Deep learning excels at managing spatial and temporal time series with variable patterns for streamflow forecasting, but traditional machine learning algorithms may struggle with complicated data, including non-linear and multidimensional complexity. Empirical heterogeneity within watersheds and limitations inherent to each estimation methodology pose challenges in effectively measuring and appraising hydrological statistical frameworks of spatial and temporal variables. This study emphasizes streamflow forecasting in the region of Johor, a coastal state in Peninsular Malaysia, utilizing a 28-year streamflow-pattern dataset from Malaysia’s Department of Irrigation and Drainage for the Johor River and its tropical rainforest environment. For this dataset, wavelet transformation significantly improves the resolution of lag noise when historical streamflow data are used as lagged input variables, producing a 6% reduction in the root-mean-square error. A comparative analysis of convolutional neural networks and artificial neural networks reveals these models’ distinct behavioral patterns. Convolutional neural networks exhibit lower stochasticity than artificial neural networks when dealing with complex time series data and with data transformed into a format suitable for modeling. However, convolutional neural networks may suffer from overfitting, particularly in cases in which the structure of the time series is overly simplified. Using Bayesian neural networks, we modeled network weights and biases as probability distributions to assess aleatoric and epistemic variability, employing Markov chain Monte Carlo and bootstrap resampling techniques. This modeling allowed us to quantify uncertainty, providing confidence intervals and metrics for a robust quantitative assessment of model prediction variability.

In recent years, electromagnetic pollution has become more and more serious, resulting in a very negative impact on people’s health. Therefore, it is important to develop efficient microwave absorbers to reduce electromagnetic pollution. Here, we construct a novel absorbing material of the polymer gel-derived porous carbon decorated by rare earth compounds (Ce (CO3) OH). When the thickness is 2.2 mm, the composite exhibits excellent microwave absorption performance with the optimal RLmin value and EAB reached up to −47.67 dB and 5.52 GHz, respectively, covering the Ku band. The high-efficiency microwave absorption is mainly attributed to the synergistic effect of dipole polarization, defect polarization and interfacial polarization. This work not only provides a new view for designing superior absorber materials, but also lay a foundation for their real applications.

Human esophageal cancer has a global impact on human health due to its high incidence and mortality. Therefore, there is an urgent need to develop new drugs to treat or prevent the prominent pathological subtype of esophageal cancer, esophageal squamous cell carcinoma (ESCC). Based upon the screening of drugs approved by the Food and Drug Administration, we discovered that Arbidol could effectively inhibit the proliferation of human ESCC in vitro. Next, we conducted a series of cell-based assays and found that Arbidol treatment inhibited the proliferation and colony formation ability of ESCC cells and promoted G1-phase cell cycle arrest. Phosphoproteomics experiments, in vitro kinase assays and pull-down assays were subsequently performed in order to identify the underlying growth inhibitory mechanism. We verified that Arbidol is a potential ataxia telangiectasia and Rad3-related (ATR) inhibitor via binding to ATR kinase to reduce the phosphorylation and activation of minichromosome maintenance protein 2 at Ser108. Finally, we demonstrated Arbidol had the inhibitory effect of ESCC in vivo by a patient-derived xenograft model. All together, Arbidol inhibits the proliferation of ESCC in vitro and in vivo through the DNA replication pathway and is associated with the cell cycle.

Abstract Model Intercomparison Projects (MIPs) are fundamental to our understanding of how the land surface responds to changes in climate. However, MIPs are challenging to conduct, requiring the organization of multiple, decentralized modeling teams throughout the world running common protocols. We explored centralizing these models on a single supercomputing system. We ran nine offline terrestrial biosphere models through the Terrestrial Biosphere Model Farm: CABLE, CENTURY, HyLand, ISAM, JULES, LPJ‐GUESS, ORCHIDEE, SiB‐3, and SiB‐CASA. All models were wrapped in a software framework driven with common forcing data, spin‐up, and run protocols specified by the Multi‐scale Synthesis and Terrestrial Model Intercomparison Project (MsTMIP) for years 1901–2100. We ran more than a dozen model experiments. We identify three major benefits and three major challenges. The benefits include: (a) processing multiple models through a MIP is relatively straightforward, (b) MIP protocols are run consistently across models, which may reduce some model output variability, and (c) unique multimodel experiments can provide novel output for analysis. The challenges are: (a) technological demand is large, particularly for data and output storage and transfer; (b) model versions lag those from the core model development teams; and (c) there is still a need for intellectual input from the core model development teams for insight into model results. A merger with the open‐source, cloud‐based Predictive Ecosystem Analyzer (PEcAn) ecoinformatics system may be a path forward to overcoming these challenges.

The flower pollination algorithm (FPA) is a novel heuristic optimization algorithm inspired by the pollination behavior of flowers in nature. However, the global and local search processes of the FPA are sensitive to the search direction and parameters. To solve this issue, an improved flower pollination algorithm based on cosine cross-generation differential evolution (FPA-CCDE) is proposed. The algorithm uses cross-generation differential evolution to guide the local search process, so that the optimal solution is achieved and sets cosine inertia weights to increase the search convergence speed. At the same time, the external archiving mechanism and the adaptive adjustment of parameters realize the dynamic update of scaling factor and crossover probability to enhance the population richness as well as reduce the number of local solutions. Then, it combines the cross-generation roulette wheel selection mechanism to reduce the probability of falling into the local optimal solution. In comparing to the FPA-CCDE with five state-of-the-art optimization algorithms in benchmark functions, we can observe the superiority of the FPA-CCDE in terms of stability and optimization features. Additionally, we further apply the FPA-CCDE to solve the robot path planning issue. The simulation results demonstrate that the proposed algorithm has low cost, high efficiency, and attack resistance in path planning, and it can be applied to a variety of intelligent scenarios.

Esophageal squamous cell carcinoma (ESCC) and esophageal adenocarcinoma (EAC) are the two major types of esophageal cancer (EC). ESCC accounts for 90% of EC. Recurrence after primary treatment is the main reason for poor survival. Therefore, recurrence prevention is a promising strategy for extending the 5-year survival rate. Here, we found tegaserod maleate could inhibit ESCC proliferation both in vivo and in vitro. Proteomics analysis revealed that tegaserod maleate suppressed the peroxisome signaling pathway, in which the key molecules peroxisome membrane protein 11B (PEX11B) and peroxisome membrane protein 13 (PEX13) were downregulated. The immunofluorescence, catalase activity assay, and reactive oxygen species (ROS) confirmed that downregulation of these proteins was related to impaired peroxisome function. Furthermore, we found that PEX11B and PEX13 were highly expressed in ESCC, and knockout of PEX11B and PEX13 further demonstrated the antitumor effect of tegaserod maleate. Importantly, tegaserod maleate repressed ESCC tumor growth in a patient-derived xenograft (PDX) model in vivo. Our findings conclusively demonstrated that tegaserod maleate inhibits the proliferation of ESCC by suppressing the peroxisome pathway.

Knowledge about the quality of data and metadata is important to support informed decisions on the (re)use of individual datasets and is an essential part of the ecosystem that supports open science. Quality assessments reflect the reliability and usability of data. They need to be consistently curated, fully traceable, and adequately documented, as these are crucial for sound decision- and policy-making efforts that rely on data. Quality assessments also need to be consistently represented and readily integrated across systems and tools to allow for improved sharing of information on quality at the dataset level for individual quality attribute or dimension. Although the need for assessing the quality of data and associated information is well recognized, methodologies for an evaluation framework and presentation of resultant quality information to end users may not have been comprehensively addressed within and across disciplines. Global interdisciplinary domain experts have come together to systematically explore needs, challenges and impacts of consistently curating and representing quality information through the entire lifecycle of a dataset. This paper describes the findings of that effort, argues the importance of sharing dataset quality information, calls for community action to develop practical guidelines, and outlines community recommendations for developing such guidelines. Practical guidelines will allow for global access to and harmonization of quality information at the level of individual Earth science datasets, which in turn will support open science.

Information about data quality helps potential data users to determine whether and how data can be used and enables the analysis and interpretation of such data. Providing data quality information improves opportunities for data reuse by increasing the trustworthiness of the data. Recognizing the need for improving the quality of citizen science data, we describe quality assessment and quality control (QA/QC) issues for these data and offer perspectives on aspects of improving or ensuring citizen science data quality and for conducting research on related issues.

Esophageal squamous cell carcinoma (ESCC) has a worldwide impact on human health, due to its high incidence and mortality. Therefore, identifying compounds to increase patients' survival rate is urgently needed. Mefloquine (MQ) is an FDA-approved anti-malarial drug, which has been reported to inhibit cellular proliferation in several cancers. However, the anti-tumor activities of the drug have not yet been completely defined. In this study, mass spectrometry was employed to profile proteome changes in ESCC cells after MQ treatment. Sub-cellular localization and gene ontology term enrichment analysis suggested that MQ treatment mainly affect mitochondria. The KEGG pathway enrichment map of down-regulated pathways and Venn diagram indicated that all of the top five down regulated signaling pathways contain four key mitochondrial proteins (succinate dehydrogenase complex subunit C (SDHC), succinate dehydrogenase complex subunit D, mitochondrially encoded cytochrome c oxidase III and NADH: ubiquinone oxidoreductase subunit V3). Meanwhile, mitochondrial autophagy was observed in MQ-treated KYSE150 cells. More importantly, patient-derived xenograft mouse models of ESCC with SDHC high expression were more sensitive to MQ treatment than low SDHC-expressing xenografts. Taken together, mefloquine inhibits ESCC tumor growth by inducing mitochondrial autophagy and SDHC plays a vital role in MQ-induced anti-tumor effect on ESCC.

The seasonal-cycle amplitude (SCA) of the atmosphere–ecosystem carbon dioxide (CO2) exchange rate is a useful metric of the responsiveness of the terrestrial biosphere to environmental variations. It is unclear, however, what underlying mechanisms are responsible for the observed increasing trend of SCA in atmospheric CO2 concentration. Using output data from the Multi-scale Terrestrial Model Intercomparison Project (MsTMIP), we investigated how well the SCA of atmosphere–ecosystem CO2 exchange was simulated with 15 contemporary terrestrial ecosystem models during the period 1901–2010. Also, we made attempt to evaluate the contributions of potential mechanisms such as atmospheric CO2, climate, land-use, and nitrogen deposition, through factorial experiments using different combinations of forcing data. Under contemporary conditions, the simulated global-scale SCA of the cumulative net ecosystem carbon flux of most models was comparable in magnitude with the SCA of atmospheric CO2 concentrations. Results from factorial simulation experiments showed that elevated atmospheric CO2 exerted a strong influence on the seasonality amplification. When the model considered not only climate change but also land-use and atmospheric CO2 changes, the majority of the models showed amplification trends of the SCAs of photosynthesis, respiration, and net ecosystem production (+0.19 % to +0.50 % yr−1). In the case of land-use change, it was difficult to separate the contribution of agricultural management to SCA because of inadequacies in both the data and models. The simulated amplification of SCA was approximately consistent with the observational evidence of the SCA in atmospheric CO2 concentrations. Large inter-model differences remained, however, in the simulated global tendencies and spatial patterns of CO2 exchanges. Further studies are required to identify a consistent explanation for the simulated and observed amplification trends, including their underlying mechanisms. Nevertheless, this study implied that monitoring of ecosystem seasonality would provide useful insights concerning ecosystem dynamics.

YesWorkflow requires neither the use of a workflow engine nor the overhead of adapting code to run effectively in such a system. Instead, YesWorkflow enables scientists to annotate existing scripts with special comments that reveal the computational modules and dataflows otherwise implicit in these scripts. YesWorkflow tools extract and analyze these comments, represent the scripts in terms of entities based on the typical scientific workflow model, and provide graphical renderings of this workflow-like view of the scripts. However, in order to perform such analyses on scientific results as part of extended research collaborations, an adequate environment and tools are required. Concretely, the need arises for a repository that will facilitate the sharing of scientific workflows and their associated execution traces in an interoperable manner, also enabling querying and visualization. Furthermore, such functionality should be supported while taking performance and scalability into account. With this purpose in mind, we introduce PBase: a scientific workflow provenance repository implementing the ProvONE proposed standard, which extends the emerging W3C PROV standard for provenance data with workflow specific concepts. PBase is built on the Neo4j graph database, thus offering capabilities such as declarative and efficient querying. Our experiences demonstrate the power gained by supporting various types of queries for provenance data. In addition, PBase is equipped with a user friendly interface tailored for the visualization of scientific workflow provenance data, making the specification of queries and the interpretation of their results easier and more effective.

Understanding what drives the interannual variability (IAV) of the land carbon sink is crucial for improving future predictions of this important, yet uncertain, component of the climate system. While drivers of global and hemispheric-scale net ecosystem exchange (NEE) IAV have been investigated, our understanding of the drivers of NEE IAV at regional scales (e.g. sub-continental, biome-level) is quite poor. Here we explore the biome-level attribution and drivers of North American NEE using inverse estimates derived from a dense network of atmospheric CO _2 observations. We find that deciduous broadleaf and mixed forests are the primary regions responsible for North American NEE IAV, which differs from the ecoregions identified for the globe and Northern Hemisphere. We also find that a suite of terrestrial biosphere models (TBMs) do not agree on the dominant biome contributing to NEE IAV, with TBMs falling along an apparent spectrum ranging between those with IAV dominated primarily by forested ecosystems to those with IAV dominated by non-forested ecosystems. Furthermore, this regional trade-off in TBM NEE IAV is found to be linked to differing regional responses to environmental drivers among TBMs. This work displays the importance of extra-tropical forests in driving continental NEE IAV and also highlights the challenges and limitations of using TBMs to inform regional-scale carbon flux dynamics.

Scientific workflows and their supporting systems are becoming increasingly popular for compute-intensive and data-intensive scientific experiments. The advantages scientific workflows offer include rapid and easy workflow design, software and data reuse, scalable execution, sharing and collaboration, and other advantages that altogether facilitate “reproducible science”. In this context, provenance – information about the origin, context, derivation, ownership, or history of some artifact – plays a key role, since scientists are interested in examining and auditing the results of scientific experiments. However, in order to perform such analyses on scientific results as part of extended research collaborations, an adequate environment and tools are required. Concretely, the need arises for a repository that will facilitate the sharing of scientific workflows and their associated execution traces in an interoperable manner, also enabling querying and visualization. Furthermore, such functionality should be supported while taking performance and scalability into account. With this purpose in mind, we introduce PBase: a scientific workflow provenance repository implementing the ProvONE proposed standard, which extends the emerging W3C PROV standard for provenance data with workflow specific concepts. Climate variability associated with the El Niño-Southern Oscillation (ENSO) and its consequent impacts on land carbon sink interannual variability have been used as a basis for investigating carbon cycle responses to climate variability more broadly, and to inform the sensitivity of the tropical carbon budget to climate change. Past studies have presented opposing views about whether temperature or precipitation is the primary factor driving the response of the land carbon sink to ENSO. To address the need for published data, considerable effort has gone into formalizing the process of data publication. From funding agencies to publishers, data publication has rapidly become a requirement. Digital Object Identifiers (DOI) and data citations have enhanced the integration and availability of data. The challenge facing data publishers now is to deal with the increased number of publishable data products and most importantly the difficulties of publishing diverse data products into an online archive. The Oak Ridge National Laboratory Distributed Active Archive Center (ORNL DAAC), a NASA-funded data center, faces these challenges as it deals with data products created by individual investigators. This paper summarizes the challenges of curating data and provides a summary of a workflow solution that ORNL DAAC researcher and technical staffs have created to deal with publication of the diverse data products. The workflow solution presented here is generic and can be applied to data from any scientific domain and data located at any data center.

We examined natural and anthropogenic controls on terrestrial evapotranspiration (ET) changes from 1982 to 2010 using multiple estimates from remote sensing-based datasets and process-oriented land surface models. A significant increasing trend of ET in each hemisphere was consistently revealed by observationally-constrained data and multi-model ensembles that considered historic natural and anthropogenic drivers. The climate impacts were simulated to determine the spatiotemporal variations in ET. Globally, rising CO _2 ranked second in these models after the predominant climatic influences, and yielded decreasing trends in canopy transpiration and ET, especially for tropical forests and high-latitude shrub land. Increasing nitrogen deposition slightly amplified global ET via enhanced plant growth. Land-use-induced ET responses, albeit with substantial uncertainties across the factorial analysis, were minor globally, but pronounced locally, particularly over regions with intensive land-cover changes. Our study highlights the importance of employing multi-stream ET and ET-component estimates to quantify the strengthening anthropogenic fingerprint in the global hydrologic cycle.
