5. Deuterated Clathrate Hydrates as a Novel Moderator Material for Very Cold Neutrons. Project Aperçu and First Results

Clathrate hydrates are water-based solids with large crystallographic unit cells that show promise as potential moderators for use in new, more intense sources of very cold neutrons (VCN), which would enhance neutron scattering techniques and increase sensitivity of particle physics experiments. These so-called inclusion compounds seem particularly suitable for this application due to the low-energy modes of guest molecules encaged in nano-voids formed by a crystalline network of hydrogen bonded water molecules. In this article we present first results of an ongoing, extensive experimental campaign with the aim of characterizing the scattering properties of deuterated clathrate hydrates relevant for moderator applications. Experiments include neutron diffraction (carried out at Institut Laue-Langevin (ILL) using the instrument D20), as well as measurements of the temperature-dependent dynamical structure factor S (q, ω) in absolute units (carried out on ILL’s Panther and IN5). These measurements will serve as a benchmark for development of new scattering kernels.

6. Hygrothermal conditions in ventilated attics with different air change rates and ceiling constructions

A recently Danish study reported that no vapour barrier is needed in ceilings, if the attic is well ventilated and the ceiling towards the dwelling is airtight. Based on that study, new investigations were initiated with focus on the hygrothermal behaviour in ventilated attics with different air change rates. A test house with three sets of four different ceiling constructions – all airtight – was used in this study. The ventilation rate was reduced in two of the sets with approx. 35 % and 50 %, respectively. Air change rates were measured with tracer gas. Furthermore, temperature and relative humidity was measured every hour. Measurements in similar ceilings with mineral wool or cellulose-based insulation material show that hygroscopic properties of the insulation have very limited effect on relative humidity. Furthermore, only at low ventilation rate the effect of a vapour barrier could be measured with minor impact. Based on the short-measured period the calculations of the risk of mould growth showed no risk. The results indicate that even when the ventilation is reduced by 50 %, the ventilated attic still performs well if the ceiling is highly airtight. However, the importance of vapour barriers becomes more important at lower air change rates.

7. Towards moisture safe ventilated cold attics – Monitored conditions in a full-scale test building

Existing building stock in Europe accounts for approx. 40% of the total energy consumption. Upgrading the thermal insulation of the existing buildings is an important measure to reduce heat losses through the building envelope. In some cases, increasing the thermal resistance of the construction may compromise the hygrothermal performance of the retrofitted construction. In particular, if vapour barrier is necessary for the good performance and it is practically difficult, if not even impossible, to install a well-sealed air- and vapour tight layer. To investigate the robustness of the hygrothermal performance of ventilated cold attics – with or without a vapour barrier – a monitoring campaign in a full-scale test building was set up. Also role of number of other parameters like moisture buffering capacity of the insulation material and thermal resistance was investigated. This paper presents part of this measuring campaign, which includes conditions both in the attic space and inside the insulation layer. The monitored data covers a period with two winters. The results show that it in temperate climate is practically indifferent for the hygrothermal performance of the monitored, well-ventilated attics with air-tight ceilings whether there is a vapour barrier or not and if the insulation material has moisture buffering capacity or not.

8. Measurements behind Internal Insulation on Solid Masonry Walls with Different Orientations and Degrees of Repair in a Danish Multi-story Building

Internal insulation is used in many heritage buildings to ensure a better indoor environment and reduce energy use. This article describes measurement results from installing an internal insulation system in a Danish multi-story building. The internal insulation system consists of a fully glued vapour tight insulated plasterboard. To reduce the risk for wood decay and mould growth at the wooden beam ends in the floor construction, a 100-200 mm uninsulated gap was left above and below the floor construction. Measurements include the effect of orientation, degree of façade repair and influence of indoor moisture content. Results show in general acceptable hygrothermal conditions behind the insulation system. However, a gradual increase in relative humidity at sun-exposed walls was registered due to ‘summer condensation’. The degree of surface repair also showed an influence on relative humidity levels during this period. At the uninsulated gap above the floor, a higher moisture content indoor resulted in a higher relative humidity, though without exceeding the threshold for mould growth. However, a large temperature range was registered. The influence of the uninsulated gap on reducing the risk of wood decay and mould growth at the beam ends could therefore not unambiguously be confirmed.

9. Investigation of the Mould Resistance Design (MRD) model for mould growth in attics

Based on measurements of mould in 17 attics in the north of Sweden, 3 were chosen as a tentative validation of the MRD-model (Mould Resistance Design) for long periods of time (1961-2012). Comparing the measurements and the results from the simulations showed great differences regarding the mould growth depending on the simplifications used in the model. How the leakage was simulated and where the moisture from the leakage was placed in the construction were the most important factors. The study could not validate the MRD model as such, but it was clear that the simulations with relatively small adjustments could give realistic data

10. Air change rate in ventilated attics – reality and input for simulations

To remove moisture by ventilating constructions is a well-known strategy, used successfully in numerous facades and roofs. However, simulating ventilation is often difficult as convection in this case is in another dimension than what is usually in focus in 1D hygrothermal simulations. There are strategies for simulating ventilation in 1D programs assuming a fixed air change rate (ACH). Unfortunately, ACH in roofs highly depend on wind direction and speed, hence a constant rate is a gross simplification. The article describes a comparative study between simulations with a 1D hygrothermal simulation program (WUFI) and simulations with BSim, a program in which the indoor climate can be simulated in different zones, in this case the ventilated attic. Focus is the effect of ACH in the attic on temperature and relative humidity. Finally, measurements of ACH in attics of ten single-family houses are presented. These measurements show that average ACH measured over more than two weeks differ very much from house to house although the ventilation openings in all houses seemed to be in accordance with current guidelines. Even measurements of the same attic differ considerably from time to time. The paper discusses what implications this should have on simulations.

11. Global report on preterm birth and stillbirth (3 of 7): evidence for effectiveness of interventions

Abstract Introduction Interventions directed toward mothers before and during pregnancy and childbirth may help reduce preterm births and stillbirths. Survival of preterm newborns may also be improved with interventions given during these times or soon after birth. This comprehensive review assesses existing interventions for low- and middle-income countries (LMICs). Methods Approximately 2,000 intervention studies were systematically evaluated through December 31, 2008. They addressed preterm birth or low birth weight; stillbirth or perinatal mortality; and management of preterm newborns. Out of 82 identified interventions, 49 were relevant to LMICs and had reasonable amounts of evidence, and therefore selected for in-depth reviews. Each was classified and assessed by the quality of available evidence and its potential to treat or prevent preterm birth and stillbirth. Impacts on other maternal, fetal, newborn or child health outcomes were also considered. Assessments were based on an adaptation of the Grades of Recommendation Assessment, Development and Evaluation criteria. Results Most interventions require additional research to improve the quality of evidence. Others had little evidence of benefit and should be discontinued. The following are supported by moderate- to high-quality evidence and strongly recommended for LMICs: • Two interventions prevent preterm births—smoking cessation and progesterone • Eight interventions prevent stillbirths—balanced protein energy supplementation, screening and treatment of syphilis, intermittant presumptive treatment for malaria during pregnancy, insecticide-treated mosquito nets, birth preparedness, emergency obstetric care, cesarean section for breech presentation, and elective induction for post-term delivery • Eleven interventions improve survival of preterm newborns—prophylactic steroids in preterm labor, antibiotics for PROM, vitamin K supplementation at delivery, case management of neonatal sepsis and pneumonia, delayed cord clamping, room air (vs. 100% oxygen) for resuscitation, hospital-based kangaroo mother care, early breastfeeding, thermal care, and surfactant therapy and application of continued distending pressure to the lungs for respiratory distress syndrome Conclusion The research paradigm for discovery science and intervention development must be balanced to address prevention as well as improve morbidity and mortality in all settings. This review also reveals significant gaps in current knowledge of interventions spanning the continuum of maternal and fetal outcomes, and the critical need to generate further high-quality evidence for promising interventions.

It allows therefore establishing structure-property relationships, which need to be understood in order to develop new and better performing materials.

13. Identification of 3 novel VHL germ-line mutations in Danish VHL patients

Abstract Background von Hippel-Lindau (VHL) disease is a hereditary cancer syndrome in which the patients develop retinal and central nervous system hemangioblastomas, pheochromocytomas and clear-cell renal tumors. The autosomal dominant disease is caused by mutations in the VHL gene. Methods VHL mutational analysis was carried out by sequencing of the coding sequence and by multiplex ligation-dependent probe amplification analysis. The functional consequence of the variants was investigated using in silico prediction tools. Results A total of 289 probands suspected of having VHL syndrome have been screened for mutations in the VHL gene. Twenty-six different VHL mutations were identified in 36 families including one in-frame duplication, two frame-shift mutations, four nonsense mutations, twelve missense mutations, three intronic mutations and four large genomic rearrangements. Three of these mutations (c.319 C > T, c.342_343dupGGT and c.520_521dupAA) were novel. Conclusions In this study we report the VHL germ-line mutations found in Danish families. We found three novel VHL mutations where two were classified as pathogenic and the latter was classified as a variant of unknown significance. Together, our findings contribute to the interpretation of the potential pathogenicity of VHL germ-line mutations.

Abstract Background The progression of implantation and placentation in ruminants is complex and is regulated by interplay between sex steroids and local signaling molecules, many of which have immune function. Chemokines and their receptors are pivotal factors in implantation and vascularization of the placenta. Based on known critical roles for chemokine receptor 4 (CXCR4) during early pregnancy in other species, we hypothesized that CXCR4 and its ligand CXCL12 would increase in the endometrium and conceptus in response to implantation in ewes. The objectives of the current study were to determine if CXCL12 and CXCR4 were upregulated in: endometrium from pregnant compared to non-pregnant ewes and in, conceptuses, cotyledons, caruncles and intercaruncular tissue. Methods Tissues were collected from sheep on Days 12, 13, 14, and 15 of either the estrous cycle or pregnancy and from pregnant ewes on Days 35 and 50. Blood samples from jugular and uterine vein were also collected on all days. Conceptuses were collected from mature ewes on Days 13, 15, 16, 17, 21 and 30 of gestation. Real time PCR was used to determine relative mRNA concentrations for CXCL12 and CXCR4 and Western blot analysis was employed to confirm protein concentration. Results Differences described are P < 0.05. In the endometrium, CXCR4 mRNA and protein was greater on Day 15 of pregnancy compared to the estrous cycle. CXCL12 and CXCR4 mRNA in conceptuses was greater on Days 21 and 30 compared to earlier days. CXCL12 mRNA was greater in cotyledons on Day 35 compared to Day 50. On Day 35 of gestation, CXCR4 was greater compared to Day 50 in caruncle and intercaruncular tissue. White blood cells obtained from jugular and uterine vein collection had the greatest mRNA concentration of CXCL12 on Day 35 of pregnancy. Conclusions A comprehensive analysis of CXCL12 and CXCR4 expression in fetal and maternal tissues during early pregnancy is reported with noteworthy differences occurring during implantation and placentation in sheep. We interpreted these data to mean that the CXCL12/CXCR4 pathway is activated during implantation and placentation in sheep and is likely playing a role in the communication between trophoblast cells and the maternal endometrium.

15. Micro-scaled high-throughput digestion of plant tissue samples for multi-elemental analysis

Abstract Background Quantitative multi-elemental analysis by inductively coupled plasma (ICP) spectrometry depends on a complete digestion of solid samples. However, fast and thorough sample digestion is a challenging analytical task which constitutes a bottleneck in modern multi-elemental analysis. Additional obstacles may be that sample quantities are limited and elemental concentrations low. In such cases, digestion in small volumes with minimum dilution and contamination is required in order to obtain high accuracy data. Results We have developed a micro-scaled microwave digestion procedure and optimized it for accurate elemental profiling of plant materials (1-20 mg dry weight). A commercially available 64-position rotor with 5 ml disposable glass vials, originally designed for microwave-based parallel organic synthesis, was used as a platform for the digestion. The novel micro-scaled method was successfully validated by the use of various certified reference materials (CRM) with matrices rich in starch, lipid or protein. When the micro-scaled digestion procedure was applied on single rice grains or small batches of Arabidopsis seeds (1 mg, corresponding to approximately 50 seeds), the obtained elemental profiles closely matched those obtained by conventional analysis using digestion in large volume vessels. Accumulated elemental contents derived from separate analyses of rice grain fractions (aleurone, embryo and endosperm) closely matched the total content obtained by analysis of the whole rice grain. Conclusion A high-throughput micro-scaled method has been developed which enables digestion of small quantities of plant samples for subsequent elemental profiling by ICP-spectrometry. The method constitutes a valuable tool for screening of mutants and transformants. In addition, the method facilitates studies of the distribution of essential trace elements between and within plant organs which is relevant for, e.g., breeding programmes aiming at improvement of the micronutrient density in edible plant parts. Compared to existing vial-in-vial systems, the new method developed here represents a significant methodological advancement in terms of higher capacity, reduced labour consumption, lower material costs, less contamination and, as a consequence, improved analytical accuracy following micro-scaled digestion of plant samples.

16. Novel de novo BRCA2 mutation in a patient with a family history of breast cancer

Abstract Background BRCA2 germ-line mutations predispose to breast and ovarian cancer. Mutations are widespread and unclassified splice variants are frequently encountered. We describe the parental origin and functional characterization of a novel de novo BRCA2 splice site mutation found in a patient exhibiting a ductal carcinoma at the age of 40. Methods Variations were identified by denaturing high performance liquid chromatography (dHPLC) and sequencing of the BRCA1 and BRCA2 genes. The effect of the mutation on splicing was examined by exon trapping in COS-7 cells and by RT-PCR on RNA isolated from whole blood. The paternity was determined by single nucleotide polymorphism (SNP) microarray analysis. Parental origin of the de novo mutation was determined by establishing mutation-SNP haplotypes by variant specific PCR, while de novo and mosaic status was investigated by sequencing of DNA from leucocytes and carcinoma tissue. Results A novel BRCA2 variant in the splice donor site of exon 21 (nucleotide 8982+1 G→A/c.8754+1 G→A) was identified. Exon trapping showed that the mutation activates a cryptic splice site 46 base pairs 3' of exon 21, resulting in the inclusion of a premature stop codon and synthesis of a truncated BRCA2 protein. The aberrant splicing was verified by RT-PCR analysis on RNA isolated from whole blood of the affected patient. The mutation was not found in any of the patient's parents or in the mother's carcinoma, showing it is a de novo mutation. Variant specific PCR indicates that the mutation arose in the male germ-line. Conclusion We conclude that the novel BRCA2 splice variant is a de novo mutation introduced in the male spermatozoa that can be classified as a disease causing mutation.

17. The estrogen hypothesis of Schizophrenia implicates glucose metabolism: Association study in three independent samples

Abstract Background Schizophrenia is a highly heritable complex psychiatric disorder with an underlying pathophysiology that is still not well understood. Metaanalyses of schizophrenia linkage studies indicate numerous but rather large disease-associated genomic regions, whereas accumulating gene- and protein expression studies have indicated an equally large set of candidate genes that only partially overlap linkage genes. A thorough assessment, beyond the resolution of current GWA studies, of the disease risk conferred by the numerous schizophrenia candidate genes is a daunting and presently not feasible task. We undertook these challenges by using an established clinical paradigm, the estrogen hypothesis of schizophrenia, as the criterion to select candidates among the numerous genes experimentally implicated in schizophrenia. Bioinformatic tools were used to build and priorities the signaling networks implicated by the candidate genes resulting from the estrogen selection. We identified ten candidate genes using this approach that are all active in glucose metabolism and particularly in the glycolysis. Thus, we tested the hypothesis that variants of the glycolytic genes are associated with schizophrenia or at least with gender-associated aspects of the illness. Results We genotyped 185 SNPs in three independent case-control samples of Scandinavian origin (a total of 765 patients and 1274 control subjects). Variants of the mitogen-activated protein kinase 14 gene (MAPK14) and the phosphoenolpyruvate carboxykinase 1 (PCK1) and fructose-1,6-biphosphatase (FBP1) were nominal significantly associated with schizophrenia, and several haplotypes within enolase 2 gene (ENO2) consist of the same SNP allele having elevated risk of schizophrenia. Importantly, we find no evidence of stratification due to nationality or gender. Conclusion Several gene variants in the Glycolysis were associated with schizophrenia in three independent samples. However, the findings are weak and not resistant to correction for multiple testing, which may indicate that they are either spurious or may relate to a particular subtype or aspect of the illness.

18. Diagnostic stability among chronic patients with functional psychoses: an epidemiological and clinical study

Abstract Background Diagnostic stability and illness course of chronic non-organic psychoses are complex phenomena and only few risk factors or predictors are known that can be used reliably. This study investigates the diagnostic stability during the entire course of illness in patients with non-organic psychoses and attempts to identify non-psychopathological risk factors or predictors. Method 100 patients with functional psychosis were initially characterised using the Operational Criteria Checklist for Psychotic Illness and Affective Illness (OPCRIT), medical records and health registers. To study the stability of diagnoses (i.e. shifts per time), we used registry data to define four measures of diagnostic variation that were subsequently examined in relation to four possible measures of time (i.e. observation periods or hospitalisation events). Afterwards, we identified putative co-variables and predictors of the best measures of diagnostic stability. Results All four measures of diagnostic variation are very strongly associated with numbers-of-hospitalisations and less so with duration-of-illness, duration-of-hospitalisation and with year-of-first-admission. The four measures of diagnostic variation corrected for numbers-of-hospitalisations were therefore used to study the diagnostic stability. Conventional predictors of illness course – e.g. age-of-onset and premorbid-functioning – are not significantly associated with stability. Only somatic-comorbidity is significantly associated with two measures of stability, while family-history-of-psychiatric-illness and global-assessment-of-functioning (GAF) scale score show a trend. However, the traditional variables age-of-first-admission, civil-status, first-diagnosis-being-schizophrenia and somatic-comorbidity are able to explain two-fifth of the variation in numbers-of-hospitalisations. Conclusion Diagnostic stability is closely linked with the contact between patient and the healthcare system. This could very likely be due to fluctuation of disease manifestation over time or presence of co-morbid psychiatric illness in combination with rigid diagnostic criteria that are unable to capture the multiple psychopathologies of the functional psychoses that results in differential diagnoses and therefore diagnostic instability. Not surprisingly, somatic-comorbidity was found to be a predictor of diagnostic variation thereby being a non-psychiatric confounder.

Mice lacking Ubp43 (null) form increased conjugated Isg15 in response to interferon. The Isg15 system has not been examined in chorioallantoic placenta (CP) or mesometrial (MM) components of implantation sites beyond 9.5 days post coitum (dpc). It was hypothesized that deletion of Ubp43 would cause disregulation of Isg15 in implantation sites, and that this would affect pregnancy rates. Methods Heterozygous (het) Ubp43 mice were mated and MM and CP implantation sites were collected on 12.5 and 17.5 days post-coitum (dpc). Results Free and conjugated Isg15 were greater on 12.5 versus 17.5 dpc in MM. Free and conjugated Isg15 were also present in CP, but did not differ due to genotype on 12.5 dpc. However, null CP had greater free and conjugated Isg15 when compared to het/wt on 17.5 dpc. Null progeny died in utero with fetal genotype ratios (wt:het:null) of 2:5:1 on 12.5 and 2:2:1 on 17.5 dpc. Implantation sites were disrupted within the junctional zone and spongiotrophoblast, contained less vasculature based on lectin B4 staining and contained greater Isg15 mRNA and VEGF protein in Ubp43 null when compared to wt placenta. Conclusion It is concluded that Isg15 and its conjugates are present in implantation sites during mid to late gestation and that deletion of Ubp43 causes an increase in free and conjugated Isg15 at the feto-maternal interface. Also, under mixed genetic background, deletion of Ubp43 results in fetal death.
