Abstract Leafy spurge (Euphorbia esula) is an important herb and potential energy source with medicinal value. Codon usage bias (CUB) is a static feature of genes and genomes that results from adaptation and selection during long-term evolution and facilitates molecular breeding in transgenic plants. Here, we used TransDecoder to identify candidate coding regions from the downloaded leafy spurge transcriptome and generate coding region annotation files based on reference genomes. The whole genome showed A/T bias, especially at terminal positions, and seven high-frequency codons were identified. We compared codon usage frequencies to identify candidate exogenous expression receptor systems for leafy spurge. The identified factors affecting leafy spurge CUB included natural selection and other factors, mutation pressure and base composition, with natural selection and other factors being dominant. The observed CUB was significantly positively correlated with the gene expression levels. Systematic analysis of whole-genome leafy spurge revealed that highly expressed protein-coding genes presented greater CUB than did less expressed protein-coding genes. Furthermore, the highly expressed genes tended to have terminal G/C bases. In summary, we conducted a series of related studies based on the leafy spurge whole-genome sequence and laid a foundation for selecting suitable exogenous expression receptor systems and improving gene expression levels.

Abstract For fractured gas reservoirs with strong water drive, gas phase trapping affects the gas recovery significantly. The recovery may be less than 50% for some reservoirs while it is only 12% for Beaver River gas field. The gas phase trapping mechanism has been revealed by the results of depletion experimental test. The residual pressure of the trapped gas is as high as 11.75 MPa with a 12.8 cm imbibition layer resulting in gas recovery deceased 49.5% compared with that without imbibition layer. A mathematical model is built to calculate the imbibition thickness based on capillary pressure and relative permeability of the matrix. The gas phase trapping are analyzed by two representative wells in Weiyuan gas field, the intermittent production reinforces the imbibition thickness and result in gas trapped in the matrix block with high residual pressure for the low performace gas wells, the extremely low gas recovery can be explained more rationally. That lays a foundation of improving the gas recovery for fractured reservoirs.

Multi-layer commingled production is the main feature of gas well development in Sulige tight sandstone gas reservoir, Ordos Basin. Identifying potential interference between layers and devising methods for their characterization are crucial considerations for optimizing the development of gas reservoirs. To address these issues, we designed a physical simulation experiment process and interlayer commingled mining schemes, implementing various interlayer combination modes. The results show that a common occurrence in the process of multi-layer commingled production of tight gas and water layers, whether it involves gas layers production alone or simultaneous production of gas and water layers. This phenomenon involves the crossflow of gas and water between layers, resulting in interlayer interference and a subsequent reduction in gas reservoir recovery. Based on these observations, the concept of an interlayer interference index in multi-layer commingled production in tight sandstone gas reservoirs is proposed. The interference index model is obtained by fitting the multiple linear regression method, showcasing its correlation with the physical properties of the reservoir. High water saturation and a significant permeability ratio of the water layer to the gas layer (exceeding the critical value of 1) can result in the early occurrence of interlayer interference and yield a higher interference index. Furthermore, based on the interference index model, a novel method for productivity evaluation of gas wells in tight gas reservoirs is established. The calculations demonstrate that the interference index curve effectively characterizes the interlayer interference performance of gas wells. The productivity and production performance predictions derived from this model align closely with historical production data, affirming the model's effectiveness and accuracy. Therefore, the interference index model emerges as a valuable tool for predicting the productivity and production performance of gas wells in Sulige tight sandstone gas reservoirs. The research results have important theoretical guidance and practical significance for the efficient development of Sulige tight sandstone gas reservoirs.

IntroductionAlkaloids are one of the main medicinal components of Dendrobium species. Dendrobium alkaloids are mainly composed of terpene alkaloids. Jasmonic acid (JA) induce the biosynthesis of such alkaloids, mainly by enhancing the expression of JA-responsive genes to increase plant resistance and increase the content of alkaloids. Many JA-responsive genes are the target genes of bHLH transcription factors (TFs), especially the MYC2 transcription factor.MethodsIn this study, the differentially expressed genes involved in the JA signaling pathway were screened out from Dendrobium huoshanense using comparative transcriptomics approaches, revealing the critical roles of basic helix-loop-helix (bHLH) family, particularly the MYC2 subfamily.Results and discussionMicrosynteny-based comparative genomics demonstrated that whole genome duplication (WGD) and segmental duplication events drove bHLH genes expansion and functional divergence. Tandem duplication accelerated the generation of bHLH paralogs. Multiple sequence alignments showed that all bHLH proteins included bHLH-zip and ACT-like conserved domains. The MYC2 subfamily had a typical bHLH-MYC_N domain. The phylogenetic tree revealed the classification and putative roles of bHLHs. The analysis of cis-acting elements revealed that promoter of the majority of bHLH genes contain multiple regulatory elements relevant to light response, hormone responses, and abiotic stresses, and the bHLH genes could be activated by binding these elements. The expression profiling and qRT-PCR results indicated that bHLH subgroups IIIe and IIId may have an antagonistic role in JA-mediated expression of stress-related genes. DhbHLH20 and DhbHLH21 were considered to be the positive regulators in the early response of JA signaling, while DhbHLH24 and DhbHLH25 might be the negative regulators. Our findings may provide a practical reference for the functional study of DhbHLH genes and the regulation of secondary metabolites.

To improve the poor understanding of the mechanism of spontaneous water imbibition in water invasion in gas reservoirs and overcome the difficulty in quantifying imbibed water and its impact on reservoir development, a study is carried out based on development simulation experiments and theoretical analysis of fluid mechanics. Spontaneous water imbibition is an important mechanical mechanism for the formation of a water seal zone in a low-permeability matrix through water invasion in gas reservoirs. Water imbibition experiments into the reservoir were carried out to clarify the trends of the imbibed distance and imbibed fluid volume to establish the theoretical model of imbibition and to make experimental corrections. An application study was performed based on the understanding of water imbibition and water sealing mechanisms and the building of an imbibition model. The water seal zones formed by imbibition during exploitation are predicted, and the seal-breaking pressure and influx of imbibed water in the water seal zone are evaluated. The research findings fully revealed the large impact of water imbibition on the development of water-driven gas reservoirs, providing technical support for the assessment of water sealing and the establishment of water control measures in water-driven gas reservoirs.

Abstract Traditional porous media imbibition models deviate from the actual imbibition process in oil and gas reservoirs. Experimental studies on gas–water imbibition in reservoirs were carried out to describe the dynamic profile variation process of wet phase saturation in reservoirs and to further reveal the variation of the imbibition front and the imbibition amount. Optimization and correction methods were established, and experimental verifications were performed. Studies have shown the following: (1) Unlike homogeneous porous media, the water phase imbibition process in oil and gas reservoirs is more complicated, and it is impossible for the maximum saturation of imbibition to reach 100%. (2) Contrary to the theoretical hypothesis, the imbibition of water is not piston-like, and there is a clear transition zone at the imbibition front. This transition zone is the main cause of water saturation variations in the imbibition zone; with the expansion of the imbibition zone, the influence of the transition zone on water saturation weakens. (3) Traditional theoretical models predict a positive correlation between the imbibition amount and the measurements; however, there is a large deviation in the numerical values, which must be corrected. (4) The L-W model was optimized and the parameter group fluid factor F and the reservoir factor R were proposed to characterize the properties of the fluid and the reservoir, respectively. These two parameters have a clear physical significance and are easy to accurately test. After experimental correction, the optimized model is favourably suitable for oil and gas reservoirs.

Abstract The development of water drive gas reservoirs (WDGRs) with fractures or strong heterogeneity is severely influenced by water invasion. Accurately simulating the rules of water invasion and drainage gas recovery countermeasures in fractured WDGRs, thereby revealing the mechanism of water invasion and an appropriate development strategy, is important for formulating water management measures and enhancing the recovery of gas reservoirs. In this work, physical simulation methods were proposed to gain a better understanding of water invasion and to optimize the water control of fractured WDGRs. Five groups of experiments were designed and conducted to probe the impacts of the distance between the fractures and the gas well, the drainage position, the drainage timing and the aquifer size on the water invasion and production performance of a gas reservoir. The gas and water production and the internal pressure drop were monitored in real time during the experiments. Based on the above experimental works, a theoretical analysis was conducted to quantitatively evaluate the performance of the gas reservoir recovery via the gas well production performance, water invasion, dynamic pressure drop and residual gas and water distribution analysis. The results show that when the fracture scale was appropriate, a gas well drilled close to a fracture (Experiment 1-3) or a high-permeability formation could also produce gas and achieve drainage efficiently. The recovery factor of Experiment 1-3 reached 62.5%, which was 24.6% and 21.1% higher than those of Experiments 1-1 and 1-2, respectively, which had wells drilled in low-permeability areas. Draining water near an aquifer can effectively inhibit water invasion during the early stage of gas recovery. The setup in Experiment 2-1 effectively inhibited water invasion and avoided the formation of water-sealed volumes of gas to recover 30% more gas than recovered with that of Experiment 1-1 without drainage wells. A shorter distance between the drainage well and the aquifer increased the drainage capacity and decreased the gas production capacity, respectively (Well 2 at Point A vs Point B). A larger aquifer had a lower gas recovery, which reduced the economic benefit. For example, due to an infinitely large aquifer, the reserves in Experiment 4-1 were developed by a single well, the gas recovery was only 33.4%. These research results are expected to be beneficial for the preparation of development plans and the optimization of water control measures for WDGRs.

Dendrobium officinale is both a traditional herbal medicine and a plant of high ornamental and medicinal value. Alkaloids, especially terpenoid indole alkaloids (TIAs), with pharmacological activities are present in the tissues of D. officinale. A number of genes involved in alkaloid biosynthetic pathways have been identified. However, the regulatory mechanisms underlying the precursor and methyl jasmonate (MeJA)-induced accumulation of alkaloids in D. officinale are poorly understood. In this study, we collected D. officinale protocorm-like bodies (PLBs) and treated them with TIA precursors (tryptophan and secologanin) and MeJA for 0 (T0), 4 (T4) and 24 h (T24); we also established control samples (C4 and C24). Then, we measured the total alkaloid content of the PLBs and performed transcriptome sequencing using the Illumina HiSeq 2,500 system. The total alkaloid content increased significantly after 4 h of treatment. Go and KEGG analysis suggested that genes from the TIA, isoquinoline alkaloid, tropane alkaloid and jasmonate (JA) biosynthetic pathways were significantly enriched. Weighted gene coexpression network analysis (WGCNA) uncovered brown module related to alkaloid content. Six and seven genes related to alkaloid and JA bisosynthetic pathways, respectively, might encode the key enzymes involved in alkaloid biosynthesis of D. officinale. Moreover, 13 transcription factors (TFs), which mostly belong to AP2/ERF, WRKY, and MYB gene families, were predicted to regulate alkaloid biosynthesis. Our data provide insight for studying the regulatory mechanism underlying TIA precursor and MeJA-induced accumulation of three types of alkaloids in D. officinale.

The WUSCHEL-related homeobox (WOX) proteins are widely distributed in plants and play important regulatory roles in growth and development processes such as embryonic development and organ development. Here, series of bioinformatics methods were utilized to unravel the structural basis and genetic hierarchy of WOX genes, followed by regulation of the WOX genes in four Euphorbiaceae species. A genome-wide survey identified 59 WOX genes in Hevea brasiliensis (H. brasiliensis: 20 genes), Jatropha curcas (J. curcas: 10 genes), Manihot esculenta (M. esculenta: 18 genes), and Ricinus communis (R. communis: 11 genes). The phylogenetic analysis revealed that these WOX members could be clustered into three close proximal clades, such as namely ancient, intermediate and modern/WUS clades. In addition, gene structures and conserved motif analyses further validated that the WOX genes were conserved within each phylogenetic clade. These results suggested the relationships among WOX members in the four Euphorbiaceae species. We found that WOX genes in H. brasiliensis and M. esculenta exhibit close genetic relationship with J. curcas and R. communis. Additionally, the presence of various cis-acting regulatory elements in the promoter of J. curcas WOX genes (JcWOXs) reflected distinct functions. These speculations were further validated with the differential expression profiles of various JcWOXs in seeds, reflecting the importance of two JcWOX genes (JcWOX6 and JcWOX13) during plant growth and development. Our quantitative real-time PCR (qRT-PCR) analysis demonstrated that the JcWOX11 gene plays an indispensable role in regulating plant callus. Taken together, the present study reports the comprehensive characteristics and relationships of WOX genes in four Euphorbiaceae species, providing new insights into their characterization.

Based on well test interpretation, production performance analysis, overburden permeability and porosity test, gas-water core flooding test and high-pressure mercury injection, a quantitative correlation has been built of in-situ effective permeability with routine permeability and water saturation, and the ranges of Main Flow Channel Index (MFCI) are determined for different permeability levels in porous sand gas reservoirs. A new method to evaluate the in-situ effective permeability of porous sand reservoir and a correlation chart of reserves producing degree and main flow channel index are established. The results reveal that the main flow channel index of porous sand gas reservoirs has close correlation with routine matrix permeability and water saturation. The lower the routine matrix permeability and the higher the water saturation, the lower the MFCI is. If the routine matrix permeability is greater than 5.0×10–3, the MFCI is generally greater than 0.5. When the routine matrix permeability is from 1.0×10–3 to 5.0×10–3, the MFCI is mainly between 0.2 and 0.5. When the routine matrix permeability is less than 1.0×10–3, the MFCI is less than 0.2. The evaluation method of in-situ effective permeability can be used to evaluate newly discovered or not tested porous sand gas reservoirs quickly and identify whether there is tight sand gas. The correlation chart of reserves producing degree and main flow channel index can provide basis for recoverable reserves evaluation and well infilling, and provide technical support for formulation of reasonable technical policy of gas reservoir.

Theaceae species are dicotyledonous angiosperms with extremely high ornamental and economic value. The chloroplast genome is traditionally used to study species evolution, expression of chloroplast genes and chloroplast transformation. Codon usage bias (CUB) analysis is beneficial for investigations of evolutionary relationships and can be used to improve gene expression efficiency in genetic transformation research. However, there are relatively few systematic studies of the CUB in the chloroplast genomes of Theaceae species. In this study, CUB and nucleotide compositions parameters were determined by the scripts written in the Perl language, CodonW 1.4.2, CU.Win2000, RStudio and SPSS 23.0. The chloroplast genome data of 40 Theaceae species were obtained to analyse the codon usage (CU) characteristics of the coding regions and the influence of the source of variation on CUB. To explore the relationship between the CUB and gene expression levels in these 40 Theaceae plastomes, the synonymous codon usage order (SCUO) and measure independent of length and composition (MILC) values were determined. Finally, phylogenetic analysis revealed the genetic evolutionary relationships among these Theaceae species. Our results showed that based on the chloroplast genomes of these 40 Theaceae species, the CUB was for codons containing A/T bases and those that ended with A/T bases. Moreover, there was great commonality in the CUB of the Theaceae species according to comparative analysis of relative synonymous codon usage (RSCU) and relative frequency of synonymous codon (RFSC): these species had 29 identical codons with bias (RSCU > 1), and there were 19 identical high-frequency codons. The CUB of Theaceae species is mainly affected by natural selection. The SCUO value of the 40 Theaceae species was 0.23 or 0.24, and the chloroplast gene expression level was moderate, according to MILC values. Additionally, we observed a positive correlation between the SCUO and MILC values, which indicated that CUB might affect gene expression. Furthermore, the phylogenetic analysis showed that the evolutionary relationships in these 40 Theaceae species were relatively conserved. A systematic study on the CUB and expression of Theaceae species provides further evidence for their evolution and phylogeny.

Shale gas storage and permeability space is between nanometers and micrometers and has strong multiscale characteristics, resulting in highly complicated shale gas flow. Therefore, the development of shale gas needs to clarify the seepage mechanism of shale gas and establish a flow model and flow equation that can analyze shale gas flow. In this paper, based on the single nanotube model, combined with the Weiyuan-Changning shale gas demonstration zone of the target layer of the Longmaxi formation shale core high-pressure mercury pore throat test results, the contribution of seepage, diffusion, transition flow, and free molecular flow to shale gas flow was calculated. The contribution of seepage and diffusion is over 95%, and seepage and diffusion are the main flow pattern. Then, a shale gas seepage and diffusion coupled flow model and coupled flow equation were established. A method for calculating shale permeability and diffusion was proposed using the relationship between flow pressure and shale gas velocity. Finally, shale gas flow experiment analysis was carried out to verify that the established shale gas flow model and flow equation can describe the shale gas flow well. The result shows that the flow rate of shale gas is composed of seepage flow rate and diffusion flow rate. The seepage flow rate is proportional to the pseudopressure difference and proportionate to the low-pressure squared difference. The diffusion velocity is proportional to the difference in shale gas density, and the pressure difference in low pressure is proportional. As the pressure of a shale gas reservoir decreases, the proportion of diffusion flow increases. The research results enrich the understanding of shale gas flow; it also has particular reference significance to the development of shale gas reservoirs.

Abstract Background Studies have demonstrated that BBX (B-BOX) genes play crucial roles in regulatory networks controlling plant growth, developmental processes and stress response. Nevertheless, comprehensive study of BBX genes in orchids (Orchidaceae) is not well studied. The newly released genome sequences of Dendrobium officinale and Phalaenopsis equestris have allowed a systematic analysis of these important BBX genes in orchids. Results Here we identified 19 (DoBBX01–19) and 16 (PeBBX01–16) BBX genes from D. officinale and P. equestris, respectively, and clustered into five clades (I-V) according to phylogenetic analysis. Thirteen orthologous, two DoBBXs paralogous and two PeBBXs paralogous gene pairs were validated. This gene family mainly underwent purifying selection, but five domains experienced positive selection during evolution. Noteworthy, the expression patterns of root, root_tips, stem, leaf, speal, column, lip, and flower_buds revealed that they might contribution to the formation of these tissues. According to the cis-regulatory elements analysis of BBX genes, qRT-PCR experiments were carried out using D. officinale PLBs (protocorm-like bodies) and displayed that these BBX genes were differentially regulated under AgNO3, MeJA (Methyl Jasmonate), ABA (abscisic acid) and SA (salicylic acid) treatments. Conclusions Our analysis exposed that DoBBX genes play significant roles in plant growth and development, and response to different environmental stress conditions of D. officinale, which provide aid in the selection of appropriate candidate genes for further functional characterization of BBX genes in plants.

Water invasion is a common occurrence in multilayer unconsolidated gas reservoirs, which results in excessive water production and reduces the economic life of gas wells. However, due to multiple layers, active edge water, and strong heterogeneity, the mechanism of water invasion and its effect in the unconsolidated sandstone gas reservoir require understanding in order to improve efficiency and minimize economic cost. In this study, an experimental study on edge water invasion of the multilayer commingled production in unconsolidated sandstone gas reservoirs was conducted to understand the water invasion process along with different permeability layers. The results show that the edge water invasion in the commingling production is mainly affected by two major factors including reservoir permeability and gas production rate, which jointly control the encroaching water advance path and speed. The nonuniform invade of edge water may occur easily and water prefers to invade toward the gas well along with high permeability layers when the commingling production is in the condition of large permeability gradient and high production rate. The bypass flow will occur when there are high permeability channels between the layers, which causes water blocking to low-permeability layers and periphery reservoirs far away from gas wells. The findings of this study can help for a better understanding of water invasion and the effects of reservoir properties so as to optimize extraction conditions and predict gas productivity in unconsolidated sandstone gas reservoirs.

Abstract Background The B-BOX (BBX) proteins have important functions in regulating plant growth and development. In plants, the BBX gene family has been identified in several plants, such as rice, Arabidopsis and tomato. However, there still lack a genome-wide survey of BBX genes in pear. Results In the present study, a total of 25 BBX genes were identified in pear (Pyrus bretschneideri Rehd.). Subsequently, phylogenetic relationship, gene structure, gene duplication, transcriptome data and qRT-PCR were conducted on these BBX gene members. The transcript analysis revealed that twelve PbBBX genes (48%) were specifically expressed in pear pollen tubes. Furthermore, qRT-PCR analysis indicated that both PbBBX4 and PbBBX13 have potential role in pear fruit development, while PbBBX5 should be involved in the senescence of pear pollen tube. Conclusions This study provided a genome-wide survey of BBX gene family in pear, and highlighted its roles in both pear fruits and pollen tubes. The results will be useful in improving our understanding of the complexity of BBX gene family and functional characteristics of its members in future study.

Ultrasonic-assisted extraction of quercetin from Dendrobium officinale was optimized by response surface methodology (RSM) using high-performance liquid chromatography as a separative method. Based on single-factor experiments and two-level factorial analysis, the ethanol concentration, solid-to-liquid ratio and ultrasonic power were selected as significant response factors. The amount of quercetin that we extracted from Dendrobium officinale was 2.506–2.594 μg/g under the extraction conditions, which showed that optimization could improve the extration rate of quercetin from Dendrobium officinale. Quercetin was extracted and detected within 12 consecutive months after the germination of Dendrobium officinale by optimizing the extraction process to analyze the accumulation of quercetin. The UV-B exposure experiments showed that the Dendrobium officinale leaves have different responses to low- and high-dose UV light. The results showed that the quercetin content in Dendrobium officinale could be changed by UV-B radiation, and the response of distinct tissue parts to varying intensities of UV-B radiation was different.

Reservoir quality and productivity of fractured gas reservoirs depend heavily on the degree of fracture development. The fracture evaluation of such reservoir media is the key to quantify reservoir characterization for the purposes such as well drilling and completion as well as development and simulation of fractured gas reservoirs. In this study, a pore-fracture network model was constructed to understand the effects of fracture on permeability in the reservoir media. The microstructure parameters of fractures including fracture length, fracture density, fracture number, and fracture radius were analyzed. Then two modes and effects of matrix and fracture network control were discussed. The results indicate that the network permeability in the fractured reservoir media will increase linearly with fracture length, fracture density, fracture number, and fracture radius. When the fracture radius exceeds 80 µm, the fracture radius has a little effect on network permeability. Within the fracture density less than 0.55, it belongs to the matrix control mode, while the fracture network control mode is dominant in the fracture density exceeding 0.55. The network permeability in the matrix and fracture network control modes is affected by fracture density and the ratio of fracture radius to pore radius. There is a great change in the critical density for the matrix network control compared with the fracture network control. This work can provide a better understanding of the relationship between matrix and fractures, and the effects of fracture on permeability so as to evaluate the fluid flow in the fractured reservoir media.

In plants, plant fructokinases (FRKs) are considered to be the main gateway of fructose metabolism as they can phosphorylate fructose to fructose-6-phosphate. Chinese white pears (Pyrus bretschneideri) are one of the popular fruits in the world market; sugar content is an important factor affecting the quality of the fruit. We identified 49 FRKs from four Rosaceae species; 20 of these sequences were from Chinese white pear. Subsequently, phylogenic relationship, gene structure and micro-collinearity were analysed. Phylogenetic and exon–intron analysis classified these FRKs into 10 subfamilies, and it was aimed to further reveal the variation of the gene structure and the evolutionary relationship of this gene family. Remarkably, gene expression patterns in different tissues or different development stages of the pear fruit suggested functional redundancy for PbFRKs derived from segmental duplication or genome-wide duplication and sub-functionalization for some of them. Additionally, PbFRK11, PbFRK13 and PbFRK16 were found to play important roles in regulating the sugar content in the fruit. Overall, this study provided important insights into the evolution of the FRK gene family in four Rosaceae species, and highlighted its roles in both pear tissue and fruits. Results presented here provide the appropriate candidate of PbFRKs that might contribute to fructose efflux in the pear fruit.

The varying law of on-way water saturation in the depletion-drive development (the formation pressure: 20 MPa depleting to abandonment pressure) of water-bearing gas reservoirs was studied through four groups of cores whose physical properties represent the rocks in Sichuan Xujiahe gas reservoirs, and cores permeability are 1.630×10−3 μm2, 0.580×10−3 μm2, 0.175×10−3 μm2, 0.063×10−3 μm2, respectively. Combined with the rock characteristics of microscopic pore structure and capillary pressure, the interaction mechanism between different permeability sandstone and water was analyzed and verified on two wells. The conclusions are as follows: (1) The water trapping effect in the sandstone reservoir is dependent on the sandstone reservoir permeability (the critical value is 0.175×10−3− 0.580×10−3 μm2). (2) The formation with permeability greater than 0.580×10−3 μm2 is characterized with big size pore and throat, low capillary pressure and weak trapping effect. So a portion of pore water would be displaced by gas and became movable water. (3) The tight formation with permeability less than 0.175×10−3 μm2 is characterized with small size pore and throat, high capillary pressure and strong trapping effect. When the rock core is saturated by moisture gas, the pore water would not be displaced by gas and would be trapped in the sandstone, which results in the water saturation increases rather than decreases. Drawdown pressure should be kept within a proper range to extend the life cycle of wells. Cellulose fibres were treated with sodium hydroxide (NaOH) to improve the accessibility of chemical reagents and the roughness of fibre surface at low temperatures. The accessibility, supermolecular structure, morphology, and physical properties of fibres were investigated using solid-state cross polarisation/magic angle spinning 13C nuclear magnetic resonance (CP/MAS 13C NMR), wide-angle X-ray diffraction (WAXD), and scanning electron microscopy (SEM). The results showed that in low temperature (-16 °C), the iodine sorption value of cellulose fibres is 145.2 mg. The shift from the C6 signal of the molecular structure to the amorphous area indicated a decrease in the crystallinity of the material from 66% to 8%, and the transformation from cellulose I to II occurred. The SEM revealed fold shapes in the fibre surface, which increased the fibre surface area.

In order to optimize the extraction of gigantol from Dendrobium officinale, the influence of methanol concentration, ultrasonic temperature, and liquid ratio on extraction efficiency was analysed by the response surface analysis method. The results show that the extraction rate reached a maximum when the methanol concentration was 92.98%, the solid-liquid ratio was 27.2 mL/g, and the extraction temperature was 41.41 °C. The content of gigantol of Dendrobium officinale in leaves was significantly higher than that in stems, reaching 4.7942 μg/g. The content of gigantol in Dendrobium huoshanensis Fengdou was significantly higher than that of other species of Fengdou. This experiment has practical significance for improving the utilization rate of Dendrobium officinale, and provides a reference for the study of the pharmacological and biological activity of gigantol.

Metabolomics technology has enabled an important method for the identification and quality control of Traditional Chinese Medical materials. In this study, we isolated metabolites from cultivated Dendrobium officinale and Dendrobium huoshanense stems of different growth years in the methanol/water phase and identified them using gas chromatography coupled with mass spectrometry (GC-MS). First, a metabolomics technology platform for Dendrobium was constructed. The metabolites in the Dendrobium methanol/water phase were mainly sugars and glycosides, amino acids, organic acids, alcohols.
