Freezing of water under nanoconfinement exhibits physical peculiarities with respect to the bulk water. However, experimental observations are extremely challenging at this scale, which limits our understanding of the effect of confinement on water properties upon freezing. In this study, we use molecular dynamic simulations to investigate how confinement affects the kinetics of growth of ice and the thermodynamic equilibrium of ice-liquid coexistence. TIP4P/Ice water model and CSH-FF model were applied to simulate ice crystallization in a confined cement system at temperatures down to 220 K. We adapted an interface detection algorithm and reparameterized the CHILL/CHILL+ algorithm to capture ice growth. The confinement leads to a shift of the maximum growth rate of ice to a higher temperature than for bulk water. Both the confinement and surface impurities contribute to slowing down the ice growth. For the ice-liquid coexistence at equilibrium, we derive a formulation of Thomson's equation adapted to statistical physics quantities accessible by molecular simulation, and we show that this adapted equation predicts accurately the melting line of bulk and confined ice Ih as a function of pressure. The confinement decreases systematically the melting temperature of ice of about 5 K compared with bulk ice Ih. A premelted water film about 1 nm thick is observed between the solid wall and ice, and its thickness is found to decrease continuously as temperature is lowered. We note that the surface impurities are key to the formation of the premelted water nanofilm when the temperature is lower than 250 K. [ABSTRACT FROM AUTHOR]

The Wattle-necked Softshell Turtle (Palea steindachneri) is the only species in the genus Palea, and is listed as a critically endangered species by IUCN Red List due to the extreme decline in its natural populations as a result of human capture and trade activities. However, the lack of studies on molecular markers is not conducive to the genetic diversity assessment and population conservation of P. steindachneri. Here, a total of 68 novel polymorphic SNP markers were developed and characterized based on whole-genome sequencing of P. steindachneri. The observed heterozygosity, expected heterozygosity, and polymorphism information content of the 68 SNP markers ranged from 0.1250 to 0.6875, 0.3047 to 0.5000, and 0.2583 to 0.3750, respectively. Only five SNPs significantly deviated from the Hardy-Weinberg equilibrium (PHWE<0.05). In this paper, a data-driven framework aimed at investigating how weather factors affect the spatio-temporal patterns of air traffic flow in the terminal maneuvering area (TMA) is presented. The framework mainly consists of three core modules, namely, trajectory structure characterization, flow pattern recognition, and association rule mining. To fully characterize trajectory structure, abnormal trajectories and typical operations are sequentially extracted based on a deep autoencoder network with two specially designed loss functions. Then, using these extracted elements as basic components to further construct and cluster per-hour-level descriptions of airspace structure, the spatio-temporal patterns of air traffic flow can be recognized. Finally, the association rule mining technique is applied to find sets of weather factors that often appear together with each flow pattern. Experimental analysis is demonstrated on two months of arrival flight trajectories at Hong Kong International Airport (HKIA). The results clearly show that the proposed framework effectively captures spatial anomalies, fine-grained trajectory structures, and representative flow patterns. More importantly, it also reveals that those flow patterns with non-conforming behaviors result from complex interactions of various weather factors. Current research on apron conflict detection is often limited to the interaction between the aircraft as a whole and other objects, making it difficult to accomplish targeted identification of vulnerable and high‐cost aircraft components. However, the implementation of detailed aircraft identification is of great significance to enhance the safety of airport surface operations. Based on the excellent performance of High‐Resolution Network (HRNet) in keypoint detection, a lightweight end‐to‐end keypoint detection network, namely Squeeze and Excitation High‐Resolution Network (SEHRNet), is proposed in this paper to solve the problems of HRNet's slower computation and more redundancy. First, the errors arising from coordinate transformations in the coding and decoding process are solved by an improved feature map coding and decoding process. Second, replace the BasicBlock in HRNet with the Depthwise separable convolutions based on the Squeeze‐and‐Excitation Networks, which drastically cuts the computational cost of the network. Third, the improved Bottleneck module is used to further enhance the capability of feature extraction. Experimental results prove that, based on the aircraft keypoint detection dataset, the SEHRNet proposed in this paper shows stronger applicability compared to the current mainstream networks. Rheumatoid arthritis (RA) is an autoimmune disease that significantly impacts quality of life by disrupting CD4+ T cell immune homeostasis. The identification of a low-side-effect drug for RA treatment is urgently needed. Our previous study suggests that Trichinella spiralis paramyosin (Ts-Pmy) has immunomodulatory effects, but its potential effect on CD4+ T cell response in RA remains unclear. In this study, we used a murine model to investigate the role of rTs-Pmy in regulating CD4+ T cell differentiation in collagen-induced arthritis (CIA). Additionally, we assessed the impact of rTs-Pmy on CD4+ T cell differentiation towards the Th1 and Th17 phenotypes, which are associated with inflammatory responses in arthritis, using in vitro assays. The results demonstrated that rTs-Pmy administration reduced arthritis severity by inhibiting Th1 and Th17 response while enhancing Treg response. Prophylactic administration of Ts-Pmy showed superior efficacy on CIA compared to therapeutic administration. Furthermore, in vitro assays demonstrated that rTs-Pmy could inhibit the differentiation of CD4+ T cells into Th1 and Th17 while inducing the production of Tregs, suggesting a potential mechanism underlying its therapeutic effects. This study suggests that Ts-Pmy may ameliorate CIA by restoring the immune balance of CD4+ T cells and provides new insights into the mechanism through which helminth-derived proteins exert their effects on autoimmune diseases. Background: The Asian yellow pond turtle (Mauremys mutica) is an important commercial freshwater aquaculture species in China. This species is a highly sexually dimorphic species, with males growing at a faster rate than females and exhibits temperature-dependent sex determination (TSD), in which the incubation temperature during embryonic development determines the sexual fate. However, the mechanisms of the sex determination or sex differentiation in the Asian yellow pond turtle are remain a mystery. Results: Temperature-specific gonadal transcriptomics of the Asian yellow pond turtle were performed during the thermosensitive period (stage 15) using RNA-seq technology to identify candidate genes that initiate gonadal differentiation. We uncovered candidates that were the first to respond to temperature. These candidates were sexually dimorphic in expression, reflecting differences in gonadal (Cirbp, Runx1) and germline differentiation (Vasa, Nanos1, Piwil2), gametogenesis (Hmgb3, Zar1, Ovoinhibitor-like, Kif4), steroid hormone biosynthesis (Hsd17b5, Hsd17b6), heat shock (Dnajb6, Hsp90b1, Hsp90aa1) and transient receptor potential channel genes (Trpm1, Trpm4, Trpm6, Trpv1). Conclusions: Our work will provide important genetic information to elucidate the mechanisms of sex control in the Asian yellow pond turtles, and will contribute important genetic resources for further studies of temperature-dependent sex determination in turtles. To promote the application of automated vehicles in large airports, in this study, we present an integrated optimization method for scheduling Unmanned follow-me cars. The scheduling process is divided into three phases: Dispatch, Guidance, and Recycle. For the Dispatch phase, we establish a vehicle assignment model, to allocate the vehicle resource equitably. For the Guidance phase, we offer an quantitative way, to measure the spacing between Unmanned follow-me car and aircraft. To optimize the efficiency of airport operation in the three phases and ensure safety, the collaborative planning model, and the conflict prediction model are established. An improved grey wolf optimization algorithm is adopted to enhance the convergence speed and generalization performance. A case study at Ezhou Huahu Airport in China demonstrates the effectiveness of the methods. The results show that the model of collaborative planning can make the balance of path selection, Unmanned follow-me car's working time, and departure sequence. The convergence speed of the improved algorithm has been increased by 18.75%. The inequity index of vehicle assignment is only 0.015731, and the spatiotemporal distribution of conflicts is influenced by the airport's surface layout. [ABSTRACT FROM AUTHOR]

The spectrum is a key environmental factor, and light-emitting diodes (LEDs) can influence the growth and development of crustaceans by altering the composition of the spectrum. This study conducted a 30-day experiment to investigate the effects of five LED spectra (red, yellow, blue, green, and white light) on the growth, antioxidant and immune enzyme activities, stress hormone levels, and the expression of α-amylase (α-AMY), ecdysone receptor (EcR) and retinoid X receptor (RXR) genes in juvenile redclaw crayfish (Cherax quadricarinatus). The results show that the survival rate of juveniles is markedly higher in the yellow and red-light groups than in the other three groups (P < 0.05). The green light group exhibits the lowest survival rate, yet it demonstrates the highest weight gain rate and specific growth rate. Regarding enzyme activity and hormone levels, the yellow light group shows the lowest malondialdehyde content, with higher superoxide dismutase and acid phosphatase activity than the other groups; no significant differences are observed in lysozyme activity among the groups (P > 0.05). The melatonin content in the green and blue light groups is significantly higher than that in the other three groups (P < 0.01). In terms of growth gene expression, the expression of α-AMY, EcR, and RXR in juvenile C. quadricarinatus is regulated by the spectrum. In conclusion, when raised under the yellow light spectrum, juvenile C. quadricarinatus displays elevated survival rates, rapid growth, and robust antioxidant and immune defenses. Aims: Soil organic carbon (SOC) stability greatly affects soil carbon sequestration capacity and carbon emissions, is related to soil bacterial communities, but it remains unclear whether such relationships exist in alpine wetland biogeomorphic succession. Methods: Here, we used a space-for-time approach across hydrological gradients, and alpine wetland succession can be divided into three types: "Swamp" (S), "Swamp Meadow" (SM), and "Meadow" (M) to examine the dynamics change of SOC fractions and bacterial communities. Results: The succession from S-SM-M resulted in decreased SOC stability (SM and M decreased by 18.65% and 31.88% compared to S). Furthermore, the decrease in SOC stability was primarily due to the decrease in soil moisture during succession and moisture-induced changes in bacterial community structure. e.g., the relative abundance of Proteobacteria decreased significantly by 40.99% and 52.87% in SM and M, while the relative abundance of Acidobacteria increased by 43.49% and 185.04% in SM and M compared to S. Co-occurrence network and random forest analyses further identified that changes in keystone taxa of bacteria, including Deltaproteobacteria, Acidimicrobiia, Bacteroidia and Gammaproteobacteria, were associated with SOC stability. Conclusions: In conclusion, bacteria shifted their community composition to reduce SOC stability in response to soil moisture variations during succession, and underscores the importance of identifying keystone taxa that are crucial for maintaining SOC stability. Simple Summary: The DKK gene family plays an important role in immune mechanisms and embryonic development, but studies on Pelodiscus sinensis have not been reported. In this study, for the first time in the context of P. sinensis, we analyzed the relevant features of the DKK gene family using bioinformatics methods and constructed transcriptional profiles and tissue expression profiles. The DKK1 and DKK3 genes were found to be highly expressed in the ovaries, while DKKL1 and DKK4 were significantly differentiated in the testes. In this study, we preliminarily investigated the function of the DKK gene family in the context of P. sinensis, and this study lays the foundation for further systematic research on the molecular mechanism of sex differentiation in P. sinensis. The DKK family is a canonical small family of WNT antagonists. Though recent studies have suggested that the DKK gene family may be involved in sex differentiation in Pelodiscus sinensis, there are still a lot of things about the DKK gene family that we do not know. In this study, we used bioinformatics methods to identify members of the DKK gene family in P. sinensis and analyzed their phylogeny, covariance, gene structure, structural domains, promoter conserved sites, signal peptides, gonadal transcription factors, transcriptional profiles, and tissue expression profiles. Additionally, qRT-PCR results were utilized for the validation and preliminary investigation of the function of the DKK gene family in P. sinensis. The results showed that the DKK gene family is divided into six subfamilies, distributed on six different chromosomal scaffolds containing different gene structures and conserved motifs with the same structural domains, and all of the members were secreted proteins. Our transcriptional profiling and embryonic expression analysis showed that DKKL1 and DKK4 were significantly expressed in the testes, whereas DKK1 and DKK3 were significantly upregulated in the ovaries. This study aims to unveil the spatiotemporal evolution of congestion within terminal airspace, offering an in-depth analysis of congestion concerns to effectively utilize airspace resources and devise targeted control strategies, thereby enhancing airspace operation safety and efficiency. Initially, converting segment flow rates into equivalent speeds serves as a quantitative benchmark for operational status. Subsequently, an enhanced version of the ST-Moran's I index model, specifically tailored to terminal airspace, is developed by incorporating improvements across spatial weight matrices, standard state parameters, and temporal dimensions. Validating this model with actual operational data from Chengdu's terminal airspace, the research demonstrates significant advancements. Compared to conventional models, the proposed model enhances recognition rates for congestion in spatial and temporal dimensions by 62.5% and 43.61%, respectively. Congestion within terminal airspace predominantly occurs at the intersection of departure-climb and approach-departure segments, exhibiting evident spatiotemporal migration behavior. Environmentally friendly powder coatings which have the advantages of being VOC-free, low-cost, and high-efficiency with a high recovery rate have been attracting increasing research attention. The introduction of antibacterial agents into the powder coatings endows them with a capacity to kill bacteria and viruses on the surface of objects; additionally, this enables them to inhibit the indirect transmission of pathogenic microorganisms. Silver, possessing broad-spectrum, strong, and stable antibacterial properties, is considered to be a promising antibacterial material for use in coating applications. Carrier materials for active silver play an important role in its activity and stability. However, there is a lack of systematic studies on the effects of different types of carriers in such coating systems, especially in green powder coating systems. In this paper, we investigated two types of carriers for active silver agents: zeolite, i.e., Linde type A (LTA) zeolite and Y-type zeolite; clay-based materials, i.e., montmorillonite and vermiculite. All the agents showed high antibacterial activity, with antibacterial rates of over 99% as compared to commercial agents. Among the four agents, the Ag-LTA zeolite antimicrobial agent showed a reduction rate of over 99.99%; additionally, it maintained a reduction rate of 99% after seven washing cycles. Thus, this agent was demonstrated to have the highest effectiveness and high durability; these features can be attributed to the high silver content and small particle size. The LTA zeolite also provides a protective effect for silver ions, protecting them from reduction, due to the restriction of elemental silver formation within the confined interior space of the α-cage structure. The Y-type zeolite antimicrobial agent exhibited a slightly lower antimicrobial performance due to its higher silicon-to-aluminum ratio and its lower cation exchange capacity. Comparatively, antimicrobial agents utilizing clay-based carriers have lower cation exchange capacity, resulting in poorer antimicrobial effectiveness than zeolite carriers. In addition, silver loaded on clay-based materials is prone to detach from the carrier and undergo a reduction reaction, making the coating yellowish in color. This study first provides information on the roles of different types of carriers in powder coating systems; then, this information guides the selection of carriers for active silver for the development of efficient antimicrobial agents and coatings. We selected seven females with good reproductive ability, which provided 16.94% of the annual contributions, while there were two females that had no offspring in four years. We also analysed the possible reasons for this difference and the importance of carrying out a family survey. Simple Summary: In aquaculture, the survival and health of aquatic organisms are influenced greatly by external environmental factors. Photoperiod is one of the key environmental factors that affect the early growth, survival, metabolism, and immune system of crustaceans significantly. The aim of the present study was to investigate the impact of photoperiod on the growth, survival, enzyme activity, body color, and expression of growth-related genes in juvenile redclaw crayfish. Five photoperiod experimental groups were established (0L:24D, 6L:18D, 12L:12D, 18L:6D, and 24L:0D), each with three replicates, and reared for 30 days. The group with an 18L:6D photoperiod had higher juvenile crayfish survival rates, better growth performance, stronger antioxidant stress response, and immune defense capabilities. Additionally, artificial photoperiods altered the body color of the redclaw crayfish. The results of the present study lay a foundation for the elucidation of the mechanisms underlying photoperiod regulation of the physiological and biochemical responses of juvenile redclaw crayfish. Through a 30-day experiment, this study investigated the effects of five photoperiods (0L:24D, 6L:18D, 12L:12D, 18L:6D, and 24L:0D) on the survival, enzyme activity, body color, and growth-related gene expression of redclaw crayfish (Cherax quadricarinatus) juveniles. The results showed that C. quadricarinatus juveniles under 18L:6D and 24L:0D photoperiods exhibited the highest survival rate, which was significantly higher than the survival rates of juveniles under the other three photoperiods (p < 0.05). However, the 0L:24D group had the highest final body weight and weight gain rate, significantly surpassing those of the 12L:12D, 18L:6D, and 24L:0D groups (p < 0.05). Regarding enzyme activity and hormone levels, juveniles under the 18L:6D photoperiod exhibited relatively higher activity of superoxide dismutase (SOD), acid phosphatase (ACP), and lysozyme (LZM) enzymes than those under other photoperiods, but their levels of melatonin and cortisol were relatively low. In addition, the 24L:0D group showed the highest malondialdehyde (MDA) content. Analysis of gene expression levels revealed that retinoid X receptor (RXR) and α-amylase (α-AMY) genes in C. quadricarinatus juveniles exhibited significantly higher expression levels under the 18L:6D photoperiod than those under the other four photoperiods (p < 0.05). With increasing daylight exposure, the body color of C. quadricarinatus changed from pale blue to yellow–brown. In summary, C. quadricarinatus juveniles achieved high survival rates, good growth performance, strong antioxidant stress response, and immune defense capabilities under an 18 h photoperiod. Therefore, in the industrial seedling cultivation of redclaw crayfish, it is recommended to provide 18 h of daily light. Further, the study demonstrated the ability to manipulate the body color of C. quadricarinatus through controlled artificial photoperiods. Male and female Chinese soft-shelled turtles (Pelodiscus sinensis) have sex-dimorphic growth patterns, and males have higher commercial value because of their larger size and thicker calipash. Thus, developing sex-specific markers is beneficial to studies on all-male breeding in P. sinensis. Here, we developed an accurate and efficient workflow for the screening of sex-specific sequences with ZW or XY sex determination systems. Based on this workflow, female and male P. sinensis reference genomes of 2.23 Gb and 2.26 Gb were obtained using de novo assembly. After aligning and filtering, 4.01 Mb female-specific sequences were finally identified. Subsequently, the seven developed sex-specific primer pairs were 100% accurate in preliminary, population, and embryonic validation. The presence and absence of bands for the primers of P44, P45, P66, P67, P68, and P69, as well as two and one bands for the PB1 primer, indicate that the embryos are genetically female and male, respectively. NR and functional annotations identified several sex-determining candidate genes and related pathways, including Ran, Eif4et, and Crkl genes, and the insulin signaling pathway and the cAMP signaling pathway, respectively. Collectively, our results reveal that a ZW-type sex-determination system is present in P. sinensis and provide novel insights for the screening of sex-specific markers, sex-control breeding, and the studies of the sex determination mechanism of P. sinensis. The Asian giant softshell turtle Pelochelys cantorii is one of the largest aquatic turtles in China and has been designated a First Grade Protected Animal in China. To advance conservation research, a combination of Illumina short-read, PacBio long-read, and Hi-C scaffolding technologies was used to develop a high-quality chromosome-level genome assembly for P. cantorii. A total of 262.77 Gb of clean data were produced (121.6 × depth) and then the genome was assembled into 2.16 Gb with a contig N50 of 41.44 Mb and scaffold N50 length of 120.17 Mb, respectively. Moreover, about 99.98% assembly genome sequences were clustered and ordered onto 33 pseudochromosomes. Genome annotation revealed that 21,833 protein-coding genes were predicted, and 96.40% of them were annotated. Salinity stress can trigger a series of physiological changes. However, the mechanism underlying the response to acute salinity stress in Macrobrachium rosenbergii remains poorly understood. In this study, osmoregulation, physiological metabolism, antioxidant capacity, and apoptosis were examined over 96 h of acute salinity stress. Hemolymph osmolality increased with increasing salinity. After 48 h of salinity exposure, the glucose, triglycerides, total protein, and total cholesterol contents in two salinity stress groups (13 and 26‰ salinity) were significantly lower than those in the 0‰ salinity group. The highest levels of these parameters were detected at 6 h; however, superoxide dismutase (SOD), total antioxidant capacity (T-AOC), and malondialdehyde (MDA) were the lowest at 96 h in the 13‰ salinity group. The activity of immunity-related enzyme alkaline phosphatase (AKP) showed a decreasing trend with increasing salinity and remained at a low level in the 26‰ salinity group throughout the experiment. No significant differences were observed in aspartate aminotransferase (AST), alanine aminotransferase (ALT), or lysozyme (LZM) among the three treatments at 96 h. After 96 h of salinity treatments, the gill filament diameter significantly decreased, and a more pronounced terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL)-positive signal was detected in the 13‰ and 26‰ groups compared to that in the 0‰ group. Expression levels of apoptosis-related genes, including Cysteine-aspartic acid protease 3 (Caspase 3), Cysteine-aspartic acid protease 8 (Caspase 8), Cytochrome c (Cyt-c), tumor suppressor gene (P53), Nuclear factor kappa-B (NF-κB), and B cell lymphoma 2 ovarian killer (Bok) were significantly higher in the 26‰ salinity group than in the other groups at 24 h, but lower than those in the 0‰ salinity group at 96 h. Cyt-c and P53 levels exhibited a significantly positive relationship with MDA, AST, and LZM activity during salinity stress. In the 13‰ salinity group, Bok expression was significantly correlated with SOD, T-AOC, AKP, acid phosphatase, and LZM activity, whereas in the 26‰ group, the AST content was positively correlated with Caspase 8, Cyt-c, and P53 expression. A significant negative relationship was observed between Caspase 3 expression and catalase (CAT) activity. These findings provide insight into the mechanisms underlying the response to acute salinity stress and will contribute to improving M. rosenbergii aquaculture and management practices. Objectives: Lung adenocarcinoma (LUAD) exhibits a higher fatality rate among all cancer types worldwide, yet the precise mechanisms underlying its initiation and progression remain unknown. Mounting evidence suggests that long non-coding RNAs (lncRNAs) exert significant regulatory roles in cancer development and progression. Nevertheless, the precise involvement of lncRNA CYP4A22-AS1 in LUAD remains incompletely comprehended. Methods: Bioinformatics analyses evaluated the expression level of CYP4A22-AS1 in lung adenocarcinoma and paracancer. The LUAD cell line with a high expression of CYP4A22-AS1 was constructed to evaluate the role of CYP4A22-AS1 in the proliferation and metastasis of LUAD by CCK8, scratch healing, transwell assays, and animal experiments. We applied transcriptome and microRNA sequencing to examine the mechanism of CYP4A22-AS1 enhancing the proliferation and metastasis of LUAD. Luciferase reporter gene analyses, west-blotting, and qRT-PCR were carried out to reveal the interaction between CYP4A22-AS1, miR-205-5p/EREG, and miR-34c-5p/BCL-2 axes. Results: CYP4A22-AS1 expression was significantly higher in LUAD tissues than in the adjacent tissues. Furthermore, we constructed a LUAD cell line with a high expression of CYP4A22-AS1 and noted that the high expression of CYP4A22-AS1 significantly enhanced the proliferation and metastasis of LUAD. We applied transcriptome and microRNA sequencing to examine the mechanism of CYP4A22-AS1 enhancing the proliferation and metastasis of LUAD. CYP4A22-AS1 increased the expression of EREG and BCL-2 by reducing the expression of miR-205-5p and miR-34-5p and activating the downstream signaling pathway of EGFR and the anti-apoptotic signaling pathway of BCL-2, thereby triggering the proliferation and metastasis of LUAD. The transfection of miR-205-5p and miR-34-5p mimics inhibited the role of CYP4A22-AS1 in enhancing tumor progression. Simple Summary: We explored the effect of maternal size on offspring size and number using a 4-year dataset based on paternity analyses in Asian yellow pond turtle, Mauremys mutica, and found that the maternal size significantly affected the offspring number but not offspring size. Our results suggest that the offspring size-number trade-off theory does not apply to cultured M. mutica and also provides basic data for the efficient artificial breeding management of M. mutica in captivity. The relationship between this trade-off and maternal size has been explored in several turtle species, except for the Asian yellow pond turtle, Mauremys mutica. To investigate how the maternal condition affects offspring size and number, we explored the relationships among the maternal body size and the number and size of cultured M. mutica hatchlings using a 4-year dataset. Our results showed that different females not only produced different sizes of offspring but also produced different numbers of offspring. No trade-off in egg size number was detected. According to regression analysis, we did not find that the maternal body size significantly influenced the offspring mass; however, we detected that the offspring size was significantly correlated with the clutch size and maternal age. The mean body mass of offspring increased with maternal age, and the clutch size varied significantly over four years, which was correlated with offspring size, maternal body size and age. However, the number of offspring per female increased with the maternal plastron length rather than age. Our results were inconsistent with the optimal offspring size theory in that females did not increase their offspring size but rather increased the offspring number to increase their fitness, which will also provide a basis for the efficient cultivation management of turtles. Different weakening buffer operators in a time-series model analysis usually result in different model sensitivities, which sometimes affect the effectiveness of relevant operator-based methods. In this paper, the stability of two classic fractional-order weakening buffer operator-based series models is studied; then, a new data preprocessing method based on a novel fractional-order bidirectional weakening buffer operator is provided, whose effect in improving the model's stability is tested and utilized in prediction problems. Practical examples are employed to demonstrate the efficiency of the proposed method in improving the model's stability in noise scenarios. The comparison indicates that the proposed method overcomes the disadvantage of many weakening buffer operators in the subjectively biased weighting of the new or old information in forecasting. Additionally, compared to hemocytes in the active state, several upregulated differentially expressed immune-related genes, such as stat1, traf3, and socs6, were identified in hemocytes during the hibernation period, thus indicating the important immune function of hemocytes. The effects of biochar on soil improvement have been widely confirmed, but its influence on soil microorganisms is still unclear. Elucidating the complex relationship and the community assembly processes of microorganisms under biochar addition is important to understand the ecological effects of this substance. We performed a one-time addition of biochar on aeolian soils and planted maize (Zea mays L.) continuously for 7 years. Afterwards, soil samples were collected, and the 16S/ITS rRNA gene sequencing technology was used to study changes in microbial community structure, network characteristics, and community assembly processes in the aeolian soils. We found that biochar addition significantly increased the maize yield and changed the soil microbial community composition (β-diversity), but had no significant effect on the microbial α-diversity. The addition of 31.5–126.0 Mg ha−1 of biochar led to a reduction of the rhizosphere bacterial network's edge number, average degree, and robustness, but had no significant effect on the fungal network properties. The bacterial community was controlled by deterministic processes, while fungi were mainly controlled by stochastic processes. The addition of 126.0 Mg ha−1 of biochar led to a transformation of the bacterial community's assembly processes from deterministic to stochastic. These results indicate that the stability of the rhizosphere bacterial community's complex network in aeolian soils diminishes under biochar addition, together changed the bacterial community's assembly processes. Fungi can instead effectively resist the environmental changes brought by biochar addition, and their network remains unchanged. These findings help clarify the effect of biochar addition on microbial interaction and assembly processes in aeolian soils characteristic of arid regions. Apron safety is critical to aviation operations and aprons are prone to wingtip scraping accidents. However, few studies have been conducted on wingtip conflicts. In this paper, a class of aircraft conflict detection models based on wingtip keypoint detection is devised to solve the problem of wingtip scraping that may occur during the aircraft taxi‐in process on the apron. Firstly, a concise camera calibration technique is designed for the conversion between the pixel coordinate and the airport actual coordinate, which allows for the acquisition of high accuracy activity posture within the apron. Secondly, the static and dynamic safety protection zone delineation methods for wingtip conflict detection are given, taking the apron standard operating procedures and aircraft kinematic model into consideration. Then, the corresponding conflict detection models and algorithms are designed for the possible scraping between the vehicle and aircraft wingtips and between the aircraft and aircraft wingtips during the aircraft taxi‐in process on the apron. Finally, HRNet is selected to detect the keypoint of the wingtip by comparison, and the corresponding models and algorithm validation are carried out based on the apron physical sandbox. The safe utilization and risk assessment of produced water (PW) from oil and gas fields for desert irrigation have received increasing attention in recent years. In this context, this study aimed to analyze structural changes in soil bacterial community, and assess the environmental impact of PW discharge and irrigation over time. High-throughput sequencing technology was employed to examine the structure of the soil bacterial community in the constructed wetland and its surrounding desert vegetation irrigation region where PW was released for a considerable amount of time (30 years). The results revealed that long-term discharge of PW and irrigation significantly reduced the abundance of the soil bacterial community but did not significantly alter the richness and diversity of the soil bacterial community. Proteobacteria was the dominant bacterial phyla in soil, but in irrigated and drained areas, the dominant bacterial phyla changed from Alphaproteobacteria to Gammaproteobacteria, the Firmicutes abundance was significantly reduced. Simple Summary: Pelochelys cantorii is critically endangered and rarely seen in the wild, and only 13 adults are being kept in captivity in China. For the purpose of reinforcing the conservation and management of P. cantorii, the Pearl River Fisheries Research Institute (Chinese Fishery Academy of Sciences) successfully bred more than 800 turtles from 2015 to 2020. In this study, we developed and characterized 10 simple sequence repeat markers from the RNA transcriptome of P. cantorii, established two multiplex PCR systems, and obtained the genetic structure and genetic diversity of the artificially conserved population. The aim was to obtain viable second-generation P. cantorii with the highest genetic diversity to implement population recovery plans for this species. To understand the genetic structure of the protected turtle species Pelochelys cantorii we used transcriptome data to design more than 30,000 tri- and tetranucleotide repeat microsatellite primer pairs, of which 230 pairs were used for laboratory experiments. After two screenings, only 10 microsatellite markers with good specificity, high amplification efficiency, and polymorphisms were obtained. Using the selected primers, two multiplex PCR systems were established to compare and analyze the genetic diversity of artificially assisted breeding generations from four parents (two females and two males) continuously bred over two years. A total of 25 alleles were detected among the 10 microsatellite loci of the offspring. The polymorphism information content (PIC) was 0.313–0.674, with an average of 0.401, among which two loci were highly polymorphic (PIC ≥ 0.5). The number of alleles was 2–5 and the number of effective alleles was 1.635–3.614. The observed heterozygosity was 0.341–0.813, with an average of 0.582, whereas the average expected heterozygosity was 0.389–0.725, with an average of 0.493. Moreover, on the basis of Nei's genetic distance and the Bayesian clustering algorithm, the 182 offspring were divided into two subgroups, and the results corresponded to the two maternal lines. This is the first study to investigate the molecular markers of P. cantorii, and the results obtained can be used to protect genetic resources and provide a genetic basis for the design of population recovery plans. HMGB2, a member of the high-mobility group (HMG) proteins, was identified as a male-biased gene and plays a crucial role in the germ cells differentiation of mammals. However, its role in spermatogenesis of turtle is still poorly understood. Here, we cloned the Pelodiscus sinensis HMGB2 and analyzed its expression profile in different tissues and in testis at different developmental ages. P. sinensis HMGB2 mRNA was highly expressed in the testis of 3-year-old turtles (P < 0.01), but was hardly detected in ovaries and other somatic tissues. The results of chemical in situ hybridization (CISH) showed that HMGB2 mRNA was specifically expressed in germ cells, where it was mainly distributed in round spermatids and sperm, but not detected in somatic cells, spermatogonia, primary spermatocytes, or secondary spermatocyte. The relative expression of HMGB2 also responded to seasonal changes in testis development in P. sinensis. In different seasons of the year, the relative expression of HMGB2 transcripts in the testis of 1 year and 2 year olds showed an overall upward trend, whereas, in the testis of 3 year old, it peaked in July and then declined in October. Moreover, in April and July, with an increase in ages, the expression of HMGB2 transcripts showed an upward trend. In response to the requirements on high flexibility of powder coatings before molding process, this paper investigates the effects of polyester resin, curing agents, flexibilizer and substrates on the film properties. By means of fomulation optimization, polyester powder coatings with high flexibility are prepared. The cell line, designated as YTO2, was then characterized through the analysis of gene expression profiles. The transcriptome analysis and the reverse transcription polymerase chain reaction (RT-PCR) showed that the cells expressed germline genes such as tdrd7, nanos1, klf5, igtb1, hsd17b4 and rad51. Moreover, the immunostaining showed that the germ cell markers, Tdrd7 and Rad51 proteins, were detected predominant in cytoplasm of perinuclear region, while proliferation marker, PCNA, was dominantly observed in the nuclei of cultured cells. Intriguingly, the cells could respond to the retinoic acid induction with significantly increasing the expression level of some meiosis genes, including vasa, dazl, figla, and dmc1. Furthermore, YTO2 cells could be efficiently transfected with the pHBAd-BHG-EGFP adenovirus and properly expressed the exogenous genes. To sum up, an ovarian cell line of the Asian yellow pond turtle had been established and could be stably propagated under in vitro culture condition, as well as being capable of efficiently expressing the exogenous gene tdrd7. Nanog is a homeodomain-containing transcription factor, and it plays a vital role in maintaining the pluripotency of embryonic stem cells; there is a lack of reporting on the Nanog gene in reptile. In this study, the cDNA sequence and promoter of a Nanog gene was identified in P. sinensis, designed as PsNanog. PsNanog gene products were mainly expressed in gonads and were especially rich in ovary. Particularly, PsNanog mRNA and protein was exclusively expressed in germ cells in ovary and testis. Collectively, this is the first report on the gene structure and expression of Nanog in turtle, which may lay a foundation for further functional analysis of the Nanog in turtle, even in the reptiles. Further, PsNanog probably has the conserved and divergent function in regulating cell pluripotency and germ cell development in turtle, compared with other species, including teleosts and mammals. However, there is lack of reporting on the Nanog gene in reptile. Here, we identified a 1032 bp cDNA sequence of a Nanog gene in Pelidiscus sinensis, known as PsNanog. PsNanog has a highly conserved HD domain and shares a high identity with that of Chelonia mydas and the lowest identity with Oryzias latipes. Similarly, PsNanog presented a tight cluster with C. mydas Nanog, but was far from those of teleosts. Additionally, we cloned a length of 1870 bp PsNanog promoter. Dual luciferase assay showed that the DNA fragment of −1560 to +1 exhibited a high promoter activity. The RT-PCR and RT-qPCR results showed that PsNanog was predominantly expressed in ovary, and then in testis. The in situ hybridization and immunohistochemical analysis showed that PsNanog was expressed in the early primary oocytes and the cytoplasm of the cortical region of stage VIII oocytes in ovary, and distributed in most stages of germ cells in testis. Spats1 acts a key function in germ cell differentiation in mammals, yet its study in P. sinensis has not been reported. To initially explore the expression of Spats1 in P. sinensis and its response to sex steroid treatment, we cloned the CDS of Spats1 for the first time and analyzed its expression profile in different tissues, including the testes in different seasons. The Spats1 cDNA fragment is 1201 base pairs (bp) in length and contains an open reading frame (ORF) of 849 bp, which codes for 283 amino acids. Spats1 mRNA was highly expressed in the testes (p < 0.01) and barely detectable in other tissues. In P. sinensis, the relative expression of Spats1 also responsive to seasonal changes in testis development. In summer (July) and autumn (October), Spats1 gene expression was significantly higher in the testes than in other seasons (p < 0.05). Spats1 mRNA was found to be specifically expressed in germ cells by chemical in situ hybridization (CISH), and it was mainly located in primary spermatocytes (Sc1), secondary spermatocytes (Sc2) and spermatozoa (St). Spats1 expression in embryos was not significantly changed after 17α-methyltestosterone (MT)and 17β-estradiol (E2) treatment. In adults, MT significantly induced Spats1 expression in male P. sinensis. However, the expression of Spats1 in testes was not responsive to E2 treatment. In addition, the expression of Spats1 in females was not affected by either MT or E2 treatment. The Chinese soft-shelled turtle (Pelodiscus sinensis) exhibits sexual dimorphism in adults and the absence of sexually dimorphic morphological characteristics in the long-lived juvenile stage. The development of sex-linked SNP markers is of great significance for the improvement and replenishment of genetic resources. Here, we developed and characterized 35 novel polymorphic SNP markers based on the gonadal transcriptome of P. sinensis. The polymorphism information content, observed heterozygosity and expected heterozygosity of the 35 SNP markers ranged from 0.023 to 0.473, 0.012 to 0.547 and 0.023 to 0.503, respectively, and 10 of the SNPs significantly deviated from Hardy–Weinberg equilibrium (PHWE < 0.01). Two sex-linked SNPs were found to be significantly associated with sex phenotypes and showed 100% concordance across all tested individuals. Overall, 7833 DE mRNAs, 619 DE lncRNAs, 231 DE circRNAs, and 520 DE miRNAs were identified. Some "star genes" associated with sex differentiation containing dmrt1, sox9, and foxl2 were identified. Additionally, some potential genes linked to sex differentiation, such as bmp2, ran, and sox3, were also isolated in P. sinensis. Functional analysis showed that the DE miRNAs and DE ncRNAs were enriched in the pathways related to sex differentiation, including ovarian steroidogenesis, the hippo signaling pathway, and the calcium signaling pathway. Remarkably, a lncRNA/circRNA–miRNA–mRNA interaction network was constructed, containing the key genes associated with sex differentiation, including fgf9, foxl3, and dmrta2. Collectively, we constructed a gender dimorphism profile of the female and male gonads of P. sinensis, profoundly contributing to the exploration of the major genes and potential ncRNAs involved in the sex differentiation of P. sinensis. Salinity is a crucial factor influencing the growth, development, immunity, and reproduction of aquatic organisms; however, little is known about the molecular mechanism of the response to salinity challenge in larvae of the giant freshwater prawn Macrobrachium rosenbergii. Herein, larvae cultured in three treatment groups with salinities of 10, 13, and 16‰ (S10, S13, and S16) were collected, and then transcriptome analysis was conducted by RNA-seq. A total of 6,473, 3,830 and 3,584 differentially expressed genes (DEGs) were identified in the S10 vs. S13 comparison, S10 vs. S16 comparison and S13 vs. S16 comparison, respectively. These genes are involved in osmoregulation, energy metabolism, molting, and the immune response. qPCR analysis was used to detect the expression patterns of 16 DEGs to verify the accuracy of the transcriptome data. Protein–protein interaction (PPI) analysis for DEGs and microsatellite marker screening were also conducted to reveal the molecular mechanism of salinity regulation. Based on the study of traditional traffic scheduling techniques and key techniques of genetic algorithms, this paper participates in the actual project of genetic algorithm-based traffic scheduling, analyzes the requirements of the project, focuses on the design and implementation of the traffic scheduling algorithm module in the genetic algorithm-based traffic scheduling system, and conducts further research on the pathfinding by constraints submodule. In this paper, the flight approach and departure sequencing problem and runway allocation problem are the main research objects. The dynamic optimal scheduling model of flight approach and departure is established by considering the interests and demands of airlines and airports, and a new scheduling algorithm is proposed. In this paper, a brief introduction to the airport terminal area is given, and the feasibility of the approach/departure optimal scheduling is introduced from the perspective of airlines with a long-range parallel two-runway airport as the research background. Secondly, through the analysis of the flight approach and departure process and the study of the approach and departure cooperative optimization strategy, a single-runway flight approach and departure traffic scheduling model under the joint sequencing strategy is established with the optimization objective of minimizing the total flight delay time, and the model is solved by using the sliding time window algorithm. Then, based on the single-runway scheduling model, a multirunway multiobjective flight optimal scheduling model is established with the objectives of minimizing total delay time, increasing runway throughput per unit time and fairness of flight delay time allocation, and a dynamic algorithm (STW-GA) combining sliding time window algorithm and dual-structured chromosome genetic algorithm is proposed to solve the model. The south slope of Fushun west open-pit mine located in Northeast China is a high-deep rock slope. In the last few years, a landsliding block has gradually formed in the south slope. Owing to the effect of sliding force caused by the sliding block, some damage has occurred on the foundation of Kengkou refinery which constructed in the west of the south slope. To improve the stability of the refinery foundation, three group micro-pile structures were employed to cut off the drag of the sliding block and reinforce the refinery on the southwest slope. In this study, a comprehensive method combining the microseismic monitoring technique and numerical simulation method was used, the stability of the southwest slope was evaluated, and reinforcement mechanism was deeply analyzed. A microseismic monitoring system is established on-site to obtain useful information about the damage energy, location, and magnitude of the rocks in the southwest slope. Thus, the west boundary of the landslide in the south slope of the open-pit is determined. By analyzing the numerical results, the stress concentration is discovered mainly around the micro-pile structures and near the west boundary. After reinforcement, the displacement decrease in the south-north direction is apparent. To investigate the interaction mechanism between the micro-pile and the surrounding rock mass, a local 3D numerical model is employed to help understand auxiliary information about the stress state and stress redistribution induced by failure in the surrounding rock mass. It is shown that the reinforcement effect becomes more and more evident with the increase in the number of employed micro-piles. The numerical results and in situ GPS displacement monitoring data illustrate that the micro-pile structures have an excellent strengthening result in controlling the deformation of the slope. Although a large number of studies have reported the importance of microbial communities in terrestrial ecosystems and their response to nitrogen (N) application, it is not clear in arid alpine wetlands, and the mechanisms involved need to be clarified. Therefore, the response of the soil microbial community in a swamp meadow to short-term (1 year) N application (CK: 0, N1: 8, N2: 16 kg⋅N⋅hm–2⋅a–1) was studied using 16S/ITS rRNA gene high-throughput sequencing technology. Results showed that N application had no significant effect on soil microbial community diversity, but significantly changed soil bacterial community structure. N1 and N2 treatments significantly reduced the relative abundance of Chloroflexi (18.11 and 32.99% lower than CK, respectively). N2 treatment significantly reduced the relative abundance of Nitrospirae (24.94% lower than CK). Meanwhile, N application reduced the potential function of partial nitrogen (N) and sulfur (S) cycling in bacterial community. For example, compared with CK, nitrate respiration and nitrogen respiration decreased by 35.78–69.06%, and dark sulfide oxidation decreased by 76.36–94.29%. N application had little effect on fungal community structure and function. The resilience and vulnerability of airport networks are significant challenges during the COVID-19 global pandemic. Previous studies considered node failure of networks under natural disasters and extreme weather. Herein, we propose a complex network methodology combined with data-driven to assess the resilience of airport networks toward global-scale disturbance using the Chinese airport network (CAN) and the European airport network (EAN) as a case study. The assessment framework includes vulnerability and resilience analyses from the network- and node-level perspectives. Subsequently, we apply the framework to analyze the airport networks in China and Europe. Specifically, real air traffic data for 232 airports in China and 82 airports in Europe are selected to form the CAN and EAN, respectively. The complex network analysis reveals that the CAN and the EAN are scale-free small-world networks, that are resilient to random attacks. However, the connectivity and vulnerability of the CAN are inferior to those of the EAN. In addition, we select the passenger throughput from the top-50 airports in China and Europe to perform a comparative analysis. By comparing the resilience evaluation of individual airports, we discovered that the factors of resilience assessment of an airport network for global disturbance considers the network metrics and the effect of government policy in actual operations. Vasa, one of the best‐studied germ cell markers plays a critical role in germ cell development and differentiation in animals. Vasa deficiency would lead to male‐specific sterility in most vertebrates, but female sterility in the fly. However, the role of the vasa gene involved in germ cell differentiation is largely elusive. Here, we first characterized the expression profile of vasa products in the Asian yellow pond turtle by quantitative reverse‐transcription polymerase chain reaction and fluorescence immunostaining. The results showed that vasa messenger RNA (mRNA) is initially detected in embryos at stage 16, and then dramatically increased in embryos at stage 19. In particular, like the sex‐related genes, vasa mRNA exhibited differential expression in embryos between the male‐producing temperature (MPT, 25°C) and the female‐producing temperature (FPT, 33°C), whereas there was no difference in methylation levels of vasa promoter detected between FPT and MPT. In contrast, in the adult Asian yellow pond, the level of vasa mRNA was much higher in the testis than ovary. Moreover, the immunostaining on testicular sections and cells showed that Vasa protein was exclusively expressed in germ cells: Weak but detectable in spermatogonia, highest in spermatocytes, moderate and concentrated in chromatid bodies in spermatids and spermatozoa, and bare in somatic cells. The expression profile of Vasa protein is similar in turtle species studied so far but distinct from those in fish species in this study. The findings of this study would provide new insights into our understanding of the conservation and divergence of the vasa gene, even other germ cell genes across phyla. Konosirus punctatus and Clupanodon thrissa are distributed in overlapping geographical areas. Their similar appearance makes it difficult to distinguish them by morphology. In recent years, overfishing has led to frequent stock collapses. The development of resource management and identification techniques is necessary for accurate statistical assessment of harvests and to protect the diversity of species and genetic resources. Considering the difficulty of distinguishing these two species, a molecular marker technique was developed to identify them. As a result, primer HB3 was developed to obtain amplification products for C. thrissa, which are not produced for K. punctatus. This study allows rapid distinction between these two species based on amplification products, providing a simple and rapid molecular method for checking products sold and illegally caught. The COVID-19 global pandemic hit the aviation industry hard since the end of 2019. It has had an immediate, dramatic impact on airport traffic and revenue. Airports are the important nodes in the aviation system network, and the failure of a single airport can often affect the surrounding airports. The purpose of our analysis is to show how is the resilience and recovery of airports in the global public health crisis. Much research on resilience can be found in air transportation networks facing natural hazards or extreme weather, which focus on the robustness of the airport network. These methods are not suitable for the global public health event. Therefore, based on the collection of existing data, we combined with existing resilience measurement methods to analyze the resilience and recovery of airports during the global public health crisis. The resilience metrics results reflect the recovery of airports very well under different strategies. Here, we analyze airport network resilience by considering the performance-based methods. We integrate some metrics such as aircraft movements, passenger throughput, and freight throughput in the resilience metrics model, comparing the resilience evaluation under different preventive and control strategies, which can reflect the airport's recovery speed during the COVID-19 pandemic. Our analysis indicates that the aviation system network deteriorates soon after the COVID-19 outbreak, but the recovery level of the aviation industry depends on what measures are taken to prevent and control the COVID-19 epidemic. In particular, the recovery of the aviation system network in Europe takes longer than in China, due to different prevention and control strategies for COVID-19. The study proves that the emergency response ability of the country for the public health crisis has a significant positive impact on speeding up the recovery of the aviation system. Helminth-derived molecules have the ability to modulate the host immune system. Our previous study identified a tetradecapeptide derived from Trichinella spiralis paramyosin (Ts -pmy) that could bind to human complement component C9 to inhibit its polymerization, making the peptide a candidate therapeutic agent for complement-related immune disorders. Here, the peptide underwent an N-terminal modification with a membrane-targeting signal (a unique myristoylated peptide) to improve its therapeutic efficacy. We found that the modified peptide had a binding affinity to human C9 that was similar to that of the original peptide, as confirmed by microscale thermophoresis assays. The binding of the modified peptide to human C9 resulted in the inhibition of C9-related complement activation, as reflected by the decreased Zn2+-induced C9 polymerization and the decreased C9-dependent lysis of rabbit erythrocytes. In addition, the original and modified peptides could both bind to recombinant mouse C9 and inhibit the C9-dependent lysis of rabbit erythrocytes in normal mouse serum (NMS), which meant that the peptides could cross the species barrier to inhibit complement activity in mice. Further in vitro and in vivo analyses confirmed that the peptide modification increased the retention time of the peptide. Furthermore, intraarticular injection of the modified peptide markedly ameliorated knee swelling and joint damage in mice with antigen-induced arthritis (AIA), as assessed histologically. These results suggested that the Ts -pmy-derived peptide modified with a membrane-targeting signal was a reasonable candidate therapeutic agent for membrane attack complex (MAC)-related diseases [such as rheumatoid arthritis (RA)] and the study presented a new modification method to improve the potential therapeutic effects of the peptide. Helminths develop strategies to escape host immune responses that facilitate their survival in the hostile host immune environment. Trichinella spiralis , a tissue-dwelling nematode, has developed a sophisticated strategy to escape complement attack. Our previous study demonstrated that T. spiralis secretes calreticulin (Ts CRT) to inhibit host classical complement activation through binding to C1q; however, the C1q binding site in Ts CRT and the specific mechanism involved with complement-related immune evasion remains unknown. Using molecular docking modeling and fragment expression, we determined that Ts CRT-S, a 153-aa domain of Ts CRT, is responsible for C1q binding. Recombinant Ts CRT-S protein expressed in Escherichia coli had the same capacity to bind and inhibit human C1q-induced complement and neutrophil activation, as full-length Ts CRT. Ts CRT-S inhibited neutrophil reactive oxygen species and elastase release by binding to C1q and reduced neutrophil killing of newborn T. spiralis larvae. Binding of Ts CRT-S to C1q also inhibited formation of neutrophil extracellular traps (NETs), which are involved in autoimmune pathologies and have yet to be therapeutically targeted.
