies have verified whether the practice of sports in childhood and adolescence could be associated with better CAM in adult life. Sports practice in childhood and/or in adolescence was associated with better CAM in adult life, both when analyzed separately and when combined. Transitional foods for infants and children of early childhood, also popularly known as baby-food, are used in order to complement the diet, gradually introducing new sources of nutrients to children in the weaning phase. The objective is to evaluate the chemical and biochemical quality of transitional foods for infants and young children such as vegetable and meat baby-food (VMBF) and vegetable and chicken baby-food (VCBF). 20 samples of VMBF and VCBF were analyzed. The levels of nitrate, sodium, pH, macronutrients (protein, lipid and carbohydrate), total caloric value (TCV), humidity, fixed mineral residue (FMR) and solid matter (SM) were determined in duplicate. The results showed that the two types of baby-food (VMBF and VCBF) did not show statistical differences (p > 0.05). Regarding the current legislation, it was found that 100% of the samples of VMBF and VCBF presented within the reference values for the pH and SM tests. The VMBF samples showed percentages of 25%, 100%, 25% and 25% for the nitrate, sodium, protein and TCV assays that were outside the permitted values, respectively. The samples of VCBF, however, presented percentage indexes of 40%, 80% and 40% for the tests of nitrate, sodium and protein outside the established reference standards, respectively. It is concluded that the high levels of nitrate and sodium are worrisome, being necessary the active inspection by Organs regulating agencies in relation to these foods due to its importance in child development. Over the past two decades, research on bat-associated microbes such as viruses, bacteria and fungi has dramatically increased. Here, we synthesize themes from a conference symposium focused on advances in the research of bats and their microbes, including physiological, immunological, ecological and epidemiological research that has improved our understanding of bat infection dynamics at multiple biological scales. We first present metrics for measuring individual bat responses to infection and challenges associated with using these metrics. We next discuss infection dynamics within bat populations of the same species, before introducing complexities that arise in multi-species communities of bats, humans and/or livestock. Finally, we outline critical gaps and opportunities for future interdisciplinary work on topics involving bats and their microbes. [ABSTRACT FROM AUTHOR]

The use of silvopastoral systems with tree legumes is a viable alternative to recover and develop pastures, as they add N to the system influencing pasture growth. This study hypothesized that the herbage and litter of signalgrass (Urochloa decumbens Stapf) is affected by legume trees in the pasture. Treatments were composed of (1) signalgrass + Mimosa caesalpiniifolia Benth.; (2) signalgrass + Gliricidia sepium Jacq.; and (3) signalgrass monoculture. The 3-year experiment followed a randomized complete block design with three replications. Tree legumes were planted in double rows (15 × 1 × 0.5 m), in 1 ha paddocks. Litter samples were taken in five distance points (0, 1.8, 3.7, 5.6 and 7.5 m) perpendicular to tree legume rows. Signalgrass was taller at longer distances from the trees (P < 0.05). Signalgrass height differed between treatments, with taller signalgrass found in pastures mixed with G. sepium (15.6 cm) compared to M. caesalpiniifolia (9 cm) (P < 0.05). Herbage N content decreased with increasing distance from tree rows (P < 0.05). Litter N content followed a similar pattern, ranging from 23 g/kg under the trees to 12 g/kg at 7.5 m away from tree rows. Signalgrass did not grow under the tree crown (0–1.8 m), especially when intercropped with M. caesalpiniifolia. The findings of this study suggest that the type of legume trees used in the silvopastoral system has the potential to modify the pattern of grass growth and content of N in pasture litter. We describe a feline sporotrichosis cluster and zoonotic transmission between one of the affected cats and a technician at a veterinary clinic in Kansas, USA. Increased awareness of sporotrichosis and the potential for zoonotic transmission could help veterinary professionals manage feline cases and take precautions to prevent human acquisition. Background: To examine the associations between motor competence (MC), moderate to vigorous physical activity (MVPA), and body mass index (BMI) changes over 1 year in preschoolers. Methods: Fifty-four preschoolers (24 girls; 42.4%) aged 4–5 years old from the metropolitan area of Porto, Portugal comprised the sample. Height, body mass, and BMI were calculated. MC was assessed according to the Movement Assessment Battery for Children-2. MVPA was measured by accelerometry. For each participant, changes in MVPA, MC, and BMI over a year were computed. Different levels of MC were calculated; and then data were analyzed. Results: The MVPA increased over time in all levels of MC in the follow-up. The BMI decreased over time for all levels in follow-up (P =.001). The preschoolers classified as at a high level of MC were more likely to spend more time in MVPA (in minutes) than their counterparts with low MC in follow-up. Multiple linear regression analyses, adjusted by sex and age, were fit to predict Δ% MC by Δ% MVPA and Δ% BMI. The Δ% MC was positively associated with Δ% MVPA. Conclusion: Increases in MC were positively associated with MVPA. Higher performance in MC increases due to time spent in MVPA. Water deficits have been considered the most restrictive environmental constraint on agricultural production worldwide. The current study aimed to investigate the role of silicon nutrition (Si) in activating defence mechanisms against drought damage in cowpea cultivars. The experiments were carried out in a randomized block design in a 2 × 2 × 4 factorial scheme, corresponding to two cowpea cultivars (BRS Novaera and BRS Tumucumaque), two water regimes (control well‐irrigated and water deficit) and four Si levels (0, 1.0, 2.0 and 4.0 mM). Plant growth and physiological and biochemical indicators were evaluated 28 days after drought imposition. Drought significantly reduced the photosynthetic pigments (Chl b and Chl total), gas exchanges (net photosynthesis, transpiration and stomatal conductance) and, consequently, all growth parameters of cowpea plants compared with well‐irrigated plants. However, Si at 2.0 mM activated critical responses in the BRS Novaera cultivar under drought, almost recovering plant performance and increasing drought tolerance. The beneficial Si‐induced effects were closely related to increased accumulation of Si, carbohydrates and free amino acids that likely promoted osmoregulation and were associated with an improved antioxidant system composed of proline and the activity of SOD, CAT and APX. These metabolic alterations were sufficient to enable enhanced net photosynthesis and plant growth. In conclusion, Si counteracts the deleterious effects of water deficit by efficiently inducing antioxidant defence and photosynthetic performance in Novaera plants. This paper presents a study conducted in São Paulo, Brazil, where the planetary boundary layer height (PBLH) was determined using ceilometer data and the wavelet covariance transform method. The retrieved PBLH values were subsequently compared with the concentrations of CO2 and CH4 measured at three distinct experimental sites in the city. The period of study was July 2021. This study also included a comparison between ceilometer data and lidar data, which demonstrated the favorable applicability of the ceilometer data for PBLH estimation. An examination of the correlation between changes in average CO2 concentrations and PBLH values revealed stronger correlations for the IAG and UNICID stations, with correlation coefficients (ρ) of approximately −0.86 and −0.85, respectively, in contrast to the Pico do Jaraguá station, which exhibited a lower correlation coefficient of −0.42. When assessing changes in CH4 concentrations against variations in PBL height, the retrieved correlation coefficients were approximately −0.78 for IAG, −0.66 for UNICID, and −0.38 for Pico do Jaraguá. The results indicated that CO2/CH4 concentrations are negatively correlated with PBL heights, with CO2 concentrations showing more significant correlation than CH4. Additionally, among the three measurement stations, IAG measurements displayed the most substantial correlation. The results from this study contribute to the understanding of the relationship between PBLH and greenhouse gas concentrations, emphasizing the potential of remote sensing systems like ceilometers in monitoring and studying atmospheric processes. Background: Prolonged hemodialysis (HD) is performed from 6 to 12 h and can last up to 24 h. To prevent system clotting some studies suggest that Regional Citrate Anticoagulation (RCA) use reduces bleeding rates relative to systemic heparin. However, there may be difficulties in the patient's clinical management and completing the prescribed HD with Genius system using RCA. Objective: To analyze safety Quality Indicators (IQs) and follow up on prolonged HD with 4% sodium citrate solution in a Genius® hybrid system. Methods: This is a retrospective cohort conducted in an intensive care unit. Results: 53 random sessions of prolonged HD with 4% sodium citrate solution of critically ill patients with AKI assessed. Evaluated safety indicators were dysnatremia and metabolic alkalosis, observed in 15% and 9.4% of the sessions, respectively. Indicators of effectiveness were system clotting which occurred in 17.3%, and the minimum completion of the prescribed HD time, which was 75.5%. Conclusion: The assessment of the indicators showed that the use of RCA with a 4% sodium citrate solution in prolonged HD with the Genius system in critically ill patients with AKI can be performed in a simple, safe, and effective way. Key learning points: What is already known about the subject?: Studies on RCA or systemic heparin usage do not demonstrate differences in the mortality of critically ill patients with AKI undergoing acute RRT. However, they suggest lower bleeding rates and greater longevity of the dialysis filter. Our working hypothesis was that magnesium (Mg) supplementation modulates plant performance under low water availability and improves drought tolerance in soybean genotypes. Plants of Bônus 8579, M8808 and TMG1180 genotypes were grown under field conditions and subjected to three water stress treatments (control, moderate and severe stress) and three Mg levels [0.9 (low), 1.3 (adequate) and 1.7 cmolc dm-³ (supplementation)]. After 28 days of drought imposition, the growth parameters, osmotic potential, relative water content, leaf succulence, Mg content and photosynthetic pigments were assessed. In general, drought drastically decreased the growth in all genotypes, and the reductions were intensified from moderate to severe stress. Under adequate Mg supply, TMG1180 was the most drought-tolerant genotype among the soybean plants, but Mg supplementation did not improve its tolerance. Conversely, although the M8808 genotype displayed inexpressive responses to drought under adequate Mg, the Mg-supplemented plants were found to have surprisingly better growth performance under stress compared to Bônus 8579 and TMG1180, irrespective of drought regime. The improved growth of high Mg-treated M8808-stressed plants correlated with low osmotic potential and increased relative water content, as well as shoot Mg accumulation, resulting in increased photosynthetic pigments and culminating in the highest drought tolerance. The results clearly indicate that Mg supplementation is a potential tool for alleviating water stress in M8808 soybean plants. Our findings suggest that the enhanced Mg-induced plant acclimation resulted from increased water content in plant tissues and strategic regulation of Mg content and photosynthetic pigments. Human activities have increased the intensity and frequency of natural stressors and created novel stressors, altering host–pathogen interactions and changing the risk of emerging infectious diseases. Despite the ubiquity of such anthropogenic impacts, predicting the directionality of outcomes has proven challenging. Here, we conduct a review and meta‐analysis to determine the primary mechanisms through which stressors affect host–pathogen interactions and to evaluate the impacts stress has on host fitness (survival and fecundity) and pathogen infectivity (prevalence and intensity). We assessed 891 effect sizes from 71 host species (representing seven taxonomic groups) and 78 parasite taxa from 98 studies. We found that infected and uninfected hosts had similar sensitivity to stressors and that responses varied according to stressor type. Specifically, limited resources compromised host fecundity and decreased pathogen intensity, while abiotic environmental stressors (e.g., temperature and salinity) decreased host survivorship and increased pathogen intensity, and pollution increased mortality but decreased pathogen prevalence. We then used our meta‐analysis results to develop susceptible–infected theoretical models to illustrate scenarios where infection rates are expected to increase or decrease in response to resource limitations or environmental stress gradients. The condensation patterns (CPs) of prophase chromosomes represent poorly explored and little understood karyotype features. Three distinct chromosome domains are observed in conventionally stained prophases of most angiosperms with small chromosomes: heterochromatin and early condensing euchromatin (ECEu), which are observed as early condensing regions (ECRs), and late condensing euchromatin or late condensing regions (LCRs). All three prophase domains have been often reported in the Annonaceae species. To gain a better understanding of these regions, we investigated the CPs, heterochromatic bands, and rDNA sites in seven Annona species with 2n  = 14, 28, and 54 and Xylopia frutescens with 2 n  = 16. Besides, histone H4K5 acetylation, telomeric sites, and DNA methylation were analysed in some of these species. LCRs corresponded to a small hyperacetylated, and hypomethylated fraction of the metaphase chromosomes. During interphase, the chromocentres displayed variable proportions of heterochromatin and ECEu. The LCRs and ECRs were conserved even between disploid and polyploid species, whereas rDNA sites and heterochromatic bands varied in number and location. Our data suggest that chromatin compartmentalization in ECR and LCR regions may represent the simplest functional organization of the small chromosomes of Annonaceae, while the remaining characters are less relevant. Key message: Campomanesia spp produce seeds with high viability and vigor which are tolerant to desiccation but not to cold temperature. The genus Campomanesia has ecological and economic importance in the Atlantic Forest, but 19 of its species are on the red list of threatened species of the International Union for Conservation of Nature (IUCN). There is a lack of studies on seed biology and considerable urgency in developing strategies for their ex-situ conservation and subsequent use in restoration. In this study, we analyzed the viability and vigor in seeds of three Campomanesia species under different temperatures to determine the ideal conditions for storage. Additionally, we analyzed the factors that influence their response in storage. Differences in germination, tetrazolium and germination speed index were compared for three Campomanesia species with different distributions in the Brazilian Atlantic Forest (C. xanthocarpa, C. reitziana and C. littoralis). Seeds of three species were stored in two water contents, three temperatures, for six period and liquid nitrogen. The three species presented seeds sensitive to freezing (−20 °C and liquid nitrogen). C. xanthocarpa seeds showed a short life during storage at 25 °C and 8 °C. C. reitziana and C. littoralis seeds with initial water content showed high germination up to 120 days of storage at 8 °C and these values were higher than seeds with water content of 0.16 g H2O. g dw− 1 stored under the same conditions. Finally, our results of the characterization of the tolerance to drying and the behavior during storage at different temperatures of C. xanthocarpa, C. littoralis and C. reitziana are the base line for the generation of successful conservation strategies, both in situ and ex situ. Extra virgin olive oil (EVOO) and avocado oil (AVO) are recognized for their unique sensory characteristics and bioactive compounds. Declared blends with other vegetable oils are legal, but undeclared mixing is a common type of fraud that can affect product quality and commercialization. In this sense, this study explored strategies to mitigate the influence of lighting in order to make digital image colorimetry (DIC) using a smartphone more robust and reliable for predicting the soybean oil content in EVOO and AVO blends. Calibration models were obtained by multiple linear regression using the images' RGB values. Corrections based on illuminance and white reference were evaluated to mitigate the lightness effect and improve the method's robustness and generalization capability. Lastly, the prediction of the built model from data obtained using a distinct smartphone was assessed. The results showed models with good predictive capacities, R2 > 0.9. Generally, models solely based on GB values showed better predictive performances. The illuminance corrections and blank subtraction improved the predictions of EVOO and AVO samples, respectively, for image acquisition from distinct smartphones and lighting conditions as evaluated by external validation. It was concluded that adequate data preprocessing enables DIC using a smartphone to be a reliable method for analyzing oil blends, minimizing the effects of variability in lighting and imaging conditions and making it a potential technique for oil quality assurance. Identifying cultivars of leguminous crops exhibiting drought resistance has become crucial in addressing water scarcity issues. This investigative study aimed to select soybean and cowpea cultivars with enhanced potential to grow under water restriction during the vegetative stage. Two parallel trials were conducted using seven soybean (AS3810IPRO, M8644IPRO, TMG1180RR, NS 8338IPRO, BMX81I81IPRO, M8808IPRO, and BÔNUS8579IPRO) and cowpea cultivars (Aracê, Novaera, Pajeú, Pitiúba, Tumucumaque, TVU, and Xique-xique) under four water levels (75, 60, 45, and 30% field capacity—FC) over 21 days. Growth, water content, membrane damage, photosynthetic pigments, organic compounds, and proline levels were analyzed. Drought stress significantly impacted the growth of both crops, particularly at 45 and 30% FC for soybean and 60 and 45% FC for cowpea plants. The BÔNUS8579IPRO and TMG1180RR soybean cultivars demonstrated the highest performance under drought, a response attributed to increased amino acids and proline contents, which likely help to mitigate membrane damage. For cowpea, the superior performance of the drought-stressed Xique-xique cultivar was associated with the maintenance of water content and elevated photosynthetic pigments, which contributed to the preservation of the photosynthetic efficiency and carbohydrate levels. The application range of polyamide 6 in lightweight part applications can be significantly increased by using appropriate processing parameters and by incorporating additives. Therefore, specimens were manufactured at varying mold temperatures and injection velocities to study the effect of injection molding parameters on the properties of polyamide 6 and its nanocomposite with nano‐calcium carbonate. Mechanical properties, degree of crystallinity, density, heat deflection temperature, and melt flow index were measured to assess the effect of injection molding conditions on material properties. Mold temperature was the process factor that most affected polyamide 6 and polyamide 6/nano‐calcium carbonate properties. Increasing this parameter increased density, impact strength, flexural strength, flexural modulus, and heat deflection temperature. The nanocomposite's degree of crystallinity, tensile modulus, and melt flow index also increased. High injection velocity reduced only the impact strength of the nanocomposite. The findings indicate that by modifying injection parameters, it is possible to improve mechanical properties and processing efficiency of polyamide 6 and its nanocomposites, bringing them closer to their maximum potential. Incorporating the nanofiller increased almost all properties except for impact strength and elongation at break. Nano‐calcium carbonate provided considerable advantages to polyamide 6 mechanical performance and processability while increasing sample weight by only 1.5%. [ABSTRACT FROM AUTHOR]

Seed physiology is related to functional and metabolic traits of the seed-seedling transition. In this sense, modeling the kinetics, uniformity and capacity of a seed sample plays a central role in designing strategies for trade, food, and environmental security. Thus, POMONA is presented as an easy-to-use multiplatform software designed to bring several logistic and linearized models into a single package, allowing for convenient and fast assessment of seed germination and or longevity, even if the data has a non-Normal distribution. POMONA is implemented in JavaScript using the Quasar framework and can run in the Microsoft Windows operating system, GNU/Linux, and Android-powered mobile hardware or on a web server as a service. The capabilities of POMONA are showcased through a series of examples with diaspores of corn and soybean, evidencing its robustness, accuracy, and performance. Featured Application: The use of microwaves in drying has been the subject of studies in the post-harvest sector. Some authors have identified damage to the seeds because of microwaves, while others have found promising results for the technique. This study verified that the use of microwaves can be safe if the appropriate temperature and power conditions are applied. In this work, readers will also be able to verify that the use of electromagnetic energy can also be more economical and efficient, as it greatly reduces the drying time, in addition to being possible to increase germination during the process, which is highly desirable. In addition to the temperature x power binomial, water content is another variable that can affect the process. This should be a stimulus for further research and studies in the area that address a broader spectrum of varieties and water content to that the technique can be safer to the point that its use reaches the public. The use of electromagnetic energy for drying seeds has been shown to be a promising alternative. However, studies show that the technique still lacks careful evaluation due to the sensitivity of soybean seeds to damage caused using microwaves. Studies have shown that the use of microwaves during drying can be safe in seeds of soybeans, corn, and beans, among others. These studies recognize that drying using microwaves has a great potential for savings in the drying process, as it considerably reduces the drying time. This work aimed to evaluate the immediate damage caused in soybean seeds subjected to drying at temperatures of 40, 50, and 60 °C, with power of 0, 0.5, and 1 W/g at a frequency of 2.45 GHz. The results of the vigor and germination tests showed that the action of microwaves on seeds was not only safe and more efficient but also brought about increases in seed germination and vigor. The aim of this study was to evaluate the influence of IR (λ850 ± 10 nm) and violet (λ405 ± 10 nm) LED phototherapy on total mast cells counts and its ability to influence mast cell degranulation. For this, 27 Wistar rats were used and were randomly distributed into three groups: control, IR LED, and violet LED. When indicated, irradiation done and they were sacrificed, had their tongue removed immediately, 20-min, 45-min, and 2-h after irradiation. Samples were processed to wax, cut, and stained with Toluidine Blue. Intact and degranulated mast cells were counted under light microscopy, and statistical analysis was carried out. In the superficial connective tissue and muscular tissues, violet LED light caused a significant increase in both total number and degranulated mast cells when compared to the control group immediately after irradiation. The degranulation indexes were higher in the groups irradiated with Violet light, both in superficial connective tissue and muscular tissues in relation to the timing. Irradiation with IR LED caused immediate increase in the total number and degranulated of mast cells when compared to the control group only in the superficial connective tissue. In all times observed, the highest total amount of mast cells was seen immediately after irradiation, except in the muscular tissue, which presented the highest amount after 20-min. It was concluded that IR and violet LED light were able to increase the number of mast cells and inducing degranulation in oral mucosa. However, considering that violet LED light can be harmful in periodontal disease, it seems that the use of IR LED light could be the best option in Dentistry. [ABSTRACT FROM AUTHOR]

The official sensory analysis of extra virgin olive oil (EVOO) must be performed by trained evaluators, but the objective of this work was to verify whether consumers perceive sensory attributes of EVOO, in a similar way to the TP. Physical-chemical and sensory analysis was performed and the means and medians were tested respectively by Tukey (p>0.01) and Kruskal-Wallis (p>0.05). Homogeneity and reproducibility were evaluated in sensory analyses. This study analyzed 4 EVOOs from Serra-da-Mantiqueira in Brazil: 2 organic and 2 conventional. Twelve experts according to COI protocols, along with 12 EVOO consumers, aged between 20 and 57 years old, volunteered. Physicochemical and sensory results met the classification requirements for EVOO. In session 1, the "consumer panel" (CP) perceived the "GreenFruitiness" attribute in a non-differentiable way to the trained ones. In session 2, the participants have been trained and doubts were resolved. In session 3, the homogeneity of the CP responses had a z-score greater than the 1 from the "trained panel" (TP), and after the training, there was a flattening in the dispersion of responses when comparing the intensities perceived by the two groups. This study evaluated the bromatological and histological features of native African grasses under grazing in Brazilian semi-arid rangelands. An experimental grid design was used in a randomised sampling method for four replicate samples of three African grasses. The grasses evaluated were: Cenchrus ciliaris L., Digitaria pentzii Stent. and Megathyrsus maximus (Jacq.). The bromatological and histological traits analysed included dry matter (DM), crude protein (CP), neutral detergent fibre corrected to ashes and protein (NDFap), lignin, mesophyll, vascular bundle, phloem, xylem, vascular sheath, sclerenchyma, bulliform cells, and the adaxial and abaxial epidermis. The type of grass and grazing cycle correlated with DM, NDF and lignin (p < 0.05), whereas CP was not affected by these factors (p > 0.05). Dry matter ranged between 128 and 304 g kg−1; CP ranged between 90 and 167 g kg−1; NDFap between 542 and 707 g kg−1, and lignin between 10 and 40 g kg−1. The proportion of xylem (8.4%), vascular sheath (30.5%) and total vascular bundles (38.1%) were greater in M. maximus (p < 0.05). The percentage of sclerenchyma did not differ between species (p > 0.05). Digitaria pentzii and C. ciliaris had a greater proportion of mesophyll than M. maximus (p < 0.05). Of the three grasses analysed. Digitaria pentzii exhibited the highest forage quality, because it had the lowest concentration of fibre and lignin, with lower proportions of lignified tissues.
