Simple Summary: The research focuses on the development and evaluation of a new immunocastration vaccine for beef cattle. This study aims to evaluate the safety, immunogenicity, immunocastration effect, and productive impact of a novel IC vaccine developed in Chile for female cattle. Two consecutive trials were conducted, the first under controlled conditions and the second under field conditions. The results demonstrated that the new vaccine is safe and effective for use in beef cattle, inducing specific immunity against GnRH-I, reducing gonadal functionality, and improving productive parameters. Sterilization and castration have been synonyms for thousands of years. Making an animal sterile meant to render them incapable of producing offspring. Castration or the physical removal of the testes was discovered to be the most simple but reliable method for managing reproduction and sexual behavior in the male. Today, there continues to be global utilization of castration in domestic animals. More than six hundred million pigs are castrated every year, and surgical removal of testes in dogs and cats is a routine practice in veterinary medicine. However, modern biological research has extended the meaning of sterilization to include methods that spare testis removal and involve a variety of options, from chemical castration and immunocastration to various methods of vasectomy. This review begins with the history of sterilization, showing a direct link between its practice in man and animals. Then, it traces the evolution of concepts for inducing sterility, where research has overlapped with basic studies of reproductive hormones and the discovery of testicular toxicants, some of which serve as sterilizing agents in rodent pests. Finally, the most recent efforts to use the immune system and gene editing to block hormonal stimulation of testis function are discussed. As we respond to the crisis of animal overpopulation and strive for better animal welfare, these novel methods provide optimism for replacing surgical castration in some species. Simple Summary: Stallion mules are infertile but still able to produce testosterone, which influences undesirable behavior. Surgical castration is required to eliminate testosterone synthesis, which results in pain and the risk of postoperative complications. To the best of our knowledge, there are no existing studies regarding immunocastration as an alternative method for surgical castration in mules. The objective of this study was to evaluate whether a gonadotropin-releasing hormone (GnRH) vaccine could be used as a method of immunocastration in stallion mules via the assessment of anti-GnRH antibody concentration, serum testosterone concentration, clinical adverse effects, and changes in the selected behaviors. Intact and unilateral cryptorchid mules received the GnRH vaccine at weeks 0, 4, and 8. The serum testosterone concentrations in GnRH-vaccinated mules were lower than before vaccination from weeks 6 to 14. Subcutaneous edema adjacent to the injection site was observed in intact mules after the second or third vaccination. The stallion mules responded to the GnRH vaccine, resulting in a temporary decrease in serum testosterone. Based on the results of this study, the GnRH vaccine may be administered as a temporary immunocastration method for stallion mules. Stallion mules have been used as working equids in several countries. Aggressiveness under the influence of testosterone results in the necessity for surgical castration before work training. The gonadotropin-releasing hormone (GnRH) vaccine may be an alternative method for immunocastration in mules. The objective of this study was to evaluate the effect of the GnRH vaccine on anti-GnRH antibody concentration, serum testosterone concentration, clinical adverse effects, and behavioral changes in response to receiving selected physical manipulations from humans. Twenty-five mules were separated into three groups: Control-intact, Control-castrated, and Treatment. The Treatment group was further divided according to condition (intact or unilateral cryptorchid) and age. The Treatment group received 195 µg of the GnRH vaccine intramuscularly at weeks 0, 4, and 8. The anti-GnRH antibody concentrations increased at weeks 6 and 10, and then they gradually decreased to baseline at week 24. The Treatment-intact-young group had the highest concentration of anti-GnRH antibody. The serum testosterone concentrations in the Treatment group were lower than before vaccination from weeks 6 to 14. Subcutaneous edema adjacent to the injection site was detected in the Treatment-intact group after booster vaccination. Simple Summary: The Italian pig sector is renowned for dry-cured ham production, which requires heavy pigs (raised for at least nine months and slaughtered at more than 160 kg). Nevertheless, efficiency and utilisation of every part of the pig, including the loin, should be maximised. To avoid boar taint, surgical castration is necessary, albeit raising welfare concerns. Immunocastration offers a promising alternative. This study investigated the effects of immunocastration compared to surgical castration on the chemical composition and fatty acid profile of loins and adipose tissue from Italian heavy pigs. Carcass parameters were monitored at slaughter, and samples of longissimus dorsi muscle and subcutaneous fat were analysed. This study showed no significant differences in carcass characteristics and proximate composition of fresh meat. However, variations were observed in the fatty acid profiles of meat and adipose tissue between groups. Notably, saturated fatty acids, particularly stearic acid (18:0), were higher in the intramuscular fat (IMF) of the immunocastrated pigs compared to the surgically castrated pigs. Conversely, monounsaturated fatty acids, predominantly oleic acid (18:1n-9), were higher in the IMF from the surgically castrated pigs compared to the immunocastrated pigs. While immunocastration may offer benefits in terms of animal growth and carcass composition, it could lead to unfavourable lipid changes in fresh loin meat for Italian heavy pigs. Simple Summary: This preliminary study investigated the sensory quality perceived by consumers (trained panellists or untrained consumers) of pork loins deriving from animals subjected to different castration methods (surgical vs. immunocastration) and fed different protein sources (soybean vs. pea). The findings revealed that the trained panel could discriminate clearly between dry-cured loin from male pigs surgically castrated and fed a soya diet compared with those fed a pea diet, and the vaccination almost completely reduced most of the sensory traits in both sexes. Additionally, the study found that immunocastration in male pigs did not negatively impact the skatole detection scoring. Moreover, the untrained consumer panel preferred the dry-cured loin from surgically castrated male pigs fed a pea-based diet. To sum up, it is possible to completely replace soybean meal with field peas when feeding heavy immunocastrated male pigs for dry-cured loin production. Two of the main issues related to cured meat products are castration to avoid boar taint and the hefty reliance on soybean meal to feed pigs. However, data on the effects of immunocastration in pigs and alternative crop protein feeds on the sensory traits and consumers' acceptance of dry-cured loin are still limited. A preliminary study was conducted on the effect of sex type (surgical castrated male pigs and immunocastrated male and female pigs, at approximately 140 kg in weight and 7 months of age) and animal diet (pea vs. soya) on dry-cured loins. The study involved a sensory evaluation of six treatments, with a 3 × 2 factorial design, conducted by trained panellists and untrained consumers in Spain (n = 126) and Portugal (n = 80). The consumers were also checked for their skatole sensitivity using a pure substance. The results showed that the pea-based diet was significantly different (p < 0.001) from the soy-based diet, as determined by a trained panel of surgically castrated male pigs, although the immunocastration treatments were not split. Dry-cured loin from immunocastrated male pigs fed with peas was considered tenderer and juicier (p < 0.05) than those fed soya-based diets. The untrained consumer panel scored higher on the sensory traits (flavour, juiciness, and overall liking) from the loin samples of surgically castrated male pigs fed a pea diet. All consumers scored similarly in boar taint detection. This study demonstrates the applicability of the pea-based diet for the feeding of pigs destined for cured meat and highlights immunocastration as a tool that does not compromise the skatole detection score. Background: Animal reproduction performance is crucial in husbandry. Immunocastrated animals serve as an ideal animal model for studying testicular function. During androgen suppression, the testis undergoes dramatic developmental and structural changes, including the inhibition of hormone secretion and spermatogenesis. Methods: To characterize this process, we investigated the effects of castration using a recombinant B2L and KISS1 DNA vaccine, and then identified functional genes in the testes of Yiling goats using RNA-seq and WGS. The experimental animals were divided into three groups: the PVAX-asd group (control), PBK-asd-immunized group, and surgically castrated group. Results: The results demonstrated that the administration of the recombinant PBK-asd vaccine in goats elicited a significant antibody response, and reduced serum follicle-stimulating hormone (FSH) and luteinizing hormone (LH), resulting in smaller scrotal circumferences and decreased sexual desire compared to the control group. In addition, RNA transcriptome sequencing (RNA-seq) analysis of the testes revealed that the biological processes after immunocastration mainly focused on the regulation of cell matrix adhesion, histone acetylation, negative regulation of developmental processes, apoptosis, and activation of the complement system and the thrombin cascade reaction system. Then, we integrated the whole-genome sequencing and testis transcriptome, and identified several candidate genes (FGF9, FST, KIT, TH, TCP1, PLEKHA1, TMEM119, ESR1, TIPARP, LEP) that influence steroidogenesis secretion and spermatogenesis. Conclusions: Multiple pathways and polygenic co-expression participate in the response to castration vaccines, altering hormone secretion and spermatogenesis. Taken together, our atlas of the immunocastration goat testis provides multiple insights into the developmental changes and key factors accompanying androgen suppression, and thus may contribute to understanding the genetic mechanism of testis function. The article investigated the dependence of the histological structure of m. Longissimus thoracis muscle in immunocastrated pigs vaccinated with the “Improvak” vaccine from the company “Zoetis”, USA, and in non-castrated pigs fattened under the conditions of industrial pork production and slaughtered at different live weights. For the study, two experimental groups of 220 gilts each were formed, derived from Irish Landrace and Irish Yorkshire sows and inseminated with semen from boars of the MakGro synthetic line. At the age of 72 days, with an average live weight of 26 kg, they were brought to a fattening farm where the piglets of the research group were vaccinated on the 112th day of life with the vaccine “Improvak” from the company “Zoetis”, USA, in a quantity of 2 ml. They were re-vaccinated on the 148th day of life with the same vaccine and in the same quantity. The pigs in the control group were not castrated. It was found that in m. Longissimus thoracis muscle of immunocastrated pigs using the “Improvak” vaccine, the diameter of the muscle fibres decreased by 15 micrometres and the fibres absorbed more sarcoplasm, which, against the background of a 14 % increase in the content of parenchymal components, indicates an increase in the juiciness and tenderness of the meat. It has been shown that the muscle fibres of the longissimus thoracis no longer grow as a result of the immunocastration of pigs, but take on useful properties, i.e. become hydrated and fat. The main purpose of the experiment carried out was to study the effects of immunological castration of gilts on the histological indicators of the muscle tissue of m. Longissimus thoracis muscle tissue compared to non-castrated animals. For the study, 440 pigs were collected, which were initially divided into two groups of 220 animals each, one of which was immunologically castrated and the other not castrated. At the end of fattening, the group of immunocastrated and non-castrated gilts was divided by weight into groups of 110 and 130 kg, resulting in four groups: noncastrated gilts with a live weight of 110 kg, non-castrated gilts with a live weight of 130 kg, immunocastrated gilts with a live weight of 110 kg and immunocastrated gilts of 130 kg. After slaughter, histological sections of the longest dorsal muscle were taken and light-optical and morphometric examinations, luminescence microscopy with microphotography of m. Longissimus thoracis muscle were carried out. As a result of the experiments, it was found that as a result of immunological castration of gilts with the Improvak vaccine, the muscle fibre diameter decreased by 15 μm and the muscle fibres absorbed a larger amount of sarcoplasm, which, against the background of an increase in the content of the parenchymal component (by 14%), indicates an increase in the juiciness and tenderness of the meat. Androgen from the testis and weak androgens from the adrenal cortex may interact with each other and affect their synthesis and secretion due to their similar functions. The purpose of this study was to investigate the compensatory effect of adrenal in rats after immunocastration and surgical castration, and the interaction between the hypothalamic–pituitary–testis (HPT) axis and the hypothalamic–pituitary–adrenal (HPA) axis. 24 male SD rats aged 8 weeks were randomly divided into three groups and accepted treatments: surgical castration group, immunocastration group and control group. In both surgical castration and immunocastration groups, the secretion of adrenocorticotropic hormone (ACTH) and dehydroepiandrosterone (DHEA) hormones was significantly increased compared with the control group (p < 0.05). In the HPT axis of the immunocastration group, the KISS1 expression was up-regulated, whereas GPR54, LH and LHR expression were down-regulated (p < 0.05). The expression levels of CRH, POMC and MC2R genes were also significantly up-regulated (p < 0.05). In addition, in the immunocastration group, the expression of adrenal LHR mRNA expression was decreased (p < 0.05). The expression of HPT axis genes and adrenal LHR were up-regulated in the surgical castration group (p < 0.05). These results show that in both immunocastration and surgical castration, adrenal androgen is increased, suggesting that the adrenal gland plays a compensatory role. Simple Summary: Alternatives to surgical castration of piglets present a challenge for control of boar taint. In this study, the proportion of animals with androstenone levels above the risk threshold for boar taint (>0.5 µg/g fat) was found to be less than 5% in immunocastrated animals (ICs), and testicular weight was a reliable indicator of androstenone-positive carcasses. In contrast, in the entire males (EMs), the proportion of animals with androstenone levels above the risk threshold was much higher (approximately 80%), and the difference in testicular size between EMs with androstenone levels below and above the risk threshold was less pronounced. However, the pelvic urogenital tract weight was reliable in these animals. We thus propose that pelvic urogenital tract and testes weights may be considered a simple, reliable, and time- as well as cost-efficient morphometric indicator for the identification of androstenone-positive carcasses for both ICs and EMs at the slaughter line. Immunocastration and rearing of entire males (EMs) are sustainable alternatives to surgical castration. However, these animal carcasses have variable risk of boar taint and should be identified at the slaughter line. We aimed to identify a simple and reliable indicator of androstenone-related boar taint by evaluating pelvic urogenital tract weight as a marker of boar-taint animals at the slaughter line. The pelvic urogenital tract, testes, and accessory sex glands of EMs and immunocastrates (ICs) were collected, dissected, and weighed, before colorimetric measurements of testicular tissue. Additionally, GnRH antibody titers and testosterone, androstenone, and skatole levels were determined. Our results showed that 81.8% of EMs had androstenone levels above the risk threshold (>0.5 µg/g fat; EM/Ahigh subgroup), whereas in ICs, the C/Ahigh subgroup with androstenone >0.5 µg/g fat accounted for only 4.3%. Androstenone levels correlated negatively with GnRH antibody titers and positively with testosterone levels and reproductive organ weights. Identification of ICs with androstenone levels above the threshold (IC/Ahigh subgroup) may be achieved via testes or pelvic urogenital tract weight measurements. However, in EMs, the latter is a more reliable parameter. A principal component analysis based on these variables and hierarchical clustering also distinguished the Ahigh from the Alow subgroup, irrespective of IC/EM. The findings highlight the possible use of pelvic urogenital tract weight along with testes weight as a simple, reliable, and efficient morphometric indicator for identifying androstenone-positive carcasses of different sex categories. The present study aimed to reveal the effects of immunocastration on the development of the immune system in rats. Seventy rats were randomly assigned into two groups: Control (n = 35) and immunized (n = 35). Twenty-day-old rats were immunized with gonadotropin-releasing hormone (GnRH) and booster immunization was administered every two weeks (three immunizations in total). From 20-day-old rats, we collected samples every two weeks, including five immunized rats and five control rats (seven collections in total). We collected blood samples, testicles, thymuses, and spleens. The results showed that GnRH immunization increased the GnRH antibody titers and reduced the testosterone concentration (both P < 0.05). Compared with the control group, the number of CD4+CD8- cells, CD4-CD8+ cells, and CD4+CD8+ cells increased (P < 0.05) whereas the number of CD4-CD8- cells and CD4+CD25+ cells reduced in the immunized group (P < 0.05) over time. GnRH immunization also increased the relative weights of thymus and spleen (P < 0.05), serum concentrations of interleukin (IL)-2, IL-4, IL-6, IL-10, IL-17 and Interferon-g (IFN-g) over time (P < 0.05), and changed the mRNA levels of IL-2, IL-4, IL-6. IL-10, IL-17, IFN-g, CD4, D8, CD19 GnRH, and GnRH receptor (GnRH-R) in thymus and spleen. Thus, GnRH immunization enhanced the immune markers in thymus, spleen, and blood immune cytokines in rats. The main objective of this study was to describe tissue thicknesses of cadaver heads from physically castrated market barrows (PC MARKET BARROWS) and immunocastrated boars (IC BOARS) at the frontal penetrating captive bolt (PCB) placement. Other objectives were to describe differences in bolt force and energy requirements to penetrate and describe potential for bolt-thalamus contact. Forty-four heads were obtained from PC MARKET BARROWS (n  = 22) and IC BOARS (n  = 22) of similar age and size that were rendered insensible with CO2. Mean HCW was 117.32 ± 3.52 kg. Snout to poll distance (cm) and maximum deflection distance (cm) were collected in duplicate. Heads were split at midline with a bandsaw and soft tissue and cranial thicknesses were measured with a digital caliper. Images of each cut surface were collected to evaluate the potential for thalamic damage. Tissue samples were retained from each half of each head and a universal tester was used to determine maximum force and energy of bolt penetration. There was no evidence to support a significant difference (P  > 0.05) in tissue thicknesses between PC MARKET BARROWS and IC BOARS. Maximum deflection distance (maximum distance from a straight edge that was placed from the tip of the snout to the poll of the head) was not different (P  = 0.10) between PC MARKET BARROWS (3.31 ± 0.10 cm) and IC BOARS (3.08 ± 0.10 cm). There was no evidence to support a difference (P =  0.77) in maximum force between PC MARKET BARROWS (7130.32 ± 483.23 N) and IC BOARS (6974.60 ± 463.70 N). There was also no evidence to support a difference (P   =  0.62) in maximum energy between PC MARKET BARROWS (33.37 ± 2.77 J) and IC BOARS (32.04 ± 2.50 J). For PC MARKET BARROWS, there was a difference (P  = 0.05) between the number of heads where the thalamus was located within the theoretical plane of bolt travel for market placement (21/21) versus mature placement (16/21). For IC BOARS, the number of heads where the thalamus was located within the plane of theoretical bolt path was not different between the two PCB placements (19/21 each). Overall, the data suggest that tissue profiles of PC MARKET BARROWS and IC BOARS do not differ at the frontal PCB placement site and the mechanical tools that are effective for PC MARKET BARROWS should also be effective for IC BOARS. Castration is frequently used to reduce aggressive behavior and improve the meat quality of animals. Traditionally, surgical and mechanical castration are used to sterilize the animals, but these approaches are associated with a high level of pain, stress, long recovery periods, and post-operative infections. Immunocastration is a new animal-friendly, painless alternative castration technique that is used to prevent undesired sexual behavior, reduce aggressive behavior, prevent unwanted pregnancy, control wildlife populations and wandering species, enhance growth performance, improve meat quality, and treat various sex hormone–dependent disorders. The mechanism of immunocastration includes the immunological block of the hypothalamic-pituitary–gonadal axis (HPG axis) which inhibits gonadotropin secretions, causes atrophy of gonadal tissues, and inhibits gametogenesis, resulting in infertility in both female and male mammals. By the mid-1990s, various immunocastration vaccines have been tested in different animal models to achieve successful castration effects. Recently, genetic immunocastration especially DNA vaccine has gained increasing attention due to its safety, being animal-friendly, and being easy to use. The breeding of entire mail pigs in modern pig farming is based on the quality of their meat, which means improving the carcass composition with more lean meat and more fat of the desired quality, while emphasizing various methods to prevent the appearance of boar taint. This review summarizes the state of research on the type and frequency of problems in the breeding and welfare of entire mail pigs. Observations of the behavior of uncastrated male pigs have shown increased activity, more frequent struggles to establish a hierarchical order, more frequent injuries and an increased degree of lameness compared to castrates and female animals. The influence of the group’s composition on sexual behavior, puberty and premature slaughter was analyzed in detail. It has been found that increased sexually motivated behavior towards pigs from the same box causes higher levels of stress. Observations of slaughtered animals with penile injuries ranging from 3% to 82% have been reported. Studies have been performed on the extent of skin lesions in different groups of animals. The possibility of vaccination against boar taint (immunocastration) was considered. It is concluded that the breeding of entire male pigs seems to be a favorable, future-oriented alternative to conventional surgical castration of pigs, provided that young intact boars are kept in a healthy and socially stable environment with sucient physical resources, as safeguarded by measures of enhanced animal care and management control. Recommendations for the practice are given, based on foreign and our research. They include: early socialisation in stable groups where entire males are separated from females; provision of more space in structured pens and provision of enrichment material for observation, keeping of the necessary hygiene and optimal microclimate, minimization of stress, etc. [ABSTRACT FROM AUTHOR]

Simple Summary: Common eland are currently underutilized regarding their meat production potential, despite their nutritious meat. Due to their temperament and adaptability, they are well-represented in commercial farms internationally, for both consumptive and ecotourism purposes. Immunocastration has been utilized as a management strategy in mixed-sex eland herds but its effects on the slaughter performance and cutting yields of common eland males have not yet been described. Furthermore, the ideal age of slaughter for eland males culled out of breeding herds relative to edible carcass product yields has not yet been explored, as it is almost impossible to control the confounding factors under current extensive production conditions within southern Africa. The Common Eland Research Facilities at The Czech University of Life Sciences Prague allows for studies under controlled conditions, with animals which have extensive records. The results indicated that while immunocastration had no influence, age affected the offal yields, primal cut, and dissection yields, as well as the high-value: low-value meat of the carcass, with the latter being in favor of juveniles. However, sub-adult males generated higher individual muscle yields. This study investigated the effects of immunocastration and slaughter age on the carcass yield performance of intensively farmed common eland males. Eighteen male eland (five immunocastrated juveniles, three intact juveniles, five immunocastrated sub-adults, and five intact sub-adults) were finished for four months, after which their carcass, offal, primal cut, and individual muscle yields were determined. Juveniles were ~6 months of age at the start of the experiment, while sub-adults were ~1.5 years old. Primal cuts were dissected to determine the percentage yields of meat, bones (with tendons), separable fat, and trimmings. Immunocastration had no effect on offal or carcass yields. While sub-adults had increased individual muscle yields, juveniles had greater proportionate yields of primal cuts and a greater total high-value: low-value meat ratio. Simple Summary: Male cattle (bulls) are often castrated as part of routine herd management. The benefits of this practice include the reduction of aggression and the elimination of unwanted pregnancies. However, castration represents an animal welfare concern as bulls are subjected to pain during and after the procedure. Surgical castration, in particular, places animals at increased risk of hemorrhage and infection. Immunocastration, a method involving vaccination against the hormones that regulate reproduction, offers a reduced-pain alternative to traditional castration, but the current products require multiple doses to effectively reduce fertility for extended periods. In an effort to improve upon current multi-dose immunocastration strategies, we evaluated the efficacy of a single-dose implantable immunocastration vaccine. This implant was designed to reduce fertility without the need for multiple doses, thus improving welfare for the animals as well as safety for producers and clinicians. The results presented here are promising and suggest that further refinement of the immunocastration implant could provide a convenient alternative to current immunocastration strategies. Immunocastration relies on the vaccine-mediated stimulation of an immune response to gonadotropin-releasing hormone (GnRH) in order to interrupt spermatogenesis. This approach offers a less painful alternative to traditional castration approaches but the current, commercially available options require multiple doses of vaccine to maintain sterility. Thus, a series of pilot studies were conducted to determine the feasibility of a single-dose immunocastration vaccine implant. These five studies utilized a total of 44 Holstein bulls to determine the optimal vaccine composition and validate the ability of a stainless-steel subcutaneous implant to deliver a vaccine. Outcome measures included the duration of implant retention, scrotal dimensions and temperature, implant site temperature, anti-GnRH antibodies, and serum testosterone concentration. Over the course of several studies, anti-GnRH antibodies were successfully stimulated by vaccine implants. No significant treatment effects on scrotal dimensions or testosterone were detected over time, but changes in spermatogenesis were detected across treatment groups. Results indicate that a single-dose implantable immunocastration vaccine elicits a humoral immune response and could impact spermatogenesis in bulls. These findings provide opportunities for the refinement of this technology to improve implant retention over longer periods of time. Taken together, this approach will offer producers and veterinarians an alternative to physical castration methods, to improve animal welfare during routine livestock management procedures. Simple Summary: Castration of piglets is routinely practiced to prevent the presence of boar taint. In Europe, around 61% of pigs are surgically castrated, 54% of which do not receive any pain-relief therapy. The increasing public awareness towards animal welfare and the ethical characteristics of animal-derived products has led to the necessity of finding more animal-friendly alternatives. Among them, immunocastration, a process of active immunisation leading to the suppression of testicular function, has shown promising results in light pig production, both in terms of animal welfare and productivity. Besides, no study has yet investigated animal welfare in immunocastrated males intended for Italian heavy pig production. This study aimed at evaluating the effect of immunocastration on the welfare of heavy pigs by monitoring behaviour and body lesions in immunocastrated and surgically castrated pigs. Our results confirmed that immunocastration is a suitable alternative to surgical castration with profitable productive performance, whereas the higher activity of animals and the higher presence of body lesions before the effective immunisation caused an impairment of animal welfare, which should be further investigated as a critical aspect in heavy pig production. Immunocastration has been pointed out as an alternative to surgical castration; though, most of the scientific studies were performed in light pig production. This study aimed to evaluate the effect of immunocastration on animal welfare in heavy pig production through the evaluation of behaviour and body lesions. A total of 188 commercial-hybrid pigs were randomly allocated into two treatment groups: surgical castration (SC) and immunocastration with Improvac® (IC). Data on behaviour, body lesions, and salivary testosterone levels were collected the day before each vaccination at 15, 22, 32, and 36 weeks of age. IC and SC pigs were slaughtered at 40 and 41 weeks of age, respectively; productive and carcass traits data were also collected. Considering productive performance, our results confirmed that IC pigs grew faster and presented a higher weight at slaughter. A critical period for pig welfare was observed before 32 weeks: testosterone concentration and body lesion score were significantly higher in IC pigs compared to SC pigs; active behaviours were significantly more frequent in IC at 15 weeks. Immunocastration may represent a suitable alternative to surgical castration with profitable productive performances, whereas the impairment of welfare during the period before the effective vaccination should be further investigated as a potential critical aspect in heavy pig production. In order to determine the influence of the castration method on the indicators of the meat cuts of pig carcasses, two groups of 74 boars were selected. Surgical castration was carried out in one of them, and immunological vaccination method was used in the other group. Both surgically castrated and immunocastrated boars were reared and fattened under the same conditions. And at the end of fattening, they were slaughtered and their carcasses evaluated for weight and proportion of meat cuts separately in the cervical-scapular, back-lumbar and pelvicfemoral thirds. The result of the assessment was the establishment of a significantly higher value of the weight indicators of such meat cuts as single-grade pork from the neck by 0.3 kg or 20.0% (P <0.01) and the weight of lard with skin by 0.6 kg or 13.64 % (P <0.05) in the shoulder-scapular third of the carcass and single-grade pork per 0.2 kg or 15.38% (P <0.05) in the pelvic-femoral third of the carcass in immunocastrated pigs. Immunocastration vaccines achieve their effects through neutralization of the endogenous hormone by the humoral antibody produced against the immunized genes, but there is little information regarding cell‐mediated immune response on the gonadal function of the immunized model is available. In this study, we used ram as a model animal to identify the cellular immune response in testicular tissues of rams immunized with intranasal KISS1 gene vaccine. The immune castration model was evaluated by sexual behaviours, spermatogenesis and serum hormone profiles after the KISS1 gene immunization. Transcriptome analysis of testicular tissues was carried out to identify the expressions of protein‐coding genes involved in cellular immunity. The results showed that we successfully constructed the KISS1 immune castration ram model, in which testicular growth and development, testosterone and kisspeptin‐54 levels, and sexual function were suppressed in immunized rams (p <.05). Using Hiseq™ 2000 high sequencing for ram testicular, we identified 21 differentially expressed genes (DEGs) related to cellular immunity, of which, 14 genes were upregulated and seven genes were downregulated in the testis of the immunized group (p <.05). The Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment showed that these differentially expressed genes were enriched in the antigen presentation process mediated by MHC class I and the cytotoxic pathway mediated by natural killer cells. Background: Salivary gland (SMG) degeneration and dysfunction are common symptoms that occur after sex hormone deprivation, but the underlying mechanisms remain largely unknown. Additionally, immunocastration, which causes drop of sex hormones, has been developed as an alternative to surgical castration, however whether it exerts similar effects as surgical castration on the salivary glands is unknown. Through histological and RNA-seq analysis, we assessed changes in morphology and transcriptome of SMG in response to immunocastration (IM) versus surgical castration (bilateral orchiectomy, ORC). Results: Compared to entire males (EM), ORC caused severe degeneration of SMG in rats, as evidenced by both decreased (P < 0.01) SMG weight and organ index, and by decreased (P < 0.01) quantity of SMG acini and ducts. IM had minimal effects (P > 0.05) on SMG weight and organ index, but it still caused degeneration (P < 0.05) of the acini and ducts. Even though, the quantity of both SMG acini and ducts was much higher (P < 0.001) in IM than in ORC. Functional enrichment analysis of the common regulated genes by ORC/IM revealed disrupted epithelial cell development, angiogenesis, anatomical structure morphogenesis and enhanced cell death are associated with SMG degeneration in deprivation of androgens. Integrated data analysis shown that there existed a selective hyperfunction of SMG ribosome and mitochondrion in ORC but not in IM, which might be associated with more severe degeneration of SMG in ORC than in IM. Conclusions: Our findings suggested that both surgical castration and immunocastration caused SMG degeneration by disrupting epithelial cell development, angiogenesis, anatomical structure morphogenesis and enhancing cell death. Neutering male piglets by surgical procedures without anaesthesia, with analgesia and/or anaesthesia and, recently, immunological-chemical castration are practices to avoid unwanted or aggressive sexual behaviour, and to prevent the development of meat boar taint. This exploratory study aims to investigate Portuguese consumer's awareness, beliefs and attitudes in issues like boar taint, piglet's castration and pork meat quality, observing possible demographic trends. It is also intended to identify clusters of consumers with similar attitudes, crossing them with demographic data to verify the existence of patterns in Portugal related to these issues. To attain this objective, a consumer's survey was performed through an online questionnaire open for 30 days. A total of 158 respondents completed the survey. Almost a half (46%) of respondents stated their unknowledge about boar taint. Surgical castration and its effects are topics with which older consumers with a rural background are more familiar with, while immunological-chemical castration is still unknown to most consumers: 65% of consumers said they were not aware of this method, and 75% did not know whether it is an effective method for eliminating boar taint. Hierarchical clustering followed by K-means analysis segmented consumers into three clusters characterized according to their opinions, mainly divided by ethical and chemical-free orientations and by a more conservative meat quality and flavour-oriented attitudes, generally independent of prevailing demographics. In general, there were no defined opinions about the subjects under study, due mainly to the lack of information or knowledge. Nevertheless, cluster classification revealed differences in consumer's opinions, especially regarding the reasons for castration and the pain inflicted, about meat quality and the willingness to buy pork from entire males or to pay more for this type of product. Objective: To determine the effect of surgical castration and immunocastration on productive parameters, carcass quality, as well as the physicochemical characteristics of the meat of fattening pigs. Design/Metodology/Aproach: Ten male pigs of the York/Pietrain/Landrace breed with a body weight of 25±5 kg were used per treatment. They were housed in individual pens and fed diets according to their physiological state. The variables evaluated were analyzed with a student's t-test for independent samples. Results: Results do not show differences (p>0.05) in the productive variables, quality of the carcass or physicochemical characteristics. Study Limitations/Implications: The study did not consider qualitative variables such as flavor and smell of the treatments. Findings/Conclusions: Immunocastration is an alternative to surgical castration since the quality of the carcass, the productive variables and the physicochemical characteristics of the meat are not affected, and it favors animal welfare. Context: Pig producers require strategies to temporarily slow growth rate, particularly when markets impose penalties on over-weight and fat carcasses. Immunocastrated pigs usually have higher feed intakes (e.g. during the 3–5-week immunocastration-to-slaughter interval, which is commonly used in Australian pig industry), resulting in higher carcass weight and backfat thickness. Shortening the immunocastration-to-slaughter interval by delaying the second vaccination by a week may be a fine-tuned strategy to reduce carcass backfat and weight of male pigs. However, postponing the age receiving immunocastration as a method to achieve a shorter immunocastration-to-slaughter interval may increase aggressive and sexual behaviours of pigs. Aims: The experiment aimed to quantify the effects of shortening the interval between the second dose of immunocastration and slaughter from 3 weeks to 2 weeks on growth performance, carcass traits, aggressive and sexual behaviours, and carcass skin blemishes in group-housed male finisher pigs slaughtered at a fixed age. Methods: Pigs (n = 58 pens; 45 pigs per pen) were selected into the experiment at 16 weeks of age. Half of the pens received the second anti-GnRH vaccine (V2) as a means of immunocastration at 19 weeks (a common immunocastration age used in Australian pig industry; control group), and the remaining half of the pens received V2 at 20 weeks of age. All the pigs were slaughtered at 22 weeks of age. Growth performance was recorded between 16 and 22 weeks of age. Behavioural observation was conducted at 20 and 21 weeks of age. Carcass weight, backfat thickness and severity of skin blemishes on carcasses were recorded in the abattoir. Key results: Results showed that shortening the immunocastration-to-slaughter interval from 3 to 2 weeks reduced average daily feed intake by 9%, growth rate by 8%, carcass weight by 3%, and backfat thickness by 7% (all P < 0.05), but the percentage of pigs that displayed sexual behaviour and carcasses with >25 lesions both increased (both P < 0.05). Conclusions: Shortening the immunocastration-to-slaughter interval from 3 to 2 weeks by delaying V2 is an effective strategy to reduce feed intake, growth rate, carcass weight and backfat in group-housed male pigs, but at a risk of increasing carcass skin blemishes. Implications: The increased sexual behaviour and carcass skin blemishes will need to be managed when using a shortened immunocastration-to-slaughter interval to temporarily reduce carcass weight and backfat. Australian pig producers require flexible strategies to temporarily slow growth rate and avoid the price penalty associated with over-weight and fat carcasses. Our study showed that shortening the immunocastration-to-slaughter interval from 3 to 2 weeks following the second vaccination can reduce growth rate, carcass weight and backfat thickness in group-housed male pigs slaughtered at 22 weeks of age. In practice, two injections of gonadotropin‐releasing hormone (GnRH) vaccine are recommended for pig immunocastration for effective outcomes. The present study aimed to investigate the expressions of cytochrome P450 aromatase (P450arom) and anti‐Müllerian hormone (AMH) in testes, testicular length and testicular histomorphometry of the fattening pigs receiving the first injection of GnRH vaccine 6 weeks earlier than the standard protocol. Based on vaccination protocol, 24 pigs were equally divided into three groups: T1 was vaccinated at 15 and 19 weeks of age, T2 received vaccine at 9 and 19 weeks of age and C remained intact. P450arom and AMH expressions were analysed using immunohistochemistry and Western blot. The results revealed that testicular length was highest in C pigs, but not different between T1 and T2 groups (6.5 ± 0.2 versus 6.9 ± 0.3 cm, p =.538). Histomorphometry demonstrated that the height of spermatogenic epithelia, the diameter of seminiferous tubules and the number of seminiferous tubules between T1 and T2 groups were not different (p >.05). For P450arom, immunohistochemistry revealed that H‐score of C group was significantly higher than that of both T1 and T2 groups. Western blot analysis showed that C group possessed the densest protein band. Moreover, H‐score between T1 and T2 groups was not significantly different. Protein band intensity between both groups was not apparently different. As for AMH, C pigs had significantly lower H‐score than both T1 and T2 pigs. Furthermore, T2 pigs possessed significantly higher H‐score than T1 pigs. Western blot analysis showed that the most intense protein band was found in T2 group. In summary, GnRH vaccine affected testicular development and functions. The first injection could be performed either at 9 or 15 weeks of age since both protocols contributed to comparable results in aspect of testicular length, histomorphometry and expressions of P450arom and AMH. The aim of this study was to compare testicle morpho-functional characteristics in bulls undergoing a single or two immunizations against GnRH. Nelore (Bos taurus indicus) bulls were randomly allocated into three experimental groups: G1 (n=12), a single 400 μg dose of anti-GnRH vaccine on day 0; G2 (n=11), a first 400 μg dose of anti-GnRH vaccine on day 0 followed by a second (boost) dose 30 days later; and control group (CG, n=12), 1 mL saline 0.9% at day 0. Every 30 days, from day 0 until slaughter at day 90, the bulls were weighed and underwent testicular biometry, semen collection and analysis, and blood sample collection for testosterone measurement. Immediately after slaughter, the testicles were removed and transport at 15°C to the laboratory for histopathological analysis. There was a decrease in testicular height (P=0.0476), width (P=0.0021), and in scrotal circumference (P=0.0001), after either a single (G1) or two (G2) immunizations against GnRH. Both G1 and G2 had lower testosterone concentrations than CG from day 60 on (P<0.01), but in G2, it was also lower than in G1 at day 90 (P=0.0006). All sperm parameters were affected by active immunization against GnRH (P<0.05), and in G2, averages were lesser (P<0.05) than in G1 from day 60 on. No signs of seminiferous tubule degeneration were found in any sample from the CG, contrasting with 75.0% and 100.0% of the samples from G1 and G2, respectively. Economic margins on pig farms are small, and changing slaughter weights may increase farm profitability. However, one can question if the optimal slaughter weight is the same for each sex. On three farms, crossbred pigs (n = 1128) were used to determine the effect of sex and slaughter weight on performance, carcass quality and gross margin per pig place per year. On each farm, an equal number of entire males (EMs), barrows (BAs), immunocastrates (IC) and gilts (GIs) were housed separately in group pens. Pens were randomly divided into three categories of different slaughter weights: 105, 117 and 130 kg BW. In BA, the high average daily feed intake (ADFI) and the lower capacity to gain muscle led to a higher feed conversion ratio (FCR) and lower lean meat percentage in comparison to EM and IC. In all sexes, ADFI and FCR increased with an increasing slaughter weight but the effect of slaughter weight on carcass quality varied between sexes. In BA and GI, slaughter weight had no effect on carcass quality, but in EM and IC, carcass quality improved at higher slaughter weights. Gross margin per pig place per year was calculated as gross margin per pig × barn turnover per year, taking into account fixed costs per round, feed costs and output price per pig. The slaughter weight that gained the highest gross margin per year differed between sexes. Slaughtering BA and GI at 130 kg BW, compared to 105 or 117 kg BW, decreased the gross margin per pig place per year due to the lower margin per pig and barn turnover at higher weights. In IC and EM, no difference in gross margin per pig place per year could be demonstrated between slaughtering at 105, 117 or 130 kg BW. In IC, the increasing gross margin per pig with increasing slaughter weights counteracted with the lower barn turnover. In EM, gross margin per pig did not differ between slaughter weights, but the effect of barn turnover was too small to demonstrate significant differences between slaughter weights on gross margin per pig place per year. In conclusion, slaughter weight has an impact on profitability in BA and GI: they should not be slaughtered at 130 kg BW but at lower weights, but no effect could be demonstrated in EM and IC. To avoid boar taint before slaughtering, late castration by immunisation against gonadotrophin releasing hormone or immunocastration has been developed. The current study aimed at determining whether differences in feed efficiency between castrated male (CM), immune castrated (IM) and entire male pigs (EM) can be explained by differences in nutrient plasma profiles after a meal. In this study, 24 male pigs (n = 8/type) were enrolled between 14 to 19 weeks of age. Entire and IM pigs ate less and were more feed efficient than CM pigs (p < 0.05). The postprandial plasma profiles of glucose, insulin, urea and amino acids (AA) were determined before (d –6), just after (d 8) and well after (d 16) the decrease in testicular hormones in IM pigs. For each test day, pigs were fasted overnight and subsequently fed a small meal (400 g). On d –6, postprandial profiles of plasma glucose of IM pigs did not differ from the two other types of pigs. On d 8, EM pigs had a greater average plasma glucose concentration than IM and CM pigs (p < 0.05) but the profiles did not differ. On d 16, the differences between profiles of glucose suggest a lower clearance of glucose in EM compared with IM and CM pigs. Plasma insulin did not significantly differ between the three types. Plasma urea profiles did not differ between CM, EM and IC pigs on d –6 whereas CM pigs showed higher plasma urea concentrations than EM and IM thereafter (p < 0.01). Among AA, Lys plasma concentrations were greater in CM than in EM and IM pigs on d –6, d 8 and d 16 (p < 0.05), whereas on d 16 plasma Hypro concentrations were lower in CM than in EM and IM pigs (p < 0.05). The finding that plasma glucose profiles were modified by immunocastration much faster and earlier than urea and AA profiles, suggest that the decrease in testicular hormones impacted energy metabolism more rapidly than protein metabolism. Heavy weight gilts commonly show signs of oestrus during the late finishing phase, which results in a period of reduced feed intake and growth rate. Immunization against gonadotropin-releasing hormone (GnRH) (IM, immunocastration) was developed for finishing boars and recently extrapolated to females. Immunocastration acts by suppressing reproductive activity and improving the growth potential. The objective of this study was to evaluate the effects of IM on growth performance, reproductive activity and carcass characteristics of late finishing gilts. Seventy-two gilts (63.49 ± 0.39 kg) were either injected with saline (Intact) or immunized against GnRH (Immunized). The study consisted of three experimental periods: between the first to second immunization (V1 to V2, 15 to 19 weeks of age), from the second immunization to the beginning of daily boar exposure (DBE) (V2 to DBE, 19 to 21 weeks of age) and from the beginning of DBE to slaughter (S) (DBE to S, 21 to 25 weeks of age). Immunized gilts showed an overall increase (from 15 to 25 weeks) of 3.90 kg (P < 0.05) of live weight, 56 g (P < 0.05) of average daily gain (ADG) and 250 g (P < 0.001) of average daily feed intake (ADFI). Immunized gilts had a greater ADFI (+240 g, P < 0.05) and worse feed conversion ratio (+0.26, P < 0.05) from 19 (V2) to 21 weeks of age (before DBE). Furthermore, those females had higher feed intake (+410 g; P < 0.001) plus greater daily weight gain (+92 g; P < 0.05) from V2 to S, and from DBE to S (+470 g of ADFI, P < 0.001; +129 g of ADG, P < 0.01, respectively). Immunocastration had no effect on backfat thickness, lean meat percentage and weight, cold carcass yield or loin depth (P > 0.05). Immunized gilts showed 4.4% increased cold carcass weight (P < 0.01) and 10.6% greater gross flank weight (P < 0.001). Immunization against GnRH did not influence shoulder, collar, loin, belly or ham weights. Nor did it influence belly fat thickness, or meat, skin plus fat and bones yields of cold ham (P > 0.05). Immunocastration reduced ovarian and uterine weights by 82% (P < 0.001) and 93% (P < 0.001), respectively, and suppressed oestrus manifestation in all gilts in the immunized group (P < 0.001). These results indicate that immunization against GnRH is a promising tool for stimulating growth performance with no detrimental effects on carcass quality of heavy weight finishing gilts, by means of oestrus suppression. This study aimed to obtain a comprehensive look at the influence of castration on mRNA expression of the genes CYP2E1, CYP1A2, CYP2A19, HSD3B, SULT2A1 and SULT1A1 and their correlation with boar taint compounds (androstenone, skatole and indole) and Improvac-specific antibodies in a Czech commercial hybrid (Large White × Landrace (sow) × Duroc (boar)). Pigs were divided into groups of entire male pigs (NC), pigs castrated surgically (SC), pigs immunologically castrated and slaughtered 8 weeks (IM8) or 15 weeks (IM15) after the second dose of Improvac, and gilts (GI). Hepatic mRNA expression, measured by quantitative real-time polymerase chain reaction, differed significantly between the control group (entire male pigs) and all groups of interest for CYP2E1, CYP1A2 and CYP2A19. The mRNA level of the HSD3B gene differed significantly between the control group and the IM8, IM15 and GI groups. SULT1A1 gene expression was significantly different between the control group and the SC, IM8 and GI. In the case of SULT2A1, a significant difference was observed only between the control group and IM8 pigs. For all genes and treatment groups described above, expression was increased relative to the control. Significant differences for Improvac-specific antibodies between IM8 and IM15 groups were observed, indicating decrease of antibodies over time. Abstract: This study aimed to investigate the immunoexpression of Ki‐67 protein, androgen receptor (AR), and estrogen receptor beta (ERβ) in testicular tissues of male pigs immunocastrated using GnRH vaccine (Improvac™, Zoetis Co., Ltd., Thailand) with different times. Totally, 30 male pigs were classified by castration protocol into three groups: T1 (n = 10) consisted of pigs immunocastrated at 14 and 18 weeks of age, T2 (n = 10) included pigs immunocastrated at 9 and 19 weeks of age, and C (n = 10) contained intact pigs. The results revealed that testicular length of pigs in C was longer than that of both T1 (8.1 ± 0.76 vs 6.5 ± 0.5 cm, p < 0.001) and T2 (8.1 ± 0.76 vs 6.9 ± 1.0, p = 0.007). Spearman correlation coefficients showed negative correlation between testicular length and H‐score of AR (r = −0.38, p = 0.037), as well as positive correlation between testicular length and Ki‐67 index (r = 0.602, p < 0.001). Generally, mean Ki‐67 index and mean H‐scores of AR and ERβ of pigs in T1 were not different from those in T2 (p > 0.05). However, mean Ki‐67 index and mean AR H‐scores of T1 and T2 were significantly different from C group (p < 0.05). In summary, the immunocastration significantly affected testicular length, including expressions of Ki‐67, AR, and ERβ in pig testes. During oestrus, fattening female pigs are more prone to lameness, fractures and wounds due to mounting and agonistic behaviours of penmates. This study assessed the effect of sexual maturity on the behaviour and welfare of heavy female pigs slaughtered at 36 weeks of age (180±10 kg) for dry-cured ham production. An immunocastrated control group was used for comparison. In all, 56 15-week-old female pigs, individually identifiable by back tattoos were equally distributed among four pens. All animals from two pens were subject to a three-dose immunocastration schedule at 16, 20 and 32 weeks of age. Skin lesions and behaviours were assessed at 18, 23, 28, 33 and 36 weeks of age. A blood sample was collected at 20, 24, 28 and 32 weeks of age for assessing health/stress parameters and GnRH antibodies. At slaughter, ovaries were weighed, measured and histologically examined; stomachs, carcasses and lungs were scored for lesions and a further blood sample was taken. Immunocastrated pigs did not significantly differ from controls in growth rate, feed efficiency and slaughter performances (lung score, gastric score, backfat thickness). However, they showed a lower frequency of aggressive interactions at 33 and 36 weeks, lower front lesions at 28 weeks, but higher at 30 weeks; a lower haptoglobin level at 28 weeks, a lower level of cortisol and back lesions at slaughter (36 weeks). Objective: The aim of this study was to investigate the reversibility and safety of KISS1 metastasis suppressor (KISS1) gene vaccine in immunocastration. Methods: Six eight-week old ram lambs were randomly divided into vaccinated and control groups. The vaccine (1 mg/ram lamb) was injected at weeks 0, 3, and 6 of the study. Blood samples were collected from the jugular vein before primary immunization and at weeks 2, 4, 6, 10, 14, 22, and 30 after primary immunization. All ram lambs were slaughtered at 38 weeks of age, and samples were collected. Results: The specific anti-KISS1 antibody titers in vaccinated animals were significantly higher and the serum testosterone level was significantly lower than those in the control groups from week 4 to 14 after primary immunization (p<0.05). No significant difference was observed at weeks 22 and 30 after the primary immunization. Similar results were also found for scrotal circumference, testicular weight, length, breadth, and spermatogenesis in seminiferous tubules in week 30 after primary immunization. KS (KISS1-hepatitis B surface antigen S) fusion fragment of KISS1 gene vaccine was not detected in host cell genomic DNA of 9 tissues of the vaccinated ram lambs by polymerase chain reaction. Problem Immunocastration or vaccination against the GnRH-I hormone is a promising alternative to reproductive control in different animal species. Given the low immunogenicity of this hormone, the use of adjuvants becomes necessary. Method of Study This study evaluated the effects of three adjuvants that induce different immune response profiles over gonadal function, fertility, and expression of GnRH-I. Female mice (n = 6) were vaccinated at days 1 and 30 with a recombinant antigen for immunocastration and different adjuvants that induced preferentially Th1/Th2, Th2, and Th1 immune profiles. Results Th1/Th2 response is the most efficient to block reproductive activity in vaccinated animals, reducing the number of luteal bodies and pre-ovulatory follicles. Th2 and Th1/Th2 responses induced an increase in GnRH-I at the hypothalamus. Conclusion The immune profile induced by different adjuvants is essential on the effects over fertility, gonadal function, and hypothalamic GnRH-I expression in immunocastrated animals. The work was aimed to evaluate testicular alterations induced by administration of immunocastration vaccine in Donkeys. Atotal of 12 adult Donkeys from the Northeast breed of Brazil were randomly distributed into control group (CG; n=04) and immunocastrationgroup (IG; n=08). Donkeys from IG received two doses of Bopriva® administered in an 30-day interval (Days 0 and 30). Bloodsamples were collected for testosterone dosage in this 30-day interval, when donkeys were weighted and measured for testicular biometry(testicular volume and tone). At the end of the experiment (Day 120), all donkeys were subjected to surgical castration in order to obtaintesticular samples for histopathological analysis and from epididymis for evaluation of sperm cells. There was no difference in live weight(P>0.05) within CG and IG groups during the experiment, however testosterone concentrations in IG donkeys were significantly (P<0.05)lower after 30 and 60 days after the first vaccine, remaining in such levels until the end of the experiment. The CG did not show variationsin testosterone levels during the experiment. No evidence of functional sperm cells was found in testisof IG donkeys, despite normal gametogenesisin CG. Testis from IG donkeys showed alterations in consistency and volume, suggesting a degenerative process, further confirmedby typical histopathological alterations of such pathology. Donkeys from the CG did not display these alterations. In IG donkeys, theductal lumen of the epididymis ductal was dilated, filled with proteinaceous and eosinophilous content alongside with acidofilous germcells. Furthermore, azoospermia was found in 75% of IG donkeys, while remaining animals showed extremely low sperm concentrationand no visible motility. Knowledge of tissue and cuts growth depending on the sex could be used to improve performance and efficiency. Computed tomography (CT) is a non-invasive technology that enables the study of the body composition of live animals during growth. The aims of the present study were (1) to evaluate variation in the body composition of four sex types (SEX) of pigs (castrated males (CM), immunocastrated males (IM), entire males (EM) and females (FE)) at the live weight of 30, 70, 100 and 120 kg, assessed using CT; (2) to model the growth of the main tissues and cuts; and (3) to predict the mature BW (MBW) of the four SEX and establish the relationships between the growth models and the MBW. There were significant phenotypic differences in the allometric growth of fat and lean among SEX. For the lean tissue, FE and EM showed higher values of the b coefficient than CM and IM (1.07 and 1.07 v. 1.00 and 1.02, respectively) all of them close to unity, indicating a proportional growth rate similar to live weight and that this tissue developed faster in FE and EM than in CM and IM. However, these differences were not related to differences in estimated MBW. There were significant differences in estimated MBW among SEX, being higher in IM and EM than in CM and FE (303 and 247 v. 219 and 216 kg), however, the MBW may have been overestimated, especially for the IM. The poorer accuracy of the MBW estimate for the IM could be due to a maximum live weight of 120 kg in the experiment, or to the fact that this particular SEX presented two clear behaviours, being more similar to EM from birth to the second injection of the vaccine (130 days) and comparable with CM from that point to the final BW. The objective was to determine the effects of immunization against gonadotropin-releasing hormone on reproductive characteristics in boars. A total of 72 boars were used in a randomized design with three treatments: single immunization (SI) (10 weeks of age) or double immunization (DI) (10 and 15 weeks of age) with Improvest® and intact controls (no Improvest®; CNT) (n=24/group). At 10, 15, 20, 25 and 40 weeks of age, blood was collected and serum harvested to evaluate testosterone concentrations. Testosterone concentrations were less for DI boars compared with CNT boars and SI boars at 20 and 25 weeks (P<0.001), but not at 40 weeks of age. At week 25, 18 pigs (n=6/group) were sacrificed and testes were removed, weighed and measured, and seminiferous tubules were examined and scored using histological slides of testes parenchyma. A sample of neck fat was assessed for boar taint aroma. All testicular measurements and weights and seminiferous tubule scores were less for DI boars compared with SI and CNT boars (P<0.001). More (P<0.05) SI and CNT boars had detectable boar taint aroma than DI boars. Libido was assessed at 32, 36, 47, 60 and 63 weeks of age and semen collected at 60 weeks of age was analyzed for indicators of quality. There were no effects of treatment (P=0.41) or treatment by week (P=0.71) on libido. Semen volume, gel weight and total number of sperm cells, determined in a subset of boars (n=3/treatment), were not different among treatments. Sperm concentration was greater for DI than SI (P=0.01), and tended to be greater for DI compared with CNT (P=0.10). Sperm motility tended to be greater for DI boars compared with CNT boars (P=0.066). The aim of this study was to evaluate the effects of immunocastration on pork meat and carcass quality, compared to meat from surgically castrated males and entire males. Ninety (Duroc x Pietrain) x (Landrace x Yorkshire) crossbred pigs were assigned to three experimental groups: surgically castrated males (barrows; castrated up to the seventh day of age), entire males (males), and vaccinated males (immunocastrates). Carcass and meat quality characteristics such as weight of hot and chilled carcass, meat yield, chilling loss and chemical parameters were examined. Surgically castrated pigs had significantly lower (p<0.01; p<0.05) weight before slaughter, than males and immunocastrates, and also lower (p<0.01) warm carcass weight than males. The average carcass meatiness of castrates was significantly lower (p<0.01) than the average meatiness of males and immunocastrates. Chilling loss of barrows was significantly lower (p<0.01) than chilling loss of males or immunocastrates. It was also found that the chilling loss of immunocastrates was significantly lower (p<0.05) than chilling loss of males. According to the results obtained, it can be concluded that immunocastration could be a good alternative to surgical castration considering meat and carcass quality characteristics. In immunocastrated (IC) pigs, revaccination (V2) increases lipid deposition (LD) because of increased voluntary feed intake; but little is known on associated effect of diet composition on partitioning of nutrients in IC pigs. Digestibility measurements, N and energy balances in respiration chambers were performed in two subsequent stages in four replicates of two male littermates to determine the changes between 85 (stage 1) and 135 (stage 2) kg live weight due to combined effect of IC, growth and increased feed intake (IC/growth). During stage 1, pigs received a standard low-fat diet (LF diet; 2.5% dry matter (DM) of fat fed at 2.27 MJ metabolizable energy (ME)/kg BW0.60 per day), whereas during stage 2, feed intake was increased to 2.47 MJ ME/kg BW0.60 per day and one littermate was fed LF diet whereas the second received a fat-enriched diet (HF diet; 8.9% DM of fat) to determine the effect of increased dietary fat content on energy utilization in IC pigs. Results from N balance and measurements of gas exchanges were used to calculate respiratory quotient (RQ), heat production (HP), nutrient contribution to fat retention, components of HP, protein deposition (PD) and LD. Nutrients and energy apparent digestibility coefficients, methane losses and N retention (P<0.05) increased with IC/growth. Despite higher ME intake, total HP remained similar (1365 kJ/kg of BW0.60 per day; P=0.47) with IC/growth. Consequently, total retained energy (RE) increased with IC/growth (from 916 to 1078 kJ/kg of BW0.60 per day; P<0.01) with a higher fat retention (625 to 807 kJ/kg BW0.60 per day; P<0.01), originating mainly from carbohydrates associated with a higher lipogenesis (536 to 746 kJ/kg BW0.60 per day; P<0.01) and RQ (1.095 to 1.145; P<0.01). Both PD (from 178 to 217 g/day; P=0.02) and LD (from 227 to 384 g/day; P<0.01) increased due to IC/growth. Feeding HF diet after IC was associated with increased crude fat digestibility (P<0.01) and increased RE as fat (807 to 914 kJ/kg BW0.60 per day; P=0.03), originating mainly from dietary fat (P<0.01) and resulting in increased LD (384 to 435 g/day; P<0.01) and lower RQ (from 1.145 to 1.073; P<0.01). Altogether, present results indicate that increased fatness of IC pigs is a result of increased daily LD caused by higher energy intake and lower basal metabolic rate. Physical castration (PC) is painful and stressful for nursing piglets. One alternative to PC is immunological castration (IC), but the pain and stress of handling associated with injections have not been assessed. The objectives of this study were to measure the pain and distress of subcutaneous (SQ) and intramuscular (IM) injections compared to PC in piglets, and to compare SQ or IM injections in finishing pigs. After farrowing, 3 to 5 d old male piglets were randomly assigned to (control) no handling treatment (NO), sham-handling (SHAM), IM, SQ, or PC. Finishing pigs were assigned to NO, SHAM, IM, or SQ. Behavior was monitored for 1 h prior and 1 h post treatment in each age group. Social, feeding behaviors, and signs of pain were recorded. Finishing pigs treated with SQ injections had higher feeding behaviors pre-treatment than they did post-treatment. Overall, physical castrations caused measurable pain-like behaviors and general behavioral dysregulation at a much higher level than the other treatment groups. SQ and IM injections did not cause either significant behavioral or physiological alterations in piglets. SQ injections caused a decrease in finishing pig feed behaviors post treatment (p = 0.02) and SHAM treated finishing pigs spent significantly more time lying than the other treatment groups. The aim of this study was to assess the effect of GnRH vaccination in boars (BO), barrows (BA), and gilts (GI) slaughtered at an average BW of 133 kg. Forty BO, 40 BA, and 40 GI were housed individually, fed ad libitum, and divided into 2 groups of 20 pigs per sex, a control group (CON) and a GnRH vaccinated group (IC). The IC group received 2 injections of GnRH vaccine (Improvac) at 70 and 105 kg. In BA, GnRH vaccination had minor effects on performance. Observed effects in BO and GI are therefore probably not caused by the GnRH vaccination itself but by the subsequent withdrawal of gonadal hormones. In BO, the increased feed intake after GnRH vaccination (P < 0.001), accompanied by a fall in estradiol and testosterone levels, led to faster growth (P = 0.014), increased back fat thickness (P = 0.021), and a tendency for a decreased meat percentage (P = 0.052). GnRH vaccination of BO decreased the boar taint-related sensory attribute scores similar to the levels of BA and GI (P < 0.001). In GI, the increased feed intake after GnRH vaccination (P < 0.001), accompanied by low progesterone levels, led to faster growth (P < 0.001), increased back fat thickness (P = 0.018), decreased meat percentage (P = 0.032), and a decreased shear force (P = 0.002) without significant differences in the sensory profile.
