The Golgi apparatus is essential for protein sorting, yet its quality control mechanisms are poorly understood. Here we show that the Dsc ubiquitin ligase complex uses its rhomboid pseudo-protease subunit, Dsc2, to assess the hydrophobic length of α-helical transmembrane domains (TMDs) at the Golgi. Thereby the Dsc complex likely interacts with orphaned ER and Golgi proteins that have shorter TMDs and ubiquitinates them for targeted degradation. Some Dsc substrates will be extracted by Cdc48 for endosome and Golgi associated proteasomal degradation (EGAD), while others will undergo ESCRT dependent vacuolar degradation. Some substrates are degraded by both, EGAD- or ESCRT pathways. The accumulation of Dsc substrates entails a specific increase in glycerophospholipids with shorter and asymmetric fatty acyl chains. Hence, the Dsc complex mediates the selective degradation of orphaned proteins at the sorting center of cells, which prevents their spreading across other organelles and thereby preserves cellular membrane protein and lipid composition. At the Golgi, the Dsc ubiquitin ligase complex targets proteins with shorter transmembrane domains for proteasomal or lysosomal degradation. Decarbonization of energy-intensive process industries (EPIs) is a central unresolved challenge for limiting global warming to 1.5°C or well-below 2°C. In this article, we investigate the alignment between government policy and applicable industry strategy in decarbonization efforts across six European countries – Denmark, Germany, Netherlands, Norway, Sweden and the United Kingdom. We distinguish between 'target alignment' (How comparable are the size of the emission reduction commitments?), 'temporal alignment' (How closely do the timelines match?) and 'solution alignment' (Are the same types of solutions prioritized?). Based on an analysis of national policy documents, company strategies of the 10 largest emitting EPI plants in each country, as well as secondary sources, we find high target alignment. However, we find substantially lower temporal alignment as emitters are reluctant to commit to intermediate targets that match the decarbonization timelines laid out in national government policy. Solution alignment is intermediate across all six countries as emitters generally pursue the decarbonization options prioritized in policy, but with most emitters remaining at the level of ambitions or plans and few examples of commercial investments so far. Key policy insights: Public policies across the analysed countries mainly prioritize CCUS, hydrogen and electrification as decarbonization solutions. Large emitters formulate ambitions or plans for the prioritized decarbonization options, but actual investment commitment remains limited. Governments should continuously survey alignment of their policy targets and prioritized decarbonization options with industry targets and activities. In particular, intermediate targets set by the largest emitters do not match those of national governments. Additional support and incentives to make industry act sufficient fast are required. Membrane proteins play crucial roles in cellular functions. However, processes such as the insertion of membrane proteins into the endoplasmic reticulum (ER), their folding into native structures, the assembly of multi-subunit membrane protein complexes, and their targeting from the ER to specific organelles are prone to errors and have a relatively high failure rate. To prevent the accumulation of defective or orphaned membrane proteins, quality control mechanisms assess folding, quantity, and localization of these proteins. This quality control is vital for preserving organelle integrity and maintaining cellular health. In this mini-review, we will focus on how selective membrane protein quality control at the Golgi apparatus, particularly through the defective for SREBP cleavage (Dsc) ubiquitin ligase complex, detects orphaned proteins and prevents their mis-localization to other organelles. Local climate change mitigation plans can be essential in enabling sustainability transitions at the municipality level. However, existing frameworks for assessing the quality of climate plans inadequately address their potential to foster sustainability transitions. We develop an assessment framework consisting of indicators and associated questions by integrating elements of the transition management framework with essential quality dimensions of climate plans and planning processes identified through a literature review. We illustrate and validate the assessment framework by applying it to the climate plans of three medium-sized cities in Switzerland. Our findings demonstrate that the local climate change mitigation plans performed well regarding the tactical and operational indicators but were less optimal concerning strategic and reflexive indicators. Nevertheless, the transition management framework provided a useful framework contributing to a comprehensive and systemic assessment highlighting the importance of stakeholder involvement, inter-sectoral coordination, and monitoring and evaluation as a means for social learning. Asset tokenisation is expected to have an impact on the financial services industry in the future. This paper examines the necessity for traditional custodians to enter tokenised markets to propose a future operating model for traditional custodians. We explore the advantages, such as leveraging regulatory expertise and existing customer bases and challenges, including the adoption of new technology stacks involved in such a model. Our proposed operating model utilises a blockchain infrastructure that relies on the use of tokens that spans both traditional and digital assets to create an automated platform. This platform can include the automation of credit risk and liquidity across multi-asset classes and multi-jurisdictional operations. Notably, our model introduces two types of tokens: value tokens (the fully digital or tokenised assets as well as tokenised money) and proxy tokens (among others, representing traditional assets). While value tokens carry value and represent tokenised assets, proxy tokens do not carry value. These proxy tokens are an internal representation of traditional assets that can facilitate managing the intra-day liquidity and extended credit lines offered to customers. Within our proposed operating model, all transaction instructions, whether in the traditional or tokenised realm, are streamlined through a unified 'instruction worklist'. Aims To assess the agreement between left ventricular end-diastolic diameter index (LVEDDi) and volume index (LVEDVi) to define LV dilatation and to investigate the respective prognostic implications in patients with heart failure (HF). Methods and results Patients with HF symptoms and LV ejection fraction (LVEF) < 50% undergoing cardiac magnetic resonance were evaluated retrospectively. LV dilatation was defined as LVEDDi or LVEDVi above the upper normal limit according to published reference values. Patients were followed up for the combined endpoint of cardiovascular death or HF hospitalization during 5 years. A total of 564 patients (median age 64 years; 79% men) were included. LVEDDi had a modest correlation with LVEDVi (r = 0.682, P < 0.001). LV dilatation was noted in 84% of patients using LVEDVi-based definition and in 73% using LVEDDi-based definition, whereas 20% of patients displayed discordant definitions of LV dilatation. During a median follow-up of 2.8 years, patients with both dilated LVEDDi and LVEDVi had the highest cumulative event rate (HR 3.00, 95% CI 1.15–7.81, P = 0.024). Both LVEDDi and LVEDVi were independently associated with the primary outcome (hazard ratio 3.29, 95%, P < 0.001 and 2.8, P = 0.009; respectively). Conclusion The majority of patients with HF and LVEF < 50% present both increased LVEDDi and LVEDVi whereas 20% show discordant linear and volumetric definitions of LV dilatation. Patients with increased LVEDDi and LVEDVi have the worst clinical outcomes suggesting that the assessment of these two metrics is needed for better risk stratification. [ABSTRACT FROM AUTHOR]

The Apiospora genus comprises filamentous fungi with promising potential, though its full capabilities remain undiscovered. In this study, we present the first genome assembly of an Apiospora arundinis isolate, demonstrating a highly complete and contiguous assembly estimated to 48.8 Mb, with an N99 of 3.0 Mb. Our analysis predicted a total of 15,725 genes, with functional annotations for 13,619 of them, revealing a fungus capable of producing very high amounts of carbohydrate-active enzymes (CAZymes) and secondary metabolites. Through transcriptomic analysis, we observed differential gene expression in response to varying growth media, with several genes related to carbohydrate metabolism showing significant upregulation when the fungus was cultivated on a hay-based medium. Facultative heterochromatin marked by histone H3 lysine 27 trimethylation (H3K27me3) is an important regulatory layer involved in secondary metabolite (SM) gene silencing and crucial for fungal development in the genus Fusarium. While this histone mark is essential in some (e.g., the rice pathogen Fusarium fujikuroi), it appears dispensable in other fusaria. Here, we show that deletion of FpKMT6 is detrimental but not lethal in the plant pathogen Fusarium proliferatum, a member of the Fusarium fujikuroi species complex (FFSC). Loss of FpKmt6 results in aberrant growth, and expression of a large set of previously H3K27me3-silenced genes is accompanied by increased H3K27 acetylation (H3K27ac) and an altered H3K36me3 pattern. Next, H3K9me3 patterns are affected in Δfpkmt6, indicating crosstalk between both heterochromatic marks that became even more obvious in a strain deleted for FpKMT1 encoding the H3K9-specific histone methyltransferase. In Δfpkmt1, all H3K9me3 marks present in the wild-type strain are replaced by H3K27me3, a finding that may explain the subtle phenotype of the Δfpkmt1 strain which stands in marked contrast to other filamentous fungi. A large proportion of SM-encoding genes is allocated with H3K27me3 in the wild-type strain and loss of H3K27me3 results in elevated expression of 49% of them. Interestingly, genes involved in the biosynthesis of the phytohormones gibberellins (GA) are among the most upregulated genes in Δfpkmt6. Although several FFSC members harbor GA biosynthetic genes, its production is largely restricted to F. fujikuroi, possibly outlining the distinct lifestyles of these notorious plant pathogens. We show that H3K27me3 is involved in GA gene silencing in F. proliferatum and at least one additional FFSC member, and thus, may serve as a regulatory layer for gene silencing under non-favoring conditions. Author summary: H3K27me3 is a hallmark of facultative heterochromatin, and as such tightly connected with gene silencing. In the genus Fusarium, a large proportion of the genome is allocated with this histone mark. Not surprisingly its removal is detrimental and even lethal for some fusaria e.g., the plant pathogen Fusarium fujikuroi for still unknown reasons. Here, we show that Kmt6 is vital but not essential in the closely related Fusarium proliferatum, allowing for the first time a comprehensive analysis of Kmt6 loss-of-function in a member of the Fusarium fujikuroi species complex (FFSC). By an "omics"-oriented approach, we show the genome-wide distribution of activating and silencing histone marks in the wild-type as well as a Δfpkmt6 strain, mapped on a newly annotated genome assembled to near chromosome level. Loss of H3K27me3 and H3K9me3 involved in facultative- and constitutive heterochromatin, respectively, revealed a crucial crosstalk between both histone marks in this genus. Purpose of the Review: The role of multimodality imaging in the evaluation of patients with aortic regurgitation is summarized in this review. Recent Findings: The etiology (mechanism) of the aortic regurgitation and the severity of aortic regurgitation and hemodynamic consequences are key in the decision making of patients with severe aortic regurgitation. While echocardiography remains as the leading technique to assess all these parameters, other imaging techniques have become essential for the accurate assessment of aortic regurgitation severity and the timing of aortic intervention. The anatomic suitability of transcatheter aortic valve implantation in inoperable patients with severe aortic regurgitation is usually assessed with computed tomography. Summary: Aortic regurgitation is a prevalent disease with various pathophysiological mechanisms that need a personalized treatment. The evaluation of the mechanism and severity of aortic regurgitation can be initially performed with echocardiography. Three-dimensional techniques, including echocardiography, have become very relevant for accurate assessment of the regurgitation severity and its hemodynamic consequences. Assessment of myocardial tissue characteristics with cardiac magnetic resonance is key in the risk stratification of patients and in the timing of aortic intervention. LAT1 and 4F2hc form a heterodimeric membrane protein complex, which functions as one of the best characterized amino acid transporters. Since LAT1–4F2hc is required for the efficient uptake of essential amino acids and hormones, it promotes cellular growth, in part, by stimulating mTORC1 (mechanistic target of rapamycin complex 1) signalling and by repressing the integrated stress response (ISR). Gain or loss of LAT1–4F2hc function is associated with cancer, diabetes, and immunological and neurological diseases. Hence, LAT1–4F2hc represents an attractive drug target for disease treatment. Specific targeting of LAT1–4F2hc will be facilitated by the increasingly detailed understanding of its molecular architecture, which provides important concepts for its function and regulation. Here, we summarize (i) structural insights that help to explain how LAT1 and 4F2hc assemble to transport amino acids across membranes, (ii) the role of LAT1–4F2hc in key metabolic signalling pathways, and (iii) how derailing these processes could contribute to diseases. The literature on the catching up of latecomer countries has pointed at windows of opportunity as a precondition for catching up. Previous research has however failed to illuminate the determining factors affecting the number of firms benefitting from windows of opportunity. The current article addresses this gap by combining insights on the nature of knowledge spillovers with sectoral characteristics. This perspective is applied empirically by analyzing the number of firms benefitting from a 'green' window of opportunity in the Chinese biomass power plant industry, specifically related to changes in the institutional framework conditions in the form of a feed-in tariff and specified targets for renewable energy. The article finds that while a single Chinese firm constituted the initial phase of the catch-up cycle, domestic knowledge spillovers allowed a larger number of Chinese firms to benefit from the window of opportunity in the later stage of the catch-up cycle. The article points at the importance of combining sectoral characteristics with the degree of domestic knowledge spillovers as key determinants for the number of firms profiting from windows of opportunity. Aims: Dapagliflozin improves the prognosis of patients with heart failure (HF), regardless of left ventricular ejection fraction (LVEF). However, its effect on cardiac remodelling parameters, specifically left atrial (LA) remodelling, is not well established. Methods and results: The DAPA‐MODA trial (NCT04707352) is a multicentre, single‐arm, open‐label, prospective and interventional study that aimed to evaluate the effect of dapagliflozin on cardiac remodelling parameters over 6 months. Patients with stable chronic HF receiving optimized guideline‐directed therapy, except for any sodium–glucose cotransporter 2 inhibitor, were included. Echocardiography was performed at baseline, 30 and 180 days, and analysed by a central core‐lab in a blinded manner to both patient and time. The primary endpoint was the change in maximal LA volume index (LAVI). A total of 162 patients (64.2% men, 70.5 ± 10.6 years, 52% LVEF >40%) were included in the study. At baseline, LA dilatation was observed (LAVI 48.1 ± 22.6 ml/m2) and LA parameters were similar between LVEF‐based phenotypes (≤40% vs. >40%). LAVI showed a significant reduction at 180 days (−6.6% [95% confidence interval −11.1, −1.8], p = 0.008), primarily due to a decrease in reservoir volume (−13.8% [95% confidence interval −22.5, −4], p = 0.007). Left ventricular geometry improved with significant reductions in left ventricular mass index (−13.9% [95% confidence interval −18.7, −8.7], p < 0.001), end‐diastolic volume (−8.0% [95% confidence interval −11.6, −4.2], p < 0.001) and end‐systolic volume (−11.9% [95% confidence interval −16.7, −6.8], p < 0.001) at 180 days. N‐terminal pro‐B‐type natriuretic peptide (NT‐proBNP) showed a significant reduction at 180 days (−18.2% [95% confidence interval −27.1, −8.2], p < 0.001), without changes in filling Doppler measures. Conclusion: Dapagliflozin administration in stable out‐setting patients with chronic HF and optimized therapy results in global reverse remodelling of cardiac structure, including reductions in LA volumes and improvement in left ventricular geometry and NT‐proBNP concentrations. 6-methylsalicylic acid (6-MSA) is a small, simple polyketide produced by a broad spectrum of fungal species. Since fungi obtained the ability to synthesize 6-MSA from bacteria through a horizontal gene transfer event, it has developed into a multipurpose metabolic hub from where numerous complex compounds are produced. The most relevant metabolite from a human perspective is the small lactone patulin as it is one of the most potent mycotoxins. Other important end products derived from 6-MSA include the small quinone epoxide terreic acid and the prenylated yanuthones. The most advanced modification of 6-MSA is observed in the aculin biosynthetic pathway, which is mediated by a non-ribosomal peptide synthase and a terpene cyclase. In this short review, we summarize for the first time all the possible pathways that takes their onset from 6-MSA and provide a synopsis of the responsible gene clusters and derive the resulting biosynthetic pathways. We perform, to our best knowledge, the first systematic mapping of the foundational economy (FE) at the sub-national level by looking at the FE employment in Swedish regions between 2007 and 2016. We show that the FE itself not only suffered less than traded activities from employment decline during the Great Recession of 2007–2009 but was also a domain of substantial job creation in the post-crisis recovery. At the same time, regions with higher dependence on foundational employment were hit harder during the crisis in terms of overall labour market performance. We demonstrate that it is specific compositions of foundational and traded activities in the regional employment mix that relate differently to regional employment growth in times of crisis and recovery. The Penicillia are known to produce a wide range natural products—some with devastating outcome for the agricultural industry and others with unexploited potential in different applications. However, a large-scale overview of the biosynthetic potential of different species has been lacking. In this study, we sequenced 93 Penicillium isolates and, together with eleven published genomes that hold similar assembly characteristics, we established a species phylogeny as well as defining a Penicillium pangenome. A total of 5612 genes were shared between ≥ 98 isolates corresponding to approximately half of the average number of genes a Penicillium genome holds. We further identified 15 lateral gene transfer events that have occurred in this collection of Penicillium isolates, which might have played an important role, such as niche adaption, in the evolution of these fungi. The comprehensive characterization of the genomic diversity in the Penicillium genus supersedes single-reference genomes, which do not necessarily capture the entire genetic variation. Problem-solving capacity is continuously gathering prominence in the Engineering education programmes of most institutions of higher learning. Embracing problem-based learning is necessary for Electrical Engineering lecturers in technical and vocational education and training (TVET) so as to shift from teacher-centred to student-centred teaching approaches which engage students in the learning process. Despite this, research on the use of problembased learning in TVET contexts by Electrical Engineering lecturers is limited. In this study, we conducted a scoping review to identify those key components which are necessary for the effective implementation of problem-based learning in a TVET Electrical Engineering programme. Our findings indicated that the problem, the facilitator and the students are the three components noted as essential constituents that are vital to the effective implementation of a problem-based learning strategy in TVET Electrical Engineering programmes. Our findings stress that, in addition to the three components, the type and nature of the problem, including the philosophy of the subject or the programme, need to be considered before inculcating problem-solving capabilities in TVET Electrical Engineering lecturers. Aqueous organic redox flow batteries (AORFBs) have gained increased interest as a promising solution to store energy from sustainable energy sources. Inspired by naturally occurring bio‐quinones, we here propose a new electrolyte based on the fungal compound phoenicin. Phoenicin was produced using the filamentous fungus Penicillium atrosanguineum at a concentration of 1.24 g L−1 liquid medium and extracted using ethyl acetate to a purity exceeding 95 %. The fungus may provide a benefit of high scalability of the biosynthesis‐based production of the electroactive substance. Here, we demonstrate the performance of biologically produced phoenicin as a negative electrolyte in an RFB against ferro/ferricyanide, as a proof of concept, giving an initial capacity of 11.75 Ah L−1 and a capacity decay of 2.85 % day−1. For a deeper investigation of the battery setup, in situ attenuated total reflection infrared (ATR‐IR) spectra of the phoenicin electrolyte were recorded. Climate change is causing problems for agriculture, but the effect of combined abiotic stresses on crop nutritional quality is not clear. Here we studied the effect of 10 combinations of climatic conditions (temperature, CO2, O3 and drought) under controlled growth chamber conditions on the grain yield, protein, and mineral content of 3 wheat varieties. Results show that wheat plants under O3 exposure alone concentrated + 15 to + 31% more grain N, Fe, Mg, Mn P and Zn, reduced K by − 5%, and C did not change. Ozone in the presence of elevated CO2 and higher temperature enhanced the content of Fe, Mn, P and Zn by 2–18%. Water-limited chronic O3 exposure resulted in + 9 to + 46% higher concentrations of all the minerals, except K. The effect of climate abiotic factors could increase the ability of wheat to meet adult daily dietary requirements by + 6% to + 12% for protein, Zn and Fe, but decrease those of Mg, Mn and P by − 3% to − 6%, and K by − 62%. Cells are constantly exposed to various chemical and physical stimuli. While much has been learned about the biochemical factors that regulate secretory trafficking from the endoplasmic reticulum (ER), much less is known about whether and how this trafficking is subject to regulation by mechanical signals. Here, we show that subjecting cells to mechanical strain both induces the formation of ER exit sites (ERES) and accelerates ER‐to‐Golgi trafficking. We found that cells with impaired ERES function were less capable of expanding their surface area when placed under mechanical stress and were more prone to develop plasma membrane defects when subjected to stretching. Thus, coupling of ERES function to mechanotransduction appears to confer resistance of cells to mechanical stress. Furthermore, we show that the coupling of mechanotransduction to ERES formation was mediated via a previously unappreciated ER‐localized pool of the small GTPase Rac1. Mechanistically, we show that Rac1 interacts with the small GTPase Sar1 to drive budding of COPII carriers and stimulates ER‐to‐Golgi transport. This interaction therefore represents an unprecedented link between mechanical strain and export from the ER. Sphingolipid levels are crucial determinants of neurodegenerative disorders and therefore require tight regulation. The Orm protein family and ceramides inhibit the rate-limiting step of sphingolipid biosynthesis—the condensation of L-serine and palmitoyl-coenzyme A (CoA). The yeast isoforms Orm1 and Orm2 form a complex with the serine palmitoyltransferase (SPT). While Orm1 and Orm2 have highly similar sequences, they are differentially regulated, though the mechanistic details remain elusive. Here, we determine the cryoelectron microscopy structure of the SPT complex containing Orm2. Complementary in vitro activity assays and genetic experiments with targeted lipidomics demonstrate a lower activity of the SPT-Orm2 complex than the SPT-Orm1 complex. Our results suggest a higher inhibitory potential of Orm2, despite the similar structures of the Orm1- and Orm2-containing complexes. The high conservation of SPT from yeast to man implies different regulatory capacities for the three human ORMDL isoforms, which might be key for understanding their role in sphingolipid-mediated neurodegenerative disorders. The foundational economy approach to economic development suggests that the production of goods and services that are critical to human welfare (e.g., water, energy, education and elderly care) should be the central concern for policymaking. This paper examines whether this approach could address three core challenges for regional development: social polarization, interregional inequality and environmental sustainability. It concludes that the foundational economy approach is particularly promising in addressing challenges relating to social polarization, but could benefit from further engagement with alternative theories on innovation, work in political science on policy for improving government quality, and various literatures on environmental sustainability. Climate change is causing problems for agriculture, but the effect of combined abiotic stresses on crop nutritional quality is not clear. Here we studied the effect of 10 combinations of climatic conditions (temperature, CO2, O3 and drought) under controlled growth chamber conditions on the grain yield, protein, and mineral content of 3 wheat varieties. Results show that wheat plants under O3 exposure alone concentrated + 15 to + 31% more grain N, Fe, Mg, Mn P and Zn, reduced K by − 5%, and C did not change. Ozone in the presence of elevated CO2 and higher temperature enhanced the content of Fe, Mn, P and Zn by 2–18%. Water-limited chronic O3 exposure resulted in + 9 to + 46% higher concentrations of all the minerals, except K. The effect of climate abiotic factors could increase the ability of wheat to meet adult daily dietary requirements by + 6% to + 12% for protein, Zn and Fe, but decrease those of Mg, Mn and P by − 3% to − 6%, and K by − 62%. This article explores whether and how innovation policies promote transitions to more sustainable configurations in socio-technical systems. Empirically it departs from an analysis of the bioeconomy policy strategies and instruments in four Nordic countries. The analysis highlights that while a transformative approach is present at the policy strategy level in the Nordic countries, the bioeconomy policy instruments implemented are significantly less transformative. The article argues that a bioeconomy transition that contributes to sustainable development will require a redirection of policies towards transformative failures (directionality, policy coordination, demand articulation, and reflexivity). In this regard, it is important that policymakers experiment with and explore ways of balancing between traditional market and innovation system approaches and new policy approaches for transformative change. The paper contributes to debates on dark phases of regional industry path development processes. We focus on the role of industry legitimacy and study legitimacy spillovers from contextual structures in the case of the biogas industry in Scania, Sweden. Previously presented as a success case, we show how legitimacy spillovers from various contextual structures influenced the path development process, in both the emergence and acceleration phases, and in the recent decline period. The analysis highlights how previously accumulated resources in the form of knowledge, finance and market were insufficient to prevent industry decline when it suffered a loss of legitimacy. The biosynthetic pathways for the fungal polyketides bikaverin and bostrycoidin, from Fusarium verticillioides and Fusarium solani respectively, were reconstructed and heterologously expressed in S. cerevisiae alongside seven different phosphopantetheinyl transferases (PPTases) from a variety of origins spanning bacterial, yeast and fungal origins. In order to gauge the efficiency of the interaction between the ACP-domains of the polyketide synthases (PKS) and PPTases, each were co-expressed individually and the resulting production of target polyketides were determined after 48 h of growth. In co-expression with both biosynthetic pathways, the PPTase from Fusarium verticillioides (FvPPT1) proved most efficient at producing both bikaverin and bostrycoidin, at 1.4 mg/L and 5.9 mg/L respectively. Furthermore, the remaining PPTases showed the ability to interact with both PKS's, except for a single PKS-PPTase combination. The results indicate that it is possible to boost the production of a target polyketide, simply by utilizing a more optimal PPTase partner, instead of the commonly used PPTases; NpgA, Gsp and Sfp, from Aspergillus nidulans, Brevibacillus brevis and Bacillus subtilis respectively. The phylogenetic relationship of the Arthrinium genus has changed throughout the years. For many years, the Arthrinium genus included the Apiospora genus as well. New evidence has now showed that these two genera in fact are phylogenetically different and belong to two different clades. Here, we present the first genome draft within the Arthrinium genus. This genome was sequenced using the MinION platform from Oxford Nanopore Technologies and the assembly was contiguous. The assembly comprises ten contigs totaling 39.8 Mb with an N50 length of 7.9. In the assembly, 11,602 genes were predicted whereof 10,784 were functionally annotated. A total of 37 rRNA genes were observed in the assembly and repeat elements spanning 7.39% of the genome were found. A total of 99 secondary metabolite gene clusters were predicted, showing a high potential of novel secondary metabolites. This genome sequence will not only be useful for further investigation of the Arthrinium clade, but also for discovery of novel secondary metabolite compounds that could be of high interest for the food, agricultural, or pharmaceutical industry. Background: This study aimed to examine the prevalence of atrial tricuspid regurgitation (ATR) and atrial mitral regurgitation (AMR) in the setting of atrial fibrillation (AFib) and identify variables related to the severity of both types of regurgitation. Methods: Cross‐sectional study evaluating data from transthoracic echocardiograms performed during 2019. We included patients with AFib during the examination, and without primary valve disease or other significant heart disease. Results: Four‐hundred and thirty‐two patients fulfilled the inclusion criteria (mean age 77.5±9.2 years, 49.1% women). We observed significant ATR in 14.8%, and significant AMR in 1.4% of patients. ATR and AMR severities were equal in 49.3% of patients, and 41% displayed greater ATR severity. ATR prevalence was significantly greater among women (23.1% vs 6.8%, p < 0.001), but AMR prevalence was similar between genders (1.9% vs.9%, p = 0.443). Variables related to greater ATR severity were: female sex (OR: 2.61, 95%CI: 1.60–4.24), left atrial (LA) volume (OR: 3.58, 95%CI: 1.50–8.55), systolic pulmonary artery pressure (OR: 1.10, 95%CI: 1.07–1.13), and moderate AMR (OR: 2.21, 95%CI: 1.22–4.00). Variables related to greater AMR severity were female sex (OR: 1.96, 95%CI: 1.24–3.09), LA volume (OR: 11.68, 95%CI: 5.29–25.80), and body mass index (OR:.94, 95%CI:.90–.98). Conclusions: In the context of AFib, ATR was more prevalent than AMR and prevailed in women. LA enlargement was associated with higher degrees of both AMR and ATR. Pulmonary hypertension was also independently associated with ATR, as well as greater AMR severity, suggesting possible adaptive changes in leaflets that might modify the atrial regurgitation incidence. The target of zero emissions sets a new standard for industry and industrial policy. Industrial policy in the twenty-first century must aim to achieve zero emissions in the energy and emissions intensive industries. Sectors such as steel, cement, and chemicals have so far largely been sheltered from the effects of climate policy. A major shift is needed, from contemporary industrial policy that mainly protects industry to policy strategies that transform the industry. For this purpose, we draw on a wide range of literatures including engineering, economics, policy, governance, and innovation studies to propose a comprehensive industrial policy framework. The policy framework relies on six pillars: directionality, knowledge creation and innovation, creating and reshaping markets, building capacity for governance and change, international coherence, and sensitivity to socio-economic implications of phase-outs. Complementary solutions relying on technological, organizational, and behavioural change must be pursued in parallel and throughout whole value chains. Current policy is limited to supporting mainly some options, e.g. energy efficiency and recycling, with some regions also adopting carbon pricing, although most often exempting the energy and emissions intensive industries. An extended range of options, such as demand management, materials efficiency, and electrification, must also be pursued to reach zero emissions. New policy research and evaluation approaches are needed to support and assess progress as these industries have hitherto largely been overlooked in domestic climate policy as well as international negotiations. Key policy insights Energy and emission intensive industries can no longer be complacent about the necessity of zero greenhouse gas (GHG) emissions. Zero emissions require profound technology and organizational changes across whole material value chains, from primary production to reduced demand, recycling and end-of-life of metals, cement, plastics, and other materials. New climate and industrial policies are necessary to transform basic materials industries, which are so far relatively sheltered from climate mitigation. It is important to complement technology R&D with the reshaping of markets and strengthened governance capacities in this emerging policy domain. This paper focuses on path development processes after a sudden path exhaustion. We analyse the decline, the closure and the attempts at the re-orientation of a forestry-based industry agglomeration in Southern Norway, located around the municipality of Hønefoss. In particular, this paper focuses on the Treklyngen holding company in Hønefoss. This paper explores how policy may be influenced by and built upon regional capabilities to support new path development in the aftermath of path exhaustion. Due to a spatial turn in the socio-technical transition literature, the geography of energy transitions has recently been taken increasingly seriously, leading to burgeoning research output on regional energy transitions since early 2010. Amidst this wealth of publications, however, it can be difficult to keep track of its diverse and constantly evolving landscape. This editorial therefore aims at developing a framework that allows for bringing multiple approaches to regional energy transitions into conversation with each other and that helps to understand and explain the complexity of these interdependencies in ways that go beyond observing regional variety in energy transitions. The rapidly expanding stream of path development studies recognises that translating observations from past paths to conscious path creation requires conceptually linking agency to path development frameworks. Actors frame issues about and for the future, coordinate their actions in the present and make sense of what may have transpired in the past. The main objective of the paper is to explore the roles that actors play in green path development by answering the following main research questions: (a) Who are the core actors in green path development in the Nordic regions, ie. in industrial development around products, solutions or technologies that make regional economies more sustainable? (b) What are their main roles in relation to other actors? (c) What are the differences and similarities between the regions in terms of agency? The paper explores whether similar actors take on different roles in different regions and whether different actors may assume similar roles. For the empirical analysis, we identified seven roles in change agency. The empirical results showed that institutional entrepreneurship was the core of change agency in conjunction with innovative entrepreneurship and place-based leadership. The other four roles supported path development efforts. The results also show that institutional entrepreneurship is not a solo activity but a collective form of agency and that well-functioning shared institutional entrepreneurship may have a chance to change institutions for green path development. Aims: Systolic pulmonary artery pressure (SPAP) and right heart adaptation in relation to pre-existing preload are often disregarded. To determine volume-related changes in the pulmonary-right ventricle (RV) unit and the preload dependence of its components, we analysed pulmonary haemodynamics and right ventricular performance, taking advantage of the plasma volume removal associated to haemodialysis (HD). Methods and results: Fifty-three stable patients on chronic HD with LVEF > 50% and without heart failure were recruited (mean age 63.0 ± 12.4 years; 31.2% women; hypertension in 89% and diabetes in 53%) and evaluated just before and after HD (mean ultrafiltration volume 2.4 ± 0.7 l). SPAP from both times were available in 39 patients. After HD, SPAP decreased (42.2 ± 12.6 to 33.7 ± 11.6 mmHg, p < 0.001) without modification of non-invasive pulmonary vascular resistance (1.75 ± 0.44 to 1.75 ± 0.40 eWU, p = 0.94). Age and drop in the E/eʹ ratio were the variables associated with greater reduction in PASP (p = 0.022 and p = 0.049, respectively). A significant reduction of right chamber sizes was observed, along with a diminution in measures of RV contractility, excluding RV longitudinal strain. Functional tricuspid regurgitation (FTR) diminution was observed in 26% of patients, occurring in every case with more than mild FTR. On multivariate analyses, left atrial size was the only predictor of pulmonary hypertension (defined as SPAP > 40 mmHg) (OR 1.29 (1.07–1.56), p = 0.006). Conclusion: Rapid volemic changes may affect FTR grading, RV size and contractility, with RV longitudinal strain being less variable than conventional parameters. The regulation of nutrient uptake into cells is important, as it allows to either increase biomass for cell growth or to preserve homoeostasis. A key strategy to adjust cellular nutrient uptake is the reconfiguration of the nutrient transporter repertoire at the plasma membrane by the addition of nutrient transporters through the secretory pathway and by their endocytic removal. In this review, we focus on the mechanisms that regulate selective nutrient transporter endocytosis, which is mediated by the α‐arrestin protein family. In the budding yeast Saccharomyces cerevisiae, 14 different α‐arrestins (also named arrestin‐related trafficking adaptors, ARTs) function as adaptors for the ubiquitin ligase Rsp5. They instruct Rsp5 to ubiquitinate subsets of nutrient transporters to orchestrate their endocytosis. The ART proteins are under multilevel control of the major nutrient sensing systems, including amino acid sensing by the general amino acid control and target of rapamycin pathways, and energy sensing by 5′‐adenosine‐monophosphate‐dependent kinase. The function of the six human α‐arrestins is comparably under‐characterised. Here, we summarise the current knowledge about the function, regulation and substrates of yeast ARTs and human α‐arrestins, and highlight emerging communalities and general principles. When cultivating the kelp Saccharina latissima, knowledge on the availability of seeding material for the production is essential. Applying a spore seeding approach requires spores from the reproductive organs of the fertile sporophytes (sori). As sori are generally not present during the time of seeding in late summer, the production of spores (sporogenesis) can be artificially induced by removing the meristematic part of the sporophyte and keeping the sporophyte under short day, temperate, and nutrient-replete conditions. Only limited information is available on the effect of light intensities including darkness on the sporogenesis of S. latissima. This study examined the natural pattern of sporogenesis in S. latissima from Middelfart, Denmark, and the effect of four different light regimes (0, 20, 60, or 120 μmol photons m−2 s−1) on the artificial induction of sporogenesis in S. latissima. Natural reproductivity and availability of spores in Denmark peaked in early winter, with 86% of the population being reproductive in November. Reproductive material was available from October until late spring, but with a variable spore release from 11 × 103 to 1.2 × 106 spores cm−2 sori. The artificial induction of sporogenesis was optimal in darkness with > 90% of sporophytes developing sori after 49 days, with an average spore release density of 1.15 ± 0.38 × 106 spores cm−2 sori. The results confirmed that S. latissima in Denmark follows the general pattern of reproduction of S. latissima in North Atlantic regions and demonstrated for the first time that sporogenesis in S. latissima can be efficiently induced in darkness. How should manufacturers motivate their retailers to provide customer services? The vertical restraints literature tells us that retail competition distorts service incentives in the short run. We consider how repeated interaction mitigates this problem, and particularly how a manufacturer can provide service incentives with discretionary lump‐sum payments. We find that these payments may allow the manufacturer to sustain optimal service levels even if retailers are very impatient. We also show that banning reverse lump‐sum payments may deprive consumers of the chance to enjoy high‐quality services, and thereby reduce their welfare. [ABSTRACT FROM AUTHOR]

Evidence regarding any association of HDL-particle (HDL-P) derangements and HDL-cholesterol content with cardiovascular (CV) death in chronic heart failure (HF) is lacking. To investigate the prognostic value of HDL-P size (HDL-Sz) and the number of cholesterol molecules per HDL-P for CV death in HF patients. Outpatient chronic HF patients were enrolled. Baseline HDL-P number, subfractions and HDL-Sz were measured using 1H-NMR spectroscopy. The HDL-C/P ratio was calculated as HDL-cholesterol over HDL-P. Endpoint was CV death, with non-CV death as the competing event. 422 patients were included and followed-up during a median of 4.1 (0–8) years. CV death occurred in 120 (30.5%) patients. Mean HDL-Sz was higher in CV dead as compared with survivors (8.39 nm vs. 8.31 nm, p < 0.001). This change in size was due to a reduction in the percentage of small HDL-P (54.6% vs. 60% for CV-death vs. alive; p < 0.001). HDL-C/P ratio was higher in the CV-death group (51.0 vs. 48.3, p < 0.001). HDL-Sz and HDL-C/P ratio were significantly associated with CV death after multivariable regression analysis (HR 1.22 [95% CI 1.01–1.47], p = 0.041 and HR 1.04 [95% CI 1.01–1.07], p = 0.008 respectively). SATB2 is a schizophrenia risk gene and is genetically associated with human intelligence. How it affects cognition at molecular level is currently unknown. Here, we show that interactions between SATB2, a chromosomal scaffolding protein, and the inner nuclear membrane protein LEMD2 orchestrate the response of pyramidal neurons to neuronal activation. Exposure to novel environment in vivo causes changes in nuclear shape of CA1 hippocampal neurons via a SATB2‐dependent mechanism. The activity‐driven plasticity of the nuclear envelope requires not only SATB2, but also its protein interactor LEMD2 and the ESCRT‐III/VPS4 membrane‐remodeling complex. Furthermore, LEMD2 depletion in cortical neurons, similar to SATB2 ablation, affects neuronal activity‐dependent regulation of multiple rapid and delayed primary response genes. In human genetic data, LEMD2‐regulated genes are enriched for de novo mutations reported in intellectual disability and schizophrenia and are, like SATB2‐regulated genes, enriched for common variants associated with schizophrenia and cognitive function. Hence, interactions between SATB2 and the inner nuclear membrane protein LEMD2 influence gene expression programs in pyramidal neurons that are linked to cognitive ability and psychiatric disorder etiology. SYNOPSIS: Cognitive processes depend on gene transcription in pyramidal neurons, which is stimulated by neuronal activity. Here, the interactions between the chromosomal scaffold protein SATB2, the inner nuclear membrane protein LEMD2, and the ESCRT‐III/VPS4 machinery are shown to orchestrate these activity‐dependent gene transcription and nuclear shape changes. The advent of the Fourth Industrial Revolution (4IR) affirms that the transformation and advancement of all industries and society are progressively driven by emergent and rapidly changing technologies. In order to help establish a technologically literate society, it is crucial for lecturers in Technical Engineering to stay abreast of the latest trends and technological advancements in their area of specialisation. This article reports on the findings of a sub-section of a nationwide survey that collected data from lecturers in Technical Engineering at 52 TVET college campuses across South Africa (n = 577) that offer TVET Engineering Study programmes. The purpose of the survey was to profile TVET Engineering Study lecturers' knowledge and pedagogical practices in engineering programmes across South Africa. The sub-section of the survey specifically probed the participants' awareness and understanding of discipline-specific technological advancements and digital educational enterprises, and also the potential impact of these on teaching technical subjects. The findings show that 52,3% (n = 302) of the participating lecturers in Technical Engineering are unaware of any technological advancements in their area of specialisation and that they do not know what the potential impact of this might be on future technical training. When Engeström's Cultural Historical Activity Theory (CHAT model) is applied to the activity systems that are involved, the underlying tension between these systems is revealed. This article examines the possible implications of these findings for the renewal of the Technical Engineering curriculum, considering that these lecturers demonstrate limited awareness of the technological advancements needed to participate effectively in the 4IR era. The foundational economy perspective suggests that industries, which provide services essential to all citizens' well-being and participation in everyday life, should be placed centrally in economic development policy. This article studies the extent to which local governments put emphasis on foundational industries in their strategies for development. Moreover, drivers behind priority-setting are examined. Based on an analysis of all 98 Danish municipalities' planning strategies and semi-structured interviews with relevant actors from two rural municipalities, we find that foundational industries are to a great extent emphasised, even if they are not characterised as the foundation for economic development. Rather, foundational industries are prioritised in the absence of other options or when municipalities are not compelled to put local job creation as a crucial focus to attract and maintain inhabitants. The parasitic salmon louse, and its documented resistance to chemotherapeutants, represents the most persistent environmental challenge to global salmonid aquaculture. We used a genetic marker associated with pyrethroid resistance to analyse ∼15 000 lice collected from the North Atlantic in the period 2000–2017. The genotype associated with resistance was not detected in lice collected from throughout the North Atlantic in the year 2000 or 2002. However, by the year 2009 onwards, it was found in lice from fish farms throughout much of the North Atlantic. It was also found in modest frequencies in lice collected from wild Atlantic salmon captured off Greenland. The most recent samples displayed very high frequencies of the genotype associated with resistance, particularly in intensive aquaculture regions of Norway (>90%) and Scotland (>70%). These results closely align with observations from the field. We suggest that pyrethroid resistance first emerged in Europe just before or around the year 2000 and was thereafter dispersed throughout much of the North Atlantic where its increased frequency was driven by extensive pyrethroid use. Although the resistant genotype was not detected in lice from Canada, it is likely to occur in very low frequencies that would quickly increase if pyrethroids were to be used in that region. Although most point sources of persistent organic pollutants (POPs), including polycyclic aromatic hydrocarbons (PAHs), are at lower latitudes, the Arctic region is contaminated. In particular, PAHs now dominate the POP body burden of the region's marine biota at the lower trophic levels. Greenlandic Inuits have the most elevated levels of POPs in their blood compared to any other population, due to their consumption of seal meat and other marine mammals. PAHs, the by-products of the incomplete combustion of petroleum products, are known carcinogens and have been shown to affect the immune system, reproduction, endocrine functions, and the nervous system. With industrial activities and climate change set to increase local PAH emissions, it is paramount to document changes in atmospheric PAH deposition to further investigate PAH exposure in the region and attribute contaminations to their sources. As a measure of atmospheric pollution, we sampled bryophyte herbarium specimens of three common and widespread species collected in Greenland between the 1920s and 1970s after which time new collections were not available. They were analyzed for 19 PAHs using GC-MS (gas chromatography mass spectrometry). The presence of more low-molecular-weight PAHs than high-molecular-weight PAHs is evidence that the PAH contamination in Greenland is due to long-range transport rather than originating from local sources. The results show peaks in PAH atmospheric deposition in the first part of the 19th century followed by a trend of decrease, which mirror global trends in atmospheric pollution known from those periods. PAHs associated with wood and fossil-fuel combustion decrease in the 1970s coinciding with the disappearance of charcoal pits and foundries in Europe and North America, and a shift away from domestic heating with wood during the 19th century. The results highlight the value of bryophytes as bioindicators to measure PAH atmospheric pollution as well as the unrealized potential of herbaria as historical records of environmental change.
