Delivering piglets is one of the most energy-demanding activities sows undergo in their lifetime. Sows can have myometrial contractions from 2 to 12 h before the first piglet is expelled as well as a nest-building behavior. Thus, when the first piglet is delivered, the female has already used part of her energy supply. When the sow gets exhausted due to lack of energy, the farrowing process can be interrupted, causing damage to the viability and vitality of the piglets. In the present study, we evaluated the effects of feeding sows an energy supplement at the onset of farrowing on farrowing kinetics and piglet vitality. The energy supplement consisted of a blend of carbohydrates and glycerol which provides 439 kJ of metabolizable energy per kg of metabolic weight. A total of 180 sows were used. At the onset of farrowing, sows were assigned to one of the following treatments: sows that were not supplied energy at the onset of farrowing, serving as controls (CON, n = 85); sows fed the energy supplement at the onset of farrowing (ESP, n = 95). Farrowing kinetics, blood glucose concentration, and piglet vitality were recorded for each sow. Blood glucose concentration was assessed by puncturing the auricular vein and using a portable glucometer at four different time points: after the birth of the 1st piglet (T0), and at 20 (T20), 40 (T40), 80 (T80), and 180 (T180) min after the birth of the 1st piglet. The vitality of the 1st, 6th, 12th, 17th, and 20th piglet born was evaluated using the Apgar score. Piglet birth weight and average colostrum intake were measured. The farrowing duration was 20 min shorter (P 0.05). In conclusion, feeding a blend of carbohydrates and glycerol as an energy supplement for farrowing sows improved farrowing kinetics and piglet vitality score.

Osteoporosis is a major complication of chronic cholestatic liver disease (CCLD). We evaluated the efficacy of using disodium pamidronate (1.0 mg/kg body weight) for the prevention (Pr) or treatment (Tr) of cholestasis-induced osteoporosis in male Wistar rats: sham-operated (Sham = 12); bile duct-ligated (Bi = 15); bile duct-ligated animals previously treated with pamidronate before and 1 month after surgery (Pr = 9); bile duct-ligated animals treated with pamidronate 1 month after surgery (Tr = 9). Rats were sacrificed 8 weeks after surgery. Immunohistochemical expression of IGF-I and GH receptor was determined in the proximal growth plate cartilage of the left tibia. Histomorphometric analysis was performed in the right tibia and the right femur was used for biomechanical analysis. Bone material volume over tissue volume (BV/TV) was significantly affected by CCLD (Sham = 18.1 ± 3.2 vs Bi = 10.6 ± 2.2%) and pamidronate successfully increased bone volume. However, pamidronate administered in a preventive regimen presented no additional benefit on bone volume compared to secondary treatment (BV/TV: Pr = 39.4 ± 12.0; Tr = 41.2 ± 12.7%). Moreover, the force on the momentum of fracture was significantly reduced in Pr rats (Sham = 116.6 ± 23.0; Bi = 94.6 ± 33.8; Pr = 82.9 ± 22.8; Tr = 92.5 ± 29.5 N; P < 0.05, Sham vs Pr). Thus, CCLD had a significant impact on bone histomorphometric parameters and pamidronate was highly effective in increasing bone mass in CCLD; however, preventive therapy with pamidronate has no advantage regarding bone fragility.

Bone mass loss is a major complication of chronic cholestatic liver disease (CCD). However, the long-term impact of CCD on bone mass acquisition is unknown. We longitudinally assessed bone mineral density (BMD) and factors involved in bone remodeling in 9 children and adolescents with CCD Child-Pugh A (5 boys/4 girls) and in 13 controls (6 boys/7 girls). The groups were evaluated twice, at baseline (T0) and after 3 years (T1), when osteocalcin, deoxypyridinoline, 25-hydroxyvitamin-D, parathyroid hormone, insulin-like growth factor-I (IGF-I), and BMD (L1-L4, proximal femur and total body) were determined. Serum levels of receptor activator for nuclear factor kB ligand (RANKL) and osteoprotegerin were measured only at T1. Lumbar spine BMD was reanalyzed twice: after adjustment for bone age and to compensate for the height factor. Volumetric density was also estimated mathematically in L2-L4. The BMD of L1-L4 was lower in the CCD group (Z-score at T0: control = -1.2 ± 0.8 vs CCD = -2.2 ± 1.4, P < 0.05; T1: control = -0.7 ± 0.8 vs CCD = -2.1 ± 1.1, P < 0.05). Osteocalcin and deoxypyridinoline were similar for the two groups. The CCD group presented lower IGF-I (Z-score at T1: control = 1.4 ± 2.8 vs CCD = -1.5 ± 1.0, P < 0.05) and RANKL (control = 0.465 ± 0.275 vs CCD = 0.195 ± 0.250 pM, P < 0.05) than control. Children with compensated CCD Child-Pugh A showed early impairment of bone acquisition, with the impact being more severe in an initial phase and then tapering in a slowly progressive way. Reduction in endocrine IGF-I has a crucial role in this process.

Avaliaram-se os efeitos da ractopamina e de dois níveis de lisina digestível na dieta sobre o desempenho e características de carcaça de leitoas com peso corporal a partir de 85kg, durante 21 e 28 dias antes do abate, os quais foram distribuídos em delineamento experimental de blocos ao acaso com cinco repetições de dois animais por unidade experimental. Os tratamentos consistiram de arranjo fatorial 2x2, com dois níveis de lisina digestível (LD), 0,67 e 0,87%, e dois níveis de ractopamina (0 e 5ppm). Observou-se efeito significativo do nível de LD sobre o ganho de peso e peso corporal e interação significativa (P0,05) do nível de LD sobre a espessura de toucinho, profundidade de lombo e rendimento em carne magra da carcaça de acordo com avaliação da carcaça, feita in vivo. Não houve efeito (P>0,05) da ractopamina sobre o consumo de ração, o ganho de peso diário e o peso corporal aos 21 e 28 dias de experimento. A ractopamina reduziu a espessura de toucinho no ponto P2 aos 21 dias e aumentou o rendimento de carne magra e a profundidade de lombo na avaliação da carcaça no frigorífico.The effects of ractopamine and digestible lysine levels on performance and carcass traits of gilts with initial body weight average of 85kg, recorded during 21 and 28 day before slaughtering time were evaluated in a completely randomized block design with five replicates of two animals per experiment unit. The treatments consisted in a 2 x 2 factorial scheme, two levels of digestible lysine (DL) (0.67 and 0.87%) and two ractopamine levels (0 and 5ppm). Significant effects of DL on daily weight gain and body weight and significant effects of DL and ractopamine level were observed for feed:weight gain ratio and lean meat carcass deposition. No significant effects (P>0.05) of DL on fat thickness, muscle depth and lean meat yield according to the in vivo carcass evaluation were observed. No significant effect of ractopamine (P>0.05) on feed intake, daily weight gain and body weight (21 and 28 days) were observed. Ractopamine reduced back fat thickness at P2 point (21days) and increased lean meat yield and muscle depth in frigorific carcass evaluation.

Data about the impact of bariatric surgery (BS) and subsequent weight loss on bone are limited. The objective of the present study was to determine bone mineral density (BMD), bone remodeling metabolites and hormones that influence bone trophism in premenopausal women submitted to BS 9.8 months, on average, before the study (OGg, N = 16). The data were compared to those obtained for women of normal weight (CG, N = 11) and for obese women (OG, N = 12). Eight patients in each group were monitored for one year, with the determination of BMD, of serum calcium, phosphorus, magnesium, parathyroid hormone, 25-hydroxyvitamin D, insulin-like growth factor-I (IGF-I) and osteocalcin, and of urinary calcium and deoxypyridinoline. The biochemical determinations were repeated every three months in the longitudinal study and BMD was measured at the end of the study. Parathyroid hormone levels were similar in the three groups. IGF-I levels (CG = 332 ± 62 vs OG = 230 ± 37 vs OGg = 128 ± 19 ng/mL) were significantly lower in the operated patients compared to the non-operated obese women. Only OGg patients presented a significant fall in BMD of 6.2% at L1-L4, of 10.2% in the femoral neck, and of 5.1% in the forearm. These results suggest that the weight loss induced by BS is associated with a significant loss of bone mass even at sites that are not influenced by weight overload, with hormonal factors such as IGF-I being associated with this process.

We assessed the effect of chronic hyperglycemia on bone mineral density (BMD) and bone remodeling in patients with type 2 diabetes mellitus. We investigated 42 patients with type 2 diabetes under stable control for at least 1 year, 22 of them with good metabolic control (GMC: mean age = 48.8 ± 1.5 years, 11 females) and 20 with poor metabolic control (PMC: mean age = 50.2 ± 1.2 years, 8 females), and 24 normal control individuals (CG: mean age = 46.5 ± 1.1 years, 14 females). We determined BMD in the femoral neck and at the L2-L4 level (DEXA) and serum levels of glucose, total glycated hemoglobin (HbA1), total and ionic calcium, phosphorus, alkaline phosphatase, follicle-stimulating hormone, intact parathyroid hormone (iPTH), 25-hydroxyvitamin D (25-OH-D), insulin-like growth factor I (IGFI), osteocalcin, procollagen type I C propeptide, as well as urinary levels of deoxypyridinoline and creatinine. HbA1 levels were significantly higher in PMC patients (12.5 ± 0.6 vs 7.45 ± 0.2% for GMC and 6.3 ± 0.9% for CG; P < 0.05). There was no difference in 25-OH-D, iPTH or IGFI levels between the three groups. BMD values at L2-L4 (CG = 1.068 ± 0.02 vs GMC = 1.170 ± 0.03 vs PMC = 1.084 ± 0.02 g/cm²) and in the femoral neck (CG = 0.898 ± 0.03 vs GMC = 0.929 ± 0.03 vs PMC = 0.914 ± 0.03 g/cm²) were similar for all groups. PMC presented significantly lower osteocalcin levels than the other two groups, whereas no significant difference in urinary deoxypyridine was observed between groups. The present results demonstrate that hyperglycemia is not associated with increased bone resorption in type 2 diabetes mellitus and that BMD is not altered in type 2 diabetes mellitus.

Determinaram-se as exigências de lisina para suínos machos castrados utilizando-se 40 animais distribuídos em delineamento experimental de blocos ao acaso, com cinco tratamentos (0,5; 0,6; 0,7; 0,8 e 0,9% de lisina total), quatro repetições e dois animais por baia. Adicionaram-se aminoácidos sintéticos para se manter o nível de acordo com o perfil da proteína ideal. Observou-se efeito quadrático dos tratamentos sobre o ganho de peso diário, espessura do toucinho na 10ª costela, espessura do toucinho na última costela e conversão alimentar. Houve efeito linear sobre o consumo de lisina diário e taxa de deposição de carne magra diária. O modelo linear response plateau ajustou-se melhor aos resultados da taxa de deposição de carne magra diária. Não houve efeito sobre o consumo de ração diário, espessura de toucinho no P1, espessura de toucinho no P2, profundidade de lombo, porcentagem de carne magra, rendimento de carcaça, comprimento de carcaça pelo método brasileiro, comprimento de carcaça pelo método americano e rendimento de pernil. A exigência de lisina total para suínos machos castrados de 95 a 122kg, selecionados para deposição de carne magra na carcaça, é de 0,76%, correspondendo ao consumo de 25,99g/dia.Lysine requirements of crossbred barrows (AG-PIC 412 X C-22), using 40 animals in a completely randomized block design with five dietary treatments (.50; .60; .70; .80 and .90% of total lysine), four replicates and two animals per experimental unit were estimated. Synthetic amino acids were added to keep amino acids levels according to the ideal protein profile. Quadratic effect of treatments on average daily gain, on feed conversion, on 10th rib fat depth and on last rib fat depth were observed. Linear effects of treatments on daily lysine intake and on daily lean gain, were observed. No effects of lysine level on daily feed intake, P1 backfat thickness, P2 backfat thickness, loin depth, lean percentage, carcass yield, carcass length, carcass length measured by Brazilian method and ham percentage were found. The total dietary lysine requirement for 95 to 122kg high lean barrows is .76%, which corresponds to approximately 25.99g/day of lysine intake.

Abstract The extensive marine biodiversity has proved to be a promising source of substances with biomedical potential. In this study, the cytotoxicity of the Brazilian octocoral Phyllogorgia dilatata (Gorgoniidae) was evaluated against two tumor cell lines and three bacterial strains. The methanol/dichloromethane crude extract presented no antibacterial activity up to the highest concentration tested (512 µg/mL), however it revealed a noteworthy antiproliferative effect against HCT-116 (80%) and MCF-7 (54%) cell lines at 50 μg/mL. Therefore, guided by the cytotoxic activity, a multistep chemical fractionation of the extract provided the subfraction 5 (PDPH2-5) with IC50 values of 3.18 and 17.80 μg/mL against HCT-116 and MCF-7, respectively. The LC-HRMS/MS analysis of PDPH2-5 showed ions of m/z 219.1742 and 219.1743, characterized as (E,E) and (Z,E) germacrone, after a LC-DAD-SPE/NMR analysis of the hexanic fraction and comparisons of NMR data with the literature. Previously reported assessments to the cytotoxic activity of the (E,E)-diastereoisomer disclosed higher IC50 values than that obtained for the PDPH2-5 fraction, suggesting, herein, a potentiated effect of the diastereoisomeric mixture. Such remark encourage further bioactivity studies with stereoisomer mixtures and reduce the urge for compound isolation.

Avaliou-se o efeito da ractopamina e de diferentes métodos de formulação de dietas sobre o desempenho e as características de carcaça de leitoas em terminação. Foram utilizadas 60 leitoas, híbridas comerciais, distribuídas em delineamento experimental de blocos ao acaso, em esquema fatorial 2x3, sendo dois níveis de ractopamina e três métodos de formulação de dietas. Houve interação de métodos de formulação versus ractopamina para ganho de peso diário, peso corporal, conversão alimentar e taxa de deposição de carne magra (TDCM) na avaliação de carcaça in vivo por ultrassom. A dieta formulada com alta proteína bruta proporcionou maior consumo de dieta (CRD), de lisina (CLD), ganho de peso diário (GPD) e peso corporal final. A suplementação de ractopamina não influenciou o CRD e o CLD, entretanto aumentou o GPD. O método de formulação da dieta não influenciou as características de carcaça aos 28 dias, enquanto a suplementação de ractopamina reduziu a espessura de toucinho, aumentou a profundidade de lombo e o rendimento de carne magra. A dieta formulada com alta proteína bruta proporcionou melhores resultados de desempenho e taxa de deposição de carne magra na carcaça de leitoas em terminação, suplementadas com ractopamina.

Leishmania amazonensis is the causative agent of American cutaneous leishmaniasis, an important neglected tropical disease. Once Leishmania amazonensis is inoculated into the human host, promastigotes are exposed to the extracellular matrix (ECM) of the dermis. However, little is known about the interaction between the ECM and Leishmania promastigotes. In this study we established L. amazonensis promastigote culture in a three-dimensional (3D) environment mainly composed of Collagen I (COL I). This 3D culture recreates in vitro some aspects of the human host infection site, enabling the study of the interaction mechanisms of L. amazonensis with the host ECM. Promastigotes exhibited “freeze and run” migration in the 3D COL I matrix, which is completely different from the conventional in vitro swimming mode of migration. Moreover, L. amazonensis promastigotes were able to invade, migrate inside, and remodel the 3D COL I matrix. Promastigote trans-matrix invasion and the freeze and run migration mode were also observed when macrophages were present in the matrix. At least two classes of proteases, metallo- and cysteine proteases, are involved in the 3D COL I matrix degradation caused by Leishmania. Treatment with a mixture of protease inhibitors significantly reduced promastigote invasion and migration through this matrix. Together our results demonstrate that L. amazonensis promastigotes release proteases and actively remodel their 3D environment, facilitating their migration. This raises the possibility that promastigotes actively interact with their 3D environment during the search for their cellular “home”—macrophages. Supporting this hypothesis, promastigotes migrated faster than macrophages in a novel 3D co-culture model.

A Chlamydia trachomatis, Ureaplasma urealyticum e Mycoplasma hominis, são bactérias potencialmente sexualmente transmissíveis, causadoras de endocervicite e outras infecções genitais. As complicações por estes patógenos podem culminar em endometrites, doença inflamatória pélvica, bartholinites, uretrites posteriores, esterilidade e até infecções graves no neonato transmitidas de modo vertical (de mãe para o feto). Mesmo sendo infecções altamente prevalentes, permanecem desconhecidas devido à sua pequena ou ausência de sintomatologia, prosseguindo assim, sem tratamento. Carecem estudos em nosso meio, que revelem a verdadeira incidência destas infecções. Objetivos: Determinar a freqüência de C. trachomatis, U. urealyticum e M. hominis na endocérvice de mulheres atendidas em clínica privada de ginecologia do Município de Teresópolis – RJ. Metodologia: Foi realizado um estudo prospectivo com mulheres sexualmente ativas com idade entre 13 e 49 anos que procuraram atendimento ginecológico por motivos diversos, não tendo usado medicação antibiótica oral ou vaginal até 15 dias antes. O estudo envolveu dados como: anamnese, exame ginecológico, coleta de material para colpocitologia oncótica e de material endocervical para detecção das bactérias já citadas. A metodologia de detecção foi realizada através de imuno-ensaio enzimático (Elisa) – Sistema Automatizado Mini-Vidas para C. trachomatis e cultura para M. hominis e U. urealyticum (Bio-Merieux). Resultados: A amostra constituiu-se de 100 pacientes. Detectaram-se antígenos clamidiais em 5 mulheres (5%). Para U. urealyticum a positividade foi de 33% e para M. hominis foi de 4%. Os casos de positividade para C. trachomatis e M. hominis foram totalmente curados com 1 grama de azitromicina via oral em dose única, enquanto que para U. urealyticum obtivemos 81,8% de cura. Conclusões: A maior freqüência encontrada foi para U. urealyticum (33%). Os principais motivos da consulta médica foram leucorréia e rotina. Obtivemos excelentes índices de cura com o uso da azitromicina 1 grama via oral em dose única.
