Abstract In this work, methylammonium lead triiodide (CH3NH3PbI3) perovskite solar cells with efficiencies higher than 18% were achieved using a new nanocomposite hole transport layer (HTL) by doping poly(ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) with a mixed dopant of polyaniline (PANI) and graphene oxide (GO). A synergistic engineering between GO, PANI, and PEDOT:PSS was accomplished to introduce additional energy levels between perovskite and PEDOT:PSS and increase the conductivity of PEDOT:PSS. Kelvin probe force microscope results confirmed that adding GO to PEDOT:PSS/PANI composite significantly reduced the average surface potential. This increased the open circuit voltage (Voc) to 1.05 V for the GO/PEDOT:PSS/PANI nanocomposite perovskite solar cells from the pristine PEDOT:PSS (Voc = 0.95 V) and PEDOT:PSS/PANI (Voc = 0.99 V). In addition, adding PANI to the HTLs substantially enhanced short circuit current density (Jsc). This was supported by the current sensing‐atomic force microscopy (CS‐AFM) and conductivity measurements. The PANI doped films showed superior electrical conductivity compared with those without PANI as indicated by CS‐AFM results. PANI can fill the gaps between the microflakes of GO and give rise to more compact hole transport material (HTM) layer. This led to a higher Jsc after doping with PANI, which was consistent with the incident photon‐to‐current efficiency and electrochemical impedance spectroscopy results. The results of X‐ray diffraction (XRD) and AFM indicated the GO/PANI doped HTMs significantly improved the crystallinity, topography, and crystal size of the perovskite film grown on their surface. A higher efficiency of 18.12% for p‐i‐n perovskite solar cells has been obtained by adding the mixed dopant of GO, PANI, and PEDOT:PSS, demonstrating better stability than the pristine PEDOT:PSS cell.

Abstract In this work, grain boundary (GB) potential barrier ( Δφ GB), dopant density (Pnet), and filled trap state density (PGB,trap) were manipulated at the nanoscale by exposing the fabricated perovskite films to various relative humidity (RH) environments. Spatial mapping of surface potential in the perovskite film revealed higher positive potential at GBs than inside the grains. The average Δφ GB, Pnet, and PGB,trap in the perovskite films decreased from 0% RH to 25% RH exposure, but increased when the RH increased to 35% RH and 45% RH. This clearly indicated that perovskite solar cells fabricated at 25% RH led to the lowest average GB potential, smallest dopant density, and least filled trap states density. This is consistent with the highest photovoltaic efficiency of 18.16% at 25% RH among the different relative humidities from 0% to 45% RH.

Traditional testing method of mine-used equipment on/off transducer needs three-phase AC power supply and electric fires or bulbs with large power to get current for transducer acting, which cannot get continuously adjustable current with complex wiring and low efficiency. For above problems, the paper proposed a design scheme of test tooling of mine-used equipment on/off transducer, gave design demands, and introduced composition and main circuits of the test tooling in details. The testing result of a mine-used equipment on/off transducer showed that the test tooling can generate continuously adjustable current with simple and efficient operation, and can be calibrated through qualified clamp meter for correctness and traceability of testing data.

Abstract Economic impacts of seasonal influenza vary across US counties, but little estimation has been conducted at the county level. This research computed annual economic costs of seasonal influenza for 3143 US counties based on Census 2010, identified inherent spatial patterns, and investigated cost-benefits of vaccination strategies. The computing model modified existing methods for national level estimation, and further emphasized spatial variations between counties, in terms of population size, age structure, influenza activity, and income level. Upon such a model, four vaccination strategies that prioritize different types of counties were simulated and their net returns were examined. The results indicate that the annual economic costs of influenza varied from $13.9 thousand to $957.5 million across US counties, with a median of $2.47 million. Prioritizing vaccines to counties with high influenza attack rates produces the lowest influenza cases and highest net returns. This research fills the current knowledge gap by downscaling the estimation to a county level, and adds spatial variability into studies of influenza economics and interventions. Compared to the national estimates, the presented statistics and maps will offer detailed guidance for local health agencies to fight against influenza.

Abstract Background Non-pharmaceutical strategies are vital in curtailing impacts of influenza and have been intensively studied in public health. However, few strategies have explicitly utilized the weekend effect, which has been widely reported to be capable of reducing influenza infections. This study aims to explore six weekend-extension strategies against seasonal and pandemic flu outbreaks. Methods The weekend-extension strategies were designed to extend regular two-day weekend by one, two and three days, respectively, and in combination with either a continuous or discontinuous pattern. Their effectiveness was evaluated using an established agent-based spatially explicit simulation model in the urbanized area of Buffalo, NY, US. Results If the extensions last more than two days, the weekend-extension strategies can remarkably reduce the overall disease attack rate of seasonal flu. Particularly, a three-day continuous extension is sufficient to suppress the epidemic and confine the spread of disease. For the pandemic flu, the weekend-extension strategies only produce a few mitigation effects until the extensions exceed three days. Sensitivity analysis indicated that a compliance level above 75% is necessary for the weekend-extension strategies to take effects. Conclusion This research is the first attempt to incorporate the weekend effect into influenza mitigation strategies. The results suggest that appropriate extensions of the regular two-day weekend can be a potential measure to fight against influenza outbreaks, while minimizing interruptions on normal rhythms of socio-economy. The concept of weekend extension would be particularly useful if there were a lack of vaccine stockpiles, e.g., in countries with limited health resources, or in the case of unknown emerging infectious diseases.

Background and Aim. We recently investigated whether Poria cocos water extract modulates ketamine-induced Rho signaling regulation and reverses ketamine-inhibited cell mobility and F-actin reconstruction in B35 and C6 cells. Various studies have mentioned that drugs of abuse induce changes in neuronal plasticity in the brain’s reward circuitry. Modulations in neuronal plasticity are closely related to Rho signaling regulation in cells. Rho signaling has also been implicated in the addictive behavior induced by chronic opiate or morphine administration. MK-801 could induce Rho signaling regulation to further modulate cell migration and actin reorganization in neuronal and glial cells. In this study, we investigated the effects of Poria cocos water extract on Rho signal regulation in MK-801-treated B35 and C6 cells. Methods. B35 neuronal cells and C6 glial cells were incubated with MK-801 for 7 days followed by MK-801, MK801 in combination with water extracts of P. cocos (PRP for P. cocos cum Radix Pini or WP for White Poria) treatment for an additional 7 days. Analysis of cell mobility, F-actin aggregation, and Rho signaling modulation was performed to clarify the roles of PRP or WP in MK-801-treated B35 and C6 cells. Results. MK-801 decreases B35 cell mobility, whereas the inhibited cell migration ability and F-actin aggregation in MK-801-treated B35 or C6 cells could be reversed by PRP or WP. The CDC42 expression in B35 or C6 cells would be reduced by MK-801 and restored by treating with PRP or WP. The RhoA expression was increased by MK-801 in both B35 and C6 cells but was differentially regulated by PRP or WP. In B35 cells, downregulation of PFN1, N-WASP, PAK1, and ARP2/3 induced by MK-801 can be reversely modulated by PRP or WP. PRP or WP reduced the increase in the p-MLC2 expression in B35 cells treated with MK-801. The reduction in ROCK1, PFN1, p-MLC2, and ARP2/3 expression in C6 cells induced by MK-801 was restored by PRP or WP. Reduced N-WASP and PAK1 expression was differentially regulated by PRP or WP in MK-801-treated C6 cells.

Objective. The glutamate system plays a major role in the development of neuropsychiatric disorders such as addiction, epilepsy, dementia, and psychosis. MK-801 (dizocilpine), an uncompetitive N-methyl-D-aspartate (NMDA) receptor antagonist, could increase locomotor activity and stereotyped neurobehaviors mimicking schizophrenic-like features in the mouse model. The study would explore the neuropharmacological differences of risperidone and valproic acid on the MK-801-induced neurobehavioral changes. Methods. The subjects were male C57BL/6J mice obtained from the National Laboratory Animal Center. Drug effects were assessed using the open field with a video-tracking system and gaiting tests. After habitation, risperidone (0, 0.1 mg/kg) or valproic acid (0, 200 mg/kg) was injected and ran locomotion for 30 mins. Sequentially, mice were followed by intraperitoneal injection (i.p.) with MK-801 (0, 0.2 mg/kg) and ran locomotion for 60 mins. Gaiting behaviors such as step angles, stride lengths, and stance widths were measured following the study drugs. Results. The results showed that risperidone and valproic acid alone could not alter the locomotor activities. Following the MK-801 injection, the travelled distance and speed in the entire open field dramatically increased. Background and Aim. MK801-induced psychotic symptoms and also the Ras homolog family member A (RhoA) expression and cell division control protein 42 (cdc42) mRNA modulation in the rat brain have been investigated. Antipsychotic drugs (APDs) have been reported to induce Rho GDP-dissociation inhibitor (RhoGDI) pathway regulation related to cytoskeleton reorganization in neuronal cells. It will be necessary to clarify the effects of APDs on MK801-induced RhoGDI signaling regulation in neuronal cells. Methods. B35 neuronal cells were treated with MK801 for 7 days then treated with MK801 in combination with haloperidol or clozapine for a further 7 days. Cell migration, F-actin condensation, and RhoGDI signaling regulation were examined to investigate the regulatory effects of MK801, haloperidol, and clozapine in B35 neuronal cells. Results. MK801 reduced B35 cell migration, whereas both haloperidol and clozapine reversed the reduction in cell migration induced by MK801. Haloperidol and clozapine restored F-actin condensation after it was diminished by MK801 in B35 cell nuclei. MK801 increased the RhoGDI1 and RhoA expression, which was diminished by the addition of haloperidol and clozapine. MK801 reduced the CDC42 expression, which was restored by haloperidol and clozapine. MK801 reduced the Rho-associated coiled-coil containing protein kinase 1 (ROCK1), profilin1 (PFN1), and neuronal Wiskott–Aldrich Syndrome protein (N-WASP) expression, which was further reduced by haloperidol and clozapine. MK801 also increased the phosphorylated myosin light chain 2 (p-MLC2), postsynaptic density protein 95 (PSD-95), and c-jun expression, which was decreased by haloperidol and clozapine. p21 (RAC1-) activated kinase 1 (PAK1) expression was not affected by MK801.

This work aims to explore the dynamical well-killing process of a vertical H2S-containing natural gas well. A dynamical well-killing model considering an H2S solubility was established to simulate the overflow and well-killing process of a vertical H2S-containing natural gas well. The mass and momentum equations of the coupled model were solved using finite difference method, while the transient temperature prediction model was solved using finite volume method. The coupled model was validated by reproducing experimental data and field data of Well Tiandong #5. The effect of H2S content, mud displacement, drilling fluid density, and initial overflow volume on the dynamical well-killing process of an H2S-containing natural gas well were obtained and analyzed in this work. Results showed that H2S will gasify near wellhead during well killing when casing pressure decreases. To balance the bottom hole pressure, when H2S releases, the casing pressure increases as H2S content increases. As initial overflow volume increases, the annular temperature, annular pressure and the casing pressure increase significantly. When H2S gasifies, the casing pressure applied at wellhead should be higher at lower initial overflow volume to balance bottom hole pressure. In the well-killing process, the annular pressure and temperature decrease as drilling fluid density increases and a lower casing pressure is needed for balancing bottom hole pressure. The casing pressure is lower at a higher displacement for higher friction resistance. Besides, as well-killing displacement increases H2S will gasify at an earlier time. When drilling for H2S-containing natural gas well, early detection of gas kick should be more frequent to avoid severe overflow. Besides, higher displacement and density of drilling fluid should be considered to avoid stratum fracturing and prevent leakage accidents under the premise of meeting drilling requirements.

A considerable number of studies for VIV under the uniform flow have been performed. However, research on VIV under shear flow is scarce. An experiment for VIV under the shear flow with the same shear parameter at the two different Reynolds numbers was conducted in a deep-water offshore basin. Various measurements were obtained by the fiber bragg grating strain sensors. Experimental data were analyzed by modal analysis method. Results show several valuable features. First, the corresponding maximum order mode of the natural frequency for shedding frequency is the maximum dominant vibration mode and multi-modal phenomenon is appeared in VIV under the shear flow, and multi-modal phenomenon is more apparent at the same shear parameter with an increasing Reynolds number under the shear flow effect. Secondly, the riser vibrates at the natural frequency and the dominant vibration frequency increases for the effect of the real-time tension amplitude under the shear flow and the IL vibration frequency is the similar with the CF vibration frequency at the Reynolds number of 1105 in our experimental condition and the IL dominant frequency is twice the CF dominant frequency with an increasing Reynolds number. In addition, the displacement trajectories at the different locations of the riser appear the same shape and the shape is changed at the same shear parameter with an increasing Reynolds number under the shear flow. The diagonal displacement trajectories are observed at the low Reynolds number and the crescent-shaped displacement trajectories appear with an increasing Reynolds number under shear flow in the experiment.

Background: This study investigated the mechanism by which tazarotene-induced gene 1 (TIG1) inhibits melanoma cell growth. The main focus was to analyze downstream genes regulated by TIG1 in melanoma cells and its impact on cell growth. Methods: The effects of TIG1 expression on cell viability and death were assessed using water-soluble tetrazolium 1 (WST-1) mitochondrial staining and lactate dehydrogenase release assays. RNA sequencing and Western blot analysis were employed to investigate the genes regulated by TIG1 in melanoma cells. Additionally, the correlation between TIG1 expression and its downstream genes was analyzed in a melanoma tissue array. Results: TIG1 expression in melanoma cells was associated with decreased cell viability and increased cell death. RNA-sequencing (RNA-seq), quantitative reverse transcription PCR (reverse RT-QPCR), and immunoblots revealed that TIG1 expression induced the expression of Endoplasmic Reticulum (ER) stress response-related genes such as Homocysteine-responsive endoplasmic reticulum-resident ubiquitin-like domain member 1 (HERPUD1), Binding immunoglobulin protein (BIP), and DNA damage-inducible transcript 3 (DDIT3). Furthermore, analysis of the melanoma tissue array revealed a positive correlation between TIG1 expression and the expression of HERPUD1, BIP, and DDIT3. Additionally, attenuation of the ER stress response in melanoma cells weakened the impact of TIG1 on cell growth. Conclusions: TIG1 expression effectively hinders the growth of melanoma cells. TIG1 induces the upregulation of ER stress response-related genes, leading to an increase in caspase-3 activity and subsequent cell death. These findings suggest that the ability of retinoic acid to prevent melanoma formation may be associated with the anticancer effect of TIG1.

Hebius sauteri (Boulenger, 1909) has been long recognized as a widely distributed species with three subspecies, namely Hebius sauteri sauteri from Taiwan Island and southeastern China, Hebius sauteri bourreti (Malnate, 1962) from northern Vietnam, and Hebius sauteri maximus (Malnate, 1962) from Sichuan, southwestern China. However, the validity of these subspecies of the H. sauteri complex has not been evaluated. In the current study, we re-evaluate the taxonomic status of H. s. maximus based on morphological and molecular evidence. Molecular phylogenetic results indicate that H. s. maximus is a distantly diverged clade with respect to H. s. sauteri, and that H. s. maximus is morphologically distinguishable from the nominative subspecies. Therefore, our results support the validity of H. s. maximus and we elevate it to full species status, namely Hebius maximus comb. nov. The distribution range of H. sauteri is revised according to our proposed taxonomic change. Furthermore, detailed redescription, natural history, and coloration of both H. sauteri and H. maximus and comments on the validity of H. s. bourreti are also provided.

In order to develop high strength Mg-Gd-Y-Zn alloys, key experiments coupled with CALPHAD (CALculation of PHAse Diagrams) calculations were carried out in the current work to provide critical understanding of this important alloy system. Three Mg-10Gd-xY-yZn (x = 4 or 5, y = 3 or 5, wt.%) were mapped on Mg-Gd-Y-Zn phase diagrams for phase equilibria and microstructure investigation. Electron microscopy was performed for phase identification and phase fraction determination in as-cast and solution treated conditions. In all three alloys, the major phases were Mg-matrix and long period stacking order (LPSO) 14H phase. With ST at 400 and 500 °C, the phase fraction of LPSO 14H increased, particularly the fine lamellar morphology in the Mg matrix. The as-cast and 400 °C Mg10Gd5Y3Zn samples had Mg5(Gd,Y) present. At 500 °C, Mg5(Gd,Y) is not stable and transforms into LPSO 14H. The Mg10Gd5Y5Zn alloy included the W-Phase, which showed a reduction in phase fraction with solution treatment. These experimental results were used to validate and improve the thermodynamic database of the Mg-Gd-Y-Zn system. Thermodynamic calculations using the improved database can well describe the available experimental results and make accurate predictions to guide the development of promising high-strength Mg-Gd-Y-Zn alloys.

BackgroundPost-stroke hemiparesis strongly affects stroke patients’ activities of daily living and health-related quality of life. Scalp acupuncture (SA) is reportedly beneficial for post-stroke hemiparesis. However, there is still no standard of SA for the treatment of post-stroke hemiparesis. Apriori algorithm-based association rule analysis is a kind of “if-then” rule-based machine learning method suitable for investigating the underlying rules of acupuncture point/location selections. This study aimed to investigate the core SA combinations for the treatment of post-stroke hemiparesis by using a systematic review and Apriori algorithm-based association rule analysis.MethodsWe conducted a systematic review to include relevant randomized controlled trial (RCT) studies investigating the effects of SA treatment in treating patients with post-stroke hemiparesis, assessed by the Fugl-Meyer Assessment (FMA) score. We excluded studies using herbal medicine or manual acupuncture.ResultsWe extracted 33 SA locations from the 35 included RCT studies. The following SA styles were noted: International Standard Scalp Acupuncture (ISSA), WHO Standard Acupuncture Point Locations (SAPL), Zhu’s style SA, Jiao’s style SA, and Lin’s style SA. Sixty-one association rules were investigated based on the integrated SA location data.ConclusionsSAPL_GV20 (Baihui), SAPL_GV24 (Shenting), ISSA_MS6_i (ISSA Anterior Oblique Line of Vertex-Temporal, lesion-ipsilateral), ISSA_MS7_i (ISSA Posterior Oblique Line of Vertex-Temporal, lesion-ipsilateral), ISSA_PR (ISSA Parietal region, comprised of ISSA_MS5, ISSA_MS6, ISSA_MS7, ISSA_MS8, and ISSA_MS9), and SAPL_Ex.HN3 (Yintang) can be considered the core SA location combination for the treatment of post-stroke hemiparesis. We recommend a core SA combination for further animal studies, clinical trials, and treatment strategies.

Hepatitis B virus X protein (HBx) and hepatic stellate cells (HSCs) are critical for liver fibrosis development. Anti-fibrosis occurs via reversion to quiescent-type HSCs or clearance of HSCs via apoptosis or ferroptosis. We aimed to elucidate the role of chrysophanol in rat HSC-T6 cells expressing HBx and investigate whether chrysophanol (isolated from Rheum palmatum rhizomes) influences cell death via ferroptosis in vitro. Analysis of lipid reactive oxygen species (ROS), Bip, CHOP, p-IRE1α, GPX4, SLC7A11, α-SMA, and CTGF showed that chrysophanol attenuated HBx-repressed cell death. Chrysophanol can impair HBx-induced activation of HSCs via endoplasmic reticulum stress (ER stress) and ferroptosis-dependent and GPX4-independent pathways.

AIM: To analyse the curative effect and influencing factors of Nd:YAG laser lacrimal duct angioplasty combined with lacrimal duct stent implantation on the treatment of lacrimal duct obstruction. METHODS: Totally 150 eyes of 150 patients with lacrimal duct obstruction admitted to our hospital from June 2017 to June 2019 were selected and treated with Nd:YAG laser lacrimal duct plasty combined with lacrimal duct stent placement. Follow-up 6mo after extubation, the patients were divided into effective group(135 cases, 135 eyes)and ineffective group(15 cases, 15 eyes)according to the clinical efficacy. General information, complications, causes of lacrimal duct blockage, postoperative lacrimal duct flushing frequency and clinical data such as the condition of bony lacrimal duct, analysis of related factors affecting the therapeutic effect.RESULTS: Follow-up 6mo after extubation, the effective rate and recurrence rate were 90.0%(135 eyes)and 1.3%(2 eyes)respectively. The course of the disease in the effective group was significantly shorter than that of the ineffective group, and the incidence of complications was significantly lower than in the ineffective group, the length of osseous lacrimal duct obstruction was significantly shorter than that of the ineffective group, the minimum diameter of the obstruction was significantly longer than that of the ineffective group, there was a significant difference in the frequency of tract flushing(all POR=4.600; 95%CI: 3.448-6.136), complications(OR=2.524; 95% CI: 1.481-4.302),the length of osseous lacrimal duct obstruction(OR=3.785; 95% CI: 1.844-7.770)and the minimum diameter of the obstruction(OR=2.330; 95% CI 1.464-3.708), as well as flushing frequency(OR=4.773; 95% CI: 2.227-10.231)were related factors affecting the therapeutic effect of Nd:YAG laser lacrimal duct plasty combined with lacrimal duct stent implantation.CONCLUSION: Nd:YAG laser lacrimal duct angioplasty combined with lacrimal duct stent implantation is effective in the treatment of lacrimal duct obstruction, with the lower incidence of complications and recurrence rate. The course of disease, complications, postoperative lacrimal passage irrigation frequency and the condition of bone lacrimal passage are all influencing factors, among which the effective rate can be improved by properly increasing the number of lacrimal duct expansion irrigation.

AIM:To study the microscopic changes of visual pathology in ocular hypertension(OHT)patients using magnetic resonance diffusion tensor imaging(DTI)technology and three-dimensional optical coherence tomography(3D-OCT)technology. METHODS: Twenty-six patients with 52 eyes diagnosed as OHT in our hospital from January 2016 to October 2019 were included in the case group. Twenty-six healthy patients who were matched with age and gender in the same period were included in the control group. All eyes were examined for best corrected visual acuity, central corneal thickness(CCT), non-contact intraocular pressure, while all eyes were examined by optical disc 3D-OCT combined optic nerve, optic chiasma, optic tract, and optic radiation DTI. Compared the differences between the two groups. RESULTS: The CCT of the OHT group was thinner than that of the normal control group; the intraocular pressure was higher than that of the normal control group, and the difference was statistically significant(all PPP>0.05). The FA values of bilateral optic nerve and optic radiation decreased compared with the normal control group, and the difference was statistically significant(all PP>0.05). CONCLUSION: 3D-OCT can obtain the thickness of CP-RNFL and the parameters of optic disc, DTI can reconstruct the intracranial visual pathway and can detect the microscopic changes of optic nerve, optic chiasma, optic tract, and optic radiation at early stage. Combining 3D-OCT and DTI technology can effectively understand the microscopic changes of the visual pathway and provide new models for clinical research of ocular hypertension patients.

Multi-pass hot extrusion was used to prepare the Al2O3/AZ31 composite.The microstru-cture was characterized by optical microscopy, scanning electron microscopy, transmission electron microscopy, and mechanical properties were tested by Vickers hardness tester and electron universal strength tester. The results show that Al2O3 particles are uniformly distributed in AZ31 magnesium matrix by multi-pass hot extrusion. The grain refining effect of Al2O3 particles on the matrix is enhanced, and the grain size of the composite decreases significantly with the increase of the pass. During the hot extrusion process, the dislocation density around the Al2O3 particles increases, and the high-density dislocation region facilitates the dynamic recrystallization nucleation, so that the grains of the Al2O3/AZ31 composite are significantly refined. The Al2O3 particles are gradually distributed into a long strip from the initial island distribution, then distributed in a linear pattern, and finally distributed uniformly in the form of particles in the AZ31 magnesium matrix. After fourth-pass hot extrusion, the mechanical properties of Al2O3/AZ31 composite are significantly improved, and hardness, tensile strength and yield strength are 89HV, 305MPa and 198MPa, respectively. The hardness, tensile strength and yield strength increase by 19.2%, 14.8%, and 14.1%, respectively, compared with the first-pass hot extrusion.

MicroRNAs (miRNAs) have been widely identified in porcine testicular tissues and implicated as crucial regulators of proliferation, apoptosis, and differentiation in porcine spermatogenesis related cells. However, the function roles of most of the miRNAs that have been identified in Sertoli cells are poorly understood. In the present study, six experiments were conducted to study the regulatory role of miR-10b in porcine immature Sertoli cells. In experiment 1, the results showed that the relative mRNA expression level of miR-10b in porcine testicular tissues decreased quadratically (P0.05) on porcine immature Sertoli cell apoptosis. Abstract The activation of hepatic stellate cells (HSCs) is an important step in the progress of liver fibrosis. Fibrosis can be impeded by HSC reversion to a quiescent state or HSC clearance through apoptosis. To investigate the apoptotic effects of hsian‐tsao (Mesona procumbens Hemsl) on human HSCs, the expression levels of cleaved caspase‐3, p38, and c‐Jun N‐terminal kinase (JNK) were assessed using Western blotting, and the caspase‐3 activity was measured using caspase‐3/CPP32 colorimetric assay kit. Hsian‐tsao extract (HTE) increased the activity of caspase‐3 and the level of activated caspase‐3, indicating the activation of apoptosis. The intracellular reactive oxygen species (ROS) level increased in a dose‐dependent manner. This increase was prevented by an antioxidant, suggesting that HTE induces ROS accumulation. In addition, we found that HTE induced the phosphorylation of the mitogen‐activated protein kinases JNK and p38. These collective data indicate that HTE induces apoptosis via ROS production through the p38, JNK, and caspase‐3‐dependent pathways. HTE may decrease HSC activation in liver fibrosis and may have a therapeutic potential.

MicroRNAs (miRNAs) are implicated in swine spermatogenesis via their regulations of cell proliferation, apoptosis, and differentiation. Recent studies indicated that miR-34c is indispensable in the late steps of spermatogenesis. However, whether miR-34c plays similar important roles in immature porcine Sertoli cells remain unknown. In the present study, we conducted two experiments using a completely randomised design to study the function roles of miR-34c. The results from experiment I demonstrated that the relative expression level of miR-34c in swine testicular tissues increased (P=0.0017) quadratically with increasing age, while the relative expression level of SMAD family member 7 (SMAD7) decreased (P=0.0009) with curve. Furthermore, miR-34c expression levels showed a significant negative correlation (P=0.013) with SMAD7 gene expression levels. Safflower extract is commonly used as a traditional Chinese medicine to promote blood circulation and remove blood stasis. The antioxidant and anticancer properties of safflower extracts have been extensively studied, but their antiaggregative effects have been less analyzed. We found that safflower extract inhibited human platelet aggregation induced by ADP. In addition, we further analyzed several safflower extract compounds, such as hydroxysafflor yellow A, safflower yellow A, and luteolin, which have the same antiaggregative effect. In addition to analyzing the active components of the safflower extract, we also analyzed their roles in the ADP signaling pathways. Safflower extract can affect the activation of downstream conductors of ADP receptors (such as the production of calcium ions and cAMP), thereby affecting the expression of activated glycoproteins on the platelet membrane and inhibiting platelet aggregation. According to the results of this study, the effect of safflower extract on promoting blood circulation and removing blood stasis may be related to its direct inhibition of platelet activation.

AIM: To measure the macular thickness of normal young people by 3D 1000 optical coherence tomography(OCT)and study the repeatability of measuring results and the relationship between the thicknesses of macular and gender. At the same time, to compare our result with the data of other types of OCT, and to understand the consistency of the measuring results of macular thickness of different types of OCT. METHODS: Totally 222 eyes in 111 young people were detected using 3D scan mode of Topcon 3D OCT 1 000(ver 2.4). Twelve cases(24 eyes)underwent repeatability check. We took transverse comparison between our measured results with other research's results. RESULTS:There were 111 cases of young people, whose age were from 18-27 years old, all uncorrected and corrected visual acuity were ≥1.0, all intraocular pressure were 3. Center thickness was 161-264μm, and the average thickness was 200.13±18.81μm. Central macular thickness were 188-273μm, and the average thickness was 229.00±18.20μm. The central macular thickness in men was significantly greater than that in women, and there was statistical difference. The results of repeated check of 12 cases(24 eyes)in the macular area were no statistical difference except the outer ring of nasal quadrant, and the repeatability of average thickness in central macular thickness was better than in center thickness. CONCLUSION:The repeatability of macular examination is good. The central macular thickness can be better repeated than the center thickness. The central macular thickness is 229.00±18.20μm in young people, according to the 3D 1000 OCT measurements. There are statistical difference of central macular thickness between different genders.

In this work, the Al-10Si-2.5Cu-0.8Fe alloy was used as the base alloy to squeeze casting with different content Mg-10Al master alloy addition. The effects of microstructure and mechanical properties were investigated by XRD, OM, SEM, and tensile test analysis. The results show that with the increase of Mg addition, the morphology of eutectic Si in the alloy changes from strip to fiber and the size is obviously reduced. Al _8 Mg _3 FeSi _6 phase in the microstructure gradually grows, and the splitting effect on the matrix structure is increased. The tensile strength and elongation of the alloy increased first and then decreased with the increase of Mg content. When the Mg content is 1.38%, the tensile strength of the alloy reached a maximum of 289 MPa and the elongation is 2.24%. The hardness of the alloy gradually increases. When the Mg content is 2.51%, the hardness reaches 121.8 HV. The cleavage surface in the fracture becomes fine with the increase of Mg content, the number of dimples increases, and the fracture mode transitions from quasi-cleavage fracture to brittle fracture.

H-rev107 is a member of the HREV107 type II tumor suppressor gene family and acts as a phospholipase to catalyze the release of fatty acids from glycerophospholipid. H-rev107 has been shown to play an important role in fat metabolism in adipocytes through the PGE2/cAMP pathway, but the detailed molecular mechanism underlying H-rev107-mediated lipid degradation has not been studied. In this study, the interaction between H-rev107 and cytochrome P450 reductase (POR), which is involved in hepatic lipid content regulation, was determined by yeast two-hybrid screen and confirmed by using in vitro pull down assays and immunofluorescent staining. The expression of POR in H-rev107-expressing cells enhanced the H-rev107-mediated release of arachidonic acid. However, H-rev107 inhibited POR activity and relieved POR-mediated decreased triglyceride content in HtTA and HeLa cervical cells. The inhibitory effect of H-rev107 will be abolished when POR-expressing cells transfected with PLA2-lacking pH-rev107 or treated with PLA2 inhibitor. Silencing of H-rev107 using siRNA resulted in increased glycerol production and reversion of free fatty acid-mediated growth suppression in Huh7 hepatic cells. In summary, our results revealed that H-rev107 is also involved in lipid accumulation in liver cells through the POR pathway via its PLA2 activity.

The complete molecule of the title compound, [MnCl2(C18H10N4)2], is generated by crystallographic twofold symmetry with the Mn atom lying on the rotation axis. The Mn coordination geometry is a distorted cis-MnCl2N4 octahedron, arising from two N,N′-bidentate dipyrido[3,2-a:2′,3′-c]phenazine (DPPZ) ligands and two chloride ions. In the crystal structure, neighbouring mononuclear units pack together through π–π contacts between the DPPZ rings [shortest centroid–centroid distance = 3.480 (2) Å], leading to a chain-like structure along [001]. C—H...Cl hydrogen bonds complete the structure.

The Ir centre in the title complex, [Ir(C11H6F2N)2(C6H4NO2)], is six-coordinated in a slightly distorted octahedral IrC2N3O fashion.

The carboxylate dianion in the title compound, [Pb(C14H8O5)(C19H12N4O)2]·2H2O, uses one carboxylate group to O,O′-chelate a bis[4-(1H-1,3,7,8-tetraazacyclopenta[l]phenanthren-2-yl)phenol]-chelated PbII atom and uses its other carboxylate group to bind to another PbII atom in an irregular monodentate manner. The PbII atom exists in an undefined seven-coordinate geometry in the chain structure; the lone pair is stereochemically active. Adjacent chains are linked by intermolecular O—H...N, N—H...O and O—H...O hydrogen bonds that involve the uncoordinated water molecules to form a three-dimensional network.

One of the two 1,4-dicarboxylate dianions in the title compound, [Mn2(C12H6O4)2(C19H12N4)2]·C3H7NO, uses its two carboxylate groups to chelate two N-heterocycle-chelated Mn atoms; the other 1,4-dicarboxylate dianion binds to four such metal centers. The octahedrally coordinated Mn atoms are linked through the two dianions into a layer motif; the dimethylformamide molecules occupy the spaces between adjacent layers. Ten C atoms and attached H atoms of one dianion are disordered equally over two positions.

The 1,4-dicarboxylate dianions in the title compound, [Mn(C12H6O4)(C19H12N4O)2]·0.5C12H8O4·H2O, bond to two 4-(1H-1,3,7,8-tetraazacyclopenta[l]phenanthren-2-yl)phenol-chelated Mn atoms to form a chain that features the metal atom in an octahedral coordination geometry. Adjacent chains interact with the uncoordinated water molecules to form a three-dimensional network. The naphthalene-1,4-dicarboxylic acid solvent molecule, which is disordered about a centre of inversion, occupies the space within the network but is not bonded to the network. One NH group is disordered equally over two positions.

In the title polymer, [Co3(C6H8O4)3(C19H12N4)2]n, two adipate dianions (C6H8O42−) occupy general positions and two are situated on different inversion centres. The two on general positions bind through their four O atoms to five 2-phenyl-1H-1,3,7,8-tetraazacyclopenta[l]phenanthrene-chelated CoII ions, whereas the two on special positions bind to only four. Of the three Co atoms, two are chelated by N-heterocycles; the third is bonded to six O atoms. The bonding mode of the dianion gives rise to a three-dimensional network structure; the network is further consolidated by N—H...O hydrogen bonds.
