High blood sugar is the main feature of diabetes. In order to effectively regulate blood glucose levels, in this study, we used Gracilaria as raw material and found that 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) were beneficial for decreasing fasting blood glucose, improving insulin resistance, enhancing the function of islet β-cells, and regulating the abnormal increase of TC, TG, LDL-c caused by hyperglycemia in diabetic mice. Moreover, 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) affected the gut microbiota structure of diabetic mice, decreased the Firmicutes/Bacteroidetes ratio, increased the abundance of Lactobacillus and Collinsella etc. and reduced the abundance of Helicobacter and Oscillibacter etc. In addition, the content of acetic acid and butyric acid in metabolites of short-chain fatty acids had increased, which had the effect of regulating blood glucose level. The results have showed that 70% ethanol extract of Gracilaria (GE) and complex protease hydrolysate of Gracilaria (GH) are potential marine foods for maintaining blood sugar health.

ObjectiveTo investigate ear canal microflora’s structure, composition and function in patients with chronic bacterial otitis externa.MethodsA case-control study design method was used to collect the ear canal secretions from 14 patients with chronic bacterial external otitis (CB group) and 14 healthy controls (H group) treated in the Department of Otolaryngology and Head and Neck Surgery in the First Affiliated Hospital of Hainan Medical University. 16S rRNA high-throughput sequencing technology was used to sequence the ear canal microflora’s V3 ~ V4 region gene amplification products in the participating population. The α diversity of ear canal microflora in 2 groups was analyzed. Based on the weighted Unifrac distance, principal coordinate analysis was performed to compare the β diversity of ear tract microflora between the two groups. The differences in ear microflora at phylum and genus levels were analyzed. PICRUSt2 function prediction and BugBase phenotype prediction were also performed.Resultsα diversity analysis showed that the diversity and richness of auricular microflora in the CB group were significantly lower than those in the H group. β diversity analysis showed that there were some differences between the two groups. At the phylum level, the relative abundance of Bdellovibrionota, Campylobacterota, and WPS-2 in the microbiota of patients in the CB group was significantly lower than that in the H group, and the differences were statistically significant. At the genus level, the relative abundance of Pseudomonas, Acinetobacter, Pelomonas, Sphingomonas, Bradyrhizobium, Brevundimonas, Enhydrobacter, Actinomyces, Paracoccus and Chryseobacterium in the ear canal of Group H is significantly higher than that of Group CB. Functional prediction of PICRUSt2 suggests that amino acid biosynthesis and bacterial microbiota may be related. In BugBase phenotypic prediction, the contribution of aerobic phenotype in group CB was significantly lower than that in group H.ConclusionThe diversity and abundance of the ear canal flora of patients with chronic bacterial otitis externa were significantly lower than those of the healthy population, and their bacterial colony structure was significantly altered. Dysbiosis of the ear canal flora may be an important cause of chronic bacterial otitis externa.

Abstract In this study, according to the Location, Diameter, Risk factor (LDRf) classification principle, the influence and effect of esophageal varices diameter on the degree of complete ligation of multicyclic ligator were investigated. Methods. The esophageal veins of healthy piglets were filled with methylene blue solution, and the in vitro pig esophageal varices model was made, which were divided into three groups according to the diameter of esophageal varices: D1, 0.4–1.0 cm; D2, 1.1–1.5 cm; and D3, 1.6–2.0 cm. Finally, the ligation effect of each group was analyzed statistically. A total of 407 ligations were performed on the simulated esophageal variceal model. There were 103 ligations in the D1 group and 98 were complete (95.15%, 98/103); 151 ligations in the D2 group and 47 were complete (31.13%, 47/151); and 153 ligations in the D3 group but none were complete (0%, 0/153). There was significant difference in the degree of complete ligation between the two groups (χ2 = 38.0014, P ≤ 0.001). In the varicose ligation model, the complete ligation effect was the most complete and robust when the varicose diameter was 0.4–1.0 cm. This study showed that the varicose vein diameter in LDRf classification was reasonable and feasible to guide endoscopic varicose vein ligation.

Abstract This letter presents a sub‐6 GHz wideband low noise amplifier (LNA) based on a double L‐type load network and utilizes the negative feedback technique. Using the cascode structure, along with the aforementioned techniques, it is possible to achieve a single‐stage wideband LNA with high gain and low noise figure (NF). Fabricated in 110‐nm SOI CMOS technology, the proposed LNA achieves a maximum power gain of 15.2 dB, noise figure of 1.0–1.56 dB. The 3‐dB bandwidth ranges from 3.05 to 4.55 GHz. The minimum power input at 1 dB compression point (IP1dB) is −17.1 dBm. The LNA area is 0.18 mm2 and dissipates a total power of 11.5 mW from a 1.4‐V power supply.

This study aimed to assess the hypoglycemic efficacy of low molecular weight polysaccharides fractions obtained from Laminaria japonica (LJOO) in a model of type 2 diabetes mellitus (T2DM) constructed using mice. Biochemical parameters were measured after 4 weeks of continuous gavage, and fasting blood glucose (FBG) concentrations were analyzed. Pathological changes in tissues were assessed. The intestinal contents were obtained for 16S rDNA high-throughput sequencing analysis and detection of short-chain fatty acids (SCFAs). LJOO lowered FBG and insulin concentrations. It altered the gut microbiota composition, as evidenced by enriched probiotic bacteria, along with an increase in the Bacteroidetes/Firmicutes ratio and a decrease in the population of harmful bacteria. LJOO stimulated the growth of SCFA—producing bacteria, thereby increasing cecal SCFAs levels. LJOO can potentially aid in alleviating T2DM and related gut microbiota dysbiosis. LJOO may be used as a food supplement for patients with T2DM.

Let R be a ring with nonzero identity. The unit graph of R is a simple graph whose vertex set is R itself and two distinct vertices are adjacent if and only if their sum is a unit of R. In this paper, we study the radius of unit graphs of rings. We prove that there exists a ring R such that the radius of unit graph can be any given positive integer. We also prove that the radius of unit graphs of self-injective rings are 1, 2, 3, ∞. We classify all self-injective rings via the radius of its unit graph. The radius of unit graphs of some ring extensions are also considered.

Background: The DTaP-IPV/Hib combination vaccine can replace the acellular tetanus vaccine, polio vaccine, and the Haemophilus influenzae type B vaccine. Data on the safety of DTaP-IPV/Hib vaccines are required. We aimed to evaluate the safety of the vaccination program. Methods: Using the National Adverse Events Following Immunization (AEFI) surveillance system (CNAEFIS) in Guangzhou, China, a retrospective study was performed from May 11, 2011, to December 31, 2017. There were 376 cases of adverse events after vaccination with the DTaP IPV/Hib vaccine. The primary analysis indicators were the number of vaccines used, the number of AEFI reports received, and the reporting rate (per 100,000). Results: From May 1, 2011, to December 31, 2017, 516,000 doses of vaccine were inoculated, and 376 cases of adverse reactions were reported; the reporting rate was 72.8 per 100,000 vaccines. There were eight cases of serious AEFIs (1.5 per 100,000), with four cases of thrombocytopenic purpura (0.8 per 100,000); three cases of cyanosis of the lips, stiffness, and flexion of limbs, and convulsions (0.6 per 100,000); and one case of a high fever (0.2 per 100,000). The highest incidence of AEFIs occurred after the fourth dose (n = 207, 55.0%, 40.1 per 100,000), followed by the first dose (n = 81, 21.5%, 15.7 per 100,000), second dose (n = 48, 12.8%, 9.3 per 100,000) and third dose (n = 40, 10.6%, 7.7 per 100,000). The AEFI incidence was higher after injection of the vaccine into the deltoid muscle of the upper arm (n = 276, 73.4%, 53.5 per 100,000) than after injection of the vaccine into the thigh (n = 100, 26.6%, 19.4 per 100,000). There was a significant difference between AEFIs after injection into the deltoid of the upper arm deltoid and the thigh (x2 = 164.8, P < 0.05). Conclusions: Most of the reported AEFIs after DTaP-IPV/Hib vaccination are not serious. There were four cases of TP in this study; vaccination may be a rare cause of thrombocytopenic purpura.

From 2017 to 2018, Guangzhou experienced a shortage in 3–6 types of National Immunization Program (NIP) vaccines. To evaluate the current situation and causes of the NIP vaccine shortage, we analysed the duration, intensity and causes of the shortage from data in the vaccine information system of the Guangzhou Center for Disease Control and Prevention; we also proposed countermeasures to resolve the shortage. In 2017, there were shortages of three types of NIP vaccines in Guangzhou. The most affected vaccines were inactivated poliovirus (IPV) vaccine and meningococcal group AC (MenAC) vaccine, accounting for 39.5% and 16.5% of the reported shortages, respectively. In 2018, the shortage extended to six vaccine types; the most affected were diphtheria, tetanus and pertussis (DTP) vaccine, MenAC vaccine, and Bacille Calmette Guerin (BCG) vaccine. The four main causes for the shortage were: 1) an insufficient production capacity; 2) a delay in batch issuance; 3) vaccine-related events; and 4) an extended bidding procurement cycle. Four solutions are proposed: 1) expand production output; 2) exempt creditworthy enterprises from batch inspections; 3) establish alternative enterprises and emergency use authorizations; and 4) establish public health funds and stockpile storage systems.

In order to improve the temperature stability of DC-contact RF MEMS switch, a thermal buckle-beam structure is implemented. The stability of the switch pull-in voltage versus temperature is not only improved, but also the impact of stress and stress gradient on the drive voltage is suppressed. Test results show that the switch pull-in voltage is less sensitive to temperature between -20 °C and 100 °C. The variable rate of pull-in voltage to temperature is about -120 mV/°C. The RF performance of the switch is stable, and the isolation is almost independent of temperature. After being annealed at 280 °C for 12 hours, our switch samples, which are suitable for packaging, have less than 1.5% change in the rate of pull-in voltage.

Enterovirus 71 (EV71) is a neurotropic virus capable of inducing severe neurological symptoms and death. No direct targeting antivirals are useful in the treatment of severe EV71 infection. Because of low toxicity and good specificity, monoclonal antibodies (MAb) are a potential candidate for the treatment of viral infections. Therefore, we developed an EV71-specific conformational MAb with high in vitro cross-neutralization activity to heterologous EV71 subgenotypes. The in vivo treatment experiment at different days post-infection indicated that a single treatment of MAb CT11F9 within day 3 post-infection fully protected mice from morbidity and mortality (0% PBS vs. 100% at 10 µg/g per body weight ***P

Highlights Steel tip arrays were used as current collectors to replace planar conductors. Optimal flow and electric fields reduced barriers for electron and ion transport. Desalination performance of flow-electrode capacitive deionization is enhanced by the tip-array current collectors.

Microorganisms from poorly explored environments are promising sources for the development of novel drugs. In our continuous efforts to screen for mangrove actinomycetes that produce metabolites with potential pharmaceutical applications, Streptomyces sp. Y009 was isolated from mangrove sediments in Guangxi, China. The phenotypic, physiological, biochemical, and phylogenetic characteristics of this strain were investigated. Analysis of phylogenetic and 16S rRNA gene sequences showed that it had the highest sequence similarity to Streptomyces thermolilacinus NBRC 14274 (98.95%). Further, the Y009 extract exhibited antioxidant activity, as indicated by DPPH and superoxide dismutase assays. The extract showed broad-spectrum and potent anticancer potential against six human cancer cell lines, with IC50 values ranging from 5.61 to 72.15 μg/mL. Furthermore, the selectivity index (SI) demonstrated that the Y009 extract exhibited less toxicity toward normal cell lines in comparison to the lung cancer cell line (A549) and hepatoma cell line (HepG2). GC–MS analysis revealed that the extract contained some biologically important secondary metabolites, mainly cyclic dipeptides and esters, which might be responsible for the antioxidant and anticancer properties. 3-Isobutylhexahydropyrrolo[1,2-a]pyrazine-1,4-dione (28.32%) was the major chemical compound available in the extract. The effect on cancer cells was then confirmed using nuclear staining and in silico docking. This study suggests that further exploration of the bioactive compounds of the newly isolated strain may be a promising approach for the development of novel chemopreventive drugs.

Organic light-emitting diodes (OLEDs) have demonstrated remarkable advancements in both device lifetime and luminous efficiency. However, insufficient operation lifetime due to device degradation remains a major hurdle, especially for brighter devices. Understanding the degradation mechanisms of OLEDs due to the degradation of functional materials and the formation of defects in device architectures continues to be a significant challenge. Herein, we evaluate the degradation characteristics by scrutinizing the electrical and optical properties, as well as analyzing the charge carrier dynamics in pristine and aged states of phosphorescent OLEDs (PhOLEDs). We show that degradation mechanisms in PhOLEDs can be elucidated in terms of the ideality factors of current and luminance in pristine and aged device states. The consistent shifts in distinct ideality factors across various states points out that the device degradation is attributed to the deterioration of the guest material, i.e. green-light-emitting phosphorescent material. Conversely, the incongruity in ideality factor changes between the two states indicates that the degradation results from the deterioration of non-light-emitting material. Subsequent characterization experiments provide further evidence that this degradation is primarily attributed to the deterioration of CBP-host material. The thorough understanding of degradation mechanisms established in this study can contribute to realizing the highly reliable PhOLEDs with a long lifetime.

Abstract Memristors offer a promising solution to address the performance and energy challenges faced by conventional von Neumann computer systems. Yet, stochastic ion migration in conductive filament often leads to an undesired performance tradeoff between memory window, retention, and endurance. Herein, a robust memristor based on oxygen‐rich SnO2 nanoflowers switching medium, enabled by seed‐mediated wet chemistry, to overcome the ion migration issue for enhanced analog in‐memory computing is reported. Notably, the interplay between the oxygen vacancy (Vo) and Ag ions (Ag+) in the Ag/SnO2/p++‐Si memristor can efficiently modulate the formation and abruption of conductive filaments, thereby resulting in a high on/off ratio (>106), long memory retention (10‐year extrapolation), and low switching variability (SV = 6.85%). Multiple synaptic functions, such as paired‐pulse facilitation, long‐term potentiation/depression, and spike‐time dependent plasticity, are demonstrated. Finally, facilitated by the symmetric analog weight updating and multiple conductance states, a high image recognition accuracy of ≥ 91.39% is achieved, substantiating its feasibility for analog in‐memory computing. This study highlights the significance of synergistically modulating conductive filaments in optimizing performance trade‐offs, balancing memory window, retention, and endurance, which demonstrates techniques for regulating ion migration, rendering them a promising approach for enabling cutting‐edge neuromorphic applications.

Cytochrome P450 (CYP) enzymes are crucial for the detoxification of xenobiotics, cellular metabolism, and homeostasis. This study investigated the molecular characterization of CYP enzymes in the black-spotted frog, Pelophylax nigromaculatus, and examined the regulation of CYP expression in response to chronic exposure to the antibiotic sulfamethoxazole (SMX) at various environmental concentrations (0, 1, 10, and 100 μg/L). The full-length cDNA of Pn-CYP26B1 was identified. The sequence included open reading frames of 1,536 bp, encoding proteins comprising 511 amino acids. The signature motif, FxxGxxxCxG, was highly conserved when compared with a number of selected animal species. SMX significantly upregulated the expression of the protein CYP26B1 in frog livers at concentrations of 1 and 10 μg/L. SMX showed an affinity for CYP26B1 of −7.6 kcal/mol, indicating a potential mechanism for SMX detoxification or adaptation of the frog. These findings contributed to our understanding of the environmental impact of antibiotics on amphibian species and underscored the importance of CYP enzymes in maintaining biochemical homeostasis under exposure to xenobiotic stress.

Abstract Durable superhydrophobic anti‐erosion/anticorrosion coatings are highly demanded across various applications. However, achieving coatings with exceptional superhydrophobicity, mechanical strength, and corrosion resistance remains a grand challenge. Herein, a robust microstructure coating, inspired by the cylindrical structures situated on the surface of conch shell, for mitigating erosion and corrosion damages in gas transportation pipelines is reported. Specifically, citric acid monohydrate as a pore‐forming agent is leveraged to create a porous structure between layers, effectively buffering the impact on the surface. As a result, the coating demonstrates remarkable wear resistance and water repellency. Importantly, even after abrasion by sandpaper and an erosion loop test, the resulting superhydrophobic surfaces retain the water repellency. The design strategy offers a promising route to manufacturing multifunctional materials with desired features and structural complexities, thereby enabling effective self‐cleaning and antifouling abilities in harsh operating environments for an array of applications, including self‐cleaning windows, antifouling coatings for medical devices, and anti‐erosion/anticorrosion protection, among other areas.

BackgroundLiver cancer, especially hepatocellular carcinoma (HCC), remains a significant global health challenge. Traditional prognostic indicators for HCC often fall short in providing comprehensive insights for individualized treatment. The integration of genomics and radiomics offers a promising avenue for enhancing the precision of HCC diagnosis and prognosis.MethodsFrom the Cancer Genome Atlas (TCGA) database, we categorized mRNA of HCC patients by Forkhead Box M1 (FOXM1) expression and performed univariate and multivariate studies to pinpoint autonomous HCC risk factors. We deployed subgroup, correlation, and interaction analyses to probe FOXM1’s link with clinicopathological elements. The connection between FOXM1 and immune cells was evaluated using the CIBERSORTx database. The functions of FOXM1 were investigated through analyses of Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG). After filtering through TCGA and the Cancer Imaging Archive (TCIA) database, we employed dual-region computed tomography (CT) radiomics technology to noninvasively predict the mRNA expression of FOXM1 in HCC tissues. Radiomic features were extracted from both tumoral and peritumoral regions, and a radiomics score (RS) was derived. The performance and robustness of the constructed models were evaluated using 10-fold cross-validation. A radiomics nomogram was developed by incorporating RS and clinical variables from the TCGA database. The models’ discriminative abilities were assessed using metrics such as the area under the curve (AUC) of the receiver operating characteristic curves (ROC) and precision-recall (PR) curves.ResultsOur findings emphasized the overexpression of FOXM1 as a determinant of poor prognosis in HCC and illustrated its impact on immune cell infiltration. After selecting arterial phase CT, we chose 7 whole-tumor features and 3 features covering both the tumor and its surroundings to create WT and WP models for FOXM1 prediction. The WT model showed strong predictive capabilities for FOXM1 expression by PR curve. Conversely, the WP model did not demonstrate the good predictive ability. In our study, the radiomics score (RS) was derived from whole-tumor regions on CT images. The RS was significantly associated with FOXM1 expression, with an AUC of 0.918 in the training cohort and 0.837 in the validation cohort. Furthermore, the RS was correlated with oxidative stress genes and was integrated with clinical variables to develop a nomogram, which demonstrated good calibration and discrimination in predicting 12-, 36-, and 60-month survival probabilities. Additionally, bioinformatics analysis revealed FOXM1’s potential role in shaping the immune microenvironment, with its expression linked to immune cell infiltration.ConclusionThis study highlights the potential of integrating FOXM1 expression and radiomics in understanding HCC’s complexity. Our approach offers a new perspective in utilizing radiomics for non-invasive tumor characterization and suggests its potential in providing insights into molecular profiles. Further research is needed to validate these findings and explore their clinical implications in HCC management.

Abstract Hollow structuring has been identified as an effective strategy to enhance the cycling stability of electrodes for rechargeable batteries due to the outstanding volume expansion buffering efficiency, which motivates ardent pursuing on the synthetic approaches of hollow materials. Herein, an intriguing route, combining solid precursor transition and Ostwald ripening (SPTOR), is developed to craft nano single‐crystal (SC)‐constructed MnCO3 submicron hollow spindles homogeneously encapsulated in a reduced graphene oxide matrix (MnCO3 SMHSs/rGO). It is noteworthy that the H‐bonding interaction between Mn3O4 nanoparticles (NPs) and oxygen‐containing groups on GO promotes uniform anchoring of Mn3O4 NPs on GO, mild reductant ascorbic acid triggers the progressive solid‐to‐solid transition from Mn3O4 NPs to MnCO3 submicron solid spindles (SMSSs) in situ on GO, and the Ostwald ripening process induces the gradual dissolution of interior polycrystals of MnCO3 SMSSs and subsequent recrystallization on surface SCs of MnCO3 SMHSs. Remarkably, MnCO3 SMHSs/rGO delivers a 500th lithium storage capacity of 2023 mAh g−1 at 1000 mA g−1, which is 10 times higher than that of MnCO3 microspheres/rGO fabricated from a conventional Mn2+ salt precursor (202 mAh g−1). The ultrahigh capacity and ultralong lifespan of MnCO3 SMHSs/rGO can be primarily attributed to the superior reaction kinetics and reversibility combined with exceptional interfacial and capacitive lithium storage capability, enabled by the fast ion/electron transfer, large specific surface area, and robust electrode pulverization inhibition efficacy. Moreover, fascinating in‐depth lithium storage reactions of MnCO3 are observed such as the oxidation of Mn2+ in MnCO3 to Mn3+ in charge process after long‐term cycles and the further lithiation of Li2CO3 in discharge process. As such, the SPTOR approach may represent a viable strategy for crafting various hollow functional materials with metastable nanomaterials as precursors.

Abstract Single-metal-atom catalysts (SMACs) have garnered extensive attention for various electrocatalytic applications, owing to their maximum atom-utilization efficiency, tunable electronic structure, and remarkable catalytic performance. In particular, carbon-based SMACs exhibit optimal electrocatalytic activity for the oxygen reduction reaction (ORR) which is of paramount importance for several sustainable energy conversion and generation technologies, such as fuel cells and metal-air batteries. Despite continuous endeavors in developing various advanced carbon-based SMACs for electrocatalytic ORR, the rational regulation of coordination structure and thus the electronic structure of carbon-based SMACs remains challenging. In this review, we critically examine the role of coordination structure, including local coordination structure (i.e., metal atomic centers and the first coordination shell) and extended local coordination structure (i.e., the second and higher coordination shells), on the rational design of carbon-based SMACs for high-efficiency electrocatalytic ORR. Insights into the relevance between coordination structures and their intrinsic ORR activities are emphatically exemplified and discussed. Finally, we also propose the major challenges and future perspectives in the rational design of advanced carbon-based SMACs for electrocatalytic ORR. This review aims to emphasize the significance of coordination structure and deepen the insightful understanding of structure-performance relationships.

SiO-based materials represent a promising class of anodes for lithium-ion batteries (LIBs), with a high theoretical capacity and appropriate and safe Li-insertion potential. However, SiO experiences a large volume change during the electrochemical reaction, low Li diffusivity, and low electron conductivity, resulting in degradation and low rate capability for LIBs. Here, we report on the rapid crafting of SiO–Sn2Fe@C composites via a one-step plasma milling process, leading to an alloy of Sn and Fe and in turn refining SiO and Sn2Fe into nanoparticles that are well dispersed in a nanosized, few-layer graphene matrix. The Sn and Fe nanoparticles generated during the first Li-insertion process form a stable network to improve Li diffusivity and electron conductivity. As an anode material, the SiO–Sn2Fe@C composite manifests high reversible capacities, superior cycling stability, and excellent rate capability. The capacity retention is found to be as high as 95% and 84% at the 100th and 300th cycles under 0.3 ​C. During rate capability testing at 3, 6, and 11 ​C, the capacity retentions are 71%, 60%, and 50%, respectively. This study highlights that this simple, one-step plasma milling strategy can further improve SiO-based anode materials for high-performance LIBs.

The traditional eight arts include film and dance. Dance is often included in the creation of films. With the progress of the times, dance has shown unprecedented vitality in film. This also puts forward higher requirements for shooting technology in film creation. To solve the contradiction between high performance of equipment and energy sensing, a new energy aware scheduling framework is proposed based on Cyber Physical System, which utilizes the balance between performance and energy consumption optimization, and uses allocation technology and list scheduling to ensure energy constraints. Besides, a highly energy-efficient and stable scheduling algorithm for film creation is constructed. The algorithm problems are mainly divided into functional safety requirements, verification problems, and energy consumption optimization problems under functional safety. The experimental results show that the system can obtain better schedulability at a lower time complexity and reasonably reflect the dynamic and static energy constraints ratio. The basic framework system based on dynamic step size also achieves better time performance than other step sizes. According to the experiment's findings, the energy consumption of all scheduling components is relatively low and can be maintained within a specific range. The research and analysis of this study can provide a theoretical reference for the equipment algorithm of film dance creation practice, promote interaction with technical practice, and assist in promoting the development process of film dance creation.

Abstract Background As a main drug for diseased thrombus, some clinically used thrombolytic agents have various disadvantages, safer novel thrombolytic agents are of great demand. This study aimed to achieve high and efficient production of a fibrinolytic enzyme with superior enzymatic properties, by a combination strategy of codon optimization, gene dosage and process optimization in Komagataella phaffii (K. phaffii). Results After codon optimization, the fibase from a marine Bacillus subtilis was expressed and secreted in K. phaffii GS115. Recombinant strains harboring different copies of the fib gene (fib-nc) were successfully obtained via Geneticin (0.25–4 mg/ml) screening on minimal dextrose selection plates and assessment via real-time quantitative PCR. The respective levels of fibase produced by strains expressing fib-5.4c, fib-6c, fib-8c, fib-9c, and fib-12c were 4428, 5781, 7323, 7930, and 2472 U/ml. Levels increased as the copy number increased from 4 to 9, but decreased dramatically at copy number 12. After high cell density fermentation optimization, the highest fibase activity of the strain expressing fib-9c was 7930 U/ml in a shake flask and increased to 12,690 U/ml after 3 days of continuous culture in a 5-L fermenter, which is one of the highest levels of production reported. The recombinant fibase was maximally active at pH 9.0 and 45 °C, and was remarkably stable at pH levels ranging from 5 to 10 and temperatures up to 50 °C. As a metal-dependent serine protease, fibase did not cause hemolysis in vitro and preferentially degraded fibrin directly. Conclusions The combination of codon optimization, gene dosage, and process optimization described herein could be used for the expression of other therapeutic proteins difficult to express. The characteristics of the recombinant fibase suggest that it has potential applications for thrombosis prevention and therapy.

ObjectivesTo test the validity and reliability of the Secondary Traumatic Stress Scale—Chinese version in clinical nurses.MethodsAccording to the translation principles of the Brislin Scale, the original scale was translated, back translated and cross-culturally adapted to form the Chinese version of the Secondary Traumatic Stress Scale. Nurses in three general hospitals in Changsha, Hunan province were surveyed by convenient sampling method from July 2020 to September 2021. Exploratory factor analysis, confirmatory factor analysis, content validity and criterion validity was used to evaluate the validity of the scale. Internal consistency Cronbach's α coefficient, split-half reliability and test-retest reliability were used to evaluate the reliability of the scale.ResultsA total of 678 nurses were included in the study. There were 460 people in sample 1 and 218 people in sample 2. Two common factors were extracted by exploratory factor analysis. The cumulative contribution was 65.560%. The two-factor structure model was good (χ2/df = 3.137, CFI = 0.928, IFI = 0.929, GFI = 0.842, TLI = 0.917, RMSEA = 0.099). The I-CVI of the scale was 0.8–1.0. The S-CVI/Ave was 0.94. The Cronbach's alpha coefficient is 0.956. The broken half reliability is 0.920. The retest reliability is 0.910.ConclusionThis study identified two components of the Secondary Traumatic Stress Scale—Chinese version, which has 2 dimensions and 17 items. With good validity and reliability, it is suitable for the assessment of secondary traumatic stress among clinical nurses in the Chinese context.

Abstract Lithium–sulfur batteries (LSBs) hold great promise as one of the next‐generation power supplies for portable electronics and electric vehicles due to their ultrahigh energy density, cost effectiveness, and environmental benignity. However, their practical application has been impeded owing to the electronic insulation of sulfur and its intermediates, serious shuttle effect, large volume variation, and uncontrollable formation of lithium dendrites. Over the past decades, many pioneering strategies have been developed to address these issues via improving electrodes, electrolytes, separators and binders. Remarkably, polymers can be readily applied to all these aspects due to their structural designability, functional versatility, superior chemical stability and processability. Moreover, their lightweight and rich resource characteristics enable the production of LSBs with high‐volume energy density at low cost. Surprisingly, there have been few reviews on development of polymers in LSBs. Herein, breakthroughs and future perspectives of emerging polymers in LSBs are scrutinized. Significant attention is centered on recent implementation of polymers in each component of LSBs with an emphasis on intrinsic mechanisms underlying their specific functions. The review offers a comprehensive overview of state‐of‐the‐art polymers for LSBs, provides in‐depth insights into addressing key challenges, and affords important resources for researchers working on electrochemical energy systems.

The ability to create one-dimensional (1D) nanomaterials with precisely controlled dimensions and compositions enable investigation into the relationship between their 1D structure and intriguing properties. However, it remains challenging to develop a general approach for producing robust 1D nanomaterials of different aspect ratios. Here, we report the crafting of hairy 1D carbon nanotube (CNT)/polymer/Au nanocomposites with tunable length by capitalizing on rationally-designed amphiphilic, crosslinkable triblock copolymers as nanoreactors. Specifically, poly(ethylene glycol)-block-poly(acrylic acid)-block-poly(4-vinylbenzylazide-co-styrene) triblock copolymer (denoted PEG-b-PAA-b-(PVBA-co-PS)) with a well-controlled molecular weight of each block is synthesized and exploited to ligate on CNT via strong π-π stacking and hydrophobic interactions. Subsequent crosslinking of PVBA blocks via UV irradiation, followed by the introduction of Au precursors, leads to the formation of PEG-capped 1D CNT/crosslinked PVBA-co-PS/Au (i.e., core/shell 1/shell 2) nanocomposites. In stark contrast to conventional methods for yielding CNT-based 1D nanomaterials, our amphiphilic crosslinkable block copolymer nanoreactor strategy circumvents the alteration and/or destruction of intrinsic properties of CNT. The thickness of the outer Au shell anchored on CNT is dictated by the length of PAA blocks. Furthermore, the plasmonic property of 1D CNT/crosslinked PVBA-co-PS/Au nanocomposites can be tailored by selective separation via centrifugation. The resulting hydrophilic 1D CNT/crosslinked PVBA-co-PS/Au nanocomposite demonstrates improved catalytic activity over the Au nanoparticle counterpart. This strategy can be readily extended to construct other appealing low-dimensional hybrid nanomaterials of interest for diverse applications including sensors, catalysis, energy conversion and storage, etc.

Monocrystalline metallic nanoparticles are preferable for plasmonic applications, but grow only in polyhedral shapes, which deviate from the predominantly employed spherical shape. Through numerical simulations we have investigated possibilities for using octahedral instead of spherical Au nanoparticles in plasmonics. Our results show that the dimers of octahedral Au nanoparticles can provide higher field enhancement compared to spherical nanoparticles and that placing the octahedrons close to a metal plate can also have certain advantages compared to spheres.

Fibrinolytic enzymes are the most effective agents for the treatment of thrombotic diseases. In the present study, we purified and characterized an extracellular fibrinolytic serine metalloprotease (named Velefibrinase) that is produced by marine Bacillus velezensis Z01 and assessed its thrombolysis in vivo. SDS-PAGE and MALDI-TOF-MS analyses showed that the molecular mass of Velefibrinase was 32.3 KDa and belonged to the peptidase S8 family. The optimal fibrinolytic activity conditions of Velefibrinase were 40 °C and pH 7.0. Moreover, Velefibrinase exhibited high substrate specificity to fibrin, and a higher ratio of fibrinolytic/caseinolytic (1.48) values, which indicated that Velefibrinase had excellent fibrinolytic properties. Based on the degradation pattern of fibrin and fibrinogen, Velefibrinase could be classified as α/β-fibrinogenase. In vitro, Velefibrinase demonstrated efficient thrombolytic ability, anti-platelet aggregation, and amelioration of blood coagulation (APTT, PT, TT, and FIB), which were superior to those of commercial anticoagulant urokinase. Velefibrinase showed no hemolysis for erythrocyte in vitro and no hemorrhagic activity in vivo. Finally, Velefibrinase effectively prevented mouse tail thrombosis in a dose-dependent (0.22–0.88 mg/kg) manner. These findings suggested that Velefibrinase has the potential to becoming a new thrombolytic agent.

Abstract Poly-γ-glutamic acid (γ-PGA) is a promising microbial polymer with wide applications in industry, agriculture and medicine. In this study, a novel glutamate-independent γ-PGA producing strain with thermotolerant characteristics was isolated and identified as Bacillus subtilis GXG-5, then its product was also characterized. The fermentation process was optimized by single-factor tests, and results showed that high temperature (50 °C) was especially suitable for the γ-PGA production by GXG-5. The γ-PGA yield reached 19.50 ± 0.75 g/L with substrate conversion efficiency of 78% at 50 °C in 10 L fermentor. Comparison of GXG-5 and GXA-28 (glutamate-dependent strain) under respective optimal fermentation conditions, the γ-PGA yield of GXG-5 was 19.0% higher than that of GXA-28, and GXG-5 was also superior to GXA-28 in the availability of carbon sources and substrates. Furthermore, the glutamate dependent difference between GXA-28 and GXG-5 was analyzed by genomic sequencing, results indicated that genes related to the glutamate dependent difference mainly involved in carbohydrate transport and metabolism and amino acid metabolism, and 13 genes related to γ-PGA synthesis were mutated in GXG-5. This study provided a potential glutamate-independent strain to replace glutamate-dependent strain for γ-PGA production, and shared novel information for understanding the glutamate dependent difference at the genomic level.

The morphology control of metal halide perovskite crystalline films is of importance to enable high-performance solar cells. Here, Heet al. use a meniscus-assisted solution-based method to print microsized perovskite grains at 60 °C, which results in high optoelectronic device efficiency of 20%.

This study characterized the biosynthetic pathway of the secondary metabolite 1-deoxynojirimycin (DNJ) from Streptomyces lavendulae. The results revealed that glucose was a preferable precursor for DNJ synthesis, and its carbon skeleton underwent a C2-N-C6 cyclization reaction during synthesis. The biosynthetic pathway was related to the glycolysis pathway, and started from fructose-6-phosphate, and involved amination, dephosphorylation, oxidation, cyclization, dehydration, and reduction reaction steps, yielding DNJ. Then, based on clarified biosynthetic pathway information, precursors, analogs, and metabolism inhibitors were used as novel regulators to enhance the production of DNJ. The results demonstrated that the titer of DNJ could reach 296.56 mg/L, which was 3.3-fold higher than that of a control group (90 mg/L) when sodium citrate (0 h, 5 g/L), sorbose (0 h, 1 g/L), iodoacetic acid (20 h, 50 mg/L), and glucose (26 h, 7 g/L) were added during the fermentation process. This study provides a new understanding of the biosynthetic pathway of DNJ, and also provides an efficient strategy to regulate the production of DNJ based on this biosynthetic pathway, which is a new perspective for the regulation of other secondary metabolites.

Microbial fibrinolytic enzyme is a promising candidate for thrombolytic therapy. Non-sterile production of fibrinolytic enzyme by marine Bacillus subtilis D21-8 under submerged fermentation was realized at a mild temperature of 34°C, using a unique combination of starvation strategy and self-production of antibacterial agents. A medium composed of 18.5 g/L glucose, 6.3 g/L yeast extract, 7.9 g/L tryptone, and 5 g/L NaCl was achieved by conventional and statistical methods. Results showed efficient synthesis of fibrinolytic enzyme and antibacterial compounds required the presence of both yeast extract and tryptone in the medium. At shake-flask level, the non-sterile optimized medium resulted in higher productivity of fibrinolytic enzyme than the sterile one, with an enhanced yield of 3,129 U/mL and a production cost reduced by 24%. This is the first report dealing with non-sterile submerged fermentation of fibrinolytic enzyme, which may facilitate the development of feasible techniques for non-sterile production of raw materials for the preparation of potential drugs with low operation cost.

The wireless sensor network, which has a low cost, low energy consumption and wide coverage, is currently involved in many interdisciplines, and becomes a hot research field with rich knowledge contents. It has more broad application prospects in the odor source localization research, and is of great significance to promote people’s emergency response capacity. This paper studies the application of wireless sensor network in odor source location by plume model and locating algorithm, and carries out a simulation experiment. The experimental results show that the number of sensor nodes and the scope of environments influence the estimation of odor source location. Among them, as the number of sensor nodes increases, positioning errors decrease. However, when the number of nodes exceeds 20, the locating errors tend to be moderate. With the expansion of environmental ranges, the number of sensor nodes required to obtain stable estimation results is increasing, and errors occurred before the stability also tend to increase. When the number of nodes is limited, the sensor node can be combined with the autonomous mobile robot to carry out simple movement, and an effective estimation of odor source location can also be carried out.
