Polygonum minus Huds. has massive potential to be commercialized following many current discoveries of its medicinal properties. We investigated the growth and physiology changes of P. minus under different shading and treatment durations. The effects of four shading levels: 0% (T1), 30% (T2), 50% (T3) and 70% (T4) and two treatment durations of 10 weeks and 20 weeks were studied. Significant effects of shading levels and treatment durations were observed on the transpiration rate (E), leaf resistance, pigment content in leaves (Chl a, Chl b and Car), carbohydrate, ascorbic acid, total anthocyanin, phenolic and flavonoid contents. Enzyme activity of superoxide dismutase and ascorbate peroxidase were also significantly affected by shading levels and treatment durations. The highest antioxidant activities were recorded under T4 of both treatment durations along with the high content of Chl a, Chl b and Car. The leaf, stem and root extracts of P. minus were also subjected to HPLC analysis for quantification of selected flavonoid compounds. Azadirachta indica or neem is recognized for its unique properties, both against insects and in improving human health. However, proteomics studies of this plant's response to abiotic stresses have yet to be reported. Application of this technique highly depends on efficient and optimized sample preparation, in deriving highly reproducible proteomics data. The present study highlights the comparison of protein extraction method which effectively recovers proteins from recalcitrant phenolic-rich tissues of neem callus. Four protocols based on TCA-acetone or/and phenol extraction methods were compared. The TCA-acetone complemented with PVPP method was the most effective approach and generated the greatest total protein yields, spot number, good resolution, and spot intensities. Subsequently, the differential proteomics performed revealed that proteins associated with oxidoreduction, energy, transcriptional, stress response, respiration, and cell division were differentially regulated in salinity stress treatment. Meanwhile, the expression of proteins classified in the group of energy, photosynthesis, protein synthesis, oxidoreduction, and post-transcriptional regulation was altered in the JA-stressed callus. The accumulation of AZA was also noticeably reduced in salinity- and JA-treated callus, suggesting these tested stress parameters were not effective in improving AZA production in the neem callus. Overall, this analysis provides valuable information on the application of plant tissue culture as an alternative for sustainable production of major bioactive constituents in A. indica. By 2050, the global population is projected to exceed 9.5 billion, posing a formidable challenge to ensure food security worldwide. To address this pressing issue, mutation breeding in horticultural crops, utilizing physical or chemical methods, has emerged as a promising biotechnological strategy. However, the efficacy of these mutagens can be influenced by various factors, including biological and environmental variables, as well as targeted plant materials. This review highlights the global challenges related to food security and explores the potential of mutation breeding as an indispensable biotechnological tool in overcoming food insecurity. This review also covers the emergence of CRISPR-Cas9, a breakthrough technology offering precise genome editing for the development of high-yield, stress-tolerant crops. Together, mutation breeding and CRISPR can potentially address future food demands. Colorectal cancer (CRC) is the most common cancer in both men and women and is associated with increased telomerase levels and activity. The potential downstream effects of TERT and/or TERC downregulation by berberine (a telomerase inhibitor) or RNA interference (RNAi) on various target RNAs, proteins, relative telomerase activity (RTA), relative telomere length (RTL), hydrogen peroxide concentration [H2O2], percentage of cell cycle distribution, cell size and granularity as well as cellular metabolites were explored in HCT 116 cell line. Knockdown of TERT decreased TERC. The downregulation of TERT and/or TERC caused increment of [H2O2], G0/G1 phase arrest in addition to decreased S and G2/M phases, as well as diminished cell size. RTL was later reduced as a result of TERT, TERT and/or TERC downregulation which decreased RTA. It was discovered that xanthine oxidase (XO) was significantly and positively correlated at FDR-adjusted p value < 0.05 with RTA, TERT, TERT, TERC, and RTL. HCT 116 with decreased RTA was closely clustered in the Principal Component Analysis (PCA) indicating similarity of the metabolic profile. A total of 55 metabolites were putatively annotated in this study, potentially associated with RTA levels. The Debiased Sparse Partial Correlation (DSPC) Network revealed that RTA was directly correlated to TERT. There were 4 metabolic pathways significantly affected by low level of RTA which include (1) purine metabolism, (2) glycine, serine, and threonine metabolism, (3) glyoxylate and dicarboxylate metabolism, and (4) aminoacyl-tRNA biosynthesis. The Gene-Metabolite Interaction Network implied that reduced RTA level was related to the mechanism of oxidative stress. This study reveals the linkages between RTA to various selected RNAs, proteins, metabolites, oxidative stress mechanism and subsequently phenotypic changes in HCT 116 which is valuable to understand the intricate biological interactions and mechanism of telomerase in CRC. Soil salinization is a major problem affecting agriculture globally. One approach to restore saline soil is through the application of biochar which is great at adsorbing salt ions and thus amending salt-affected soils. Biochar produced using forestry wastes (Gigantochloa levis and Melaleuca cajuputi) were pyrolyzed at 300, 450 and 600 ℃ for 2 h. Proximate and ultimate analyses were done followed with physical and chemical analyses of the biochar and their raw materials before undergoing the sodium adsorption study. The fixed carbon was observed to increase from 9.636 to 72.919% (G. levis) and 9.473–69.950% (M. cajuputi) upon carbonization of the raw materials. These values correlated with the increasing C content of the samples, which also resulted in lower H/C value especially for G-600 and M-600 samples. It was found that biochar aromaticity and stability increased with increasing temperature. In addition, field emission scanning electron microscopy (FESEM) elucidated that higher pyrolysis temperature increased the porosity and surface area. The pH observed to increase from 5.36 to 7.97 (G. levis) and 4.19–9.05 (M. cajuputi) following higher pyrolysis temperature. Green synthesis of NPs is preferred due to its eco-friendly procedures and non-toxic end products. However, unintentional release of NPs can lead to environmental pollution affecting living organisms including plants. NPs accumulation in soil can affect the agricultural sustainability and crop production. In this context, we report the morphological and biochemical response of spinach nanoprimed with MgO–NPs at concentrations, 10, 50, 100, and 150 µg/ml. Nanopriming reduced the spinach root length by 14–26%, as a result a reduction of 20–74% in the length of spinach shoots was observed. The decreased spinach shoot length inhibited the chlorophyll accumulation by 21–55%, thus reducing the accumulation of carbohydrates and yield by 46 and 49%, respectively. The reduced utilization of the total absorbed light further enhanced ROS generation and oxidative stress by 32%, thus significantly altering their antioxidant system. Additionally, a significant variation in the accumulation of flavonoid pathway downstream metabolites myricitin, rutin, kaempferol-3 glycoside, and quercitin was also revealed on MgO-NPs nanopriming. Additionally, NPs enhanced the protein levels of spinach probably as an osmoprotectant to regulate the oxidative stress. However, increased protein precipitable tannins and enhanced oxidative stress reduced the protein digestibility and solubility. Overall, MgO-NPs mediated oxidative stress negatively affected the growth, development, and yield of spinach in fields in a concentration dependent manner. [ABSTRACT FROM AUTHOR]

A growing world population means greater pressure on earth's resources. Currently, 30% of food is wasted, which poses a significant risk to both humans and the environment. One way to offset the growth in food waste (FW) is through the process of microbial bioconversion, whereby FW is transformed into a range of nutrient-dense biofertilisers. This approach not only promotes a highly desirable circular economy, but it can also reduce the use of inorganic fertilisers, which adversely impact the environment through increased greenhouse gases, changes in soil and water characteristics, and loss of biodiversity. The bioconversion of FW to biofertiliser relies on the processes of aerobic (composting) and anaerobic digestion. Recently, alternative decomposition techniques included growing specific beneficial microbes, such as effective microorganisms, to speed up the breakdown process. Microorganisms can act as biostimulants and biodecomposers, possessing nutrient-fixing abilities and providing protection from biotic and abiotic stresses, thus enhancing plant growth and overall health. Due to their sessile nature, plants are exposed to various environmental stressors such as exposure to high levels of harmful ultraviolet (UV), ionizing, and non-ionizing radiations. This exposure may result in various damages, ranging from DNA and chromosomal aberrations to phenotypic abnormalities. As an adaptation, plants have evolved efficient DNA repair mechanisms to detect and repair any damage caused by exposure to these harmful stressors to ensure their survival. In this study, the effects of gamma radiation (as a source of ionizing radiation) on clonal Ananas comosus var. MD2 was evaluated. The morphology and physiology of the clonal plantlets before and after exposure to gamma radiation were monitored at specific time intervals. The degree of genetic variation between the samples pre- and post-irradiation was also analyzed by using inter-simple sequence repeat (ISSR) markers. The resulting data revealed that the heights of the irradiated plantlets were significantly reduced (compared to control), but improved with the recovery period. Irradiated samples also exhibited relatively good photosynthetic efficiency that further improved as the plantlets recover. These observations were supported by the ISSR analysis, where the genetic dissimilarities between the irradiated samples and control were reduced by 0.1017, after 4 weeks of recovery. Overall, our findings suggested that the phenotype recovery of the clonal A. comosus var. MD2 plantlets was contributed by their ability to detect and repair the DNA lesions (as exemplified by the reduction in genetic dissimilarity after 4 weeks) and hence allow the plantlets to undergo phenotype reversion to normal plant stature. Purpose The purpose of this study is to evaluate the pattern of carotenoids distribution in three underutilized Malaysian 'ulam' or traditional vegetables in Malaysia (Averrhoa carambola, Manihot esculenta and Ipomoea batatas) and their valuable pro-vitamin A activities. These assessments will yield valuable knowledge and insight into the importance of these underutilized traditional vegetables and highlight their potential for applications in medicinal and functional colorant industries.Design/methodology/approach In this study, the authors have evaluated the distribution of carotenoid compounds in aerial organs of three underutilized traditional vegetables (Averrhoa carambola, Manihot esculenta and Ipomoea batatas). The content of the individual carotenoids were quantified using high-performance liquid chromatography (HPLC) and the organ with the highest accumulation of these compounds were identified. Their valuable pro-vitamin A activities were also reported to indicate their medicinal potential which can further be exploited as pharmacologically active natural colorants and in other applications.Findings In total, three major chromatographic peaks corresponding to lutein, violaxanthin and β-carotene were observed through HPLC. Among the compounds detected, lutein and β-carotene were the most abundant carotenoids found in both shoots and petioles of all three species. Violaxanthin was only detected in I. batatas shoots. Overall, carotenoid content was observed to be higher in the shoots than in the petioles, where I. batatas contained the highest amount of total carotenoid, followed by M. esculenta and A. carambola. The opposite trend was observed in the petioles, where A. carambola petioles had the highest carotenoid content, while I. batatas contained the least.Research limitations/implications The distribution and abundance of these individual carotenoids suggested that the petioles contained the highest amount of carotenoid, contributing to its high pro-vitamin A activity, and could be potentially useful for medicinal application, as it can act as storage site that is not as prone to natural drying or degradation during harvest and sample storage. Future research work should include improvements in the extraction and purification procedures as well as robust identification methods which may lead to better detection and identification of other compounds that could attribute to its bioactivity, to complement the findings of the current study.Practical implications This analysis provides valuable information on the importance of underutilized traditional vegetables as important biofactories for sustainable production of valuable pigments (such as carotenoids) with medicinal benefits and can further be exploited in various industries, such as in formulation of functional natural colorants. This study also highlights the importance of petiole as a storage site of pharmacologically active compounds that is not as prone to natural drying or degradation during harvest and sample storage.Originality/value To date, there is no previous report found on comparative analysis of carotenoid content and quantification of individual carotenoid concentration in the edible aerial parts of Averrhoa carambola, Manihot esculenta and Ipomoea batatas, although they have been traditionally consumed as "ulam" in Malaysia. Purpose The purpose of this study is to evaluate the content of bioactive pigments in coloured callus of Azadirachta indica and to understand the correlation between the callus colours with their bioactive constituents, antioxidant properties and cytotoxicity. These assessments will yield valuable insight into the use of in vitro-derived pigments for possible use as functional natural colourants.Design/methodology/approach In this study, the authors have successfully developed a protocol to produce leaf-derived callus of various colours with enhanced content of bioactive pigments in A. indica through plant tissue culture. Comparative analysis of the pigments content (chlorophyll, carotenoid, phenolics and anthocyanins) in the coloured callus was conducted, followed by evaluation of its bioactive properties. The antioxidant properties against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, ferric reducing antioxidant power and cytotox activities of the coloured callus extracts were also reported.Findings Callus of various colours were successfully produced in A. indica through plant tissue culture, and their valuable pigment content and bioactivity were evaluated. The green callus contained the highest amount of anthocyanin, followed by brown and cream callus. The total anthocyanin contents in both the green and brown callus was more than two-fold higher than that in cream callus. Contrasting observation was obtained for total phenolic content (TPC), where the TPC of cream callus was significantly higher than that in brown callus. Nevertheless, the green callus also exhibited the highest TPC. Green callus also contained the highest amount of total chlorophyll and carotenoid, as well as exhibited the highest antioxidant potential, and was found to be the only extract with active cytotox activity against SKOV-3 cells. Correlation analysis revealed that the excellent bioactivity exhibited by the coloured extracts was strongly correlated with the bioactive pigments present in the callus.Research limitations/implications The major bioactive compounds identified in the methanolic extracts of A. indica coloured callus are anthocyanins, phenolics, chlorophylls and carotenoids. Vermicompost is the product of composting or breaking down organic matter through the use of earthworms. It is rich in essential plant nutrients, and has the ability to enhance the condition and increase the quality of the soil. In the present study, a field trial was conducted from January 2015 until March 2016 to elucidate the effects of vermicompost application (compared to supplementation with chemical fertilizer and no fertilizer) on the sandy loam soil and plant nutrients of pineapple (Ananas comosus var. MD2). The morphophysiology and yield performance of MD2 pineapple grown with vermicompost in the field were also evaluated. In this study, vermicompost was applied onto the sandy loam soils during transplanting, followed by a second application at seven MAP (months after planting) at the rate of 10 t ha-1. On the other hand, the chemical fertilizer was applied based on the normal conventional cultivation practice. The soil and D-leaf samples at six MAP (S1) and during the red bud stage (S2; 10 MAP) were used to determine the soil and plant nutrient contents. The morphology of the plants was evaluated every month, and the fruits that were produced were subjected to quality analysis. Data analysis revealed that soil pH was increased after a second supplementation of vermicompost and contained significantly higher total N (0.15%) in the soils compared to the control (0.07%). There was no significant difference between plants supplied with chemical fertilizer and vermicompost in terms of plant height, number of leaves, or the length and width of D-leaves. However, different fertilization treatments were found to affect the yield and physical characteristics of the resulting fruits. Plants supplied with chemical fertilizer produced the highest fruit yield (136.97 t ha-1) with the largest fruit size, followed by vermicompost (121.39 t ha-10) and the control (94.93 t ha-1). However, fruits supplied with vermicompost were observed to have the smallest crowns. Taken together, these results indicated that the use of vermicompost produced pineapple plants with excellent growth performance, comparable to that obtained when chemical fertilizer was used. Also, based on the cost analysis conducted, it was shown that the total cost (fertilizer and labor) for plants grown with vermicompost was lower than plants grown with chemical fertilizer. However, the usage of vermicompost as the single source of nutrients is not suggested for this type of soil and field conditions, but can be used as a supplement to maintain the soil quality and ensure agricultural sustainability. Purpose The purpose of the present study is to understand the role of auxin and cytokinin in stimulating the production of pigmented callus in Orthosiphon stamineus and to gain correlation between the callus colours with their antioxidant capacity and bioactive constituents.Design/methodology/approach In this study, plant tissue culture was used to induce production of callus of various colours from leaf explants of O. stamineus, via manipulation of plant hormones (0-2.0 mg L−1 indole-3-acetic acid [IAA] and Kinetin [Kin]). The coloured callus was subjected to solvent extraction and used for quantification of its carotenoid, chlorophyll, anthocyanin and phenolic contents. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity of the extracts was also evaluated, before and after four weeks of storage at −20°C.Findings The highest mean (per cent) explants that produced roots (93.33 ± 0.05 per cent) were observed when the cultures were supplemented with 2.0 mg L−1 IAA. The colour of the callus changed with time, from green to cream to brown after two and four months of culture, respectively. Optimum production of green callus was achieved with addition of 2.0 mg L−1 Kin plus 1.0-2.0 mg L−1 IAA to the media, while cream callus in 0.5 mg L−1 Kin plus 2.0 mg L−1 IAA and brown callus in 0.5 mg L−1 Kin plus 1.5 mg L−1 IAA. Green callus was found to contain the highest amount of chlorophylls, carotenoid and anthocyanin, while cream callus contained the highest amount of phenolic compounds. The amount of pigments and secondary metabolites in the callus extracts decreased after four weeks of storage, except anthocyanin. The antioxidant potential of the extracts also increased after storage.Research limitations/implications The major compounds identified in the methanolic extracts of O. stamineus-coloured callus are chlorophylls, carotenoids, flavonoids and phenolic acids. Somaclonal and phenotypic variation caused by genetic and/or epigenetic modifications, are a valuable source of genetic variation to improve desirable polygenetic traits in crops. In this study, we induced somaclonal variation in vitro pineapple (Ananas comosus var. MD2) through hormonal induction, NaCl, and abscisic acid (ABA) supplementation. Our results showed that supplementation of high concentration of 6-benzylaminopurine (4.0 mg/L BAP) alone or combined with indole-butyric acid (IBA) produced the highest percentage of dwarf variants (100%). Murashige and Skoog (MS) media containing 4.0 mg/L BAP plus 2.0 mg/L IBA produced the shortest plantlets (1.9 ± 0.1 cm). In comparison, MS media containing 1.0% NaCl induced formation of dwarf plantlets with a mean plantlet height of 1.4 ± 0.3 cm, whereas 1.0 mg/L ABA generated plantlets with a mean plantlet height of 1.7 ± 0.1 cm. We then analyzed the histone deacetylase (HDAC) enzyme activity for dwarf and non-dwarf plantlets. In general, dwarf plantlets exhibited higher HDAC activity than non-dwarf plantlets. The highest HDAC activity (109, 333.33 ± 4.40 ng/min/mg) was recorded for dwarf plantlets grown on media supplemented with 1.0 mg/L ABA. The dwarf variants also underwent phenotypic recovery to normal phenotype within 8 months after transferred to MS basal media. No ploidy alteration was detected in these dwarf plantlets after analyzed by flow cytometry. Taken together, although the generated dwarf plantlets showed higher HDAC activity compared to non-dwarf plantlets, their capability of reverting to non-dwarf phenotype suggested that it might be due to epigenetic modulation. Phoenix dactylifera or date palm fruits are reported to contain natural compounds that exhibit antioxidant and antibacterial properties. This research aimed to study the effect of fruit chilling at 4 °C for 8 weeks, extract storage at 20 °C for 5 weeks, and extraction solvents (methanol or acetone) on total phenolic content (TPC), antioxidant activity and antibacterial properties of Saudi Arabian P. dactylifera cv Mabroom, Safawi and Ajwa, as well as Iranian P. dactylifera cv Mariami. The storage stability of total anthocyanin content (TAC) was also evaluated, before and after storing the extracts at 20 °C and 4 °C respectively, for 5 weeks. Mariami had the highest TAC (3.18 ± 1.40 mg cyd 3-glu/100 g DW) while Mabroom had the lowest TAC (0.54 ± 0.15 mg cyd 3-glu/100 g DW). The TAC of all extracts increased after storage. The chilling of date palm fruits for 8 weeks prior to solvent extraction elevated the TPC of all date fruit extracts, except for methanolic extracts of Mabroom and Mariami. All IC50 values of all cultivars decreased after the fruit chilling treatment. Methanol was a better solvent compared to acetone for the extraction of phenolic compounds in dates. The TPC of all cultivars extracts decreased after 5 weeks of extract storage. IC50 values of all cultivars extracts increased after extract storage except for the methanolic extracts of Safawi and Ajwa. Different cultivars exhibited different antibacterial properties. Only the methanolic extract of Ajwa exhibited antibacterial activity against all four bacteria tested: Staphylococcus aureus, Bacillus cereus, Serratia marcescens and Escherichia coli. The leaf of Gardenia jasminoides Ellis was used as explants and was cultured on MS and WPM media supplemented with various concentrations of NAA, IAA, 2,4-D, IBA, TDZ, and Kn (0 to 5mg L-1 with 0.5 increment). After six months, the higher percentage of callus (100%) and the best dry and fresh weight of callus were formed onWPMmedium supplemented with 2,4-D andNAA (2.0-3.0mg L-1) and this amount was decreased from (84%) to (69%) when this media supplemented with Kinetin and TDZ (1mg L-1) respectively were used. Leaf segments cultured on WPM media added with Kn (1mg L-1) and TDZ (2mgL-1) yielded the least amount of callus. It was found that WPM media added with IAA (4.5-5.0mg L-1) were optimum for root induction from G. jasminoides plantlets. Antibacterial screening of leaf extracts (in vivo) showed no inhibitory effect against E. coli, P. aeruginosa, S. aureus, and B. cereus, in contrast to callus extracts from leaf cultures supplemented with NAA, which showed inhibition activity against E. coli and B. cereus. Various explants (stem, leaf, and root) of Citrus assamensis were cultured on MS media supplemented with various combinations and concentrations (0.5-2.0 mgL-1) of NAA and BAP. Optimum shoot and root regeneration were obtained from stem cultures supplemented with 1.5 mgL-1 NAA and 2.0 mgL-1 BAP, respectively. Explant type affects the success of tissue culture of this species, whereby stem explants were observed to be the most responsive. Addition of 30 gL-1 sucrose and pH of 5.8 was most optimum for in vitro regeneration of this species. Photoperiod of 16 hours of light and 8 hours of darkness was most optimum for shoot regeneration, but photoperiod of 24 hours of darkness was beneficial for production of callus. The morphology (macro and micro) and anatomy of in vivo and in vitro/ex vitro Citrus assamensis were also observed to elucidate any irregularities (or somaclonal variation) that may arise due to tissue culture protocols. Orthosiphon stamineus is a well-known medicinal herb that has long been used as a traditional treatment for a variety of ailments and disorders. In this study, in vitro grown O. stamineus plantlets were subjected to various stress factors to elucidate the effects of these stresses on the occurrence of somaclonal variation in this species. Murashige and Skoog (MS) media supplemented with 2.0 mg/L kinetin plus 0.5 mg/L IAA was identified as the optimum in vitro regeneration media (OM). The data analysis confirmed the clonal nature of the resulting plantlets. The plantlets were also exposed to various stress factors such as high salinity, abscisic acid (ABA), and high cytokinin concentrations, by subculturing the plantlets on OM added with various concentrations of NaCl, ABA, and high kinetin levels. Exposure to the various stress factors was observed to result in the occurrence of somaclonal variation in O. stamineus, where the incidence of hyperhydricity was found to be the most prominent. An ISSR analysis revealed 70.12% polymorphism among the variants. The intensive amount of chemical usage in agricultural practices could contribute to a significant impact on food safety issues and environmental health. Over-usage of chemical fertilisers may alter soil characteristics and contaminate water sources, leading to several human and animal health issues. Recently, there have been efforts to use microbial biofertilisers as a more sustainable and environmentally friendly agricultural practice in the common household of Southeast Asia. Traditionally, this method tends to utilise leftover food materials and readily available bacterial cultures, such as yoghurt drinks, and ferment them under a specific period in either solid or liquid form. So far, most of the testimonial-based feedbacks from local communities have been positive, but only limited information is available in the literature regarding the usage of biofertiliser fermented food (BFF). Previously, raw food waste has been used in the agriculture system to promote plant growth, however, the functional role of fermented food in enhancing plant growth have yet to be discovered. An understanding of the symbiotic relationship between fermented food and plants could be exploited to improve agricultural plant production more sustainably. Fermented food is known to be rich in good microbial flora (especially lactic acid bacteria (LAB)). LAB exist in different sources of fermented food and can act as a plant growth-promoting agent, improving the nutrient availability of food waste and other organic materials. Therefore, in this review, the potential use of seafood-based, plant-based, and animal-based fermented food as biofertiliser, especially from Southeast Asia, will be discussed based on their types and microbial and nutritional contents. The different types of fermented food provide a wide range of microbial flora for the enrichment of proteins, amino acids, vitamins, and minerals content in enhancing plant growth and overall development of the plant. Amelioration of soil acidity can boost soil fertility, hence increasing nutrient uptake, secondary metabolite, and its antioxidant potential. In the present study, the effectiveness of food waste compost and palm kernel biochar was assessed as soil amendments for Melastoma malabathricum L. grown in acidic soil conditions. A six-month greenhouse study was conducted using completely randomized design (CRD) with three treatment groups, including control plants (T1), plants amended with palm kernel biochar (T2), and plants amended with food waste compost (T3). Data analysis revealed that Melastoma malabathricum L. amended with T3 recorded the highest total chlorophyll content (433.678 ± 13.224 µg g−1 DW), followed by T2 and T1. The increase in chlorophyll content was contributed by the increase in soil pH. This was shown by the positive significant correlations between soil pH and chlorophyll a (r2 = 0.96; p ≤ 0.01) and chlorophyll b (r2 = 0.778; p ≤ 0.01). In addition, the same treatment exhibited the highest total anthocyanin content (leaves; 36.1 × 10−2 ± 0.034 mg/g DW and root extract; 8.9 × 10−2 ± 0.020 mg/g DW), total phenolic content (stem extract; 4930.956 ± 16.025 mg GAE/g DE), and total flavonoid content (stem extract; 209.984 ± 0.572 mg QE/g DE). Moreover, this study also found that the highest antioxidant potential against 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2-Azinobis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radicals was exhibited by samples supplemented with food waste compost (T3), followed by palm kernel biochar (T2). This indicates that the soil amendments have the capacity to enhance the secondary metabolites that protect plants, therefore ameliorating Melastoma malabathricum L.'s response towards acidic stress, and resulting in better antioxidant properties. Furthermore, this study also recorded better nutrient uptake in T3. With the significantly higher levels of macronutrient in the soil, the food waste compost could enhance the nutrient properties, secondary metabolites, and antioxidant capacity of Melastoma malabathricum L. grown in acidic soil conditions. The growing demand for high value aromatic herb Polygonum minus-based products have increased in recent years, for its antioxidant, anticancer, antimicrobial, and anti-inflammatory potentials. Although few reports have indicated the chemical profiles and antioxidative effects of Polygonum minus, no study has been conducted to assess the benefits of micro-environmental manipulation (different shading levels) on the growth, leaf gas exchange and secondary metabolites in Polygonum minus. Therefore, two shading levels (50%:T2 and 70%:T3) and one absolute control (0%:T1) were studied under eight weeks and 16 weeks of exposures on Polygonum minus after two weeks. It was found that P. minus under T2 obtained the highest photosynthesis rate (14.892 µmol CO2 m−2 s−1), followed by T3 = T1. The increase in photosynthesis rate was contributed by the enhancement of the leaf pigments content (chlorophyll a and chlorophyll b). This was shown by the positive significant correlations observed between photosynthesis rate with chlorophyll a (r2 = 0.536; p ≤ 0.05) and chlorophyll b (r2 = 0.540; p ≤ 0.05). As the shading levels and time interval increased, the production of total anthocyanin content (TAC) and antioxidant properties of Ferric Reducing Antioxidant Power (FRAP) and 2,2-Diphenyl-1-picrylhydrazyl (DPPH) also increased. The total phenolic content (TPC) and total flavonoid content (TFC) were also significantly enhanced under T2 and T3. The current study suggested that P.minus induce the production of more leaf pigments and secondary metabolites as their special adaptation mechanism under low light condition. Although the biomass was affected under low light, the purpose of conducting the study to boost the bioactive properties in Polygonum minus has been fulfilled by 50% shading under 16 weeks' exposure. Colorectal cancer (CRC) is the most common cancer among males and females, which is associated with the increment of telomerase level and activity. Some plant-derived compounds are telomerase inhibitors that have the potential to decrease telomerase activity and/or level in various cancer cell lines. Unfortunately, a deeper understanding of the effects of telomerase inhibitor compound(s) on CRC cells is still lacking. Therefore, in this study, the aspects of telomerase inhibitors on a CRC cell line (HCT 116) were investigated. Screening on HCT 116 at 48 h showed that berberine (10.30 ± 0.89 µg/mL) is the most effective (lowest IC50 value) telomerase inhibitor compared to boldine (37.87 ± 3.12 µg/mL) and silymarin (>200 µg/mL). Further analyses exhibited that berberine treatment caused G0/G1 phase arrest at 48 h due to high cyclin D1 (CCND1) and low cyclin-dependent kinase 4 (CDK4) protein and mRNA levels, simultaneous downregulation of human telomerase reverse transcriptase (TERT) mRNA and human telomerase RNA component (TERC) levels, as well as a decrease in the TERT protein level and telomerase activity. The effect of berberine treatment on the cell cycle was time dependent as it resulted in a delayed cell cycle and doubling time by 2.18-fold. Telomerase activity and level was significantly decreased, and telomere erosion followed suit. Vermicompost is a nutrient-rich organic waste produced from earthworms that is beneficial in enhancing the soil condition and has been reported to aid in improving the crop yield and quality. In the present study, a field trial was conducted using a randomized complete block design with four replicates to elucidate the effects of vermicompost application (compared to supplementation with chemical fertilizer and no fertilizer) on the productivity of ex vitro MD2 pineapple plants. Vermicompost was applied on the sandy loam soils at transplanting followed by a second application at 7 months after planting (MAP) at the rate of 10 t·ha−1, while chemical fertilizer was applied based on the recommended cultivation practice. Data analysis revealed that there was no significant difference between the plants treated with vermicompost and chemical fertilizer in terms of the plant height, number of leaves, length and width of D-leaves, stomatal density and stomatal size. However, the fruits produced with vermicompost amendment were smaller in size but contained higher total soluble solids, titratable acidity, total solids, ascorbic acid and total chlorophyll content compared to the fruits produced from plants supplied with chemical fertilizer. Based on the DPPH, ABTS and FRAP assays, the methanolic fruit extracts from the control plants showed the highest antioxidant potential, followed by those of plants treated with vermicompost and chemical fertilizer. On the other hand, the application of vermicompost reduced soil acidity and produced macro- and micronutrient contents (N, P, K, Mg, Ca, S, Fe, Zn, B and Al) in the soil and plants that were comparable to or higher than those produced by the chemical fertilizer treatment. However, some of the nutrient contents observed in all treatments were lower than the recommended range for pineapple plant growth, suggesting that vermicompost or chemical fertilizer should not be used alone as a source of nutrients for ex vitro MD2 pineapple plants under these soil and field conditions. For centuries, Azadirachta indica or neem has been utilized as a primary source of medicine due to its antimicrobial, larvacidal, antimalarial and antifungal properties. Recently, its potential as an effective biopesticide has garnered attention, especially towards efficient and continuous production of its bioactive compounds. The present study investigated the effect of the plant growth regulators (PGRs) thiadiazuron (TDZ) and 2,4-dichlorophenoxyacetic acid (2,4-D) on the induction of colored callus formation and subsequent accumulation of azadirachtin (AZA) in A. indica. An efficient protocol was established for micropropagation and colored callus production of this species, followed by quantification of AZA (a mixture of azadirachtin A and B) and its safety assessment. For induction of the callus, leaf and petiole explants obtained from a young growing neem plant were excised and cultured on Murashige and Skoog (MS) medium supplemented with TDZ (0.2–0.6 mg L−1) and 2,4-D (0.2–0.6 mg L−1), either applied singly or in combination. Callus was successfully induced from both explant types at different rates, where media with 0.6 mg L−1 of TDZ resulted in the highest fresh weight (3.38 ± 0.08 g). In general, media with a single hormone (particularly TDZ) was more effective in producing a high mass of callus compared to combined PGRs. A culture duration of six weeks resulted in the production of green, brown and cream colored callus. The highest callus weight and accumulation of AZA was recorded in green callus (214.53 ± 33.63 mg g−1 dry weight (DW)) induced using TDZ. On the other hand, small amounts of AZA were detected in both brown and cream callus. Further experimentation indicated that the green callus with the highest AZA was found to be non-toxic (LC50 at 4606 µg mL−1) to the zebrafish animal model. These results suggested that the addition of different PGRs during in vitro culture could prominently affect callus and secondary metabolite production and can further be manipulated as a sustainable method for the production of a natural and environmentally friendly pesticide. Orthosiphon stamineus has been widely used as traditional remedy for various illnesses and diseases, such as cardiovascular diseases and epileptic seizures. In this study, direct regeneration through nodal segment of this species was attempted using Kinetin (6-Furfurylaminopurine) and IAA (indole-3-acetic acid). Optimum regeneration media was identified as MS media supplemented with 2.0 mg L−1 Kin plus 0.5 mg L−1 IAA. This yielded the highest number of shoots (5.57 ± 0.42) and leaves (20.53 ± 1.91) per explant. Acclimatization of the resulting in vitro regenerants was successful in all potting mixtures tested. However, potting mixture PF (1:1:1 ratio of black soil/red soil/compost) was identified as the best medium for acclimatization of this species, as it yielded 100% survival percentage after 90 days of acclimatization. Ten in vitro regenerants of O. stamineus were randomly collected after the third subculture and subjected to genetic variation analysis using inter-simple sequence repeat (ISSR) markers. Out of 20 ISSR markers tested, 10 working primers were observed to produce satisfactory amplification of bands, with an average of 7.11 bands per primer. A total of 610 bands were produced by the 10 primers. The percentage of polymorphism was observed to be very low, yielding only 7.32% polymorphism among all samples. Jaccard dissimilarity analysis was also conducted and very low genetic distance (about 0.1) was found among the in vitro regenerants and between the regenerants with the mother plant, thus ascertaining the clonal nature of the plantlets produced in this study. Vermicompost is an organic waste produced from earthworms that can enhance the soil condition and is rich with essential plant nutrients, thus increasing produce quality and shelf life. In this study, a one-year field trial was conducted to elucidate the effects of vermicompost supplementation on the composition of bioactive compounds and antioxidant activities of pineapple (Ananas comosus var. MD2) fruits, compared to control and application of chemical fertilizer. Based on the results, pineapple fruits produced from plants supplemented with chemical fertilizer showed the strongest radical scavenging properties against 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), followed by vermicompost and control plants. Application of chemical fertilizer and vermicompost also produced fruits with a very high content of chlorophylls and β-carotene compared to control plants. However, the amounts of bioactive compounds present in fruits produced with chemical fertilizer are higher than in fruits produced with vermicompost. Total phenolics content and Ferric Reducing Antioxidant Power (FRAP) reducing power were lowest in fruit extracts produced from pineapple plants supplemented with vermicompost. These results suggested that vermicompost cannot completely replace chemical fertilizer for the production of fruits with a high content of phytoconstituents but could be used as an additional supplement to reduce environmental pollution and ensure agricultural sustainability. This project studied the effect of vermicompost application on the composition of bioactive anthocyanin and phenolic compounds, and the antioxidant activity of Clinacanthus nutans. The correlation between the bioactive constituents and antioxidant capacity was also evaluated. In this project, a field study was conducted using a randomized complete block design (RCBD) with four treatment groups, including control plants (CC), plants supplied with chemical fertilizer (CF), plants supplied with vermicompost (VC), and plants supplied with mixed fertilizer (MF). The leaves of C. nutans from all treatment groups were harvested, subjected to solvent extraction, and used for quantification of total anthocyanin content (TAC), total phenolic content (TPC), and total flavonoid content (TFC). The initial antioxidant activity of the extracts was evaluated using 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays, as well as after two and four weeks of storage at −20 °C and 4 °C. Data analysis showed that CC plants contained the highest TAC (2180.14 ± 338.43 µg/g dry weight) and TFC (276.25 ± 3.09 mg QE/g dry weight). On the other hand, CF plants showed the highest TPC (181.53 ± 35.58 mg GAE/g dry weight). Moreover, we found that CC plants had the highest antioxidant potential against DPPH radicals whereas MF plants showed the lowest antioxidant potential. After four weeks of extract storage at −20 °C and 4 °C, the TPC, TFC, TAC, and antioxidant potential of the extracts decreased. Extracts from VC showed the lowest percentage of total phenolic and total flavonoid loss after extract storage at −20 °C and 4 °C compared with other plant extracts. At this juncture, it could be deduced that the application of vermicompost had little effect on the expression of phenolics, flavonoids, or anthocyanin in C. nutans. Various explants (stem, leaf, and root) of Citrus assamensis were cultured on MS media supplemented with various combinations and concentrations (0.5-2.0 mg L(-1)) of NAA and BAP. Optimum shoot and root regeneration were obtained from stem cultures supplemented with 1.5 mg L(-1) NAA and 2.0 mg L(-1) BAP, respectively. Explant type affects the success of tissue culture of this species, whereby stem explants were observed to be the most responsive. Addition of 30 gL(-1) sucrose and pH of 5.8 was most optimum for in vitro regeneration of this species. Photoperiod of 16 hours of light and 8 hours of darkness was most optimum for shoot regeneration, but photoperiod of 24 hours of darkness was beneficial for production of callus. The morphology (macro and micro) and anatomy of in vivo and in vitro/ex vitro Citrus assamensis were also observed to elucidate any irregularities (or somaclonal variation) that may arise due to tissue culture protocols. Mantled fruits as a result of somaclonal variation are often observed from the oil palm plantlets regenerated via tissue culture. The mantling of fruits with finger-like and thick outer coating phenotypes significantly reduces the seed size and oil content, posing a threat to oil palm planters, and may jeopardize the economic growth of countries that depend particularly on oil palm plantation. The molecular aspects of the occurrence of somaclonal variations are yet to be known, possibly due to gene repression such as DNA methylation, histone methylation and histone deacetylation. Histone deacetylases (HDACs), involved in eukaryotic gene regulation by catalyzing the acetyl groups are removal from lysine residues on histone, hence transcriptionally repress gene expression. This paper described the total protein polymorphism profiles of somaclonal variants of oil palm and the effects of histone deacetylation on this phenomenon. Parallel to the different phenotypes, the protein polymorphism profiles of the mantled samples (leaves, fruits, and florets) and the phenotypically normal samples were proven to be different. Higher HDAC activity was found in mantled leaf samples than in the phenotypically normal leaf samples, leading to a preliminary conclusion that histone deacetylation suppressed gene expression and contributed to the development of somaclonal variants. The present study deals with the cytological investigations on the meristematic root cells of carnation (Dianthus caryophyllus Linn.) grown in vivo and in vitro. Cellular parameters including the mitotic index (MI), chromosome count, ploidy level (nuclear DNA content), mean cell and nuclear areas, and cell doubling time (Cdt) were determined from the 2 mm root tip segments of this species. The MI value decreased when cells were transferred from in vivo to in vitro conditions, perhaps due to early adaptations of the cells to the in vitro environment. The mean chromosome number was generally stable (2n = 2x = 30) throughout the 6-month culture period, indicating no occurrence of early somaclonal variation. Following the transfer to the in vitro environment, a significant increase was recorded for mean cell and nuclear areas, from 26.59 ± 0.09  μm² to 35.66 ± 0.10  μm² and 142.90 ± 0.59  μm² to 165.05 ± 0.58  μm², respectively. However, the mean cell and nuclear areas of in vitro grown D. caryophyllus were unstable and fluctuated throughout the tissue culture period, possibly due to organogenesis or rhizogenesis. Ploidy level analysis revealed that D. caryophyllus root cells contained high percentage of polyploid cells when grown in vivo and maintained high throughout the 6-month culture period.
