The surface-enhanced Raman scattering (SERS) spectra of three amphiphilic oligopeptides derived from EAK16 (AEAEAKAK)2 were examined to study systematic amino acid substitution effects on the corresponding interaction with Ag colloidal nanoparticles. Such self-assembling molecular systems, known as “molecular Lego”, are of particular interest for their uses in tissue engineering and as biomimetic coatings for medical devices because they can form insoluble macroscopic membranes under physiological conditions. Spectra were collected for both native and gamma-irradiated samples. Quantum mechanical data on two of the examined oligopeptides were also obtained to clarify the assignment of the prominent significative bands observed in the spectra. In general, the peptide–nanoparticles interaction occurs through the COO− groups, with the amide bond and the aliphatic chain close to the colloid surface. After gamma irradiation, mimicking a free oxidative radical attack, the SERS spectra of the biomaterials show that COO− groups still provide the main peptide–nanoparticle interactions. However, the spatial arrangement of the peptides is different, exhibiting a systematic decrease in the distance between aliphatic chains and colloid nanoparticles.

A new powder formulation obtained from bovine blood (Fe-heme) was tested on cucumber (Cucumis sativus L.) to investigate its effectiveness as iron supply in comparison with two synthetic iron-chelates fertilizers: ethylenediaminetetraacetic acid (EDTA/Fe3+) and ethylenediamine-N’N’-bis(2-hydroxyphenyl acetic acid) (o,oEDDHA/Fe3+). Green stressed cucumber plants were evaluated in their recovery (SPAD index and weight variations) and to test the iron reduction capacity of the roots at pH 7.5 and 6.0 using each iron treatment as iron supply. The blood-derived product showed similar effects on decreasing iron-deficiency symptoms: SPAD increments and the weights of plants were similar. Noteworthy, the average of Fe3+ reduction capacity in roots was higher for EDTA/Fe3+, while it was similar for o,oEDDHA/Fe3+, and Fe-heme at pH 7.5. Fe-heme showed a complex behavior due to aggregation and low solubility at pH 6 and showed an unexpectedly high contribution of root exudates to iron reduction.

The present review highlights the recent applications of mid-infrared spectroscopy and in particular of diffuse reflectance spectroscopy (DRIFT) and attenuated total reflectance (ATR) and processing methods (e.g., deconvolution, derivative and chemometrics) to rapidly provide valuable information on soil composition and organic geochemistry. Research has demonstrated that both DRIFT and ATR techniques can be considered useful tools for the analysis of a large number of soil samples, giving not only typical spectral patterns but permitting an accurate prediction of quantitative parameters such as, e.g., total carbon, total nitrogen, C/N ratio, lignin, dissolved OC, carbonyl-C, aromatic-C, O-alkyl-C, and alkyl-C contents. Based on literature results, infrared spectroscopy can be recognized as one of the most promising analytical techniques for investigating soil science.

Mitochondrial neurogastrointestinal encephalomyopathy (MNGIE) is a rare autosomal recessive mitochondrial disease associated with mutations in the nuclear TYMP gene. As a result, the thymidine phosphorylase (TP) enzyme activity is markedly reduced leading to toxic accumulation of thymidine and therefore altered mitochondrial DNA. MNGIE is characterized by severe gastrointestinal dysmotility, neurological impairment, reduced life expectancy and poor quality of life. There are limited therapeutic options for MNGIE. In the attempt to restore TP activity, allogenic hematopoietic stem cell transplantation has been used as cellular source of TP. The results of this approach on ∼ 20 MNGIE patients showed gastrointestinal and neurological improvement, although the 5-year mortality rate is about 70%. In this study we tested whether the liver may serve as an alternative source of TP. We investigated 11 patients (7M; 35-55 years) who underwent hepatic resection for focal disorders. Margins of normal liver tissue were processed to identify, quantify and localize the TP protein by Western Blot, ELISA, and immunohistochemistry, and to evaluate TYMP mRNA expression by qPCR. Western Blot identified TP in liver with a TP/GAPDH ratio of 0.9 ± 0.5. ELISA estimated TP content as 0.5 ± 0.07 ng/μg of total protein. TP was identified in both nuclei and cytoplasm of hepatocytes and sinusoidal lining cells. Finally, TYMP mRNA was expressed in the liver. Overall, our study demonstrates that the liver is an important source of TP. Orthotopic liver transplantation may be considered as a therapeutic alternative for MNGIE patients.
