AbstractAs a processed product of traditional Chinese medicine Curcumae Radix, Curcumae Radix Carbonisata (CRC) has been widely used in the treatment of liver diseases in ancient medical books. In this study, novel carbon dots (CDs) extending from 1.0 to 4.5 nm were separated from fluid extricates of CRC. Meanwhile, a liver fibrosis model induced by carbon tetrachloride (CCl4) was utilized to determine the inhibitory effects of CRC-CDs against liver fibrosis. The results exhibited the CRC-CDs with a quantum yield of 1.34% have a significant inhibitory effect on CCl4-induced liver fibrosis, as demonstrated by improving hepatocyte degeneration and necrosis, inflammatory cell infiltration and fibrotic tissue hyperplasia, downregulating the levels of alanine transaminase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL), direct bilirubin (DBIL), total bile acid (TBA), triglyceride (TG), tumour necrosis factor-α (TNF-α), interleukin (IL)-6 and IL-1β in the serum, upregulating the contents of superoxide dismutase (SOD), reduced glutathione (GSH), and downregulating the concentration of malondialdehyde (MDA), which lays an important foundation for the development of CRC-CDs as a novel drug for the treatment of liver fibrosis, and provide a certain experimental basis for the clinical application of CRC-CDs in the future.

AbstractChinese herbs contain substances that regulate female hormones. Our study confirmed that Zingiberis rhizoma carbonisata contains Zingiberis rhizoma-based carbon dots (ZR-CDs), which exert regulatory effects on serum oestradiol and FSH in mice and show impacts on endometrial growth and follicular development that potentially affect the ability of female fertility. ZR-CDs were characterized to clarify the microstructure, optical features, and functional group characteristics. It shows that ZR-CDs are spherical carbon nanostructures ranging from 0.97 to 2.3 nm in diameter, with fluorescent properties and a surface rich in functional groups. We further investigated the impact of ZR-CDs on oestradiol and FSH in serum, growth, and the development of ovarian and uterine using normal female mice and exogenous oestradiol intervention model. It was observed that ZR-CDs accelerated oestrogen metabolism and attenuated oestradiol-induced endometrial hyperplasia. Simultaneously, ZR-CDs triggered an increase in FSH, even in the presence of high-serum oestradiol that inhibits FSH secretion. Our findings suggest that ZR-CDs could be a potential therapeutic treatment for anovulatory menstruation.

Objective To evaluate the effectiveness of a machine learning based on computed tomography (CT) radiomics to distinguish nontuberculous mycobacterial pulmonary disease (NTM-PD) from pulmonary tuberculosis (PTB).Methods In this retrospective analysis, medical records of 99 individuals afflicted with NTM-PD and 285 individuals with PTB in Zhejiang Chinese and Western Medicine Integrated Hospital were examined. Random numbers generated by a computer were utilized to stratify the study cohort, with 80% designated as the training cohort and 20% as the validation cohort. A total of 2153 radiomics features were extracted using Python (Pyradiomics package) to analyse the CT characteristics of the large disease areas. The identification of significant factors was conducted through the least absolute shrinkage and selection operator (LASSO) regression. The following four supervised learning classifier models were developed: random forest (RF), support vector machine (SVM), logistic regression (LR), and extreme gradient boosting (XGBoost). For assessment and comparison of the predictive performance among these models, receiver-operating characteristic (ROC) curves and the areas under the ROC curves (AUCs) were employed.Results The Student’s t-test, Levene test, and LASSO algorithm collectively selected 23 optimal features. ROC analysis was then conducted, with the respective AUC values of the XGBoost, LR, SVM, and RF models recorded to be 1, 0.9044, 0.8868, and 0.7982 in the training cohort. In the validation cohort, the respective AUC values of the XGBoost, LR, SVM, and RF models were 0.8358, 0.8085, 0.87739, and 0.7759. The DeLong test results noted the lack of remarkable variation across the models.Conclusion The CT radiomics features can help distinguish between NTM-PD and PTB. Among the four classifiers, SVM showed a stable performance in effectively identifying these two diseases.

Quantitative assessment and trade-off/synergy analysis of land use multifunctions can effectively identify regional conflicts and dominant functions, providing decision support for promoting sustainable socio-economic and land use development. However, current research in this field still faces challenges due to coarse scale of studies and limited availability of accurate data. Taking Harbin City as a research case, this research employed an improved mutation level method, Pearson correlation analysis, and a multi-scale geographically weighted regression model to comprehensively investigate the profiling of land use multifunctions and their trade-off /synergy relationships. The comparative advantage theory was adopted to identify dominant functional zones using the NRCA index at a grid scale, in order to achieve a territorial spatial functional zoning delineation. The results showed that there were intricate trade-off/synergy relationships among production–living–ecology functions. Moreover, the types and intensity of trade-off/synergy evolved continuously with socio-economic development and regional resource endowment disparities. Due to its exceptional resource endowment, the agricultural dominated, urban dominated, and ecological dominated functional areas accounted for a significant proportion of 29%, 7%, and 26%, respectively. However, owing to the intricate trade-offs/synergies inherent in land use multifunctions, only a mere 2% (agricultural), 1% (urban), and 1% (ecological) of the area were identified as Optimization Guidance Zones. Conversely, Remediation Improvement Zones constituted the largest share at 63% of the total area, with agricultural, urban, and ecological Remediation Improvement Zones accounting for approximately 33%, 12%, and 18%, respectively. Based on the results of the type and intensity of trade-off/synergy among production–living–ecological functions, as well as the dominant zones and the integrated territorial spatial functional zoning delineation, this article provided targeted recommendations for the sustainable development of the region. These recommendations took into account both resource endowment and socio-economic development characteristics specific to the study area. The study aims to contribute to related research gaps, while providing valuable insights for other regional studies.

Background: Liver fibrosis represents an intermediate stage in the progression of liver disease, and as of now, there exists no established clinical therapy for effective antifibrotic treatment.Purpose: Our aim is to explore the impact of Carbon dots derived from Vaccaria Semen Carbonisata (VSC-CDs) on carbon tetrachloride-induced liver fibrosis in mice.Methods: VSC-CDs were synthesized employing a modified pyrolysis process. Comprehensive characterization was performed utilizing various techniques, including transmission electron microscopy (TEM), multiple spectroscopies, X-ray photoelectron spectroscopy (XPS), and high-performance liquid chromatography (HPLC). A hepatic fibrosis model induced by carbon tetrachloride was utilized to evaluate the anti-hepatic fibrosis effects of VSC-CDs.Results: VSC-CDs, exhibiting a quantum yield (QY) of approximately 2.08%, were nearly spherical with diameters ranging from 1.0 to 5.5 nm. The VSC-CDs prepared in this study featured a negative charge and abundant chemical functional groups. Furthermore, these particles demonstrated outstanding dispersibility in the aqueous phase and high biocompatibility. Moreover, VSC-CDs not only enhanced liver function and alleviated liver damage in pathomorphology but also mitigated the extent of liver fibrosis. Additionally, this study marks the inaugural demonstration of the pronounced activity of VSC-CDs in inhibiting inflammatory reactions, reducing oxidative damage, and modulating the TGF-β/Smad signaling pathway.Conclusion: VSC-CDs exerted significant potential for application in nanodrugs aimed at treating liver fibrosis.

Numerous natural compounds exhibit low bioavailability due to suboptimal water solubility. The solubilization methods of the modern pharmaceutical industry in contemporary pharmaceutical research are restricted by low efficiency, sophisticated technological requirements, and latent adverse effects. There is a pressing need to elucidate and implement a novel solubilizer to ameliorate these challenges. This study identified natural biomass-derived carbon dots as a promising candidate. We report on natural fluorescent carbon dots derived from Aurantia Fructus Immatures (AFI-CDs), which have exhibited a remarkable solubilization effect, augmenting naringin (NA) solubility by a factor of 216.72. Subsequent analyses suggest that the solubilization mechanism is potentially contingent upon the oration of a nanostructured complex (NA-AFI-CDs) between AFI-CDs and NA, mediated by intermolecular non-covalent bonds. Concomitantly, the synthesized NA-AFI-CDs demonstrated high biocompatibility, exceptional stability, and dispersion. In addition, NA-AFI-CDs manifested superior free radical scavenging capacity. This research contributes foundational insights into the solubilization mechanism of naringin-utilizing AFI-CDs and proffers a novel strategy that circumvents the challenges associated with the low aqueous solubility of water-insoluble drugs in the field of modern pharmaceutical science.

Abstract Background Acute kidney injury (AKI) refers to a tricky clinical disease, known by its high morbidity and mortality, with no real specific medicine for AKI. The carbonization product from Pollen Typhae (i.e., Pu-huang in China) has been extensively employed in clinic, and it is capable of relieving the renal damage and other diseases in China since acient times. Results Inspired by the carbonization process of Traditional Chinese Medicine (TCM), a novel species of carbon dots derived from Pollen Typhae (PT-CDs) was separated and then collected using a one-pot pyrolysis method. The as-prepared PT-CDs (4.85 ± 2.06 nm) with negative charge and abundant oxygenated groups exhibited high solubility, and they were stable in water. Moreover, the rhabdomyolysis (RM)-induced AKI rat model was used, and it was first demonstrated that PT-CDs had significant activity in improving the level of BUN and CRE, urine volume and kidney index, and histopathological morphology in RM-induced AKI rats. It is noteworthy that interventions of PT-CDs significantly reduced degree of inflammatory reaction and oxidative stress, which may be correlated with the basial potential mechanism of anti-AKI activities. Furthermore, cytotoxicity assay and biosafety evaluation exhibited high biocompatibility of PT-CDs. Conclusion This study offers a novel relieving strategy for AKI based on PT-CDs and suggests its potential to be a related candidate for clinical applications. Graphical Abstract

Introduction: Ulcerative colitis (UC) is a chronic and progressive inflammatory disease of the intestines. The primary symptoms, such as bloody diarrhea, can result in weight loss and significantly diminish the patient’s quality of life. Despite considerable research endeavors, this disease remains incurable. The scrambled Coptidis Rhizoma (SCR) has a rich historical background in traditional Chinese medicine as a remedy for UC. Drawing from a wealth of substantial clinical practices, this study is focused on investigating the protective effects and underlying mechanisms of the active component of SCR, namely SCR-based carbon dots (SCR-CDs), in the treatment of UC.Methods: SCR-CDs were extracted and isolated from the decoction of SCR, followed by a comprehensive characterization of their morphological structure and functional groups. Subsequently, we investigated the effects of SCR-CDs on parameters such as colonic length, disease activity index, and histopathological architecture using the dextran sulfate sodium (DSS)-induced colitis mice model. Furthermore, we delved into the assessment of key aspects, including the expression of intestinal tight junction (TJ) proteins, inflammatory cytokines, oxidative stress markers, and gut microbial composition, to unravel the intricate mechanisms underpinning their therapeutic effects.Results: SCR-CDs displayed a consistent spherical morphology, featuring uniform dispersion and diameters ranging from 1.2 to 2.8 nm. These SCR-CDs also exhibited a diverse array of surface chemical functional groups. Importantly, the administration of SCR-CDs, particularly at higher dosage levels, exerted a noteworthy preventive influence on colonic shortening, elevation of the disease activity index and colonic tissue impairment caused by DSS. These observed effects may be closely associated with the hygroscopic capability and hemostatic bioactivity inherent to SCR-CDs. Concurrently, the application of SCR-CDs manifested an augmenting impact on the expression of intestinal TJ proteins, concomitantly leading to a significant reduction in inflammatory cell infiltration and amelioration of oxidative stress. Additionally, SCR-CDs treatment facilitated the restoration of perturbed gut microbial composition, potentially serving as a fundamental mechanism underlying their observed protective effects.Conclusion: This study demonstrates the significant therapeutic potential of SCR-CDs in UC and provides elucidation on some of their mechanisms. Furthermore, these findings hold paramount importance in guiding innovative drug discovery for anti-UC agents.

Introduction:Fuligo Plantae (FP), the ash that sticks to the bottom of pots or chimneys after weeds burn, has long been used for its hemostatic effects and treatment of gastrointestinal bleeding. Nevertheless, the active ingredient of FP still needs to be further explored.Methods: The microstructure, optical and chemical properties of FP-CDs were characterized. An alcohol-induced gastric ulcer model was utilized to evaluate whether pre-administration of FP-CDs alleviated gastric bleeding symptoms and ameliorated gastric mucosal barrier disruption. In addition, the feces of each group of rats were extracted for 16S rDNA genome sequencing of intestinal flora.Results: FP-CDs with a diameter ranging from 1.4–3.2 nm had abundant chemical groups, which may be beneficial to the exertion of inherent activity. FP-CDs alleviated alcohol-induced gastric ulcer, as demonstrated by activating the extrinsic coagulation pathway, alleviating inflammation, and suppressing oxidative stress levels. More interestingly, FP-CDs can improve the diversity and dysbiosis of intestinal flora in rats with alcohol-induced gastric ulcer.Conclusion: These comes about illustrate the momentous inhibitory effects of FP-CDs on alcoholic gastric ulcer in rats, which give a modern methodology for investigating the effective ingredient of FP, and lay an experimental basis for the application of FP-CDs in the clinical treatment of alcoholic gastric ulcer.

Drought is a damaging and costly natural disaster that will become more serious in the context of global climate change in the future. Constructing a reliable drought risk assessment model and presenting its spatial pattern could be significant for agricultural production. However, agricultural drought risk mapping scientifically still needs more effort. Considering the whole process of drought occurrence, this study developed a comprehensive agricultural drought risk assessment model that involved all risk components (exposure, hazard, vulnerability and mitigation capacity) and their associated criteria using geospatial techniques and fuzzy logic. The comprehensive model was applied in Songnen Plain to justify its applicability. ROC and AUC techniques were applied to evaluate its efficiency, and the prediction rate was 88.6%. The similar spatial distribution of water resources further verified the model’s reliability. The southwestern Songnen Plain is a very-high-risk (14.44%) region, determined by a high vulnerability, very high hazardousness and very low mitigation capacity, and is the region that should be paid the most attention to; the central part is a cross-risk region of high risk (24.68%) and moderate risk (27.28%) with a serious disturbance of human agricultural activities; the northeastern part is a dry grain production base with a relatively optimal agricultural production condition of very low risk (22.12%) and low risk (11.48%). Different drought mitigation strategies should be adopted in different regions due to different drought causes. The findings suggest that the proposed model is highly effective in mapping comprehensive drought risk for formulating strong drought mitigation strategies and could be used in other drought-prone areas.
