Buffalobur (Solanum rostratum Dunal) is an invasive species that seriously endangers crop production and the ecological environment. Seeds are the primary source of infestation; therefore, understanding the molecular basis of buffalobur seed dormancy, and germination is crucial for precision weed management. In this study, high-throughput RNA-Seq was performed on buffalobur seeds, which imbibed under 0.35 mmol/L giberellic acid (GA) and 0.35 mmol/L abscisic acid (ABA). In total, 3658 differentially expressed genes (DEGs) were identified during seed germination. Gene annotation revealed that the DEGs were significantly enriched during the protein metabolic process, as well as the macromolecular complex and cytoplasmic part for ABA versus GA. Pathway analysis predicted that the DEGs were associated with metabolic pathways, the biosynthesis of secondary metabolites and ribosome. Nine germination-related genes involved in the biosynthesis and metabolism of the phytohormones and encoding of the endo-β-mannanase (EBM) were identified. Gene expression indicated that GA upregulated GA3OX1 and MAN2 expression to increase the EBM activity, which caused the endosperm cap to weaken and lowered the puncture force to trigger the germination of buffalobur. The obtained results would be helpful to clarify the regulation of seed dormancy and the germination of buffalobur, and could serve as a valuable resource when unravelling the genetic basis of seed biology of this weed species. The green foxtail, Setaria viridis (L.) P. Beauv. (Poales: Poaceae), is a troublesome and widespread grass weed in China. The acetolactate synthase (ALS)-inhibiting herbicide nicosulfuron has been intensively used to manage S. viridis, and this has substantially increased the selection pressure. Here we confirmed a 35.8-fold resistance to nicosulfuron in an S. viridis population (R376 population) from China and characterized the resistance mechanism. Molecular analyses revealed an Asp-376-Glu mutation of the ALS gene in the R376 population. The participation of metabolic resistance in the R376 population was proved by cytochrome P450 monooxygenases (P450) inhibitor pre-treatment and metabolism experiments. To further elucidate the mechanism of metabolic resistance, eighteen genes that could be related to the metabolism of nicosulfuron were obtained bythe RNA sequencing. The results of quantitative real-time PCR validation indicated that three ATP-binding cassette (ABC) transporters (ABE2, ABC15, and ABC15-2), four P450 (C76C2, CYOS, C78A5, and C81Q32), and two UDP-glucosyltransferase (UGT) (UGT13248 and UGT73C3), and one glutathione S-transferases (GST) (GST3) were the major candidates that contributed to metabolic nicosulfuron resistance in S. viridis. However, the specific role of these ten genes in metabolic resistance requires more research. Solanum nigrum is a noxious weed in agricultural ecosystem that limits many crops' production. The aim of the present study was to understand the level of genetic diversity and population structure of S. nigrum in China. A specific-locus amplified fragment (SLAF) sequencing method was conducted to detect single-nucleotide polymorphisms (SNPs) in the genomes of S. nigrum from 66 populations in China. A total of 616,533 high-quality SNPs were identified from 189,840 SLAFs, with an average sequencing depth of 10.59× fold and a Q30 value of 93% and a GC content of 42.78%. It showed a considerable amount of genetic diversity and genetic variability of S. nigrum among samples. The genetic differentiation of S. nigrum indicated that there was a low level of genetic differentiation (Fst < 0.1000) among geographical populations. A cluster analysis showed that populations of S. nigrum were divided into two subgroups, with some samples from adjacent position roughly clustered together, which showed some correlation between geographic origins. A population structure analysis suggested the 66 S. nigrum samples could have originated from three different genetic clusters. The Xinjiang site was the only location where all genetic clusters were found, which suggested these populations were genetically diverse. These results showed that there was a high degree of genetic diversity and low difference among the different geographical populations of S. nigrum. The results from the genetic structure of the SNP markers indicated that wide genetic variability exists among the population of S. nigrum in China, which may contribute to the adaptation and infestation of this weed species. Sodium chloride (NaCl), as a eustressor, can trigger relevant pathways to cause plants to produce a series of metabolites, thus improving the quality of crops to a certain extent. However, there are few reports on the improvement of nutrient quality and flavor of hydroponic Chinese chives (Allium tuberosum Rottler) by sodium chloride. In this study, five NaCl concentrations were used to investigate the dose-dependent effects on growth, nutritional quality and flavor in Chinese chives. The results show that 10 mM NaCl had no significant effect on the growth of Chinese chives, but significantly decreased the nitrate content by 40% compared with 0 mM NaCl treatment, and the content of soluble protein and vitamin C was increased by 3.6% and 2.1%, respectively. In addition, a total of 75 volatile compounds were identified among five treatments using headspace solid-phase microextraction gas chromatography/mass spectrometry (HS-SPME/GC-MS). Compared with the 0 mM NaCl treatment, 10 mM NaCl had the greatest effect on the quantity and content of volatile compounds, with the total content increased by 27.8%. Furthermore, according to the odor activity values (OAVs) and odor description, there were 14 major aroma-active compounds (OAVs > 1) in Chinese chives. The "garlic and onion" odor was the strongest among the eight categories of aromas, and its highest value was observed in the 10 mM NaCl treatment (OAVs = 794).Taken together, adding 10 mM NaCl to the nutrient solution could improve the nutritional quality and flavor of Chinese chives without affecting their normal growth. Orobanche cumana Wallr. is the most serious parasitic weed that threatens sunflower production in China, it infects sunflower roots and causes severe yield and economic losses. This study determined the effects of O. cumana infection on sunflower growth, physiological, biochemical, photosynthesis indexes and used RNA-Seq to investigate the potential regulatory factors involved in the parasitic interaction. Results showed that O. cumana infestation significantly inhibited sunflower height, fresh weight, chlorophyl contents, photosynthetic and the MDA content, SOD and POD activities were increased in different degrees, which might be related to sunflower resistance to O. cumana infestation. Additionally, six parasite-related genes were selected, which markedly enriched in plant hormone signal transduction, photorespiration and phenylpropanoid metabolism pathway. Among these genes, HsP90A, MYC2 and HAO were discovered for the first time in sunflowers and O. cumana parasitic interaction. Main conclusion: NO enhances the resistance of tomato seedlings to salt stress through protein S-nitrosylation and transcriptional regulation, which involves the regulation of MAPK signaling and carbohydrate metabolism. Nitric oxide (NO) regulates various physiological and biochemical processes and stress responses in plants. We found that S-nitrosoglutathione (GSNO) treatment significantly promoted the growth of tomato seedling under NaCl stress, indicating that NO plays a positive role in salt stress resistance. Moreover, GSNO pretreatment resulted in an increase of endogenous NO level, S-nitrosothiol (SNO) content, S-nitrosoglutathione reductase (GSNOR) activity and GSNOR expression under salt stress, implicating that S-nitrosylation might be involved in NO-alleviating salt stress. To further explore whether S-nitrosylation is a key molecular mechanism of NO-alleviating salt stress, the biotin-switch technique and liquid chromatography/mass spectrometry/mass spectrometry (LC–MS/MS) were conducted. A total of 1054 putative S-nitrosylated proteins have been identified, which were mainly enriched in chloroplast, cytoplasm and mitochondrion. Among them, 15 and 22 S-nitrosylated proteins were involved in mitogen-activated protein kinase (MAPK) signal transduction and carbohydrate metabolism, respectively. In MAPK signaling, various S-nitrosylated proteins, SAM1, SAM3, SAM, PP2C and SnRK, were down-regulated and MAPK, MAPKK and MAPKK5 were up-regulated at the transcriptional level by GSNO treatment under salt stress compared to NaCl treatment alone. The GSNO pretreatment could reduce ethylene production and ABA content under NaCl stress. In addition, the activities of enzyme identified in carbohydrate metabolism, their expression at the transcriptional level and the metabolite content were up-regulated by GSNO supplication under salt stress, resulting in the activation of glycolysis and tricarboxylic acid cycle (TCA) cycles. Thus, these results demonstrated that NO might beneficially regulate MAPK signaling at transcriptional levels and activate carbohydrate metabolism at the post-translational and transcriptional level, protecting seedlings from energy deficiency and salinity, thereby alleviating salt stress-induced damage in tomato seedlings. Silene conoidea L. is an annual troublesome broadleaf weed in winter wheat fields in China. In recent years, field applications of tribenuron-methyl have been ineffective in controlling S. conoidea in Hebei Province, China. The aim of this study was to determine the molecular basis of tribenuron-methyl resistance in S. conoidea. Whole-plant response assays revealed that the resistant population (R) exhibited a higher level of resistance (382.3-fold) to tribenuron-methyl. The R population also showed high cross-resistance to other acetolactate synthase (ALS) inhibitors, including imazethapyr, bispyribac-sodium and florasulam. However, the R population could be controlled by the field-recommended rates of bentazone, MCPA, fluroxypyr, carfentrazone-ethyl and bromoxynil. In vitro ALS activity assays indicated that the tribenuron-methyl I50 value for the R population was 18.5 times higher than those for the susceptible population (S). ALS gene sequencing revealed an amino acid mutation, Trp-574-Leu, in the R population. Pretreatment with the P450 inhibitor malathion indicated that the R population might have cytochrome P450-mediated metabolic resistance. These results suggest that the Trp-574-Leu mutation and P450-mediated enhanced metabolism coexist in S. conoidea to generate tribenuron-methyl resistance. The evolved acetolactate synthase (ALS) inhibiting herbicide-resistant redroot amaranth has been confirmed in China and caused a great loss in soybean production. This study was conducted to evaluate the growth and competitiveness of ALS-resistant (R) and ALS-susceptible (S) redroot amaranth biotypes. Seeds of both R and S biotypes were subjected to different temperature regimes. Data revealed that the germination percentage and seedling vigor of both biotypes did not differ largely from each other at 10/20 to 30/40 °C. Under noncompetitive conditions, there were no significant leaf number, plant height, or dry weight differences between the R and S biotypes. Moreover, replacement series experiment results indicated that the R and S biotypes have a similar competitive ability. This study shows that there are no significant differences in growth or competitiveness between the R and S redroot amaranth biotypes regarding the physiological characteristics evaluated. Therefore, the proportion and distribution of the R biotype will not be affected in the absence of the ALS-inhibiting herbicide. Black grass (Alopecurus myosuroides Huds.) is a highly competitive weed in winter wheat fields of China. Due to repeated use of acetolactate synthase (ALS) inhibitors, many A. myosuroides populations have evolved resistance to pyroxsulam in some wheat fields. Research was conducted to determine the molecular basis of herbicide resistance in the AH93 A. myosuroides population. Whole-plant dose–response assay confirmed that the AH93 population was resistant to pyroxsulam with a resistance index of 4.2. Cross- and multiple-resistance assays indicated that the AH93 population was cross-resistant to mesosulfuron-methyl and multiple-resistant to pinoxaden. Sequencing of the ALS and ACCase gene revealed that there was no target-site mutation in ALS, but Trp-2027-Cys and Cys-2088-Arg amino acid mutations in ACCase in the AH93 population. A malathion pretreatment study indicated that the AH93 population might have cytochrome P450–mediated herbicide metabolic resistance. This is the first report of pyroxsulam resistance in a multiple-resistant A. myosuroides population in China, and the Cys-2088-Arg mutation is the first reported case of an ACCase mutant conferring herbicide resistance in A. myosuroides. In crop fields, resistance to acetolactate synthase (ALS)-inhibiting herbicides found in many troublesome weed species, including Bromus japonicus Thunb, is a worldwide problem. In particular, the development of herbicide resistance in B. japonicus is a severe threat to wheat production in China. The purpose of this research was to investigate the physiological and molecular basis of B. japonicus resistance to flucarbazone-sodium. Dose-response analysis demonstrated that, compared with the susceptible B. japonicus (S) population, the resistant (R) population exhibited a 120-fold increase in flucarbazone-sodium resistance. Nucleotide sequence alignment of the ALS gene indicated that the Pro-197-Ser mutation in ALS was associated with resistance to flucarbazone-sodium in the R population. The results of a malathion pretreatment study showed that B. japonicus might also have remarkable cytochrome P450 monooxygenase (P450)-mediated metabolic resistance. Volatile compounds could affect the flavor and ornamental quality of cut flowers, but the flavor change occurring during the vase period of the cut flower is unclear. To clarify the dynamic changes during the vase period of cut lily (Lilium spp. 'Manissa') flowers, comprehensive flavor profiles were characterized by the electronic nose (E-nose) and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME/GC-MS). The response value of sensor W2W was significantly higher than other sensors, and its response value reached the highest on day 4. A total of 59 volatiles were detected in cut lilies by HS-SPME/GC-MS, mainly including aldehydes, alcohols, and esters. There were 19 volatiles with odor activity values (OAVs) greater than 1. Floral and fruity aromas were stronger, followed by a pungent scent. Principal component analysis (PCA) and hierarchical cluster analysis (HCA) could effectively discriminate lily samples derived from different vase times on the basis of E-nose and HS-SPME-GC-MS. In summary, our study investigates the flavor change profile and the diversity of volatile compounds during the vase period of cut lilies, and lilies on day 4 after harvest exhibited excellent aroma and flavor taking into consideration of the flavor intensity and diversity. This provided theoretical guidance for the assessment of scent volatiles and flavor quality during the vase period of cut lily flowers and will be helpful for the application of cut lilies during the postharvest process. Nitric oxide (NO), as a ubiquitous gas signaling molecule, modulates various physiological and biochemical processes and stress responses in plants. In our study, the NO donor nitrosoglutathione (GSNO) significantly promoted tomato seedling growth under NaCl stress, whereas NO scavenger 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide potassium (cPTIO) treatment reversed the positive effect of NO, indicating that NO plays an essential role in enhancing salt stress resistance. To explore the mechanism of NO-alleviated salt stress, the transcriptome of tomato leaves was analyzed. A total of 739 differentially expressed genes (DEGs) were identified and classified into different metabolic pathways, especially photosynthesis, plant hormone signal transduction, and carbon metabolism. Of these, approximately 16 and 9 DEGs involved in plant signal transduction and photosynthesis, respectively, were further studied. We found that GSNO increased the endogenous indoleacetic acid (IAA) and salicylic acid (SA) levels but decreased abscisic acid (ABA) and ethylene (ETH) levels under salt stress conditions. Additionally, GSNO induced increases in photosynthesis pigment content and chlorophyll fluorescence parameters under NaCl stress, thereby enhancing the photosynthetic capacity of tomato seedlings. Moreover, the effects of NO mentioned above were reversed by cPTIO. Together, the results of this study revealed that NO regulates the expression of genes related to phytohormone signal transduction and photosynthesis antenna proteins and, therefore, regulates endogenous hormonal equilibrium and enhances photosynthetic capacity, alleviating salt toxicity in tomato seedlings. In order to rapidly and precisely identify the volatile compounds in Chinese chive (Allium tuberosum Rottler), seven key parameters of headspace solid-phase micro-extraction conditions (HS-SPME) from Chinese chive were optimized. A total of 59 volatile compounds were identified by using the optimized method, including 28 ethers, 15 aldehydes, 6 alcohols, 5 ketones, 2 hydrocarbons, 1 ester, and 2 phenols. Ethers are the most abundant, especially dimethyl trisulfide (10,623.30 μg/kg). By calculating the odor activity values (OAVs), 11 volatile compounds were identified as the major aroma-active compounds of Chinese chive. From the analysis of the composition of Chinese chive aroma, the "garlic and onion" odor (OAV = 2361.09) showed an absolute predominance over the other 5 categories of aroma. The results of this study elucidated the main sources of Chinese chive aroma from a chemical point of view and provided the theoretical basis for improving the flavor quality of Chinese chive. BACKGROUND: It remains an open question if the long‐term application of single chemical herbicides would inevitably lead to increased weed populations and result in out‐of‐control weeds. The annual dynamics of weed seed bank responses to different weeding measures (chemical herbicide, hand weeding and no weeding) in rice–wheat cropping systems were compared to observe the succession of weed communities under different weed selection pressures for 17 years. RESULTS: In unweeded rice–wheat cropping plots, the initially dominant broadleaf weeds were overtaken by grasses and eventually by sedges, while in plots subjected to chemical herbicide or hand weeding, broadleaf weeds remained dominant followed by grasses. The rice–wheat cropping system favoured the spread of paddy weed species; weeding had little effect on the composition of the dominant rice weeds but greatly influenced that of wheat weeds. Total seed density tended to decrease in both weeded and unweeded plots, but the species density and composition of the seed banks differed among plots treated differently. Weeding slightly increased weed species diversity and decreased weed community evenness and dominance in the first several years, but this scenario could have negative consequences in the long term; however, without weeding, stronger interspecific competition led to a decrease in weed species diversity whereas weed community evenness and dominance increased. CONCLUSION: Long‐term and repeated application of pre‐emergence chemical herbicides and hand weeding had similar effects on the weed community dynamics, indicating that exclusive application of pre‐emergence herbicide could maintain the weed community at a durable relatively low infestation level. © 2019 Society of Chemical Industry [ABSTRACT FROM AUTHOR]

Johnsongrass ( Sorghum halepense [L.]Pers.), an exotic invasive weed in China, secretes the phenolic compounds, p-hydroxybenzoic acid ( p-HBA) and p-hydroxybenzaldehyde ( p-HBAL), as the dominant allelochemicals in the root exudates. To better understand how these two allelochemicals affect the soil microbial community in the rhizosphere of S. halepense, the fate of these compounds in the invaded soil and the effect of these phytotoxins on the soil bacterial community were evaluated. The concentrations of the allelochemicals in the soil were determined by a high-performance liquid chromatography-ultraviolet/photodiode array after 1, 2, 4, 6, 12 and 24 h of treatment. MiSeq sequencing was undertaken to understand how the bacterial populations in the soil were affected by the allelochemicals. The HPLC results indicated that p-HBA was degraded by the microorganisms that were present in the soil after 1 h and disappeared after 6 h of incubation. The compound, p-HBAL, initially was converted to p-HBA and then the p-HBA broke down, disappearing after 12 h of incubation in non-sterile soil. Both p-HBA and p-HBAL were stable under sterile soil conditions for up to 24 h. The relative abundance of Proteobacteria was significantly inhibited. However, those of Acidobacteria, Chloroflexi, Verrucomicrobia and Cyanobacteria were increased by the p-HBAL treatment. Glyphosate is an important non-selective herbicide that is in common use worldwide. However, evolved glyphosate-resistant ( GR) weeds significantly affect crop yields. Unfortunately, the mechanisms underlying resistance in GR weeds, such as goosegrass ( Eleusine indica (L.) Gaertn.), an annual weed found worldwide, have not been fully elucidated. In this study, transcriptome analysis was conducted to further assess the potential mechanisms of glyphosate resistance in goosegrass. The RNA sequencing libraries generated 24 597 462 clean reads. De novo assembly analysis produced 48 852 UniGenes with an average length of 847 bp. All UniGenes were annotated using seven databases. Sixteen candidate differentially expressed genes selected by digital gene expression analysis were validated by quantitative real-time PCR (qRT-PCR). Among these UniGenes, the EPSPS and PFK genes were constitutively up-regulated in resistant (R) individuals and showed a higher copy number than that in susceptible (S) individuals. The expressions of four UniGenes relevant to photosynthesis were inhibited by glyphosate in S individuals, and this toxic response was confirmed by gas exchange analysis. Two UniGenes annotated as glutathione transferase ( GST) were constitutively up-regulated in R individuals, and were induced by glyphosate both in R and S. In addition, the GST activities in R individuals were higher than in S. Our research confirmed that two UniGenes ( PFK, EPSPS) were strongly associated with target resistance, and two GST-annotated UniGenes may play a role in metabolic glyphosate resistance in goosegrass. The phenolic allelochemicals from the root exudates of Johnson grass (Sorghum halepense) were collected at different developmental stages and analyzed using high performance liquid chromatography-ultraviolet/photo-diode array. The results revealed that three simple phenolic components, p-hydroxybenzoic acid, p-hydroxybenzaldehyde and ethyl p-hydroxybenzoate, along with two flavonoids, diosmetin (3',5,7-trihydroxy-4'- methoxyflavone) and tricin (5,7,4'-trihydroxy-3',5'-dimethoxyflavone), were found in the root exudates of Johnson grass. However, the major phenolic compounds varied and the quality of each compound was dynamic at different developmental stages: the only phenolic compound that was detected in the seedling stage was p-hydroxybenzoic acid; three simple phenolic allelochemicals, p-hydroxybenzoic acid, p-hydroxybenzaldehyde and ethyl p-hydroxybenzoate, were found in the jointing stage; and five compounds, including the two flavonoids, tricin and diosmetin, appeared in the reproductive stage along with the other three simple phenolic allelochemicals. The highest diversity in phenolic constituents was in the reproductive stage, with five allelochemicals. Only p-hydroxybenzoic acid was found in the whole life cycle of Johnson grass, with a range of 2.9 nmol to 6.8 nmol per plant per day. p-Hydroxybenzaldehyde had the highest quality among all the compounds, with the highest releasing rate of 26.8 nmol per plant per day. The lowest-content compounds were the two flavonoids, with the highest quality of 1.8 nmol per plant per day. A 1142-bp upstream sequence (named CaEPSPS- P, GenBank accession number: KC107822) of the EPSPS gene from Convolvulus arvensis L was obtained by genome walking. The full-length sequence of the CaEPSPS- P and four deletion mutants were fused to the β-glucuronidase (GUS) gene and introduced into Arabidopsis via Agrobacterium-mediated transformation. Histochemical GUS staining of the transgenic plants showed that the CaEPSPS- P could drive GUS expression in the roots, stems, and leaves, but no visible GUS staining was detected in seeds. Further deletion analysis revealed two positive regulatory regions (−900 to −632 and −632 to −418) responsible for the basal activity of the EPSPS promoter. GUS activity assays indicated that GUS expression can be stimulated by light and glyphosate. In addition, a region between −632 and −400 was necessary for light-induced expression, while a region from −900 to −632 was necessary for glyphosate-induced GUS expression. These results suggested that the CaEPSPS- P was modulated by multiple cis-regulatory elements in distinct and complex patterns to regulate transgene expression. [ABSTRACT FROM AUTHOR]

Main conclusion: Field-evolved resistance of goosegrass to glyphosate is due to double or single mutation in EPSPS , or amplification of EPSPS leads to increased transcription and protein levels. Glyphosate has been used widely in the south of China. The high selection pressure from glyphosate use has led to the evolution of resistance to glyphosate in weeds. We investigated the molecular mechanisms of three recently discovered glyphosate-resistant Eleusine indica populations (R1, R2 and R3). The results showed that R1 and R2 had double Thr102Ile and Pro106Ser mutation and a single mutation of Pro106Leu in the 5-enolpyruvylshikimate-3-phosphate synthase ( EPSPS) gene, respectively. Escherichia coli containing the mutated EPSPS genes was tolerant to glyphosate. EPSPS activity in R1 and R2 plants was higher than in the sensitive plants. There was no amino acid substitution in EPSPS gene in R3. However, expression of EPSPS in R3 plants was higher than in glyphosate-susceptible (S) population (13.8-fold) after glyphosate treatment. EPSPS enzyme activity in both R3 and S plants was inhibited by glyphosate, while shikimate accumulation in R3 was significantly lower than for the S population. Further analysis revealed that the genome of R3 contained 28.3-fold more copies of the EPSPS gene than that of susceptible population. EPSPS expression was positively correlated with copy number of EPSPS. Glucosinolates (GLS) are important anionic secondary metabolites that are rich in thiocyanin in cabbage, Brassica oleracea L. var. capitata. GLS are important in food flavor, plant antimicrobial activity, insect resistance, disease resistance, and human anti-cancer effects. Sulfur is an important raw material of GLS, directly affecting their synthesis. However, the mechanism of sulfur regulation of GLS biosynthesis in cabbage is unclear. In the present study, cabbage was treated with sulfur-free Hoagland nutrient solution (control; −S), and normal Hoagland nutrient solution (treatment; +S). Through joint transcriptomic and proteomic analyses, the effect of exogenous S on GLS synthesis was explored. S application induced GLS accumulation; especially, indole glycosides. Transcriptome analysis showed that +S treatment correlated positively with differentially expressed genes and proteins involved in amino acid biosynthesis, carbon metabolism, and plant hormone signal transduction. Compared with −S treatment, the mRNA expression of GLS synthesis genes (CYP, GSTU, UGT, and FMO) and those encoding transcription factors (RLK, MYB, AP2, bHLH, AUX/IAA, and WRKY) were upregulated significantly in the +S group. Combined transcriptome and proteome analysis suggested that the main pathway influenced by S during GLS synthesis in cabbage is amino acid biosynthesis. The biological activities of the primary metabolites and secondary metabolites of 69 green cabbage varieties were tested. The LC-MS detection method was used to determine the content of 19 free amino acids (lysine, tryptophan, phenylalanine, methionine, threonine, isoleucine, leucine, valine, arginine, asparagine, glycine, proline, tyrosine, glutamine, alanine, aspartic acid, serine, and glutamate). The content of 10 polyphenols (chlorogenic acid, gallic acid, 4-coumaric acid, ferulic acid, gentisic acid, cymarin, erucic acid, benzoic acid, rutin, and kaempferol) was determined by the HPLC detection method. Considering the complexity of the data obtained, variance analysis, diversity analysis, correlation analysis, hierarchical cluster analysis (HCA), and principal component analysis (PCA) were used to process and correlate amino acid or polyphenol data, respectively. The results showed that there were significant differences between the different amino acids and polyphenols of the 69 cabbage varieties. The most abundant amino acids and polyphenols were Glu and rutin, respectively. Both amino acids and polyphenols had a high genetic diversity, and multiple groups of significant or extremely significant correlations. The 69 cabbage varieties were divided into two groups, according to 19 amino acid indexes, by PCA. Among them, seven varieties with high amino acid content all fell into the fourth quadrant. The HCA of amino acids also supports this view. Based on 10 polyphenols, the 69 cabbage varieties were divided into two groups by HCA. Based on 29 indexes of amino acids and polyphenols, 69 cabbage varieties were evaluated and ranked by PCA. Therefore, in this study, cabbage varieties were classified in accordance with the level of amino acids and polyphenols, which provided a theoretical basis for the genetic improvement of nutritional quality in cabbage. Aegilops tauschii Coss. is known as a noxious grass weed seriously affecting wheat quality and yield. To investigate its present occurrence in wheat fields and the potential genetic diversity of the grass weed in China, a filed survey covering major wheat production regions was conducted during 2017–2019. Seeds of different Ae. tauschii populations collected from the survey were analyzed with Simple Sequence Repeats (SSRs) technique. Results showed that Ae. tauschii was occurring in each of the provinces surveyed with varied occurrence frequency ranging from 0.91% in Sichuan Province to 92.85% in Henan Provinces. Eighty alleles with size ranging from 98 bp to 277 bp were detected from the 192 collected Ae. tauschii populations with 17 SSR markers. Ae. tauschii, in this study, exhibited a moderately high level of genetic diversity, high differentiation, deficient heterozygosity and limited gene flow. Compared with other provinces, Hubei populations possessed relatively low genetic diversity. Dendrogram analysis showed that genetic distance did not seem to be related to geographic distribution. Additionally, STRUCTURE analysis suggested that Ae. tauschii populations in wheat fields of China can be divided into three groups, which was further supported by cluster analysis. Among the three groups, solely 7% of the total variation was detected, whereas the majority variation (67%) occurred among different populations within same group. Undoubtedly, such information will help us to better understand population relationships and spread of Ae. tauschii in China and will provide a new perspective for its integrated management. Convolvulus arvensis is a troublesome weed that is naturally tolerant to glyphosate. This weed tolerates glyphosate at a rate 5.1 times higher than that of glyphosate-susceptible Calystegia hederacea. Glyphosate-treated C. arvensis plants accumulated less shikimic acid than C. hederacea plants. The overexpression of EPSPS genes from the two species in transgenic Arabidopsis thaliana resulted in similar glyphosate tolerance levels. qPCR of genomic DNA revealed that the EPSPS copy number in C. arvensis was approximately 2 times higher than that in C. hederacea. Moreover, glyphosate treatment caused a marked increase in EPSPS mRNA in C. arvensis compared to C. hederacea. GUS activity analysis showed that the promoter of CaEPSPS (CaEPSPS-P) highly improved GUS expression after glyphosate treatment, while no obvious differential GUS expression was observed in ChEPSPS-P transgenic A. thaliana in the presence or absence of glyphosate. Based on the obtained results, two coexisting mechanisms may explain the natural glyphosate tolerance in C. arvensis: (i) high EPSPS copy number and (ii) specific promoter-mediated overexpression of EPSPS after glyphosate treatment. Buffalobur (Solanum rostratum Dunal), which belongs to the Solanaceae family, is a worldwide noxious invasive weed and is listed as one of the top 10 alien invasive species in China. It is harmful to humans and livestock because the entire plant is covered with spines containing toxins. Many studies have analysed the gene expression in this weed species under different stress conditions using quantitative real-time PCR (qPCR). However, until now, there has been no report on suitable reference genes in buffalobur. Herein, 14 candidate reference genes were selected and evaluated for their expression stability in buffalobur in different tissues, at different developmental stages, and in response to several stress conditions using the geNorm, NormFinder, BestKeeper and RefFinder statistical algorithms. The results showed that EF1α, ACT and SAND are suitable reference genes across all samples tested. We recommend the normalization of target gene expression under different experimental conditions using these three genes together. Validation of selected reference genes was achieved by assessing the relative expression levels of P5CS and GI. Goosegrass (Eleusine indica) is one of the most serious annual grassy weeds worldwide, and its evolved herbicide-resistant populations are more difficult to control. Quantitative real-time PCR (qPCR) is a common technique for investigating the resistance mechanism; however, there is as yet no report on the systematic selection of stable reference genes for goosegrass. This study proposed to test the expression stability of 9 candidate reference genes in goosegrass in different tissues and developmental stages and under stress from three types of herbicide. The results show that for different developmental stages and organs (control), eukaryotic initiation factor 4 A (eIF-4) is the most stable reference gene. Chloroplast acetolactate synthase (ALS) is the most stable reference gene under glyphosate stress. Under glufosinate stress, eIF-4 is the best reference gene. Ubiquitin-conjugating enzyme (UCE) is the most stable reference gene under quizalofop-p-ethyl stress. The gene eIF-4 is the recommended reference gene for goosegrass under the stress of all three herbicides. Moreover, pairwise analysis showed that seven reference genes were sufficient to normalize the gene expression data under three herbicides treatment.
