In order to mitigate the risk of antibiotic resistance in poultry, scientists nowadays consider plant secondary metabolites to be a major organic antibacterial substitute. This study aimed to characterize and investigate the in silico, in vitro, and in vivo antibacterial effects of peppermint essential oil (PEO) in the form of a nanoemulsion (NE), termed PEONE. Menthol as a major compound of PEO has been identified by gas chromatography and mass spectroscopy (GCMS) analysis as 32.3 %, while lower droplet size, polydispersity Index (PDI), and optimum zeta potential values depicted the stability of PEONE have been observed and validated by transmission electron microscopy (TEM) micrograph image. In silico antibacterial activity was studied by molecular docking of menthol and enrofloxacin with Topoisomerase IV protein (PDB: 1s16;) of Escherichia coli K12 MG1655 and this effect was validated by in vitro and in vivo analysis. In vitro analysis, sustained release of PEONE has been observed against Escherichia coli and Staphylococcus aureus. In this study for in vivo experiments (n = 90) day-old broiler chicks were distributed into 6 dietary treatments with 5 replicates of 3 birds per replication. Dietary treatments included 1) Negative control (basal diet), 2) Positive control (basal diet + 200 µl enrofloxacin), 3) 25 µl PEONE + basal diet, 4) 50 µl PEONE + basal diet, 5) 75 µl PEONE + basal diet, and 6) 100 µl PEONE + basal diet. Analyzed data by different statistical tools confirmed that PEONE significantly affected body weight gain (BWG) with an improved feed conversion ratio (FCR) compared to the control group. A significant increase in cecal Lactobacillus count and a decrease in total coliform was observed. Positive effects on physiological parameters, visceral organs, and meat quality characteristics have been observed. In conclusion, our experiments suggest that PEONE can be used in the broiler industry as a substitute for antibiotics to minimize bacterial resistance.

Colitis, an inflammatory condition of the colon that encompasses ulcerative colitis (UC) and Crohn’s disease, presents significant challenges due to the limitations and side effects of current treatments. This study investigates the potential of natural products, specifically AH and NSO, as organic therapeutic agents for colitis. Molecular docking studies were conducted to identify the binding affinities and interaction mechanisms between the bioactive compounds in AH and NSO and proteins implicated in colitis, such as those involved in inflammation and oxidative stress pathways. An in vivo experiment was performed using an albino mouse model of colitis, with clinical symptoms, histopathological assessments, and biochemical analyses conducted to evaluate the therapeutic effects of the compounds both individually and in combination. Results from the molecular docking studies revealed promising binding interactions between fructose and Prostaglandin G/H synthase 2 (Ptgs2) and between fructose and cellular tumor antigen p53, with docking energy measured at −6.0 kcal/mol and −5.1 kcal/mol, respectively. Meanwhile, the presence of glucose molecule glucokinase chain A (−6.3 kcal/mol) and chain B (−5.8 kcal/mol) indicated potential efficacy in modulating inflammatory pathways. Experimental data demonstrated that treatment with AH and NSO significantly reduced inflammation, improved gut health, and ameliorated colitis symptoms. Histopathological evaluations confirmed reduced mucosal damage and immune cell infiltration, while biochemical analyses showed normalization of inflammatory markers and oxidative stress levels. This study provides compelling evidence for the potential of AH and NSO as natural, complementary treatments for colitis, suggesting their future role in integrative therapeutic strategies. However, further research into long-term safety, optimal dosing, and mechanisms of action is warranted to translate these findings into clinical applications.

This research characterized and explored the effect of cumin essential oil nanoemulsion (CEONE) on broiler growth performance, serum biochemistry, hematological parameters, and cecal microbial count. Day-old (n = 96) broilers (Ross 308) were randomly assigned to six treatments with five replicates of three broilers each. The dietary treatments consisted of negative control (only basal diet), positive control (basal diet + 200 µL of enrofloxacin), 25 µL (basal diet + 25 µL of CEONE), 50 µL (basal diet + 50 µL of CEONE), 75 µL (basal diet + 75 µL of CEONE), and 100 µL (basal diet + 100 µL of CEONE). The broiler’s body weight gain (BWG) after 42 days of treatment exhibited increased weight in the CEONE group (976.47 ± 11.82–1116.22 ± 29.04). The gain in weight was further evidenced by the beneficial microbe load (107 log) compared to the pathogenic strain. All the biochemical parameters were observed in the normal range, except for a higher level of HDL and a lower LDL value. This safety has been validated by pKCSM toxicity analysis showing a safe and highly tolerable dose of cuminaldehyde. In conclusion, this research observed the potential of CEONE as a multifunctional agent. It is a valuable candidate for further application in combating bacterial infections and enhancing animal health and growth.

Streptococcus agalactiae and Klebsiella pneumoniae are emerging as major milk-borne pathogens. Additionally, resistance to antibiotics of pathogens is of concern. Therefore, this study investigated the prevalence and drug resistance of S. agalactiae and K. pneumoniae in mastitis milk samples and assessed the antimicrobial potential of sodium alginate (G)-stabilized MgO nanoparticles (M) and antibiotics (tylosin [T] and ampicillin [A]) against both of these pathogens. A total of n = 200 milk samples from cattle were collected using purposive sampling, and standard microbiological approaches were adopted to isolate target bacteria. Parametric and non-parametric statistical tests were used to analyze the obtained data. Four preparations, GT (gel-stabilized tylosin), GA (gel-stabilized ampicillin), GTM (tylosin and MgO nanoparticles stabilized in gel), and GAM (ampicillin and MgO nanoparticles stabilized in gel), were evaluated against both bacteria through well diffusion and broth microdilution method. The analysis revealed that 45.24% (95/210) of the milk samples were positive for mastitis, of which 11.58% (11/95) were positive for S. agalactiae and 9.47% (9/95) were positive for K. pneumoniae. S. agalactiae had a significantly higher zone of inhibition (ZOI) than K. pneumoniae against penicillin, tetracycline, and amoxicillin, whereas the opposite was observed against imipenem and erythromycin. All gel (G)-based preparations showed an increase in the percentage of ZOI compared with antibiotics alone, with GTM presenting the highest of all, i.e., 59.09 and 56.25% ZOI compared with tylosin alone against S. agalactiae and K. pneumoniae, respectively. Similarly, in a broth microdilution assay, the lowest MIC was found for K. pneumoniae (9.766 ± 0.0 μg/mL) against GTM, followed by GT, GAM, and GA after incubation for 24 h. A similar response was noted for preparations against S. agalactiae but with a comparatively higher MIC. A significant reduction in MIC with respect to incubation time was found at 8 h and remained until at 20 h against both pathogens. The cytotoxicity of the MgO nanoparticles used in this study was significantly lower than that of the positive control. Overall, this study found that K. pneumoniae and S. agalactiae appeared higher in prevalence and antimicrobial resistance, and sodium alginate-based antibiotics and MgO nanoparticles were effective alternative approaches for tackling antimicrobial resistance.

The aim of the current study was to evaluate the toxic effect of silver nanoparticles (Ag-NPs) on biochemical biomarkers, immune responses, and the curative potential effects of vitamin C and E on grass carp. Fish (n = 420) with an average initial body weight of 8.045 ± 0.13 g were shifted to glass aquaria (36 x 18 x 18 inches, filled with 160-L tap water) in triplicates. Aquaria were randomly designated as A, B, C, D with alone Ag-NPs (Control (0), 0.25, 0.50, 0.75 mg/L) and E, F, G with Ag-NPs + Vit. C + Vit. E (0.25+0.25+0.25, 0.50+0.50+0.50, 0.75+0.75+0.75 mg/L). NPs particles were administrated viz, oral and intravenous routes for 7 days. The results indicated that both routes had non-significant effect, but levels of Ag-NPs had significant effect. Treatments C, D and G showed significant decrease in levels of RBC, HGB and HCT except for WBC and NEUT levels, which significantly increased. ALT, ALP, AST, urea, and creatinine showed significant increase in activity in the C, D, and G groups. CAT, SOD decreased significantly in all Ag-NPs alone groups, while significantly increased with vitamin E and C. LYZ, TP, ALB, GLB showed significant low activity in the B, C, and D groups while significantly high activity in the E, F, and G groups. Cortisol, glucose and triglycerides showed significant increase in the B, C, and D groups, while E, F, and G groups showed significant low levels of triglycerides, COR, and GLU. Cholesterol level was same across all treatment groups. In conclusion, vitamin E and C as powerful antioxidants protect the fish against Ag-NPs except high dose level of 0.75mg/L, while 0.25mg/L of Ag-NPs was presumably safe for C. idella.

The present study was conducted on Head Punjnad (HP) and Head Taunsa (HT) to evaluate the contamination of Pb, Cr, As, Hg, and Cd in water, soil, sediment, fish as a whole and fish organs. Fish, water, soil and sediment samples were collected from different sites of HT and HP on a monthly basis for 8 months. Heavy metals in water, soil, and sediment were determined by a polarized Zeeman atomic absorption spectrophotometer and in fish and fish organs by an atomic absorption spectrophotometer. هذه الدراسة الموسومة ( الإدمان الرقمي وعلاقته بالعزلة الاجتماعية لدى أطفال طيف التوحد من وجهة نظر أولياء امورهم ) تناول فيها الباحث موضوعا مهما ، هو معرفة الإدمان الرقمي لدى طفل طيف التوحد وما علاقته بالعزلة الاجتماعية لديهم ، إذ ترمي الدراسة الى معرفة "الإدمان الرقمي عند طفل طيف التوحد من وجهة نظر أولياء أمورهم ، وذلك عن طريق معرفة مدى إدمان اطفال طيف التوحد ،ومعرفة العلاقة ما بين الإدمان الرقمي وما بين العزلة الاجتماعية لدى أطفال طيف التوحد . وقدمت الدراسة فرضيات عدةِ وهي : يوجد ادمان رقمي عند اطفال طيف التوحد من وجهة نظر اولياء الامور؟ توجد عزلة اجتماعية عند اطفال طيف التوحد ؟ هناك علاقة ارتباطية بين الإدمان الرقمي والعزلة الاجتماعية لدى أطفال طيف التوحد؟ وتحقيقا لأهداف الدراسة أستخدم الباحث المنهج المسحي في الدراسة ، و اعتمد على أداة جمع المعلومات والبيانات وهي، الاستبانة التي تتعلق بمشكلة الدراسة، وقد توصلت الدراسة الى أهم النتائج الآتية: يتضح من نتائج الدراسة وجود إدمان رقمي لدى طفل طيف التوحد. ومن نتائج الفرضيات التي ظهرت بأنه توجد عزلة اجتماعية لدى طفل طيف التوحد. ومن نتائج الفرضيات، هي وجود علاقة ارتباطية موجبة دالة احصائيا ما بين الإدمان الرقمي والعزلة الاجتماعية لدى طفل طيف التوحد.

Abstract Phyto-synthesized nanoparticles (NPs) having reduced chemical toxicity have been focused globally and become essential component of nanotechnology recently. We prepared green phytochemically (ginger and garlic) reduced NiO-NPs to replace synthetic bactericidal and catalytic agent in textile industry. NPs were characterized using ultra-violet visible spectroscopy (UV-Vis), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), Fourier-transform infrared spectroscopy (FTIR), energy-dispersive X-ray spectroscopy (EDS), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). The synthesis of NPs was confirmed by XRD and UV-Vis having strong absorption at 350 nm with size ranged between 16–52 nm for ginger and 11–59 nm for garlic. Scanning and transmission electron microscopy confirmed pleomorphism with cubic- and more spherical-shaped NPs. Moreover, exact quantities of garlic and ginger extracts (1:3.6 ml) incorporated to synthesize NiO-NPs have been successfully confirmed by FTIR. Phytochemically reduced NPs by garlic presented enhanced bactericidal activity against multiple drug-resistant Staphylococcus aureus at increasing concentrations (0.5, 1.0 mg/50 μl) and also degraded methylene blue (MB) dye efficiently. Conclusively, green synthesized NiO-NPs are impending activists to resolve drug resistance as well as environment friendly catalytic agent that may be opted at industrial scale.

The intestinal tract microbiota influences many aspects of the dietary components on colon health and during enteric infections, thus, playing a pivotal role in the colon health. Therefore, the eugenol (EU) nano-emulsion effective concentration reported in our previous study against cancer cells should be explored for safety against beneficial microbes. We evaluated the sensitivity of Bifidobacterium breve and B. adolescentis against EU-loaded nano-emulsions at 0, 300, 600 and 900 µm, which were effective against colon and liver cancer cells. Both B. breve and B. adolescentis showed comparable growth ranges to the control group at 300 and 600 µm, as evident from the plate count experimental results. However, at 900 µm, a slight growth variation was revealed with respect to the control group. The real-time inhibition determination through flow cytometry showed B. breve viable, sublethal cells (99.49 and 0.51%) and B. adolescentis (95.59 and 0.15%) at 900 µm, suggesting slight inhibition even at the highest tested concentration. Flow cytometry proved to be a suitable quantitative approach that has revealed separate live, dead, and susceptible cells upon treatment with EU nano-emulsion against Escherichia coli. Similarly, in the case of B. breve and B. adolescentis, the cells showed only live cells that qualitatively suggest EU nano-emulsion safety. To judge the viability of these sublethal populations of B. breve and B. adolescentis, Fourier transforms infrared spectroscopy was carried out, revealing no peak shift for proteins, lipids, DNA and carbohydrates at 900 µm EU nano-emulsion compared to the control. On the other hand, EU-loaded nano-emulsions (900 µm)-treated E. coli showed a clear peak shift for a membrane protein, lipids, DNA and carbohydrates. This study provides insights to utilize plant phenols as safe medicines as well as dietary supplements.

The plant Caralluma edulis is traditionally used against diabetes and inflammatory conditions in Pakistan. This study was designed to provide scientific validation of the traditional use of Caralluma edulis. Phytochemicals were extracted from the plant by different solvents (distilled water, methanol, ethanol, and acetone) using the Soxhlet’s extraction method. Bioactive compounds were detected by gas chromatography–mass spectrometry (GC-MS). The in vitro anti-inflammatory activities (albumin denaturation, membrane stabilization, and proteinase inhibition) and antioxidant capacity (DPPH scavenging activity, FRAP reducing activity) of different extracts from Caralluma edulis were assessed. The antidiabetic potential of Caralluma edulis plant extracts was determined in acute and subacute diabetic rabbit models. Oxidative stress and enzymatic antioxidant status were also estimated in MDA, CAT, and SOD levels. Results showed that the methanol extract yielded the highest contents of phenolics, flavonoids, alkaloids, and terpenoids. The in vitro anti-inflammatory activity and antioxidant potential of the methanol extract were the highest among the tested solvents. The tested extracts did not show any remarkable antidiabetic activity in the acute diabetic model. However, all tested extracts demonstrated antidiabetic potential in the subacute diabetic model. No adverse effect was observed at the tested dose (200 mg/kg) of Caralluma edulis extracts in experimental animals. It is concluded that methanol is the key solvent for extracting bioactive compounds from Caralluma edulis. The plant can be used against inflammatory disorders and may prove a potential candidate for drug development. Long-term use of Caralluma edulis at the tested dose (200 mg/kg) showed antidiabetic properties in the animal model.

The binding capacity of Pb on modified rice bran-insoluble fibers RBIF-0.2, RBIF-1.25, and RBIF-2.0 (0.2%, 1.25%, and 2.0% H2SO4 with 1.25% KOH) having the variable specific surface area and cation-exchange capacity was evaluated. The Pb binding in terms of maximum binding capacity (BCmax) with RBIF increased with pH (2–7). However, the presence of sodium and calcium reduced the binding of Pb to RBIFs. Further, adsorption kinetics followed pseudo-second-order kinetic model when fitted to real Pb sorption data. However, sorption mechanism data (scanning electron microscopy images) were well fitted to Langmuir model instead of Freundlich and Dubinin–Radushkevich that suggests monomolecular adsorption of Pb to RBIF (

A cellulosic rice bran insoluble dietary fibre (RBIDF) with different structures and physicochemical properties is shown to inhibit the activity of pancreatic lipase (PL) at different levels. The adsorption rate and saturation level to PL were greatest for RBIDF-2.0 (modified with 2.0% H2SO4 + 1.25% KOH), which had the highest specific surface area and oil holding capacity. The conformational deformation of unbound PL in the supernatant of PL-RBIDF confirmed by circular dichroism (CD) suggested that not only bound but also unbound PL activities were altered. Free energy calculations using data from fluorescence spectroscopy revealed that binding of PL to fibre depends primarily on electrostatic interactions, and the binding process is spontaneous and exothermic. This study indicates that dietary intake of cellulosic fractions of rice bran may be useful in reducing the bioavailability of dietary fat by altering the conformation and activity of PL that might be useful to control obesity.

Wheat flour fortification can be a novel and effective food based approach to improve effective micronutrient deficiencies that affect millions of people worldwide especially in the developing countries. Wheat is an important cereal crop grown worldwide and its per capita consumption is high even in the developing countries. Being a most popular dietary food component, fortification of wheat flour with micronutrients like iron, vitamin A, folic acid, zinc, and iodine is expected to be the most effective strategy to overcome the related deficiencies and, if mandated, could be helpful in achieving the international health goals. However, on the other hand food fortification (Direct mixing) with micronutrients might cause unwanted sensory changes and interaction with food components resulting in a lower bioavailability. Microencapsulation may be helpful to prevent unwanted sensory changes and diminish micronutrients interactions with wheat flour components. The current review will focus on the technical issues related to the fortification (Direct mixing) of wheat flour and prospects of microencapsulation technology in fortification.
