Green mold caused by Penicillium digitatum is the leading postharvest disease in citrus fruit. While chemical fungicides are commonly used for management, yeast-based biological control strategy offers a promising and eco-friendly alternative. In this study, yeast strains were isolated from citrus orchards and the strain with the best biocontrol efficacy was screened to investigate its mechanisms for controlling postharvest green mold in citrus fruit. In total, 30 yeast strains were isolated from fruits, leaves and soil of two citrus orchards. Subsequently, 8 yeast strains tested in vivo effectively inhibited green mold in citrus fruit, with Kurtzmaniella quercitrusa BS-AY-S1 achieving the best biocontrol efficacy by reducing the disease incidence (DI) by 58.33 % and lesion diameter (LD) by 73.36 % compared to the control on the 6th day. BS-AY-S1 exhibited excellent colonization ability, laying the foundation for its biocontrol efficacy. Furthermore, the antifungal properties of BS-AY-S1 metabolites were evaluated. The fermentation supernatant of BS-AY-S1 effectively inhibited the growth of P. digitatum and controlled green mold in citrus fruit. Additionally, the volatile organic compounds (VOCs) from BS-AY-S1 markedly reduced the spore germination, spore production and mycelial growth of P. digitatum, and slowing down the progression of green mold in citrus fruit. In total, 20 VOCs, including alcohols, aldehydes, esters, ethers and ketones produced by BS-AY-S1, were identified by gas chromatography-ion mobility spectrometry (GC-IMS). In conclusion, BS-AY-S1 demonstrated potential for controlling postharvest green mold in citrus fruit, with its mechanisms of action including the ability to colonize citrus fruit wounds and produce antifungal metabolites and VOCs.

Highlights The main effects of deformation of flexible catalytic materials on the catalytic hydrogen evolution reaction performance are discussed, and a series of novel strategies to design highly active catalysts based on the mechanical flexibility of low-dimensional nanomaterials are summarized in detail. This review provides a strategic choice for the rational design of low-cost and high-performance industrialized electrocatalysts.

The rapid development of science and technology, as well as the trend toward intelligence, has resulted in a surge in the demand for excellent energy storage devices for smart devices. At present, the most widely used lithium ion batteries have limited further development because of their high cost and various disadvantages of electrolytes (flammable and toxic). In spite of the low costs and safety of aqueous zinc-ion batteries, the high radius of the zinc ions and strong electrostatic interactions make cathode materials without excellent performance difficult to find. Various oxidation states, structural diversity, excellent multiplicity performance and high capacity have attracted researchers to vanadium-based oxides in recent years. However, vanadium-based oxides have disadvantages such as poor structural stability and low electrical conductivity, so this paper summarizes the progress of research on vanadium oxide cathode materials such as V2O5, VO2, V2O3, etc., and provides an outlook on future development directions.

Fresh-cut vegetables are widely consumed, but there is no food preservative available to selectively inhibit vancomycin-resistant E. faecalis, which is a serious health menace in fresh-cut vegetables. To develop a promising food biopreservative, a bacteriocin, paracin wx7, was synthesized, showing selective inhibition against E. faecalis with MIC values of 4–8 μM. It showed instant bactericidal mode within 1 h at high concentrations with concomitant cell lysis against vancomycin-resistant E. faecalis. Its lethal effect was visualized in a dose-dependent manner by PI/SYTO9 staining observation. The results of an in vivo control experiment carried out on E. faecalis in fresh-cut lettuce showed that 99.97% of vancomycin-resistant E. faecalis were dead after 64 μM paracin wx7 treatment for 7 days without influencing total bacteria. Further, the action mechanism of paracin wx7 was investigated. Confocal microscopy showed that paracin wx7 was located both on the cell envelope and in cytoplasm. For the cell envelope, the studies of membrane permeability using SYTOX Green dyeing and DNA leakage revealed that paracin wx7 damaged the membrane integrity of E. faecalis. Simultaneously, it exhibited membrane depolarization after analysis using DiSC3(5). Damage to the cell envelope resulted in cell deformation observed by scanning electron microscopy. On entering the cytoplasm, the paracin wx7 induced the production of endogenous reactive oxygen species.

During storage and transportation after harvest, the jujube fruit is susceptible to black spot rot, which is caused by Alternaria alternata. The present study aimed to evaluate the effectiveness of the yeast Meyerozyma caribbica in controlling A. alternata in postharvest jujube fruits, and to explore the biofilm formation mechanism. The results showed that M. caribbica treatment significantly reduced the A. alternata decay in jujube fruits. M. caribbica could rapidly colonize jujube fruit wounds, adhering tightly to hyphae of A. alternata, and accompanied by the production of extracellular secretions. In in vitro experiments, we identified that M. caribbica adhered to polystyrene plates, indicating a strong biofilm-forming ability. Furthermore, we demonstrated that M. caribbica can secrete phenylethanol, a quorum sensing molecule which can affect biofilm development. Phenylalanine (a precursor substance for phenylethanol synthesis) enhanced the secretion of phenylethanol and promoted the formation of M. caribbica biofilms. Meanwhile, phenylalanine enhanced the biological control performance of M. caribbica against jujube black spot rot. Our study provided new insights that enhance the biological control performance of antagonistic yeast.

In this study, the transcriptional profiling of Penicillium digitatum after C12O3TR treatment was analyzed by RNA-Seq technology. A total of 2562 and 667 genes in P. digitatum were differentially expressed after 2 and 12 h treatment, respectively. These genes were respectively mapped to 91 and 79 KEGG pathways. The expression patterns of differentially expressed genes (DEGs) at 2 and 12 h were similar, mainly were the metabolic processes in cell wall, cell membrane, genetic information and energy. Particularly, the main metabolic process which was affected by C12O3TR stress for 2 and 12 h was cell integrity, including cell wall and cell membrane. The changes of chitin in cell wall was observed by Calcofluor White (CFW) staining assay. The weaker blue fluorescence in the cell wall septa, the decrease of β-1, 3-glucan synthase activity and the increase of chitinase and AKP activity showed that C12O3TR could damage the cell wall integrity. In conclusion, these results suggested that C12O3TR could inhibit the growth of P. digitatum through various mechanisms at transcriptional level, and could influence the cell wall permeability and integrity.

Utilizing antagonistic yeasts is a promising approach for managing postharvest decay of fruits. However, it is well established that various severe stresses encountered in the environment and production process cause the intracellular reactive oxygen species (ROS) accumulation in yeast cells, resulting in cell damage and loss of vitality. Here, proline has been shown to function as a cell protectant and inducer of biofilm formation able to increase the oxidative stress tolerance and the biocontrol ability of the antagonistic yeast Metschnikowia citriensis. Addition of proline to M. citriensis cells induced a significant rise in superoxide dismutase (SOD) and catalase (CAT) activity in the early and late stages of oxidative stress, respectively, and increased the maroon pigment production that directly reduced intracellular iron content and indirectly diminished intracellular ROS levels and thus inhibited ROS- and iron-induced apoptosis. Treating cells with iron chelator tropolone yielded similar results. Pigment production induced by proline also enhanced the capability of biofilm formation of M. citriensis. These results suggested an important role for pigment of M. citriensis in response to oxidative stress. The abilities of proline to scavenge intracellular ROS and inhibit apoptosis, increase pigment production, and promote biofilm formation contribute to the improvements in oxidative stress tolerance and biocontrol efficacy of M. citriensis.

Loss and waste of postharvest vegetables are the main challenges facing the world’s vegetable supply. In this study, an innovative method of value-added transformation was provided: production of bacteriocin from vegetable waste, and then its application to preservation of vegetables. Antibacterial activity to soft rot pathogen Pectobacterium cartovorum (Pcb BZA12) indicated that tomato performed best in the nutrition supply for bacteriocin production among 12 tested vegetables. Moreover, the antibacterial activity was from Lactobacillus paracasei WX322, not components of vegetables. During a fermentation period of 10 days in tomato juice, L. paracasei WX322 grew well and antibacterial activity reached the maximum on the tenth day. Thermostability and proteinase sensitivity of the bacteriocin from tomato juice were the same with that from Man-Rogosa-Sharpe broth. Scanning electron microscope images indicated that the bacteriocin from tomato juice caused great damage to Pcb BZA12. At the same time, the bacteriocin from tomato juice significantly reduced the rotten rate of Chinese cabbage from 100% ± 0% to 20% ± 8.16% on the third day during storage. The rotten rate decrease of cucumber, tomato, and green bean was 100% ± 0% to 0% ± 0%, 70% ± 14.14% to 13.33% ± 9.43%, and 76.67% ± 4.71% to 26.67% ± 4.71%, respectively. Bacteriocin treatment did not reduce the rotten rate of balsam pear, but alleviated its symptoms.

Backgroud: The growing emergence of antibiotic-resistant pathogens including the most dangerous superbugs requires quick discovery of novel antibiotics/biopreservatives for human health and food safety. Bacteriocins, a subgroup of antimicrobial peptides, have been considered as promising alternatives to antibiotics. Abundant novel bacteriocins are stored in genome sequences of lactic acid bacteria. However, discovery of novel bacteriocins still mainly relies on dubious traditional purification with low efficiency. Moreover, sequence alignment is invalid for novel bacteriocins which have no homology to known bacteriocins in databases. Therefore, an efficient, simple, universal, and time-saving method was needed to discover novel bacteriocins.Methods and Results: Crude bacteriocins from both cell-related and culture supernatant of Lactobacillus crustorum MN047 fermentation were applied to LC-MS/MS for peptidome assay, by which 131 extracellular peptides or proteins were identified in the complete genome sequence of L. crustorum MN047. Further, the genes of suspected bacteriocins were verified by expressed in Escherichia coli BL21 (DE3) pLysS. Thereafter, eight novel bacteriocins and two nonribosomal antimicrobial peptides were identified to be broad-spectrum activity against both Gram-positive and Gram-negative bacteria, including some multidrug-resistant strains. Among them, BM1556 located within predicted bacteriocin gene cluster. The most active bacteriocin BM1122 had low MIC values of 13.7 mg/L against both Staphylococcus aureus ATCC29213 and E. coli ATCC25922. The BM1122 had bactericidal action mode by biofilm-destruction, pore-formation, and membrane permeability change.Conclusions: The combination of complete genome and peptidome is a valid approach for quick discovery of novel bacteriocins without/with-low homology to known ones. This method will contribute to deep exploitation of novel bacteriocins in genome of bacteria submitted to GenBank.

The carbon-supported bimetallic Au-Pd catalyst with core-shell structure is prepared by successive reduction method. The core-shell structure, surface morphology, and electrochemical performances of the catalysts are characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM), ultraviolet-visible absorption spectrometry, linear sweep voltammetry, and chronopotentiometry. The results show that the Au-Pd/C catalyst with core-shell structure exhibits much higher catalytic activity for the direct oxidation of NaBH4 than pure Au/C catalyst. A direct borohydride-hydrogen peroxide fuel cell, in which the Au-Pd/C with core-shell structure is used as the anode catalyst and the Au/C as the cathode catalyst, shows as high as 68.215 mW cm−2 power density.
