Multiple propagation modes progress along the basilar membrane and Reissner’s membrane, called basilar membrane mode (BM mode) and Reissner’s membrane mode (RM mode), respectively. This study focuses on the effects of the RM mode on the hearing process and investigates the difference in the attenuation characteristics between the RM and BM modes in the vicinity of the cochlea base by using modal analysis. Results indicated that the RM mode has fewer effects on the hearing process, except otoacoustic emissions, due to its bigger attenuation constant than the BM mode in the vicinity of the cochlea base. The structural dependency of the attenuation constant of the RM mode is also investigated.

Commercially available cochlear implants are designed to aid profoundly deaf people in understanding speech and environmental sounds. A typical cochlear implant uses a bank of bandpass filters to decompose an audio signal into a set of dynamic signals. These filters’ critical center frequencies f0 imitate the human cochlea’s vibration patterns caused by audio signals. Gammatone filters (GTFs), with two unique characteristics: (a) an appropriate “pseudo resonant” frequency transfer function, mimicking the human cochlea, and (b) realizing efficient hardware implementation, could demonstrate them as unique candidates for cochlear implant design. Although GTFs have recently attracted considerable attention from researchers, a comprehensive exposition of GTFs is still absent in the literature. This paper starts by enumerating the impulse response of GTFs. Then, the magnitude spectrum, |H(f)|, and bandwidth, more specifically, the equivalent rectangular bandwidth (ERB) of GTFs, are derived. The simulation results suggested that optimally chosen filter parameters, e.g., critical center frequencies,f0; temporal decay parameter, b; and order of the filter, n, can minimize the interference of the filter bank frequencies and very likely model the filter bandwidth (ERB), independent of f0b. Finally, these optimized filters are applied to delineate a filter bank for a cochlear implant design based on the Clarion processor model.

Age-related hearing loss (ARHL) is the most common sensory degenerative disease and can significantly impact the quality of life in elderly people. A previous study using GeneChip miRNA microarray assays showed that the expression of miR-29a changes with age, however, its role in hearing loss is still unclear. In this study, we characterized the cochlear phenotype of miR-29a knockout (miR-29a–/–) mice and found that miR-29a-deficient mice had a rapid progressive elevation of the hearing threshold from 2 to 5 months of age compared with littermate controls as measured by the auditory brainstem response. Stereocilia degeneration, hair cell loss and abnormal stria vascularis (SV) were observed in miR-29a–/– mice at 4 months of age. Transcriptome sequencing results showed elevated extracellular matrix (ECM) gene expression in miR-29a–/– mice. Both Gene Ontology (GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis revealed that the key differences were closely related to ECM. Further examination with a transmission electron microscope showed thickening of the basilar membrane in the cochlea of miR-29a–/– mice. Five Col4a genes (Col4a1-a5) and two laminin genes (Lamb2 and Lamc1) were validated as miR-29a direct targets by dual luciferase assays and miR-29a inhibition assays with a miR-29a inhibitor. Consistent with the target gene validation results, the expression of these genes was significantly increased in the cochlea of miR-29a–/– mice, as shown by RT-PCR and Western blot. These findings suggest that miR-29a plays an important role in maintaining cochlear structure and function by regulating the expression of collagen and laminin and that the disturbance of its expression could be a cause of progressive hearing loss.

For a deeper understanding of the inner ear dynamics, a Finite-Element model of the human cochlea is developed. To describe the unsteady, viscous creeping flow of the liquid, a pressure-displacement-based Finite-Element formulation is used. This allows one to efficiently compute the basilar membrane vibrations resulting from the fluid-structure interaction leading to hearing nerve stimulation. The results show the formation of a travelingwave on the basilar membrane propagating with decreasing velocity towards the peaking at a frequency dependent position. This tonotopic behavior allows the brain to distinguish between sounds of different frequencies. Additionally, not only the middle ear, but also the transfer behavior of the cochlea contributes to the frequency dependence of the auditory threshold. Furthermore, the fluid velocity and pressure fields show the effect of viscous damping forces and allow us to deeper understand the formation of the pressure difference, responsible to excite the basilar membrane.

Abstract Background Despite significant anatomical variation amongst patients, cochlear implant frequency-mapping has traditionally followed a patient-independent approach. Basilar membrane (BM) length is required for patient-specific frequency-mapping, however cochlear duct length (CDL) measurements generally extend to the apical tip of the entire cochlea or have no clearly defined end-point. By characterizing the length between the end of the BM and the apical tip of the entire cochlea (helicotrema length), current CDL models can be corrected to obtain the appropriate BM length. Synchrotron radiation phase-contrast imaging has made this analysis possible due to the soft-tissue contrast through the entire cochlear apex. Methods Helicotrema linear length and helicotrema angular length measurements were performed on synchrotron radiation phase-contrast imaging data of 14 cadaveric human cochleae. On a sub-set of six samples, the CDL to the apical tip of the entire cochlea (CDLTIP) and the BM length (CDLBM) were determined. Regression analysis was performed to assess the relationship between CDLTIP and CDLBM. Results The mean helicotrema linear length and helicotrema angular length values were 1.6 ± 0.9 mm and 67.8 ± 37.9 degrees, respectively. Regression analysis revealed the following relationship between CDLTIP and CDLBM: CDLBM = 0.88(CDLTIP) + 3.71 (R 2 = 0.995). Conclusion This is the first known study to characterize the length of the helicotrema in the context of CDL measurements. It was determined that the distance between the end of the BM and the tip of the entire cochlea is clinically consequential. A relationship was determined that can predict the BM length of an individual patient based on their respective CDL measured to the apical tip of the cochlea.

The similarity of acoustic tasks performed by odontocete (toothed whale) and microchiropteran (insectivorous bat) biosonar suggests they may have common ultrasonic signal reception and processing mechanisms. However, there are also significant media and prey dependent differences, notably speed of sound and wavelengths in air vs. water, that may be reflected in adaptations in their auditory systems and peak spectra of out-going signals for similarly sized prey. We examined the anatomy of the peripheral auditory system of two species of FM bat (big brown bat Eptesicus fuscus; Japanese house bat Pipistrellus abramus) and two toothed whales (harbor porpoise Phocoena phocoena; bottlenose dolphin Tursiops truncatus) using ultra high resolution (11–100 micron) isotropic voxel computed tomography (helical and microCT). Significant differences were found for oval and round window location, cochlear length, basilar membrane gradients, neural distributions, cochlear spiral morphometry and curvature, and basilar membrane suspension distributions. Length correlates with body mass, not hearing ranges. High and low frequency hearing range cut-offs correlate with basilar membrane thickness/width ratios and the cochlear radius of curvature. These features are predictive of high and low frequency hearing limits in all ears examined. The ears of the harbor porpoise, the highest frequency echolocator in the study, had significantly greater stiffness, higher basal basilar membrane ratios, and bilateral bony support for 60% of the basilar membrane length. The porpoise’s basilar membrane includes a “foveal” region with “stretched” frequency representation and relatively constant membrane thickness/width ratio values similar to those reported for some bat species. Both species of bats and the harbor porpoise displayed unusual stapedial input locations and low ratios of cochlear radii, specializations that may enhance higher ultrasonic frequency signal resolution and deter low frequency cochlear propagation.

Cochlear implants (CIs) have become the standard treatment for severe-to-profound sensorineural hearing loss. Conventional CIs have some challenges, such as the use of extracorporeal devices, and high power consumption for frequency analysis. To overcome these, artificial basilar membranes (ABMs) made of piezoelectric materials have been studied. This study aimed to verify the conceptual idea of a totally implantable ABM system. A prototype of the totally implantable system composed of the ABM developed in previous research, an electronic module (EM) for the amplification of electrical output from the ABM, and electrode was developed. We investigated the feasibility of the ABM system and obtained meaningful auditory brainstem responses of deafened guinea pigs by implanting the electrode of the ABM system. Also, an optimal method of coupling the ABM system to the human ossicle for transducing sound waves into electrical signals using the middle ear vibration was studied and the electrical signal output according to the sound stimuli was measured successfully. Although the overall power output from the ABM system is still less than the conventional CIs and further improvements to the ABM system are needed, we found a possibility of the developed ABM system as a totally implantable CIs in the future.

The roles of the medial olivocochlear reflex (MOCR) in human hearing have been widely investigated but remain controversial. We reason that this may be because the effects of MOCR activation on cochlear mechanical responses can be assessed only indirectly in healthy humans, and the different methods used to assess those effects possibly yield different and/or unreliable estimates. One aim of this study was to investigate the correlation between three methods often employed to assess the strength of MOCR activation by contralateral acoustic stimulation (CAS). We measured tone detection thresholds (N = 28), click-evoked otoacoustic emission (CEOAE) input/output (I/O) curves (N = 18), and distortion-product otoacoustic emission (DPOAE) I/O curves (N = 18) for various test frequencies in the presence and the absence of CAS (broadband noise of 60 dB SPL). As expected, CAS worsened tone detection thresholds, suppressed CEOAEs and DPOAEs, and horizontally shifted CEOAE and DPOAE I/O curves to higher levels. However, the CAS effect on tone detection thresholds was not correlated with the horizontal shift of CEOAE or DPOAE I/O curves, and the CAS-induced CEOAE suppression was not correlated with DPOAE suppression. Only the horizontal shifts of CEOAE and DPOAE I/O functions were correlated with each other at 1.5, 2, and 3 kHz. A second aim was to investigate which of the methods is more reliable. The test–retest variability of the CAS effect was high overall but smallest for tone detection thresholds and CEOAEs, suggesting that their use should be prioritized over the use of DPOAEs. Many factors not related with the MOCR, including the limited parametric space studied, the low resolution of the I/O curves, and the reduced numbers of observations due to data exclusion likely contributed to the weak correlations and the large test–retest variability noted. These findings can help us understand the inconsistencies among past studies and improve our understanding of the functional significance of the MOCR.

To date, objective measurements and psychophysical experiments have been used to measure frequency dependent basilar membrane (BM) delays in humans; however, in vivo measurements have not been made. This study aimed to measure BM delays by performing intracochlear electrocochleography in cochlear implant recipients. Sixteen subjects with various degrees of hearing abilities were selected. Postoperative Computer Tomography was performed to determine electrode locations. Electrical potentials in response to acoustic tone pips at 0.25, 0.5, 1, 2, and 4 kHz and clicks were recorded with electrodes at the frequency specific region. The electrode array was inserted up to the characteristic cochlear frequency region of 250 Hz for 6 subjects. Furthermore, the array was inserted in the region of 500 Hz for 15 subjects, and 1, 2, and 4 kHz were reached in all subjects. Intracochlear electrocochleography for each frequency-specific tone pip and clicks showed detectable responses in all subjects. The latencies differed among the cochlear location and the cochlear microphonic (CM) onset latency increased with decreasing frequency and were consistent with click derived band technique. Accordingly, BM delays in humans could be derived. The BM delays increased systematically along the cochlea from basal to apical end and were in accordance with Ruggero and Temchin, 2007.

Objective: We used synchrotron radiation phase contrast imaging (SR-PCI) to study the 3D microanatomy of the basilar membrane (BM) and its attachment to the spiral ligament (SL) (with a conceivable secondary spiral lamina [SSL] or secondary spiral plate) at the round window membrane (RWM) in the human cochlea. The conception of this complex anatomy may be essential for accomplishing structural preservation at cochlear implant surgery. Material and methods: Sixteen freshly fixed human temporal bones were used to reproduce the BM, SL, primary and secondary osseous spiral laminae (OSL), and RWM using volume-rendering software. Confocal microscopy immunohistochemistry (IHC) was performed to analyze the molecular constituents. Results: SR-PCI reproduced the soft tissues including the RWM, Reissner’s membrane (RM), and the BM attachment to the lateral wall (LW) in three dimensions. A variable SR-PCI contrast enhancement was recognized in the caudal part of the SL facing the scala tympani (ST). It seemed to represent a SSL allied to the basilar crest (BC). The SSL extended along the postero-superior margin of the round window (RW) and immunohistochemically expressed type II collagen. Conclusions: Unlike in several mammalian species, the human SSL is restricted to the most basal portion of the cochlea around the RW. It anchors the BM and may influence its hydro-mechanical properties. It could also help to shield the BM from the RW. The microanatomy should be considered at cochlear implant surgery.

Gammatones have had a long history in auditory studies, and recent theoretical work suggests they may play an important role in cochlear mechanics as well. Following this lead, the present paper takes five examples of basilar membrane impulse responses and uses a curve-fitting algorithm to decompose them into a number of discrete gammatones. The limits of this ‘sum of gammatones’ (SOG) method to accurately represent the impulse response waveforms were tested and it was found that at least two and up to six gammatones could be isolated from each example. Their frequencies were stable and largely independent of stimulus parameters. The gammatones typically formed a regular series in which the frequency ratio between successive members was about 1.1. Adding together the first few gammatones in a set produced beating-like waveforms which mimicked waxing and waning, and the instantaneous frequencies of the waveforms were also well reproduced, providing an explanation for frequency glides. Consideration was also given to the impulse response of a pair of elastically coupled masses—the basis of two-degree-of-freedom models comprised of coupled basilar and tectorial membranes—and the resulting waveform was similar to a pair of beating gammatones, perhaps explaining why the SOG method seems to work well in describing cochlear impulse responses. A major limitation of the SOG method is that it cannot distinguish a waveform resulting from an actual physical resonance from one derived from overfitting, but taken together the method points to the presence of a series of closely spaced local resonances in the cochlea.

Auditory sensory outer hair cells are thought to amplify sound-induced basilar membrane vibration through a feedback mechanism to enhance hearing sensitivity. For optimal amplification, the outer hair cell-generated force must act on the basilar membrane at an appropriate time at every cycle. However, the temporal relationship between the outer hair cell-driven reticular lamina vibration and the basilar membrane vibration remains unclear. By measuring sub-nanometer vibrations directly from outer hair cells using a custom-built heterodyne low-coherence interferometer, we demonstrate in living gerbil cochleae that the reticular lamina vibration occurs after, not before, the basilar membrane vibration. Both tone- and click-induced responses indicate that the reticular lamina and basilar membrane vibrate in opposite directions at the cochlear base and they oscillate in phase near the best-frequency location. Our results suggest that outer hair cells enhance hearing sensitivity through a global hydromechanical mechanism, rather than through a local mechanical feedback as commonly supposed.

This paper presents a digital implementation of the Cascade of Asymmetric Resonators with Fast-Acting Compression (CAR-FAC) cochlear model. The CAR part simulates the basilar membrane's (BM) response to sound. The FAC part models the outer hair cell (OHC), the inner hair cell (IHC), and the medial olivocochlear efferent system functions. The FAC feeds back to the CAR by moving the poles and zeros of the CAR resonators automatically. We have implemented a 70-section, 44.1 kHz sampling rate CAR-FAC system on an Altera Cyclone V Field Programmable Gate Array (FPGA) with 18% ALM utilization by using time-multiplexing and pipeline parallelizing techniques and present measurement results here. The fully digital reconfigurable CAR-FAC system is stable, scalable, easy to use, and provides an excellent input stage to more complex machine hearing tasks such as sound localization, sound segregation, speech recognition, and so on.

In this paper we present two mathematical models for the basilar membrane. In the first model the membrane is represented as an annular region. In the second model the basilar membrane is treated as a rectangular region. Comparison of the two models allows us to study the effect of the curvature of the basilar membrane on the range of the frequencies of hearing. The differential equation of both models is a fourth order partial differential equation derived from the classical plate theory. Boundary conditions are defined as a region with four sides. The conditions are different on each side and together form an interesting physiological combination, relative to standard engineering problems. Eigenvalues of the differential equations of the two models are obtained numerically. A comparison of the eigenvalues of the two models clearly shows that the range of the hearing frequencies of the first model is larger than that of the second model. The results indicate strongly that the curvature of the basilar membrane plays an important role in the hearing process. Curvature and measurement of curvature should be allowed in future models and experiments of the inner ear.

Abstract Background Exposure to intense noise causes the excessive movement of the organ of Corti, stretching the organ and compromising sensory cell functions. We recently revealed changes in the transcriptional expression of multiple adhesion-related genes during the acute phases of cochlear damage, suggesting that the disruption of cell-cell junctions is an early event in the process of cochlear pathogenesis. However, the functional state of cell junctions in the sensory epithelium is not clear. Here, we employed graded dextran-FITC, a macromolecule tracer that is impermeable to the organ of Corti under physiological conditions, to evaluate the barrier function of cell junctions in normal and noise-traumatized cochlear sensory epithelia. Results Exposure to an impulse noise of 155 dB (peak sound pressure level) caused a site-specific disruption in the intercellular junctions within the sensory epithelium of the chinchilla cochlea. The most vulnerable sites were the junctions among the Hensen cells and between the Hensen and Deiters cells within the outer zone of the sensory epithelium. The junction clefts that formed in the reticular lamina were permeable to 40 and 500 but not 2,000 kDa dextran-FITC macromolecules. Moreover, this study showed that the interruption of junction integrity occurred in the reticular lamina and also in the basilar membrane, a site that had been considered to be resistant to acoustic injury. Finally, our study revealed a general spatial correlation between the site of sensory cell damage and the site of junction disruption. However, the two events lacked a strict one-to-one correlation, suggesting that the disruption of cell-cell junctions is a contributing, but not the sole, factor for initiating acute sensory cell death. Conclusions Impulse noise causes the functional disruption of intercellular junctions in the sensory epithelium of the chinchilla cochlea. This disruption occurs at an early phase of cochlear damage. Understanding the role of this disruption in cochlear pathogenesis will require future study.
