Epidural anesthesia in dogs is a locoregional anesthesia technique used in veterinary medicine, becoming an important integrated application in the anesthetic protocol to provide safer and more effective analgesia to patients. For this, professionals must adhere to rigorous guidelines and possess technical skills. In this context, in veterinary education, the development of practical clinical skills represents a crucial aspect in the training of these professionals. However, traditional teaching methods have proven insufficient to ensure a consistent level of competence among recent graduates. The introduction of non-animal alternatives for educational purposes has contributed to the development of simulation-based teaching, an innovative and accessible field capable of enhancing pre-clinical proficiency in students and reducing the use of live animals and cadavers. Despite its application in various areas of veterinary education, there are no conclusive results regarding the development of accessible simulators capable of effectively enhancing training in epidural anesthesia in dogs. Therefore, this article represents a pioneering study aimed at sharing a method for creating SimuVet, a realistic simulator for training epidural anesthesia in dogs. The simulator was fully developed by veterinary researchers with limited experience in 3D printing and, after preliminary analysis, demonstrated excellent performance and ultrasonographic anatomy. Future work will focus on the formal validation of this simulator with the aim of improving the teaching and learning process for students and experts in performing epidural anesthesia in companion animals.

NETosis is a key host immune process against a pathogenic infection during innate immune activation, consisting of a neutrophil “explosion” and, consequently, NET formation, containing mainly DNA, histones, and other nuclear proteins. During sepsis, an exacerbated immune host response to an infection occurs, activating the innate immunity and NETosis events, which requires histone H3 citrullination. Our group compared the circulating histone levels with those citrullinated H3 levels in plasma samples of septic patients. In addition, we demonstrated that citrullinated histones were less cytotoxic for endothelial cells than histones without this post-translational modification. Citrullinated histones did not affect cell viability and did not activate oxidative stress. Nevertheless, citrullinated histones induced an inflammatory response, as well as regulatory endothelial mechanisms. Furthermore, septic patients showed elevated levels of circulating citrullinated histone H3, indicating that the histone citrullination is produced during the first stages of sepsis, probably due to the NETosis process.

Considering the high prevalence of cartilage-associated pathologies, low self-repair capacity and limitations of current repair techniques, tissue engineering (TE) strategies have emerged as a promising alternative in this field. Three-dimensional culture techniques have gained attention in recent years, showing their ability to provide the most biomimetic environment for the cells under culture conditions, enabling the cells to fabricate natural, 3D functional microtissues (MTs). In this sense, the aim of this study was to generate, characterize and compare scaffold-free human hyaline and elastic cartilage-derived MTs (HC-MTs and EC-MTs, respectively) under expansion (EM) and chondrogenic media (CM). MTs were generated by using agarose microchips and evaluated ex vivo for 28 days. The MTs generated were subjected to morphometric assessment and cell viability, metabolic activity and histological analyses. Results suggest that the use of CM improves the biomimicry of the MTs obtained in terms of morphology, viability and extracellular matrix (ECM) synthesis with respect to the use of EM. Moreover, the overall results indicate a faster and more sensitive response of the EC-derived cells to the use of CM as compared to HC chondrocytes. Finally, future preclinical in vivo studies are still needed to determine the potential clinical usefulness of these novel advanced therapy products.

The SWAN wave model is widely used in coastal waters and the main focus of this work is on its application in a harbor. Its last released version - SWAN 40.51 - includes an approximation to compute diffraction, however, so far there are few published works that discuss this matter. The performance of the model is therefore investigated in a harbor where reflection and diffraction play a relevant role. To assess its estimates, a phase-resolving Boussinesq wave model is employed as well, together with measurements carried out at a small-scale model of the area behind the breakwater. For irregular, short-crested waves with broad directional spreading, the importance of diffraction is relatively small. On the other hand, reflection of the incident waves is significant, increasing the energy inside the harbor. Nevertheless, the SWAN model does not achieve convergence when it is set to compute diffraction and reflection simultaneously. It is concluded that, for situations typically encountered in harbors, with irregular waves near reflective obstacles, the model should be set without the diffraction option.O modelo de ondas SWAN é amplamente empregado em simulações na região costeira e o presente trabalho investiga sua aplicação dentro de um porto. A última versão disponibilizada para a comunidade - SWAN 40.51 - inclui uma aproximação para computar a difração, embora, até o momento, poucos trabalhos abordando este tema foram publicados. O desempenho do modelo é estudado em um porto onde os fenômenos de reflexão e difração são importantes. Para avaliar suas estimativas, um modelo do tipo Boussinesq também é empregado, juntamente com medições realizadas em um modelo em escala reduzida da área atrás do quebramar. Para ondas irregulares, com cristas curtas e espalhamento direcional mais amplo, a importância da difração é relativamente menor. Contudo, o modelo SWAN não alcança convergência quando programado para estimar difração e reflexão simultaneamente. Conclui-se que, para situações normalmente encontradas em portos, com ondas irregulares próximas a obstáculos refletivos, o modelo deve ser empregado sem a opção de difração.

Duodenal hematoma is a rare complication of endoscopic duodenal biopsy that occurs mainly in children or adults with impaired coagulation. The clinical presentation consists of signs of intestinal obstruction, and pancreatitis and direct hyperbilirubinemia are possible complications caused by ampullary obstruction. A case of a six-year-old girl who presented with a duodenal hematoma and acute pancreatitis after having an endoscopic duodenal biopsy is reported. A review of the literature and data from all similar cases reported so far are briefly presented and discussed.
