Abstract Collective spin excitations in magnetically ordered crystals, called magnons or spin waves, can serve as carriers in novel spintronic devices with ultralow energy consumption. The generation of well-detectable spin flows requires long lifetimes of high-frequency magnons. In general, the lifetime of spin waves in a metal is substantially reduced due to a strong coupling of magnons to the Stoner continuum. This makes metals unattractive for use as components for magnonic devices. Here, we present the metallic antiferromagnet CeCo2P2, which exhibits long-living magnons even in the terahertz (THz) regime. For CeCo2P2, our first-principle calculations predict a suppression of low-energy spin-flip Stoner excitations, which is verified by resonant inelastic X-ray scattering measurements. By comparison to the isostructural compound LaCo2P2, we show how small structural changes can dramatically alter the electronic structure around the Fermi level leading to the classical picture of the strongly damped magnons intrinsic to metallic systems. Our results not only demonstrate that long-lived magnons in the THz regime can exist in bulk metallic systems, but they also open a path for an efficient search for metallic magnetic systems in which undamped THz magnons can be excited.

Concurrent genital-anal human papillomavirus (HPV) infections may impose an increased anal cancer risk in women with HPV-related genital lesions. High viral load may facilitate genital-anal HPV concurrence. Genital and anal HPV is reduced by a bivalent HPV16/18 vaccine, yet the effect on concurrent genital-anal HPV remains unclear.This study analyzed viral load in concurrent genital-anal HPV infections, relative to genital-only and anal-only HPV infections and the impact of vaccination in young women. We included 1074 women, who provided both genital and anal swabs. HPV detection and genotyping was performed using the SPF10-DEIA-LiPA25. Introduction Sleep disturbances are common following traumatic brain injury (TBI) worsening morbidity and other neuropsychiatric symptoms. Post-TBI alterations in sleep architecture require further study. Objectives (1) To evaluate polysomnographic measures of sleep architecture in participants with history of TBI compared to controls and as meta-analyses of pooled means. (2) To evaluate effects of timing and severity of TBI on polysomnographic outcomes. Methods PRISMA compliant systematic review was conducted of MEDLINE, PsycINFO, EMBASE and Scopus. Inclusion criteria: 1) reporting polysomnography in the context of TBI and 2) operationalizing TBI using clear/formalized criteria. Data were pooled in random-effects meta-analyses with outcomes expressed as mean differences (MD). Results In participants with TBI, sleep was comprised of 19.39% REM sleep, 8.13% N1, 51.18% N2, and 17.53% N3, as determined by meta-analyses of single means. Introduction Neuroimaging has been a highly utilized technique for studying traumatic brain injury (TBI) and post-traumatic stress disorder (PTSD) independently of one another, however, neuroimaging has increasingly been identified as a useful tool in better understanding TBI-related psychiatric conditions, such as PTSD. Objectives To complete a systematic review of the literature examining neuroimaging findings in TBI-related PTSD and to highlight the current literature’s limitations in order to strengthen future research. Methods A PRISMA compliant literature search was conducted in PubMed (MEDLINE), PsychINFO, EMBASE, and Scopus databases prior to May of 2019. The initial database query yielded 4388 unique articles, which were narrowed down based on specified inclusion criteria (e.g., clear TBI definition, clinician-diagnosed PTSD, statistically analyzed relationship between neuroimaging and PTSD, quantified time interval between TBI and neuroimaging). Results A final cohort of 10 articles met inclusion criteria, comprising the findings of 482 participants with TBI. Key neuroanatomical findings among the included articles suggest that PTSD is associated with significant changes in whole-brain networks of resting state connectivity and disruptions in bilateral frontal and temporal white matter tracts, fronto-limbic pathways, the internal capsule, and the uncinate fasciculus (Figure 1). Conclusions Additional inquiry with attention to specified imaging timing post-injury, consistent TBI definitions, clinician-diagnosed TBI and PTSD, and control groups is crucial to extrapolating discrepancies between primary and TBI-related PTSD. Introduction Traumatic brain injury (TBI) may alter dementia progression, although co-occurring neuropsychiatric symptoms (NPS) have received less attention. The mild behavioral impairment (MBI) construct relates NPS to underlying neural circuit disruptions, representing an important area of inquiry regarding TBI and dementia. Objectives (1) to examine the influence of prior TBI history (preceding study enrollment) on MBI incidence in all-cause dementia (prior to dementia diagnosis, i.e. MBI’s original definition) and (2) to utilize MBI domains as a construct for examining the influence of TBI on related NPS across the course of dementia onset and progression. Methods Using National Alzheimer’s Coordinating Center data, individuals progressing from normal cognition to all-cause dementia over 7.6±3.0 years were studied to estimate MBI incidence and symptom domains in 124 participants with prior TBI history compared to 822 without. Results Moderate-severe TBI was associated with the social inappropriateness MBI domain (ORadj.=4.034; p=0.024) prior to dementia onset, and the abnormal perception/thought content domain looking across dementia progression (HRadj.=3.703, p=0.005). TBI (all severities) was associated with the decreased motivation domain looking throughout dementia progression (HRadj.=1.546, p=0.014). Conclusions TBI history is associated with particular MBI domains prior to onset and throughout progression of dementia. Understanding TBI’s impact on inter-related NPS may help elucidate underlying neuropathology. Disclosure No significant relationships.

Over the Plio-Pleistocene interval a strong linkage exists between northern African climate changes and the supply of dust over the surrounding oceans and continental runoff towards the Mediterranean Sea. Both these signatures in the sedimentary record are determined by orbital cycles influencing glacial variability on the one hand and northern African monsoon intensity on the other hand. In this paper, we use the intermediate-complexity model CLIMBER-2 to simulate African climate during the Plio-Pleistocene between 3.2 and 2.3 million years ago (Ma) and compare our simulations with existing and new climate reconstructions. The CLIMBER-2 model is externally forced with atmospheric CO2 concentrations, ice sheet topography, and orbital variations, all of which strongly influence climate during the Pliocene and Pleistocene. Our simulations indicate that the records of northern Africa climate oscillate in phase with climatic precession. For the Earth's obliquity cycle, the time lag between the 41 000-year component in insolation forcing and the climatic response increased after inception of Northern Hemisphere (NH) glaciation around 2.8 Ma. To test the outcome of our simulations, we have put emphasis on the comparison between the simulated runoff of grid boxes encompassing the Sahara desert and the Sahel region and the sedimentary records of marine sediment cores from ODP Site 659 (Atlantic Ocean) and ODP Site 967 (Mediterranean). In this study we will show for the first time an extended Ti∕Al record of Site 967 down to 3.2 Ma. This record strongly correlates with runoff in the Sahara and Sahel regions, whereas correlation with the dust record of Site 659 is moderate and slightly improves after NH ice sheet inception. We investigated the transient variability of the individual and combined contributions of the Sahel and Sahara regions and found significant transient behaviour overlapping the inception of NH ice sheets (2.8 Ma) and the Plio-Pleistocene transition (2.6 Ma). Prior to 2.8 Ma, a larger contribution from the Sahara region is required to explain the variability of Mediterranean dust input. After this transition, we found that a more equal contribution of the two regions is required, representing an increased influence of Sahel runoff and wet periods.

Abstract Background Around 20% of patients undergoing spinal fusion surgery have persistent back or leg pain despite surgery. Pain catastrophizing is the strongest psychological predictor for chronic postsurgical pain. Psychological variables are modifiable and could be target for intervention. However, randomized controlled trials evaluating the effectiveness of psychological interventions to reduce chronic pain and disability after spinal fusion in a population of patients with high preoperative pain catastrophizing scores are missing. The aim of our study is to examine whether an intervention targeting pain catastrophizing mitigates the risk of chronic postsurgical pain and disability. Our primary hypothesis is that targeted perioperative cognitive behavioral therapy decreases the risk of chronic postsurgical pain and disability after spinal fusion surgery in high catastrophizing patients. Methods We will perform a two-center prospective, single-blind, randomized, controlled study comparing lumbar spinal fusion surgery outcome between 2 cohorts. Adult patients selected for lumbar spinal fusion with decompression surgery and a minimum score of 24 on the pain catastrophizing scale will be randomized with 1:1 allocation for either perioperative cognitive behavioral therapy (intervention group) or a perioperative education plus progressive exercise program (control group). Patients randomized to the intervention group will receive six individual sessions of cognitive behavioral therapy, two sessions before the operation and four after. Primary outcome is the Core Outcome Measures Index at 12 months. Secondary outcomes include pain, disability, depression and quality of life. Discussion This is the first trial that evaluates the effectiveness of cognitive behavioral therapy as a perioperative tool to improve pain and disability after spinal fusion surgery in comparison with an educational/exercise control intervention, in patients with high levels of pain catastrophizing. If perioperative cognitive behavioral therapy proves to be effective, this might have important clinical implications, reducing the incidence of chronic postsurgical pain and improving outcome after spinal fusion surgery. Abstract The Aids to Management are a product of the Global Campaign against Headache, a worldwide programme of action conducted in official relations with the World Health Organization. Developed in partnership with the European Headache Federation, they update the first edition published 11 years ago. The common headache disorders (migraine, tension-type headache and medication-overuse headache) are major causes of ill health. They should be managed in primary care, firstly because their management is generally not difficult, and secondly because they are so common. These Aids to Management, with the European principles of management of headache disorders in primary care as the core of their content, combine educational materials with practical management aids. They are supplemented by translation protocols, to ensure that translations are unchanged in meaning from the English-language originals. The Aids to Management may be individually downloaded and, as is the case for all products of the Global Campaign against Headache, are available without restriction for non-commercial use.

Background: In recent years, more awareness is raised about sex-specific dilemmas in inherited bleeding disorders. However, no large studies have been performed to assess differences in diagnosis, bleeding phenotype and management of men and women with bleeding disorders. Therefore, we investigated sex differences in a large cohort of well-defined patients with autosomal inherited bleeding disorders (von Willebrand disease (VWD), rare bleeding disorders (RBDs) and congenital platelet defects (CPDs)). Methods: We included patients from three nationwide cross-sectional studies on VWD, RBDs and CPDs in the Netherlands, respectively the WiN, RBiN and TiN study. In all studies a bleeding score (BS) was obtained, and patients filled in an extensive questionnaire on the management and burden of their disorder. Findings: We included 1092 patients (834 VWD; 196 RBD; 62 CPD), of whom 665 (60.9%) were women. Women were more often referred because of a bleeding diathesis than men (47.9% vs 36.6%, p = 0.002). Age of first bleeding was similar between men and women, respectively 8.9 ± 13.6 (mean ±sd) years and 10.6 ± 11.3 years (p = 0.075). However, the diagnostic delay, which was defined as time from first bleeding to diagnosis, was longer in women (11.6 ± 16.4 years) than men (7.7 ± 16.6 years, p = 0.002). Similar results were found when patients referred for bleeding were analyzed separately. Of women aging 12 years or older, 469 (77.1%) had received treatment because of sex-specific bleeding. Interpretation: Women with autosomal inherited bleeding disorders are more often referred for bleeding, have a longer diagnostic delay, and often require treatment because of sex-specific bleeding. Abstract We evaluated whether cortical interruptions classified as vascular channel (VC) on high-resolution peripheral quantitative computed tomography (HR-pQCT) could be confirmed by histology. We subsequently evaluated the image characteristics of histologically identified VCs on matched single and multiplane HR-pQCT images. Four 3-mm thick portions in three anatomic metacarpophalangeal joint specimens were selected for histologic sectioning. First, VCs identified with HR-pQCT were examined for confirmation on histology. Second and independently, VCs identified by histology were matched to single and multiplane HR-pQCT images to assess for presence of cortical interruptions. Only one out of five cortical interruptions suggestive for VC on HR-pQCT could be confirmed on histology. In contrast, 52 VCs were identified by histology of which 39 (75%) could be classified as cortical interruption or periosteal excavation on matched single HR-pQCT slices. On multiplane HR-pQCT images, 11 (21%) showed a cortical interruption in at least two consecutive slices in two planes, 36 (69%) in at least one slice in two planes and five (10%) showed no cortical interruption. Substantially more VCs were present in histology sections than initially suggested by HR-pQCT. The small size and heterogeneous presentation, limit the identification as VC on HR-pQCT.

The use of in situ measurements is essential in the validation and evaluation of the algorithms that provide coastal water quality data products from ocean colour satellite remote sensing. Over the past decade, various types of ocean colour algorithms have been developed to deal with the optical complexity of coastal waters. Yet there is a lack of a comprehensive intercomparison due to the availability of quality checked in situ databases. The CoastColour Round Robin (CCRR) project, funded by the European Space Agency (ESA), was designed to bring together three reference data sets using these to test algorithms and to assess their accuracy for retrieving water quality parameters. This paper provides a detailed description of these reference data sets, which include the Medium Resolution Imaging Spectrometer (MERIS) level 2 match-ups, in situ reflectance measurements, and synthetic data generated by a radiative transfer model (HydroLight). These data sets, representing mainly coastal waters, are available from doi:10.1594/PANGAEA.841950. The data sets mainly consist of 6484 marine reflectance (either multispectral or hyperspectral) associated with various geometrical (sensor viewing and solar angles) and sky conditions and water constituents: total suspended matter (TSM) and chlorophyll a (CHL) concentrations, and the absorption of coloured dissolved organic matter (CDOM). Inherent optical properties are also provided in the simulated data sets (5000 simulations) and from 3054 match-up locations. The distributions of reflectance at selected MERIS bands and band ratios, CHL and TSM as a function of reflectance, from the three data sets are compared. Match-up and in situ sites where deviations occur are identified. The distributions of the three reflectance data sets are also compared to the simulated and in situ reflectances used previously by the International Ocean Colour Coordinating Group (IOCCG, 2006) for algorithm testing, showing a clear extension of the CCRR data which covers more turbid waters.

Global land cover is a key variable in the earth system with feedbacks on climate, biodiversity and natural resources. However, global land cover data sets presently fall short of user needs in providing detailed spatial and thematic information that is consistently mapped over time and easily transferable to the requirements of earth system models. In 2009, the European Space Agency launched the Climate Change Initiative (CCI), with land cover (LC_CCI) as 1 of 13 essential climate variables targeted for research development. The LC_CCI was implemented in three phases: first responding to a survey of user needs; developing a global, moderate-resolution land cover data set for three time periods, or epochs (2000, 2005, and 2010); and the last phase resulting in a user tool for converting land cover to plant functional type equivalents. Here we present the results of the LC_CCI project with a focus on the mapping approach used to convert the United Nations Land Cover Classification System to plant functional types (PFTs). The translation was performed as part of consultative process among map producers and users, and resulted in an open-source conversion tool. A comparison with existing PFT maps used by three earth system modeling teams shows significant differences between the LC_CCI PFT data set and those currently used in earth system models with likely consequences for modeling terrestrial biogeochemistry and land–atmosphere interactions. The main difference between the new LC_CCI product and PFT data sets used currently by three different dynamic global vegetation modeling teams is a reduction in high-latitude grassland cover, a reduction in tropical tree cover and an expansion in temperate forest cover in Europe. The LC_CCI tool is flexible for users to modify land cover to PFT conversions and will evolve as phase 2 of the European Space Agency CCI program continues.

As global demand for livestock products (such as meat, milk and eggs) is expected to double by 2050, necessary increases to future production must be reconciled with negative environmental impacts that livestock cause. This paper describes the LivestockPlus concept and demonstrates how the sowing of improved forages can lead to the sustainable intensification of mixed crop-forage-livestock-tree systems in the tropics by producing multiple social, economic and environmental benefits. Sustainable intensification not only improves the productivity of tropical forage-based systems but also reduces the ecological footprint of livestock production and generates a diversity of ecosystem services (ES) such as improved soil quality and reduced erosion, sedimentation and greenhouse gas (GHG) emissions. Integrating improved grass and legume forages into mixed production systems (crop-livestock, tree-livestock, crop-tree-livestock) can restore degraded lands and enhance system resilience to drought and waterlogging associated with climate change. When properly managed tropical forages accumulate large amounts of carbon in soil, fix atmospheric nitrogen (legumes), inhibit nitrification in soil and reduce nitrous oxide emissions (grasses), and reduce GHG emissions per unit livestock product. The LivestockPlus concept is defined as the sustainable intensification of forage-based systems, which is based on 3 interrelated intensification processes: genetic intensification - the development and use of superior grass and legume cultivars for increased livestock productivity; ecological intensification - the development and application of improved farm and natural resource management practices; and socio-economic intensification - the improvement of local and national institutions and policies, which enable refinements of technologies and support their enduring use. Increases in livestock productivity will require coordinated efforts to develop supportive government, non-government organization and private sector policies that foster investments and fair market compensation for both the products and ES provided. Effective research-for-development efforts that promote agricultural and environmental benefits of forage-based systems can contribute towards implemention of LivestockPlus across a variety of geographic, political and socio-economic contexts.Keywords: Eco-efficiency, environmental benefits, livestock and environment, mixed farming, pastures, smallholders.DOI: 10.17138/TGFT(3)59-82

Nitrogen (N), the most critical and essential nutrient for plant growth, largely determines the productivity in both extensive and intensive grassland systems. Nitrification and denitrification processes in the soil are the primary drivers of generating reactive N (NO3-, N2O and NO), largely responsible for N loss and degradation of grasslands. Suppressing nitrification can thus facilitate retention of soil N to sustain long-term productivity of grasslands and forage-based production systems. Certain plants can suppress soil nitrification by releasing inhibitors from roots, a phenomenon termed ‘biological nitrification inhibition’ (BNI). Recent methodological developments [e.g. bioluminescence assay to detect biological nitrification inhibitors (BNIs) from plant-root systems] led to significant advances in our ability to quantify and characterize BNI function in pasture grasses. Among grass pastures, BNI capacity is strongest in low-N environment grasses such as Brachiaria humidicola and weakest in high-N environment grasses such as Italian ryegrass (Lolium perenne) and B. brizantha. The chemical identity of some of the BNIs produced in plant tissues and released from roots has now been established and their mode of inhibitory action determined on nitrifying Nitrosomonas bacteria. Synthesis and release of BNIs is a highly regulated and localized process, triggered by the presence of NH4+ in the rhizosphere, which facilitates release of BNIs close to soil-nitrifier sites. Substantial genotypic variation is found for BNI capacity in B. humidicola, which opens the way for its genetic manipulation. Field studies suggest that Brachiaria grasses suppress nitrification and N2O emissions from soil. The potential for exploiting BNI function (from a genetic improvement and a system perspective) to develop production systems, that are low-nitrifying, low N2O-emitting, economically efficient and ecologically sustainable, is discussed.

Phaeocystis globosa (Prymnesiophyceae) is an ecologically dominating phytoplankton species in many areas around the world. It plays an important role in both the global sulfur and carbon cycles, by the production of dimethylsulfide (DMS) and the drawdown of inorganic carbon. Phaeocystis globosa has a polymorphic life cycle and is considered to be a harmful algal bloom (HAB) forming species. All these aspects make this an interesting species to study the effects of increasing carbon dioxide (CO2) concentrations, due to anthropogenic carbon emissions. Here, the combined effects of three different dissolved carbon dioxide concentrations (CO2(aq)) (low: 4 μmol kg−1, intermediate: 6–10 μmol kg−1 and high CO2(aq): 21–24 μmol kg−1) and two different light intensities (low light, suboptimal: 80 μmol photons m−2 s−1 and high light, light saturated: 240 μmol photons m−2 s−1) are reported. The experiments demonstrated that the specific growth rate of P. globosa in the high light cultures decreased with increasing CO2(aq) from 1.4 to 1.1 d−1 in the low and high CO2 cultures, respectively. Concurrently, the photosynthetic efficiency (FV/FM) increased with increasing CO2(aq) from 0.56 to 0.66. The different light conditions affected photosynthetic efficiency and cellular chlorophyll a concentrations, both of which were lower in the high light cultures as compared to the low light cultures. These results suggest that in future inorganic carbon enriched oceans, P. globosa will become less competitive and feedback mechanisms to global change may decrease in strength.

The quality of life (QoL) of informal caregivers can be adversely affected by a number of factors. This issue, however, has not been well explored for carers of people with Parkinson's (PwP), with research largely restricted to the assessment of caregiver burden and caregiver strain. This study aims to determine the main influences on carer QoL in this population and consider results in the context of current clinical guidelines for the management of Parkinson's disease (PD). Carers completed the newly validated PDQ-Carer, and PwP completed the PDQ-39. The sample comprised 238 carers (mean age 68.20 years) and 238 PwP (mean age 71.64). Results suggest multiple influences on caregiver QoL. These include carer age, gender, health status, and duration of the caregiving role. PwP levels of mobility and cognitive impairment are also significant influences on carer QoL. Not only should practitioners and service providers be particularly aware of the heightened impact of PD on carers over time and as PwP symptoms deteriorate, but this should also be reflected in clinical guidelines for the management of PD.

Biological aerosol particles affect human health, are essential for microbial and gene dispersal, and have been proposed as important agents for atmospheric processes. However, the abundance and size distributions of atmospheric biological particles are largely unknown. In this study we used a laser-induced fluorescence instrument to measure fluorescent biological aerosol particle (FBAP) concentrations for 18 months (October 2020–April 2022) at a rural, forested site in Sweden. The aim of this study was to investigate FBAP number concentrations (NFBAP) over time and analyze their relationship with meteorological parameters. NFBAP was highest in summer and lowest in winter, exhibiting a ∼ 5-fold difference between these seasons. The median NFBAP was 0.0050, 0.0025, 0.0027, and 0.0126 cm−3 in fall, winter, spring, and summer, respectively, and constituted ∼ 0.1–0.5 % of the total supermicron particle number concentration. NFBAP was dominated by the smallest measured size fraction (1–3 µm), suggesting that the main portions of the biological particles measured were due to single bacterial cells, fungal spores, and bacterial agglomerates. NFBAP was significantly correlated with increasing air temperature (P<0.01) in all seasons. For most of the campaign NFBAP was seen to increase with wind speed (P<0.01), while the relationship with relative humidity was for most of the campaign nonsignificant (46 %) but for a large part (30 %) negative (P<0.05). Our results indicate that NFBAP was highest during warm and dry conditions when wind speeds were high, suggesting that a major part of the FBAP in spring and summer was due to mechanical aerosol generation and release mechanisms. In fall, relative humidity may have been a more important factor in bioaerosol release. This is one of the longest time series of atmospheric FBAPs, which are greatly needed for estimates of bioaerosol background concentrations in comparable regions.

Abstract Background Four large community-randomized trials examining universal testing and treatment (UTT) to reduce HIV transmission were conducted between 2012–2018 in Botswana, Kenya, Uganda, Zambia and South Africa. In 2014, the UNAIDS 90–90-90 targets were adopted as a useful metric to monitor coverage. We systematically review the approaches used by the trials to measure intervention delivery, and estimate coverage against the 90–90-90 targets. We aim to provide in-depth understanding of the background contexts and complexities that affect estimation of population-level coverage related to the 90–90-90 targets. Methods Estimates were based predominantly on “process” data obtained during delivery of the interventions which included a combination of home-based and community-based services. Cascade coverage data included routine electronic health records, self-reported data, survey data, and active ascertainment of HIV viral load measurements in the field. Results The estimated total adult populations of trial intervention communities included in this study ranged from 4,290 (TasP) to 142,250 (Zambian PopART Arm-B). The estimated total numbers of PLHIV ranged from 1,283 (TasP) to 20,541 (Zambian PopART Arm-B). By the end of intervention delivery, the first-90 target (knowledge of HIV status among all PLHIV) was met by all the trials (89.2%-94.0%). Three of the four trials also achieved the second- and third-90 targets, and viral suppression in BCPP and SEARCH exceeded the UNAIDS target of 73%, while viral suppression in the Zambian PopART Arm-A and B communities was within a small margin (~ 3%) of the target. Conclusions All four UTT trials aimed to implement wide-scale testing and treatment for HIV prevention at population level and showed substantial increases in testing and treatment for HIV in the intervention communities. This study has not uncovered any one estimation approach which is superior, rather that several approaches are available and researchers or policy makers seeking to measure coverage should reflect on background contexts and complexities that affect estimation of population-level coverage in their specific settings. All four trials surpassed UNAIDS targets for universal testing in their intervention communities ahead of the 2020 milestone. All but one of the trials also achieved the 90–90 targets for treatment and viral suppression. UTT is a realistic option to achieve 95–95-95 by 2030 and fast-track the end of the HIV epidemic.

Abstract The use of supplementary cementitious materials (SMCs) is one of the three essential factors for the sustainability of the cement industry. To reduce CO2 emissions, several studies have introduced new methods for the utilization of industrial wastes such as the addition into cementitious materials. The objectives of this study were to chemically and physically characterize refractory ceramic industry waste and determine its viability as an SMC. The pozzolanic characteristics or SMC characteristics of the refractory waste were investigated using standard tests. The results revealed that the refractory waste was mainly composed of Al2O3 and SiO2. The mineralogical analysis revealed that the mullite was the major phase of the waste, followed by small amounts of cristobalite and quartz phases. The results of the modified Chapelle test and pozzolanic activity index with lime after 7 days indicated that the waste did not meet the minimum pozzolanicity required by the standard. However, the waste improved the mechanical resistance of the cement specimens containing the waste after 28 days compared with the reference sample, indicating the promising potential of the material for this application. This result indicated that the refractory waste shows a filler type effect, and thus, can be employed as an SCM.

Abstract Background Haemophilic arthropathy is a chronic and debilitating joint disease caused by recurrent spontaneous joint bleeds in patients with haemophilia. Understanding how characteristics of individual joint bleeds relate to the subsequent development of arthropathy could improve management and prevention of this joint disease. Here, we aimed to explore relations between joint bleed characteristics and development of bone pathology in a mouse model of haemophilic arthropathy by using novel in vivo imaging methodology. Methods We characterised induced knee bleeds in a murine model of haemophilic arthropathy by quantitative in vivo fluorescence molecular tomography (FMT) and by measurements of changes in the diameter of the injured knee. Wild-type mice and non-injured haemophilic mice acted as controls. Development of arthropathy was characterised by post mortem evaluation of bone pathology by micro-CT 14 days after bleed-induction. In an in vitro study, we assessed the effect of blood on the quantification of fluorescent signal with FMT. Results In most injured haemophilic mice, we observed significant loss of trabecular bone, and half of the mice developed pathological bone remodelling. Development of pathological bone remodelling was associated with significantly increased fluorescent signal and diameter of the injured knee just 1 day after induction of the bleed. Further, a correlation between the fluorescent signal 1 day after induction of the bleed and loss of trabecular bone reached borderline significance. In the in vitro study, we found that high concentrations of blood significantly decreased the fluorescent signal. Conclusion Our results add novel insights on the pathogenesis of haemophilic arthropathy and underline the importance of the acute phase of joint bleeds for the subsequent development of arthropathy.

Acute pancreatitis can result in fat necrosis, typically occurring in the peripancreatic region within weeks to months, and it generally appears as a low attenuation collection, with minimal heterogeneity. There are no specific imaging features that can diagnose retroperitoneal fat necrosis which may imitate other entities including certain malignancies, which may lead to invasive studies for diagnosis. Herein, we present a case of extensive retroperitoneal fat necrosis beyond the peripancreatic region that developed 10 days after an episode of acute pancreatitis.

Museums and their collections have specially customized databases in order to optimally gather and record their contents and associated metadata associated with their specimens. To share, exchange, and publish data, an appropriate data standard is essential. ABCD (Access to Biological Collection Data) is a standard for biological collection units, including living and preserved specimen, together with field observation data. Its extension, EFG (Extension for Geoscience), enables sharing and publishing data related to paleontological, mineralogical, and petrological objects. The standard is very granular and allows detailed descriptions, including information about the collection event itself, the holding institution, stratigraphy, chemical analysis, and host rock. The standard extension was developed in 2006 and has been used since then by different initiatives and applied for the publication of collection-related data in domain-specific and interdisciplinary portals.

Abstract Two important and large non-point sources of nitrogen in residential areas that adversely affect water quality are stormwater runoff and effluent from on-site treatment systems. These sources are challenging to control due to their variable flow rates and nitrogen concentrations. Denitrifying bioreactors that employ a lignocellulosic wood chip medium contained within a saturated (anoxic) zone are relatively new technology that can be implemented at the local level to manage residential non-point nitrogen sources. In these systems, wood chips serve as a microbial biofilm support and provide a constant source of organic substrate required for denitrification. Denitrifying wood chip bioreactors for stormwater management include biofilters and bioretention systems modified to include an internal water storage zone; for on-site wastewater, they include upflow packed bed reactors, permeable reactive barriers, and submerged wetlands. Laboratory studies have shown that these bioreactors can achieve nitrate removal efficiencies as high as 80–100% but could provide more fundamental insight into system design and performance. For example, the type and size of the wood chips, hydraulic loading rate, and dormant period between water applications affects the hydrolysis rate of the lignocellulosic substrate, which in turn affects the amount and bioavailability of dissolved organic carbon for denitrification. Additional field studies can provide a better understanding of the effect of varying environmental conditions such as ambient temperature, precipitation rates, household water use rates, and idle periods on nitrogen removal performance. Long-term studies are also essential for understanding operations and maintenance requirements and validating mathematical models that integrate the complex physical, chemical, and biological processes occurring in these systems. Better modeling tools could assist in optimizing denitrifying wood chip bioreactors to meet nutrient reduction goals in urban and suburban watersheds.

ABSTRACT: Control of Conyza spp. prior to soybean sowing has faced difficulties because of the resistance cases in Brazil, hence new herbicides as halauxifen-methyl + diclosulam are important to manage this specie. The objective of this research was to evaluate the effectiveness of the herbicide halauxifen-methyl + diclosulam applied at pre-planting of soybean. The experiments were set up in three locations in Paraná State, Brazil, in the 2015/2016 season. The herbicide halauxifen-methyl + diclosulam was associated with glyphosate and compared to other commercial herbicide to Conyza spp. control. The application occurred 15 days before soybean sowing, in plants of Conyza spp. with an average height of 20-35 cm in the three areas. None of the treatments promoted Conyza spp. control above 90%, when soybean was at the V1 stage; however, the findings showed that halauxifen-methyl + diclosulam promoted better control in comparison to the other treatments. The control reached >90% at V3 soybean development stage, in which treatments containing glyphosate + halauxifen-methyl + diclosulam were more effective than the other treatments. In addition, treatments with glyphosate + halauxifen-methyl + diclosulam decreased dry matter content of Conyza spp. between 87 and 93%, depending on the location, when compared to the control. The burndown treatment to Conyza spp. with glyphosate + halauxifen-methyl + diclosulam did not decrease soybean yield, and it was always higher than the untreated check.

Abstract In the germ line and during early embryogenesis, DNA methylation (DNAme) undergoes global erasure and re-establishment to support germ cell and embryonic development. While DNAme acquisition during male germ cell development is essential for setting genomic DNA methylation imprints, other intergenerational roles for paternal DNAme in defining embryonic chromatin are unknown. Through conditional gene deletion of the de novo DNA methyltransferases Dnmt3a and/or Dnmt3b, we observe that DNMT3A primarily safeguards against DNA hypomethylation in undifferentiated spermatogonia, while DNMT3B catalyzes de novo DNAme during spermatogonial differentiation. Failing de novo DNAme in Dnmt3a/Dnmt3b double deficient spermatogonia is associated with increased nucleosome occupancy in mature sperm, preferentially at sites with higher CpG content, supporting the model that DNAme modulates nucleosome retention in sperm. To assess the impact of altered sperm chromatin in formatting embryonic chromatin, we measure H3K4me3 occupancy at paternal and maternal alleles in 2-cell embryos using a transposon-based tagging approach. Our data show that reduced DNAme in sperm renders paternal alleles permissive for H3K4me3 establishment in early embryos, independently of possible paternal inheritance of sperm born H3K4me3. Together, this study provides evidence that paternally inherited DNAme directs chromatin formation during early embryonic development.

Abstract Background Ketamine is a promising therapy for treatment-resistant depression due to its rapid onset, although benefits are often transitory, with patients needing maintenance therapy to prevent relapse. Most data supporting ketamine for treatment-resistant depression refers to the intravenous route of administration, leaving alternative routes lacking in data, especially as maintenance regimens. Moreover, the safety of ketamine maintenance therapy is poorly defined. This report aims to describe and evaluate a novel hospital-to-outpatient intranasal racemic ketamine maintenance therapy program. Methods This was an observational program evaluation study. Participants were adult inpatients with treatment-resistant depression who had been successfully treated with intranasal racemic ketamine in hospital and were being referred for outpatient maintenance therapy with an intranasal racemic ketamine spray, administered at a specialized community treatment centre. Effectiveness was assessed with the Self-Report Quick Inventory of Depressive Symptomatology, the Quality of Life Scale, and the Clinical Global Impression-Improvement scale. Results Five patients were enrolled, completing up to 14 treatment sessions over 192 days. The mean dose administered throughout treatment was 220 mg (100 to 400 mg). All patients had decreased (or stable) depressive symptoms and increased (or stable) quality of life. There were no serious adverse events or discontinuations due to adverse effects. Reported adverse effects included anxiety and nausea. Slight blood pressure increases were seen during treatment, none of which required intervention. Conclusions Intranasal racemic ketamine maintenance therapy for treatment-resistant depression appeared to be feasible and well tolerated, although effectiveness conclusions cannot be drawn from this small pilot study. Further investigations regarding the safety and effectiveness of intranasal ketamine maintenance therapy are warranted.

Abstract This systematic review examines four themes of device-based remote monitoring (DRM): technology, patient monitoring and support, integration of DRM into clinical care, and patient engagement, and their impact on hospital service use. We included randomized controlled trials (RCTs) until 2024 comparing hospital service use in DRM with usual care. Hospital service use decreased in DRM in 72% of the 116 included RCTs. Non-implantable devices were most commonly used to measure data, but showed a lower decrease in hospital service use than implanted or mobile devices (69% vs 89% and 76%). Providing 24/7 support for patients led to a decrease (81% of the studies). DRM replacing usual care, involving designated healthcare providers, and patient-performed data transmission led to a greater decrease in hospital service use. DRM has the potential to further reduce hospital service use with additional factors such as sufficient support, automated processes, and optimized care redesign.

Ozone plays a crucial role in tropospheric chemistry, is the third largest contributor to greenhouse radiative forcing after carbon dioxide and methane and also a toxic air pollutant affecting human health and agriculture. Long-term measurements of tropospheric ozone have been performed globally for more than 30 years with UV photometers, all relying on the absorption of ozone at the 253.65 nm line of mercury. We have re-determined this cross-section and report a value of 11.27 x 10−18 cm2 molecule−1 with an expanded relative uncertainty of 0.86% (coverage factor k= 2). This is lower than the conventional value currently in use and measured by Hearn (1961) with a relative difference of 1.8%, with the consequence that historically reported ozone concentrations should be increased by 1.8%. In order to perform the new measurements of cross-sections with reduced uncertainties, a system was set up to generate pure ozone in the gas phase together with an optical system based on a UV laser with lines in the Hartley band, including accurate path length measurement of the absorption cell and a careful evaluation of possible impurities in the ozone sample by mass spectrometry and Fourier transform infrared spectroscopy. This resulted in new measurements of absolute values of ozone absorption cross-sections of 9.48 x 10−18, 10.44 x 10−18 and 11.07 x 10−18 cm2 molecule−1, with relative expanded uncertainties better than 0.7%, for the wavelengths (in vacuum) of 244.06, 248.32, and 257.34 nm respectively. The cross-section at the 253.65 nm line of mercury was determined by comparisons using a Standard Reference Photometer equipped with a mercury lamp as the light source. The newly reported value should be used in the future to obtain the most accurate measurements of ozone concentration, which are in closer agreement with non-UV-photometry based methods such as the gas phase titration of ozone with nitrogen monoxide.

Abstract Background Escherichia coli is the most common cause of human bloodstream infections and bacterial sepsis/septic shock. However, translation of preclinical septic shock resuscitative therapies remains limited mainly due to low-fidelity of available models in mimicking clinical illness. To overcome the translational barrier, we sought to replicate sepsis complexity by creating an acutely critically-ill preclinical bacterial septic shock model undergoing active 48-h intensive care management. Aim To develop a clinically relevant large-animal (ovine) live-bacterial infusion model for septic shock. Methods Septic shock was induced by intravenous infusion of the live antibiotic resistant extra-intestinal pathogenic E. coli sequence type 131 strain EC958 in eight anesthetised and mechanically ventilated sheep. A bacterial dose range of 2 × 105–2 × 109 cfu/mL was used for the dose optimisation phase (n = 4) and upon dose confirmation the model was developed (n = 5). Post-shock the animals underwent an early-vasopressor and volume-restriction resuscitation strategy with active haemodynamic management and monitoring over 48 h. Serial blood samples were collected for testing of pro-inflammatory (IL-6, IL-8, VEGFA) and anti-inflammatory (IL-10) cytokines and hyaluronan assay to assess endothelial integrity. Tissue samples were collected for histopathology and transmission electron microscopy. Results The 2 × 107 cfu/mL bacterial dose led to a reproducible distributive shock within a pre-determined 12-h period. Five sheep were used to demonstrate consistency of the model. Bacterial infusion led to development of septic shock in all animals. The baseline mean arterial blood pressure reduced from a median of 91 mmHg (71, 102) to 50 mmHg (48, 57) (p = 0.004) and lactate levels increased from a median of 0.5 mM (0.3, 0.8) to 2.1 mM (2.0, 2.3) (p = 0.02) post-shock. The baseline median hyaluronan levels increased significantly from 25 ng/mL (18, 86) to 168 ng/mL (86, 569), p = 0.05 but not the median vasopressor dependency index which increased within 1 h of resuscitation from zero to 0.39 mmHg−1 (0.06, 5.13), p = 0.065, and. Over the 48 h, there was a significant decrease in the systemic vascular resistance index (F = 7.46, p = 0.01) and increase in the pro-inflammatory cytokines [IL-6 (F = 8.90, p = 0.02), IL-8 (F = 5.28, p = 0.03), and VEGFA (F = 6.47, p = 0.02)]. Conclusions This critically ill large-animal model was consistent in reproducing septic shock and will be applied in investigating advanced resuscitation and therapeutic interventions.

Abstract The capacity to perceive tactile input at the fingertips, referred to as tactile sensitivity, is known to diminish with age due to regressive changes to mechanoreceptor density and morphology. Sensitivity is measured as perceptual responses to stimuli of varying intensity. Contrary to traditional sensitivity monitoring instruments, smartphones are uniquely suited for remote assessment and have shown to deliver highly calibrated stimuli along a broad spectrum of intensity, which may improve test reliability. The aim of this study was to evaluate a vibration-emitting smartphone application, the Vibratus App, as a mode of estimating tactile sensory thresholds in the aging adult. The peripheral nerve function of 40 neurologically healthy volunteers (ages 18–71) was measured using monofilaments, a 128-Hz tuning fork, the Vibratus App, and nerve conduction studies (NCS). Between group differences were analyzed to determine each measurement’s sensitivity to age. Spearman correlation coefficients depicted the associative strength between hand-held measurements and sensory nerve action potential (SNAP) amplitude. Abstract Owing to its roles in cellular signal transduction, protein phosphorylation plays critical roles in myriad cell processes. That said, detecting and quantifying protein phosphorylation has remained a challenge. We describe the use of a novel mass spectrometer (Orbitrap Astral) coupled with data-independent acquisition (DIA) to achieve rapid and deep analysis of human and mouse phosphoproteomes. With this method, we map approximately 30,000 unique human phosphorylation sites within a half-hour of data collection. The technology is benchmarked to other state-of-the-art MS platforms using both synthetic peptide standards and with EGF-stimulated HeLa cells. We apply this approach to generate a phosphoproteome multi-tissue atlas of the mouse. Altogether, we detect 81,120 unique phosphorylation sites within 12 hours of measurement. With this unique dataset, we examine the sequence, structural, and kinase specificity context of protein phosphorylation. Finally, we highlight the discovery potential of this resource with multiple examples of phosphorylation events relevant to mitochondrial and brain biology.

Galaxy formation theory identifies superwinds as a key regulator of star formation rates, galaxy growth, and chemical enrichment. Thermal and radiation pressure are known to drive galactic-scale winds in dusty starbursting galaxies (e.g., M82), but modern numerical simulations have recently highlighted that cosmic-ray (CR)–driven winds may be especially important in normal galaxies with modest star formation rate surface densities. However, CR-driven winds have yet to be conclusively observed—leaving significant uncertainty in their detailed microphysics. We present MeerKAT radio continuum and H i spectral-line observations of one such normal galaxy, NGC 1532; a nearby ( D ~ 15 Mpc) and nearly edge-on ( i ≳ 80°) spiral galaxy tidally interacting with its smaller elliptical companion, NGC 1531. We find magnetized, highly ordered radio-continuum loops extending ~10 kpc above and below the disk, visibly connecting discrete star-forming regions in the disk to the center. The deep MeerKAT H i observations place an upper limit on the column density of neutral gas coincident with the outflow of N _H I ≲ 3 × 10 ^19 cm ^−2 . Unlike previously observed outflows—for which ejected gas and dust can be traced across multiple wavelengths—the loops in NGC 1532 show no detectable signs of dust or gas coincident with the radio emission far from the disk. We explore multiple possible mechanisms for driving this magnetic wind and favor an explanation where CR pressure plays a significant role in launching these outflows.

ABSTRACT Gene expression systems that transcend species barriers are needed for cross-species analysis of gene function. In particular, expression systems that can be utilized in both model and pathogenic bacteria underpin comparative functional approaches that inform conserved and variable features of bacterial physiology. In this study, we develop replicative and integrative vectors alongside a novel, IPTG-inducible promoter that can be used in the model bacterium Escherichia coli K-12 as well as strains of the antibiotic-resistant pathogen, Acinetobacter baumannii. We generate modular vectors that transfer by conjugation at high efficiency and either replicate or integrate into the genome, depending on design. Embedded in these vectors, we also developed a synthetic, IPTG-inducible promoter, PabstBR, that induces to a high level but is less leaky than the commonly used trc promoter. We show that PabstBR is titratable at both the population and single-cell levels, regardless of species, highlighting the utility of our expression systems for cross-species functional studies. Finally, as a proof of principle, we use our integrating vector to develop a reporter for the E. coli envelope stress σ factor, RpoE, and deploy the reporter in E. coli and A. baumannii, finding that A. baumannii does not recognize RpoE-dependent promoters unless RpoE is heterologously expressed. We envision that these vector and promoter tools will be valuable for the community of researchers who study the fundamental biology of E. coli and A. baumannii.IMPORTANCEAcinetobacter baumannii is a multidrug-resistant, hospital-acquired pathogen with the ability to cause severe infections. Understanding the unique biology of this non-model bacterium may lead to the discovery of new weaknesses that can be targeted to treat antibiotic-resistant infections. In this study, we provide expression tools that can be used to study the gene function in A. baumannii, including in drug-resistant clinical isolates. These tools are also compatible with the model bacterium, Escherichia coli, enabling cross-species comparisons of gene function. We anticipate that the use of these tools by the scientific community will accelerate our understanding of Acinetobacter biology.

Neste trabalho, apresenta-se o estudo experimental em escala laboratorial e industrial da incorporação de resíduo do tratamento de Drenagem Ácida de Mina (DAM) na formulação de massa de cerâmica vermelha, com substituição parcial das argilas. Foram realizados algumas análises e ensaios nas amostras dos blocos cerâmicos: análise dilatométrica a verde, absorção de água, resistência à compressão, eflorescência de sais solúveis e emissões atmosféricas (MP e SOx). Os resultados das análises nos corpos de prova ensaiados indicam aumento de resistência a compressão. Os ensaios de eflorescência de sais solúveis indicam maior intensidade de cristalização de sais solúveis na superfície dos corpos de prova e blocos cerâmicos à medida que se aumenta o percentual de resíduo. As análises realizadas nos blocos cerâmicos ensaiados em escala industrial, recomendam uma proporção de até 2,5% de resíduo na massa cerâmica.An experimental laboratory and industry scale study of the incorporation of residue from Acid Drainage Mine (ADM) in the ceramics mass, with partial substitution of clays, is presented. Some analyses and assays of the ceramic blocks were been carried out: dilatometry, water absorption, compressive strength, efflorescence and emissions (MP and SOx). The results show an increase of compressive strength. The efflorescence assays indicate greater intensity of crystallization of soluble salts in the surface of the ceramic blocks for increasing residue content. The analyses in the ceramic blocks in industrial scale recommend a ratio of up to 2.5% of waste in bulk ceramics.

Abstract Candida albicans and Staphylococcus aureus are two commonly associated pathogens that cause nosocomial infections with high morbidity and mortality. Our prior and current work using a murine model of polymicrobial intra-abdominal infection (IAI) demonstrates that synergistic lethality is driven by Candida-induced upregulation of functional S. aureus α-toxin leading to polymicrobial sepsis and organ damage. In order to determine the candidal effector(s) mediating enhanced virulence, an unbiased screen of C. albicans transcription factor mutants was undertaken revealing that zcf13Δ/Δ fails to drive augmented α-toxin or lethal synergism during co-infection. A combination of transcriptional and phenotypic profiling approaches shows that ZCF13 regulates genes involved in pentose metabolism, including RBK1 and HGT7 that contribute to fungal ribose catabolism and uptake, respectively. Subsequent experiments reveal that ribose inhibits the staphylococcal agr quorum sensing system and concomitantly represses toxicity. Unlike wild-type C. albicans, zcf13Δ/Δ did not effectively utilize ribose during co-culture or co-infection leading to exogenous ribose accumulation and agr repression. Forced expression of RBK1 and HGT7 in the zcf13Δ/Δ mutant fully restores pathogenicity during co-infection. Collectively, our results detail the interwoven complexities of cross-kingdom interactions and highlight how intermicrobial metabolism impacts polymicrobial disease pathogenesis with devastating consequences for the host.

The organophosphate chlorpyrifos is a commonly used pesticide for fruits and vegetables despite its association with neurotoxicity in humans. While some studies suggest that organophosphates may impact the gut microbiota, no studies to date have investigated the direct effect of chlorpyrifos on the gut microbiota with doses that approximate environmentally relevant dietary concentrations (EPA chronic reference dose: 0.3 µg/kg/day in humans and EPA acute reference dose: 5 µg/kg/day in humans). Thus, we examined the influence of chlorpyrifos on the gut microbiota by assessment of bacterial physiology and metabolism using flow cytometry, 1H NMR-based metabolomics, and changes in the cecal microbiota community with 16S rRNA amplicon sequencing and analysis. Chlorpyrifos did not directly damage bacteria but rather perturbed bacterial metabolism. Chlorpyrifos exposure to bacteria increased the concentration of amino acids, carbohydrates, and nucleic acids. The relative abundances of Lactobacillus, Allobaculum, Roseburia, and Butyricicoccus increased after exposure to chlorpyrifos. Analyses of the 16S rRNA gene amplicon data predicted decreased amino acid biosynthesis and nucleic acid degradation and increased glycolysis which was supported by 1H NMR-based metabolomics. Collectively, these results demonstrate that environmentally relevant doses of chlorpyrifos can impact the metabolic activity of isolated gut microbes which may result in an imbalance in overall gut metabolic activity.

Abstract Sphingolipids play vital roles in metabolism and regulation. Previously, the aryl hydrocarbon receptor (AHR), a ligand‐activated transcription factor, was reported to directly regulate ceramide synthesis genes by binding to their promoters. Herein, sphingosine kinase 2 (SPHK2), responsible for producing sphingosine‐1‐phosphate (S1P), was found to interact with AHR through LXXLL motifs, influencing AHR nuclear localization. Through mutagenesis and co‐transfection studies, AHR activation and subsequent nuclear translocation was hindered by SPHK2 LXXLL mutants or SPHK2 lacking a nuclear localization signal (NLS). Similarly, an NLS‐deficient AHR mutant impaired SPHK2 nuclear translocation. Silencing SPHK2 reduced AHR expression and its target gene CYP1A1, while SPHK2 overexpression enhanced AHR activity. SPHK2 was found enriched on the CYP1A1 promoter, underscoring its role in AHR target gene activation. Additionally, S1P rapidly increased AHR expression at both the mRNA and protein levels and promoted AHR recruitment to the CYP1A1 promoter. Using mouse models, AHR deficiency compromised SPHK2 nuclear translocation, illustrating a critical interaction where SPHK2 facilitates AHR nuclear localization and supports a positive feedback loop between AHR and sphingolipid enzyme activity in the nucleus. These findings highlight a novel function of SPHK2 in regulating AHR activity and gene expression.
