The ultraviolet (UV) radiation triggers a pigmentation response in human skin, wherein, melanocytes rapidly activate divergent maturation and proliferation programs. Using single-cell sequencing, we demonstrate that these 2 programs are segregated in distinct subpopulations in melanocytes of human and zebrafish skin. The coexistence of these 2 cell states in cultured melanocytes suggests possible cell autonomy. Luria–Delbrück fluctuation test reveals that the initial establishment of these states is stochastic. Tracking of pigmenting cells ascertains that the stochastically acquired state is faithfully propagated in the progeny. A systemic approach combining single-cell multi-omics (RNA+ATAC) coupled to enhancer mapping with H3K27 acetylation successfully identified state-specific transcriptional networks. This comprehensive analysis led to the construction of a gene regulatory network (GRN) that under the influence of noise, establishes a bistable system of pigmentation and proliferation at the population level. This GRN recapitulates melanocyte behaviour in response to external cues that reinforce either of the states. Our work highlights that inherent stochasticity within melanocytes establishes dedicated states, and the mature state is sustained by selective enhancers mark through histone acetylation. While the initial cue triggers a proliferation response, the continued signal activates and maintains the pigmenting subpopulation via epigenetic imprinting. Thereby our study provides the basis of coexistence of distinct populations which ensures effective pigmentation response while preserving the self-renewal capacity. An eco-friendly approach of creating nanoparticles has emerged in order to get around the drawbacks of conventional techniques. Today, a wide range of nanoparticles have been created by employing various microbes, and their potential in numerous cutting-edge technological fields have been investigated. These particles have well-defined chemical compositions, sizes, and morphologies. The green production of nanoparticles mostly uses plants and microbes. Hence, the use of microbial nanotechnology in agriculture and plant science is the main emphasis of this review. In the present scenario, growing population demands more food, resulting in the need for sustainable agriculture. Numerous approaches are explored in response to dangers and obstacles to sustainable agriculture. A viable approach is to be exploiting microbial consortium, which generate diverse biostimulants with growth-promoting characteristics for plants. These bioinoculants play an indispensable role in optimizing nutrient uptake efficiency mitigating environmental stress. Plant productivity is mostly determined by the microbial associations that exist at the rhizospheric region of plants. The engineered consortium with multifunctional attributes can be effectively employed to improve crop growth efficacy. A number of approaches have been employed to identify the efficient consortia for plant growth and enhanced crop productivity. Various plant growth-promoting (PGP) microbes with host growth-supporting characteristics were investigated to see if they might work cohesively and provide a cumulative effect for improved growth and crop yield. The effective microbial consortia should be assessed using compatibility tests, pot experimentation techniques, generation time, a novel and quick plant bioassay, and sensitivity to external stimuli (temperature, pH). The mixture of two or more microbial strains found in the root microbiome stimulates plant growth and development. The present review deals with mechanism, formulation, inoculation process, commercialization, and applications of microbial consortia as plant bioinoculants for agricultural sustainability. Heavy metal pollution is one of the modern environmental problems that contaminate soil, water, and air. Heavy metal pollution not only results in significant crop yield losses, but also poses health risks. When subjected to heavy metal toxicity, which happens when the quantity of particular heavy metals in the soil or water exceeds their tolerance levels, plants can exhibit a variety of reactions. Metal accumulation in the tissues of plants causes oxidative stress. The oxidative stress in turn impacts cellular homeostasis and adversely affects growth and development. Plants exhibit different mechanisms to fight stressful conditions and lessen the negative consequences of heavy metal toxicity. In this regard, the first line of defence in response to heavy metal toxicity provided by the plant is restricting the uptake of the metals. The second line of defence includes various detoxification mechanisms. Microbes especially associated with the rhizospheric region of the crops have role in remediation of the soils from metal contaminants. The present research work is primarily carried out to select the suitable and better variants or functions of ETAS models to describe the energetic offshore earthquake swarms following 26 December 2004 (MW 9.1) and 21 March 2014 (MW 6.5) along the Andaman–Sumatra subduction zone. An attempt has also been made to inspect the variability of the selected model parameters from this robust estimation to discuss the physical mechanism differences between the SSE-related and fluid- or magma-related swarm sequences. For this purpose, different variants of ETAS models, such as single ETAS model, combined ETAS model, Boxcar swarm model, and exponential swarm model, are utilized for investigating the seismic anomalies by estimating the model parameters from the earthquake occurrence times using maximum likelihood method. The estimated AIC (Akaike information criterion) values for all these models suggest that the exponential swarm model outperforms the other models in successfully explaining the transient changes of background seismicity for the five energetic earthquake swarms triggered by the volcano-tectonic activities in the Andaman Sea. The reason is undoubtedly related to the localized nature of magmatic intrusion or fluid migration that caused the swarms and the exponential decay patterns of earthquakes after the initiation of the swarms. Although the swarms exhibit the shape of exponential function, the background seismicity shown by the parameter μ increases by one order during the swarm as compared to the pre-swarm period in the sub-catalogue. Therefore, the procedure of identifying improved variants or functions of ETAS models and estimating model parameters using earthquake occurrence times enhances our understanding of earthquake-triggering mechanisms, potential swarm activity patterns, and the intricate interplay between seismicity and volcanic processes in submarine volcanic arcs. While this approach might not facilitate real-time forecasting, it remains instrumental in characterizing seismic patterns and providing essential information for effective hazard assessment and the formulation of mitigation strategies within the coastal areas of the Andaman–Nicobar Islands region. Endophytic microbes are plant-associated microorganisms that reside in the interior tissue of plants without causing damage to the host plant. Endophytic microbes can boost the availability of nutrient for plant by using a variety of mechanisms such as fixing nitrogen, solubilizing phosphorus, potassium, and zinc, and producing siderophores, ammonia, hydrogen cyanide, and phytohormones that help plant for growth and protection against various abiotic and biotic stresses. The microbial endophytes have attained the mechanism of producing various hydrolytic enzymes such as cellulase, pectinase, xylanase, amylase, gelatinase, and bioactive compounds for plant growth promotion and protection. The efficient plant growth promoting endophytic microbes could be used as an alternative of chemical fertilizers for agro-environmental sustainability. Endophytic microbes belong to different phyla including Euryarchaeota, Ascomycota, Basidiomycota, Mucoromycota, Firmicutes, Proteobacteria, and Actinobacteria. The most pre-dominant group of bacteria belongs to Proteobacteria including α-, β-, γ-, and δ-Proteobacteria. The least diversity of the endophytic microbes have been revealed from Bacteroidetes, Deinococcus-Thermus, and Acidobacteria. Among reported genera, Achromobacter, Burkholderia, Bacillus, Enterobacter, Herbaspirillum, Pseudomonas, Pantoea, Rhizobium, and Streptomyces were dominant in most host plants. The present review deals with plant endophytic diversity, mechanisms of plant growth promotion, protection, and their role for agro-environmental sustainability. Cannabis misuse and opioid use disorder (OUD) are highly comorbid but under-treated and associated with poorer outcomes. This paper reports a double-blind, parallel-group randomized controlled trial to determine the efficacy of single-session, clinician-delivered screening and brief intervention (SBI) for reducing cannabis risk. The primary outcome was the cannabis-specific Alcohol, Smoking, and Substance Involvement Screening Test (ASSIST) scores, measured at three-month post-intervention. The secondary objectives were to determine the efficacy of SBI in reducing the frequency of cannabis use, in risk transition from moderate to low risk, and in prescription and non-prescription opioid use. One hundred forty-three participants were randomly allocated to receive either SBI (n = 72) or control (n = 71) interventions. We performed Per-protocol (PP) (n = 125) and Intention-to-treat (ITT) analysis (n = 143). We adjusted our analysis for age, sex, and baseline ASSIST score. The ITT showed that the SBI group had a significant reduction (F = 39.46, p <.001, Effect size 0.22) in the mean ASSIST at follow-up. PP analyses too revealed a similar positive effect of SBI (F = 53.1; p <.001, Effect size 0.31). At follow-up, the SBI group had a higher number of days of cannabis abstinence. The leachates emanating from the landfills are high in organic loads and thus become potential sources of contamination for both surface and groundwater. As the landfill ages, the nature of leachate changes from acidic to alkaline. The change in pH level affects the chemical oxygen demand (COD)/biochemical oxygen demand (BOD) ratio and when it is less than 0.63, chemical treatments are more effective over the biological treatment methods such as upflow anaerobic sludge blankets (UASB). The existing literature suggests coagulation–flocculation and advanced oxidation process (Fenton) as effective methods for treating wastewater but no comparison of the two are available. Thus, the present study attempts to identify the most efficient coagulants out of ferric chloride (FeCl3), ferrous sulphate (FeSO4) and alum [Al2(SO4)3]. Ferric chloride leading to 99% colour removal, 98% COD removal, 99% decrease in total organic carbon, 94.3% removal in NH3–N and 91.4% removal in total Kjeldahl nitrogen is observed to be the most efficient coagulant and surprisingly, proves to be even better than Fenton. To understand the field applicability of the two treatment procedures, coagulation with FeCl3 and Fenton are compared with the UASB method which is currently employed at Gazipur landfill site, Delhi. With lesser operational cost than UASB, both FeCl3 and Fenton perform better on cost-efficiency scale. Switching from in-suit UASB method to the FeCl3 method of treatment may result in decreasing the operational cost by 71.9% and to conventional Fenton may result in decreasing the operational cost by 76.8%. High-performance thin-layer chromatography-mass spectrometry (HPTLC-MS) is one of the most modern hyphenated analytical methods available today. HPTLC-MS has recently been used to perform extensive analytical work using advanced features and still more analyses are in progress using these advanced features. Herbal medicines contain a wide range of bioactives that require proper identification for the activity and quality control. Recently, herbal medicine identification and quality control have become increasingly popular using high-performance liquid chromatography mass spectrometry. The aim of this paper is to provide a brief overview of recent method developments in analysis, 15 most significant herbal drug applications with their chromatographic conditions, pharmacological actions, and solvents used in the present paper. An extensive literature search was performed incorporating several databases, notably, Web of Knowledge, PubMed and Google Scholar, and other relevant published materials. In previous research, HPTLC and its hyphenation with MS allowed for quantification of analytes in complex matrixes at nanogram and picogram concentrations. Quantifying a wide variety of analytes using these techniques has been extremely accurate, selective, and sensitive. Throughout this review, the HPTLC-MS technique is discussed in relation to the quality control of herbal drugs. Hence, 15 herbal drugs were identified based on their RF values and m/z ratio by mass spectrometry by HPTLC-MS for the first time. In this compilation, researchers can gain insights into HPTLC-MS techniques for resolving quality control issues with herbal drugs using their fingerprint spectra. Besides, the application of HPTLC-MS methods could to be sufficiently precise and reproducible for established conditions and after validation may be used for routine quality control of herbal drugs/formulations in herbal industries. Isolation of seismomagnetic effect arising from the stress-induced modulations of magnetisation becomes difficult due to natural variations of magnetic disturbances. It has been critical to find out the precursory earthquake behaviour in any of the data including geomagnetic, for the last 50 years. To search seismomagnetic signals in the present study, we have analysed four earthquakes (Mw ≥ 4.9) during 2007–2011 related to Ghuttu MPGO situated in NW Himalayas, India. These four earthquakes were found to be eligible for which Ghuttu station is in the preparation zone and an index of magnetic variation (Imv), which quantifies anomalous variations of the magnetic field from geomagnetic data, has been estimated. For this endeavour, an assumption has been made considering the average of magnetic data for the quiet days in order to compare the analysis with respect to the magnetic storms. Using the magnetic data recorded at Ghuttu MPGO for the interval of nearly two months within the range of occurrence of four moderate earthquakes, the Imv has been computed. As a result, it has been observed that the proposed Imv computed is still sensitive to global magnetic disturbances as evidenced by comparing to Dst and sum Kp for the earthquakes considered in the present study. Therefore, it is summarised that the best way to isolate seismomagnetic effect is by comparing the data of pair of stations in differential mode, one located close to the area of stress built up and the other immediately outside the seismic zone, so that the background noise can be eliminated by analysing this kind of pair stations in order to achieve the goal. Several natural hazards, including earthquakes, may trigger disasters and the presence of disaster drivers further lead to the massive loss of life and property, every year around the world. The earthquakes are unavoidable, as exact earthquake prediction in terms of date, and time is difficult. However, with the advancement in technology, earthquake early warning (EEW) has emerged as a life-saving guard in many earthquake-prone countries. Unlike other warning systems (where hours of warning are possible), only a few seconds of warning is possible in the EEW system, but this warning may be very helpful in saving human lives by taking the proper action. The concept of EEW relies on using the initial few seconds of information from nearby instruments, performing basic calculations, and issuing the warning to the farther areas. A dense network or enough network coverage is the backbone of an EEW system. Because of insufficient station coverage, the estimated earthquake location is error-prone, which in turn may cause problems for EEW in terms of estimating strong shaking for the affected areas. Seismic instrumentation for EEW has improved significantly in the last few years considering the station coverage, data quality, and related applications. Many countries including the USA, Mexico, Japan, Taiwan, and South Korea have developed EEW systems and are issuing a warning to the public and authorities. Several other countries, namely China, Turkey, Italy, and India are in process of developing and testing the EEW system. This article discusses the challenges and future EEW systems developed around the world along with different parameters used for EEW. Background: Nepal has implemented the social health security program in certain health facilities to improve access to quality health services. The aim of the study is to understand the utilization pattern of social health security schemes and community's perception of the social health security program. Method: A descriptive cross-sectional mixed-method study was conducted among 300 households of Illam district who had enrolled in the social health security scheme. A multi-stage random sampling method was used. A semi-structured questionnaire was used to collect quantitative data whereas focus group discussions (FGDs) were conducted for qualitative data. Descriptive analysis, bivariate and multivariate analysis was done. FGDs were transcribed and thematic analysis was done. Findings: The utilization rate of social health security scheme was 88.7%. Factors associated with the utilization of program included: presence of under-five children and chronic illness in households, sex and age group. Focus group discussion revealed that people were aware of social health insurance and pleased with program implemented by government. However, the hospitals under the social health insurance were not able to meet their expectations. Conclusion: Although the utilization of social health security scheme was high, people were dissatisfied with the service provided by the hospital under the social health security scheme. Groundwater withdrawal at very fast rate poses threat on existing groundwater resources in different parts of the world. This reduction in groundwater levels significantly disturbed the natural aquifer flow rate and thereby different hydrogeochemical processes, which may further impair the groundwater quality. The groundwater quality in rural area of Chhattisgarh State is degraded, and the problem of saline water poses health risk to people. In this research investigation, suitability of groundwater of Bemetara District, Chhattisgarh, India, has been evaluated for drinking purpose through water quality index (WQI) method and principal component analysis (PCA). Total 116 groundwater samples were collected during the pre-monsoon (June 2019) and post-monsoon season (December 2019) and analyzed for physicochemical parameters. Total dissolved solids ranged from 250 to 10,440 mg/L and 289 to 3583 mg/L during pre-monsoon and post-monsoon, respectively, and 55% of the total samples exceeded acceptable BIS limit in pre-monsoon, while about 66% samples exceeded in post-monsoon season. SO42− concentrations varied from 3 to 5734 mg/L during pre-monsoon and 4.5 to 2002 mg/L during post-monsoon, respectively. Total 28% samples in pre-monsoon and 18% samples in post-monsoon season exceeded the maximum permissible BIS limit (400 mg/L) of SO42− ion in the study area. On the basis of WQI, the quality of groundwater varies from "Excellent water" to "Good water" category. The groundwater of northeastern part of the district is not suitable for drinking, and therefore, it is recommended to treat this groundwater before human consumption with special reference to SO42− contamination. PCA inferred that four components are sufficient to explain the variance in chemistry of groundwater that is mainly governed by dissolution of gypsum mineral, other rock–water interaction and anthropogenic activities. Further, water quality was improved in the direction of groundwater flow in the study area, establishing a direct relationship between groundwater flow and water quality of the Bemetara District. Hydrodynamic and heat transfer characteristics of long-term stable nanofluids are very crucial for their industrial applications. Also utilization of coils is beneficial for industries as it provides a high rate of heat transfer and is compact in size too. So, the main focus of this study is to investigate the hydrodynamic and heat transfer characteristics of long-term stable f-CNT nanofluids when flowing inside a coil-based heat exchanger. f-CNT nanofluids were prepared by utilizing modified two-step method. To analyze the effect of different parameters on hydrodynamic and convective heat transfer characteristics, f-CNT concentration, D c , and Re were varied from 0 to 0.048 vol%, 95 to 175 mm, and 2300 to 9500, respectively. According to results, f-CNT concentration, D c , and Re substantially influenced the hydrodynamic and heat transfer characteristics of f-CNT nanofluids. It was found that improvement in h (152%) was much higher than the enhancement in friction factor (49%) when f-CNT nanofluid at 0.048 vol% was flowing through the coil of 95 mm diameter. Based on the heat transfer and hydrodynamic data, performance index was evaluated. The maximum performance index was calculated ⁓ 2.5, suggesting that the utilization of helical coils and f-CNT nanofluids is an excellent choice in industrial applications. Based on the experimental data, empirical correlations have been proposed to calculate the friction factor and Nusselt number for f-CNT nanofluids when flowing inside coils of different diameters and at different f-CNT concentrations. This study aims to estimate attenuation characteristics of the central Himalayan region of India concerning various strong-motion parameters such as Kappa value (κ) and site effects. We have tried to elaborate on the regional structural heterogeneities and their implications towards the seismic hazard assessment of the study region. A total of 81 earthquakes recorded at 50 stations situated in the central Himalayan region of India are used for the purpose. The particular focus is kept on Kappa value, which shows variability from 0.03 s to 0.095 s, inferring the higher values obtained in plains with deep sediment accumulations proving high-frequency energy dissipation and stiff-soil/rocky sites exhibit comparatively limited attenuation accordingly. To substantiate these results various attenuation parameters such as coda wave quality factor (Qc), intrinsic attenuation parameter (Qi), and scattering attenuation parameter (Qs), have been estimated for two regions in the central seismic gap Himalayan region of India employing the single backscattering model and Wennerberg formulation. The estimated values of Qc, Qi, and Qs are found to be highly dependent on frequency in the frequency range 1.5–24 Hz for both the regions. The average frequency-dependent relationships ( Q = Q 0 f η ) estimated for both regions are Q c = 158 f 1.18 and Q c = 194 f 1.2 , respectively. The low value of Q0 shows that the region is highly heterogeneous while the higher value of η indicates higher seismicity in the area. It is also found that intrinsic attenuation is predominant over the scattering attenuation, envisaging the behavior of the wave attenuation through the absorption within the granitic layer at shallow depths. At lower frequencies, Qc values are found close to Qs values, which is in agreement with the theoretical measurements suggesting the presence of complex crustal heterogeneities beneath the region affecting the propagation of seismic waves experiencing considerable decay of energy through scattering. To confirm the aggregate attenuation on the stations, the site characteristics are also determined for examining the behavior of the amplification as the ground motion is comprised of the combined effect of the source, path, and site. The sites are amplified at a predominant frequency (fpeak) in between 1.5 to 10 Hz for the central Himalayan region. The different attenuation and amplification parameters like kappa, Q, and site effects can be utilized for detailed seismic hazard analysis (based on ground motion prediction equations) of the area as this region is of great importance from a socio-economic point of view. The statistically point process model known as epidemic-type aftershock sequence (ETAS) model is employed for systematically investigating the seismic quiescence or seismic anomalies around the focal regions of large/strong earthquakes for NW Himalaya. For this propose, the model predicted the expected occurrence rates of earthquakes by estimating the model parameters from the earthquake occurrences times using maximum likelihood method, has been used. Then the exhibited relative quiescence due to decreasing occurrence rates from the modeled ones can be identified by inspecting the abnormally downward deviated plot from the extended cumulative curve of the Residual Point Process (RPP) events. Examination of such RPP events in the whole time interval exhibits significant 1.5 years and 2.0 years of relative seismic quiescence before the strong 1991 Uttarkashi (MW 6.8) and 1999 Chamoli (MW 6.6) earthquakes, respectively. Considering the optimally oriented planes of Uttarkashi earthquake, the Coulomb stress changes (ΔCFS) have been investigated to check the rate of seismicity around the focal region of Chamoli earthquake. It has been found that ΔCFS of Uttarkashi earthquake exhibits stress shadow in or near the source zone of Chamoli earthquake and eventually decreases seismicity rates due to seismic quiescence in the source zone. On the other hand, the detected quiescence and activation relative to the predicted seismicity rate are consistent with the obtained Coulomb stress to depict the associated anomalies being sensitive enough to detect a slight stress change in the study region. Henceforth, the increased or decreased seismic activity due to seismic activation or quiescence is found to be consistent with the patterns of the Coulomb's stress changes calculated on the ruptured fault planes of Uttarkashi earthquake. Hence, this ETAS model based on statistical technique can thus be incorporated with other sensitive geophysical instruments for identifying seismically quiet period not only in the seismic gaps, but also in its neighborhoods along the Himalayan range for mitigating seismic hazards due to impending great earthquakes. ICP-OES is a powerful, versatile, and advanced analytical technique with excellent detection properties. Due to its extraordinary features, it has been widely employed for the analysis of a wide variety of chemical elements in the past few years with great success. It offers the least detection time, lower detection limits, broader linear dynamic range, and greater matrix tolerability as well as negligible chemical interferences. Beyond this, it can handle multiple varieties of samples including aqueous, inorganic, organic liquids, and solids as well. It comprises complex instrumental makeup which enables it to detect up to 2 to 70 elements simultaneously with great accuracy. Recent reports evidenced that this hyphenated technique has been employed in several analytical determinations including food analysis, agricultural investigations, geological studies, drug/metabolite analysis, and environmental and forensic sciences. In the present compilation, a detailed description of the fundamental principles of ICP-OES has been provided along with its various sophisticated functional components. Also, the reported applications of this technique in different fields have been discussed highlighting the basic experimental setups and outcomes by presenting the data as tables. Site effect estimation using recorded ground motion is an effective approach to assess the seismic hazard of a region. Keeping the same thing in mind, an endeavor is being made to study the local site effects at different locations in the North East region of India through analysis of recorded strong ground motion data provided by Indian strong motion network. The data recorded at 25 sites from 37 earthquakes with a magnitude range M L 4.0–6.9 have been utilized. The estimated predominant frequencies ( f p e a k ) using horizontal to the vertical spectral ratio (HVSR) is well observed for various sites placed in various geological formations like the Precambrian, Tertiary and Quaternary Consequently, the pseudo velocity response (PSV) for 5% critical damping is estimated and compared with the regional geological formations, especially in the Brahmaputra Valley region. It is further noticed that the HVSR, as well as PSV, show a noticeable correlation according to the geological set up of the region for most of the sites, giving a clear idea about the site effect evaluation. Analyzed strong motion data also show the effect of non-linearity, which is an important parameter as it explains the inelastic behavior of the soil causing a reduced or non-linear amplification. Since some of the sites used in present work do not correlate with geology, the sites used for HVSR estimation are used to classify them according to the classical approach employing f p e a k . All the sites are classified in four different classes namely A, B, C, and D based on shear wave velocity in the upper 30 m. The outcome of the present study is being shown in the form of contour maps for f p e a k and site amplification for various types of buildings in order to assess the seismic hazard and risk mitigation. Background: The content and nature of media reports could influence suicide prevention measures. Aim: To evaluate contemporary online media reports' compliance with guidelines for responsible reporting of suicidal acts from Indian resources. Methods: We included English and Hindi articles for reports concerning suicide, published by local and national media sources for 30 consecutive days from the day of death of a celebrity by alleged suicide. The search was performed in Google News with predefined search queries and selection criteria. Two independent investigators did data extraction. Subsequently, each news report was assessed against guidelines for the responsible reporting of suicide by the WHO and the Press Council of India. Results: We identified 295 articles (Hindi n = 172, English n = 123). Results showed more than 80% of the media reports deviated from at least one criterion of the recommendations. A maximum breach was seen in the news article's headlines, sensational reportage and detailed suicide methods description. Significant differences were seen in the quality of English and Hindi reporting and reporting celebrity and noncelebrity suicide. Additional items revealed were reporting suicide pacts, linking multiple suicides in a single news report, allowing user-generated threads and linking Religion and suicide. Bioremediation is a promising technology for the treatment of environmental contaminants and paving new avenues for the betterment of the environment. Over the last some years, several approaches have been employed to optimize the genetic machinery of microorganisms relevant to bioremediation. Metabolic engineering is one of them that provides a new insight for bioremediation. This review envisages the critical role of these techniques toward exploring the possibilities of the creation of a new pathway, leading to pathway expansion to new substrates by assembling of catabolic modules from different origins in the same microbial cell. The recombinant DNA technology and gene editing tools were also explored for the construction of metabolically engineered microbial strains for the degradation of complex pollutants. Moreover, the importance of CRISPR-Cas system for knock-in and knock-out of genes was described by using recent studies. Further, the idea of the cocultivation of more than one metabolic engineered microbial communities is also discussed, which can be crucial in the bioremediation of multiple and complex pollutants. The recorded strong motion data in the Delhi region provide an excellent opportunity to study high-frequency decay parameter, kappa (κ) for the National Capital (Delhi) region and to further understand its implications to study the site effects characterized by different stations within the vicinity of the study region. The kappa values are estimated at 30 locations from 99 accelerograms of 19 earthquakes recorded in the Delhi region and are found to vary from place to place depending upon the controlling parameters, primarily the site characterization. The estimated average values of 'κ' lie in the range 0.0118–0.0537 s for the various locations of the region depending upon the source, path, and site characteristics of earthquakes considered in the present study. The distance dependence is found insignificant, while there is a scatter in the variation of κ values with that of magnitude which indicates that κ is more related to the site characteristic for the entire Delhi region which in turn reveals the fact of the basic criterion of the κ parameter. To affirm the total attenuation on the instruments, the site effects demonstrate the behavior of amplification to the geological exposure. It has been found that the various sites under consideration for the study area amplify between 0.6 and 7.0 Hz predominant frequency ( f peak ) and agree with the geological arrangements of the region. Based on the present study, the most vulnerable areas are the northeastern region of Delhi which lies in proximity to the flood plains of Yamuna river and alluvial deposits of younger origins of the foreland basin along with the southwestern part of Delhi capital which is comprised of the water-saturated alluvial deposits. The estimated 'κ' values are found to be correlated with those of the estimated site amplifications and are useful in strong ground motions simulation for the proper evaluation of seismic hazard to build a seismic risk resilient society. Background: Media portrayal of mental illness may influence public stigma and service utilization. Aim: This study aims to explore the overall tone and content of the news articles on mental illnesses in India. Materials and Methods: This descriptive cross-sectional study was conducted on online English and Hindi news portals for articles which made reference to mental illness, published by local and national media sources between September 2019 and August 2020. The search was performed using location and news-only filters in the "Google" search engine; we used predefined search queries and selection criteria. A previously published checklist was used for the content analysis; it was done by two independent investigators. Results: A total of 273 news reports met inclusion criteria (Hindi n = 164, English n = 109). Results showed that more than half (54.9%) had a positive tone. Nearly a third (30.8%) of these articles were stigmatizing in tone. Persons with mental illness were portrayed as violent, unreliable, and unpredictable. Uses of blanket terms (33.3%) such as mentally ill, mentally unstable, mentally retarded, and stigmatizing photographs (36.9%) were also common. Overall, Hindi media had a significantly better quality of reporting than the English language media. National media reports had a less negative and stigmatizing tone than local articles. Aims We conducted a cross-sectional survey to estimate the prevalence and clinical manifestation of disulfiram ethanol reaction (DER) and isopropanol toxicity (IT) in patients with alcohol use disorders, on disulfiram. Alcohol-based hand rub contains either ethanol or isopropanol or both. COVID-19 pandemic has led to wide scale usage of sanitizers. Patients with alcohol use disorders, on disulfiram, might experience disulfiram ethanol like reactions with alcohol-based sanitizers. Methods We telephonically contacted 339 patients, prescribed disulfiram between January 2014 and March 2020. The assessment pertained to the last 3 months (i.e. third week of March to third week of June 2020). Result The sample consisted of middle-aged men with a mean 16 years of alcohol dependence. Among the 82 (24%) patients adherent to disulfiram, 42 (12.3%) were using alcohol-based hand rubs. Out of these, a total of eight patients (19%; 95% CI 9–33) had features suggestive of DER; four of whom also had features indicative of IT. Five patients (62.5%) had mild and self-limiting symptoms. Severe systemic reactions were experienced by three (37.5%). Severe reactions were observed with exposure to sanitizers in greater amounts, on moist skin or through inhalation. Objective  Carbapenems are last resort antibiotics for multidrug-resistant Enterobacteriaceae. However, resistance to carbapenem is increasing at an alarming rate worldwide leading to major therapeutic failures and increased mortality rate. Early and effective detection of carbapenemase producing carbapenem-resistant Enterobacteriaceae (CRE) is therefore key to control dissemination of carbapenem resistance in nosocomial as well as community-acquired infection. The aim of present study was to evaluate efficacy of Modified strip Carba NP (CNP) test against Modified Hodge test (MHT) for early detection of carbapenemase producing Enterobacteriaceae (CPE). Material and Methods Enterobacteriaceae isolated from various clinical samples were screened for carbapenem resistance. A total of 107 CRE were subjected to MHT and Modified strip CNP test for the detection of CPE. Statistical Analysis  It was done on Statistical Package for the Social Sciences (SPSS) software, IBM India; version V26. Nonparametric test chi-square and Z -test were used to analyze the results within a 95% level of confidence. Results  Out of 107 CRE, 94 (88%) were phenotypically confirmed as carbapenemase producer by Modified strip CNP test and 46 (43%) were confirmed by Modified Hodge Test (MHT). Thirty-eight (36%) isolates showed carbapenemase production by both MHT and CNP test, 56 isolates (52%) were CNP test positive but MHT negative, eight (7%) isolates were MHT positive but CNP test negative and five (5%) isolates were both MHT and CNP test negative. There is statistically significant difference in efficiency of Modified CNP test and MHT (p < 0.05). Conclusion  Modified strip CNP test is simple and inexpensive test which is easy to perform and interpret and gives rapid results in less than 5 minutes. It has high degree of sensitivity and specificity. Modified strip CNP test shows significantly higher detection capacity for carbapenemase producers as compared with MHT. [ABSTRACT FROM AUTHOR]

Delhi, National Capital Region (NCR) of India, falls in the seismic Zone IV (Zone factor 0.24) on the seismic zoning map prepared by the Bureau of Indian Standards (BIS), and this region may experience devastating intensities in case of plausible moderate to the major earthquake in the vicinity. The strategic geological, geomorphological, and geographic characteristics make this seat of administrative power more vulnerable toward the earthquake disaster right from the ancient periods. Therefore, we study the impact of the M 6.0 earthquake sourced at Sohna fault in the neighborhood of the capital region by generating synthetic accelerograms through semiempirical envelop technique. The observed accelerograms of November 25, 2007 ( M w 4.7) earthquake have been modeled to utilize the reliability of the semiempirical approach. To analyze the actual scenario, the ground motions at the surface have been generated after the incorporation of the site effects because different soil conditions of NCR fascinate different degrees of damage in case of a destructive earthquake. As a result, the obtained peak ground acceleration (PGA) varies between 100 and 600 cm/s2, and some of the sites exhibit even higher PGA values being situated on the sediments of river-oriented plain areas and proximity of the source. The spatial distribution of estimated values of PGA and spectral accelerations at different periods show that sites in NCR like Delhi (600 cm/s2), Sonipat (633 cm/s2), Gurgaon (461 cm/s2), and Faridabad (300 cm/s2) exhibit high to severe seismic hazard in case of M 6.0 at Sohna fault and it is suggested that a population of about 4.78 million along with the infrastructure of this region is exposed to high risk. The estimated seismic exposure of the population is important to utilize the resources properly before the destructive earthquake incidence. The hazard maps for PGA and different structural periods in the NCR region reveal the level of seismic hazard and risk of the study region. These hazard maps are very helpful for administrators, stakeholders, and civil engineers to construct earthquake-resistant structures to minimize the risk generated by the future impending earthquakes. The exponential growth of the buildings, industries, businesses, etc., attracts the attention of urban area planners because of high seismic risk due to the damaging earthquakes, and its severity must be understood to save the life and property to mitigate the natural disaster like an earthquake by proper disaster mitigation plans, especially in the metropolitan cities like Delhi NCR. Bioremediation is a better alternative and widely accepted approach used for efficient degradation of environmental pollutants released from industries, urban and agricultural activities due to its eco-friendly nature. Systems biology helps in the identification of new genes, proteins, metabolites, and metabolic pathways involved in bioremediation. Such information can be used for designing synthetic microbial communities that can degrade multiple recalcitrant pollutants simultaneously. This review gives a brief insight into various systems biology tools towards providing a greater understanding of microbial behaviour and improving the way of bioremediation. These techniques alone or in combination, provide a way to understand and improve the genetic potential of microorganisms to remediate various environmental contaminants efficiently. Further, this review also describes the successful employment of synthetic microbial consortium in the bioremediation. Moreover, In-silico tools are also described to analyse the data obtained through different laboratory experiments as well to predict the behaviour of microbial consortium towards the pollutants using different databases. Today, environmental pollution is a serious problem, and bioremediation can play an important role in cleaning contaminated sites. Remediation strategies, such as chemical and physical approaches, are not enough to mitigate pollution problems because of the continuous generation of novel recalcitrant pollutants due to anthropogenic activities. Bioremediation using microbes is an eco-friendly and socially acceptable alternative to conventional remediation approaches. Many microbes with a bioremediation potential have been isolated and characterized but, in many cases, cannot completely degrade the targeted pollutant or are ineffective in situations with mixed wastes. This review envisages advances in systems biology (SB), which enables the analysis of microbial behavior at a community level under different environmental stresses. By applying a SB approach, crucial preliminary information can be obtained for metabolic engineering (ME) of microbes for their enhanced bioremediation capabilities. CNT nanofluids are getting attention in heat transfer applications due to their very high thermal conductivity in comparison with conventional fluids. For commercial exploitation of CNT nanofluids as heat transfer media, they must have long-term stability. In this study, the two-step method was modified to prepare dynamically stable CNT nanofluids by utilizing commercial grade multiwalled carbon nanotubes and SDBS as a surfactant. The modified technique consists of separation of coarse agglomerates of CNT from the CNT nanofluids by applying centrifugal action after its preparation. The effect of relative centrifugal force was also studied for the very first time on the stable concentration of CNT nanofluids. The stability of CNT nanofluids was analyzed by measurement of their CNT concentration and Zeta potential. Results showed that CNT nanofluids possess good stability and remain stable for more than 15 months. In addition to stability, thermo-physical properties such as thermal conductivity, density, and viscosity of CNT nanofluids were also measured. The results of this study elucidated the effect of RCF on the stable concentration of CNT nanofluids. It is expected that the results obtained in this study may significantly contribute to the proper tailoring of CNT nanofluids, by providing long-term stable CNT nanofluids which are suitable for industrial heat transfer applications. Plants are continually facing biotic and abiotic stresses, and hence, they need to respond and adapt to survive. Plant response during multiple and combined biotic and abiotic stresses is highly complex and varied than the individual stress. These stresses resulted alteration of plant behavior through regulating the levels of microRNA, heat shock proteins, epigenetic variations. These variations can cause many adverse effects on the growth and development of the plant. Further, in natural conditions, several abiotic stresses causing factors make the plant more susceptible to pathogens infections and vice-versa. A very intricate and multifaceted interactions of various biomolecules are involved in metabolic pathways that can direct towards a cross-tolerance and improvement of plant’s defence system. Systems biology approach plays a significant role in the investigation of these molecular interactions. The valuable information obtained by systems biology will help to develop stress-resistant plant varieties against multiple stresses. Thus, this review aims to decipher various multilevel interactions at the molecular level under combinatorial biotic and abiotic stresses and the role of systems biology to understand these molecular interactions. PURPOSE: Pseudomonas aeruginosa causes a wide spectrum of infections including bacteremia, pneumonia, urinary tract infection, etc., Metallo-beta-lactamase (MBL) producing P. aeruginosa is an emerging threat and cause of concern as they have emerged as one of the most feared resistance mechanisms. This study was designed to know the prevalence of MBL production in P. aeruginosa and to evaluate the four phenotypic tests for detection of MBL production in imipenem-resistant clinical isolates of P. aeruginosa. METHODS: Totally, 800 isolates of P. aeruginosa isolated from various clinical samples were evaluated for carbapenem resistance and MBL production. All imipenem-resistant strains were tested for carabapenemase production by modified Hodge test. Screening for MBL production was done by double-disc synergy test and combined disc test (CDT). Confirmation of MBL production was done by the E-test (Ab BioDisk, Solna, Sweden). RESULTS: Out of the 800 isolates of P. aeruginosa, 250 isolates were found resistant to imipenem. Based on the results of E-test, 147 (18.37%) isolates of P. aeruginosa were positive for MBL production. The CDT has the highest sensitivity and specificity for the detection of MBL production as compared to other tests. CONCLUSION: The results of this study are indicative that MBL production is an important mechanism of carbapenem resistance among P. aeruginosa. Use of simple screening test like CDT will be crucial step toward large-scale monitoring of these emerging resistant determinants. In the present work, acceleration response spectra are determined from earthquakes which have occurred in the NE region and the effect of local geology on its shape is studied. One hundred and ninety-five strong ground motion time histories from 45 earthquakes which have occurred in the NE region having a magnitude range of 3.5 ≤ Mw ≤ 6.9 and a distance range of 20-600 kms are used. It is observed that the shape of the normalized acceleration response spectra is influenced by the local site conditions and regional geology. The influence of magnitude and distance on the spectra is also studied. The present study is carried out for three categories of rocks: Pre-Cambrian, Tertiary and Quaternary. It is inferred that the acceleration response spectra in the current Indian code designed for the entire country are applicable for NE region as it is within the spectral limits prescribed in Indian code. The ground motion is amplified at higher frequencies for stations located on hard rock, while for stations located on alluvium sites, it is amplified at lower frequencies. The sites located on hard rock show lowest values of spectral acceleration than the sites located on alluvium sites. The results obtained in the present study are compared with the similar results obtained in the stable continent region like Gujarat. It is found that the dominating period of response spectrum of similar rock types is found to be at higher side for NE region as compared to Gujarat region. Introduction: Fungal respiratory infections are important cause of mortality and morbidity among HIV positive individuals. They account for up to 70% of illness in Acquired Immunodeficiency Disease Syndrome cases (AIDS). The range of illness varies from asymptomatic mucosal candidiasis to overwhelming disseminated infections. In these patients dissemination of fungus leads to very serious outcomes hence, it is important to have the knowledge of prevailing profile of fungus causing infections, so that it can be treated at the onset. Low CD4+T lymphocyte count is an excellent indicator of decreased immunity and can also be helpful to predict opportunistic fungal respiratory infections and other complications. Aim: To define the fungal aetiology of lower respiratory tract infections in HIV positive patients and to correlate the occurrence of different fungi with CD4+ T lymphocyte count. Materials and Methods: This was a cross sectional study conducted between May 2014 to April 2015, on 180 treatment naive HIV seropositive patients with lower respiratory tract infections attending the Integrated Counselling and Testing Centre, SMS Medical College, Jaipur, Rajasthan. Early morning expectorated and induced sputum samples were collected and processed for isolation and identification of fungal species. CD4+ T lymphocyte count estimation was done by BD FACS Calibur. Results: Fungal species were isolated from 155 (86.1%) patients. The most common isolate was Candida albicans (31.7%), followed by Aspergillus niger (17.7%) and Aspergillus flavus (10%). The fungal species were most commonly isolated from patients with CD4+ T lymphocyte cell less than 200 cells/μl. Conclusion: Fungal infections were seen in 86.1% of HIV positive patients with lower respiratory tract infections hence, high level of clinical suspicion for fungal aetiology of respiratory infections in HIV positive patients should be kept in mind. Earthquakes are deadliest among all the natural disasters. The areas that have experienced great/large earthquakes in the past may experience big event in future. In this study, we have simulated Kangra earthquake (1905, Mw 7.8) and a hypothetical great earthquake (Mw 8.5) in the north-west Himalaya using Empirical Green's Function (EGF) technique. Recordings of Dharamsala earthquake (1986, Mw 5.4) are used as Green function with a heterogeneous source model and an asperity. It has been observed that the towns of Kangra and Dharamsala can expect ground accelerations in excess of 1 g in case of a Mw 8.5 earthquake and could have experienced an acceleration close to 1 g during 1905 Kangra earthquake. The entire study region can expect acceleration in excess of 100 cm/s in case of Mw 7.8 and 200 cm/s in case of Mw 8.5. The sites located near the rupture initiation point can expect accelerations in excess of 1 g for the magnitudes simulated. For validation, the estimates of the PGA for Mw 7.8 simulation are compared with isoseismal studies carried out in the same region after the Kangra earthquake of 1905 by converting PGA values to intensities. It was found that the results are comparable. The target earthquakes (Mw 7.8 and Mw 8.5) are simulated at depth of 20 km and 30 km to examine the effect of PGA for different depths. The PGA values obtained in the present analysis gave us an idea about the level of accelerations experienced in the area during 1905 Kangra earthquake. Tuberculosis is caused by Mycobacterium tuberculosis (M.tb) as well as Non-tubercular mycobacterium (NTM) with similar clinical presentation. Infections due to NTM are reported to have increased in the past few years. Growth of M.tb is inhibited by p-Nitrobenzoic acid (PNB), whereas, NTM are resistant. One hundred and nine isolates from various clinical samples were identified up to species level by their growth rate, pigmentation, and a battery of biochemical tests, including niacin accumulation, nitrate reduction, and heat-stable catalase (680C) reactions. Para-nitrobenzoic acid (PNB) inhibition test was performed to differentiate between M.tb and NTM. PNB was added to the Lowenstein-Jensen (LJ) medium and BACTECTM MIGIT (Mycobacteria Growth Indicator Tube)960 medium to a final concentration of 500 μg/ml. All the M.tb isolates, including Mycobacterium tuberculosis H37Rv (standard strain), were inhibited by PNB on both LJ and MGIT 960. Of the NTM isolates, all were resistant to PNB on MGIT 960 and on LJ PNB, except one isolate of Mycobacterium marinum that was resistant to MGIT 960 PNB, but was susceptible to LJ PNB. The reporting time for M.tb ranged from 4-11 days (median 5.9 days) by MGIT 960 and for NTM it was 2-10 days with an average of 4.5 days. Malaria is caused by the protozoan Plasmodium. The parasite Plasmodium completes its life cycle inside two hosts, i.e. human and mosquito. Among all known Plasmodium species, Plasmodium falciparum is known to cause maximum mortality. Various studies done on the mosquito stages of the parasite suggest that the proteins present on the parasite's surface are responsible for its survival under the adverse conditions prevailing in the mosquito midgut. When human blood containing Plasmodium gametocytes enters the mosquito gut, the gametocytes form gametes which then fuse to form zygotes. At this stage two closely related proteins, Pfs25 and Pfs28 are expressed on the surface of the parasite which continue to express up to the young oocyst stage. These proteins present on zygotes, ookinetes and young oocysts of Plasmodium are categorized in P25 and P28 families and are well known malaria vaccine candidate proteins. In this study, we have done sequence analysis, homology modeling and docking studies of a typical member of the P25 family of ookinete surface protein, i.e. Pfs25 from Plasmodium falciparum. We have built a 3D model of Pfs25 based on the X-ray crystallographic structure of Pvs25 from Plasmodium vivax. Also we have modeled the Fv region of the malaria transmission blocking monoclonal antibody 4B7. This antibody is the transmission blocking monoclonal antibody for Pfs25 protein. Pfs25 and 4B7 scFv (single chain variable fragment only) docking results indicate that EGF domain III of the Pfs25 protein interacts with the scFv region of modeled 4B7 antibody forming seven hydrogen bonds out of which six are formed with heavy chain of scFv region. Antioxidants can remove damaging reactive oxygen species produced as by-products of desiccation and respiration during late embryogenesis, imbibition of dormant seeds and germination. We have expressed a protein, PER1, encoded by the Balem ( barley aleurone and embryo) transcript previously called B15C, and show it to reduce oxidative damage in vitro. PER1 shares high similarity to a novel group of thiol-requiring antioxidants, named peroxiredoxins, and represents a subgroup with only one conserved cysteine residue (1-Cys). PER1 is the first antioxidant belonging to the 1-Cys subgroup shown to be functionally active, and the first peroxiredoxin of any kind to be functionally described in plants. The steady state level of the transcript, Per1, homologous to a dormancy-related transcript (pBS128) from bromegrass ( Bromus secalinus), increases considerably in imbibed embryos from dormant barley ( Hordeum vulgare L.) grains. Our investigations also indicate that Per1 transcript levels are dormancy-related in the aleurone layer of whole grains. In contrast to most seed-expressed antioxidants Per1 disappears in germinating embryos, and in the mature aleurone the transcript is down-regulated by the germinating embryo or by gibberellic acid (GA). Our data show that the barley seed peroxiredoxin is encoded by a single Per1 gene. Possible roles of the PER1 peroxiredoxin in barley grains during desiccation, dormancy and imbibition are discussed. Diffuse cutaneous leishmaniasis (DCL) is characterized by the presence of a large number of lesions at several anatomic sites (head, limbs and trunk). The lesions include papules, nodules and areas of diffuse infiltration that do not ulcerate and reveal abundant parasites on histopathological examination. DCL and human immunodeficiency virus (HIV) co-infections are seldom reported. We report two cases of DCL in HIV positive patients without visceral involvement. Introduction: Gastrointestinal Tract (GIT) infections are among the most frequent infections in HIV/AIDS patients. The intestinal opportunistic parasitic infections in HIV-infected subjects present most commonly as diarrhoea. A study was conducted to determine the prevalence of enteric parasitic infections in HIV infected patients with diarrhoea, with different levels of immunity. Methods: This study was carried out at the HIV Lab of the Microbiology Department of a tertiary care teaching hospital in Jaipur, Rajasthan, between June-October 2009 among consecutively enrolled 75 HIV infected patients who presented with diarrhoea. Stool samples were collected and examined for enteric parasites by using microscopy and special staining methods. The CD4 + cell counts were estimated by using the FACS count system. Results: Intestinal parasitic pathogens were detected in 38.66% patients, Cryptosporidium species was the most common enteric opportunistic parasite which accounted for 37.93 % of the total parasites, followed by Isospora belli 31.03 %. In the HIV infected patients with CD4+ counts of < 200 cells/µl, parasites were identified in 56.25 % patients and in HIV patients with CD4+ counts between 200-499 cells /µl, parasites could be identified in 27.5 % of the patients . No parasite was detected in the patients with CD4+ counts of >500 cells/ µl. Conclusion: Parasitic infections were detected in 38.66% HIV infected patients with diarrhoea and a low CD4+ count was significantly associated with opportunistic infections. Identification of the aetiological agent of diarrhoea in an HIV patient is very important, as it can help in the institution of the appropriate therapy and the reduction of the morbidity and the mortality in these patients.
