Landscape fragmentation affected the structure and function of the ecosystem, resulting in an impact on ecosystem service value (ESV). This paper analyzed the correlation between landscape fragmentation and ESV using land-use data from northern Shaanxi covering three periods from 2000 to 2020. The paper employed the granularity deduction method, spatial autocorrelation analysis, and value equivalent method to study the fragmentation characteristics of the regional landscape and the spatial-temporal evolution of ESV. The research findings indicated that the optimal granularity was 150 m, and the amplitude was 5 km × 5 km. The study found that the degree of landscape fragmentation was positively correlated with patch density (PD), division index (DIVISION), and Shannon's diversity index (SHDI), while negatively correlated with the largest patch index (LPI), patch cohesion index (COHESION), and effective mesh size (MESH). Moreover, the total ESV in the study area showed a decreasing trend, with grass, forest, and cultivated land being the three land-use types that contributed the most value. The analysis indicated that there was a negative correlation between the degree of landscape fragmentation and ESV. As the degree of landscape fragmentation increased, ESV decreased. The correlation between landscape fragmentation and ESV discussed in this paper provided valuable insights into the optimal utilization and sustainable development of regional land resources. Background: Tumor heterogeneity plays essential roles in developing cancer therapies, including therapies for breast cancer (BC). In addition, it is also very important to understand the relationships between tumor microenvironments and the systematic immune environment. Methods: Here, we performed single-cell, VDJ sequencing and spatial transcriptome analyses on tumor and adjacent normal tissue as well as axillar lymph nodes (LNs) and peripheral blood mononuclear cells (PBMCs) from 8 BC patients. Results: We found that myeloid cells exhibited environment-dependent plasticity, where a group of macrophages with both M1 and M2 signatures possessed high tumor specificity spatially and was associated with worse patient survival. Cytotoxic T cells in tumor sites evolved in a separate path from those in the circulatory system. T cell receptor (TCR) repertoires in metastatic LNs showed significant higher consistency with TCRs in tumor than those in nonmetastatic LNs and PBMCs, suggesting the existence of common neo-antigens across metastatic LNs and primary tumor cites. In addition, the immune environment in metastatic LNs had transformed into a tumor-like status, where pro-inflammatory macrophages and exhausted T cells were upregulated, accompanied by a decrease in B cells and neutrophils. Finally, cell interactions showed that cancer-associated fibroblasts (CAFs) contributed most to shaping the immune-suppressive microenvironment, while CD8+ cells were the most signal-responsive cells. Conclusions: This study revealed the cell structures of both micro- and macroenvironments, revealed how different cells diverged in related contexts as well as their prognostic capacities, and displayed a landscape of cell interactions with spatial information. We investigate the eco-environmental effects and the driving factors of transforming the production–living–ecological space (PLES) land use function and offer a scientific foundation for developing regional territorial area and environmental preservation. Eco-environment quality index and ecological contribution ratio are used to analyze the spatial–temporal evolution characteristics and eco-environment effects of land use transformation in the Yangtze River Delta Urban Agglomeration (YRDUA) over the three time periods of 2000, 2010, and 2020, and the geographic detectors are used to analyze the factors that influence the spatial difference of eco-environment quality (EEQ). The findings indicate the following: (1) The land use transformation of YRDUA is primarily shown in the shrinkage of the production land area, the stability of ecological land, and the rapid increase of living land. The area of ecological land, such as water, forest, and pasture, has remained relatively steady from the perspective of secondary land types. In contrast, the area of urban and rural living land has significantly increased. (2) Most land use environment comprises the lower-value zone, accounting for about 50%. The area of the low-value zone has continued to rise owing to the rapid urban and rural living land development, tending to continuous growth. (3) Both the ecological improvement and degradation trends are present simultaneously, although the ecological improvement trend is less prominent than the environmental degradation trend. The primary factor is improving the eco-environment by transforming agricultural production land into forest, water, and ecological pasture land. The degradation of the regional EEQ is mostly due to the occupation of agricultural production land by urban and rural living land. (4) Considering natural elements such as altitude, precipitation, and slope, the extent of land use impacts the EEQ. The combination of several factors has changed the EEQ of the YRDUA. The Grain for Green Project (GGP) has influenced Shaanxi Province's land-use pattern, resulting in a shift in ecosystem service value (ESV). Exploring the spatial and temporal evolution of the pattern of land use and ESV in Shaanxi Province, before and after the project's implementation, can give a theoretical foundation for regional land-use planning. For this study, we used the transfer matrix and the value equivalent approaches to investigate the influence of project implementation on the spatial distribution and evolution of patterns of land use and ESV in Shaanxi Province based on four periods of land-use data from 1990 to 2020. The results suggest the following: (1) Farmland, forestland, and grassland were the most common land-types in Shaanxi Province. Farmland, forestland, and grassland in Shaanxi Province were all altered dramatically over the research period due to the GGP. Farmland was turned mostly into forestland and grassland, and forestland and grassland areas progressively grew. (2) The ESVs in Shaanxi Province were USD 3802.82, 3814.90, 3836.20, and 3806.50 billion in 1990, 2000, 2010, and 2020, respectively. The most value was supplied by hydrological management, while water resources provided the lowest value. Forestland and grassland were the most valuable land-types in high-value locations, whereas built-up land was the most valuable land-type in low-value areas. (3) While the GGP has increased the ESV of forestland and grassland, it has harmed the acreage of other land-types, resulting in a decline in the total ESV in Shaanxi Province. Lung cancer remains the leading cause of cancer-related deaths worldwide. Traditional Chinese medicine (TCM) is a valuable resource of active natural products and plays an important role in cancer treatment with the advantages of high efficiency and safety. Wenxia Changfu formula (WCF) is modified from Dahuang Fuzi decoction from Han Dynasty and has been used for treating lung cancer in China. Our previous research showed that WCF had an antitumor effect in vivo and in vitro, while the mechanism has not been well illustrated. In this study, the effect of WCF on the proliferative ability in three lung cancer cells and one noncancerous human cell line was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. WCF suppressed A549, H460, and PC-9 cell viability in a dose-dependent manner, with no inhibition of noncancerous MRC-5 cells after 48 h treatment with WCF (0–50 mg/mL). Furthermore, we screened for genes in A549 cells using four WCF-treated samples and four control samples on a gene expression profile microarray. 21 genes were significantly downregulated by WCF, which may potentially play an important role in the proliferation of A549 cells. High-content screening evaluated whether silencing the 21 genes affected A549 cell growth. The results showed that PIF1 knockdown exhibited the most potent inhibition of cell proliferation compared with the other genes. Downregulation of PIF1 increased A549 cell apoptosis and the activity of caspase 3/7. Besides, RT-PCR showed that the expression levels of PIF1 mRNA decreased significantly in A549, H460, and PC-9 cells after WCF treatment. Gastrointestinal stromal tumor (GIST) is the most common mesenchymal tumor of the human gastrointestinal tract. In this study, we performed single‐cell RNA sequencing (RNA‐seq) on intra‐ and peri‐tumor tissues from GIST patients with the aim of discovering the heterogeneity of tumor cells in GIST and their interactions with other cells. We found four predominating cell types in GIST tumor tissue, including T cells, macrophages, tumor cells, and NK cells. Tumor cells could be clustered into two groups: one was highly proliferating and associated with high risk of metastasis, the other seemed "resting" and associated with low risk. Their clinical relevance and prognostic values were confirmed by RNA‐seq of 65 GIST samples. T cells were the largest cell type in our single‐cell data. Two groups of CD8+ effector memory (EM) cells were in the highest clonal expansion and performed the highest cytotoxicity but were also the most exhausted among all T cells. A group of macrophages were found polarized to possess both M1 and M2 signatures, and increased along with tumor progression. Cell‐to‐cell interaction analysis revealed that adipose endothelial cells had high interactions with tumor cells to facilitate their progression. Macrophages were at the center of the tumor microenvironment, recruiting immune cells to the tumor site and having most interactions with both tumor and nontumor cells. In conclusion, we obtained an overview of the GIST microenvironment and revealed the heterogeneity of each cell type and their relevance to risk classifications, which provided a novel theoretical basis for learning and curing GISTs. Abstract: Vegetation restoration practices play an important role in environmental management and could mitigate soil and water losses in the Chinese Loess Plateau. The main objective of this study was to ascertain the influence of vegetation durations on soil infiltration rate and other related soil properties. Undisturbed soil columns in triplicate from the same plot, with different locations, were collected to estimate the accumulated soil infiltration over different vegetation periods (9, 15, and 25 years) of grassland and forestland at different time intervals. The highest cumulative infiltration and wet front movement speed was recorded after 25 years of grassland and increased with the vegetation restoration duration. Low root biomass density (g cm−3) and soil organic matter (g kg−1) were recorded in the 9-year plantation of forestland and grassland; however, maximum root biomass densities of 1.614 ± 0.95 mg cm−3 and 0.938 ± 0.03 mg cm−3 were recorded after 25 years of forestland and grassland. Furthermore, root images scanner analysis showed that the 25 years of grassland has higher root length density and root surface area density of 5.917 ± 0.86 cm cm−3 and 2.058 ± 0.95 cm2 cm−3 at surface and subsurface soil layers. We therefore suggest that for revegetation periods of less than 25 years, grassland would be better for soil infiltration and related soil properties particularly in areas of the Chinese Loess Plateau.Graphical abstract: [ABSTRACT FROM AUTHOR]

Exploring soil water dynamics under different land use types is important for water resource management and vegetation restoration in the Loess Plateau. In this study, we investigated the hydrogen and oxygen isotopic compositions of soil water from four different land use types to explore the mechanism of soil water movement and transformation and analyse the influence of land use. The results show that the range of stable isotopes (δD and δ18O) in soil water was smaller than that in precipitation. Values for δD and δ18O in soil water showed relatively similar temporal variation, heavy isotopes were enriched in the soil water in July and depleted in October. Stable isotope values in shallow (<100 cm depth) soil water and deep (>200 cm depth) soil water were low. The δD and δ18O values in woodlands decreased gradually with increasing depth. Across the four land use types, the maximum variation in δD and δ18O was in the shallow depth of the soil profile. Groundwater was recharged mainly from precipitation and then from soil water. The ratio of groundwater recharge by soil water under different land use types followed this rank order: woodland (35.70%) > grassland (31.14%) > shrubland (29.47%) > cropland (29.18%). Matrix flow and preferential flow coexisted during infiltration, and the occurrence of preferential flow was related to the land use type. The main reason for the variation in isotopic composition in soil water is the difference in soil evaporation, which is influenced by different vegetation cover. Owing to the difference in soil evaporation and fractionation, precipitation on cropland, shrubland, and grassland can recharge more soil water than on woodland. [ABSTRACT FROM AUTHOR]
