Background: Recent studies have suggested that the composition of gut microbiota (GM) may change after intracerebral hemorrhage. However, the causal inference of GM and hemorrhagic stroke is unknown. Mendelian Randomization (MR) is an effective research method that removes confounding factors and investigates the causal relationship between exposure and outcome. This study intends to explore the causal relationship between GM and hemorrhagic stroke with the help of MR. Methods: Univariable and multivariable MR analyses were performed using summary statistics of the GM (n = 18,340) in the MiBioGen consortium vs. the FinnGen consortium R9 summary statistics (intracerebral hemorrhage, subarachnoid hemorrhage, and nontraumatic intracranial hemorrhage). Causal associations between gut microbiota and hemorrhagic stroke were analyzed using inverse variance weighted, MR-Egger regression, weighted median, weighted mode, simple mode, and MR-PRESSO. Cochran's Q statistic, MREgger regression, and leave-one-out analysis were used to test for multiplicity and heterogeneity of instrumental variables. Separate reverse MR analyses were performed for microbiota found to be causally associated with hemorrhagic stroke in the forward MR analysis. Also, multivariate MR analyses were conducted after incorporating common confounders. Results: Based on the results of univariable and multivariate MR analyses, Actinobacteria (phylum) (OR, 0.80; 95%CI, 0.66-0.97; p = 0.025) had a protective effect against hemorrhagic stroke, while Rikenellaceae RC9 gut group (genus) (OR, 0.81; 95%CI, 0.67-0.99; p = 0.039) had a potential protective effect. Furthermore, Dorea (genus) (OR, 1.77; 95%CI, 1.27-2.46; p = 0.001), Eisenbergiella (genus) (OR, 1.24; 95%CI, 1.05-1.48; p = 0.013) and Lachnospiraceae UCG008 (genus) (OR, 1.28; 95%CI, 1.01-1.62; p = 0.041) acted as potential risk factors for hemorrhagic stroke. The abundance of Dorea (genus) (β, 0.05; 95%CI, 0.002~ 0.101; p = 0.041) may increase, and that of Eisenbergiella (genus) (β, -0.072; 95%CI, -0.137~ -0.007; p = 0.030) decreased after hemorrhagic stroke according to the results of reverse MR analysis. No significant pleiotropy or heterogeneity was detected in any of the MR analyses. Conclusion: There is a significant causal relationship between GM and hemorrhagic stroke. The prevention, monitoring, and treatment of hemorrhagic stroke through GM represent a promising avenue and contribute to a deeper understanding of the mechanisms underlying hemorrhagic stroke. Hypertension (HT) is a major cause of intracerebral hemorrhage (ICH), and hypertensive cerebral hemorrhage (HCH) has a serious impact on life quality of patients and gives society a heavy burden. This study investigated the key genes involved in HT and HCH. Three patients with HT, three with HCH, and three healthy individuals were recruited in the present study. Shared and specific differentially expressed genes (DEGs) in HT and HCH were identified using RNA sequencing and bioinformatics analysis. Functional annotation and protein--protein interaction (PPI) network construction of HT- and HCH-specific DEGs were conducted. Estimation of cell infiltration, drug prediction, and real-time qPCR (RT-qPCR) validation were performed. Compared with the controls, 502 and 1649 DEGs were identified in the HT and HCH groups, respectively. Among them, 378 DEGs were HT-specific and 633 DEGs were HCH-specific, and a total of 1771 DEGs were identified between HT and HCH. In addition, there were fewer immune cell types that differed significantly among the three groups. The RT-qPCR validation results in human subjects and cell models were generally consistent with the sequencing results. TyphoonMangkhut crossed the northeastern South China Sea (SCS) in September 2018 and induced energetic near-inertial waves (NIWs) that were captured by an array of 39 current- and pressure-recording inverted echo sounders and two tall moorings with acoustic Doppler current profilers and current meter sensors. The array extended from west of the Luzon Strait to the interior SCS, with the path of the typhoon cutting through the array. NIWs in the interior SCS had lower frequency than those near the Luzon Strait. After the typhoon crossed the SCS, Mangkhut-induced near-inertial currents in the upper ocean reached over 50 cm s-1. NIWs traveled southward for hundreds of kilometers, dominated by modes 2 and 3 in the upper and deep ocean. The horizontal phase speeds of mode 2 were ~3.9 and ~2.5 m s-1 north and south of the typhoon's track, respectively, while those of mode 3 were ~2.1 and ~1.7 m s-1, respectively. Mode 5 was only identified in the north with a smaller phase speed. Owing to different vertical group velocities, the energy of mode-2 NIWs reached the deep ocean in 20 days, whereas the higher-mode NIWs required more time to transfer energy to the bottom. NIWs in the north were trapped and carried by a westward-propagating anticyclonic eddy, which enhanced the near-inertial kinetic energy at ;300 m and lengthened the duration of energetic NIWs observed in the north. Consolidated bioprocessing (CBP) is a promising approach to directly convert lignocellulose feedstocks into targeted biochemicals and biofuels, and is commonly executed by single engineered microorganisms. Alternatively, the complex metabolic functions can be compartmentalized into different specialized microorganisms by using microbial consortia. In this study, a simplified microbial consortium was reassembled consisting of the lignocellulose degrader Trichoderma asperellum and lactic acid (LA) producer Lactobacillus paracasei, which were derived from the same native microbial consortium. The novel strategy of "learning from nature" combined with "simplifying the native microbial consortium" successfully increased cellulosic LA production from 7.36 g L-1 (native microbial consortium) to 57.57 g L-1 owing to the members' co-operation. Especially, over 80% carbon flux flowed from Avicel into LA synthesis with a yield of 0.92 g g-1. Furthermore, these two members can co-exist after compartmentalization into different spatial structures based on an aggregated granule of hydrogel-embedded L. paracasei and the formed biofilm of T. asperellum, leading to a 29.41% increase of LA productivity. This successfully constructed microbial consortium creates individual ecological niches to meet different oxygen demands, which enables more efficient co-operative performance in LA production from lignocellulose biomass. This study aims to evaluate the in vitro antibacterial and anti-biofilm activities of licochalcone A on Enterococcus faecalis and to investigate the possible target genes of licochalcone A in E. faecalis. This study found that licochalcone A had antibacterial activities against E. faecalis, with the MIC50 and MIC90 were 25μM. Licochalcone A (at 4 × MIC) indicated a rapid bactericidal effect on E. faecalis planktonic cells, and killed more E. faecalis planktonic cells (at least 3-log10 cfu/ml) than vancomycin, linezolid, or ampicillin at the 2, 4, and 6 h of the time-killing test. Licochalcone A (at 10 × MIC) significantly reduced the production of E. faecalis persister cells (at least 2-log10 cfu/ml) than vancomycin, linezolid, or ampicillin at the 24, 48, 72, and 96 h of the time-killing test. Licochalcone A (at 1/4 × MIC) significantly inhibited the biofilm formation of E. faecalis. The RNA levels of biofilmformation-related genes, agg, esp, and srtA, markedly decreased when the E. faecalis isolates were treated with licochalcone A at 1/4×MIC for 6 h. To explore the possible target genes of licochalcone A in E. faecalis, the licochalcone A non-sensitive E. faecalis clones were selected in vitro by induction of wildtype strains for about 140 days under the pressure of licochalcone A, andmutations in the possible target genes were detected by whole-genome sequencing. This study found that there were 11 nucleotide mutations leading to nonsynonymous mutations of 8 amino acids, and among these amino acidmutations, there were 3mutations located in transcriptional regulator genes (MarR family transcriptional regulator, TetR family transcriptional regulator, and MerR family transcriptional regulator). In conclusion, this study found that licochalcone A had an antibacterial effect on E. faecalis, and significantly inhibited the biofilm formation of E. faecalis at subinhibitory concentrations. Background: The incidence of major surgery is on the rise globally, and more than 20% of patients are readmitted to hospital following discharge from hospital. During their hospital stay, patients are monitored for early detection of clinical deterioration, which includes regularly measuring physiological parameters such as blood pressure, heart rate, respiratory rate, temperature, and pulse oximetry. This monitoring ceases upon hospital discharge, as patients are deemed clinically stable. Monitoring after discharge is relevant to detect adverse events occurring in the home setting and can be made possible through the development of digital technologies and mobile networks. Smartwatches and other technological devices allow patients to self-measure physiological parameters in the home setting, and Bluetooth connectivity can facilitate the automatic collection and transfer of this data to a secure server with minimal input from the patient. Objective: This paper presents the protocol for the DREAMPath (Domiciliary Recovery After Medicalization Pathway) study, which aims to measure compliance with a multidevice remote monitoring kit after discharge from hospital following major surgery. Methods: DREAMPath is a single-center, prospective, observational, cohort study, comprising 30 patients undergoing major intracavity surgery. The primary outcome is to assess patient compliance with wearable and interactive smart technology in the first 30 days following discharge from hospital after major surgery. Secondary outcomes will explore the relation between unplanned health care events and physiological data collected in the study, as well as to explore a similar relationship with daily patient-reported outcome measures (Quality of Recovery-15 score). Secondary outcomes will be analyzed using appropriate regression methods. Cardiopulmonary exercise testing data will also be collected to assess correlations with wearable device data. Results: Recruitment was halted due to COVID-19 restrictions and will progress once research staff are back from redeployment. We expect that the study will be completed in the first quarter of 2022. Conclusions: Digital health solutions have been recently made possible due to technological advances, but urgency in rollout has been expedited due to COVID-19. The security of massive data has always been the focus of computer security research. With the increase of data storage, the computing platform of single node can not deal with the increasing security of massive data. It is urgent to use distributed computing platform to improve computing efficiency and detection accuracy. The physical deployment of intrusion detection system on cloud computing platform consists of monitoring server, Hadoop master server, IDS server, node and IDS terminal management. The experimental results show that the proposed intrusion detection system based on Hadoop cloud node has better detection effect. This paper searches for the optimal weights, and then begins the training of the neural network. The whole process uses the Hadoop framework of distributed computing platform to implement the genetic algorithm and the neural network algorithm in the cloud computing platform. At the same time, the algorithm is improved to improve the efficiency and accuracy of intrusion detection. Health is dependent on the homeostasis of both inner and external microenvironments. The microbiota as the external microenvironment plays a critical role in regulation of several organ systems in mammals. However, it is unclear whether the microbiota regulates homeostasis of the skeletal system and bone marrow mesenchymal stem cells (BMMSCs). Here, using a well-established germ-free (GF) mouse model, we show that the microbiota significantly alters the stemness of BMMSCs in comparison to specific-pathogen-free (SPF)-derived BMMSCs. Colonization of GF mice with SPF microbiota (conventionalized (ConvD)) normalizes the proliferation and differentiation abilities of BMMSCs. On the other hand, normal microbiota is required to maintain immunomodulatory properties of BMMSCs through induction of activated T-cell apoptosis and cytokine secretion. GF-derived BMMSCs lose the capacity to ameliorate disease phenotypes in dextran sulfate sodium-induced experimental colitis mice. Mechanistically, single-cell RNA-sequencing analysis shows that ConvD BMMSCs have a similar gene expression pattern to SPF-derived BMMSCs, which have a distinct gene distribution from GF-derived BMMSCs. Background: Overgeneral autobiographical memory has been identified as a risk factor for the onset and maintenance of depression. However, little is known about the underlying mechanisms that might explain overgeneral autobiographical memory phenomenon in depression. The purpose of this study was to test the mediation effects of rumination on the relationship between overgeneral autobiographical memory and depressive symptoms. Specifically, the mediation effects of brooding and reflection subtypes of rumination were examined in patients with major depressive disorder. Methods: Eighty-seven patients with major depressive disorder completed the 17-item Hamilton Depression Rating Scale, Ruminative Response Scale, and Autobiographical Memory Test. Bootstrap mediation analysis for simple and multiple mediation models through the PROCESS macro was applied. Results: Simple mediation analysis showed that rumination significantly mediated the relationship between overgeneral autobiographical memory and depression symptoms. Multiple mediation analyses showed that brooding, but not reflection, significantly mediated the relationship between overgeneral autobiographical memory and depression symptoms. Conclusions: Our results indicate that global rumination partly mediates the relationship between overgeneral autobiographical memory and depressive symptoms in patients with major depressive disorder. Furthermore, the present results suggest that the mediating role of rumination in the relationship between overgeneral autobiographical memory and depression is mainly due to the maladaptive brooding subtype of rumination. Background: Schizophrenia (SCZ) is a complex, heritable, and devastating psychiatric disorder. Recent genome-wide association studies have identified a single-nucleotide polymorphism (SNP; rs10503253) in the CUB and SUSHI multiple domains 1 (CSMD1) gene as a risk factor for SCZ. In this study, we investigated whether the rs10503253 in CSMD1 contributes to the risk of SCZ in a Han Chinese population. Methods: We conducted a case-control study in a population from eastern China, involving 1378 SCZ patients and 1091 unrelated healthy controls, using the ligase detection reaction-polymerase chain reaction method to genotype the rs10503253 polymorphism in the CSMD1 gene. Results: No significant association was found between the SCZ patients and controls for any allele or genotype frequency of the SNP rs10503253 (all P > 0.05). A single layer of dense (>1011 cm-2) Si nanodots on an insulating a-SiN layer was fabricated by the method combining the laser irradiation on the ultrathin amorphous Si films and subsequent thermal annealing. Raman scattering spectroscopy, planar and cross-section transmission electron microscopy were employed to characterize the formation of Si nanodots. It was found that the size of formed Si nanodots is strongly influenced by the initial amorphous Si film thickness. Visible light emission was observed from the obtained Si nanodots at room temperature and the luminescence peak is varied from 660 to 725 nm with increasing the amorphous Si film thickness. The variable luminescence can be attributed to the interface state assisted radiative recombination rather than the quantum size effect. [ABSTRACT FROM AUTHOR]
