Abstract In China, according to the ‘Technical Operating Procedures for Blood Stations (2019 Edition),’ blood stations are authorized to utilize Chemiluminescence Immunoassay (CLIA) to detect pathogen markers linked with transfusion-transmissible infections. However, currently, there is no approved CLIA reagent for the screening of blood-borne diseases in China, specifically for the detection of Hepatitis B surface antigen. The objective of this research project is to conduct a comprehensive evaluation of the performance of the Wantai Chemiluminescent Microparticle Hepatitis B surface antigen reagent. This study evaluates the performance of the Wantai Chemiluminescent Microparticle Immunoassay (CMIA) on the Wan200 + analyzer in screening for Hepatitis B Surface Antigen (HBsAg) in blood samples. The clinical trial component of this evaluation is included as part of the documentation submitted to the National Medical Products Administration (NMPA) of China for the approval of blood screening reagents. The evaluation plan of this study encompasses two main components: clinical trials and performance assessment. We adopted a controlled trial design, utilizing the WanTai Chemiluminescent Microparticle Immunoassay (CMIA) on the Wan200 + analyzer and the Enzyme-Linked Immunosorbent Assay (ELISA) to screen for Hepatitis B Surface Antigen (HBsAg) in routine blood donor samples and reference serum panel samples. To ensure the accuracy of the screening, we additionally employed Abbott's ELISA reagents and HBV DNA for validation. The assessment primarily focused on key performance indicators such as sensitivity, specificity, and analytical sensitivity. Moreover, this clinical trial data has been included as part of the submission to China's National Medical Products Administration (NMPA). In the clinical trials of this study, a total of 10,470 blood donor samples underwent simultaneous testing using both CMIA and ELISA methods. Across two clinical trials, there was remarkable concordance between CMIA and the two ELISA reagents, with Kappa values exceeding 0.82. Among the 269 samples that were double-reactive in the enzyme immunoassay (ELISA) tests, CMIA exhibited a 100% reactivity detection rate. However, CMIA produced 14 and 6 false-positive results in the respective clinical trials, resulting in specificities of 99.73% and 99.89%. In contrast, the specificities for Wantai ELISA and Xin Chuang ELISA were both greater than 99.94%.When testing samples in the gray zone serum plates, CMIA's detection limit significantly exceeded that of the two ELISA assays. CMIA had a detection cutoff of 0.05 IU/mL, while the two ELISA reagents had cutoffs of 0.1 IU/mL and 0.09 IU/mL, respectively. CMIA's detection limits for the adr and adw subtypes were 0.05 IU/mL, and for the ay subtype, it was 0.1 U/mL. The detection limit for 10 HBV mutant samples was 0.5 U/mL. In 165 cases where ELISA tested negative but HBV DNA tested positive, CMIA detected 5 HBsAg-positive samples. This study evaluated the performance of the Wantai CMIA in screening for HBsAg among blood donors. The results demonstrate outstanding performance of CMIA in both clinical trials and performance assessments, detecting all true positive samples with a sensitivity of 100%. It exhibits excellent concordance with the two ELISA assays. Of particular note is its superiority in early detection of HBsAg in the screening of early-stage hepatitis B infections, reducing the window period compared to ELISA. CMIA achieves a specificity exceeding 99.73% for negative blood donors, aligning with the European Union's standards for blood screening assay specificity. In summary, Wantai's CMIA displays high sensitivity and specificity in blood donor screening, making it suitable for screening blood donors in China.

Abstract Background Metastasis is the primary cause of recurrence and death in patients with papillary thyroid carcinoma (PTC). LncRNA ACTA2-AS1, a long non-coding RNA, acts as a tumor suppressor in multiple types of human malignancies, while the role of ACTA2-AS1 in PTC metastasis remains unclear. Methods The ACTA2-AS1 expression in PTC tissues was analyzed. The sponged roles of ACTA2-AS1 via miR-4428/KLF9 axis were identified using starBase tool. The function of ACTA2-AS1 in PTC was performed with in vitro and in vivo experiments. The correlation between DNA methylation and mRNA expressions of these gene in the TCGA dataset was explored. Results ACTA2-AS1 expression was downregulated in PTC tissues without metastasis and further decreased in PTC tissues with lymph node metastasis compared with that in normal tissues. Functionally, the overexpression of ACTA2-AS1 inhibited the growth, proliferation, and invasion of PTC cells, whereas its depletion exerted opposite effect. In vivo, ACTA2-AS1 expression inhibited PTC metastasis. Furthermore, ACTA2-AS1 acted as a competing endogenous RNA for miR-4428, thereby positively regulating the expression of miR-4428 target gene, KLF9. Finally, miR-4428 overexpression enhanced invasive potential of PTC cells and significantly weakened the effects of ACTA2-AS1 on promotion and inhibition of KLF9 expression as well as invasive ability of PTC cells, respectively. In the TCGA dataset, the methylation level of ACTA2-AS1 was significantly correlated with its mRNA expression (r = 0.21, p = 2.1 × e−6). Conclusions Our findings demonstrate that ACTA2-AS1 functions as a tumor suppressor in PTC progression at least partly by regulating the miR-4428-dependent expression of KLF9. Graphical abstract

Floods are the most destructive, widespread, and frequent natural hazards. The extent of flood events is accelerating in the context of climate change, where flood management and disaster mitigation remain important long-term issues. Different studies have been utilizing data and images from various types of sensors for mapping, assessment, forecasting, early warning, rescue, and other disaster prevention and mitigation activities before, during, and after floods, including flash floods, coastal floods, and urban floods. These monitoring processes evolved from early ground-based observations relying on in situ sensors to high-precision, high-resolution, and high-coverage monitoring by airborne and remote sensing sensors. In this study, we have analyzed the different kinds of sensors from the literature review, case studies, and other methods to explore the development history of flood sensors and the driving role of floods in different countries. It is found that there is a trend towards the integration of flood sensors with artificial intelligence, and their state-of-the-art determines the effectiveness of local flood management to a large extent. This study helps to improve the efficiency of flood monitoring advancement and flood responses as it explores the different types of sensors and their effectiveness.

Timely identification of early internal short circuit faults, commonly referred to as micro short circuits (MSCs), is essential yet poses significant challenges for the safe and reliable operation of lithium-ion battery (LIB) energy storage systems. This paper introduces an innovative diagnostic method for early internal short circuits in LIB packs, utilizing dynamic time warping (DTW) applied to incremental capacity (IC). Initially, the terminal voltages of all cells within the LIB pack are ordered at any moment to determine the median terminal voltage, which is then used to generate the median IC curve. This curve acts as a reference benchmark that represents the condition of healthy cells in the pack. Subsequently, the DTW algorithm is utilized to measure the similarity between each cell’s IC curve and the median IC curve. Cells exhibiting similarity scores that exceed a specified threshold are identified as having MSC faults. Lastly, for the cells diagnosed with MSC conditions, a method for estimating short-circuit resistance (SR) based on variations in maximum charging voltage is devised to quantitatively evaluate the severity and evolution of the MSC. Experimental findings reveal that the proposed method effectively identifies MSC cells in the LIB pack and estimates their SRs without the necessity of a battery model, thereby affirming the method’s validity.

African swine fever virus (ASFV) is one of the most complex viruses. ASFV is a serious threat to the global swine industry because no commercial vaccines against this virus are currently available except in Vietnam. Moreover, ASFV is highly stable in the environment and can survive in water, feed, and aerosols for a long time. ASFV is transmitted through the digestive and respiratory tract. Mucosal immunity is the first line of defense against ASFV. Saccharomyces cerevisiae (SC), which has been certified by the U.S. Food and Drug Administration and has a generally recognized as safe status in the food industry, was used for oral immunization in this study. ASFV antigens were effectively expressed in recombinant SC strains with high DNA copy numbers and stable growth though surface display technology and chromosome engineering (δ-integration). The recombinant SC strains containing eight ASFV antigens—KP177R, E183L, E199L, CP204L, E248R, EP402R, B602L, and B646L— induced strong humoral and mucosal immune responses in mice. There was no antigenic competition, and these antigens induced Th1 and Th2 cellular immune responses. Therefore, the oral immunization strategy using recombinant SC strains containing multiple ASFV antigens demonstrate potential for future testing in swine, including challenge studies to evaluate its efficacy as a vaccine against ASFV.

Abstract MAVS is an adapter protein involved in RIG-I-like receptor (RLR) signaling in mitochondria, peroxisomes, and mitochondria-associated ER membranes (MAMs). However, the role of MAVS in glucose metabolism and RLR signaling cross-regulation and how these signaling pathways are coordinated among these organelles have not been defined. This study reports that RLR action drives a switch from glycolysis to the pentose phosphate pathway (PPP) and the hexosamine biosynthesis pathway (HBP) through MAVS. We show that peroxisomal MAVS is responsible for glucose flux shift into PPP and type III interferon (IFN) expression, whereas MAMs-located MAVS is responsible for glucose flux shift into HBP and type I IFN expression. Mechanistically, peroxisomal MAVS interacts with G6PD and the MAVS signalosome forms at peroxisomes by recruiting TNF receptor-associated factor 6 (TRAF6) and interferon regulatory factor 1 (IRF1). By contrast, MAMs-located MAVS interact with glutamine-fructose-6-phosphate transaminase, and the MAVS signalosome forms at MAMs by recruiting TRAF6 and TRAF2. Our findings suggest that MAVS mediates the interaction of RLR signaling and glucose metabolism.

The oligomerization of light olefins is an alternative for generating clean liquid fuels. In this study, the MCM-22 zeolite featuring a partially disordered layered structure with framework combined 10 membered ring (10 MR) was discussed for 1-hexene oligomerization. MCM-22 and Y zeolite were compared to investigate the influences of the framework and acidity strength on the activity and yield in the 1-hexene oligomerization. The results indicated that the MCM-22 zeolite was an efficient catalyst for oligomerization compared to Y zeolite. The MCM-22 zeolite was characterized in detail by NMR, Py-FTIR, and TG analyses. The catalytic performance and deactivation properties of MCM-22 zeolite in the oligomerization reaction of 1-hexene were investigated. Structure–activity relationships were established through the kinetic study. Although the density of strong acid sites was essential for the oligomerization of 1-hexene, the presence of low pressure was also necessary for the formation of dimers. Thus, MCM-22 zeolite demonstrates a substantial improvement in initial conversion and catalyst lifespan for 1-hexene oligomerization.

Epithelial ovarian cancer (EOC) is a fatal gynecological malignancy with limited therapeutic options. Previous research has demonstrated that Tripterygium glycosides (GTW) can enhance effectiveness of cisplatin (DDP) chemotherapy against EOC. However, the underlying mechanism of GTW alleviating EOC still remains unclear. In this article, an ID8 cell-derived xenograft mouse model was established to evaluate the anti-tumor efficacy of GTW combined with DDP. Consistent with previous findings, the results suggested that GTW combined with DDP can exhibit a stronger tumor suppressive effect than DDP alone. Additionally, GTW was found can further exert gastrointestinal protection against DDP by reducing pathological damage on colon tissue. Secondly, to verify whether gut microbiota play an instrumental role in GTW’s anticancer effect, we treated mice models with antibiotic to eliminate gut microbiota. And our experimental results indicated that all drug groups showed a weaker tumor suppressive effect and more severe gastrointestinal damage post antibiotic supplement. At genus level, the relative abundance of Lactobacillus was dramatically diminished by the antibiotic treatment, while combined treatment of GTW and DDP can significantly restore the level. Moreover, we performed Lactobacillus acidophilus transplantation and healthy mice fecal microbiota transplantation experiments to further investigate the link between the anticancer effect of GTW and gut microbiota. Our results suggested that both cisplatin-sensitizing and intestinal barrier-protecting effects of GTW can be recovered to a different extent. In conclusion, our results indicated that GTW is a promising chemosensitization and intestinal barrier repair drug for EOC, and the potential mechanism may corelate with the restoration of the compromised intestinal microbial balance.

Objective: This study aimed to assess the efficacy of individual-donation nucleic acid testing (ID-NAT) in detecting occult hepatitis B virus infection (OBI) among anti-HBc positive blood donors, compared to minipool nucleic acid testing (MP-NAT). Methods: The present study analyzed data from the Shandong Blood Center in China during the period from January 2018 to June 2022, where HBsAg-negative blood donors were screened using the 6-sample minipool nucleic acid testing (6-sample MP NAT) method. NAT-positive samples underwent subsequent anti-HBc and anti-HBs testing. Approximately 5000 samples that passed the nucleic acid mixing test were randomly selected for anti-HBc testing, and over 100 anti-HBc positive samples underwent individual donor nucleic acid testing (ID-NAT). Any HBV DNA positive samples detected by ID-NAT were subsequently confirmed using alternative nucleic acid testing methods. Results: Among 220,445 HBsAg-negative blood donors, the positivity rate of HBV DNA detection using the 6-sample minipool nucleic acid testing (MP NAT) method was found to be 0.031% (69/220,445). Of the 67 HBV DNA positive samples, 55 (82.09%) and 25 (37.31%) were found to be positive for anti-HBc and anti-HBs, respectively, using the supplementary chemiluminescent microparticle immunoassay (CMIA). Among the 4797 HBsAg-negative/MP NAT-negative samples, 909 (18.95%) tested positive for anti-HBc. Further NAT testing was performed on 164 arbitrarily selected anti-HBc-positive/MP HBV DNA-negative samples, revealing a HBV DNA positivity rate of 1.22% (2/164). Conclusion: Using individual donation nucleic acid testing can significantly increase the detection rate of occult hepatitis B virus infection in anti-HBc-positive blood donors, resulting in a detection rate of 0.22% (1.22 × 0.1895). This rate is 8.10 times higher than the detection rate achieved by mixed testing methods (0.031%) [calculated as (0.22 + 0.031)/0.031]. Therefore, it is recommended to perform single HBV DNA testing on anti-HBc-positive blood donors, discard plasma with weakly positive or negative anti-HBs but positive anti-HBc, or avoid transfusing anti-HBc-positive plasma to recipients with weakly positive or negative anti-HBs to prevent HBV infection.

BackgroundAlmost 40% of patients with kidney renal clear cell carcinoma (KIRC) with advanced cancers eventually develop to metastases, and their 5-year survival rates are approximately 10%. Aberrant DNA methylations are significantly associated with the development of KIRC. The aim of our present study was to identify suitable ferroptosis- and immune-related (FI) biomarkers correlated with aberrant methylations to improve the prognosis and diagnosis of KIRC.MethodsChAMP and DESeq2 in R (3.6.2) were used to screen the differentially expressed methylation probes and differentially expressed genes, respectively. Univariate and multivariate Cox regression were used to identify the overall survival (OS)–related biomarkers.ResultsWe finally identified five FI biomarkers (CCR4, CMTM3, IFITM1, MX2, and NR3C2) that were independently correlated with the OS of KIRC. The area under the curve value of the receiver operating characteristic value of prognosis model was 0.74, 0.68, and 0.72 in the training, validation, and entire cohorts, respectively. The sensitivity and specificity of the diagnosis model were 0.8698 and 0.9722, respectively. In addition, the prognosis model was also significantly correlated with several immune cells and factors.ConclusionOur present study suggested that these five FI-DEGs (CCR4, CMTM3, IFITM1, MX2, and NR3C2) could be used as prognosis and diagnosis biomarkers for patients with KIRC, but further cross-validation clinical studies are still needed to confirm them.

The COVID-19 epidemic poses a significant challenge to the operation of society and the resumption of work and production. How to quickly track the resident location and activity trajectory of the population, and identify the spread risk of the COVID-19 in geospatial space has important theoretical and practical significance for controlling the spread of the virus on a large scale. In this study, we take the geographical community as the research object, and use the mobile phone trajectory data to construct the spatiotemporal profile of the potential high-risk population. First, by using the spatiotemporal data collision method, identify, and recover the trajectories of the people who were in the same area with the confirmed patients during the same time. Then, based on the range of activities of both cohorts (the confirmed cases and the potentially infected groups), we analyze the risk level of the relevant places and evaluate the scale of potential spread. Finally, we calculate the probability of infection for different communities and construct the spatiotemporal profile for the transmission to help guide the distribution of preventive materials and human resources. The proposed method is verified using survey data of 10 confirmed cases and statistical data of 96 high-risk neighborhoods in Chengdu, China, between 15 January 2020 and 15 February 2020. The analysis finds that the method accurately simulates the spatiotemporal spread of the epidemic in Chengdu and measures the risk level in specific areas, which provides an objective basis for the government and relevant parties to plan and manage the prevention and control of the epidemic.

Background and objective Non-small cell lung cancer (NSCLC) is the most common histological type of lung cancer, and one of the malignant tumor with the highest mortality. As the main part of the optical molecular imaging probe, peptide can realize the early screening and diagnosis of tumor and improve the survival rate of patients. The aim of this study was to screen the small-molecule peptide that highly binds to NSCLC NCI-H1299 cells using in vivo phage display technology and to identify their binding specificity by in vitro experiment. Methods To prepare a tumor-bearing nude mouse model of NCI-H1299 cells, after 3 rounds of in vivo screening with Ph.D.-C7CTM Peptide Library, phage clones were randomly picked, using immunohistochemistry and enzyme-linked immunosorbent assay (ELISA) to identify the affinity of phage clones to NCI-H1299 cells. The positive monoclonal phages DNA was extracted and sequenced to obtain the amino acid sequence of the peptides. The peptides with the highest repetition rate was chemically synthesized and labeled with fluorescein (FITC) to prepare optical molecular probe. We preliminary identified the specificity of the probe binding to lung cancer cells by in vitro experiment. Results After three rounds of in vivo screening, the phages enrichment rate was 341.3 times compared with the first round. Immunohistochemical staining showed that with the increase of screening times, the phages binding to tumor tissues continued to increase, and the binding amount was significantly higher than normal tissues; ELISA results showed that 20 clones among the 30 randomly selected phage clones were positive. After sequencing, the peptide with the highest repetition rate was synthesized and named NSP1; Methyl thiazolyl tetrazolium assay (MTT) and would healing assay showed that NSP1 will not affect cell proliferation and migration. Flow cytometry and immunofluorescence showed specific binding of NSP1 to NCI-H1299 cells. Conclusion We successfully obtained the peptide NSP1 that specifically binds to lung cancer NCI-H1299 cells by in vivo phage display, which provide a theoretical basis for NSCLC early diagnosis and targeted therapy.

Background: The antiviral effects of Novaferon, a potent antiviral protein drug, on COVID-19 was evaluated in the laboratory, and in a randomized, open-label, parallel-group trial. Methods: In the laboratory, Novaferon's inhibition of viral replication in cells infected with SARS-CoV-2, and prevention of SARS-CoV-2 entry into healthy cells was determined. Antiviral effects of Novaferon in COVID-19 patients with treatment of Novaferon, Novaferon plus Lopinavir/Ritonavir, or Lopinavir/Ritonavir were evaluated. The primary endpoint was the SARS-CoV-2 clearance rates on day six of treatment, and the secondary endpoint was the time to SARS-CoV-2 clearance. Results: Novaferon inhibited viral replication (EC50 = 1.02 ng/ml), and prevented viral infection (EC50 = 0.10 ng/ml). Results from the 89 enrolled COVID-19 patients showed that both Novaferon and Novaferon plus Lopinavir/Ritonavir groups had significantly higher viral clearance rates on day six than Lopinavir/Ritonavir group (50.0% vs. 24.1%, p = 0.0400, and 60.0% vs. 24.1%, p = 0.0053). The median time to viral clearance was six days, six days, and nine days for three groups, respectively, a 3-day reduction in both the Novaferon and Novaferon plus Lopinavir/Ritonavir groups compared with the Lopinavir/Ritonavir group. Conclusions: Novaferon exhibited anti-SARS-CoV-2 effects in vitro and in COVID-19 patients. These data justify further evaluation of Novaferon. In this paper, the effects of sodium sulfosalicylate on hemimorphite activation were studied. Sulfosalicylic acid ions enhanced the sulfidation-flotation process, which increased the flotation recovery by 49.70%, and after lead ion assisted activation, the flotation recovery subsequently increased by 13.11%. Time-of-flight secondary ion spectrometry (TOF-SIMS) surface analysis revealed that the surface structure of the hemimorphite was greatly damaged by sodium sulfosalicylate, which could promote ZnS film formation for obtaining hemimorphite hydrophobicity. The ZnS film thickness on the hemimorphite surface was approximately 41.78 nm with direct sulfidation. After activating sulfidation, excellent flotation effects were obtained as the ZnS film reached about 418.19 nm. 3D images showed a heterogeneous and homogeneous ZnS film on the hemimorphite surface after direct sulfidation and activating sulfidation, respectively. The X-ray photoelectron spectroscopy (XPS)-based analysis of the substances on the hemimorphite surface further indicated that the atomic concentration of sulfur on the surface of the activating sulfidation hemimorphite nearly double that of the direct sulfidation hemimorphite (1.57% versus 2.38%, respectively). After activating sulfidation, the XPS spectra indicated that a deep complexation reaction occurred with sodium sulfosalicylate on the hemimorphite surface with various elements and significantly influenced elemental distribution on the hemimorphite surface.

Abstract. Parkinson disease (PD) is the second-most common neurodegenerative disorder. Its main pathological mechanism is the selective degeneration and deletion of dopaminergic neurons in the dense part of the substantia nigra and the damage of dopaminergic neurons caused by the abnormal deposition of a Lewy body, leading to a decreased dopamine level. Positron emission computed tomography (PET)/single photon emission computed tomography (SPECT) is a molecular imaging technology that can directly or indirectly reflect changes in molecular levels by using a specific tracer. With the research and development on the tracers of related enzymes for labeling dopamine transporter and dopamine receptor and for being involved in dopamine formation, this imaging technology has been applied to all aspects of PD research. It not only contributes to clinical work but also provides an important theoretical basis for exploring the pathological mechanism of PD at a molecular level. Therefore, this review discusses the application value of PET/SPECT in PD in terms of early diagnosis, disease severity evaluation, clinical manifestations, differential diagnosis, and pathological mechanism.

Introduction: Some encouraging findings of Chinese herbal medicine (CHM) in management of idiopathic membranous nephropathy (IMN) obtained in the setting of clinical trials are hard to validate in the daily clinical practice due to a complicated treatment scenario of CHM in practice. The primary objective of this registry is to provide a description of treatment patterns used in management of IMN and assess clinical remission in daily practice in a Chinese population sample with IMN.Methods and analysis: This is a prospective, multicenter cohort which will comprise 2000 adults with IMN regardless of urinary protein levels that will be recruited from 11 nephrology centers across China. The participants will be followed for up to at least 2 years. Primary outcome is composite remission (either complete remission or partial remission) 24 months after enrolment. The secondary outcomes are complete remission, partial remission, time to remission, no response, relapse, proteinuria, annual change of glomerular filtration rate, antibodies against PLA2R, and composite endpoint of 40% reduction of glomerular filtration rate, doubling of serum creatinine, end-stage renal disease, and death. Propensity score analysis will be used for matching and adjustment.Ethics and dissemination: This study has been approved by the Ethics Committee of Guangdong Provincial Hospital of Chinese Medicine (BF2020-094-01). Results of the study will be published in both national and international peer-reviewed journals, and presented at scientific conferences. Investigators will inform the participants as well as other IMN patients of the findings via health education.Study registration: ChiCTR2000033680 (prospectively registered).

Two novel betaine surfactants with distinct hydrophilic headgroups were synthesized, including carboxybetaine surfactant (DCB) and sulfobetaine surfactant (DSB). Their properties of reducing the interfacial tension (IFT) of Xinjiang crude oil/water were studied under alkaline-free conditions, as were their thermal stability, wettability, and emulsification properties. The chemical structures of the target products were characterized and analyzed by using 1H-NMR and 13C-NMR. The experimental results indicate that the introduction of a benzene ring to the hydrophobic group can improve the solubility and high-temperature resistance of the betaine surfactant. Thermogravimetric analysis showed that the degradation temperature of the synthesized betaine was above 190 °C. As the concentration of the betaine solution increased, DSB (0.0750 mmol/L) showed a lower critical micelle concentration (CMC) than DCB (0.1852 mmol/L). The wetting ability of DCB was significantly higher than that of DSB, and their contact angles on paraffin film decreased to 28.36° and 35.26°. In addition, both DCB and DSB can reduce the IFT of Xinjiang crude oil/water to ultra-low levels (10−3 mN/m) in the absence of alkali. The appropriate ion concentration has a synergistic effect on the surfactant to reduce the interfacial tension of oil/water and the effect of the three ions on the interfacial tension was as follows: Na+ < Ca2+ < Mg2+.

The purpose of this work is to significantly improve the vanadium grade of vanadium-bearing shale after flotation preconcentration, which is conducive to reducing the acid consumption and industrial costs in the subsequently enhanced acid leaching of vanadium. Vanadium concentrate from vanadium-bearing shale enriched by flotation is used for acid-leaching feed. The leaching effects of two kinds of acid-leaching systems were compared, and the mechanism of acid leaching on the mineral structure was also described. The difficulty of spontaneous reactions of vanadium-bearing minerals such as garnet in an acid-leaching system was studied through thermodynamics. Additionally, several characterization methods were used to evaluate the improvement of leaching performance. The addition of oxidants and fluorinating aids strengthens the acid-leaching process, which greatly destroys the structure of a garnet, which is conducive to the extraction of vanadium in a flotation concentrate. The leaching efficiency can reach 94.86%, and the acid consumption is also reduced. Through the mechanism study of the leaching system, it is expected that when the enhanced acid-leaching process is put into industrial production, the effective leaching of vanadium can be accurately controlled, and the difficulty of subsequent vanadium enrichment and purification can be reduced.

In shale vanadium ore, vanadium is mainly formed as a low-V(III) homogeneous phase to partially replace Si and exists in the lattice structure of aluminosilicate minerals such as biotite. During the acid leaching of shale vanadium ore, an activator is needed to effectively extract vanadium. Using biotite as a tetrahedral structure doped with Aluminum-Silicon tetrahedron structure, the interaction between leaching-agent ions (H+), activator ions (F−), and vanadium-containing biotite surface was discussed by DFT quantum chemical calculations. The dissolution behavior of activator fluoride ions on the surface of vanadium-bearing biotite during sulfuric acid leaching was revealed. According to the simulated leaching results, the oxygen on the biotite surface first absorbed hydrogen ions to undergo complete hydroxylation, and then combined with hydrogen ions to form water molecules. However, in the presence of activator (NaF), fluoride ions were adsorbed on the surface cations, which catalyzed the formation of water molecules and promoted the dissolution of surface cations. SEM–EDS analysis showed that the surface of vanadium-bearing minerals became very irregular, and the number of voids and cracks greatly increased. At the same time, XPS showed that the addition of activator fluoride ions destroyed the Al-O tetrahedron structure. Many Al and V atoms dissolved in the solution, which improved the leaching rate of vanadium.

Tungsten is a commercially important metal element that usually coexists with a variety of non-ferrous metals, which makes its extraction difficult. Scheelite is a commonly occurring tungsten-containing ore with the formula CaWO4. Improving the surface properties of scheelite to increase its adsorption of the collector for flotation separation is the focus of our current research. In this paper, the effects of manganese ions on scheelite flotation in benzohydroxamic acid (BHA) system were studied by micro-flotation tests, adsorption tests, fourier transform infrared spectroscopy (FTIR), zeta potential, and X-ray photoelectron spectroscopy (XPS) analysis. The addition of Mn2+ was found to improve the recovery of scheelite. The addition of Mn2+ greatly improved the recovery of scheelite. Infrared spectroscopy, adsorption tests, zeta potential measurements and XPS analysis all confirmed that BHA had a higher adsorption capacity and a stronger bond to the surface of scheelite after the addition of manganese ions, increasing the floatability of scheelite particles. Therefore, Mn2+ shows great potential for the improvement of the flotation index of scheelite in a system with BHA.

Semantic segmentation, as a pixel-level recognition task, has been widely used in a variety of practical scenes. Most of the existing methods try to improve the performance of the network by fusing the information of high and low layers. This kind of simple concatenation or element-wise addition will lead to the problem of unbalanced fusion and low utilization of inter-level features. To solve this problem, we propose the Inter-Level Feature Balanced Fusion Network (IFBFNet) to guide the inter-level feature fusion towards a more balanced and effective direction. Our overall network architecture is based on the encoder–decoder architecture. In the encoder, we use a relatively deep convolution network to extract rich semantic information. In the decoder, skip-connections are added to connect and fuse low-level spatial features to restore a clearer boundary expression gradually. We add an inter-level feature balanced fusion module to each skip connection. Additionally, to better capture the boundary information, we added a shallower spatial information stream to supplement more spatial information details. Experiments have proved the effectiveness of our module. Our IFBFNet achieved a competitive performance on the Cityscapes dataset with only finely annotated data used for training and has been greatly improved on the baseline network.

Spatiotemporal graph convolution has made significant progress in skeleton-based action recognition in recent years. Most of the existing graph convolution methods take all the joints of the human skeleton as the overall modeling graph, ignoring the differences in the movement patterns of various parts of the human, and cannot well connect the relationship between the different parts of the human skeleton. To capture the unique features of different parts of human skeleton data and the correlation of different parts, we propose two new graph convolution methods: the whole graph convolution network (WGCN) and the part graph convolution network (PGCN). WGCN learns the whole scale skeleton spatiotemporal features according to the movement patterns and physical structure of the human skeleton. PGCN divides the human skeleton graph into several subgraphs to learn the part scale spatiotemporal features. Moreover, we propose an adaptive fusion module that combines the two features for multiple complementary adaptive fusion to obtain more effective skeleton features. By coupling these proposals, we build a whole and part adaptive fusion graph convolution neural network (WPGCN) that outperforms previous state-of-the-art methods on three large-scale datasets: NTU RGB+D 60, NTU RGB+D 120, and Kinetics Skeleton 400.

Abstract Hymenoptera is an order accounting for a large proportion of species in Insecta, among which Chalcidoidea contains many parasitoid species of biocontrol significance. Currently, some species genomes in Chalcidoidea have been assembled, but the chromosome-level genomes of Aphelinidae are not yet available. Using Illumina, PacBio HiFi and Hi-C technologies, we assembled a genome assembly of Eretmocerus hayati (Aphelinidae, Hymenoptera), a worldwide biocontrol agent of whiteflies, at the chromosome level. The assembled genome size is 692.1 Mb with a contig N50 of 7.96 Mb. After Hi-C scaffolding, the contigs was assembled onto four chromosomes with a mapping rate of > 98%. The scaffold N50 length is 192.5 Mb, and Benchmarking Universal Single-Copy Orthologues (BUSCO) value is 95.9%. The genome contains 370.8 Mb repeat sequences and total of 24471 protein coding genes. P450 gene families were identified and analyzed. In conclusion, our chromosome-level genome assembly provides valuable support for future research on the evolution of parasitoid wasps and the interaction between hosts and parasitoid wasps.

Pulsar halos (also termed “TeV halos”) are a new class of γ -ray sources in the Galaxy, which manifest as extended γ -ray emission around middle-aged pulsars, as discovered around the Geminga pulsar, the Monogem pulsar, and PSR J0622+3749 by the High-Altitude Water Cherenkov Observatory and the Large High-Altitude Air Shower Observatory. A consensus has been reached that the teraelectronvolt emission comes from the inverse Compton scattering of escaping electrons/positrons from the pulsar wind nebula of the soft background radiation field, while the particle transport mechanism in the halo is still in dispute. Currently, there are mainly three interpretations: the isotropic, suppressed diffusion model; the isotropic, unsuppressed diffusion model that considers the ballistic propagation of newly injected particles; and the anisotropic diffusion model. While the predicted γ -ray surface brightness profiles of all three models can be more or less consistent with the observations, the implications of the three models for cosmic-ray transport mechanisms and the properties of the interstellar magnetic field are quite different. In this study, we calculate the anticipated X-ray emission of pulsar halos under the three models. We show that the synchrotron radiation of these escaping electrons/positrons can produce a corresponding X-ray halo around the pulsar and that the expected surface brightness profiles are distinct in the three models. We suggest that sensitive X-ray detectors of a large field of view (such as eROSITA and the Einstein Probe) with a reasonably long exposure time are crucial to understanding the formation mechanism of pulsar halos and can serve as a probe of the properties of interstellar turbulence.

Cell senescence is closely related to autophagy. In this article, we identified a natural nucleoside analogue, cordycepin, that has the ability to significantly improve lysosomal function, enhance the activity of the lysosomal representative protease cathepsin B (CTSB), and promote the expression of the functional protein lysosomal‐associated membrane protein 2 (LAMP2) on the lysosomal membrane. Cordycepin then restores the damaged autophagy level of aging cells by activating the classic AMPK and mTOR–p70S6K signaling pathways, thus inhibiting cell senescence in an H2O2‐induced stress‐induced premature senescence (SIPS) cell model. This study provides new theoretical support for the further development of cordycepin and clinical antiaging drugs to inhibit cell senescence and suggests that the regulatory mechanisms of lysosomes in senescent cells should be considered when treating age‐related diseases.

Abstract Activation of adipose tissue macrophages (ATMs) contributes to chronic inflammation and insulin resistance in obesity. However, the transcriptional regulatory machinery involved in ATM activation during the development of obesity is not fully understood. Here, we profiled the chromatin accessibility of blood monocytes and ATMs from obese and lean mice using assay for transposase-accessible chromatin sequencing (ATAC-seq). We found that monocytes and ATMs from obese and lean mice exhibited distinct chromatin accessibility status. There are distinct regulatory elements that are specifically associated with monocyte or ATM activation in obesity. We also discovered several transcription factors that may regulate monocyte and ATM activation in obese mice, specifically a predicted transcription factor named ETS translocation variant 5 (ETV5). The expression of ETV5 was significantly decreased in ATMs from obese mice and its downregulation was mediated by palmitate stimulation. The decrease in ETV5 expression resulted in macrophage activation. Our results also indicate that ETV5 suppresses endoplasmic reticulum (ER) stress and Il6 expression in macrophages. Our work delineates the changes in chromatin accessibility in monocytes and ATMs during obesity, and identifies ETV5 as a critical transcription factor suppressing ATM activation, suggesting its potential use as a therapeutic target in obesity-related chronic inflammation.

Paeoniflorin, a monoterpene glucoside, is increasingly used in the clinical treatment of many diseases because it has a variety of pharmacological activities. Besides, paeoniflorin has been considered the characteristic chemical constituent of Paeoniaceae plants since it was first reported in 1963. Therefore, how to better develop and utilize paeoniflorin in Paeoniaceae has always been a research hotspot. We reviewed the current knowledge on paeoniflorin in Paeoniaceae, with particular emphasis on its distribution and influencing factors. Moreover, the limited understanding of the biosynthesis pathway has restricted the production of paeoniflorin by synthetic biology. This review provides insights into the post-modification pathway of paeoniflorin biosynthesis and proposes directions for further analysis in the future.

ObjectiveTo evaluate the pregnancy outcomes of progestin-primed ovarian stimulation (PPOS) protocol for patients with endometrioma underwent in vitro fertilization/intra-cytoplasmic sperm injection embryo transfer (IVF/ICSI-ET).DesignObservational retrospective cohort study.SettingUniversity affiliated reproductive center.Study Participants605 infertile patients with endometrioma underwent IVF/ICSI-ET from January 2016 to March 2021 were included in this study.MethodsMultivariable logistic regression analyses were conducted to determine the independent effect of controlled ovarian stimulation (COS) protocols on reproductive outcomes of first embryo transfer (ET) cycles. The live birth was primary outcome, the implantation rate, biochemical pregnancy, clinical pregnancy and ongoing pregnancy were secondary outcomes.ResultsCompared to PPOS protocol, the probability of implantation showed no significant difference with ultra-long gonadotrophin-releasing hormone agonist (GnRHa) protocol and gonadotrophin-releasing hormone antagonist (GnRHant) protocol (OR 1.7, 95% CI 0.9-3.1, OR 1.2, 95% CI 0.7-2.1, respectively). The PPOS protocol was correlated with a significantly lower biochemical pregnancy and clinical pregnancy than ultra-long GnRHa protocol in the multivariable logistic regression analysis (OR 2.3, 95% CI 1.1-4.9, OR 2.4, 95% CI 1.1-5.3, respectively). However, there was no significant difference in terms of biochemical pregnancy, clinical pregnancy and ongoing pregnancy between PPOS and GnRHant protocol (OR 1.4, 95% CI 0.7-2.7, OR 1.3, 95% CI 0.7-2.4, OR 1.1, 95% CI 0.6-2.3, respectively). In addition, compared to PPOS protocol, ultra-long GnRHa protocol and GnRHant protocol demonstrated no statistical difference in ongoing pregnancy (OR 2.0, 95% CI 0.9-4.5, OR 2.1, 95% CI 0.6-2.3, respectively). Notably, the ultra-long GnRHa protocol was associated with a significant higher probability of live birth than PPOS protocol both in crude analysis and multivariable logistic regression analysis (OR 2.6, 95% CI 1.3-5.1, OR 2.5, 95% CI 1.1-5.7, respectively). Nevertheless, no statistical difference was found in live birth between PPOS and GnRHant protocol either in crude analysis and multivariable logistic regression analysis (OR1.2, 95% CI 0.6-2.3, OR 1.2, 95% CI 0.6-2.5, respectively).ConclusionsBased on the reproductive outcomes of the first ET cycles in patients with endometrioma, PPOS protocol may associated with inferior reproductive outcomes in terms of biochemical pregnancy, clinical pregnancy and live birth than ultra-long GnRHa protocol. However, there was no significant difference in implantation rate, clinical pregnancy, ongoing pregnancy and live birth between PPOS and GnRHant protocol.

Pulsar wind nebula (PWN) Boomerang and the associated supernova remnant (SNR) G106.3+2.7 are among candidates for the ultra-high-energy (UHE) gamma-ray counterparts published by LHAASO. Although the centroid of the extended source, LHAASO J2226+6057, deviates from the pulsar’s position by about 0.3∘, the source partially covers the PWN. Therefore, we cannot totally exclude the possibility that part of the UHE emission comes from the PWN. Previous studies mainly focus on whether the SNR is a PeVatron, while neglecting the energetic PWN. Here, we explore the possibility of the Boomerang Nebula being a PeVatron candidate by studying its X-ray radiation. By modeling the diffusion of relativistic electrons injected in the PWN, we fit the radial profiles of X-ray surface brightness and photon index. The solution with a magnetic field B=140μG can well reproduce the observed profiles and implies a severe suppression of IC scattering of electrons. Hence, if future observations reveal part of the UHE emission originating from the PWN, we propose to introduce a proton component to account for the UHE emission in light of the recent LHAASO measurement on Crab Nebula. In this sense, Boomerang Nebula would be a hadronic PeVatron.
