Abstract The human yolk sac (hYS) is essential for embryo nutrient biosynthesis/transport and development. However, there lacks a comprehensive study of hYS nutrient-gene interactions. Here we performed a secondary analysis of hYS transcript profiles (n = 9 samples) to identify nutrient-sensitive hYS genes and regulatory networks, including those that associate with adverse perinatal phenotypes with embryonic origins. Overall, 14.8% highly expressed hYS genes are nutrient-sensitive; the most common nutrient cofactors for hYS genes are metals and B vitamins. Functional analysis of highly expressed hYS genes reveals that nutrient-sensitive hYS genes are more likely to be involved in metabolic functions than hYS genes that are not nutrient-sensitive. Through nutrient-sensitive gene network analysis, we find that four nutrient-sensitive transcription regulators in the hYS (with zinc and/or magnesium cofactors) are predicted to collectively regulate 30.9% of highly expressed hYS genes. Lastly, we identify 117 nutrient-sensitive hYS genes that associate with an adverse perinatal outcome with embryonic origins. Among these, the greatest number of nutrient-sensitive hYS genes are linked to congenital heart defects (n = 54 genes), followed by microcephaly (n = 37). Collectively, our study characterises nutrient-sensitive hYS functions and improves understanding of the ways in which nutrient-gene interactions in the hYS may influence both typical and pathological development.

IntroductionThere is a need to rebuild soil health by implementing regenerative agricultural practices across cropping systems dependent on agrochemicals, such as in United States corn production. One increasingly utilized regenerative practice is the application of biostimulants, or microbial inoculants that can rebuild soil health and productivity. In this study, we describe results from a multi-year corn trial conducted to quantify the impact of Re-Gen, a biostimulant invented to recycle plant biomass and increase nutrient bioavailability in the soil.MethodsRe-Gen was applied across four fields (VA01, BH01, VA04, and LA05) at a dairy cattle farm in Ferrisburgh, VT.ResultsOver the two consecutive years (2022-2023) of the trial, Re-Gen application on corn stover and cover crop residues increased ear corn yield by 24% and corn silage yield by 12.5-30%, depending on the field. Analysis of soil nutrients and plant tissues showed increased nutrients, particularly in field LA-05. Multi-year Re-Gen application increased yield and generated additional economic value, indicating that the effects of Re-Gen do not diminish with multiple applications. The increased corn silage yield correlated to increased potential milk yield from cows fed silage grown in Re-Gen-treated fields. Further investigation into the mechanism suggests that increased phosphatase production Re-Gen could contribute to increased phosphorus bioavailability in the soil and uptake in the tissue, potentially increasing yields.DiscussionThese results highlight the potential for Re-Gen to foster regenerative agriculture processes on cropland and livestock farms while also increasing corn and milk yield and, therefore, revenue for corn farmers in the United States and for similar cropping systems worldwide.

The biofortification of edible crops with selenium (Se) is a common and effective strategy to address inadequate Se intake, which is suffered by millions of people worldwide. However, there is little information regarding the effects of this practice on crops belonging to the important Brassica family. To evaluate the efficacy of foliar Se application on broccoli, four treatments with varying Se concentrations were tested: 0%, 0.05%, 0.10%, and 0.15% (w/v), applied as sodium selenate during the early flowering stage. Although no overall effects on growth and biomass parameters were observed, the results indicate that the lowest Se dose (0.05-Se) was sufficient to notably increase Se concentration in the florets, even after boiling. Based on the increase to 14.2 mg Se kg−1 of dry matter in this broccoli fraction, it was estimated that consuming a 100-gram portion of boiled florets biofortified with 0.05% Se would provide approximately 140 µg of Se, which could be sufficient to potentially improve human selenium status, as previously documented. Moreover, the results obtained underscore how the application of this small dose was also adequate to reduce phytate concentration in the florets and to increase antioxidant and polyphenol concentrations, thereby improving the concentration and bioavailability of other essential nutrients, including Ca, Mg, Fe, and Zn, along with improving its quality as an antioxidant food.

Point- and non-point source nutrients are likely to have different ecological impacts in receiving waters, due to differences in the concentration and proportions of nutrient fractions. However, the direct comparison of their ecological impacts in receiving waters has barely been quantified. We undertook algal bioassays with algal communities from river sites and showed that there was a photosynthetic yield (Fv/Fm) response to nutrient enrichment when river nutrient concentrations were relatively low, but not at higher nutrient concentrations. To combat this variability in the photosynthetic state of algae, we developed a standardized algal bioassay (3-day), using a cultured species of algae which was starved of nitrogen, to compare the photosynthetic response to three nitrogen sources: treated wastewater, aquaculture farm discharges, and soil erosion-derived nutrient sources. This study showed that the nutrient parameter that had the highest correlation with algal photosynthetic response was total dissolved nitrogen (TDN), i.e., the sum of dissolved inorganic and organic nitrogen, rather than dissolved inorganic nitrogen alone. This was true across all three nutrient sources (R2 = 0.58–0.78). Additionally, the same concentrations of TDN from soil erosion-derived and aquaculture samples resulted in a significantly higher algal photosynthetic response, compared to the treated wastewater. This indicates that TDN from soils and aquaculture farms was significantly more bioavailable to the cultured algae than treated wastewater. When a range of parameters were correlated with algal responses, organic and inorganic nutrients, and organic carbon were the parameters that had the highest explanatory power for soil erosion-derived and aquaculture samples (R2 = 0.75–0.87). The importance of organic compounds in these equations points to the potential of microbial transformation of organic nutrients into more bioavailable forms during the 3-day bioassay. This highlights the need to understand the relationship between algal and microbial communities in natural systems for nutrient source impact assessment. This study provides an improved understanding and metrics for comparing the algal growth response to different nutrient sources.

This study aimed to verify the potential of calcium lactate for the performance, blood index, shinbone quality, jejunal immunity, and egg quality of aged laying hens. A total of 360 62-week-old Hy-Line Brown laying hens were randomly divided into four treatments, with six replicates and 15 chickens per replicate. Experimental groups were fed with 0.25%, 0.5%, and 1.0% calcium lactate to substitute limestone in the control group (maintaining the same amount of calcium). The feeding trial lasted for 12 weeks. The laying rate and daily egg mass of laying hens fed the diets supplemented with calcium lactate was increased relative to those of the control group. The dietary addition of calcium lactate for laying hens enhanced the eggshell ratio, eggshell thickness, eggshell strength, and albumen height of eggs, and the addition level of 0.5% had the best effect. Dietary calcium lactate increased the number of red blood cells, corpuscular hemoglobin, mononuclear leucocytes and basophilic granulocytes, and decreased heterophils in the blood of laying hens. The activities of serum alanine transaminase and creatine kinase in laying hens was reduced by the dietary addition of calcium lactate. Calcium lactate supplementation in diets increased the serum calcium and phosphorus contents of laying hens. The dietary inclusion of calcium lactate increased the contents of IgA, IgG, lysozyme, and sIgA in the jejunal mucosa, and the 0.5% addition level worked best, but the IL-2 content decreased. The addition of 0.5% calcium lactate to the diet reduced the maximal force of the shinbone and increased the work required for shinbone rupture in laying hens. In conclusion, the dietary addition of calcium lactate improved the performance and egg quality of laying hens, probably by its positive effects on body health, intestinal digestible ability, calcium bioavailability, and jejunal mucosal immunity. The optimum amount of calcium lactate in the diet of laying hens is recommended to be 0.5%.

Raw bars have become popular among health-conscious consumers due to their nutrient-dense ingredients and lack of additives and preservatives. However, the effect of simulated gastrointestinal digestion on the nutrient content of these bars has yet to be extensively studied. In this study, four different raw bar recipes were subjected to simulated gastrointestinal digestion to evaluate the impact on their nutrient content. The recipes have dates and almond flour as base ingredients and specific ingredients such as Maca root powder, Ginger powder, Aronia powder, Pollen, Propolis extract, Astragalus powder, and Cacao powder. These variations were intended to provide diverse flavors and potential health benefits to cater to different preferences and needs. The in vitro digestion model was designed to mimic the conditions of the human gastrointestinal tract, including the mouth, stomach, and small intestine. The results showed that the simulated gastrointestinal digestion significantly impacted the nutrient content of the bars, with varying degrees of nutrient loss observed depending on the recipe. The highest phenolic content and antioxidant activity were observed in the salivary phase for all samples. Vitamin B content generally decreases from the salivary to the intestinal stage. After digestion, the recovery rates of total phenols, antioxidant capacity, and vitamins B1, B3, and B6 varied across the recipes. The recovery rates of vitamins B1, B3, and B6 were generally high across all recipes, indicating their stability and retention during digestion. The findings suggest that simulated GI digestion provides insights into the nutrient bioavailability of raw bars. These results can inform the formulation and optimization of raw bars to enhance nutrient absorption and nutritional value. Further research is warranted to investigate the effects of different processing techniques and ingredient combinations on nutrient bioavailability.

The present study evaluated the effects of butyric acid supplementation and Saccharomyces boulardii (alone or in combination) on growth performance, nutrient digestibility, bone mineralization, and blood hormones of male broiler chickens fed a diet including reduced levels of nonphytate phosphorus (NPP). The chickens were allocated to 6 dietary treatments: 1) positive control diet with adequate amounts of NPP (PC; 0.48, 0.43, and 0.39% in the starter, grower, and finisher period, respectively); 2) negative control diet with low amounts of NPP (NC; 0.38, 0.33, and 0.29% in the starter, grower, and finisher period, respectively); 3) NC plus 500 FTU/kg microbial phytase (PHY); 4) NC plus 0.2% butyric acid (BA); 5) NC plus 1 × 108 cfu/kg S. boulardii (SB); 6) NC plus butyric acid and S. boulardii (BA+SB). Nannochloropsis gaditana is a microalga with interesting nutritional and functional value due to its high content of protein, polyunsaturated fatty acids, and bioactive compounds. However, the hardness of its cell wall prevents accessibility to these components. This work aimed to study the effect of a treatment to increase the fragility of the cell wall on the bioavailability of its nutrients and functional compounds. The antioxidant and antiproliferative capacity of functional extracts from treated and untreated N. gaditana was assessed, and the profile of bioactive compounds was characterized. Furthermore, to study the effect of treatment on its nutrient availability and functional capacity, an in vivo experiment was carried out using a rat experimental model and a 20% dietary inclusion level of microalgae. Functional extracts from treated N. gaditana exhibited higher antioxidant activity than the untreated control. Furthermore, the treated microalga induced hypoglycemic action, higher nitrogen digestibility, and increased hepatic antioxidant activity. In conclusion, N. gaditana has interesting hepatoprotective, antioxidant, and anti-inflammatory potential, thus proving itself an ideal functional food candidate, especially if the microalga is treated to increase the fragility of its cell wall before consumption.

The health of the majority of South Africa’s population is seriously threatened by hunger and micronutrient deficiency, with impaired immune response a real threat, which the current SARSCoV-2 virus pandemic has highlighted. Traditional household food-processing techniques can, amongst other advantages, increase nutrient bioavailability in affordable staple foods and hence provide a way, in part, to alleviate malnutrition for food-insecure communities. In this way, immune defence and pathogen resilience of the food insecure could be enhanced so that they can better survive both COVID-19 and future threats.

Fermented food products, especially those derived from cereals and legumes are important contributors to diet diversity globally. These food items are vital to food security and significantly contribute to nutrition. Fermentation is a process that desirably modifies food constituents by increasing the palatability, organoleptic properties, bioavailability and alters nutritional constituents. This review focuses on deciphering possible mechanisms involved in the modification of nutritional constituents as well as nutrient bioavailability during the fermentation of cereals and legumes, especially those commonly consumed in developing countries. Although modifications in these constituents are dependent on inherent and available nutrients in the starting raw material, it was generally observed that fermentation increased these nutritive qualities (protein, amino acids, vitamins, fats, fatty acids, etc.) in cereals and legumes, while in a few instances, a reduction in these constituents was noted. A general reduction trend in antinutritional factors was also observed with a corresponding increase in the nutrient bioavailability and bioaccessibility. Notable mechanisms of modification include transamination or the synthesis of new compounds during the fermentation process, use of nutrients as energy sources, as well as the metabolic activity of microorganisms leading to a degradation or increase in the level of some constituents. A number of fermented products are yet to be studied and fully understood. Further research into these food products using both conventional and modern techniques are still required to provide insights into these important food groups, as well as for an overall improved food quality, enhanced nutrition and health, as well as other associated socioeconomic benefits.

The mining of oil sands in Alberta, Canada is a large-scale disturbance that requires land reclamation to equivalent land capability. Cover soils used for upland forest reclamation are forest floor mineral-mix (FFM) sourced from upland forest ecosystems and peat mineral-mix (PM) sourced from lowland ecosystems. Spatial heterogeneity and quantity of soil resources, especially nutrient bioavailability, is important because it affects the establishment of native vegetation in forest ecosystems and soil respiration is an indicator for overall soil biologic activity. We studied spatial heterogeneity of soil nutrients as well as seasonal and spatial patterns of soil respiration in two sites reclaimed either with FFM or PM and two reference sites recovering either from harvest or fire. Contrary to our initial hypothesis, we identified spatial heterogeneity in some bioavailable nutrients in FFM and PM cover soil, indicating that standard placement processes can recreate spatial heterogeneity. However, some nutrients such as P, K and S showed no heterogeneity in PM at all. P and K availability was significantly lower and S was significantly higher in PM than in FFM and reference sites. Seasonal pattern of respiration showed variability on natural reference sites and on FFM, indicating that disturbance had not removed belowground function completely. PM reclaimed sites showed no strong seasonal respiration patterns indicating homogeneous belowground function. Surprisingly PM treatments were not characterized by highest rates of soil respiration, while having highest amounts of total organic carbon. We conclude that FFM reclaimed sites may be more successfully reclaimed than PM sites because they are more similar to reference sites in terms of nutrient status and seasonal respiration patterns.
