Concerns surrounding potential health and environmental impacts of per- and polyfluoroalkyl substances (PFAS) are growing at tremendous rates because adverse health impacts are expected with trace-level exposures. Extreme measures are required to mitigate potential PFAS contamination and minimize exposures. Extensive PFAS use results in the release of diverse PFAS species from domestic, industrial, and municipal effluents to wastewater, which partition to biosolids throughout secondary treatment. Biosolids generated during municipal wastewater treatment are a major environmental source of PFAS due to prevailing disposal practices as fertilizers. Pyrolysis is emerging as a viable, scalable technology for PFAS removal from biosolids while retaining nutrients and generating renewable, raw materials for energy generation. Despite early successes of pyrolysis in PFAS removal, significant unknowns remain about PFAS and transformation product fates in pyrolysis products and emissions. Applicable PFAS sampling methods, analytical workflows, and removal assessments are currently limited to a subset of high-interest analytes and matrices. Further, analysis of exhaust gases, particulate matter, fly ashes, and other pyrolysis end-products remain largely unreported or limited due to cost and sampling limitations. This paper identifies critical knowledge gaps on the pyrolysis of biosolids that must be addressed to assess the effectiveness of PFAS removal during pyrolysis treatment.

Pharmaceuticals in natural waters are considered emerging pollutants due to their low concentrations and the negative effects they pose to the environment. Common sources of such pollutants include untreated wastewater from hospitals, residential, industrial, and agricultural sources. Many wastewater treatment methods only remove a subset of all pharmaceuticals from the wastewater; remaining pharmaceuticals are discharged into natural waters, and ultimately drain into coastal areas. Regions without proper wastewater treatment are especially susceptible to such contamination. This study deals with the distribution, sources, and seasonal variability of pharmaceuticals in key aquatic systems in the Philippines. Two watershed continuums (Davao Gulf, Davao City; Macajalar Bay, Cagayan de Oro City); two tourist areas (Boracay Island, Aklan; Mabini, Batangas); and one pristine atoll (Tubbataha Reefs, Palawan)—all with varied prevailing human population pressures—were studied. Samples of hospital wastewater as well as groundwater, surface and bottom water samples from rivers and coastal seas collected during dry and wet seasons were analyzed using solid-phase extraction and liquid chromatography-tandem mass spectrometry. Thirty-four target pharmaceutical residues and antibiotics were extracted and quantified. Acetaminophen was detected at concentrations of up to 289.17 ppb in freshwater samples, and at concentrations of up to 253.39 ppb in seawater samples. Ubiquitous to all the sites was caffeine, reaching 1848.57 ppb. Sulfamethazine, a commonly used veterinary antibiotic, was detected at 764.91 ppb in a river site in Cagayan de Oro. Untreated hospital wastewater contained metformin, iopamidol, sulfamethoxazole, acetylsulfamethoxazole, ciprofloxacin, and azithromycin, but these pharmaceuticals were not detected in other river and coastal waters. Samples collected during the dry season exhibited higher concentrations than those from the wet season, which appears to be related to increase in transient populations from tourism activities as well as dilution. The presence of pharmaceutical residues and antibiotics in these areas and the potential impact on the environment indicate the need for stricter wastewater management measures, particularly in communities located near water bodies. As the results of this study show, such measures might be most beneficial and effective if imposed during dry season and in areas open to tourism.

The application of animal manures to cropland is an important nutrient recycling strategy in many parts of the world. Commonly, aggregated manure wastes contain chemical stressors including veterinary antimicrobials, heavy metals, and antimicrobial resistance genes (ARGs) that can stimulate the development and proliferation of antimicrobial resistance (AMR). While the presence of antimicrobials in manure is well-documented, the co-occurrence of other potentially impactful chemical stressors in swine manure remains underreported. This study quantifies and analyzes correlations between antimicrobials, metals, and certain ARGs present in manure samples from swine farms in Iowa, United States. Relationships between chemical stressors and different stages of swine production or feed composition are also investigated. Results revealed substantial levels of tetracyclines [up to 1,260 µg g−1 dry weight (d.w.) of manure for oxytetracycline] detected in all samples. Tiamulin, two ionophores (monensin and lasalocid), and one macrolide (tilmicosin) were detected at maximum class concentrations of 9.4, 0.547, and 0.472 µg g−1 d.w., respectively. The median relative abundances of ermB and tetM were 0.13 and 0.17 copies g−1 wet weight (w.w.) manure (normalized to 16S gene), respectively. Additionally, high levels of copper (Cu), iron (Fe), and zinc (Zn) were detected in all samples, with maximum concentrations of 887, 1,900, and 2,100 µg g−1 d.w., respectively. Notably, uranium (U) was detected in 11 samples, at concentrations up to 0.77 µg g−1. A global analysis of AMR-stressor relationships using Spearman’s rank correlation indicates Cu, and Ba are the most positively and significantly correlated with cytotoxic anhydrotetracycline (ATC) and/or anhydrochlortetracycline (ACTC) concentrations in all tested facilities (Cu-ATC: ρ = 0.67, p = 0.0093; Cu-ACTC: ρ = 0.75, p = 0.0022; Ba-ATC: ρ = 0.84, p = 0.0002). Interestingly, ermB and tetM genes were strongly, positively correlated to each other (ρ = 0.92, p < 0.0001), suggesting possible co-selection, despite the absence of correlation between ARGs and tetracycline concentrations. This study demonstrates the complexity of interactions between antimicrobials, metals, and ARGs in multiple manure storage pits prior to cropland application.

This study reveals unique information that fluorine nuclear magnetic resonance (19F-NMR) spectroscopy provides in the analysis of per- and polyfluoroalkyl substances (PFASs). Our results demonstrate that the intensity of the terminal -CF3 signal can be used to determine the total PFASs regardless of headgroup. Characteristic chemical shifts of different classes of PFASs can also be observed, and are useful for differentiating between classes of PFAS. The NMR spectra of PFASs with ether linkages (e.g. GenX) show characteristic reference signals for both -CF2 and -CF3 signals that are useful for detection. Notably, 19F-NMR can differentiate between PFASs, non-PFAS, and F− ions, eliminating the need for sample clean-up even for complex samples. To illustrate this, the 19F-NMR spectra of perfluorooctane sulfonic acid (PFOS) in biosolids extract and in clean solvent spiked with PFOS standard were compared, and showed a difference of < 0.3 % in their signal intensities. The lack of matrix effect is contrary to the suppression or enhancement observed in PFAS analysis by liquid chromatography with mass spectrometry, the most commonly used method for quantifying PFASs. This paper presents 19F-NMR reference spectra for 34 PFASs and discusses the complementarity of this method with other approaches for the total and class-specific analysis of PFASs.

Per- and polyfluoroalkyl substances (PFASs) are exceptionally stable chemicals due to their strong CF bonds. Nanoscale zero-valent iron (nZVI) particles have the potential to remove and degrade PFASs through redox activity. In this study, we deposited nZVI onto two-dimensional reduced graphene oxide (rGO) nanosheets and tested these synthesized rGO-nZVI nanohybrid (NH) for the treatment of a mixture of short- and long-chain PFASs in water with and without H2O2. All PFASs were removed at a higher efficiency by the rGO-nZVI NH than by the parent materials rGO and nZVI. Notably, the long-chain PFASs were removed at a faster rate than the short-chain PFASs. After a 10 min exposure to the rGO-nZVI NH without H2O2, the long-chain PFASs (perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA)) were removed by 85 % and 39 %, respectively, while short-chain PFASs (perfluoropentane sulfonic acid and perfluoropentanoic acid) were removed by 19 % and 18 %, respectively. The addition of H2O2 enhanced the PFAS treatment performance by 10–18 %, which can be attributed to the generation of reactive oxygen species by the rGO-nZVI NH. Liquid chromatography high-resolution mass spectrometry analysis confirmed the formation of unique shorter chain and partially defluorinated PFAS-Fe complexes from both PFOS and PFOA.

The vast number of chemicals potentially reaching aquatic environment pose a challenge in maintaining the quality of water resources. However, best management practices to improve water quality are typically focused on reducing nutrient transport without assessing how these practices may impact the occurrence of micropollutants. The potential for co-management of nutrients and organic micropollutants exists, but few studies have comprehensively evaluated the suite of contaminants associated with different water quality management practices (riparian zone restoration, stormwater management, etc.). Furthermore, most studies dealing with the determination of micropollutants in environmental samples include only a limited number of target analytes, leaving many contaminants undetected. To address this limitation, there has been a gradual shift in environmental monitoring from using target analysis to either suspect screening analysis (SSA) or non-targeted analysis (NTA), which relies on accurate mass measurements, mass spectral fragmentation patterns, and retention time information obtained using liquid chromatography coupled to high-resolution mass spectrometry. The work presented in this paper focuses on a wide-scope detection of micropollutants in surface water samples from the Potomac River watershed (United States). An in-house database composed of 1039 compounds based on experimental analysis of primary standards was established, and SSA workflow was optimized and applied to determine the presence of micropollutants in surface water. A total of 103 micropollutants were detected in the samples, some of which are contaminants that were not previously monitored and belong to various classes such as pharmaceuticals, personal care products, per-and polyfluoroalkyl substances and other persistent industrial chemicals. The impact of best management practices being implemented for nitrogen and phosphorus reductions were also assessed for their potential to reduce micropollutant transport. This work illustrates the advantages of suspect screening methods to determine a large number of micropollutants in environmental samples and reveals the potential to co-manage a diverse array of micropollutants based on shared transport and transformation mechanisms in watersheds.

Wastewater treatment plants (WWTPs) receive a confluence of sewage containing antimicrobials, antibiotic resistant bacteria, antibiotic resistance genes (ARGs), and pathogens and thus are a key point of interest for antibiotic resistance surveillance. WWTP monitoring has the potential to inform with respect to the antibiotic resistance status of the community served as well as the potential for ARGs to escape treatment. However, there is lack of agreement regarding suitable sampling frequencies and monitoring targets to facilitate comparison within and among individual WWTPs. The objective of this study was to comprehensively evaluate patterns in metagenomic-derived indicators of antibiotic resistance through various stages of treatment at a conventional WWTP for the purpose of informing local monitoring approaches that are also informative for global comparison. Relative abundance of total ARGs decreased by ∼50% from the influent to the effluent, with each sampling location defined by a unique resistome (i.e., total ARG) composition. However, 90% of the ARGs found in the effluent were also detected in the influent, while the effluent ARG-pathogen taxonomic linkage patterns identified in assembled metagenomes were more similar to patterns in regional clinical surveillance data than the patterns identified in the influent. Analysis of core and discriminatory resistomes and general ARG trends across the eight sampling events (i.e., tendency to be removed, increase, decrease, or be found in the effluent only), along with quantification of ARGs of clinical concern, aided in identifying candidate ARGs for surveillance. Relative resistome risk characterization further provided a comprehensive metric for predicting the relative mobility of ARGs and likelihood of being carried in pathogens and can help to prioritize where to focus future monitoring and mitigation. Most antibiotics that were subject to regional resistance testing were also found in the WWTP, with the total antibiotic load decreasing by ∼40–50%, but no strong correlations were found between antibiotics and corresponding ARGs. Overall, this study provides insight into how metagenomic data can be collected and analyzed for surveillance of antibiotic resistance at WWTPs, suggesting that effluent is a beneficial monitoring point with relevance both to the local clinical condition and for assessing efficacy of wastewater treatment in reducing risk of disseminating antibiotic resistance.

Antifouling (AF) coatings containing booster biocides are used worldwide as one of the most cost-effective ways to prevent the attachment of marine organisms to submerged structures. Nevertheless, many of the commercial biocides, such as Econea® (tralopyril), are toxic in marine environments. For that reason, it is of extreme importance that new efficient AF compounds that do not cause any harm to non-target organisms and humans are designed. In this study, we measured the half-maximal inhibitory concentration (IC50) of a promising nature-inspired AF compound, a triazolyl glycosylated chalcone (compound 1), in an immortalized human retinal pigment epithelial cell line (hTERT-RPE-1) and compared the results with the commercial biocide Econea®. We also investigated the effects of these biocides on the cellular lipidome following an acute (24 h) exposure using liquid chromatography quadrupole time-of-flight mass spectrometry (LC-Q-TOF/MS). Our results showed that compound 1 did not affect viability in hTERT-RPE-1 cells at low concentrations (1 μM), in contrast to Econea®, which caused a 40% reduction in cell viability. In total, 71 lipids were found to be regulated upon exposure to 10 µM of both compounds. Interestingly, both compounds induced changes in lipids involved in cell death, membrane modeling, lipid storage, and oxidative stress, but often in opposing directions. In general, Econea® exposure was associated with an increase in lipid concentrations, while compound 1 exposure resulted in lipid depletion. Our study showed that exposure to human cells at sublethal Econea® concentrations results in the modulation of several lipids that are linked to cell death and survival.

Persistent organic pollutants (POPs) accumulate in the food web of the Great Lakes ecosystem, causing concern that these chemicals play a role in the decline of avian species such as colonial-nesting waterbirds. In this study, samples from four life stages of the common tern (Sterna hirundo), a threatened species in New York State, were collected post-mortem in the Buffalo-Niagara region (United States). Brains (n = 26) and livers (n =27) were analyzed for polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and the insecticide p,p’-dichlorodiphenyltrichloroethane (DDT) and its two metabolites p,p’-dichlorodiphenyldichloroethylene (DDE) and p,p’-dichlorodiphenyldichloroethane (DDD). Detectable concentrations of PCBs and PBDEs were observed in all samples; concentrations of total PCBs ranged from 5.59 to 2,490 ng/g wet weight (ww), total PBDEs ranged from 1.09 to 494 ng/g ww, and DDE metabolites ranged from 0.56 to 637 ng/g ww. Analysis of the primary food source for terns in the Buffalo-Niagara region, emerald shiners (Notropis atherinoides), revealed that all three classes of POPs had similar contaminant profiles in the fish to those in the brains and livers of tern samples. Overall, small chicks contained greater concentrations of pollutants than medium chicks and juveniles, likely from maternal loading to eggs. These results underline the persistence of these legacy contaminants in the Great Lakes, despite their banning, and their biomagnification in threatened species through food web interactions.

Background: Antimicrobial resistance (AMR) is a growing public health concern. Recent research has suggested that interactions between pathogens and antibiotic residues in various environmental matrices promote the development and spread of AMR in the environment. The levels of antibiotic residues in the aquatic environment have been analysed globally. Recently, Predicted No Effect Environmental Concentration (PNEC) values for many antibiotics have been suggested, based on their estimated minimal selective concentrations for selected bacterial species. The PNEC values can serve as a guide on the maximum levels of antibiotic residues in an environmental matrix, below which resistance is unlikely to develop. Aim: We aimed to determine which of the antibiotics, considered as “priority antibiotics” by the World Health Organisation (WHO), most frequently exceeded their PNEC values in the global aquatic environment. Methods: We obtained data from the German Environment Agency pharmaceutical database on means, medians or single values of 12 antibiotic types in five different environmental matrices [municipal wastewater treatment plant effluent, industrial wastewater effluent, hospital wastewater effluent, surface water, and drinking water] across 47 countries. We compared the mean levels of the 12 antibiotics in each environmental matrix to their suggested PNEC values to determine which antibiotic types exceeded PNEC and were most likely to select for resistance. We also determined which environmental matrices and countries had the highest burden of antibiotic residues. Results: Our study revealed that 7.9% of all analyses of antibiotic residues performed in the environmental matrices globally exceeded PNEC. Ciprofloxacin and clarithromycin had the greatest proportion (>30%) of residues exceeding PNEC. Hospital wastewater and industrial wastewater had the highest burden of antibiotic residues exceeding PNEC. No antibiotics exceeded PNEC in drinking water. Conclusion: While most environmental monitoring studies have focused on municipal wastewater treatment plants, the limited number of studies on hospital wastewater and industrial wastewater revealed that a large number of antibiotic residues coming from these sources exceeded their PNEC values. Our study highlights the importance of implementing on-site treatment systems that aim to destroy antibiotics prior to discharging wastewater to surface waters. Attention needs to be focused on the role that environmental matrices, particularly our wastewater sites, play in promoting antibiotic resistance. Novel treatment technologies need to be developed and implemented to increase the removal efficiencies of treatment plants and from antibiotic manufacturing, and decrease the discharge of antibiotic residues into aquatic environments.

Antimicrobial resistance is a worldwide problem that is both pressing and challenging due to the rate at which it is spreading, and the lack of understanding of the mechanisms that link human, animal and environmental sources contributing to its proliferation. One knowledge gap that requires immediate attention is the significance of antimicrobial residues and other pharmaceuticals that are being discharged from wastewater treatment plants (WWTPs) on the dissemination of antimicrobial resistance in the environment. In this work we provide an approach to develop a harmonized analytical method for 8 classes of antimicrobials and other pharmaceuticals that can be used for global monitoring in wastewater and receiving waters. Analysis of these trace organic chemicals in the influent and effluent wastewater, and in the respective upstream and downstream receiving waters from different countries across the globe is not trivial. Here, we demonstrated that sample preparation using solid-phase extraction (SPE) not only provides a convenient and cost-effective shipping of samples, but also adds stability to the analytes during international shipping. It is important that SPE cartridges are maintained at cold temperature during shipment if the duration is longer than 7 days because a significant decrease in recoveries were observed after 7 days in the cartridges stored at room temperature, especially for sulfonamides and tetracyclines. To compensate for sample degradation during shipment, and matrix effects in liquid chromatography/mass spectrometry, the use of stable isotope labeled compounds should be employed when available and affordable. The importance of applying a defined tolerance for the ion ratios (Q/q) that have been optimized for wastewater and surface water is discussed. The tolerance range was set to be the mean Q/q of the analyte standard at various concentrations ±40% for the influent, and ±30% for the effluent, upstream, and downstream samples; for tetracyclines and quinolones, however, the tolerance range was ±80% in order to minimize false negative and false positive detection. The optimized procedures were employed to reveal differences in antimicrobial and pharmaceutical concentrations in influent, effluent, and surface water samples from Hong Kong, India, Philippines, Sweden, Switzerland, and United States. The antimicrobials with the highest concentrations in influent and effluent samples were ciprofloxacin (48,103 ng/L, Hong Kong WWTP 1) and clarithromycin (5178 ng/L, India WWTP 2), respectively. On the other hand, diclofenac (108,000 ng/L, Sweden WWTP 2), caffeine (67,000 ng/L, India WWTP 1), and acetaminophen (28,000 ng/L, India WWTP 1) were the highest detected pharmaceuticals in the receiving surface water samples. Hong Kong showed the highest total antimicrobial concentrations that included macrolides, quinolones, and sulfonamides with concentrations reaching 60,000 ng/L levels in the influent. The occurrence of antibiotics in the environment from discharges of wastewater treatment plants (WWTPs) and from the land application of antibiotic-laden manure from animal agriculture is a critical global issue because these residues have been associated with the increased emergence of antibiotic resistance in the environment. In addition, other classes of pharmaceuticals and personal care products (PPCPs) have been found in effluents of municipal WWTPs, many of which persist in the receiving environments. Analysis of antibiotics by liquid chromatography-tandem mass spectrometry (LC-MS/MS) in samples from different countries presents unique challenges that should be considered, from ion suppression due to matrix effects, to lack of available stable isotopically labeled standards for accurate quantification. Understanding the caveats of LC-MS/MS is important for assessing samples with varying matrix complexity. Ion ratios between quantifying and qualifying ions have been used for quality assurance purposes; however, there is limited information regarding the significance of setting criteria for acceptable variabilities in their values in the literature. Upon investigation of 30 pharmaceuticals in WWTP influent and effluent samples, and in receiving surface water samples downstream and upstream of the WWTP, it was found that ion ratios have higher variabilities at lower concentrations in highly complex matrices, and the extent of variability may be exacerbated by the physicochemical properties of the analytes. In setting the acceptable ion ratio criterion, the overall mean, which was obtained by taking the average of the ion ratios at all concentrations (1.56 to 100 ppb), was used. Then, for many of the target analytes included in this study, the tolerance range was set at 40% for WWTP influent samples and 30% for WWTP effluent, upstream, and downstream samples. A separate tolerance range of 80% was set for tetracyclines and quinolones, which showed higher variations in the ion ratios compared to the other analytes.
