Lipids are a class of compounds with unique physiological functions and activities. They participate in the regulation of a variety of plant responses to abiotic stress, which are essential for maintaining physiological homeostasis in plants. Since its proposal in 2003, lipidomics has rapidly developed as an emerging discipline that provides novel technical support for traditional metabolomics. Lipidomics can aid in clarifying the mechanisms of lipid metabolic regulation in plants. Moreover, the advantages of being label-free, non-specific, highly sensitive an d allowing for simultaneous elemental and molecular analysis mass spectrometry imaging (MSI) has been widely used to decipher the spatial distribution of various lipid molecules in plant tissues. Here, we introduce the current research status of lipidomics and MSI technologies, and comprehensively review recent progress in applying lipid analysis techniques to study lipid metabolic regulation in plant seeds, especially the application of MSI techniques for mapping lipid species in plants. Lipidomics and MSI are important complements to commonly used multi-omics technologies, and they will provide new opportunities for deeper exploration of plant metabolic pathways and regulatory mechanisms. The continuous development of high-spatial-resolution MSI technologies is expected to further promote the deep analysis and prospecting applications of lipid metabolism network regulation at the spatially resolved level in plants.

Digital spatial profiling (DSP) is an emerging powerful technology for proteomics and transcriptomics analyses in a spatially resolved manner for formalin-fixed paraffin-embedded (FFPE) samples developed by nanoString Technologies. DSP applies several advanced technologies, including high-throughput readout technologies (digital optical barcodes by nCounter instruments or next generation sequencing (NGS)), programmable digital micromirror device (DMD) technology, and microfluidic sampling technologies into traditional immunohistochemistry (IHC) and RNA in situ hybridization (ISH) approaches, creating an innovative tool for discovery, translational research, and clinical uses. Since its launch in 2019, DSP has been rapidly adopted, especially in immuno-oncology and tumor microenvironment research areas, and has revealed valuable information that was inaccessible before. In this article, we report the successful setup and validation of the first DSP technology platform in China. Both DSP spatial protein and RNA profiling approaches were validated using FFPE colorectal cancer tissues. Regions of interest (ROIs) were selected in the areas enriched with tumor cells, stroma/immune cells, or normal epithelial cells, and multiplex spatial profiling of both proteins and RNAs were performed. DSP spatial profiling data were processed and normalized accordingly, validating the high quality and consistency of the data. Unsupervised hierarchical clustering as well as principal component analysis (PCA) grouped tumor, stroma/immune cells, and normal epithelial cells into distinct clusters, indicating that the DSP approach effectively captured the spatially resolved proteomics and transcriptomics profiles of different compartments within the tumor microenvironment. In summary, the results confirmed the expected sensitivity and robustness of the DSP approach in profiling both proteins and RNAs in a spatially resolved manner. As a novel technology in highly complex spatial analyses, DSP endows refined analytical power from the tumor microenvironment perspective with the potential of scaling up to more analyzable targets at relatively low cell input levels. We expect that the DSP technology will greatly advance a wide range of biomedical research, especially in immuno-oncology and tumor microenvironment research areas.

In the last decades, Raman spectroscopy has become an important tool to identify and investigate minerals, gases, glasses, and organic material at room temperature. In combination with high-temperature and high-pressure devices, however, the in situ investigation of mineral transformation reactions and their kinetics is nowadays also possible. Here, we present a novel approach to in situ studies for the sintering process of silicate ceramics by hyperspectral Raman imaging. This imaging technique allows studying high-temperature solid-solid and/or solid-melt reactions spatially and temporally resolved, and opens up new avenues to study and visualize high-temperature sintering processes in multi-component systems. After describing in detail the methodology, the results of three application examples are presented and discussed. These experiments demonstrate the power of hyperspectral Raman imaging for in situ studies of the mechanism(s) of solid-solid or solid-melt reactions at high-temperature with a micrometer-scale resolution as well as to gain kinetic information from the temperature- and time-dependent growth and breakdown of minerals during isothermal or isochronal sintering.

A piezoelectric tuning fork sensor is evaluated as a physical excitation and sensing device aiming at the detection of crystals and their nucleation behavior in joint fluids, as observed in gout. This device allows to study the crystallization tendency at varying temperatures or at increased uric acid concentrations as predominant in real human joints. In this work, a tuning fork resonator is characterized to enable spatially resolved viscosity and density measurements by calculating the mode shape of the vibration from resonance frequency measurements when immersed in test liquids.
