The cotton industry in Xinjiang, China, is limited by irrigation. In cotton production, 1,1-dimethyl-piperidinium chloride (DPC) is used as a growth regulator to improve plant shape, resistance, yield, and quality. However, few studies have investigated the mechanisms by which DPC affects the growth and yield of cotton when combined with different irrigation water volumes. In this study, a split-zone design was used to conduct field experiments over two years using Zhongmiansuo 92 and Zhongmiansuo 087. Three irrigation volumes (3750, 4500, and 5250 m3 hm−2; W1, W2, and W3, respectively) and four DPC applications (0, 120, 240, and 480 g hm−2) were evaluated. The SPAD (Soil and Plant Analyzer Development) values, photosynthesis, dry matter accumulation and partitioning, agronomic traits, yield, and water use efficiency of cotton leaves were assessed. W2 increased the chlorophyll content and stomatal opening of leaves, improved photosynthetic rates, promoted the accumulation of aboveground dry matter, and increased plant height, main stem node number, and fruit branch platform. The best yields were obtained using W1 and W2 with 120 g hm−2 DPC and W3 with 240 g hm−2 DPC. These results can be applied practically to improve cotton production in Xinjiang. Soil microbes are crucial in shaping the root-associated microbial communities. In this study, we analyzed the effect of the soil–root niche gradient on the diversity, composition, and assembly of the bacterial community and co-occurrence network of two cotton varieties. The results revealed that the bacterial communities in cotton soil–root compartment niches exhibited a skewed species abundance distribution, dominated by abundant taxa showing a strong spatial specificity. The assembly processes of the rhizosphere bacterial communities were mainly driven by stochastic processes, dominated by the enrichment pattern and supplemented by the depletion pattern to recruit bacteria from the bulk soil, resulting in a more stable bacterial community. The assembly processes of the endosphere bacterial communities were determined by processes dominated by the depletion pattern and supplemented by the enrichment pattern to recruit species from the rhizosphere, resulting in a decrease in the stability and complexity of the community co-occurrence network. The compartment niche shaped the diversity of the bacterial communities, and the cotton variety genotype was an important source of diversity in bacterial communities within the compartment niche. We suggest that the moderate taxa contribute to significantly more changes in the diversity of the bacterial community than the rare and abundant taxa during the succession of bacterial communities in the cotton root–soil continuum. Pseudomonas aeruginosa is a highly pathogenic bacterium known for its ability to sense and coordinate the production of virulence factors in response to host immune responses. However, the regulatory mechanisms underlying this process have remained largely elusive. In this study, we investigate the two-component system CprRS in P. aeruginosa and unveil the crucial role of the sensor protein CprS in sensing the human host defense peptide LL-37, thereby modulating bacterial virulence. We demonstrate that CprS acts as a phosphatase in the presence of LL-37, leading to the phosphorylation and activation of the response regulator CprR. The results prove that CprR directly recognizes a specific sequence within the promoter region of the HigBA toxin-antitoxin system, resulting in enhanced expression of the toxin HigB. Importantly, LL-37-induced HigB expression promotes the production of type III secretion system effectors, leading to reduced expression of proinflammatory cytokines and increased cytotoxicity towards macrophages. Moreover, mutations in cprS or cprR significantly impair bacterial survival in both macrophage and insect infection models. This study uncovers the regulatory mechanism of the CprRS system, enabling P. aeruginosa to detect and respond to human innate immune responses while maintaining a balanced virulence gene expression profile. Additionally, this study provides new evidence and insights into the complex regulatory system of T3SS in P. aeruginosa within the host environment, contributing to a better understanding of host-microbe communication and the development of novel strategies to combat bacterial infections. However, the underlying mechanisms that enable P. aeruginosa to adapt to capricious environments of the human body are still poorly understood. In this study, we identified the CprRS, one of TCSs in P. aeruginosa, as an essential virulence factor. The sensor protein CprS directly interacts with human host defense peptide LL-37 and activates the response regulator CprR, which then coordinates the activities of type III secretion system for higher cytotoxicity. Loss of cprS or cprR significantly impairs bacterial survival in macrophage and host colonization. The transcription factor LIM‐only protein 4 (LMO4) is overexpressed in the psoriatic epidermis and regulates keratinocyte proliferation and differentiation. High LMO4 expression levels are induced by interleukin‐23 (IL‐23) to activate the AKT/STAT3 signaling pathway. Interleukin‐6 (IL‐6) is mainly involved in regulating T cell functions and development in patients with psoriasis. However, whether LMO4 expression is regulated by IL‐6 remains unclear. Therefore, the purpose of this study is to explore the role and molecular mechanisms of IL‐6 in regulating LMO4 expression. The interleukin‐6 (IL‐6) levels in human plasma were determined using a chemiluminescence immunoassay system. A psoriasis‐like mouse model was established using imiquimod induction. Epidermal keratinocytes (HaCaT) were cultured in defined keratinocyte‐serum‐free medium and stimulated by IL‐6 alone or with inhibitors. The proteins of interest were detected using western blot analysis, immunofluorescence, and immunohistochemistry. The 5‐ethynyl‐2′‐deoxyuridine assay was used to detect cell proliferation. The results revealed that IL‐6 levels were markedly increased in the plasma of patients with psoriasis, compared to healthy control. The high expression of LMO4 was consistent with high levels of IL‐6, p‐AKT, and p‐STAT3 in the lesions of both psoriasis patients and imiquimod‐induced psoriasis‐like mice. IL‐6 activates the AKT/STAT3 signaling pathway, followed by LMO4 high‐expression in HaCaT cells. IL‐6 induces HaCaT proliferation and differentiation via AKT/STAT3 signaling pathway activation. We think that the high expression of LMO4 in psoriatic keratinocytes requires IL‐6 to activate the AKT/STAT3 signaling pathway and leads to epidermal keratinocytes abnormal proliferation and differentiation. Background: The orthopedic treatment of the stiff clubfoot is challenging for clinicians, and the purpose of this study was to explore the preliminary findings of 3D printing-assisted patient-specific instrument (PSI) osteotomy guide for use in the orthopedic treatment of the stiff clubfoot. Material and methods: There were 20 patients (25 feet) with stiff clubfoot admitted from December 2018 to June 2022, including 13 males (16 feet) and 7 females (9 feet), aged 24–52 years, mean 40.15 years; 8 left feet, 7 right feet, 5 bipedal. All patients underwent triple arthrodesis and were divided into 10 cases (12 feet) in the PSI group (n = 12) and 10 cases (13 feet) in the conventional surgery group (n = 13) according to the surgical approach. The duration of surgery and the number of radiation exposures were recorded in all cases, and the American Orthopedic Foot and Ankle Society (AOFAS), and International Congenital Clubfoot Study Group (ICFSG) scoring systems were applied postoperatively to assess the effect of corrective treatment. All measurement data were expressed as mean ± standard deviation, and differences between groups were determined by Student's t test. All count data between the two groups were compared using the chi-square test or Fisher's exact test analysis. Results: All 20 patients (25 feet) were followed up for 1 year. No major complications related to osteotomy, such as overcorrection, incomplete correction, or bone nonunion, were observed in the PSI and conventional surgery groups at the final follow-up, and the PSI group had the advantage of shorter operative time (P < 0.01), less radiation exposure (P < 0.01), and higher excellent rate compared with the conventional surgery group. The AOFAS score (P > 0.05) and ICFSG score (P > 0.05) at the last follow-up were not statistically significant in both groups, but the excellent rate at the last follow-up was 91.7% in the PSI group which was significantly higher than that of the conventional surgery group at 76.9%. Conclusions: The utilization of 3D printing-assisted PSI osteotomy guide in orthopedic surgery for stiff clubfoot offers a safe and effective surgical tool for triple joint fusion treatment. Background: Platelet‐rich plasma (PRP) has been increasingly accepted as a potential therapy in the treatment of androgenetic alopecia (AGA), However, there remains a dearth of data on the effectiveness of PRP prepared by automatic blood cell separator with a combination of topical minoxidil for the treatment of AGA. Objective: To evaluate the efficacy and safety of PRP prepared by automatic blood cell separator combined with topical 5% minoxidil therapy in male AGA. Methods: Thirty male patients with mild/moderate AGA were enrolled in a randomized double‐blind controlled study. Patients were randomly divided into two treatment arms: (group A) PRP prepared by automatic blood cell separator combined with topical 5% minoxidil group; (group B) PRP prepared by automatic blood cell separator combined with a topical placebo group. Trichoscopic assessments regarding hair density/quantity and mean hair diameter were performed at baseline and follow‐up. Clinical efficacy of global photography and patient satisfaction were conducted to verify the therapeutic efficacy of the treatment, and the occurrence of adverse reactions was recorded. Results: We detected a significant increase in all patients in hair density and quantity after PRP treatment (p < 0.05), and there was no significant difference in mean hair diameter. Although hair density/quantity was more pronounced in group A than in group B, the difference between groups was not statistically significant (p > 0.05). In terms of clinical efficacy and patient satisfaction, group A was superior to group B, and no serious adverse reactions occurred. Conclusion: We hereby conclude that the injections of PRP prepared by an automated method are effective and safe in the treatment of mild‐to‐moderate male AGA patients, and its combination with topical 5% minoxidil therapy was superior to PRP monotherapy with better clinical efficacy and higher patient satisfaction. Key message: Genomic analysis of upland cotton revealed that cold tolerance was associated with ecological distribution. GhSAL1 on chromosome D09 negatively regulated cold tolerance of upland cotton. Cotton can undergo low-temperature stress at the seedling emergence stage, which adversely affects growth and yield; however, the regulatory mechanism underlying cold tolerance remains nebulous. Here, we analyze the phenotypic and physiological parameters in 200 accessions from 5 ecological distributions under constant chilling (CC) and diurnal variation of chilling (DVC) stresses at the seedling emergence stage. All accessions were clustered into four groups, of which Group IV, with most germplasms from the northwest inland region (NIR), had better phenotypes than Groups I–III under the two kinds of chilling stresses. A total of 575 significantly associated single-nucleotide polymorphism (SNP) were identified, and 35 stable genetic quantitative trait loci (QTL) were obtained, of which 5 were associated with traits under CC and DVC stress, respectively, while the remaining 25 were co-associated. The accumulation of dry weight (DW) of seedling was associated with the flavonoid biosynthesis process regulated by Gh_A10G0500. The emergence rate (ER), DW, and total length of seedling (TL) under CC stress were associated with the SNPs variation of Gh_D09G0189 (GhSAL1). GhSAL1HapB was the elite haplotype, which increased ER, DW, and TL by 19.04%, 11.26%, and 7.69%, respectively, compared with that of GhSAL1HapA. The results of virus-induced gene silencing (VIGS) experiment and determination of metabolic substrate content preliminarily illustrated that GhSAL1 negatively regulated cotton cold tolerance through IP3-Ca2+ signaling pathway. Key message: Two regions located at chromosome A05 and D04 were found to be significantly associated with 0–0.5 mm and 0.5–2 mm diameter roots, respectively, and two candidate genes related to root development were identified. Roots absorb water and nutrients, and play an important role in plant growth. However, there are few genetic developmental studies on cotton root structural traits. In this study, we used 200 upland cotton (Gossypium hirsutum L.) varieties to analyze the phenotypic variation of 43 traits. A total of 2001 related single-nucleotide polymorphism (SNP) sites located within or near 1046 genes were detected through a genome-wide association study (GWAS). The 32 root traits were linked to SNPs that corresponded to 317 nonrepetitive genes. For SNPs associated with root length and 0–0.5 mm diameter root traits, a significant peak appeared on chromosome A05 (between 21.91 and 22.24 Mb). For SNPs associated with root surface area, root volume and 0.5–2 mm diameter root traits, a significant peak appeared on chromosome D04 (between 7.35 and 7.70 Mb). Within these two key regions, SNPs were detected in the promoter and coding regions of two candidate genes, GhTRL1-A05 and GhPIN8-D04. The expression levels of these two genes also changed significantly according to transcriptome sequencing and quantitative real-time PCR (qRT–PCR). After silencing the GhTRL1 and GhPIN8 genes via virus-induced gene silencing (VIGS), we found that the plants expressing TRV2::GhTRL1 and TRV2::GhPIN8 had a reduced root length, surface area. Moreover, the contents of cis-12-oxo-phytodienoic acid (cis-OPDA), isopentenyl adenosine (iPR) and cis-zeatin (cZ) in the roots of the plants expressing TRV2::GhTRL1 decreased. Cold stress is a major environmental factor affecting plant growth and development. Although some plants have developed resistance to cold stress, the molecular mechanisms underlying this process are poorly understood. Using genome-wide association mapping with 200 cotton accessions collected from different regions, we identified variations in the short chain alcohol dehydrogenase gene, GhSAD1 , that responds to cold stress. Virus-induced gene silencing and overexpression in Arabidopsis revealed that GhSAD1 fulfils important roles in cold stress responses. Ectopic expression of a haploid genotype of GhSAD1 (GhSAD1 HapB ) in Arabidopsis increased cold tolerance. Silencing of GhSAD1 HapB resulted in a decrease in abscisic acid (ABA) content. Conversely, overexpression of GhSAD1 HapB increased ABA content. GhSAD1 HapB regulates cold stress responses in cotton through modulation of C-repeat binding factor activity, which regulates ABA signalling. GhSAD1 HapB induces the expression of COLD - REGULATED (COR) genes and increases the amount of metabolites associated with cold stress tolerance. Overexpression of GhSAD1 HapB partially complements the phenotype of the Arabidopsis ABA2 mutant, aba2-1. Background: To study the biomechanical effects of femoral prostheses at different coronal positions using finite element analysis and provide a clinical reference for unicompartmental knee arthroplasty (UKA). Methods: A normal knee joint model was established and verified, establishing 13 working conditions for the femoral prosthesis: the standard position, varus and valgus angles of 3°, 6° and 9° and medial and lateral translations of 1 mm, 3 mm and 5 mm. The stress changes at different positions were analysed, including the polyethylene (PE) insert upper surface, the surface of lateral compartment cartilage and the surface of cancellous bone under tibial prosthesis. Results: The stresses on the PE insert upper surface and the cancellous bone surface increased with increasing femoral prosthesis valgus/varus, and the stress increased gradually during medial to lateral translation. The stress change is more significant during valgus and lateral translation. However, the stress on the cartilage surface decreases in the process of varus to valgus and medial translation to lateral translation. Conclusion: The fixed-bearing femoral prosthesis of the medial UKA should avoid translation or varus/valgus tilt on the coronal plane as much as possible. The obvious misalignment of the femoral prosthesis will significantly affect the stress on the internal structure of the knee joint, especially the PE insert and cartilage surface. A femoral prosthesis coronal tilt of more than 6° may significantly increase the stress on the PE surface, and varus of more than 6° may significantly increase the stress on the cartilage surface. Roots play an important role in the response to nitrogen stress in cotton. To better understand the molecular and physiological mechanisms underlying the adaptation of cotton roots to nitrogen stress, the responses of genes in roots to nitrogen changes were analysed by RNA sequencing (RNA‐seq), and morphological and physiological indicators were measured for verification. This study revealed that lack of nitrogen stress (LN, 0 mm) and excess nitrogen stress (HN, 8 mm) inhibited root growth. Moderate nitrogen (MN, 4 mm) resulted in the largest values for root length, projected area and volume. Nitrogen stress regulated root growth via two mechanisms. Nitrogen stress decreased root growth by regulating cell wall growth via alterations in the cell wall composition and the expression of cell wall‐related genes. Under LN and HN, the contents of cellular polysaccharide and glucan reached their lowest levels, whereas the contents of lignin and cellulose reached their highest levels. Lower expression of EXPA17, EXLB1 and PME3 as well as higher expression of EXLA1, WAT1, CESA1, CESA3, CAD6, COMT1, SAMS, LAC4 and NAC081 were obtained under nitrogen stress compared to MN, indicating that nitrogen stress increases lignin and cellulose synthesis and promotes root ageing. Nitrogen stress also decreased root growth by affecting the contents and regulation of salicylic acid (SA), jasmonic acid (JA) and abscisic acid (ABA). Nitrogen significantly upregulated the expression of PAD4, ALD1, WRKY70, WAKL10, MYB44, FER, LHY and BHLH112 as well as significantly downregulated that of TGA6, BT1, BT2 and PYL4. The changes in the expression of these genes under nitrogen stress resulted in significant increases in the SA, JA and ABA contents in roots. Our results suggested that nitrogen stress inhibits the growth of the cotton root system by affecting cell wall growth and hormone regulation. These results contribute to our understanding of how nitrogen regulates cotton root growth at the genetic and physiological levels, and they provide a theoretical basis for the efficient utilization of nitrogen resources. Cotton yield is affected by drought stress. We performed widely targeted metabolomics to explore metabolic changes under drought stress in the roots and leaves of two cotton varieties differing in drought tolerance—XLZ8 as sensitive and G95079 as tolerant variety. A total of 537 metabolites were detected at different stages of drought stress. Abscisic acid (ABA) was significantly enriched in all stages of drought stress in both roots and leaves of XLZ8. Amino acids showed enrichment in leaves of G95079 in the early stage of drought stress. Flavonoids were significantly enriched in the leaves of the two varieties in the late stage of drought stress. In roots, nucleotides and their derivatives were highly enriched in XLZ8 in the late stage of drought stress. The biosynthetic pathways of kynurenine and polyamine and glycerophospholipid metabolism may play an important role in cotton drought tolerance by scavenging reactive oxygen species (ROS) and reconstructing cell membranes. Spraying appropriate concentrations of proline, kynurenine, S‐adenosylmethionine, glucose and 1‐oleoyl‐2‐hydroxy‐sn‐glycero‐3‐phosphocholine (lyso PC18:1) significantly increased the survival rate of cotton seedlings under drought stress. A hydroponic experiment was conducted to explore the effects of nitrogen on cotton root morphology and related hormone contents and to analyse the correlation between the indexes. The effects of different nitrogen concentrations (N0, N1, N2, and N3 with 0, 2, 4, and 8 mmol l−1, respectively) on main and lateral roots were evaluated. A moderate nitrogen concentration (N2, 4 mmol l−1) promotes root growth. Lateral roots responded quickly to changes in nitrogen concentrations. Compared with N0 treatment, N2 treatment resulted in significantly greater main root volume after 12 days as well as increased lateral root length, root projected area, root surface area, and root volume after 8 days. Nitrogen significantly decreased abscisic acid (ABA), salicylic acid (SA), and isopentenyladenine (iP) content, which were the lowest in N2 treatment (33.92% lower than N0). The ABA and SA content were negatively correlated with root morphology. This shows that a moderate nitrogen concentration (4 mmol l−1) significantly decreased ABA and SA content in the main and lateral roots and promoted root growth in cotton plants. These results will contribute to root growth theory regarding efficient nitrogen utilization by cotton plants and will provide a theoretical basis to ensure nitrogen resources are utilized efficiently. Postherpetic neuralgia (PHN) is the most common complication of acute herpes zoster. The treatment of PHN remains a challenge for clinical pain management. The present study investigated the P2X7 receptor antagonist brilliant blue G (BBG) whether inhibits endoplasmic reticulum stress and pyroptosis (a necrotic form of cell death) and alleviates PHN. Varicella zoster virus (VZV)-infected CV-1 cells were used to induce PHN model. Mechanical paw withdrawal thresholds were measured using an ascending series of von Frey filaments. Immunohistochemistry was used to detect the expression of P2X7R in nerve tissues. Western blot was used to determine the expression of endoplasmic reticulum (ER) stress and pyroptosis-related molecules. The expression of IL-1β and IL-18 in tissue homogenate was detected by ELISA. The PHN rat has the lower paw withdrawal threshold, but higher expression of P2X7 in nerve tissues. And, endoplasmic reticulum stress was activated and pyroptosis was increased in PHN rats. BBG can decrease pain thresholds and reduce ER stress and pyroptosis in PHN rats. In addition, ER stress activator tunicamycin (TM) can reverse the effect of BBG on the paw withdrawal thresholds, endoplasmic reticulum stress, and pyroptosis. The mutants resulted from the ongoing SARS-CoV-2 epidemic have showed resistance to antibody neutralization and vaccine-induced immune response. The present study isolated and identified two novel SARS-CoV-2 neutralizing antibodies (nAbs) from convalescent COVID-19 patients. These two nAbs (XG81 and XG83) were then systemically compared with nine nAbs that were reconstructed by using published data, and revealed that, even though these two nAbs shared targeting epitopes on spike protein, they were different from any of the nine nAbs. Compared with XG81, XG83 exhibited a higher RBD binding affinity and neutralization potency against wild-typed pseudovirus, variant pseudoviruses with mutated spike proteins, such as D614G, E484Q, and A475V, as well as the authentic SARS-CoV-2 virus. To explore potential broadly neutralizing antibodies, heavy and light chains from all 18 nAbs (16 published nAbs, XG81 and XG83) were cross-recombined, and some of the functional antibodies were screened and studied for RBD binding affinity, and neutralizing activity against pseudovirus and the authentic SARS-CoV-2 virus. The results demonstrated that several recombined antibodies had a more potent neutralization activity against variant pseudoviruses compared with the originally paired Abs. Taken together, the novel neutralizing antibodies identified in this study are a likely valuable addition to candidate antibody drugs for the development of clinical therapeutic agents against SARS-CoV-2 to minimize mutational escape. The relationships among the generation of acoustic emission, electromagnetic emission, and the fracture stress of rock grain are investigated, which are based on the mechanism of acoustic emission and electromagnetic emission produced in the process of indenting rock. Based on the relationships, the influence of loading rate on the characteristics of acoustic emission and electromagnetic emission of rock fragmentation is further discussed. Experiment on rock braking was carried out with three loading rates of 0.001 mm/s, 0.01 mm/s, and 0.1 mm/s. The results show that the phenomenon of acoustic emission and electromagnetic emission is produced during the process of loading and breaking rock. The wave forms of the two signals and the curve of the cutter indenting load show jumping characteristics. Both curves have good agreement with each other. With the increase of loading rate, the acoustic emission and electromagnetic emission signals are enhanced. Through analysis, it is found that the peak count rate, the energy rate of acoustic emission, the peak intensity, the number of pulses of the electromagnetic emission, and the loading rate have a positive correlation with each other. The experimental results agree with the theoretical analysis. The proposed studies can lead to an in-depth understanding of the rock fragmentation mechanism and help to prevent rock dynamic disasters. [ABSTRACT FROM AUTHOR]

Plant population density (PPD) and nitrogen (N) application rate (NAR) are two controllable factors in cotton production. We conducted field experiments to investigate the effects of PPD, NAR and their interaction (PPD × NAR) on yield, N uptake and N use efficiency (NUE) of cotton using a split-plot design in the North China Plain during 2013 and 2014. The main plots were PPDs (plants m−2) of 3.00 (low), 5.25 (medium) and 7.50 (high) and the subplots were NARs of 0 (N-free), 112.5 (low), 225.0 (moderate) and 337.5 (high). During both 2013 and 2014, biological yield and N uptake of cotton increased significantly, but harvesting index decreased significantly with NAR and PPD increasing. With NAR increasing, internal nitrogen use efficiency(NUE) decreased significantly under three PPDs and agronomical NUE, physiologilal NUE, nitrogen recovery efficiency(NRE) and partial factor productivity from applied nitrogen (PFPN) also decreased significantly under high PPD between two years. Lint yield increment varied during different PPDs and years, but NAR enhancement showed less function under higher PPD than lower PPD in general. Taken together, moderate NAR under medium PPD combined higher lint yield with higher agronomic NUE, physiological NUE, and NRE, while low NAR with high PPD would achieve a comparable yield with superior NRE and PFPN and high NAR under high PPD and medium PPD produced higher biological yield but lower harvest index, lint yield and NUE compared to moderate NAR with medium PPD. The biodegradation characteristics of dimethyl phthalate (DMP) by three freshwater unicellular organisms were investigated in this study. The findings revealed that all the organisms were capable of metabolizing DMP; among them, Cyanothece sp. PCC7822 achieved the highest degradation efficiency. Lower concentration of DMP supported the growth of the Cyanobacteria; however, with the increase of DMP concentration growth of Cyanobacteria was inhibited remarkably. Phthalic acid (PA) was detected to be an intermediate degradation product of DMP and accumulated in the culture solution. The optimal initial pH value for the degradation was detected to be 9.0, which mitigated the decrease of pH resulting from the production of PA. The optimum temperature for DMP degradation of the three species of organisms is 30°C. After 72 hours’ incubation, no more than 11.8% of the residual of DMP aggregated in Cyanobacteria cells while majority of DMP remained in the medium. Moreover, esterase was induced by DMP and the activity kept increasing during the degradation process. This suggested that esterase could assist in the degradation of DMP. [ABSTRACT FROM AUTHOR]

Salinity is a major abiotic stress that affects plant growth and development. In this study, we performed a proteomic analysis of cotton roots and leaf tissue following exposure to saline stress. 611 and 1477 proteins were differentially expressed in the roots and leaves, respectively. In the roots, 259 (42%) proteins were up-regulated and 352 (58%) were down-regulated. In the leaves, 748 (51%) proteins were up-regulated and 729 (49%) were down-regulated. On the basis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis, we concluded that the phenylalanine metabolism and starch and sucrose metabolism were active for energy homeostasis to cope with salt stress in cotton roots. Moreover, photosynthesis, pyruvate metabolism, glycolysis / gluconeogenesis, carbon fixation in photosynthetic organisms and phenylalanine metabolism were inhabited to reduce energy consumption. Background and Aims Cotton (Gossypium hirsutum) has indeterminate growth. The growth regulator mepiquat chloride (MC) is used worldwide to restrict vegetative growth and promote boll formation and yield. The effects of MC are modulated by complex interactions with growing conditions (nutrients, weather) and plant population density, and as a result the effects on plant form are not fully understood and are difficult to predict. The use of MC is thus hard to optimize. Methods To explore crop responses to plant density and MC, a functional–structural plant model (FSPM) for cotton (named CottonXL) was designed. The model was calibrated using 1 year's field data, and validated by using two additional years of detailed experimental data on the effects of MC and plant density in stands of pure cotton and in intercrops of cotton with wheat. CottonXL simulates development of leaf and fruits (square, flower and boll), plant height and branching. Crop development is driven by thermal time, population density, MC application, and topping of the main stem and branches. Key Results Validation of the model showed good correspondence between simulated and observed values for leaf area index with an overall root-mean-square error of 0·50 m2 m−2, and with an overall prediction error of less than 10 % for number of bolls, plant height, number of fruit branches and number of phytomers. Canopy structure became more compact with the decrease of leaf area index and internode length due to the application of MC. Moreover, MC did not have a substantial effect on boll density but increased lint yield at higher densities. Conclusions The model satisfactorily represents the effects of agronomic measures on cotton plant structure. Varicella and herpes zoster are mild symptoms-associated diseases caused by varicella–zoster virus (VZV). They often cause severe complications (disseminated zoster), leading to death when diagnoses and treatment are delayed. However, most commercial VZV diagnostic tests have low sensitivity, and the most sensitive tests are unevenly available worldwide. Here, we developed and validated a highly sensitive VZV diagnostic kit based on the chemiluminescent immunoassay (CLIA) approach. VZV-glycoprotein E (gE) was used to develop a CLIA diagnostic approach for detecting VZV-specific IgA, IgG, and IgM. The kit was tested with 62 blood samples from 29 VZV-patients classified by standard ELISA into true-positive and equivocal groups and 453 blood samples from VZV-negative individuals. The diagnostic accuracy of the CLIA kit was evaluated by receiver-operating characteristic (ROC) analysis. The relationships of immunoglobulin-isotype levels between the two groups and with patient age ranges were analyzed. Overall, the developed CLIA-based diagnostic kit demonstrated the detection of VZV-specific immunoglobulin titers depending on sample dilution. From the ELISA-based true-positive patient samples, the diagnostic approach showed sensitivities of 95.2%, 95.2%, and 97.6% and specificities of 98.0%, 100%, and 98.9% for the detection of VZV-gE-specific IgA, IgG, and IgM, respectively. Combining IgM to IgG and IgA detection improved diagnostic accuracy. Comparative analyses on diagnosing patients with equivocal results displaying very low immunoglobulin titers revealed that the CLIA-based diagnostic approach is overall more sensitive than ELISA. In the presence of typical VZV symptoms, CLIA-based detection of high titer of IgM and low titer of IgA/IgG suggested the equivocal patients experienced primary VZV infection. Furthermore, while no difference in IgA/IgG level was found regarding patient age, IgM level was significantly higher in young adults. The CLIA approach-based detection kit for diagnosing VZV-gE-specific IgA, IgG, and IgM is simple, suitable for high-throughput routine analysis situations, and provides enhanced specificity compared to ELISA. This paper presents the results of an experimental investigation on the deformation and cracking behaviors of ring-shaped granite specimens. Diametrical compression tests were conducted on ring-shaped granite specimens with inclusion and those without inclusion for comparison. The inclusion materials were of different strengths, stiffness, and sizes. Strain gauges (SGs) were amounted at the boundary of the hole to trace the strain evolutions, and digital image correlation (DIC) technique was used to identify the crack initiation and propagation process. The results indicated that the inclusion has a positive effect on the enhancement of the carrying capacity. The maximum carrying capacity of the specimen with low-strength inclusion decreases with increasing the inclusion diameter, while it first decreases and then increases when the inclusion diameter increases from 10 to 20 mm for the high-strength infilled specimens. The results of the strain evolutions analyzed by the SGs and DIC methods indicate that a lager hole diameter and a lower inclusion strength lead to a more intensive strain concentration around the hole. Due to the mismatch in the deformation at the rock–cement mortar interface, serious strain concentrations are induced, and hence leading to three types of failure, i.e., debonding between the interface, tensile crack in the granite, and tensile crack in the inclusion material. Gossypium hirsutum L., is a widely cultivated cotton species around the world, but its production is seriously threatened by its susceptibility to chilling stress. Low temperature affects its germination, and the underlying molecular mechanisms are rarely known, particularly from a transcriptional perspective. In this study, transcriptomic profiles were analyzed and compared between two cotton varieties, the cold-tolerant variety KN27-3 and susceptible variety XLZ38. A total of 7535 differentially expressed genes (DEGs) were identified. Among them, the transcripts involved in energy metabolism were significantly enriched during germination based on analysis of Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways, such as glycolysis/gluconeogenesis, tricarboxylic acid cycle (TCA cycle), and glyoxylate cycle (GAC). Results from further GO enrichment analysis show the earlier appearance of DNA integration, meristem growth, cotyledon morphogenesis, and other biological processes in KN27-3 compared with XLZ38 under chilling conditions. The synthesis of asparagine, GDP-mannose, and trehalose and the catabolic process of raffinose were activated. DEGs encoding antioxidants (spermidine) and antioxidase (CAT1, GPX4, DHAR2, and APX1) were much more up-regulated in embryos of KN27-3. The content of auxin (IAA), cis-zeatin riboside (cZR), and trans-zeatin riboside (tZR) in KN27-3 are higher than that in XLZ38 at five stages (from 12 h to 54 h). GA3 was expressed at a higher level in KN27-3 from 18 h to 54 h post imbibition compared to that in XLZ38. And abscisic acid (ABA) content of KN27-3 is lower than that in XLZ38 at five stages. Results from hormone content measurements and the related gene expression analysis indicated that IAA, CTK, and GA3 may promote germination of the cold-tolerant variety, while ABA inhibits it. Background: Appropriate plant architecture can improve the amount of cotton boll opening and allow increased planting density, thus increasing the level of cotton mechanical harvesting and cotton yields. The internodes of cotton fruiting branches are an important part of cotton plant architecture. Thus, studying the molecular mechanism of internode elongation in cotton fruiting branches is highly important. Results: In this study, we selected internodes of cotton fruiting branches at three different stages from two cultivars whose internode lengths differed significantly. A total of 76,331 genes were detected by transcriptome sequencing. By KEGG pathway analysis, we found that DEGs were significantly enriched in the plant hormone signal transduction pathway. The transcriptional data and qRT-PCR results showed that members of the GH3 gene family, which are involved in auxin signal transduction, and CKX enzymes, which can reduce the level of CKs, were highly expressed in the cultivar XLZ77, which has relatively short internodes. Genes related to ethylene synthase (ACS), EIN2/3 and ERF in the ethylene signal transduction pathway and genes related to JAR1, COI1 and MYC2 in the JA signal transduction pathway were also highly expressed in XLZ77. Plant hormone determination results showed that the IAA and CK contents significantly decreased in cultivar XLZ77 compared with those in cultivar L28, while the ACC (the precursor of ethylene) and JA contents significantly increased. GO enrichment analysis revealed that the GO categories associated with promoting cell elongation, such as cell division, the cell cycle process and cell wall organization, were significantly enriched, and related genes were highly expressed in L28. However, genes related to the sphingolipid metabolic process and lignin biosynthetic process, whose expression can affect cell elongation, were highly expressed in XLZ77. In addition, 2067 TFs were differentially expressed. The WRKY, ERF and bHLH TF families were the top three largest families whose members were active in the two varieties, and the expression levels of most of the genes encoding these TFs were upregulated in XLZ77. Conclusions: Auxin and CK are positive regulators of internode elongation in cotton branches. In contrast, ethylene and JA may act as negative regulators of internode elongation in cotton branches. Furthermore, the WRKY, ERF and bHLH TFs were identified as important inhibitors of internode elongation in cotton. In XLZ77(a short-internode variety), the mass synthesis of ethylene and amino acid conjugation of auxin led to the inhibition of plant cell elongation, while an increase in JA content and degradation of CKs led to a slow rate of cell division, which eventually resulted in a phenotype that presented relatively short internodes on the fruiting branches. The results of this study not only provide gene resources for the genetic improvement of cotton plant architecture but also lay a foundation for improved understanding of the molecular mechanism of the internode elongation of cotton branches.
