Purpose: The overall survival rate for metastatic osteosarcoma hovers around 20%. Responses to second-line chemotherapy, targeted therapies, and immunotherapies have demonstrated limited efficacy in metastatic osteosarcoma. Our objective is to validate differentially expressed genes and signaling pathways between non-metastatic and metastatic osteosarcoma, employing single-cell RNA sequencing (scRNA-seq) and additional functional investigations. We aim to enhance comprehension of metastatic mechanisms and potentially unveil a therapeutic target. Methods: scRNA-seq was performed on two primary osteosarcoma lesions (1 non-metastatic and 1 metastatic). Seurat package facilitated dimensionality reduction and cluster identification. Copy number variation (CNV) was predicted using InferCNV. CellChat characterized ligand-receptor-based intercellular communication networks. Differentially expressed genes underwent GO function enrichment analysis and GSEA. Validation was achieved through the GSE152048 dataset, which identified PDGFD-PDGFRB as a common ligand-receptor pair with significant contribution. Immunohistochemistry assessed PDGFD and PDGFRB expression, while multicolor immunofluorescence and flow cytometry provided insight into spatial relationships and the tumor immune microenvironment. Kaplan-Meier survival analysis compared metastasis-free survival and overall survival between high and low levels of PDGFD and PDGFRB. Manipulation of PDGFD expression in primary osteosarcoma cells examined invasion abilities and related markers. Results: Ten clusters encompassing osteoblasts, osteoclasts, osteocytes, fibroblasts, pericytes, endothelial cells, myeloid cells, T cells, B cells, and proliferating cells were identified. Osteoblasts, osteoclasts, and osteocytes exhibited heightened CNV levels. Ligand-receptor-based communication networks exposed significant fibroblast crosstalk with other cell types, and the PDGF signaling pathway was activated in non-metastatic osteosarcoma primary lesion. These results were corroborated by the GSE152048 dataset, confirming the prominence of PDGFD-PDGFRB as a common ligand-receptor pair. Immunohistochemistry demonstrated considerably greater PDGFD expression in non-metastatic osteosarcoma tissues and organoids, correlating with extended metastasis-free and overall survival. PDGFRB expression showed no significant variation between non-metastatic and metastatic osteosarcoma, nor strong correlations with survival times. Multicolor immunofluorescence suggested co-localization of PDGFD with PDGFRB. Flow cytometry unveiled a highly immunosuppressive microenvironment in metastatic osteosarcoma. Manipulating PDGFD expression demonstrated altered invasive abilities and marker expressions in primary osteosarcoma cells from both non-metastatic and metastatic lesions. Conclusions: scRNA-seq illuminated the activation of the PDGF signaling pathway in primary lesion of non-metastatic osteosarcoma. Context: Landscape sensation is essential for the delivery of cultural ecosystem services (CESs), yet the pathways through which these services are delivered remain inadequately understood. Exploring how people obtain CESs from landscapes facilitates better understanding of the tradeoffs and synergies between ecosystem services and landscape sustainability. Objectives: This study aimed to elucidate the sensory pathways that links landscape attributes to CESs, focusing on the roles of cognitive and affective experiences. Methods: We analyzed social media comments for the measurement scale of cognition. We employed partial least squares structural equation modeling to integrate sensation, cognition, affect, and satisfaction, using questionnaire data (n = 503). Results: Cognitive comprehensions and affective responses play a crucial role in interpreting CESs while sensory experiences do not directly determine people’s satisfaction with CESs. The effective pathways are achieved through the sole mediator of cognition or by serial mediators of cognition and affect. Of the two mediators, cognition has a more profound mediating effect than affect. Conclusions: Both physical and biological components, such as landscape sensory attributes, as well as cognitive and affective responses, influence human-nature interactions. Due to the harsh operating conditions experienced by 1Cr17Ni2 steel, efforts were made to optimize its performance by subjecting 1Cr17Ni2 stainless steel to nitriding treatments at temperatures of 460 °C, 500 °C, and 550 °C, each for durations of 8 and 16 h. The formation state of its cross section was observed through a metallurgical microscope and scanning electron microscope, and it was characterized by hardness measurement. Through a ball-on-disk wear experiment, the adhesive wear and friction coefficient of its non-lubricated sliding were measured. The phase composition of its surface was measured by XRD. The results revealed that nitriding led to the formation of a modified layer on the surface of the samples, with a depth of 130 μm after nitriding at 550 °C for 16 h. The hardness of the modified layer exceeded that of the matrix, reaching up to 1400 Hv0.1. X-ray diffraction (XRD) analysis of the sample surfaces indicated the presence of high-hardness phases such as CrN, γ′-Fe4N, and ε-Fe2-3N. This article predicts the mechanical properties of nitrided phases in high-alloy martensitic stainless steel through first-principles computational methods. Urban green spaces play a crucial role in enhancing the well-being of urban residents and promoting sustainable urban development. However, optimizing the planning and management of urban green spaces to meet residents' diverse needs and preferences poses a considerable challenge. This study addresses this challenge by employing a landsenses ecology approach, integrating residents' perspectives into the planning and design of urban green spaces. Starting from human needs, a conceptual framework for the meliorization model of urban green spaces is constructed, grounded in the principles of landsense creation and incorporating a "design-simulation-management" process. Through this model, the mechanisms driving the meliorization process are explored. This study contributes to improving the meliorization process in landsenses ecology, while expanding the theoretical framework and methodology of landscape ecology. By emphasizing the dynamic interactions between land planning, construction, and residents' experiences, this study provides valuable insights into the dynamic development of urban green spaces, facilitating the implementation of sustainable urban development strategies and practices. This review exploits the crucial role of computational methods in discovering and optimizing materials for redox flow batteries (RFBs). Integration of high‐throughput computational screening (HTCS) and machine learning (ML) accelerates materials discovery, guided by algorithms categorizing RFBs. A collaborative exploration, spanning macroscopic to mesoscopic scales, combines quantum machine learning with reinforcement learning, transfer learning, time series analysis, Bayesian optimization, active learning and various generative models. The collaborative integration of ML with computational techniques and experimental methods, anchored in experimentally validated Density Functional Theory (DFT) calculations and molecular dynamics (MD) simulations, proves indispensable for cost‐effective RFBs. Data collection and feature engineering are explored, emphasizing the integration of optimization goals and precise data collection within the ML framework. Feature analysis importance is highlighted, utilizing methods such as the filter, embedded, wrapper and deep learning methods for efficient energy materials exploration. Computational perspectives on materials features and operating conditions encompass membrane characteristics, fluid dynamics, temperature dependence and pressure sensitivity. Purpose: To develop a deep learning (DL) model for differentiating between benign and malignant ovarian tumors of Ovarian-Adnexal Reporting and Data System Ultrasound (O-RADS US) Category 4 lesions, and validate its diagnostic performance. Methods: A retrospective analysis of 1619 US images obtained from three centers from December 2014 to March 2023. DeepLabV3 and YOLOv8 were jointly used to segment, classify, and detect ovarian tumors. Precision and recall and area under the receiver operating characteristic curve (AUC) were employed to assess the model performance. Results: A total of 519 patients (including 269 benign and 250 malignant masses) were enrolled in the study. The number of women included in the training, validation, and test cohorts was 426, 46, and 47, respectively. The detection models exhibited an average precision of 98.68% (95% CI: 0.95–0.99) for benign masses and 96.23% (95% CI: 0.92–0.98) for malignant masses. Moreover, in the training set, the AUC was 0.96 (95% CI: 0.94–0.97), whereas in the validation set, the AUC was 0.93(95% CI: 0.89–0.94) and 0.95 (95% CI: 0.91–0.96) in the test set. The sensitivity, specificity, accuracy, positive predictive value, and negative predictive values for the training set were 0.943,0.957,0.951,0.966, and 0.936, respectively, whereas those for the validation set were 0.905,0.935, 0.935,0.919, and 0.931, respectively. In addition, the sensitivity, specificity, accuracy, positive predictive value, and negative predictive value for the test set were 0.925, 0.955, 0.941, 0.956, and 0.927, respectively. Background: Training parents to implement language and communication intervention strategies is an effective approach to promote language development for children with language delay. Aims: This study introduces an online parent training program conducted in Hubei province, China, which was designed to help parents of language‐delayed children with a diagnosis of autism spectrum disorder (ASD), developmental language disorder (DLD) or global developmental delay (GDD) apply language intervention strategies into daily interactions and promote their children's language development at home. Methods & Procedures: The Bethel Hearing and Speaking Training Center Family Training for Early Communication & Language Development (Bethel Family Training Program, BFT) (Bethel HSTC, 2020) was designed to improve the language and communication skills for children with language delay in a naturalistic way. The caregivers (including parents, grandparents and other main caregivers) participated in an 8‐h online program, including lectures on milestones in child language development, common misunderstandings of child language development, and three basic family language intervention strategies ('Looking together, playing together, and talking together') incorporating active learning through video analysis and discussion. Tongji Hospital in Hubei then continued with 3 months of online home intervention monitoring to all the caregivers via weekly online Q&As led by BFT certified speech therapists' team. The Gesell Developmental Schedules (GDS) was carried out before the online parent training program and after the 3‐month online home intervention monitoring. Outcomes & Results: 146 families whose children aged 12–68 months with language delay participated in the online training program. The results of the GDS assessments conducted before and after the program showed that not only did the developmental quotient (DQ) of language improve, but so did the DQ of social behaviour and adaptive behaviour (p < 0.001). There is no between‐group difference in the application of three strategies between the ASD group and the DLD or GDD group (p > 0.05). Furthermore, both caregivers' ability to apply 'looking together, playing together, talking together' strategies and the effective interaction time played important roles in improving the child's language abilities. Conclusion & Implications: The online parent training focusing on improving daily interaction with children through speech–language stimulation strategies promoted the development of language skills. It is an economic and practical approach for children with language delay who have limited access to local language intervention programs. Non-small cell lung cancer (NSCLC) is an aggressive and rapidly expanding lung cancer. Abnormal upregulation or knockdown of PDIA6 expression can predict poor prognosis in various cancers. This study aimed to investigate the biological function of PDIA6 in NSCLC. SOX2 and PDIA6 expression in NSCLC tissues and regulatory relationship between them were analyzed using bioinformatics. GSEA was performed on the enrichment pathway of PDIA6. qRT-PCR was utilized to examine expression of SOX2 and PDIA6 in NSCLC tissues and cells, and dual-luciferase reporter assay and ChIP experiments were performed to validate their regulatory relationship. CCK-8 experiment was conducted to assess cell viability, western blot was to examine levels of stem cell markers and proteins related to aerobic glycolysis pathway in cells. Cell sphere formation assay was used to evaluate efficiency of cell sphere formation. Reagent kits were used to measure glycolysis levels and glycolysis products. High expression of PDIA6 in NSCLC was linked to aerobic glycolysis. Knockdown of PDIA6 reduced cell viability, expression of stem cell surface markers, and cell sphere formation efficiency in NSCLC. Overexpression of PDIA6 could enhance cell viability and promote aerobic glycolysis, but the addition of 2-DG could reverse this result. Bioinformatics predicted the existence of upstream transcription factor SOX2 for PDIA6, and SOX2 was significantly upregulated in NSCLC, and they had a binding relationship. Further experiments revealed that PDIA6 overexpression restored repressive effect of knocking down SOX2 on aerobic glycolysis and cell stemness. This work revealed that the SOX2/PDIA6 axis mediated aerobic glycolysis to promote NSCLC cell stemness, providing new therapeutic strategies for NSCLC. [ABSTRACT FROM AUTHOR]

Objectives: To compare the on‐site diagnostic performance of contrast‐enhanced ultrasound (CEUS), computed tomography (CECT), and magnetic resonance imaging (CEMRI) for hepatocellular carcinoma (HCC) across diverse practice settings. Methods: Between May 2019 and April 2022, a total of 2085 patients with 2320 pathologically confirmed focal liver lesions (FLLs) were enrolled. Imaging reports were compared with results from pathology analysis. Diagnostic performance was analyzed in defined size, high‐risk factors for HCC, and hospital volume categories. Results: Three images achieved similar diagnostic performance in classifying HCC from 16 types of FLLs, including HCC ≤2.0 cm. For HCC diagnosis at low‐volume hospitals and HCC with high‐risk factors, the accuracy and specificity of CEUS were comparable to CECT and CEMRI, while the sensitivity of CEUS (77.4 and 89.5%, respectively) was inferior to CEMRI (87.0 and 92.8%, respectively). The diagnostic accuracy of CEUS + CEMRI and CEUS + CECT increased by 7.8 and 6.2% for HCC ≤2.0 cm, 8.0 and 5.0% for HCC with high‐risk factors, and 7.4 and 5.5% for HCC at low‐volume hospitals, respectively, compared with CEMRI/CECT alone. Conclusions: Compared with CECT and CEMRI, CEUS provides adequate diagnostic performance in clinical first‐line applications at high‐volume hospitals. Background: Cell-based immunotherapy shows the therapeutic potential in sarcomas, in addition to angiogenesis-targeted tyrosine kinase inhibitor (TKI) and immune checkpoint inhibitor (ICI). Multi-antigen stimulated cell therapy-I (MASCT-I) technology is a sequential immune cell therapy for cancer, which composes of multiple antigen-loaded dendritic cell (DC) vaccines followed by the adoptive transfer of anti-tumor effector T-cells. Methods: In this phase 1 study, we assessed MASCT-I plus camrelizumab (an ICI against PD-1) and apatinib (a highly selective TKI targeting VEGFR2) in patients with unresectable recurrent or metastatic bone and soft-tissue sarcoma after at least one line of prior systemic therapy. One MASCT-I course consisted of 3 DC subcutaneous injections, followed by 3 active T cell infusions administered 18–27 days after each DC injection. In schedule-I group, 3 DC injections were administered with a 28-day interval in all courses; in schedule-II group, 3 DC injections were administered with a 7-day interval in the first course and with a 28-day interval thereafter. All patients received intravenous camrelizumab 200 mg every 3 weeks and oral apatinib 250 mg daily. Results: From October 30, 2019, to August 12, 2021, 19 patients were enrolled and randomly assigned to schedule-I group (n = 9) and schedule-II group (n = 10). Of the 19 patients, 11 (57.9%) experienced grade 3 or 4 treatment-related adverse events. No treatment-related deaths occurred. Patients in schedule-II group showed similar objective response rate (ORR) with those in schedule-I group (30.0% versus 33.3%) but had higher disease control rate (DCR; 90.0% versus 44.4%) and longer median progression-free survival (PFS; 7.7 versus 4.0 months). For the 13 patients with soft-tissue sarcomas, the ORR was 30.8%, DCR was 76.9%, and median PFS was 12.9 months; for the 6 patients with osteosarcomas, the ORR was 33.3%, the DCR was 50.0%, and median PFS was 5.7 months. Conclusions: Overall, MASCT-I plus camrelizumab and apatinib was safe and showed encouraging efficacy in advanced bone and soft-tissue sarcoma, and schedule-II administration method was recommended. Objectives: Abdominal palpation is an essential examination to diagnose various digestive system diseases. This study aimed to develop an objective and standardized test based on abdominal palpation simulators, and establish a credible pass/fail standard of basic competency. Methods: Two tests were designed using the newly developed Jucheng abdominal palpation simulator (test 1) and the AbSim simulator (test 2), respectively. Validity evidence for both tests was gathered according to Messick's contemporary framework by using experts to define test content and then administering the tests in a highly standardized way to participants of different experience. Different simulator setups modified by the built-in software were selected from hepatomegaly, splenomegaly, positive McBurney's sign plus rebound tenderness, gallbladder tenderness (Murphy's sign), pancreas tenderness, and a normal setup without pathologies, with six sets used in test 1 and five sets used in test 2. Different novices and experienced were included in the tests, and test 1 was also administered to an intermediate group. Scores and test time were collected and analyzed statistically. Results: The internal consistency reliability of test 1 and test 2 showed low Cronbach's alphas of 0.35 and -0.41, respectively. Cronbach's alpha for palpation time across cases were 0.65 for test 1 and 0.76 for test 2. There was no statistical difference in total time spent and total scores among the three groups in test 1 (P-values (ANOVA) were 0.53 and 0.35 respectively), nor between novices and experienced groups in test 2 (P-values (t-test) were 0.13 and 1.0 respectively). It was not relevant to try to establish pass/fail standards due to the low reliability and lack of discriminatory ability of the tests. Conclusions: It was not possible to measure abdominal palpation skills in a valid way using either of the two standardized, simulation-based tests in our study. Assessment of the patient's abdomen using palpation is a challenging clinical skill that is difficult to simulate as it highly relies on tactile sensations and adequate responsiveness from the patients. Background: We aimed to investigate the value of a machine learning (ML) algorithm in the preoperative prediction of lymph node metastasis in patients with rectal cancer. Methods: Based on the histopathological results, 126 rectal cancer patients were divided into two groups: lymph node metastasis‐positive and metastasis‐negative groups. We collected clinical and laboratory data, three‐dimensional endorectal ultrasound (3D‐ERUS) findings, and parameters of the tumor for between‐group comparisons. We constructed a clinical prediction model based on the ML algorithm, which demonstrated the best diagnostic performance. Finally, we analyzed the diagnostic results and processes of the ML model. Results: Between the two groups, there were significant differences in serum carcinoembryonic antigen (CEA) levels, tumor length, tumor breadth, circumferential extent of the tumor, resistance index (RI), and ultrasound T‐stage (P < 0.05). The extreme gradient boosting (XGBoost) model had the best comprehensive diagnostic performance for predicting lymph node metastasis in patients with rectal cancer. Compared with experienced radiologists, the XGBoost model showed significantly higher diagnostic value in predicting lymph node metastasis; the area under curve (AUC) value of the receiver operating characteristic (ROC) curve of the XGBoost model and experienced radiologists was 0.82 and 0.60, respectively. Conclusions: Preoperative predictive utility in lymph node metastasis was demonstrated by the XGBoost model based on the 3D‐ERUS finding and related clinical information. Metastases to the thyroid gland (MTT) are uncommon in clinical practice. The ultrasound (US) features are easily confused with primary thyroid malignancy, Hashimoto's thyroiditis, and other thyroid diseases. Therefore, this study aimed to assess the role of US and analysis of prognosis of MTT. A total of 45 patients with MTT in the database between July 2009 and February 2022 at the Fujian Cancer Hospital were reviewed. US examinations were performed only on 20 patients, who were finally included in our study. Among the 20 patients, nine were male, and eleven were female. According to US characteristics, metastases to the thyroid gland were divided into nodular and diffuse types (17 and 3 cases, respectively). Three lesions (17.6%) had circumscribed margins, and 14 (82.4%) were uncircumscribed. Three lesions (17.6%) were regular in shape, and 14 (82.4%) were irregular. Nine metastases (52.9%) were a taller-than-wide shape, and eight (47.1%) were not a taller-than-wide shape. Ten lesions (58.8%) had rich vascularity, and seven (41.2%) had absence/not rich vascularity. The mean overall survival (OS) from the time of MTT diagnosis was 22 months (95% confidence interval: 5.95–38.05). The 1-, 3-, and 5-year OS after metastasis was 68.1%, 25.5%, and 17%, respectively. The prognosis of MTT was poor, which is closely related to the characteristics of the primary tumor and metastatic disease. In order to study the seismic behavior of composite fired wall boards reinforced concrete frame, the low cycle repeated loading test was carried out for the composite fired wall boards-RC frame structure. Experiment phenomena, load-displacement curves, stiffness degradation curves and displacement ductility factors were given. The results showed when the fired wall boards is flexibly connected with the reinforced concrete frame, it can effectively avoid the destruction of the wall board at the initial stage of loading. With the increase of the deformation of the frame, the wall board acts as a slant support to the frame, which helps to improve the overall seismic performance of the frame, and makes the composite fired wall board reinforced concrete frame structure show good energy dissipation capacity and ductility as a whole. Introduction: The present study aimed to evaluate the influence of biological characteristics of hepatocellular carcinoma (HCC) on the Liver Imaging Reporting and Data System (LI-RADS) v2017 category of contrast-enhanced ultrasound (CEUS) in patients with high risk and compare the outcomes among different categories after radical resection. Methods: Between June 2017 and December 2020, standardized CEUS data of liver nodules were prospectively collected from multiple centers across China. We conducted a retrospective analysis of the prospectively collected data on HCCs measuring no more than 5 cm, as diagnosed by pathology. LI-RADS categories were assigned after thorough evaluation of CEUS features. Then, CEUS LI-RADS categories and major features were compared in different differentiation, Ki-67, and microvascular invasion (MVI) statuses. Differences in recurrence-free survival (RFS) among different LI-RADS categories were further analyzed. Results: A total of 293 HCC nodules in 293 patients were included. This study revealed significant differences in the CEUS LI-RADS category of HCCs among differentiation (p < 0.001) and levels of Ki-67 (p = 0.01) and that poor differentiation (32.7% in LR-M, 12% in LR-5, and 6.2% in LR-4) (p < 0.001) and high level of Ki-67 (median value 30%) were more frequently classified into the LR-M category, whereas well differentiation (37.5% in LR-4, 15.1% in LR-5, and 11.5% in LR-M) and low levels of Ki-67 (median value 11%) were more frequently classified into the LR-4 category. No significant differences were found between MVI and CEUS LI-RADS categories (p > 0.05). With a median follow-up of 23 months, HCCs assigned to different CEUS LI-RADS classes showed no significant differences in RFS after resection. Conclusions: Biological characteristics of HCC, including differentiation and level of Ki-67 expression, could influence major features of CEUS and impact the CEUS LI-RADS category. Zinc-based hybrid-flow batteries are considered as a promising alternative to conventional electrochemical energy-storage systems for medium- to large-scale applications due to their high energy densities, safety, and abundance. However, the performance of these batteries has been limited by issues such as dendritic growth and passivation of zinc anodes during charge–discharge cycling. To address this challenge, a variety of two- and three-dimensional zinc anodes have been investigated. While two-dimensional zinc anodes have been extensively studied, there has been limited investigation into three-dimensional zinc anodes for hybrid-flow batteries. This study highlights the potential of three-dimensional zinc anodes to mitigate overpotentials and improve the mass transport of active species to promote negative electrode reactions. The performance of a membraneless flow battery based on low-cost zinc and organic quinone was herein evaluated using experimental and numerical approaches. Specifically, the use of zinc fiber was shown to yield an average coulombic efficiency of approximately 90% and an average voltage efficiency of approximately 82% over the course of 100 cycles at a current density of 30 mA cm−2. In the field of automatic speech recognition (ASR), the noisy audio data and the ambiguity in recognizing homophone lead to the degradation of model performance. In order to address the mentioned problems, a network called DMRS-transformer, a Transformer-based network, is proposed in this study. The proposed DMRS-Transformer mainly consists of two components except for the traditional Transformer network, which are denoising module and Mandarin recognition supplementary module respectively. The denoising module is used for pruning the trivial features caused by the noisy input audio data. The Mandarin recognition supplementary module, short for MRS module, tends to tackle the problem of recognizing Mandarin speech signals which have several homophones. Empirical evaluations have been conducted on two widely used datasets, which are Aishell-1 and HKUST respectively. The experimental results can validate the effectiveness of the proposed DMRS-Transformer network. Compared with the Transformer baseline, the proposed DMRS-Transformer has 0.8% CER improvement and 1.5% CER improvement in these two datasets respectively. [ABSTRACT FROM AUTHOR]

The land surface temperature (LST) is an important indicator reflecting the ecological environment condition. As a sensitive area to climate change, mastering the spatial and temporal changes of summer LST in the Bosten Lake basin (BLB) helps gain insight into the evolution of the thermal environment in the Bosten Lake basin and for long-term monitoring of the basic ecological changes in the basin. Based on MOD11A1 data from 2005 to 2020, this paper investigates the diurnal LST spatiotemporal series variation and its influencing factors in the Bosten Lake basin by using surface temperature class classification, trending analysis, the Hurst index, and geographic probes. The results show that (1) the wetland grasslands in and around the Bayinbruck steppe in the northwestern part of the study area exhibit a heat island effect during the day, while the opposite is true at night. In terms of temporal changes, LST changes in the BLB fluctuate widely, having a general rising and then decreasing trend. (2) The decreasing trend of LST from 2005 to 2020 is significant during the daytime and vice versa at night, and the change at night is greater than during the day. The areas with significantly higher diurnal LST in the future have all expanded compared to the area occupied by them now, with an overall trend of a steady increase. (3) The dominant factor of LST variation has the strongest explanatory power when altitude and NDVI are combined during the daytime and the strongest explanatory power when NPP and temperature are combined at night. Accurately mapping and monitoring the urban impervious surface area (ISA) is crucial for understanding the impact of urbanization on heat islands and sustainable development. However, less is known about ISA spectra heterogeneity and their similarity to bare land, wetland, and high-rise-building shadows. This study proposes a feature-based approach using decision tree classification (FDTC) to map ISAs and their spatio-temporal changes in a coastal city in southeast China using Landsat 5 TM, Landsat 8 OLI/TIRS, and Sentinel-2 images from 2009 to 2021. Atmospheric correction using simplified dark object subtraction (DOS) was applied to Landsat imagery, which enabled faster computation. FDTC's performance was evaluated with three sensors with different spectral and spatial resolutions, with parameter thresholds held constant across remote-sensing images. FDTC produces a high average overall accuracy (OA) of 94.53%, a kappa coefficient (KC) of 0.855, and a map-level image classification efficacy (MICE) of 0.851 for ISA mapping over the studied period. In comparison with other indices such as BCI (biophysical composition index), PISI (automated built-up extraction index), and ABEI (perpendicular impervious surface index), the FDTC demonstrated higher accuracy and separability for extracting ISA and bare land as well as wetland and high-rise buildings. The results of FDTC were also consistent with those of two open-source ISA products and other remote sensing indices. The study found that the ISA in Xiamen City increased from 16.33% to 26.17% over the past 13 years due to vegetation occupation, encroachment onto bare land, and reclamation of coastal areas. While the expansion significantly reduced urban vegetation in rapidly urbanizing areas of Xiamen, ambitious park greening programs and massive redevelopment of urban villages resulted in a modest but continuous increase in urban green space. Accurately estimating land-use demand is essential for urban models to predict the evolution of urban spatial morphology. Due to the uncertainties inherent in socioeconomic development, the accurate forecasting of urban land-use demand remains a daunting challenge. The present study proposes a modeling framework to determine the scaling relationship between the population and urban area and simulates the spatiotemporal dynamics of land use and land cover (LULC). An allometric scaling (AS) law and a Markov (MK) chain are used to predict variations in LULC. Random forest (RF) and cellular automata (CA) serve to calibrate the transition rules of change in LULC and realize its micro-spatial allocation (MKCARF-AS). Furthermore, this research uses several shared socioeconomic pathways (SSPs) as scenario storylines. The MKCARF-AS model is used to predict changes in LULC under various SSP scenarios in Jinjiang City, China, from 2020 to 2065. The results show that the figure of merit (FoM) and the urban FoM of the MKCARF-AS model improve by 3.72% and 4.06%, respectively, compared with the MKCAANN model during the 2005–2010 simulation period. For a 6.28% discrepancy between the predicted urban land-use demand and the actual urban land-use demand over the period 2005–2010, the urban FoM degrades by 21.42%. The growth of the permanent urban population and urban area in Jinjiang City follows an allometric scaling law with an exponent of 0.933 for the period 2005–2020, and the relative residual and R2 are 0.0076 and 0.9994, respectively. From 2020 to 2065, the urban land demand estimated by the Markov model is 19.4% greater than the urban area predicted under scenario SSP5. At the township scale, the different SSP scenarios produce significantly different spatial distributions of urban expansion rates. Thermal comfort in outdoor spaces is of vital importance to human health and well-being, and affects the livability of cities. In a humid-hot region with elevated temperatures, it is difficult to improve the thermal experience of urban residents by changing objective conditions such as the microclimate. This study created a set of experience indices in different dimensions including visual, auditory, and olfactory perceptions and psychological cognitions, to explore the influencing factors and mechanisms of visitor's thermal comfort in an urban park. We found that visual, auditory, and olfactory experience indices and psychological cognition indices were both strongly correlated with thermal comfort (Pearson correlation coefficients were above 0.3). By analyzing a survey data with a structural equation model, we found that Visual, auditory, and olfactory experiences have a complete, mediating effect on thermal comfort. Explicitly, visual, auditory, and olfactory perceptions affect thermal comfort through two mediating variables: thermal sensation and psychological cognitions. In turn, thermal sensation and psychological cognitions have a direct effect on visitor's thermal comfort. Psychological cognitions has the largest direct effect on thermal comfort. Quantitatively, a 1-unit increase in psychological cognitions produces an increase of 0.601 units in thermal comfort. A potential approach is to improve multisensory modalities that affect necessary sensory perception and psychological cognitions of visitors. OBJECTIVES: To present a method combining ultrasound (US) and contrast-enhanced ultrasound (CEUS) features for differential diagnosis of cervical tuberculous lymphadenitis (CTL) and primary lymphoma. METHODS: A total of 155 patients with CTL (n = 49) and lymphoma (n = 106) who underwent US and CEUS were retrospectively included. The features extracted from US and CEUS and the significant clinical data were created three models using the least absolute shrinkage and selection operator and logistic regression analysis. The diagnostic performance of the models was assessed using the area under the curve (AUC). RESULTS: The combined model outperformed US model and CEUS model in distinguish CTL from lymphoma achieved favorable performances in training set and validation set with AUCs of 0.958 and 0.946 as well as high accuracies (91.7% and 87.2%), sensitivities (95.9% and 84.4%) and specificities (82.4% and 93.3%). Delong's test showed that among the three models, combined model was significantly different from the other two models in training set (p = 0.011 and 0.029, respectively) and validation set (p = 0.018 and 0.001, respectively). CONCLUSIONS: A combination of US and CEUS achieved good diagnostic performance in differentiating lymphoma and CTL, which might aid in clinical decision-making. [ABSTRACT FROM AUTHOR]

Background: Colorectal cancer (CRC) is a prevalent malignancy of the gastrointestinal. Circular RNAs (circRNAs) act as important roles in CRC malignant progression. However, the role of circ_0039857 in CRC is still unclear. Therefore, this study aimed to explore the function and mechanism of hsa_circ_0039857 in the CRC. Methods: The mRNA and protein expression were measured via RT-qPCR. RNase R assay and Actinomycin D were employed to evaluate the stability of circ_0039857. Functional experiments, such as proliferation and apoptosis, were applied to study the function of circ_0039857 in CRC cells. The underlying mechanisms of circ_0039857 were then analyzed by bioinformatics, dual-luciferase reporter gene assay, RNA pull-down and rescue experiments. Results: We revealed that circ_0039857 was significantly enhanced in CRC. Circ_0039857 was stabler than linear RNA in cells and valuable for the disease diagnosis. In addition, circ_0039857 knockdown inhibited proliferation and promoted apoptosis. Mechanistically, circ_0039857 positively regulated the expression of RAB32 via sponging miR-338-3p. Conclusion: This study demonstrated that circ_0039857 knockdown suppressed CRC malignant progression through miR-338-3p/RAB32 axis. To investigate the impact of marriage on multiple myeloma (MM) survival, and the role of factors such as age, sex, and income in this relationship. The Surveillance, Epidemiology, and End Results (SEER) database was searched for eligible MM patients between 2007 and 2016. We compared overall survival (OS) and cancer-specific survival (CSS) differences by the Kaplan–Meier method and log-rank tests. The hazard ratios (HRs) with 95% confidence intervals (CIs) for OS and CSS were estimated by Cox regression models. A 1:1 propensity score matching (PSM) analysis was used to minimize the covariates differences between married and unmarried group. This study identified 48,952 eligible patients diagnosed with MM, comprising 29,607 married patients, 5,147 divorced/separated patients, 6,851 single patients, and 7,347 widowed patients. Married MM patients were found to be an independent protective prognostic factor for OS and CSS (all P < 0.001). The survival advantage of marriage still remained in 1:1 matched-pair analysis. The stratified analysis further revealed that the OS and CSS was poorer in unmarried than married MM patients across different age of diagnosis, sex, income, race, and period of diagnosis subgroups (all P < 0.05). The beneficial effect of marriage on the survival of MM was particularly prominent in younger (<65 years) patients, males, higher income patients, non-Hispanic whites, and more recent years. Married patients with MM tend to have better OS and CSS likely due to their higher income, education level, insurance and receipt of chemotherapy. [ABSTRACT FROM AUTHOR]

Purpose: Malignant pleural effusion (MPE) is an adverse prognostic factor in patients with osteoblastic osteosarcoma; however, the cellular contexts of MPE are largely unknown. Experimental design: We performed single‐cell RNA‐sequencing (scRNA‐seq) on 27 260 cells from seven MPE samples and 91 186 cells from eight osteosarcoma tissues, including one recurrent, one lung metastasis and six primary tumour (PT) samples, to characterize their tumour microenvironment. Results: Thirteen main cell groups were identified in osteosarcoma tumour and MPE samples. Immune cells dominate the cellular contexts in MPE with more T/NK cells and less osteoclasts compared to PT samples. Of T/NK cells, CD8+GNLY+, CD8+KLRC2+ T cells and FCGR3A+NK cells were enriched in MPE but CD4+FOXP3+ Tregs were enriched in PT samples. Naïve IGHD+ B and immune regulatory IGHA1+ B cells were largely identified in MPE, whereas bone metabolism‐related CLEC11A+ B cells were significantly enriched in osteosarcoma PT. M2‐type TAMs, including CLEC11A_TAM, C1QC_TAM and Prolif_TAMs, among myeloid cells were enriched in PT, which may suppress cytotoxicity activities of T cells through multiple ligand–receptor interactions. Mature LAMP3+ DCs were transformed from CD1C+ DC and CLEC9A+ DC sub‐clusters when exposure to tumour alloantigens, which may improve T cell cytotoxicity activities on tumour cells under anti‐PD‐L1 treatments. In further, immune cells from MPE usually present up‐regulated glycolysis and down‐regulated oxidative phosphorylation and riboflavin metabolism activities compared to those in PT samples. Objective: To evaluate the diagnostic performance of contrast‐enhanced ultrasound (CEUS) combined with thyroglobulin (Tg) levels in fine‐needle aspirates (FNA) washout fluid (FNA‐Tg) in diagnosing cervical lymph node (LN) metastasis in papillary thyroid cancer (PTC) patients. Methods: Data from 190 LNs in 167 patients suspected of metastasis from the US between November 2018 and September 2020 were included. All subjects underwent FNA, CEUS, and FNA‐Tg examinations. The final outcomes were confirmed by histopathological or cytological examination or follow‐up imaging. Data were analyzed using the Wilcoxon rank‐sum or chi‐squared test. The diagnostic efficacy of FNA, CEUS, and FNA‐Tg in diagnosing LNs was compared. Results: A cutoff value of 6.15 ng/ml (AUC 0.925, 95% confidence interval (CI) 0.885–0.966) successfully identified metastatic LNs. FNA missed 58 LN metastases, of these, 94.8% (55/58) were correctly diagnosed using the combination of CEUS and FNA‐Tg. FNA‐Tg showed higher sensitivity (90.2%), NPV (86.1%) and accuracy (88.9%) than either FNA (48.2, 57.4 and 69.5%, respectively) or CEUS (82.1, 67.7 and 70.5%, respectively) alone. The combination of CEUS, FNA and FNA‐Tg resulted in maximal sensitivity (100%) and NPV (100%) but reduced specificity (51.3%) and overall diagnostic accuracy (80.0%). After adding FNA‐Tg to discordant samples between CEUS and FNA, 81.9% of LNs (77/94) were correctly diagnosed. Purpose: The options for the second-line treatment of metastatic osteosarcoma are still limited. Anlotinib is a multi-kinase inhibitor which has shown promising efficacy and good tolerability in various cancer types. This retrospective study was conducted to evaluate the efficacy and safety of anlotinib combined with gemcitabine/docetaxel (GD) in patients with metastatic osteosarcoma who have failed first-line chemotherapy. Patients and Methods: The data of patients who received anlotinib combined with GD or GD were collected. The primary endpoint was progression-free survival. Secondary endpoints included objective response rate and safety. Results: From July 2013 to November 2020, a total of 32 patients were enrolled, 13 received anlotinib combined with GD and 19 received GD. Median PFS was 9.0 months (95% CI 6.7– 39.1) in the combination group and 5.0 months (95% CI 1.2– 6.7) in the chemotherapy group. ORR were 38.4% and 15.8%, DCR were 69.2% and 38.1% in the combination and chemotherapy group, respectively. The most common adverse events included fatigue (78.9% in the combination group vs 69.2% in the chemotherapy group), hypertension (46.2% vs 10.5%), diarrhea (38.5% vs 21.1%), hypothyroidism (38.5% vs 15.8%), neutropenia (23.1% vs 36.8%) and AST elevation (30.8% vs 21.1%). The most common grade 3 or worse adverse events included hand-foot reaction (7.7% vs 5.3%), hypothyroidism (15.4% vs 0), neutropenia (0 vs 10.5%). Conclusion: The combination of anlotinib and GD showed favorable efficacy with manageable toxicities compared with GD in the second-line treatment for metastatic osteosarcoma. A variety of factors may affect the planning and design of urban infrastructure but urban population usually is the primary consideration given scaling laws between population and infrastructure. Although urban physical spatial form and infrastructure are physically attached, their allometric scaling has not been systematically addressed. In this study, we examine how five types of infrastructure, including roads, water supply pipelines, drainage pipelines, buses, and schools, affect urban spatial form characterized with landscape metrics derived for 78 large cities in China. The results suggest that urban infrastructure has scaling relations with landscape shape index (LSI), area-weighted mean perimeter-area ratio (PARA_AM), and largest patch index. Specifically, at the mean value of PARA_AM, road area increases about 2.5 times when doubling LSI. Under the same population size, cities with more fragmented spatial form need more investments in infrastructure, particularly roads and other traffic components. It is concluded that urban infrastructure experiences a super-linear quantitative relationship with landscape metrics, and a city with a more complex urban form spatially requires more infrastructure. The coefficients of the regression models show a lot of variations but the allometric scaling is meaningful for the planning and design of urban infrastructure. The single‐phase oxides with elemental complexity and compositional diversity, usually named high entropy oxides, feature homogeneously dispersed multi‐metallic elements in equiatomic concentration. The unusual properties of high entropy oxides endow their potential application in clean‐energy‐related electrocatalysis. However, the possible fundamental relationship between configuration entropy and the underlying catalytic mechanism is still not well understood and established. Herein, a high entropy perovskite cobaltate consisting of five equimolar metals in the B‐site (Mg, Mn, Fe, Co, and Ni) is employed as an electrocatalyst for oxygen evolution reaction (OER). The configuration entropy serves as an effective tool to promote the intrinsic activity of the Co reactive site and manipulate the OER mechanism. The high entropy cobaltate demonstrates a lower overpotential of 320 mV at a current density of 10 mA cm−2, outperforming other counterparts. The X‐ray spectroscopies disclose the synergistic charge‐exchange effect among different cations and the formation of a new oxygen hole state. Combinatorially computational and experimental results unveil the enigma that the high configuration entropy leads to the random occupation of cations, facilitates the surface reconstruction, and benefits the formation of stable surface oxygen vacancies. Purpose: Emerging evidence shows that immune checkpoint inhibitors lead to durable responses in a variety of cancers, including nasopharyngeal carcinoma (NPC), however, combination approaches (i.e., stereotactic body radiation therapy, SBRT) are required to extend this benefit beyond a subset of patients. This study retrospectively evaluated eight recurrent/metastatic NPC patients, to investigate how radiation could potentiate PD-1 checkpoint inhibition therapy.Methods: Between September 2016 and July 2017, eight consecutive cases with histologically confirmed PDL1-positive status, for which prior standard therapy had been ineffective (five patients), were treated at our institution and Macao Clinics and two patients had disease progression within 6 months of completion of definitive chemoradiation, or one patient refused to receive chemoradiotherapy. All received PD-1 inhibitors first, seven of them accepted SBRT with an unmodified PD-1 inhibitors regimen after first evaluation as they were unresponsive to PD-1 inhibitors alone. Treatment was discontinued as long as patients were experiencing a clinical benefit in the opinion of the physicians and at least five cycles were given before stoppage.Results: Median follow-up time was 56.7 months. The confirmed objective response rate based on RECIST-v1.1 at first evaluation was 12.5% (1/8). For the seven cases who received SBRT, six of them experience an objective response (6/7, 85.7%) after SBRT. Only one patient showed rapid progress and die within 95 days after the initiation of SBRT intervention. Three patients who did not have all lesions exposed to irradiation were available to evaluate the incidence of an abscopal effect, however, it did not occur as expected. Median PFS and OS for the seven patients were 8.0 and 30.8 months after SBRT intervention, respectively. Two-year OS as indicated was 71.0%.Conclusions: PD-1 inhibitors combined with SBRT demonstrated promising antitumor activity in patients with PD-L1 positive RM-NPC. A model of environmental evolution of cities, especially in East Asia, is said to follow four stages. The pathways to the fourth stage of eco-city are not always clear, but need to be contextual. This research extended the original stage model of environmental evolution to describe urban development and its impact on the environment, especially integrated a model comprise of density, mix and accessibility (DMA) for urbanization and applied Ecological Footprint (EF) concept for pressure on eco-environment. The research analyzed the Xiamen City in China through the first three stages in order to gain insights how the city might be able to hasten its transition to the fourth stage of an eco-city. As expected, the development of Xiamen City and its environmental impacts has very obvious stage characteristic. Before 2010, Xiamen City got through a long time of Stage I (poverty stage); and in the period of 2010 to 2019, Xiamen experienced the Stage II (industrial pollution stage) and Stage III (mass consumption stage) crossly. The new built-up land in the decade of 2000-2010 is much larger than the previous 10 years from 1989 to 2000, 5 times more. Meanwhile, compared with 1980-1995, the ecological deficit also enlarged greatly in period of 1995-2010. Considering up to 2019 Xiamen still has large amount of energy consumption and high level output of industry solid waste, Xiamen should take measures of adjusting the industrial structure, promoting the green industry and planning the urban growth boundary to achieve the eco-city stage. Objective: Anlotinib, a novel multitarget kinase inhibitor of VEGFR, FGFR, PDGFR and c-Kit, has proven to be effective and safe for refractory soft tissue sarcoma patients, but has not been examined in recurrent or metastatic primary malignant bone tumors in a clinical trial setting. Methods: This is a multicenter single-arm trial. Patients with pathologically proven recurrent or metastatic primary malignant bone tumors were eligible. Anlotinib was administered orally at 12 mg per day. Each cycle consisted of 2 weeks of treatment followed by 1-week off-treatment. The primary endpoint was progression-free survival (PFS), as assessed in the intention-to-treat (ITT) population. Secondary endpoints included objective response rate (ORR), disease control rate (DCR) and overall survival (OS). Adverse events (AEs) were assessed per NCI CTCAE version 4.03. Results: A total of 42 patients were enrolled. Median PFS was 5.3 months (95% CI 3.5-8.4 months) in the overall analysis, 4.8 months (95%CI 3.5-7.1 months) in osteosarcoma patients and 2.8 months [95%CI 1.3 months to not reached (NR)] in chondrosarcoma patients. The median OS was 11.4 months (95% CI 10.1 months to NR) in the overall analysis, not reached (95% CI, NR, NR) in osteosarcoma patients and 11.4 months (95% CI 1.8 to 21.1 months) in chondrosarcoma patients. The ORR was 9.52% and DCR was 78.57%. Grade 3 or above AEs occurred in 54.76% of the patients, and included hypertension (19.05%), hypertriglyceridemia (9.52%) and pustulosis palmaris et plantaris (7.14%). No treatment-related death was reported. Conclusion: Anlotinib demonstrated promising antitumor activities in recurrent or metastatic primary malignant bone tumors with manageable AEs. Objectives. Evidence suggests that about 80% of all salivary gland tumors involve the parotid glands, with approximately 20% of parotid gland tumors (PGTs) being malignant. Discriminating benign and malignant parotid gland lesions preoperatively is vital for selecting the appropriate treatment strategy. This study explored the diagnostic performance of deep learning system for discriminating benign and malignant PGTs in ultrasonography images and compared it with radiologists. Methods. A total of 251 consecutive patients with surgical resection and proven parotid gland malignant or benign tumors who underwent preoperative ultrasound examinations were enrolled in this study between January 2014 and November 2020. Next, we compared the diagnostic accuracy of deep learning methods (ViT-B\16, EfficientNetB3, DenseNet121, and ResNet50) and radiologists in parotid gland tumor. In addition, the area under the curve (AUC), specificity, sensitivity, positive predictive value, and negative predictive value were calculated. Results. Among the 251 patients, 176/251 were the training set, whereas 75/251 were the validation set. Results showed that 74/251 patients had malignant tumor. Deep learning models achieved good performance in differentiating benign from malignant tumors, with the diagnostic accuracy and AUCs of ViT-B\16, EfficientNetB3, DenseNet121, and ResNet50 model being 81% and 0.81, 80% and 0.82, 77% and 0.81, and 79% and 0.80, respectively. On the other hand, the diagnostic accuracy and AUCs of radiologists were 77%-81% and 0.68-0.75, respectively. It was evident that the diagnostic accuracy of deep learning methods was higher than that of inexperienced radiologists, but there was no significant difference between deep learning methods and experienced radiologists. Conclusions. This study shows that the deep learning system can be used for diagnosing parotid tumors. Purpose: Inflammatory response and nutritional status are associated with cancer development and progression. The present study aimed to evaluate the predictive ability of the fibrinogen–albumin ratio index (FARI) to the efficacy of neoadjuvant chemotherapy (NAC) for osteosarcoma. Patients and methods: A retrospective analysis involving 752 consecutive osteosarcoma patients between 2012 and 2020 was performed. Data on serum fibrinogen, albumin levels, white blood cell count, platelet count, and alkaline phosphatase (ALP) before and after NAC were collected. The predictive value of the NAC efficacy in osteosarcoma was assessed by constructing a receiver operating characteristic (ROC) curve and calculating the area under the curve (AUC). Prognosis and its predictive factors were analyzed by Kaplan–Meier method and COX regression analysis. Nomogram was established according to selected variables. The predictive performance of the nomogram model was assessed using C-statistics. Results: A total of 203 patients were included. ROC analysis showed that both FARI before NAC (preFARI; AUC = 0.594, p = 0.032) and the change in FARI before and after NAC (dfFARI = preFARI-postFARI; AUC = 0.652, p = 0.001) exhibited more favorable predictive ability than ALP and other inflammation markers. The preFARI was divided into the high group (> 6.1%) and the low group (≤ 6.1%) based on the optimal cut-off value of 6.1%. Patients with a high preFARI showed significantly decreased metastasis-free survival (MFS) and disease-free survival (DFS) (all p< 0.01). In multivariable analysis, preFARI was an independent prognostic marker for patients with osteosarcoma. Predictive nomograms exhibited good ability to predict MFS (C-index = 0.748, se = 0.028) and DFS (C-index=0.727, se = 0.030). Conclusion: Our findings indicated that FARI exhibits the favorable predictive ability for the efficacy of NAC for osteosarcoma, which could support clinicians and patients in clinical decision-making and treatment optimization. In the era of big data, the data is collected and applied in every aspect of life. Establishing a reasonable architectural heritage evaluation indicators system is the key of architectural heritage evaluation. According to the connotation of architectural heritage and the standard of eliminating duplicate information and the standard of the maximum weighted R cluster grade, this paper constructs an architectural heritage evaluation indicators system through quantitative approaches of R cluster and rough set analysis. The contribution lies in the following: Firstly, it uses the method of square-sum of dispersion to classify the evaluation indicator criterion of the same guideline, which ensures the different indicators reflect information that is different. Secondly, it uses the rough set analysis to solve the weighted rough set grade of each indicator of the similar, then screening the indicator that contain the maximum weighted rough set, in which ensuring the selected indicator is the most significant influence to the evaluation result. Thirdly, through quantitative approaches of R cluster and rough set analysis, establishing the architectural heritage evaluation indicators system composed of 18 indicators includes three guidelines: heritage historical value, heritage artistic value, and heritage scientific value. Steam activation treatments were introduced in the preparation of activated carbon fiber from liquefied wood (LWACF), to enlarge its specific surface area and develop the pore size distribution. With increasing activation time, the average fiber diameter of LWACF decreased from 27.2 µm to 13.2 µm, while the specific surface area increased from 1025 to 2478 m2/g. Steam activation predominantly enhanced the development of microporosity, without significant pore widening. Prolonging the steam activation time exponentially increased the removal efficiency of Cu2+ at a constant adsorbent dose, as a result of an increase in the number of micropores and acidic-oxygenated groups. Moreover, for LWACF activated for 220 min at 800 °C, the removal efficiency of Cu2+ increased from 55.2% to 99.4%, when the porous carbon fiber dose went from 0.1 to 0.5 g/L. The synthesized LWACF was proven to be a highly efficient adsorbent for the treatment of Cu2+ ion-contaminated wastewater. [ABSTRACT FROM AUTHOR]

Osteosarcoma (OS) is the most common primary bone sarcoma, chemoresistance becomes an obstacle to its treatment. Metabolic reprogramming is a hallmark of malignancy, targeting the metabolic pathways might provide a reasonable therapeutic strategy for OS. Here we demonstrated that Ailanthone (AIL), a major component of the Chinese medicine Ailanthus altissima, significantly suppressed OS cell growth in vitro and in vivo. Furthermore, AIL dose-dependently inhibited cell migration and invasion, induced cell cycle arrest and apoptosis in OS cells. Combined transcriptomics, proteomics and metabolomics analyses revealed that AIL induced widespread changes in metabolic programs in OS cells, while the serine biosynthetic pathway (SSP) was the most significantly altered pathway. qRT-PCR and Western blot assay confirmed that the transcript and protein levels of the SSP genes (PHGDH , PSAT1 and PSPH) were downregulated dose-dependently by AIL. In addition, we found out that many downstream pathways of the SSP including the one-carbon pool by folate, purine metabolism, pyrimidine metabolism, DNA replication and sphingolipid metabolism were downregulated after AIL treatment. In the revere test, PHGDH overexpression but not exogenous serine supplementation clearly attenuated the effects of AIL on OS cells. Taken together, AIL exerts antitumor effects on OS through mediating metabolic reprogramming, at least in part, by suppressing the SSP. In this study, the chemical constituents of jujube and jujube fermentation broth were compared by chromatography tandem high‐resolution mass spectrometry for the first time. A total of 68 compounds were detected, and six of them were quantified. The results showed that rutin was hydrolyzed and converted into quercetin during the fermentation process. Further, the antioxidant experiments in vitro were carried out, and the IC50 value of the fermentation broth was lower than that of the jujube extract. Twenty‐three potential bioactive components from jujube samples were identified by a target cell‐based screening method, and triterpenic acid compounds exhibited high cell binding property. These results will help to understand the different biological activities of jujube and its fermented products and will benefit the discovery of new functional components. [ABSTRACT FROM AUTHOR]

Purpose: The aim of this prospective study was to evaluate the value of the combination of contrast-enhanced ultrasonography (CEUS) and blue dye (BD) for SLN detection in patients with clinically negative node breast cancer. Methods: Patients with clinically negative node breast cancer were randomized into two cohorts for SLN biopsy (SLNB): the combination method cohort using CEUS and BD together, and the single BD method cohort. Standard axillary lymph node dissection was performed if any of the SLNs confirmed positive by pathology. The identification rate, the number of SLNs removed and recurrence-free survival (RFS) rates were evaluated between two cohorts. In addition, we assessed the sensitivity, specificity, accuracy, false-negative rate of CEUS for diagnosis of SLNs based on patterns of CEUS enhancement. Results: 144 consecutive patients with clinically negative node breast cancer were randomized into two cohorts. Each cohort consisted of 72 cases. In the combination method cohort, contrast-enhanced lymphatic vessels were clearly visualized and SLNs were accurately localized in 72 cases. The identification rate and the mean number of SLNs detected by the combination method were 100% (72/72) and 3.26 (1–9), respectively. In contrast, in the single BD method cohort, SLNs in 69 cases were successfully identified. The identification rate and the mean number of SLNs using BD alone were 95.8% (69/72) and 2.21 (1–4), respectively. According to patterns of CEUS enhancement, the sensitivity, specificity, accuracy, and the FNR of CEUS for SLN diagnosis were 69.2%, 96.6%, 91.7%, and 30.8%, respectively. After a median follow-up of 50 months for the combination method cohort and 51 months for the blue dye alone cohort, five patients in the combination method cohort and nine in the blue dye alone cohort had recurrence. RFS rates showed no significant difference (P = 0.26) between two cohorts. Conclusion: The combination of CEUS and BD is more effective than BD alone for SLNB in clinically negative node patients with an identification rate as high as 100%. Use of BD and CEUS in combination may provide the possibility of a non-radioactive alternative method for SLNB in centers without access to radioisotope. [ABSTRACT FROM AUTHOR]

Using antibodies to block the programmed cell death 1 (PD-1)/programmed cell death-ligand 1 (PD-L1) pathway as an immunotherapy has achieved great success in the clinical treatment of various types of carcinoma. However, the efficacy is limited because of tumor-mediated immune immunosuppression and evasion. This study demonstrated that inhibiting the PI3K pathway with (−)-4-O-(4-O-β-D-glucopyranosylcaffeoyl) quinic acid (QA), a new compound from endophytic fungus Penicillium citrinum of Avicennia marina, enhanced the therapeutic efficacy of anti-PD-L1 antibody against esophageal tumors. mEC25 cells were injected into C57BL/6 mice to establish a syngeneic esophageal tumor model. Tumor infiltration lymphocytes (TILs) were analyzed by flow cytometry. Gene and protein expression was detected by qPCR and western blot, respectively. Moreover, the therapeutic effects of QA combining with anti-PD-L1 antibody were evaluated in the tumor model. These data demonstrated that inhibition of PI3K with QA could overcome immunosuppression and promote the response of T-lymphocytes, resulting in the restoration of cytotoxic T cell-mediated tumor control. QA and anti-PD-L1 combination therapy significantly delayed tumor growth. Our results provide a scientific basis to develop combination therapies involving anti-PD-L1 and PI3K inhibitors to improve responses in patients with esophageal cancer. [ABSTRACT FROM AUTHOR]

Alternative splicing (AS) occurs extensively in eukaryotes as an essential mechanism for regulating transcriptome complexity and diversity, but the AS landscape regulating potassium (K) use efficiency in plants is unclear. In this study, we performed high-throughput transcriptome sequencing of roots and shoots from allopolyploid Nicotiana tabacum under K+ deficiency. Preliminary physiological analysis showed that root system architecture was dramatically changed due to potassium deficiency and that IAA content was significantly reduced in root and shoot. AS analysis showed that a total of 28,179 genes exhibited 54,457 AS events, and 1,510 and 1,732 differentially alternatively spliced (DAS) events were identified in shoots and roots under low K+ stress. Nevertheless, only 120 DAS events occurred in both shoots and roots, implying that most DAS events were tissue-specific. Both in shoot and the root, the proportion of DAS genes in differentially expressed (DE) genes equaled that in non-DE genes, which indicated that AS might play a unique regulatory role in response to low potassium. Gene ontology analysis further indicated that transcription regulation and AS modulation worked independently in response to low K+ stress in tobacco, as their target biological processes were different. Totally 45 DAS transcription factors (TFs) were found, which were involved in 18 TF families. Five Auxin response factor (ARF) TFs were significantly DAS in root, suggesting that response to auxin was probably subject to AS regulation in the tobacco root. Our study shows that AS variation occurs extensively and has a particular regulatory mechanism under K+ deficiency in tobacco. The study also links changes in root system architecture with the changes in AS of ARF TFs, which implied the functional significance of these AS events for root growth and architecture. Based on the theory of landsenses ecology, this paper makes a detailed analysis of the architecture pattern of 'The House of Hundred Rooms'(HHR) in the East Dragon Village, a traditional ancient Chinese village, so as to provide a reference for the research on reasonable land use and sustainable development in modern times. By the principle of the vein-compliance in orientation and bearing in landsense creation, the paper analyses the causes of the north-facing feature and the 'counter-tradition' room setting feature lie in the architectural pattern of HHR, revealing that people's psychological perceptions of orientation and bearing is distinct from the physical one. It points out these features of this building complex are the results of man's active adaptation to nature on the basis of respecting nature, which together manages to form a process of constructing the harmonious relationship between man and nature during land use. By the principle of the multiscale (trans-scale or cross-scale) spatiotemporal combination in landsense creation, the paper continues to analyze on the pattern of HHR and finds that it has both the function of cultural inheritance and the innovation of adaptation to new environment, which can promote the protection of historical and cultural elements during the course of development, and bring reference for the realization of sustainable development from the perspective of contemporary historical and cultural heritage protection. The development of economic catalysts for aerobic oxidation procedure has attracted extensive attention. In this work, a novel Ni/Zn supported defective carbon with multi-functional catalytic sites was fabricated via a two-step pyrolysis-H2O2 treatment. The catalyst was applied to the Baeyer–Villiger (B–V) oxidation using ambient air as a green and safe oxidant. The catalyst with optimal Ni/Zn ratio of (2:1) delivers a high catalytic activity (> 92%) and perfect selectivity (> 99%) for the conversion of a wide range of substituted cyclic-ketones to the corresponding lactones. The characterization results have clarified that the H2O2 treatment leads to the formation of active N/O-group on the catalyst surface, which facilitates the adsorption of substrate/intermediate molecules and benefits the reaction. Moreover, the synergistic effect between multi-functional sites results in the buffering/stabilizing of free radicals, enhanced efficiency of oxygen insertion to form lactone. Supply chain system experiences variance amplification in order replenishment and inventory level, leading to severe inefficiencies of the system. Information distortion is universally known as a fundamental reason for the variance amplification phenomenon. The purpose of this paper is to study the effect of demand information sharing in reducing bullwhip effect and improving the robustness of supply chain systems. The "automatic pipeline inventory and order-based production control system, APIOBPCS" is adopted to model supply chains with different information-sharing strategies. The stochastic factors in the supply chain system lead to poor performance in system robustness. Taguchi design is adopted to find out the optimal setting of ordering parameters in the APIOBPCS model for a robust supply chain. An extension of Taguchi design is adopted to solve the multi-response problems. The weighted signal-to-noise ratio is used as the performance index of the overall performance of the supply chain, including inventory cost, customer service level, and inventory variance amplification. The results show that full demand information transparency helps to improve the overall performance of supply chain. Furthermore, the sensitivity analysis of stochastic lead times verifies the results. This research gives some insights to improve the overall performance of supply chain via information sharing.
