2. Single-cell analysis of matrisome-related genes in breast invasive carcinoma: new avenues for molecular subtyping and risk estimation.

Background: The incidence of breast cancer remains high and severely affects human health. However, given the heterogeneity of tumor cells, identifying additional characteristics of breast cancer cells is essential for accurate treatment. Purpose: This study aimed to analyze the relevant characteristics of matrix genes in breast cancer through the multigroup data of a breast cancer multi-database. Methods: The related characteristics of matrix genes in breast cancer were analyzed using multigroup data from the breast cancer multi database in the Cancer Genome Atlas, and the differential genes of breast cancer matrix genes were identified using the elastic net penalty logic regression method. The risk characteristics of matrix genes in breast cancer were determined, and matrix gene expression in different breast cancer cells was evaluated using real-time fluorescent quantitative polymerase chain reaction (PCR). A consensus clustering algorithm was used to identify the biological characteristics of the population based on the matrix molecular subtypes in breast cancer, followed by gene mutation, immune correlation, pathway, and ligand-receptor analyses. Results: This study reveals the genetic characteristics of cell matrix related to breast cancer. It is found that 18.1% of stromal genes are related to the prognosis of breast cancer, and these genes are mostly concentrated in the biological Frontiers in processes related to metabolism and cytokines in protein. Five different matrixrelated molecular subtypes were identified by using the algorithm, and it was found that the five molecular subtypes were obviously different in prognosis, immune infiltration, gene mutation and drug-making gene analysis. Conclusions: This study involved analyzing the characteristics of cell-matrix genes in breast cancer, guiding the precise prevention and treatment of the disease. [ABSTRACT FROM AUTHOR]

4. Gut microbiota dysbiosis in ankylosing spondylitis: a systematic review and meta-analysis.

Background: Ankylosing spondylitis (AS) is a connective tissue disease that primarily affects spinal joints, peripheral joints, and paravertebral soft tissues, leading to joint stiffness and spinal deformity. Growing evidence has implicated gutmicrobiota in the regulation of AS, though the underlying mechanisms remain poorly understood. Methods: We conducted a comprehensive search of PubMed, Embase, Web of Science, the Cochrane Library, MEDLINE, Wanfang Data, China Science and Technology Journal Database (VIP), and China National Knowledge Internet (CNKI) databases from the time the databases were created until 30 July 2023. To evaluate changes in a-diversity and the abundance of certain microbiota families in AS, standardized mean difference (SMD) and 95% confidence interval (CI) calculations were made. Meta-analyses were performed using STATA 12.0 and the quality of the literature was assessed by following systematic review guidelines. Results: This systematic review and meta-analysis included 47 studies, providing insights into the gut microbiota composition in patients with AS compared to healthy controls (HCs). Our findings indicate a significant reduction in gut microbial diversity in patients with AS, as evidenced by a decrease in both richness and evenness. Specifically, the Shannon index showed a moderate decrease (SMD = -0.27, 95% CI: -0.49, -0.04; P < 0.001), suggesting a less diverse microbial ecosystem in patients with AS. The Chao1 index further confirmed this trend, with a larger effect size (SMD = -0.44, 95% CI: -0.80, -0.07; P < 0.001), indicating a lower species richness. The Simpson index also reflected a significant reduction in evenness (SMD = -0.30, 95% CI: -0.53, -0.06; P < 0.001). Additionally, patients with AS who received anti-rheumatic combination treatment exhibited a more pronounced reduction in α-diversity compared to untreated patients, highlighting the potential impact of this treatment on gut microbiota balance. In terms of specific microbial families, we observed a significant decrease in the abundance of Bifidobacterium (SMD = -0.42, 95% CI: -2.37, 1.52; P < 0.001), which is known for its beneficial effects on gut health. Conversely, an increase in the abundance of Bacteroidetes was noted (SMD = 0.42, 95% CI: -0.93, 1.76; P < 0.001), suggesting a possible shift in the gut microbiota composition that may be associated with AS pathophysiology. Conclusion: Our analysis revealed changes in α-diversity and the relative abundance of specific bacteria in AS. Aquaculture of red swamp crayfish Procambarus clarkii is an emerging farming industry in China. As shown recently, one of the most significant diseases affecting the red swamp crayfish is caused by overabundance of epibiotic peritrichs, a group of ciliated protists that are commonly found living as epibionts on aquatic animals. Nevertheless, few studies focusing on this issue have been performed. As part of a faunal study carried out in the Lake Weishan Wetland, China, four colonial sessilid peritrichs, namely Epicarchesium granulatum, Epistylis bimarginata, Epistylis semiciculus, and Zoothamnium procerius, attached to P. clarkii are documented. For each species, redescriptions and improved diagnoses are supplied. As a further aid to identification, 14 new rDNA sequences of these four and a closely related species were obtained. In addition, as an aid for future studies on epibiotic ciliates, we supply a list and information about recent integrative studies that document the geographic distributions of related taxa. Aqueous Mg-ion batteries (AMIBs) featuring advantages of good safety, low cost, and high specific energy have been recognized as a promising energy-storage technology. However, the performance of AMIBs is consistently limited by sluggish diffusion kinetics and structural degradation of cathode materials arising from the strong electrostatic interactions between high-charge-density Mg2+ and host materials. Here, layered-structured NiOOH, as traditional cathodes for alkaline batteries, is initially demonstrated to realize proton-assisted Mg-(de)intercalation chemistry with a high discharge platform (0.57 V) in neutral aqueous electrolytes. Benefiting from the unique core/shell structure, the resulting NiOOH/CNT cathodes achieve a high capacity of 122.5 mAh g
-1 and long cycle stability. Further theoretical calculations reveal that the binding energy of hydrated Mg2+ is higher than that of Mg2+ with NiOOH, resulting in that Mg2+ is easily intercalated/de-intercalated into/from NiOOH. Benefiting from the freestanding design, the assembled fiber-shaped "rocking-chair" NaTi2 (PO4 )3 //NiOOH AMIB shows a high energy density and satisfactory mechanical flexibility, which could be woven into a commercial fabric and power for fiber-shaped photoelectric sensors. Li metal batteries (LMBs) with LiNi
0.8 Co0.1 Mn0.1 O2 (NMC811) cathodes could release a specific energy of >500 Wh kg-1 by increasing the charge voltage. However, high-nickel cathodes working at high voltages accelerate degradations in bulk and at interfaces, thus significantly degrading the cycling lifespan and decreasing the specific capacity. Here, we rationally design an all-fluorinated electrolyte with addictive tri(2,2,2-trifluoroethyl) borate (TFEB), based on 3, 3, 3-fluoroethylmethylcarbonate (FEMC) and fluoroethylene carbonate (FEC), which enables stable cycling of high nickel cathode (LiNi0.8 Co0.1 Mn0.1 O2 , NMC811) under a cut-off voltage of 4.7 V in Li metal batteries. The electrolyte not only shows the fire-extinguishing properties, but also inhibits the transition metal dissolution, the gas production, side reactions on the cathode side. Therefore, the NMC811jjLi cell demonstrates excellent performance by using limited Li and high-loading cathode, delivering a specific capacity >220 mA h g-1 , an average Coulombic efficiency >99.6% and capacity retention >99.7% over 100 cycles. Background: Metabolic dysregulation is a hallmark of type 2 diabetes mellitus (T2DM), in which the abnormalities in brown adipose tissue (BAT) play important roles. However, the cellular composition and function of BAT as well as its pathological signifi- cance in diabetes remain incompletely understood. Our objective is to delineate the single-cell landscape of BAT-derived stromal vascular fraction (SVF) and their characteristic alterations in T2DM rats. Methods: T2DM was induced in rats by intraperitoneal injection of low-dose streptozotocin and high-fat diet feeding. Single-cell mRNA sequencing was then performed on BAT samples and compared to normal rats to characterize changes in T2DM rats. Subsequently, the importance of key cell subsets in T2DM was elucidated using various functional studies. Results: Almost all cell types in the BAT-derived SVF of T2DM rats exhibited enhanced inflammatory responses, increased angiogenesis, and disordered glucose and lipid metabolism. The multidirectional differentiation potential of adipose tissue-derived stem cells was also reduced. Moreover, macrophages played a pivotal role in intercellular crosstalk of BAT-derived SVF. A novel Rarres2+macrophage subset promoted the differentiation and metabolic function of brown adipocytes via adipose-immune crosstalk. Conclusion: BAT SVF exhibited strong heterogeneity in cellular composition and function and contributed to T2DM as a signifi- cant inflammation source, in which a novel macrophage subset was identified that can promote brown adipocyte function. 12. Investigation on the improved absolute nodal coordinate formulation for curved shell with variable cross-section.

Curved shell structures with variable cross-sections are extensively applied in options for deep space exploration and on-orbit operation, due to their better mechanical properties compared to traditional materials. However, these structures also pose challenging dynamic modeling issues due to their nonlinear geometry and high flexibility. In this paper, the improved absolute nodal coordinate formulation (S-A-ANCF) is proposed for analyzing the dynamic behaviors of curved shells with variable cross-sections. Firstly, the neutrosphere of the curved shell is corrected to accurately describe the displacement field. Secondly, reduction factors for meshing are proposed by considering the aspect ratio criterion and shear performance criterion, which are then combined with the Enhanced Assumed Strain (EAS) method to establish the S-A-ANCF model. Finally, the correctness of the proposed method is verified through numerical analysis, and the optimized criterion for the modeling process is analyzed. According to the numerical analysis, the proposed method effectively improves modeling accuracy and efficiency. The coupling relationships between geometry, materials, boundary conditions, and stress are illustrated. The research outcomes of this paper can provide theoretical guidance for spacecraft systems with curved shells, which are of great significance and urgently need to be studied. [ABSTRACT FROM AUTHOR]

13. Host–microbe interactions in the nasal cavity of dogs with chronic idiopathic rhinitis.

Chronic rhinitis (CR) is a frustrating clinical syndrome in dogs and our understanding of the disease pathogenesis in is limited. Increasingly, host– microbe interactions are considered key drives of clinical disease in sites of persistent mucosal inflammation such as the nasal and oral cavities. Therefore, we applied next generation sequencing tools to interrogate abnormalities present in the nose of dogs with CR and compared immune and microbiome profiles to those of healthy dogs. Host nasal cell transcriptomes were evaluated by RNA sequencing, while microbial communities were assessed by 16S rRNA sequencing. Correlation analysis was then used to identify significant interactions between nasal cell transcriptomes and the nasal microbiome and how these interactions were altered in animals with CR. Notably, we observed significant downregulation of multiple genes associated with ciliary function in dogs with CR, suggesting a previously undetected role for ciliary dysfunction in this syndrome. We also found significant upregulation of immune genes related to the TNF-α and interferon pathways. The nasal microbiome was also significantly altered in CR dogs, with overrepresentation of several potential pathobionts. Interactome analysis revealed significant correlations between bacteria in the genus Porphyromonas and the upregulated host inflammatory responses in dogs with CR, as well as defective ciliary function which was correlated with Streptococcus abundance. These findings provide new insights into host–microbe interactions in a canine model of CR and indicate the presence of potentially causal relationships between nasal pathobionts and the development of nasal inflammation and ciliary dysfunction. 14. Identification and validation of Rab GTPases RAB13 as biomarkers for peritoneal metastasis and immune cell infiltration in colorectal cancer patients.

Background: As one of the most common cancer, colorectal cancer (CRC) is with high morbidity and mortality. Peritoneal metastasis (PM) is a fatal state of CRC, and few patients may benefit from traditional therapies. There is a complex interaction between PM and immune cell infiltration. Therefore, we aimed to determine biomarkers associated with colorectal cancer peritoneal metastasis (CRCPM) and their relationship with immune cell infiltration. Methods: By informatic analysis, differently expressed genes (DEGs) were selected and hub genes were screened out. RAB13, one of the hub genes, was identificated from public databases and validated in CRC tissues. The ESTIMATE, CEBERSORT and TIMER algorithms were applied to analyze the correlation between RAB13 and immune infiltration in CRC. RAB13's expression in different cells were analyzed at the single-cell level in scRNA-Seq. The Gene Set Enrichment Analysis (GSEA) was performed for RAB13 enrichment and further confirmed. Using oncoPredict algorithm, RAB13's impact on drug sensitivity was evaluated. Results: High RAB13 expression was identified in public databases and led to a poor prognosis. RAB13 was found to be positively correlated with the macrophages and other immune cells infiltration and from scRNA-Seq, RAB13 was found to be located in CRC cells and macrophages. GSEA revealed that high RAB13 expression enriched in a various of biological signaling, and oncoPredict algorithm showed that RAB13 expression was correlated with paclitaxel sensitivity. Conclusion: Our study indicated clinical role of RAB13 in CRC-PM, suggesting its potential as a therapeutic target in the future. [ABSTRACT FROM AUTHOR]

Introduction: Gut microbiota (GM) influences the occurrence and development of pancreatic cancer (PC), potentially through the involvement of inflammatory cytokines (IC) and immune cells (IM). We aimed to investigate the causal impact of the gut microbiota (GM) on pancreatic cancer (PC) and identify potential IC and IM mediators. Methods: The summary statistics data from whole-genome association studies of gut microbiota, immune cells, inflammatory cytokines, and four types of pancreatic tumors (MNP: Malignant neoplasm of pancreas; BNP: Benign neoplasm of pancreas; ADCP: Adenocarcinoma and ductal carcinoma of pancreas; NTCP: Neuroendocrine tumor and carcinoma of pancreas). Twosample univariable Mendelian randomization (UVMR), multivariable Mendelian randomization (MVMR), and mediation analysis were employed to assess the causal relationship between gut microbiota (GM) and pancreatic cancer (PC), as well as potential IC and IM mediators. Results: The two-sample UVMR analysis showed causal relationships between 20 gut microbiota species and pancreatic cancer, with pancreatic cancer affecting the abundance of 37 gut microbiota species. Mediation analysis revealed that Interleukin-6 (IL-6), "CD4 on naive CD4+ T cell" and "SSC-A on HLA DR+ Natural Killer" mediated the causal effects of gut microbiota on pancreatic cancer. Conclusion: This Mendelian randomization study demonstrates causal relationships between several specific gut microbiota and pancreatic cancer, as well as potential mediators (IC, IM). 18. Trimethylamine N-oxide predicts cardiovascular events in coronary artery disease patients with diabetes mellitus: a prospective cohort study.

Background: Gut microbiota has significant impact on the cardio-metabolism and inflammation, and is implicated in the pathogenesis and progression of atherosclerosis. However, the long-term prospective association between trimethylamine N-oxide (TMAO) level and major adverse clinical events (MACEs) in patients with coronary artery disease (CAD) with or without diabetes mellitus (DM) habitus remains to be investigated. Methods: This prospective, single-center cohort study enrolled 2090 hospitalized CAD patients confirmed by angiography at Beijing Hospital from 2017-2020. TMAO levels were performed using liquid chromatography-tandem mass spectrometry. The composite outcome of MACEs was identified by clinic visits or interviews annually. Multivariate Cox regression analysis, Kaplan-Meier analysis, and restricted cubic splines were mainly used to explore the relationship between TMAO levels and MACEs based on diabetes mellitus (DM) habitus. Results: During the median follow-up period of 54 (41, 68) months, 266 (12.7%) developed MACEs. Higher TMAO levels, using the tertile cut-off value of 318.28 ng/mL, were significantly found to be positive dose-independent for developing MACEs, especially in patients with DM (HR 1.744, 95%CI 1.084-2.808, p = 0.022). Conclusions: Higher levels of TMAO are significantly associated with long-term MACEs among CAD patients with DM. The combination of TMAO in patients with CAD and DM is beneficial for risk stratification and prognosis. [ABSTRACT FROM AUTHOR]

Background: Phosphodiesterase 7 (PDE7) plays a role in neurological function. Increased expression and activity of PDE7 has been detected in several central nervous system diseases. However, the role of PDE7 in regulating stress levels remains unclear. Thus, this study aimed to determine whether and how PDE7 involved in the stress-induced behavioral and neuron morphological changes. Methods: The single prolonged stress (SPS) was used to build a stress exposure model in C57BL/6 J mice and detected PDE7 activity in hippocampus, amygdala, prefrontal cortex and striatum. Next, three doses (0.2, 1, and 5 mg/kg) of the PDE7 inhibitor BRL-50481 were intraperitoneally administered for 10 days, then behavioral, biochemical, and morphological tests were conducted. Results: PDE7 activity in hippocampus of mice significantly increased at all times after SPS. BRL-50481 significantly attenuated SPS induced anxiety-like behavior and fear response in both context and cue. In addition, BRL-50481 increased the levels of key molecules in the cAMP signaling pathway which were impaired by SPS. Immunofluorescent staining and Sholl analysis demonstrated that BRL-50481 also restored the nucleus/cytoplasm ratio of hippocampal neurons and improved neuronal plasticity. These effects of BRL-50481 were partially blocked by the TrkB inhibitor ANA-12. Conclusion: PDE7 inhibitors attenuate stress-induced behavioral changes by protecting the neuron cytoarchitecture and the neuronal plasticity in hippocampus, which is mediated at least partly through the activation of BDNF/TrkB signaling pathway. 20. Left Pulmonary Vein Trunk Length as a Robust Predictor of Long-Term Success of Atrial Fibrillation Catheter Ablation.

- Abstract
Background: Radiofrequency catheter ablation (RFCA) is a commonly used treatment for atrial fibrillation (AF), but the long-term recurrence rate remains relatively high. Given the inconsistent results regarding the role of left pulmonary vein (PV) ostial anatomy in post-ablative recurrence of RFCA in previous studies, we sought to investigate the role of left PV trunk length using an alternative methodology. Methods: A total of 369 AF patients undergoing catheter ablation were included. The left/right trunk length (LTL/RTL) of the PV was measured from pre-ablative computed tomography (CT) using three-dimensional reconstruction techniques. We constructed three multivariable Cox models, with the inclusion of the LTL, RTL, and no LTL/RTL, and used the Delong test, integrated discrimination index (IDI), and net reclassification index (NRI) to assess model improvement. We identified optimal cut-off values for LTL with the receiver operating characteristic (ROC) curve, and estimated outcomes using the Kaplan-Meier survival curve. We also used subgroup analysis to evaluate interactions. Results: The results of the Delong test, IDI, and NRI indicated that LTL had a favorable impact on the performance of the multivariate model. Subsequently, the multivariate Cox regression analysis identified LTL as a significant risk factor for post-ablative recurrence of AF (adjusted hazard ratio (HR) = 1.08, 95% CI: 1.05–1.12, p < 0.001). According to the ROC curve, the optimal cut-off value for LTL is 11.15 mm, and the Kaplan-Meier estimator revealed different outcomes (p < 0.001). We calculated p for interaction between LTL and other factors, and no significant interaction terms were observed. Conclusions: LTL is a robust prognostic indicator for post-ablative outcome in AF patients receiving RFCA, with a longer LTL indicating a higher risk of recurrence. [ABSTRACT FROM AUTHOR]

21. Spatial distribution and source apportionment of soil heavy metals in the areas affected by non-ferrous metal slag field in southwest China.

The spatial distribution and source analysis of heavy metals in soil are of great significance for the risk assessment and control of heavy metal pollution. Herein, a total of 87 topsoil samples (0-30 cm) were collected in the areas adjacent to a non-ferrous metal slag field of Gejiu City, Yunnan Province. The typical heavy metal concentrations, available phosphorus, available potassium, ammonium nitrogen, nitrate nitrogen, and pH in each soil sample were determined. Based on the above basic data, distribution and origins of heavy metals was analyzed. The results indicated that the mean contents of Pb, As, Cu, Zn, Ni and Cr in the topsoil of the study site were 247.82, 204.36, 104.83, 411.38, 29.73 and 67.59 mg kg-1, respectively. Exception of the Ni element, the contents of the remaining five heavy metals surpassed their respective background levels in study site. Especially, As element (with the highest accumulation effect) is 11.1l times of the background level, and the remaining four elements are 1.04-6.10 times of the background level in Yunnan. Single factor pollution index showed that the pollution degree of Pb, Cu and Zn was light, and the accumulation of As was a great threat to soil quality. Furthermore, the percentages of sites at mild pollution, moderate pollution and heavy pollution levels were 16.09%, 26.09% and 63.22%, respectively. Additionally, The order of potential ecological hazard degree of soil in the survey area was As > Pb > Cu = Ni > Zn = Cr. The spatial dispersion of heavy metals within the study region is impacted by both natural phenomena and human activities. The areas with severe pollution were the mining area in the northwest and planting sites in the east, middle and south of the study area. The main contribution sources of Ni were the mixed sources of natural parent material and industrial activities. The heavy metal element largely contributed by industrial activities and transportation is Pb. The main sources of metals As, Cu and Zn are the integrated sources of agricultural and industrial activities. The main source of Cr is natural source. Therefore, these useful results can guide the heavy metal pollution control and restoration of study site and this research work can also be used to other similar metal slag field and its affected area. Background: T lymphocytes in tumor microenvironment play a pivotal role in the anti-tumor immunity, and the memory of T cells contributes to the longterm protection against tumor antigens. Compared to solid tumors, studies focusing on the T-cell differentiation in the acute myeloid leukemia (AML) bone marrow (BM) microenvironment remain limited. Patients and methods: Fresh BM specimens collected from 103 adult AML patients at diagnosis and 12 healthy donors (HDs) were tested T-cell differentiation subsets by multi-parameter flow cytometry. Results: CD4 and CD8 T-cell compartments had different constituted profiles of T-cell differentiated subsets, which was similar between AML patients and HDs. Compared to HDs, AML patients as a whole had a significantly higher proportion of CD8 effector T cells (Teff, P = 0.048). Moreover, the T-cell compartment of AML patients with no DNMT3A mutations skewed toward terminal differentiation at the expense of memory T cells (CD4 Teff: P = 0.034; CD8 Teff: P = 0.030; CD8 memory T: P = 0.017), whereas those with mutated DNMT3A had a decrease in CD8 naïve T (Tn) and CD4 effector memory T cells (Tem) as well as an increase in CD4 central memory T cells (Tcm) (P = 0.037, 0.053 and 0.053). Adverse ELN genetic risk correlated with a lower proportion of CD8 Tn. In addition, the low proportions of CD4 Tem and CD8 Tn independently predicted poorer relapsefree survival (RFS, HR [95%CI]: 5.7 (1.4-22.2), P = 0.017 and 4.8 [1.3-17.4], P = 0.013) and event-free survival (EFS, HR [95% CI]: 3.3 (1.1-9.5), P = 0.029; 4.0 (1.4-11.5), P = 0.010), respectively. Conclusions: AML patients had abnormal profiles of BM T-cell differentiation subsets at diagnosis, which was related to DNMT3A mutations. High-entropy compounds have been emerging as promising candidates for electrolysis, yet their controllable electrosynthesis strategy remains a formidable challenge because of the ambiguous ionic interaction and codeposition mechanism. Herein, we report a oxygenates directionally induced electrodeposition strategy to construct high-entropy materials with amorphous features, on which the structural evolution from high-entropy phosphide to oxide is confirmed by introducing vanadate, thus realizing the simultaneous optimization of composition and structure. The representative P-CoNiMnWVOx shows excellent bifunctional catalytic performance toward alkaline hydrogen evolution reaction and ethanol oxidation reaction (EOR), with small potentials of -168 mV and 1.38 V at 100 mA cm
-2 , respectively. In situ spectroscopy illustrates that the electrochemical reconstruction of P-CoNiMnWVOx induces abundant Co--O species as the main catalytic active species for EOR and follows the conversion pathway of the C2 product. Theoretical calculations reveal the optimized electronic structure and adsorption free energy of reaction intermediates on P-CoNiMnWVOx , thereby resulting in a facilitated kinetic process. A membrane-free electrolyzer delivers both high Faradaic efficiencies of acetate and H2 over 95% and superior stability at100 mA cm-2 during 120 h electrolysis. In addition, the unique composition and structural advantages endow P-CoNiMnWVOx with multifunctional catalytic activity and realize multipathway electrosynthesis of formate-coupled hydrogen production. Heart failure (HF) prognosis depends on various regulatory factors; microRNA‐128 (miR‐128) is identified as a regulator of cardiac fibrosis, contributing to HF. MyoD family inhibitor (MDFI), which is reported to be related with Wnt/β‐catenin pathway, is supposed to be regulated by miR‐128. This study investigates the interaction between miR‐128 and MDFI in cardiomyocyte development and elucidates its role in heart injury. Gene expression profiling assessed miR‐128's effect on MDFI expression in HF using qPCR and Western blot analysis. Luciferase assays studied the direct interaction between miR‐128 and MDFI. MTT, transwell, and immunohistochemistry evaluated the effects of miR‐128 and MDFI on myocardial cells in mice HF. Genescan and luciferase assays validated the interaction between miR‐128 and MDFI sequences. miR‐128 mimics significantly reduced MDFI expression at mRNA and protein levels with decrease rate of 55%. Overexpression of miR‐128 promoted apoptosis with the increase rate 65% and attenuated cardiomyocyte proliferation, while MDFI upregulation significantly enhanced proliferation. Elevated miR‐128 levels upregulated Wnt1 and β‐catenin expression, whereas increased MDFI levels inhibited these expressions. Histological analysis with haematoxylin and eosin staining revealed that miR‐128 absorption reduced MDFI expression, hindering cell proliferation and cardiac repair, with echocardiography showing corresponding improvements in cardiac function. Our findings suggest miR‐128 interacts with MDFI, playing a crucial role in HF management by modulating the Wnt1/β‐catenin pathway. Suppression of miR‐128 could promote cardiomyocyte proliferation, highlighting the potential value of the miR‐128/MDFI interplay in HF treatment. [ABSTRACT FROM AUTHOR]

Intuition plays a crucial role in human driving decision-making, and this rapid and unconscious cognitive process is essential for improving trafic safety. We used the first proposed multi-layer network analysis method, "Joint Temporal-Frequency Multi-layer Dynamic Brain Network" (JTF-MDBN), to study the EEG data from the initial and advanced phases of driving intuition training in the theta, alpha, and beta bands. Additionally, we conducted a comparative study between these two phases using multi-layer metrics as well as local and global metrics of single layers. The results show that brain region activity is more stable in the advanced phase of intuition training compared to the initial phase. Particularly in the alart state task, the JTF-MDBN demonstrated stronger connection strength. Multi-layer network analysis indicates that modularity is significantly higher for the non-alert state task than the alert state task in the alpha and beta bands. In the W4 time window (1 second before a collision), we identified significant features that can differentiate situations where a car collision is imminent from those where no collision occurs. Single-layer network analysis also revealed statistical differences in node strength and local effciency for some EEG channels in the alpha and beta bands during the W4 and W5 time windows. Using these biomarkers to predict vehicle collision risk, the classification accuracy of a linear kernel SVM reached up to 87.5%, demonstrating the feasibility of predicting driving collisions through brain network biomarkers. System logs are instrumental in understanding computer system behavior and ensuring system stability and reliability, making anomaly detection in system logs crucial. However, with the increasing scale and complexity of modern software systems, log data is growing exponentially, rendering traditional manual log inspection methods inefficient. Moreover, the evolution of log messages over time results in a lower accuracy rate for anomaly detection. To address these issues, this paper proposes a log anomaly detection method based on multi-scale temporal convolution networks and multi-head attention. This method utilizes temporal convolution networks to extract temporal information from log data and extracts hidden features of logs through different receptive fields of multi-scale convolution kernels. By integrating the multi-head attention mechanism, the sequential dependencies of logs can be better captured. We conducted repeated experiments on the authoritative public HDFS and BGL log datasets to evaluate their detection accuracy and robustness. In the aquatic microbial food web, scuticociliates are diverse and predominant in almost all kinds of biotopes while extremely confused regarding their taxonomy and systematic relationships within the subclass Scuticociliatia. Here we provide detailed descriptions of two new and one known species, Glauconema sinica sp. nov., Pleuronema parasetigerum sp. nov., and Histiobalantium natans viridis, isolated from a freshwater wetland in northern China, along with their molecular phylogeny based on SSU rRNA gene sequences. Our results show: (1) both molecular and morphological data strongly support that the isolated lineage containing the genera Glauconema, Miamiensis, Paramesanophrys, and Anophryoides should be identified as a new taxon at the family level in the order Philasterida; hence, a new family, Glauconematidae fam. nov., is established. This new family is mainly characterized by the polymorphic life cycle of its members (macrostome and microstome stages); the trophont (or macrostome) has closely spaced membranelles 1-3, whereas in the tomite (or microstome) membranelle 1 is distant from membranelle 2. (2) Phylogenetic analyses using novel data for these species revealed that all three cluster with their congeners, supporting the validity of the genera to which they belong. This article examines the development of primary English education from teachers' and students' perspectives; this is done by investigating environment, curriculum and teacher and students' information. The study was carried out with 42 primary English teachers and 404 primary students from 90 urban and rural primary schools in 13 regions in the Hunan Province, China, the participants engaged in the questionnaire tasks to investigate the present status and problems with primary English education in the Hunan Province. Based on the assessment indicators through CIPP model, combining educational theory and sustainable development theory, the present study develops a primary school English education evaluation model and conducts a practical investigation of primary school English education in the Hunan Province, based on the established model. The findings indicate that the quality of teachers has improved, teaching methods and teaching equipment have become diversified, and student interests and English proficiencies have increased. However, there are still deficiencies in the implementation of Primary English Education in the Hunan Province. For instance, certain schools improperly implement national policies, resulting in imbalanced education. Additionally, educational inequality persists due to disparities in family economic status and importance. Moreover, regional, ideological, and management factors contribute to uneven allocation of educational resources. Furthermore, significant disparities exist between urban and rural areas in terms of teacher qualifications, teaching quality, and school operating hours. The article proposes enhancing awareness of sustainable development, strengthening supervision, and seeking educational and policy support to facilitate the sustainable development of primary English education. 29. Recurrent Hypoglycemia After Total Gastrectomy: A Case Report and Literature Analysis.

Objective: Rare disease Background: Hypoglycemia is a common complication following total gastrectomy, primarily caused by dumping syndrome and severe malnutrition, with late dumping syndrome being particularly significant. However, for recurrent fasting hypoglycemia, the possibility of insulinoma should be considered. Hypoglycemia caused by insulinoma can lead to severe consequences, including seizures and even death. Thus, it is crucial to differentially diagnose hypoglycemia occurring after total gastrectomy. Case Report: In this report, we present the case of a 36-year-old Chinese woman who underwent total gastrectomy for gastric cancer and subsequently received chemotherapy. Four months after surgery, she began experiencing recurrent seizures, and multiple tests confirmed hypoglycemia. A series of laboratory and imaging examinations ultimately led to a diagnosis of insulinoma. After surgical resection of the tumor, the patient's hypoglycemic symptoms resolved, and pathology results confirmed an insulinoma. Conclusions: This case report highlights the rapid weight loss and severe hypoglycemia observed in a patient only 4 months after total gastrectomy for gastric cancer. Although dumping syndrome was initially suspected based on the clinical course, the final diagnosis turned out to be insulinoma. The case underscores the importance of comprehensive evaluation and appropriate diagnostic investigations for patients experiencing hypoglycemia after total gastrectomy. Furthermore, the case suggests that the increased levels of enteroglucagon following changes in the gastrointestinal tract resulting from total gastrectomy may promote the development of insulinomas. The binary code similarity detection (BCSD) technique can quantitatively measure the differences between two given binaries and give matching results at predefined granularity (e.g., function), and has been widely used in multiple scenarios including software vulnerability search, security patch analysis, malware detection, code clone detection, etc. With the help of deep learning, the BCSD techniques have achieved high accuracy in their evaluation. However, on the one hand, their high accuracy has become indistinguishable due to the lack of a standard dataset, thus being unable to reveal their abilities. On the other hand, since binary code can be easily changed, it is essential to gain a holistic understanding of the underlying transformations including default optimization options, non-default optimization options, and commonly used code obfuscations, thus assessing their impact on the accuracy and adaptability of the BCSD technique. This paper presents our observations regarding the diversity of BCSD datasets and proposes a comprehensive dataset for the BCSD technique. We employ and present detailed evaluation results of various BCSD works, applying different classifications for different types of BCSD tasks, including pure function pairing and vulnerable code detection. Our results show that most BCSD works are capable of adopting default compiler options but are unsatisfactory when facing non-default compiler options and code obfuscation. Bilateral breast cancer (BBC), an infrequent breast cancer subtype, has primarily been studied in terms of incidence, prognosis, and through comparative analysis of synchronous (SBBC) and metachronous (MBBC) manifestations. The advent and application of organoid technology hold profound implications for tumor research and clinical management. This study represents the pioneering use of organoid models in BBC research. We established organoid lines from two surgical tumor specimens of a BBC patient, with one line undergoing detailed pathological and genomic analysis. The BBC organoid from the right breast demonstrated a marker expression profile of ER (-), PR (-), HER-2 (0), and Ki67 index 10%, indicating that it may derived from the TNBC tissue. Whole Exome Sequencing (WES) displayed consistent set of Top10 cancer driver genes affected by missense mutations, frameshift mutation, or splice site mutations in three tumor tissues and the organoid samples. The organoids' single nucleotide polymorphisms (SNPs) were more closely aligned with the TNBC tissue than other tumor tissues. Evolutionary analysis suggested that different tumor regions might evolve from a common ancestral layer. In this case, the development of BBC organoids indicated that simultaneous lesions with diverse molecular profiles shared a high degree of consistency in key tumor-driving mutations. These findings suggest the feasibility of generating BBC organoids representing various molecular types, accurately replicating significant markers and driver mutations of the originating tumor. Introduction: The gut microbiota (GM) influences the occurrence and progression of lung cancer (LC), with potential involvement of immune cells (IC). We aimed to investigate the causal impact of GM on LC and identify potential immune cell mediators. Methods: The utilized data for the Genome-Wide Association Studies (GWAS) were summarized as follows: gut microbiota data from the Dutch Microbiome Project (DMP) (N = 7,738), lung cancer data from the Transdisciplinary Research in Cancer of the Lung (TRICL) and International Lung Cancer Consortium (ILCCO) (Ncase = 29,266, Ncontrol = 56,450) included four types of cancer: NSCLC, LUAD, LUSC, and SCLC, and immune cell data from European populations (N = 3,757). We employed bi-directional two-sample univariable Mendelian randomization (UVMR), multivariable Mendelian randomization (MVMR), and mediation analysis to assess the causal relationship between GM and LC and potential immune cell mediators. Results: Bi-directional UVMR analysis revealed that 24 gut microbiota species can affect LC, while LC can affect the abundance of 17 gut microbiota species. Mediation analysis demonstrated that six immune cells mediated the causal relationships of seven gut microbiota species on LC: "CCR7 on naive CD8+ T cell" mediated the causal relationship between s_Alistipes_putredinis and LUAD, with a mediation proportion of 9.5% and P = 0.018; "IgD-CD27-B cell %lymphocyte" mediated the causal relationships between g_Gordonibacter and s_Gordonibacter_pamelaeae with LUSC, with mediation proportions of 11.8% and 11.9%, respectively and P = 0.029; "CD20-CD38-B cell %lymphocyte" mediated the causal relationship between s_Bacteroides_clarus and SCLC, with a mediation proportion of 13.8% and P = 0.005; "CD20 on IgD+ CD38- unswitched memory B cell" mediated the causal relationship between s_Streptococcus_thermophilus and SCLC, with a mediation proportion of 14.1% and P = 0.023; "HLA DR on CD14- CD16+ monocyte" mediated the causal relationship between s_Bifidobacterium_bifidum and SCLC, with a mediation proportion of 8.7% and P = 0.012; "CD45 on Granulocytic Myeloid-Derived Suppressor Cells" mediated the causal relationship between f_Lactobacillaceae and SCLC, with a mediation proportion of 4.0% and P = 0.021. Recently, there has been a significant interest in the study of highly active and stable transition metalbased electrocatalysts for the oxygen evolution reaction (OER). Non-noble metal nanocatalysts with excellent inherent activity, many exposed active centers, rapid electron transfer, and excellent structural stability are especially promising for the displacement of precious-metal catalysts for the production of sustainable and "clean" hydrogen gas through water-splitting. Herein, efficient electrocatalyst N-doped nickel molybdate nanorods were synthesized on Ni foam by a hydrothermal process and effortless chemical vapor deposition. The heterostructure interface of N-NiMoO
4 /NiO2 led to strong electronic interactions, which were beneficial for enhancing the OER activity of the catalyst. Excellent OER catalytic activity in 1.0 M KOH was shown, which offered a small overpotential of 185.6 mV to acquire a current density of 10 mA cm-2 (superior to the commercial benchmark material RuO2 under the same condition). This excellent electrocatalyst was stable for 90 h at a constant current density of 10 mA cm-2 . We created an extremely reliable and effective OER electrocatalyst without the use of noble metals by doping a nonmetal element with nanostructured heterojunctions of various active components. Purpose: In the current study, the clinical efficacy of iodine-125 (125I) radioactive particle implantation with deep hyperthermia in malignant tumor treatment was retrospectively analyzed. Material and methods: Sixty patients with malignant tumors treated at Zhongshan Hospital affiliated with Dalian University, from January 2021 to January 2023, were included in this retrospective analysis. Computed tomography (CT)-guided 125I radioactive particles were implanted into lesions, followed by deep hyperthermia administered for 3 days after surgery. Lesion size was evaluated monthly using CT or magnetic resonance imaging (MRI), and objective response rate (ORR) and local control rate (DCR) were calculated. Mean pain numerical score and European Organization for Research and Treatment of Cancer quality of life core scale C30 (EORTC QLQ-C30 v. 3.0) were assessed pre-treatment and post-treatment, and post-operative adverse reactions were comparatively analyzed. Results: After 6 months, the combined treatment resulted in a 73.33% ORR and an 81.67% DCR among 60 patients experiencing pre-treatment pain, and 49 (81.67%) exhibited effective pain relief. EORTC QLQ-C30 v. 3.0 evaluation revealed improved quality of life post-treatment. Adverse reactions during combined treatment were mitigated with targeted supportive measures, and no adverse reactions of level 2 or higher were observed. Conclusions: 125I radioactive particle implantation with deep hyperthermia is shown to be an effective combination therapy for malignant tumors, yielding significant clinical efficacy. Notably, it enhances patient quality of life, reduces tumor burden, mitigates adverse reactions, and alleviates pain. Objective: The objective of this study is to investigate the causal relationship between gut microbiota and juvenile idiopathic arthritis, and to identify and quantify the potential role of plasma metabolites as mediators. Methods: Using summary-level data from genome-wide association studies, a two-sample Mendelian randomization was conducted involving 131 gut microbiota genus, 1,400 plasma metabolites, and juvenile idiopathic arthritis. Additionally, a two-step approach was employed to quantify the proportion of the effect of gut microbiota on juvenile idiopathic arthritis mediated by plasma metabolites. Effect estimation primarily utilized Inverse Variance Weighting, with further validation using Bayesian weighted Mendelian randomization. Results: In our MR analysis, a positive correlation was observed between Rikenellaceae and the risk of juvenile idiopathic arthritis, while Dorea showed a negative correlation with juvenile idiopathic arthritis risk. Mediation analysis indicated that Furaneol sulfate levels acted as a mediator between Dorea and juvenile idiopathic arthritis, with an indirect effect proportion of 19.94, 95% CI [8.86-31.03%]. Conclusion: Our study confirms a causal relationship between specific microbial genus and juvenile idiopathic arthritis, and computes the proportion of the effect mediated by plasma metabolites, offering novel insights for clinical interventions in juvenile idiopathic arthritis. Cancer and diabetes are significant diseases that pose a threat to human health. Their interconnection is complex, particularly when they coexist, often necessitating multiple therapeutic approaches to attain remission. Sodiumglucose cotransporter protein two inhibitors (SGLT-2i) emerged as a treatment for hyperglycemia, but subsequently exhibited noteworthy extraglycemic properties, such as being registered for the treatment of heart failure and chronic kidney disease, especially with co-existing albuminuria, prompting its assessment as a potential treatment for various non-metabolic diseases. Considering its overall tolerability and established use in diabetes management, SGLT-2i may be a promising candidate for cancer therapy and as a supplementary component to conventional treatments. This narrative review aimed to examine the potential roles and mechanisms of SGLT-2i in the management of diverse types of cancer. Future investigations should focus on elucidating the antitumor efficacy of individual SGLT-2i in different cancer types and exploring the underlying mechanisms. Additionally, clinical trials to evaluate the safety and feasibility of incorporating SGLT-2i into the treatment regimen of specific cancer patients and determining appropriate dosage combinations with established antitumor agents would be of significant interest. Background: Nodal T-follicular helper cell lymphomas (nTFHLs) represent a new family of peripheral T-cell lymphomas (PTCLs), and comparative studies of their constituents are rare. Methods: This study retrospectively enrolled 10 patients with nTFHL-F and 30 patients with nTFHL-NOS diagnosed between December 2017 and October 2023 at six large comprehensive tertiary hospitals; 188 patients with nTFHL-AI were diagnosed during the same period at the First Affiliated Hospital of Zhengzhou University for comparison. Results: Compared with nTFHL-AI, nTFHL-NOS patients exhibited better clinical manifestations, lower TFH expression levels, and a lower Ki-67 index. However, no differences in clinicopathological features were observed between nTFHL-F and nTFHL-AI patients as well as nTFHL-NOS patients. According to the survival analysis, the median OS for patients with nTFHL-NOS, nTFHL-AI, and nTFHL-F were 14.2 months, 10 months, and 5 months, respectively, whereas the median TTP were 14 months, 5 months, and 3 months, respectively. Statistical analysis revealed differences in TTP among the three subtypes(P=0.0173). Among the population of patients receiving CHOP-like induction therapy, there were significant differences in the OS and TTP among the nTFHL-NOS, nTFHL-AI, and nTFHL-F patients (P=0.0134, P=0.0205). Both the GDPT and C-PET regimens significantly improved the ORR, OS, and PFS in nTFHL patients. Conclusion: There are significant differences in the clinical manifestations, pathology, and survival outcomes among the three subtypes of nTFHLs. However, further research with a larger sample size, and involving clinical pathology and molecular genetics is needed to determine the distinctive biological characteristics of these tumors. Myxobacteria are special bacteria with wide adaptability, which are rich sources of structurally diverse natural products with intriguing biological properties. Here, a gram-negative myxobacterium strain s54d21T was isolated from the sediment of a wetland park in China using the Escherichia coli baiting method. Based on 16S rRNA gene sequence and genomic data, the strain was demonstrated to be a novel species of a rare genus Hyalangium, designated Hyalangium ruber sp. nov (type strain s54d21T = GDMCC 1.1945T = JCM 39263T). The subsequent chemical investigation of the strain s54d21T led to the isolation of three rare 3,5,6-trisubstituted 2(1H)-pyrazinones, namely, hyalanones A-C (1-3), together with a known macrolactin A (4). Those new structures and their absolute configurations were unambiguously assigned by extensive analyses of spectroscopic data and density functional theory (DFT) calculations. In biological assays, compound 4 exhibited moderate cytotoxic activities against human cell lines RKO, A549, and NCM460 with IC50 values ranging from 27.21 to 32.14 µM. [ABSTRACT FROM AUTHOR]

Human pluripotent stem cells (hPSCs) dynamically respond to their chemical and physical microenvironment, dictating their behavior. However, conventional in vitro studies predominantly employ plastic culture wares, which offer a simplified representation of the in vivo microenvironment. Emerging evidence underscores the pivotal role of mechanical and topological cues in hPSC differentiation and maintenance. In this study, we cultured hPSCs on hydrogel substrates with spatially controlled stiffness. The use of culture substrates that enable precise manipulation of spatial mechanical properties holds promise for better mimicking in vivo conditions and advancing tissue engineering techniques. We designed a photocurable polyethylene glycol-polyvinyl alcohol (PVA-PEG) hydrogel, allowing the spatial control of surface stiffness and geometry at a micrometer scale. This versatile hydrogel can be functionalized with various extracellular matrix proteins. Laminin 511-functionalized PVA-PEG gel effectively supports the growth and differentiation of hPSCs. Moreover, by spatially modulating the stiffness of the patterned gel, we achieved spatially selective cell differentiation, resulting in the generation of intricate patterned structures. Valproic acid (VPA) has been widely used as an antiepileptic drug for decades. Although VPA is effective and well-tolerated, long-term VPA treatment is usually associated with hepatotoxicity. However, the underlying mechanisms of VPAcaused hepatotoxicity remain unclear. In this study, a total of 157 pediatric patients with epilepsy were recruited and divided into normal liver function (NLF, 112 subjects) group and abnormal liver function (ABLF, 45 subjects) group. We observed that MTHFR A1298C and MTHFR C677T variants may be linked to VPA-induced liver dysfunction (p = 0.001; p = 0.023, respectively). We also found that the MTHFR A1298C polymorphism was associated with a higher serum Hcy level (p = 0.001) and a lower FA level (p = 0.001). Moreover, the serum Hcy levels was strongly correlated with the GSH and TBARS concentrations (r = -0.6065, P < 0.001; r = 0.6564, P < 0.001, respectively). Furthermore, logistic analysis indicated that MTHFR A1298C/C677T polymorphisms and increased Hcy concentrations may be risk factors for VPA-induced liver dysfunction. These results suggested that individual susceptibility to VPA-induced liver dysfunction may result from MTHFR A1298C/C677T polymorphisms and increased Hcy levels. Segmental and suprasegmental phonological awareness (PA) are closely related to word reading skills in native speakers learning to read an alphabetic script as used in English. However, their roles in English word and pseudoword reading among native Chinese (NC) speakers, and how English proficiency might affect these relationships, remain less clear. This study examined the links between English segmental/suprasegmental PA and word/pseudoword reading in NC and native English (NE) speakers. Both child and adult participants were assessed on English segmental and suprasegmental PA, alongside vocabulary, at a single time point. The results showed that both segmental PA (elision and segmenting nonwords) and suprasegmental PA (aural suffix judgment and written suffix judgment) were significantly correlated with English real word and pseudoword reading of both NE and NC children, and NC adults, but not NE adults. Moreover, for NE and NC children, segmental PA correlated stronger with real word reading than suprasegmental PA after controlling for vocabulary. Among NC adults, both segmental and suprasegmental PA significantly contributed to real word reading. For pseudoword reading, after controlling for vocabulary, segmental PA had a stronger correlation among NC children and adults, while suprasegmental PA was more influential for NE children. This research gives insights into factors influencing NC speakers' English word reading ability, bearing essential implications for enhancing second language literacy in learners from a logographic background. Precipitation infiltration serves as a significant source of groundwater in the Badain Jaran Desert. To investigate variations in precipitation infiltration within the desert, this study collected data on moisture content and temperature from the vadose zone through in-situ field monitoring. Utilizing these data, a numerical model is employed to explore the mechanism of groundwater recharge via precipitation. The results are as follows: (1) Moisture content and temperature in the shallow vadose zone exhibit significant seasonal variations, with moisture content diminishing with increasing depth; (2) Groundwater recharge via precipitation infiltration initially increases and then decreases with groundwater level depth (GWD). Peak groundwater recharge via precipitation occurs at a GWD of 0.75 m, decreasing to merely 0.012 cm at GWDs exceeding 2 m; (3) Groundwater is no longer susceptible to phreatic water evaporation when the GWD reaches approximately 3.7 m. Interferon gamma (IFNγ) is a critical cytokine known for its diverse roles in immune regulation, inflammation, and tumor surveillance. However, while IFNγ levels were elevated in sera of most newly diagnosed acute myeloid leukemia (AML) patients, its complex interplay in AML remains insufficiently understood. We aim to characterize these complex interactions through comprehensive bulk and single-cell approaches in bone marrow of newly diagnosed AML patients. We identify monocytic AML as having a unique microenvironment characterized by IFNγ producing T and NK cells, high IFNγ signaling, and immunosuppressive features. IFNγ signaling score strongly correlates with venetoclax resistance in primary AML patient cells. Additionally, IFNγ treatment of primary AML patient cells increased venetoclax resistance. Lastly, a parsimonious 47-gene IFNγ score demonstrates robust prognostic value. In summary, our findings suggest that inhibiting IFNγ is a potential treatment strategy to overcoming venetoclax resistance and immune evasion in AML patients. [ABSTRACT FROM AUTHOR]
