In recent years, a growing body of research has confirmed that the gut microbiota plays a major role in the maintenance of human health and disease. A gut microbiota imbalance can lead to the development of many diseases, such as pregnancy complications, adverse pregnancy outcomes, polycystic ovary syndrome, endometriosis, and cancer. Short-chain fatty acids are metabolites of specific intestinal bacteria and are crucial for maintaining intestinal homeostasis and regulating metabolism and immunity. Endometriosis is the result of cell proliferation, escape from immune surveillance, and invasive metastasis. There is a strong correlation between the anti-proliferative and anti-inflammatory effects of short-chain fatty acids produced by gut microbes and the development of endometriosis. Given that the mechanism of action of gut microbiota and Short-chain fatty acids in endometriosis remain unclear, this paper aims to provide a comprehensive review of the complex interactions between intestinal flora, short-chain fatty acids and endometriosis. In addition, we explored potential microbial-based treatment strategies for endometriosis, providing new insights into the future development of diagnostic tests and prevention and treatment methods for endometriosis.

Background: Epidermal growth factor-like repeats and discoidin I-like domains 3 (EDIL3) is a secreted extracellular matrix protein implicated in diverse physiological and pathological processes including embryonic development, angiogenesis, and anti-inflammatory responses. Recent reports have indicated that EDIL3 play critical roles in carcinogenesis and progression of many cancers. Herein, we performed a pan-cancer investigation to study the potential functions of EDIL3 in various cancers and experimentally validate its function in gastric cancer (GC). Methods: We analysed EDIL3 expression profiles in different tumours using The Cancer Genome Atlas database. The Kaplan–Meier Plotter was used to investigate the prognostic value of EDIL3, while receiver operating characteristic curve was performed to analyze its diagnostic efficacy. Several bioinformatics tools were used to study the association between EDIL3 and promoter methylation, gene enrichment analysis, immune infiltration, immune-related genes, and drug sensitivity. Molecular biology experiments were conducted to validate the tumorigenic effects of EDIL3. Results: EDIL3 is variably expressed in different cancers and is closely associated with clinical outcomes. An inverse correlation between EDIL3 and DNA methylation has been observed in 13 cancers. Enrichment analysis indicated that EDIL3 is correlated with many cellular pathways such as extracellular matrix receptor interactions and focal adhesion. EDIL3 was tightly associated with immune infiltration and immune checkpoints. EDIL3 knockdown can promote GC calls apoptosis while preventing proliferation, migration, and invasion in vitro. Conclusion: EDIL3 is a promising prognostic, diagnostic, and immunological biomarker in various cancers, which could be applied as a new target for cancer therapy.

Immune checkpoint inhibitors (ICIs) have been widely applicated in clinical therapy in recent years. Skin-related adverse reaction is one of the most common adverse events for ICIs. Stevens-Johnson syndrome (SJS) is one of the serious cutaneous reactions threatening the life. Here, we reported a case of 76-year-old male patient with poorly differentiated metastatic lung adenocarcinoma, after 9 weeks exposure of sintilimab (3 doses) combined with paclitaxel liposome after concurrent chemotherapy/radiotherapy, experienced Stevens-Johnson syndrome involving limbs, trunk, lip and the oral mucosa. Biopsy of the skin tissue showed infiltration of CD4 and CD8 positive T lymphocytes. We also found PD-L1 expression in the glands and the basal layer of the skin. This finding is distinct from the previously reported expression of PD-L1 on the surface of epidermal keratinocytes in patients with SJS due to immunotherapy.

Background EDIL3, which contains epidermal growth factor-like repeats and discoidin I-like domains, is a secretory protein that plays an important role in embryonic development and various illnesses. However, the biological function of EDIL3 in gastric cancer (GC) is still unclear. The objective of this research was to explore the role and potential mechanism of EDIL3 in GC. Methods In this study, we used the GEPIA, HPA, MethSurv, SMART, STRING, GeneMANIA, LinkedOmics TIMER, TIMER2.0, TISIDB, and RNAactDrug databases to comprehensively analyze the roles of EDIL3 in GC. To validate the in silico findings, EDIL3 expression was measured in our collected GC tissues. Meanwhile, several in vitro experiments were performed to test the function of EDIL3 in GC. Results We found that EDIL3 was highly expressed in GC and associated with adverse clinical features. In vitro assays revealed that EDIL3 promoted the proliferation, migration, and invasion of GC cells. The functions of EDIL3 and co-expression genes were significantly associated with extracellular structure organization and matrix receptor interaction. EDIL3 expression was positively associated with numerous tumor-infiltrating immune cells and their biomarkers. Conclusion This study determined that EDIL3 may function as an oncogene and is associated with immune infiltration in GC. EDIL3 could be used as a potential therapeutic target for GC.

Objective To explore the intrinsic mechanism of the mammalian target of rapamycin (mTOR) pathway activation and promotion of neuronal axon growth. Methods Human neuroblastoma cells, SH-SY5Y, were induced with all-trans retinoic acid (ATRA; 10 μM for three days) which differentiated the cell line into a neuronal-like state. Immunohistochemical staining was used to detect the differentiation status of the neuronal-like cells. Phosphatase and tensin homolog (PTEN) RNA interference (RNAi) experiments were performed on the differentiated cells; reverse transcription-polymerase chain reaction (RT-PCR) detected transcriptional levels of PTEN following 24 h of interference. After 36 h, western blot assay was used to detect expression levels of ribosomal protein S6 kinase (pS6k) and mTOR. To downregulate the expression of PTEN and cluster of differentiation 44 (CD44), a cell-surface glycoprotein, simultaneously, PTEN siRNA and CD44 siRNA sequences were mixed in equal proportions in co-interference experiments. RT-PCR detected the transcription level of CD44, and the relationship between the CD44 and axonal growth was observed after 48 h of interference. Results Microtubule-associated protein 2 (MAP2) expression was enhanced after three days of induction in SH-SY5Y cells. RT-PCR showed the transcription level of PTEN was significantly downregulated after 24 h of PTEN knockdown. mTOR and pS6k protein expression levels were significantly upregulated after 36 h of interference. CD44 transcription levels were upregulated after PTEN gene interference. The neurite length of the cells in the experimental interference group was significantly longer than that in the control group, and the expression level of CD44 was positively correlated with neurite growth. The neurite length of the PTEN-only interference group was significantly greater than that of the co-interference and ATRA groups. Conclusion Activation of the mTOR pathway promoted neurite growth through upregulation of CD44 expression, thus promoting neuronal regeneration.

This study mitigated the challenge of head and neck CT angiography by IPA-based time-resolved imaging of contrast kinetics. To this end, 627 cerebral hemorrhage patients with dizziness, brain aneurysm, stroke, or hemorrhagic stroke diagnosis were randomly categorized into three groups, namely, the original dataset (450), verification group (112), and in vivo testified group (65), in the Affiliated BenQ Hospital of Nanjing Medical University. In the first stage, seven risk factors were assigned: age, CTA tube voltage, body surface area, heart rate per minute, cardiac output blood per minute, the actual injected amount of contrast media, and CTA delayed trigger timing. The expectation value of the semi-empirical formula was the CTA number of the patient’s left artery (LA). Accordingly, 29 items of the first-order nonlinear equation were calculated via the inverse problem analysis (IPA) technique run in the STATISTICA 7.0 program, yielding a loss function and variance of 3.1837 and 0.8892, respectively. A dimensionless AT was proposed to imply the coincidence, with a lower AT indicating a smaller deviation between theoretical and practical values. The derived formula was confirmed for the verification group of 112 patients, reaching high coincidence, with average ATavg and standard deviation values of 3.57% and 3.06%, respectively. In the second stage, the formula was refined to find the optimal amount of contrast media for the CTA number of LA approaching 400. Finally, the above procedure was applied to head and neck CTA images of the third group of 65 patients, reaching an average CTA number of LA of 407.8 ± 16.2 and finding no significant fluctuations.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has resulted in countless infections and caused millions of deaths since its emergence in 2019. Coronavirus disease 2019 (COVID-19)-associated mortality is caused by uncontrolled inflammation, aberrant immune response, cytokine storm, and an imbalanced hyperactive immune system. The cytokine storm further results in multiple organ failure and lung immunopathology. Therefore, any potential treatments should focus on the direct elimination of viral particles, prevention strategies, and mitigation of the imbalanced (hyperactive) immune system. This review focuses on cytokine secretions of innate and adaptive immune responses against COVID-19, including interleukins, interferons, tumor necrosis factor-alpha, and other chemokines. In addition to the review focus, we discuss potential immunotherapeutic approaches based on relevant pathophysiological features, the systemic immune response against SARS-CoV-2, and data from recent clinical trials and experiments on the COVID-19-associated cytokine storm. Prompt use of these cytokines as diagnostic markers and aggressive prevention and management of the cytokine storm can help determine COVID-19-associated morbidity and mortality. The prophylaxis and rapid management of the cytokine storm appear to significantly improve disease outcomes. For these reasons, this study aims to provide advanced information to facilitate innovative strategies to survive in the COVID-19 pandemic.

It is a valuable issue to explore whether a flame can radiate microwaves, in spite of the electric field formed in the flame. Presented herein is an experimental study on a series of flames with millimeter-wave radiation in the combustion of pyrotechnic films. The pyrotechnic films were composed of ultra-fine red phosphorus (P), sodium nitrate (NaNO3), Polyvinyl Alcohol (PVA) and some additives such as chopped carbon fibers (CFs) and aluminized glass fibers (GFs). The combustion temperatures and millimeter-wave radiation signals of the flames were measured, the millimeter-wave emissivity and spectral radiant exitance were calculated to describe the millimeter-wave radiation intensity. The results demonstrate that the flame of the pyrotechnic films based on P/NaNO3/CFs can radiate millimeter waves, and different materials and their proportion have a great effect on the millimeter-wave radiation intensity.

Bronchopulmonary dysplasia (BPD) is a common pulmonary complication observed in preterm infants that is composed of multifactorial pathogenesis. Current strategies, albeit successful in moderately reducing morbidity and mortality of BPD, failed to draw overall satisfactory conclusion. Here, using a typical mouse model mimicking hallmarks of BPD, we revealed that both cord blood-derived mononuclear cells (CB-MNCs) and umbilical cord-derived mesenchymal stem cells (UC-MSCs) are efficient in alleviating BPD. Notably, infusion of CB-MNCs has more prominent effects in preventing alveolar simplification and pulmonary vessel loss, restoring pulmonary respiratory functions and balancing inflammatory responses. To further elucidate the underlying mechanisms within the divergent therapeutic effects of UC-MSC and CB-MNC, we systematically investigated the long noncoding RNA (lncRNA)–microRNA (miRNA)–messenger RNA (mRNA) and circular RNA (circRNA)–miRNA–mRNA networks by whole-transcriptome sequencing. Importantly, pathway analysis integrating Gene Ontology (GO)/Kyoto Encyclopedia of Genes and Genomes (KEGG)/gene set enrichment analysis (GSEA) method indicates that the competing endogenous RNA (ceRNA) network is mainly related to the regulation of GTPase activity (GO: 0043087), extracellular signal-regulated kinase 1 (ERK1) and ERK2 signal cascade (GO: 0070371), chromosome regulation (GO: 0007059), and cell cycle control (GO: 0044770). Through rigorous selection of the lncRNA/circRNA-based ceRNA network, we demonstrated that the hub genes reside in UC-MSC- and CB-MNC-infused networks directed to the function of cell adhesion, motor transportation (Cdk13, Lrrn2), immune homeostasis balance, and autophagy (Homer3, Prkcd) relatively. Our studies illustrate the first comprehensive mRNA–miRNA–lncRNA and mRNA–miRNA–circRNA networks in stem cell-infused BPD model, which will be valuable in identifying reliable biomarkers or therapeutic targets for BPD pathogenesis and shed new light in the priming and conditioning of UC-MSCs or CB-MNCs in the treatment of neonatal lung injury.

This study optimized the ultrasound image of carotid artery stenosis using Taguchi dynamic analysis and an indigenous water phantom. Eighteen combinations of seven essential factors of the ultrasound scan facility were organized according to Taguchi’s L18 orthogonal array. The seven factors were assigned as follows: (1) angle of probe; (2) signal gain; (3) resolution vs. speed; (4) dynamic range; (5) XRES; (6) zoom; (7) time gain compensation. An indigenous water phantom was customized to satisfy the quantified need in Taguchi’s analysis. Unlike the conventional dynamic Taguchi analysis, an innovative quantified index, the figure of merit (FOM), was proposed to integrate four specific quality characteristics, namely (i) average difference between the practical scan and theoretically preset area (78.5, 50.2 and 12.6 mm2) of stenosis, (ii) standard deviation of the average, (iii) practical scan’s sensitivity β to various stenosis diameters (10, 8, and 4 mm), and (iv) correlation coefficient r2 of the linear regressed sensitivity curve. The highest value (FOM = 0.413) was furnished by the optimal combination of factors on 18 groups under study, yielding high r2 and low β or standard deviation values and the best quality of ultrasound images for the further clinical diagnosis. The comparison between FOM and the conventional signal-to-noise (S/N) ratio in Taguchi’s analysis revealed that FOM compiled more quality characteristics that were superior by nature to fulfill the practical need in clinical diagnosis. The alternative choice in ultrasound scan optimization can be based on stenosis diameter variation from a different perspective to be explored in the follow-up study.

Severe acute respiratory syndrome coronavirus (SARS-CoV-2) first emerged in December 2019 in Wuhan, China, and has since spread rapidly worldwide. As researchers seek to learn more about COVID-19, the disease it causes, this novel virus continues to infect and kill. Despite the socioeconomic impacts of SARS-CoV-2 infections and likelihood of future outbreaks of other pathogenic coronaviruses, options to prevent or treat coronavirus infections remain limited. In current clinical trials, potential coronavirus treatments focusing on killing the virus or on preventing infection using vaccines largely ignore the host immune response. The relatively small body of current research on the virus indicates pathological responses by the immune system as the leading cause for much of the morbidity and mortality caused by COVID-19. In this review, we investigated the host innate and adaptive immune responses against COVID-19, collated information on recent COVID-19 experimental data, and summarized the systemic immune responses to and histopathology of SARS-CoV-2 infection. Finally, we summarized the immune-related biomarkers to define patients with high-risk and worst-case outcomes, and identified the possible usefulness of inflammatory markers as potential immunotherapeutic targets. This review provides an overview of current knowledge on COVID-19 and the symptomatological differences between healthy, convalescent, and severe cohorts, while offering research directions for alternative immunoregulation therapeutic targets.

Objective: To observe the effect of acne-eliminating cream on acne in a rabbit ear acne model. Methods: Thirty New Zealand rabbits of both genders were divided into normal control group, acne model group, high and low dose of acne-eliminating cream group, tretinoin cream group. Except 3 rabbits in normal control group, ears of the rest rabbits were treated topically with coal tar for 2 weeks to establish acne model. Afterwards vehicle, acne cream (high and low dose), and 0.1% tretinoin cream were applied to respective group of rabbit ears daily for 3 weeks. Ear samples were taken 24 hours after the last topical treatment, followed by fixation in 10% formaldehyde and hematoxylin-eosin staining. Histopathological changes were assessed and photographed under a microscope. Results: In comparison to untreated acne ears, high dose of acne-eliminating cream markedly reduced the intensity of ear reactions to coal tar (t=2.62, P0.05 vs. untreated acne ears). High dose of acne-eliminating cream and tretinoin cream exerted comparable efficacy in reduction of intensity of ear reactions to coal tar and improvements in histopathology (t=0.06, P>0.05 for reduction of intensity;χ2=0.34, P>0.05 for histopathology). Conclusion: High dose of acne-eliminating cream can significantly improve the acne in coal tar-induced rabbit ear acne model.

The deployment of a client–server-based distributed intelligent system involves application development in both the network domain and the device domain. In the network domain, an application server (typically in the cloud) is deployed to execute the network applications. In the device domain, several Internet of Things (IoT) devices may be configured as, for example, wireless sensor networks (WSNs), and interact with each other through the application server. Developing the network and the device applications are tedious tasks that are the major costs for building a distributed intelligent system. To resolve this issue, a low-code or no-code (LCNC) approach has been purposed to automate code generation. As traditional LCNC solutions are highly generic, they tend to generate excess code and instructions, which will lack efficiency in terms of storage and processing. Fortunately, optimization of automated code generation can be achieved for IoT by taking advantage of the IoT characteristics. An IoT-based distributed intelligent system consists of the device domain (IoT devices) and the network domain (IoT server). The software of an IoT device in the device domain consists of the Device Application (DA) and the Sensor Application (SA). Most IoT LCNC approaches provide code generation in the network domain. Very few approaches automatically generate the DA code. To our knowledge, no approach supports the SA code generation. In this paper, we propose DeviceTalk, an LCNC environment for the DA and the SA code development. DeviceTalk automatically generates the code for IoT devices to speed up the software development in the device domain for a distributed intelligent system. We propose the DeviceTalk architecture, design and implementation of the code generation mechanism for the IoT devices. Then, we show how a developer can use the DeviceTalk Graphical User Interface (GUI) to exercise LCNC development of the device software.

The thyroid biokinetic model of radioactive I-131 was re-evaluated using a refined nine-compartmental model and applied to twelve thyroid cancer patients. In contrast to the simplified four-compartmental model regulated by the ICRP-56 report, the revised model included nine compartments specified in the ICRP-128 report, namely, oral, stomach, body fluid, thyroid, whole body, liver, kidney, bladder, and remainder (i.e., the whole body minus kidney and bladder). A self-developed program run in MATLAB was designed to solve the nine first-order simultaneous linear differential equations. The model was realized in standard and simplified versions. The latter neglected two feedback paths (body fluid to oral, i31, and kidney to the whole body, i87) to reduce computations. Accordingly, the biological half-lives for the major compartments (thyroid and body fluid + whole body) were 36.00 ± 15.01, 15.04 ± 5.63, 34.33 ± 15.42, and 14.83 ± 5.91 of standard and simplified version. The correlations between theoretical and empirical data for each patient were quantified by the dimensionless AT (agreement) index and, the ATtot index integrated each individual AT of a specific organ of one patient. Since small AT values indicated a closer correlation, the obtained range of ATtot (0.048 ± 0.019) proved the standard model’s reliability and high accuracy, while the simplified one yielded slightly higher ATtot (0.058 ± 0.023). The detailed outcomes among various compartments of twelve patients were calculated and compared with other researchers’ work. The correlation results on radioactive I-131 evolution in thyroid cancer patients’ bodies are instrumental in viewpoint of radioactive protection of patients and radiological personnel.

This study tried to propose an innovated idea of solidifying the resolution of gamma camera in routine quality control and recommended a quantified index as minimum detectable difference (MDD) of gamma camera SPECT images using the Taguchi analysis and an indigenous V-shaped slit gauge. The gauge was customized to fulfill the quantitative requirement of the Taguchi analysis. The MDD among slit gauge of derived SPECT image was calculated from two overlapped peak profiles collected from a tangent slice of the V-shaped slit gauge with two nearby peaks. In particular, MDD was evaluated as minimum distance between two peak centers through the Student’s t-test with a constant, 1.96, which indicates that two peak centers separated distant enough to create a 95% confidence level of separation. Eighteen combinations of six gamma camera scanned factors were organized according to Taguchi analysis. Accordingly, (A) collimator, (B) detector to targe distance, (C) total counts, (D) acquired energy width, (E) Matrix size, and (F) zoom of collected ROI with each of two or three levels were organized into 18 groups to collect the slit gauge images according to Taguchi L18 orthogonal array. Then, three well-trained radiologists were ranked the scanned gauge images to derive the fish-bone-plot of signal-to-noise ratio (S/N, dB) and correlated ANOVA. Furthermore, the quantified MDD was proposed to verify the optimal suggestion of gamma camera scanned protocol, and obtained the MDD as 8.4, 7.9, and 7.1 mm for the second group of original L18 preset, conventional, and the optimal preset, respectively. Thus, the optimal preset of gamma camera was achieved in this study. The MDD proved to be a successful index in quantifying the imaging resolution of a gamma camera.

This study evaluated the preservation effectiveness of selected antistaling agents on lotus seeds (Nelumbo nucifera Gaertn) status and the alteration of phenolic acids during their storage. Ascorbic acid (AA), benzoic acid (BA), sodium hydrogen sulfite (SHS), BA+ AA and SHS+ AA were identified as effective to keep lotus seeds fresh with the order of SHS+ AA > BA+ AA > AA > SHS > BA in terms of general performance on not-browning degree, microorganism contamination status, sensory evaluation, and the protection of polyphenols against being oxidized. The polyphenols determined by HPLC and HPLC-MS in lotus seeds were mainly gallic, chlorogenic, gentisic, caffeic, cinnamic, p-coumaric, ferulic, rosmarinic, and salicylic acids. Purpose: The root of Cynanchum auriculatum Royle ex Wight, known as Baishouwu, has been widely used for a tonic supplement since ancient times. The current study was performed to explore the effect of Baishouwu extract on the development of experimental hepatocellular carcinoma (HCC) and the potential mechanism involved.Methods: Rats were injected diethylnitrosamine (DEN) to initiate the multistep hepatocarcinogenesis. Animals were treated concurrently with Baishouwu extract given daily by oral gavage for 20 weeks to evaluate its protective effects. Time series sera and organ samples from each group were collected to evaluate the effect of Baishouwu extract on hepatic carcinogenesis.Results: It was found that Baishouwu extract pretreatment successfully attenuated liver injury induced by DEN, as shown by decreased levels of serum biochemical indicators (AST, ALT, ALP, TP, and T-BIL). Administration of Baishouwu extract inhibited the fibrosis-related index in serum and live tissue, respectively from inflammation stage to HCC stage after DEN treatment. It significantly reduced the incidence and multiplicity of DEN-induced HCC development in a dose-dependent manner. Macroscopic and microscopic features suggested that pretreatment with Baishouwu extract for 20 weeks was effective in inhibiting DEN-induced inflammation, liver fibrosis, and HCC. Furthermore, TLR4 overexpression induced by DEN was decreased by Baishouwu extract, leading to the markedly down-regulated levels of MyD88, TRAF6, NF-κB p65, TGF-β1 and α-SMA in hepatitis, cirrhosis, and hepatocarcinoma.Conclusion: In conclusion, Baishouwu extract exhibited potent effect on the development of HCC by altering TLR4/MyD88/ NF-κB signaling pathway in the sequence of hepatic inflammation-fibrosis-cancer, which provided novel insights into the mechanism of Baishouwu extract as a candidate for the pretreatment of HCC in the future.

Smart spring support is a kind of active damping device based on piezoelectric material. It can effectively suppress the vibration of a shaft system in an overly critical state by changing the stiffness and damping of the support. The support parameters have a significant impact on the vibration of the system. By studying the influence of the smart spring support parameters on the vibration characteristics of the transmission shaft system, the support parameters can be configured more reasonably so that the vibration of the transmission system can be reduced as much as possible. Based on the finite element method, this paper studies the influence of the stiffness, damping and mass of the smart spring support on the vibration characteristics of the three-support shafting. Firstly, the smart spring shafting test bed is built, and the vibration reduction performance test of the smart spring is carried out to verify the damping effect of the smart spring. Then, the shafting dynamic model is established by the finite element method, and the inherent characteristics of the system are analyzed. Finally, the influence of the stiffness, damping, mass and other parameters of the smart spring support on the dynamic response of the system is studied. The results show that increasing the stiffness of the smart spring support can effectively reduce the vibration amplitude of the system. The damping of the smart spring support has no obvious effect on the vibration of the shafting. The smaller the mass of the smart spring support, the more favorable the system is.

Smart Spring is a kind of active vibration control device based on piezoelectric material, which can effectively suppress the vibration of the shaft system in an over-critical state, and the selection of control strategy has great influence on the vibration reduction effect of the Smart Spring. In this paper, the authors investigate the control of the over-critical vibration of the transmission shaft system with Smart Spring, based on the ADAMS and MATLAB joint simulation method. Firstly, the joint simulation model of three-support shafting with Smart Spring is established, and the over-critical speed simulation analysis of the three-support shafting under the fixed control force of the Smart Spring is carried out. The simulation results show that the maximum vibration reduction rate is 71.6%. The accuracy of the joint simulation model is verified by the experiment of the three-support shafting subcritical vibration control. On this basis, a function control force vibration control strategy with time-varying control force is proposed. By analyzing the axis orbit of the shafting, the optimal fixed control force at different speeds is obtained, the control force function is determined by polynomial fitting, and the shafting critical crossing simulation under the function control force is carried out. The simulation results show that the displacement response of the shafting under the function control force is less than that under the fixed control force in the whole speed range.

Periploca sepium Bunge is not only a traditional medicinal plant, but also a pioneer species for soil and water conservation because of its great environmental adaptability. In this study, its complete chloroplast genome was assembled from the whole genome Illumina sequencing data. The circular genome was 153,513 bp in size, containing a large single copy (LSC) region of 83,818 bp and a small single copy (SSC) region of 18,177 bp, which were separated by a pair of 25,759 bp inverted repeat (IR) regions. It encoded a total of 139 genes, including 85 protein-coding genes, 46 tRNA genes, and eight rRNA genes. The most of gene species occurred as a single copy, while 15 gene species occurred in double copies. The overall A + T content was 61.9%, while the corresponding values of the LSC, SSC, and IR regions were 63.8, 68.1, and 56.6%, respectively. Phylogenetic analysis indicated that P. sepium was relatively close to the species belonging to the subfamily Apocynoideae.

The construction of high density genetic linkage map provides a powerful tool to detect and map quantitative trait loci (QTLs) controlling agronomically important traits. In this study, simple sequence repeat (SSR) markers and Illumina 9K iSelect single nucleotide polymorphism (SNP) genechip were employed to construct one genetic linkage map of common wheat (Triticum aestivum L.) using 191 recombinant inbred lines (RILs) derived from cross Yu 8679×Jing 411. This map included 1 901 SNP loci and 178 SSR loci, covering 1 659.9 cM and 1 000 marker bins, with an average interval distance of 1.66 cM. A, B and D genomes covered 719.1, 703.5 and 237.3 cM, with an average interval distance of 1.66, 1.45 and 2.9 cM, respectively. Notably, the genetic linkage map covered 20 chromosomes, with the exception of chromosome 5D. Bioinformatics analysis revealed that 1 754 (92.27%) of 1 901 mapped SNP loci could be aligned to 1 215 distinct wheat unigenes, among which 1 184 (97.4%) were located on one single chromosome, and the rest 31 (2.6%) were located on 2 to 3 chromosomes. By performing in silico comparison, 214 chromosome deletion bin-mapped expressed sequence tags (ESTs), 1 043 Brachypodium genes and 1 033 rice genes were further added onto the genetic linkage map. This map not only integrated genetic and physical maps, SSR and SNP loci, respectively, but also provided the information of Brachypodium and rice genes corresponding to 1 754 SNP loci. Therefore, it will be a useful tool for comparative genomics analysis, fine mapping of QTL/gene controlling agronomically important traits and marker-assisted selection breeding in wheat.

This paper proposes an IoT-based intelligent hydroponic plant factory solution called PlantTalk. The novelty of our approach is that the PlantTalk intelligence can be built through an arbitrary smartphone. We show that PlantTalk can flexibly configure the connections of various plant sensors and actuators through a smartphone. One can also conveniently write Python programs for plant-care intelligence through the smart phone. The developed plant-care intelligence includes automatic LED lighting, water spray, water pump and so on. As an example, we show that the PlantTalk intelligence effectively lowers the CO2 concentration, and the reduction speed is 53% faster than a traditional plant system. PlantTalk has been extended for a plant factory called AgriTalk.

14-3-3 proteins belong to a family of phosphoserine/threonine-binding modules and participate in a wide array of signal transduction and regulatory events. Our previous study demonstrated that Ta14-3-3 was significantly down-regulated in leaf and root tissues of hybrid wheat at the tillering stage. In this paper, three homoeologous Ta14-3-3 genes were cloned from common wheat (Triticum aestivum L., 2n=6x=42, AABBDD) and mapped on chromosomes 2A, 2B, and 2D, respectively. Transgenic Arabidopsis plants ectopically overexpressing Ta14-3-3 displayed shorter primary roots, delayed flowering and retarded growth rates, indicating that Ta14-3-3 acted as a growth inhibitor in Arabidopsis. In wheat, Ta14-3-3 was down-regulated in roots and leaves of hybrids as compared to their parental lines. We proposed that Ta14-3-3 proteins might regulate growth vigor in hybrid wheat.

Plant leaves respond to day/night cycling in a number of physiological ways. At the mRNA level, the expression of some genes changes during the 24 h period. To determine which proteins exhibited a rhythmic pattern of expression, proteomic profiles in maize seedling leaves were analyzed by high-throughput two-dimensional gel electrophoresis, combined with MALDI-TOF MS technology. Of the 464 proteins that were detected with silver staining in a pH range of 4-7, 17 (3.66%) showed clock rhythmicity in their abundance. These proteins belonged to diverse functional groups and proteins involved in photosynthesis and carbon metabolism were over-represented. These findings provide a new perspective on the relationship between the physiological functions of leaves and the clock rhythmic system.

This article overviews the scientific results of the microstructural features observed in 316 L stainless steel manufactured by the laser powder bed fusion (LPBF) method obtained by the authors, and discusses the results with respect to the recently published literature. Microscopic features of the LPBF microstructure, i.e., epitaxial nucleation, cellular structure, microsegregation, porosity, competitive colony growth, and solidification texture, were experimentally studied by scanning and transmission electron microscopy, diffraction methods, and atom probe tomography. The influence of laser power and laser scanning speed on the microstructure was discussed in the perspective of governing the microstructure by controlling the process parameters. It was shown that the three-dimensional (3D) zig-zag solidification texture observed in the LPBF 316 L was related to the laser scanning strategy. The thermal stability of the microstructure was investigated under isothermal annealing conditions. It was shown that the cells formed at solidification started to disappear at about 800 °C, and that this process leads to a substantial decrease in hardness. Colony boundaries, nevertheless, were quite stable, and no significant grain growth was observed after heat treatment at 1050 °C. The observed experimental results are discussed with respect to the fundamental knowledge of the solidification processes, and compared with the existing literature data.

High-density genetic linkage maps are necessary for precisely mapping quantitative trait loci (QTLs) controlling grain shape and size in wheat. By applying the Infinium iSelect 9K SNP assay, we have constructed a high-density genetic linkage map with 269 F 8 recombinant inbred lines (RILs) developed between a Chinese cornerstone wheat breeding parental line Yanda1817 and a high-yielding line Beinong6. The map contains 2431 SNPs and 128 SSR & EST-SSR markers in a total coverage of 3213.2 cM with an average interval of 1.26 cM per marker. Eighty-eight QTLs for thousand-grain weight (TGW), grain length (GL), grain width (GW) and grain thickness (GT) were detected in nine ecological environments (Beijing, Shijiazhuang and Kaifeng) during five years between 2010-2014 by inclusive composite interval mapping (ICIM) (LOD ≥ 2.5). Among which, 17 QTLs for TGW were mapped on chromosomes 1A, 1B, 2A, 2B, 3A, 3B, 3D, 4A, 4D, 5A, 5B and 6B with phenotypic variations ranging from 2.62% to 12.08%. Four stable QTLs for TGW could be detected in five and seven environments, respectively. Thirty-two QTLs for GL were mapped on chromosomes 1B, 1D, 2A, 2B, 2D, 3B, 3D, 4A, 4B, 4D, 5A, 5B, 6B, 7A and 7B, with phenotypic variations ranging from 2.62% to 44.39%. QGl.cau-2A.2 can be detected in all the environments with the largest phenotypic variations, indicating that it is a major and stable QTL. For GW, 12 QTLs were identified with phenotypic variations range from 3.69% to 12.30%. We found 27 QTLs for GT with phenotypic variations ranged from 2.55% to 36.42%. In particular, QTL QGt.cau-5A.1 with phenotypic variations of 6.82-23.59% was detected in all the nine environments. Moreover, pleiotropic effects were detected for several QTL loci responsible for grain shape and size that could serve as target regions for fine mapping and marker assisted selection in wheat breeding programs.

OBJECTIVE: The aim of this study was to investigate the prognostic value of metastatic lymph node ratio (LNR) in patients having radical resection for stage III gastric cancer. METHODS: A total of 365 patients with stage III gastric cancer who underwent radical resection between 2002 and 2008 at Tianjin Medical University Cancer Institute and Hospital were analyzed. The cut-point survival analysis was adopted to determine the appropriate cutoffs for LNR. Kaplan-Meier survival curves and log-rank tests were used for the survival analysis. RESULTS: By cut-point survival analysis, the LNR staging system was generated using 0.25 and 0.50 as the cutoff values. Pearson's correlation test revealed that the LNR was related with metastatic lymph nodes but not related with total harvested lymph nodes. Abstract Background Depressive disorders are the most common form of mental disorders in community and health care settings. Unfortunately, the treatment of Major Depressive Disorder (MDD) is far from satisfactory. Vagus nerve stimulation (VNS) is a relatively new and promising physical treatment for depressive disorders. One particularly appealing element of VNS is the long-term benefit in mood regulation. However, because this intervention involves surgery, perioperative risks, and potentially significant side effects, this treatment has been limited to those patients with treatment-resistant depression who have failed medication trials and exhausted established somatic treatments for major depression, due to intolerance or lack of response. This double-blinded randomized clinical trial aims to overcome these limitations by introducing a novel method of stimulating superficial branches of the vagus nerve on the ear to treat MDD. The rationale is that direct stimulation of the afferent nerve fibers on the ear area with afferent vagus nerve distribution should produce a similar effect as classic VNS in reducing depressive symptoms without the burden of surgical intervention. Design One hundred twenty cases (60 males) of volunteer patients with mild and moderate depression will be randomly divided into transcutaneous vagus nerve stimulation group (tVNS) and sham tVNS group. The treatment period lasts 4 months and all clinical and physiological measurements are acquired at the beginning and the end of the treatment period. Discussion This study has the potential to significantly extend the application of VNS treatment for MDD and other disorders (including epilepsy, bipolar disorder, and morbid obesity), resulting in direct benefit to the patients suffering from these highly prevalent disorders. In addition, the results of this double-blinded clinical trial will shed new light on our understanding of acupuncture point specificity, and development of methodologies in clinical trials of acupuncture treatment.
