Abstract The Dali River basin as the representative areas of the middle reaches of the Yellow River. Accurately evaluating the degree of soil erosion control is very important for soil erosion control. In response to the problems existing in the current research on soil erosion control degree (SECD) methods, the maximum possible soil erosion modulus is introduced into the evaluation model. Thereby eliminating the uncertainty of the assumed state of the original model. This paper calculates the SECD for 2020 based on this method. The results show: (1) From the soil erosion modulus spatial distribution, it can be seen that the soil erosion intensity is stronger in the eastern and southwestern parts. (2) Based on the SECD spatial distribution, it was found that eastern and southwestern regions were well controlled. This is consistent with the actual situation. (3) As the slope increases, the area proportion of SECD within the 0.2-0.3 range in each slope zone shows an increasing trend, while that of SECD within the 0.5-1 range gradually decreases; (4) In the case of vegetation coverage greater than 80%, SECD is still concentrated between 0.3-0.5, which indicates that vegetation coverage is not the only factor affecting soil erosion control. This research provided a new perspective for the evaluation of SECD.

Soil erosion risk assessment enables the identification of areas requiring priority treatment and avoids wasting human and material resources. The factor scoring method used in existing studies has high subjectivity, and the method of expressing erosion risk according to the soil erosion intensity ignores the random nature of the occurrence of erosion; therefore, neither method accurately reflects the risk of soil erosion. In order to address this issue, this study proposes a soil erosion risk assessment method that integrates the outcome and the probability of occurrence of soil erosion by means of a probabilistic statistical model. Subsequently, experimental research is conducted in the Dali River Basin. On the basis of long time-series data, using mathematical statistics as a tool and drawing on the empirical frequency formula, the probabilistic statistical risk assessment model is combined with the Modified Universal Soil Loss Equation (RUSLE) model to account for the probability of regional soil erosion at different intensity levels in the long time-series, which is combined with the intensity of erosion to carry out soil erosion risk assessment. The results of our study show the following: (1) The central and southwestern regions of the Dali River Basin (DRB) present medium and high levels of soil erosion risk, with the proportion of low-risk areas increasing annually, accounting for 78.97% of the DRB in 2020, while extremely high-risk areas account for only 0.40% of the DRB. (2) The major components impacting soil erosion risk in the DRB, as revealed by the geodetector, are the normalized difference vegetation index (NDVI) and slope, where the interaction between the two dominated the spatial variation in soil erosion risk. (3) Comparing the soil erosion risk and its status in the coming years, the proposed assessment method based on the occurrence probability can reveal the future soil erosion risk better than the traditional assessment method.

Low-carbon travel assumes paramount significance in energy conservation and the establishment of an eco-friendly transportation ecosystem. This paper endeavors to explore the relationship between low-carbon travel intention, latent psychological variables, and sociodemographic attributes, drawing insights from responses of 602 residents in Hangzhou, China by structural equation model and multi-group model. In particular, we synthesize the theory of planned behavior, value-belief-norm theory, and view of incentive, a reflection of the public support for incentive policies. Results reveals that the primary determinants influencing the low-carbon travel intention encompass the view of incentive, attitude, and subjective norms. Individuals with diverse sociodemographic attributes manifest varying sensitivities, with males and elders exhibiting heightened responsiveness to incentive, while the presence of children decrease the attraction of incentive. These findings demonstrate that low-carbon travel intention can be increased by three ways, one is by the strong attraction of incentive especially tailor incentive policy, another is by making family-friendly policies to facilitate travel for groups with children, and the last is by improving the quality of low-carbon travel services thus increasing the attitude and other determinants.

A social network personality prediction model with multi-channel information (MCIPP) is constructed. In the framework of in-depth learning, objective behavior data are used to automatically predict users’ personality traits, and whether users’ online behavior is consistent with their offline personality traits is analyzed. Specifically, the bi-directional long-term and short-term memory network (BiLSTM) and Attention mechanism (Attention) are used to capture the context semantic features of the text, and a syntactic dependency tree is constructed through a graph convolution network (GCN) to obtain a syntactic-based structural representation. Attention is integrated into a Topic Model to extract deep semantic information, and finally, the deep semantic information is input into a Softmax layer to obtain the personality tendency of a user Weibo. The results show that the MCIPP model has a good prediction effect with the highest accuracy of 0.806 4. There is a significant positive correlation between online and offline corresponding dimensions of individuals. Therefore, this model can be used to conduct psychological modeling on user network data, so that theory-driven psychological scientific research can objectively interpret individual psychology and behavior.

The recombination of charges and thermal excitation of carriers at the interface between methylammonium lead iodide perovskite (PVK) and the carbon electrode are crucial factors that affect the optoelectronic performance of carbon-based hole transport layer (HTL)-free perovskite photodetectors. In this work, a method was employed to introduce SnS quantum dots (QDs) on the back surface of perovskite, which passivated the defect states on the back surface of perovskite and addressed the energy-level mismatch issue between perovskite and carbon electrode. Performance testing of the QDs and the photodetector revealed that SnS QDs possess energy-level structures that are well matched with perovskite and have high absorption coefficients. The incorporation of these QDs into the interface layer effectively suppresses the dark current of the photodetector and greatly enhances the utilization of incident light. The experimental results demonstrate that the introduction of SnS QDs reduces the dark current by an order of magnitude compared to the pristine device at 0 V bias and increases the responsivity by 10%. The optimized photodetector exhibits a wide spectral response range (350 nm to 750 nm), high responsivity (0.32 A/W at 500 nm), and high specific detectivity (>1 × 1012 Jones).

Abstract Photodetectors (PDs) suffer from dark current due to defects in the perovskite photosensitive layer. Contact between the photosensitive layer and carbon electrodes could result in recombination of carriers at the interface. In this work, CsPbI3 quantum dots (QDs) were added between the photosensitive layer and the carbon electrode as the interfacial layer to passivate the surface defects of perovskite layer and improve the energy level matching at the interface. The effect of QDs concentrations on the passivation of the perovskite layer was investigated. It was found that the photoluminescence intensity of perovskite films was the strongest and the decay lifetime was the longest when the QDs concentration was 3 mg/mL. Owing to QDs passivation, the dark current of perovskite PD decreased by 94% from $$2.04\; \times \;10^{ - 9}$$ 2.04 × 10 - 9 to $$1.17\; \times \;10^{ - 10}$$ 1.17 × 10 - 10 A. The responsivity (R) at 605 nm improved by 27% from 0.29 to 0.37 A/W at 0 V bias voltage. The specific detectivity (D*) increased by 420% from $$8.9\; \times \;10^{11}$$ 8.9 × 10 11 to $$4.7\; \times \;10^{12}$$ 4.7 × 10 12 Jones.

Abstract In this work, inorganic tin-doped perovskite quantum dots (PQDs) are incorporated into carbon-based perovskite solar cells (PSCs) to improve their photovoltaic performance. On the one hand, by controlling the content of Sn2+ doping, the energy level of the tin-doped PQDs can be adjusted, to realize optimized band alignment and enhanced separation of photogenerated electron–hole pairs. On the other hand, the incorporation of tin-doped PQDs provided with a relatively high acceptor concentration due to the self-p-type doping effect is able to reduce the width of the depletion region near the back surface of the perovskite, thereby enhancing the hole extraction. Particularly, after the addition of CsSn0.2Pb0.8I3 quantum dots (QDs), improvement of the power conversion efficiency (PCE) from 12.80 to 14.22% can be obtained, in comparison with the pristine device. Moreover, the experimental results are analyzed through the simulation of the one-dimensional perovskite/tin-doped PQDs heterojunction.

Objective This study aimed to explore the potential mechanism of MYC proto-oncogene, BHLH Transcription Factor (MYC) gene, on sepsis. Materials And Methods In this experimental study, rat-derived H9C2 cardiomyocyte cells were cultured in vitro, followed by lipopolysaccharide (LPS) treatment with different concentration gradients. The cholecystokinin octapeptide (CCK-8) assay, enzyme-linked immunoassay (ELISA) assay, quantitative reverse transcription polymerase chain reaction (qRT-PCR), cell transfection, Western blot and flow cytometry were used to observe the cellular apoptosis and proliferation of cells in both treated LPS groups and normal control group. Results The result of CCK-8 assay showed that silencing MYC inhibited cellular proliferation of sepsis in absence or presence of LPS treatment. ELISA assay showed that the expressions of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were decreased in MYC silenced group, but they were increased after LPS treatment. Moreover, Flow cytometry assay showed that MYC silencing contributed to the apoptosis of sepsis cells. Furthermore, the expression of inflammatory factors showed that MYC silencing elevated the expression of inflammation factors. Conclusion MYC might take part in the process of LPS induced sepsis through suppressing apoptosis and inducing cell proliferation. Moreover, MYC might reduce inflammation during the progression of LPS induced sepsis.

Abstract Background/Objectives Oral maintenance chemotherapy can effectively prolong overall survival (OS) in many types of metastatic cancer, but its role in metastatic nasopharyngeal carcinoma (mNPC) is unclear. In this study, the efficacy of oral maintenance chemotherapy in mNPC and the effectiveness of circulating tumor EBV‐DNA for screening patients were evaluated. Methods Between June 2016 and December 2017, 141 patients with mNPC who received platinum‐based systemic chemotherapy were included (median follow‐up time, 21 months). Patients were classified into two groups according to the administration of oral maintenance chemotherapy. Plasma samples were collected before, during, and after treatment for the measurement of circulating EBV DNA. Results The 2‐year OS was higher for patients who received maintenance chemotherapy than for patients without maintenance chemotherapy (78.9% vs 62.7%, P = .016). Patients with undetectable posttreatment EBV‐DNA after 4‐6 cycles of systemic chemotherapy (n = 73) had a higher 2‐year OS than that of patients with detectable EBV‐DNA (n = 68) (82.16% vs 51.45%, P = .001). For patients with undetectable posttreatment EBV‐DNA, OS was better for those with maintenance chemotherapy than for those without (86.7% vs 73%, P = .027). For patients with detectable posttreatment EBV‐DNA, maintenance chemotherapy did not improve outcomes (49.5% vs 55.4%, P = .824). The most common acute events were hematological toxicity, and all were tolerable and curable. Conclusions Oral maintenance chemotherapy with S1 or capecitabine can improve OS in mNPC. Posttreatment EBV‐DNA was not only an independent prognosis factor for mNPC but also can screen out beneficiaries of maintenance chemotherapy.

Travel time is inherently uncertain in urban networks due to volatile traffic flows, signal controls, bus stops and disturbances from pedestrians. An effective way to characterize such uncertainty is by estimating Travel Time Distribution (TTD). However, conventional TTD models are lack of considering the interactions between turning movements within the link. Whereas the phenomenon of the Channelization Section Spillover (CSS) is very common, leading to strong interactions between turning movements. In this study, by incorporating the correlation of turning movements in the TTD model, that is, considering CSS, copula-based link-level and path-level TTD models are built. First, based on the empirical data, the correlations of turning movements are analyzed, and then the applicability of the various copula models are examined. The marginal distribution of each turning movement is described using parametric and non-parametric regression analysis, respectively. Then, the best-fitting copula is determined based on correlation parameters and the goodness-of-fit tests. As a case study, the chosen model is applied in estimating link-level and path-level TTD in an arterial road in Hangzhou, China, and compared with the model that did not consider the CSS before. Both results indicate that the copula-based approach can precisely capture the positively correlated relationship between turning movements during peak hours. Furthermore, higher TTD estimation accuracy demonstrates the significance to consider CSS, particularly in peak hours.

Car sharing has become a new mode of transport during the past two decades in the world. Its rapid growth in China has attracted a wide range of users and posed some problems. The main focus is on service efficiency and user satisfaction. To explore possible service enhancement and management intervention, this study aims at capturing the user characteristics according to different user types and scrutinizing their satisfaction with station-based one-way car sharing service. The study firstly illustrates descriptive statistics of user profile. This is followed by a study of user satisfaction influenced by user rates on staffs, the efficiency of rental process, vehicle situation, the use of credit card and their familiarity towards rental station. Furthermore, by clustering users according to the total travel time and distance during one rent, two different types of users are identified and defined as User Group A (UGA) and User Group B (UGB). To examine how fully do users utilize the shared cars, ANOVA was conducted implying family car ownership, total travel distance and main travel purpose have strong impact on total rental time for UGB, while for UGA, travel purpose and age have strong impact. Finally, ordinal logistic regression was introduced to find that for UGB, "shopping" is the main travel purpose with longer rental time, whereas for UGA, "out for business", "shopping", "visit friends" or "pick up others" are the main travel purposes with longer total travel time. Based on the findings, advices for operators on how to improve service quality and suggestions for government management strategy are discussed, respectively.

Perovskite solar cells (PSCs) have been developing rapidly since they were discovered, and their excellent photoelectric properties have attracted wide attention from researchers. The compact layer is an important part of PSCs, which can transport electrons and block holes. SnO2 is an excellent and commonly used electron transport layer (ETL) material, and doping modification is an effective way to improve performance. Here, Zn with a similar radius to Sn has been introduced to the doping of the SnO2 compact layer to achieve the purposes of conductivity enhancement of the compact layer and followed photoelectric performance improvement of the device. Zn-SnO2 compact layers with different doping concentrations were prepared and applied to mesoporous architecture PSCs. When the doping content was 5%, the power conversion efficiency (PCE) of the device based on the Zn-SnO2 compact layer has increased from 9.08% to 10.21%, with an increase of 12.44%. The doping of SnO2 promotes its application in low-cost PSCs.

Abstract In this study, Ag nanoparticles with diverse particle size and concentration, fabricated via the polyol method, were embedded in a TiO2 compact film to improve the power conversion efficiency of perovskite solar cells. Obtained results showed that Ag nanoparticles embedded in the TiO2 compact film do not affect the crystal structure of TiO2, while the size of the Ag nanoparticles can strongly influence the light absorption capacity of perovskite materials. However, the absorption intensity and power conversion efficiency of perovskite cells decreased with the increase in size of Ag nanoparticles. The amount of Ag nanoparticles was also an important factor for the performance of perovskite solar cells, and Ag nanoparticles in the compact layer were optimized to measure 10 nm in diameter, being embedded at a molar ratio of 1.5% (Ag:Ti = 1.5 mol%). Compared with hole-conductor-free perovskite solar cells that use carbon as counter electrodes, without Ag nanoparticles incorporated in the compact film, the enhanced efficiency of cells developed in this study can be mainly ascribed to the accelerated charge transfer, decreased charge recombination, and enhanced light absorption of the perovskite material in the visible region.

Background: Therapeutic applications of stem cells, especially mesenchymal stem cells, were once regarded as a promising therapy for mitigating acute cerebral infarction. Unfortunately, all the stem cell clinical trials have been futile. A new stroke therapeutic strategy of combining stem cells with nanotechnology has recently gained significant attention. The objective of this study was to evaluate the application of cerium oxide nanoparticle (nanoceria)-labeled human umbilical cord mesenchymal stem cells (HucMSCs) for stroke therapy. Methods: In our study, cerium oxide nanoparticles were precovered with hyaluronic acid before labeling HucMSCs and the synergistic effects from both HucMSCs and cerium oxide nanoparticles were analyzed in in vivo and in vitro experiments Results: The nanoceria-labeled HucMSCs combined advantages from both sides, including the capacity for inflammatory modulation of HucMSCs and the antioxidant effects of nanoceria. Compared with either HucMSCs or nanoceria individually, nanoceria-labeled HucMSCs exerted significantly enhanced capacities after gaining combined antioxidant and anti-inflammatory effects. Conclusion: Our findings suggest a novel strategy with effective and well-tolerated applications of stem cells for acute cerebral infarction therapy after modification of cells with nanomaterials.

In this paper, Ag@SiO2 core-shell nanoparticles (NPs) with different shell thicknesses were prepared experimentally and introduced into the photosensitive layer of mesoscopic hole-conductor-free perovskite solar cells (PSCs) based on carbon counter electrodes. By combining simulation and experiments, the influences of different shell thickness Ag@SiO2 core-shell nanoparticles on the photoelectric properties of the PSCs were studied. The results show that, when the shell thickness of 0.1 wt% Ag@SiO2 core-shell nanoparticles is 5 nm, power conversion efficiency is improved from 13.13% to 15.25%, achieving a 16% enhancement. Through the measurement of the relevant parameters of the obtained perovskite film, we found that this gain not only comes from the increase in current density that scholars generally think, but also comes from the improvement of the film quality. Like current gain, this gain is related to the different shell thickness of Ag@SiO2 core-shell nanoparticles. Our research provides a new direction for studying the influence mechanism of Ag@SiO2 core-shell nanoparticles in perovskite solar cells.

PAL, UFGT as structural genes and MYB10 as a regulatory gene play an important role in the accumulation of anthocyanins in plants. In this experiment, ‘Cuihongli’ and ‘Qiangcuili’ were used as materials to clone PAL, UFGT and MYB10 related to anthocyanin synthesis by homologous sequence cloning. The results showed that the full length of PAL was 2160 bp, encoding 719 amino acids; the full length of UFGT was 1428 bp, encoding 475 amino acids, and the differences of PAL and UFGT between the two cultivars were one amino acid and three amino acids, respectively. The sequence length of ‘Cuihongli’ MYB10 gene is 732 bp, which encodes 243 amino acids and belongs to the superfamily of SANT family. Homology analysis and phylogenetic tree analysis showed that the proteins encoded by PAL, UFGT and MYB10 genes were short and closely related to Prunus persica, Prunus avium, Prunus armeniaca and Prunus cerasifera in Rosaceae.

MYB10, bHLH and WD40 are correlative regulatory factors of anthocyanin. In this paper, the expression of MYB10, bHLH and WD40 in the peel and the pulp of two plum cultivars were analyzed with the materials of ‘Cuihongli’ and ‘Qiangcuili’. The results showed that, with the development of the fruit of ‘Cuihongli’, the expression of MYB10 in the peel was downregulated firstly then upregulated, and the expression of MYB10 was not detected in the flesh of ‘Cuihongli’s pulp and the peel and the pulp of the ‘Qiangcuili’. The expression of bHLH and WD40 showed that the they had no significant correlation with the accumulation of anthocyanin in the two cultivars. It is suggested that the deletion of MYB10 gene expression result in the lack of anthocyanin in the flesh of ‘Qiangcuili’ .

Au@Pt@Au core-shell nanoparticles, synthesized through chemical reduction, are utilized to improve the photoelectric performance of perovskite solar cells (PSCs) in which carbon films are used as the counter electrode, and the hole-transporting layer is not used. After a series of experiments, these Au@Pt@Au core-shell nanoparticles are optimized and demonstrate outstanding optical and electrical properties due to their local surface plasmon resonance and scattering effects. PSC devices containing 1 wt.% Au@Pt@Au core-shell nanoparticles have the highest efficiency; this is attributable to their significant light trapping and utilization capabilities, which are the result of the distinctive structure of the nanoparticles. The power conversion efficiency of PSCs, with an optimal content of plasmonic nanoparticles (1 wt.%), increased 8.1%, compared to normal PSCs, which was from 12.4% to 13.4%; their short-circuit current density also increased by 5.4%, from 20.5 mA·cm−2 to 21.6 mA·cm−2. The open-circuit voltages remaining are essentially unchanged. When the number of Au@Pt@Au core-shell nanoparticles in the mesoporous TiO2 layer increases, the photovoltaic parameters of the former shows a downward trend due to the recombination of electrons and holes, as well as the decrease in electron transporting pathways.

A Ag-doped double-layer composite film with TiO2 nanoparticles (P25) as the underlayer and TiO2 nanotube (TNT) arrays with the Ag-doped nanoparticles as the overlayer was fabricated as the photoanode in dye-sensitized solar cells (DSSCs). Five different concentrations of Ag-doped TNT arrays photoelectrode were compared with the pure TNT arrays composite photoelectrode. It is found that the photoelectric conversion efficiency of the TNT arrays composite photoanode is gradually improved from 3.00% of the pure TNT arrays composite photoanode to 6.12% of the Ag-doped TNT arrays photoanode with the increasing of the doping concentration, reaching up to the maximum in the 0.04 mol/L AgNO3 solution, and then slightly decreased to 5.43% after continuing to increase the doping concentration. The reason is mainly that the cluster structure of the Ag nanoparticles with large surface area contributes to dye adsorption and the Surface Plasmon Resonance Effect of the Ag nanoparticles improved the photocatalytic ability of the TNT arrays film.

Traffic congestion varies spatially and temporally. The observation of the formation, propagation and dispersion of network traffic congestion can lead to insights about the network performance, the bottleneck dynamics etc. While many researchers use the traffic flow data to reconstruct the congestion profile, the data missing problem is bypassed. Current methods either omit the missing data or supplement the missing part by average etc. Great error may be introduced during these processes. Rather than simply discarding the missing data, this research regards the data missing event as a result of either the severe congestion which prevent the floating vehicle from entering the congested area, or a type of feature of the resulting traffic flow time series. Hence a new traffic flow operational index time series similarity measurement is expected to be established as a basis of identifying the dynamic network bottleneck. The method first measures the traffic flow operational similarity between pairs of neighboring links, and then the similarity results are used to cluster the spatial-temporal congestion. In order to get the similarity under missing data condition, the measurement is implemented in a two-stage manner: firstly the so called first order similarity is calculated given that the traffic flow variables are bounded both upside and downside; then the first order similarity is aggregated to generate the second order similarity as the output. We implement the method on part of the real-world road network; the results generated are not only consistent with empirical observation, but also provide useful insights.

Highly ordered TiO2 nanotube arrays (TNT arrays) were fabricated by two-step anodization process. In order to further improve the performance of DSSCs, TNT arrays were optimized by changing the anodization conditions to meet the requirements of high-performance photoanode. The photoelectric conversion properties of DSSCs based on P25/TNT arrays double-layer film with different anodization time were investigated and compared. The results show that the conversion efficiency of 4.20% was achieved in double-layer photoanode at 18 h, with an open-circuit voltage (Voc) of 0.65 V and short-circuit current density (Jsc) of 9.98 mA cm−2.

In this study, Ag@SiO2 nanoparticles were synthesized by a modified Stöber method for preparing the TiO2 mesoporous layer of carbon counter electrode-based perovskite solar cells (PSCs) without a hole transporting layer. Compared with normal PSCs (without Ag@SiO2 incorporated in the TiO2 mesoporous layer), PSCs with an optimal content of Ag@SiO2 (0.3 wt. % Ag@SiO2-TiO2) show a 19.46% increase in their power conversion efficiency, from 12.23% to 14.61%, which is mainly attributed to the 13.89% enhancement of the short-circuit current density, from 20.23 mA/cm2 to 23.04 mA/cm2. These enhancements mainly contributed to the localized surface Plasmon resonance effect and the strong scattering effect of Ag@SiO2 nanoparticles. However, increasing the Ag@SiO2 concentration in the mesoporous layer past the optimum level cannot further increase the short-circuit current density and incident photon-to-electron conversion efficiency of the devices, which is primarily ascribed to the electron transport pathways being impeded by the insulating silica shells inside the TiO2 network.

AbstractObjective: Todetect the abnormal distribution of B-lymphocytes between peripheral and bone marrow (BM) compartments and explore the mechanism of abnormal chemotaxis of B-lymphocytes in lupus subjects. Methods: The proportions of CXC chemokine receptor (CXCR)4+ B cells and CFDA-labeled MRL/lpr-derived B cells were detected by flow cytometry. The levels of CXC chemokine ligand (CXCL)12in peripheral blood (PB)were measured by ELISA. The migrated B cells to osteoblasts (OBs) was measured by transwell migration assay. The relative spatial position of B cells, OBs and CXCL12 was presented by Immunofluorescence assay. Results: Firstly, we found that the percentage of CXCR4+ B cells was lower in PB and higher in the BM from both MRL/lpr mice and patientswith Systemic lupus erythematosus (SLE). Secondly, OBs from MRL/lpr mice produced more CXCL12 than that from C57BL/6 mice. Besides, MRL/lpr-derived OBs demonstrated more potent chemotactic ability toward B-lymphocytes than control OBs by vitro an vivo. Additionally, more B-lymphocytes were found to co-localize with OBs within the periosteal zone of bone in MRL/lpr mice. Lastly, the percentages of CXCR4+B cells were found to be negatively correlated with serum Immunoglobulin (Ig) G concentration, moreover, BM CXCL12 levels were found to be positively correlated with SLE disease activity index Score and negatively correlated with serum Complement3 (C3) concentration. Conclusions: our results indicated that there is a shifted distribution of B-lymphocytes between BM and peripheral compartments in both SLE patients and MRL/lpr mice. Besides, the up-regulated levels of CXCL12 in OBs was indicated to contribute to the enhanced chemotactic migration and anchorage of B-lymphocytes to OBs.

Background Pseudomonas aeruginosa is a highly prevalent bacterial species known for its ability to cause various infections and its remarkable adaptability and biofilm-forming capabilities. In earlier work, we conducted research involving the screening of 33 metabolites obtained from a commercial source against two prevalent bacterial strains, Escherichia coli and Staphylococcus aureus. Through screening assays, we discovered a novel malic acid combination (MAC) consisting of malic acid, citric acid, glycine, and hippuric acid, which displayed significant inhibitory effects. However, the precise underlying mechanism and the potential impact of the MAC on bacterial biofilm formation remain unknown and warrant further investigation. Methods To determine the antibacterial effectiveness of the MAC against Pseudomonas aeruginosa, we conducted minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) assays. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) techniques were employed to observe bacterial morphology and biofilm formation. We further performed a biofilm inhibition assay to assess the effect of the MAC on biofilm formation. Whole-transcriptome sequencing and bioinformatics analysis were employed to elucidate the antibacterial mechanism of the MAC. Additionally, the expression levels of differentially expressed genes were validated using the real-time PCR approach. Results Our findings demonstrated the antibacterial activity of the MAC against P. aeruginosa. SEM analysis revealed that the MAC can induce morphological changes in bacterial cells. The biofilm assay showed that the MAC could reduce biofilm formation. Whole-transcriptome analysis revealed 1093 differentially expressed genes consisting of 659 upregulated genes and 434 downregulated genes, in response to the MAC treatment. Mechanistically, the MAC inhibited P. aeruginosa growth by targeting metabolic processes, secretion system, signal transduction, and cell membrane functions, thereby potentially compromising the survival of this human pathogen. This study provides valuable insights into the antibacterial and antibiofilm activities of the MAC, a synergistic and cost-effective malic acid combination, which holds promise as a potential therapeutic drug cocktail for treating human infectious diseases in the future.

Abstract Background and aim Autoantibody production are the main risk factors for inflammation of rheumatoid arthritis (RA). This study aimed to investigate differences in B lymphocyte subsets (native B, memory B, and plasmablasts) and several cytokines in RA patients and their correlation with the clinical parameters. Methods In total, 81 RA patients (active RA and inactive RA) and 40 healthy subjects were recruited between September 2018 and October 2020. The distribution of B lymphocyte subsets in peripheral blood samples was measured via flow cytometry and the plasma cytokines were detected by enzyme linked immunosorbent assay. The receiver operating characteristic curve (ROC) was used to evaluate the value of each index for RA diagnosis and activity prediction. Results The percentages of native B and memory B cells in RA patients did not differ significantly from the percentages of those in healthy controls. However, the percentage of plasmablasts in active RA patients was significantly higher compared with healthy subjects and inactive RA patients. The percentage of plasmablasts was significantly related to C reaction protein. ROC curve analysis showed that when the best cutoff value of plasmablasts/B cell was 1.08%, the area under the curve (AUC) for diagnosing RA was 0.831 (95% CI 0.748 ~ 0.915), the specificity was 91.4%, and the sensitivity was 67.5%. The AUC predicted by the combination of plasmablast and anti-CCP for active RA patients was 0.760, which was higher than that of plasmablast and anti-CCP. Conclusion In conclusion, the percentage of plasmablast varies among RA patients in different stages. The percentage of plasmablasts can be used as an early diagnosis marker for RA.
