Crop resistance to herbicides is crucial for agricultural productivity and sustainability amidst escalating challenges of weed resistance. Uncovering herbicide resistant genes is particularly important for rice production. In this study, we tested the resistance to three commonly used herbicides: glufosinate, glyphosate and mesotrione of 421 diverse rice cultivars and employed genome-wide association studies (GWAS) to unravel the genetic underpinnings of resistance to these three herbicides in rice. We discovered that cultivated rice exhibited rich variation in resistance to the three herbicides, and the differences among subpopulations were significant. Six identified associations harboring candidate genes for resistance to these herbicides were significant. Among them, RGlu6 and RGly8 were the major QTL for resistance to glufosinate and glyphosate, respectively. The favorable alleles of RGlu6 and RGly8 were primarily present in japonica cultivars that originated from Europe, highlighting the geographic and genetic diversity of herbicide resistance and emphasizing the localized selection pressures in European rice varieties. Moreover, our findings might suggest that traditional target genes may not contain tolerant alleles in nature, and alternative mechanisms with novel loci associated with resistance may work. By mapping the genes for herbicide resistance, our results may help develop new strategies to combat the dual challenges on effective weed management and herbicide sustainability.

Diquat, glufosinate and the combination of the latter with carfentrazone or saflufenacil are believed to be as effective for pre-harvest desiccation of soybean plants as paraquat. This work was developed with the objective of evaluating the efficacy of paraquat, diquat, and glufosinate alone or at mixtures of carfentrazone or saflufenacil in pre-harvest desiccation of soybean. Soybean defoliation, maturity and yield were evaluated. Similar efficacy was observed between glufosinate and paraquat in soybean defoliation and maturity, in addition to a greater flexibility of use, in which it could be an anticipator and standardizing agent of the harvest. The application of diquat (200 g of active ingredient [a.i.] ha-1), paraquat (200 g a.i. ha-1) or glufosinate (400 or 500 g a.i. ha-1) was effective for pre-harvest desiccation of soybean at the R7.2 stage. As well, it was observed promising results for application of glufosinate + saflufenacil (300 + 24.5 g a.i. ha-1), glufosinate + carfentrazone (200 + 25 g a.i. ha-1), glufosinate + saflufenacil (200 + 35 g a.i. ha-1). Diquat and glufosinate can replace paraquat in pre-harvest desiccation of soybean plants, and glufosinate also provide greater management flexibility, in anticipating the harvest.

Abstract The objective of this work was to evaluate the effectiveness of herbicides when applied alone and in combinations at sourgrass (Digitaria insularis) post-emergence, as well as to identify a substitute to paraquat in sequential application. Field and greenhouse experiments were conducted during the 2019/2020 and 2020/2021 crop seasons. The herbicides applied alone and in combinations were: atrazine, clethodim, clodinafop, diquat, glufosinate, haloxyfop, imazapic, imazapyr, mesotrione, nicosulfuron, paraquat, glyphosate, saflufenacil, tembotrione, and tepraloxydim. In the 2019/2020 crop season, in the experiment conducted in the field, the control of sourgrass was considered low due to the dry weather condition and to the full flowering of the plants. In the greenhouse, a satisfactory control above 80% was observed at 28 days after herbicide application for most treatments. In the 2020/2021 crop season, under field conditions, the application of glyphosate combined with haloxyfop, with a sequential application of glufosinate, resulted in the highest weed control. In the greenhouse, most treatments were effective and, of these, all contained glufosinate. The sequential application of glufosinate or in combinations favors a better control of sourgrass. However, diquat and glufosinate do not differ in efficacy in sequential application and are an option for the control of the weed.

Abstract A putative glufosinate‐resistant Amaranthus palmeri population was reported in 2015 in Anson County, North Carolina. The results from dose–response assays conducted in the field suggested plants were surviving lethal rates of glufosinate. Dose–response assays conducted in the glasshouse determined the Anson County accession exhibited reduced susceptibility to glufosinate compared to three glufosinate‐susceptible populations. The LD50 values (210–316 g ai ha−1) for the Anson County population were always higher than the LD50 values (118–158 g ai ha−1) for the tested susceptible populations from the dose–response assays. Anson County plants that survived lethal glufosinate rates were reciprocally crossed with susceptible plants to create F1 genotypes and treated with a lethal rate of glufosinate (267 g ai ha−1; ascertained from glasshouse dose–response assay) to determine the distribution of injury and survival for each cross compared to a cross of susceptible parents. The distribution of injury was non‐normal for the crosses containing an Anson County plant compared to the cross with a susceptible parent. Survival was 68%–84% for crosses containing an Anson County plant, whereas the survival was significantly reduced to 35% for the susceptible plant cross. Chi‐square goodness of fit tests were used to test inheritance models to describe the responses of the genotypes. The resistant × susceptible crosses were best described with a heterozygous two loci with incomplete dominance model compared to the resistant × resistant cross that was best described with a heterozygous single locus with incomplete dominance model. The Anson County population has evolved resistance to glufosinate that is heritable and likely conferred by an oligogenic mechanism with incomplete dominance.

Glyphosate and glufosinate are among the most widely used pesticides in agriculture worldwide. Their extensive use leads to the presence of their residues on crops and in the surrounding environment. Beehives, bees, and apiculture products can represent potential sources for the accumulation of these substances and their metabolites, and the consequences for bee health, as well as the level of risk to human health from consuming contaminated food, are still unclear. Furthermore, information on the contamination levels of honey and other beehive products by these compounds remains poorly documented. This study is part of a broader research effort aimed at developing specific analytical methods for monitoring the level of these contaminants in bee products. The methodology employed enabled the acquisition of preliminary information concerning the levels of glyphosate and glufosinate contamination in honey samples obtained from various retailers in Italy to assess compliance with the limits established by Regulation 293/2013. The liquid chromatography tandem mass spectrometry analysis of the 30 honey samples revealed quantifiable levels of glyphosate in eight samples, with contamination ranging from 5.4 to 138.5 ng/g. Notably, one sample of the wildflower type showed residue levels nearly three times the maximum residue limit. Additionally, trace levels of glyphosate contamination were detected in another ten samples. It is noteworthy that glufosinate and its metabolites were not detected in any of the analyzed samples within the established method’s detection ranges.

ObjectiveTo analyze the clinical features， treatment and prognosis of patients with respiratory depression caused by glufosinate poisoning.MethodsThe clinical data of patients with respiratory depression caused by glufosinate poisoning admitted to the ICU of Xiangshan first people’s hospital medical and health group from March 2018 to January 2022 were retrospectively analyzed.ResultsA total of 21 patients with respiratory depression caused by glufosinate poisoning were included. The median （interquartile） intake of glufosinate was 30 （20， 40） g， and the median （interquartile） visit time was within 2.0 （1.0， 2.8） h. The initial symptoms were nausea and vomiting in 16 cases （76.2%）， and sore throat in 8 cases （38.1%）. Respiratory depression， convulsions and shock occurred 6‒48 hours after ingestion of glufosinate. Convulsion occurred in 13 cases （61.9%）， shock in 10 cases （47.6%） and bradycardia in 5 cases （23.8 %）. Among the patients with convulsion or shock， respiratory depression occurred earlier than convulsion and shock in 10 cases （76.9%） and 9 cases （90.0%）， respectively. All patients were treated with gastric lavage， catharsis， mechanical ventilation and symptomatic support. Blood purification was performed in 14 cases. The duration of mechanical ventilation was 5.0 （4.0， 7.0） d，and no patient died. The patients were divided into blood purification group and routine treatment group. There was no significant difference in complications and duration of mechanical ventilation between the blood purification group and the routine treatment group （P>0.05）.ConclusionRespiratory depression caused by glufosinate poisoning usually occurs earlier than convulsion and shock. The overall prognosis of patients with respiratory depression caused by glufosinate poisoning is good， which mainly depends on the early recognition and intervention of respiratory depression.

Despite potential health implications, data on the presence of Glyphosate (GLY) and other non-GLY herbicides in human matrices remain scarce. This study aimed to develop a simple and cost-effective methodology for detecting and quantifying GLY, its primary biodegradation product; aminomethylphosphonic acid (AMPA); and glufosinate (GLU) in plasma and urine of environmentally and occupationally exposed populations from the province of Córdoba (Argentina). Different alternatives of pre-treatment, derivatization with FMOC-Cl, solid phase extraction, and final sample conditioning steps were evaluated to improve the quantification of the herbicides by a high-performance liquid chromatography system coupled to a triple-quadrupole mass spectrometer. Recoveries ranged from 39 to 84% in both matrices, while limits of quantification were 3, 1, and 0.3 ng/mL and 3.6, 5.1, and 0.3 ng/mL for AMPA, GLY, and GLU in plasma and urine, respectively. In plasma samples, GLY was the most frequently detected analyte (32%), followed by GLU (10%). In urine samples, GLU was the most frequently detected herbicide (13%), followed by GLY (6%). No differences between group or matrix correlations were found. This study is the first report of GLU in human biological matrices and should be used to establish baseline values for future surveillance systems.

Nozzle type and herbicide application timing can affect herbicide efficacy. Prickly sida (Sida spinosa) and barnyardgrass (Echinochloa crus-galli) are problematic weeds in eastern Mississippi cotton production and have reduced yield in recent years. Field studies were conducted at two locations – Brooksville, MS (2018, 2019) and Starkville, MS (2019) to understand the nozzle type and herbicide application timing effects on prickly sida and barnyardgrass control in cotton. Studies also compared applications made by an eight-nozzle tractor-mounted sprayer with a four-nozzle backpack sprayer. Herbicide applications were made at four timings: preemergence (PRE), early-postemergence (EPOST), mid-postemergence (MPOST), and late-postemergence (LPOST) corresponding to the preemergence (immediately after planting), two-to-three leaf, four-to-six leaf, and early-bloom stages, respectively. Treatments were made at 140 l · ha−1 applied at each growth stage, with nozzle type and sprayer as variables by each timing. Results showed no differences in treatments applied with backpack and tractor-mounted sprayers. Control of barnyardgrass was significantly affected by nozzle type, but control of prickly sida was not significantly influenced by nozzle type. In all three site-years, plots receiving a MPOST only herbicide application resulted in less weed control than areas receiving a two-pass POST herbicide program. Cotton yield was significantly affected by the herbicide program at one site-year, but was not significantly affected by the herbicide program except where cotton injury exceeded 15%. A two- or three-pass herbicide program was most effective in controlling prickly sida and barnyardgrass in Mississippi cotton.

Objective Among Iran's climate-well-adapted oily plants, safflower (Carthamus tinctorius L.) is considered as a top-ranked plant since it is well-adapted to areas with water shortage. Safflower is known as a model crop with a variety of fatty acids with a relatively high tolerance to salinity and drought, and also due to its high nutritional value that is related to the composition of 90 percent unsaturated fatty acids in its oil content. It has always been considered as a desirable and valuable oilseed plant and can play an important role in expanding the area under planting of oilseeds in the country. Although, Iran with its diverse climate, is one of the richest regions in the world in terms of safflower genetic resources, safflower planting is not very common in our country. One of the main reasons is probably the lack of promotion and low grain yield of its cultivars. In this study, For the first time the floral dipping was performed in Agrobacterium suspension in order that the glufosinate gene would be transferred to the safflower plant. Actually, the access to appropriate engineered cultivars will play a critical role in the development of safflower planting. Materials and methods Nine safflower lines were selected from a world-wide collection genotypes based on the different agronomic and phenotypic traits. They were submerged in Agrobacterium suspensions with optical density (OD600 nm) of 1 and in the presence of two different amounts of silwet-77. Results The genomic analysis of T1 generations using glufosinate resistance gene primers indicated the success of glufosinate transfer in safflower plant through flower immersion. Conclusions Among 9 studied genotypes and 17 transformed plants using the floral dipping method, 3 transgenic plants were obtained with sequencing confirmation: one seedling belongs to Mahali Yazd genotype, one seedling belongs to the Mahali Kerman and one seedling belongs to the Paeize 12 genotype. All transgenic plants were inoculated with 40μl concentration of silwet-77.

A method was developed to determine glyphosate, aminomethylphosphonic acid (AMPA), and glufosinate in soil. The worldwide use of this herbicide in agricultural activities, and its known negative effects on both the environment and health, have generated interest in the establishment of methodologies for its determination in several matrices at trace level. The development of analytical methods for the determination of glyphosate, AMPA, and glufosinate is challenging due to its present amphoteric properties, high solubility in water, low molecular weight, high affinity to the ions presents in the soil, and lack of chromophore groups in its structure, making its quantification difficult. The proposed method exhibits a linear range from 5.0 to 600 µg/kg with limits of detection of 1.37, 0.69 and 1.22 μg/kg, limits of quantification of 4.11, 2.08, and 3.66 μg/kg for glyphosate, AMPA, and glufosinate, respectively, and adequate repeatability and reproducibility (coefficients of variation

Although genetic transformation in Glycine max has been intensively investigated, the process remains ineffective. In the present work, for the first time, the in vitro selection of transgenic lines of soybean variety Incasoy-27 is addressed. From somatic embryos transformed with Agrobacterium tumefaciens strain EHA105, the regeneration of three lines of supposedly transformed plants that showed resistance to phosphinothricin 6.0 mg l-1 was achieved. GUS histochemical analysis demonstrated stable expression of the uidA gene in leaves of two plant lines. Subsequently, the presence of the bar and uidA genes in two regenerated lines was confirmed with the polymerase chain reaction; while in the third line the band referring to the uidA gene was found but it was not possible to demonstrate the presence of the bar gene. In conclusion, is possible to in vitro select transgenic lines of soybean variety Incasoy-27, with expression of the uidA and bar genes in the genome of the transformed plants and 0.30% efficiency.

Hair fescue (Festuca filiformis) is a tuft-forming perennial grass that reduces yields in lowbush blueberry (Vaccinium angustifolium) fields. Nonbearing year foramsulfuron applications suppress hair fescue, but there is interest in increasing suppression through foramsulfuron use in conjunction with fall-applied herbicides. The objective of this research was to determine the main and interactive effects of fall-bearing year herbicide applications and spring-nonbearing year foramsulfuron applications on hair fescue. The experiment was a 5 × 2 factorial arrangement of fall-bearing year herbicide (none, terbacil, pronamide, glufosinate, dichlobenil) and spring-nonbearing year foramsulfuron application (0, 35 g·ha−1) arranged in a randomized complete block design at lowbush blueberry fields in Portapique and Stewiacke, Nova Scotia, Canada. Spring-nonbearing year foramsulfuron applications did not reduce total tuft density or consistently reduce flowering tuft density, flowering tuft inflorescence number, or flowering tuft seed production. Fall-bearing year pronamide applications reduced hair fescue density for the 2-year production cycle, although additional bearing year density reductions occurred when pronamide was followed by spring-nonbearing year foramsulfuron applications at Stewiacke. Fall-bearing year dichlobenil applications reduced total and flowering tuft density at each site, although reductions in flowering tuft inflorescence number and seed production were most consistent when followed by spring-nonbearing year foramsulfuron applications at Stewiacke. Suppression extended into the bearing year at each site, and dichlobenil should be examined further for hair fescue control. Fall-bearing year glufosinate applications reduced hair fescue total tuft density at each site and flowering tuft density and flowering tuft seed production at Stewiacke. Fall-bearing year glufosinate applications followed by spring-nonbearing year foramsulfuron applications also reduced nonbearing year flowering tuft inflorescence number and bearing year hair fescue seedling density at Stewiacke, indicating that this treatment may reduce hair fescue seedling recruitment at some sites. Fall-bearing year terbacil applications did not suppress hair fescue and are not recommended for hair fescue management in lowbush blueberry.

Abstract Glutamine synthetase (GS) generates glutamine from glutamate and controls the release of inflammatory mediators. In macrophages, GS activity, driven by IL10, associates to the acquisition of M2‐like functions. Conditional deletion of GS in macrophages inhibits metastasis by boosting the formation of anti‐tumor, M1‐like, tumor‐associated macrophages (TAMs). From this basis, we evaluated the pharmacological potential of GS inhibitors in targeting metastasis, identifying glufosinate as a specific human GS inhibitor. Glufosinate was tested in both cultured macrophages and on mice bearing metastatic lung, skin and breast cancer. We found that glufosinate rewires macrophages toward an M1‐like phenotype both at the primary tumor and metastatic site, countering immunosuppression and promoting vessel sprouting. This was also accompanied to a reduction in cancer cell intravasation and extravasation, leading to synchronous and metachronous metastasis growth inhibition, but no effects on primary tumor growth. Glufosinate treatment was well‐tolerated, without liver and brain toxicity, nor hematopoietic defects. These results identify GS as a druggable enzyme to rewire macrophage functions and highlight the potential of targeting metabolic checkpoints in macrophages to treat cancer metastasis.

Hive products have numerous beneficial properties; however, the hive’s health is affected by the surrounding environment. The widespread use of herbicides in agriculture, such as glyphosate and glufosinate, has raised alarm among consumers, beekeepers, and environmentalists due to their potential to harm bees and humans through the consumption of bee products. This review aims to provide a comprehensive overview of the presence of glyphosate, glufosinate, and their metabolites in hive products, collecting and comparing available data from peer-reviewed research and surveys conducted across several countries. Moreover, it analyzes and discusses the potential impacts of these substances on human and bee health, analytical aspects, and recent regulatory developments. The data has revealed that these substances can be present in the different matrices tested, but the concentrations found are usually lower than the maximum residue limits set. However, the use of different methodologies with non-uniform analytical performances, together with an incomplete search for regulated analytes, leads to heterogeneity and makes comparisons challenging. In addition to the completion of studies on the toxicology of herbicide active ingredients, further monitoring actions are necessary, harmonizing analytical methodologies and data management procedures.

Natural astaxanthin is known to be produced by green microalgae, a potent producer of the most powerful antioxidant. To increase the productivity of astaxanthin in microalgae, random mutagenesis has been extensively used to improve the yield of valuable substances. In the presented work, a newly isolated Coelastrum sp. was randomly mutagenized by exposure to ethyl methane sulfonate and further screened using two approaches; an approach for high growth mutant and an approach for high astaxanthin producing mutant with a high-throughput screening method using glufosinate. Among these, mutant G1-C1 that was selected using glufosinate showed the highest of total carotenoids (45.48±1.5 mg/L) and astaxanthin (28.32±2.5 mg/L) production, which was almost 2-fold higher than that of wild type. This study indicates that random mutagenesis via chemical mutation strategy and screening using glufosinate successfully expedited astaxanthin production in a mutated strain of a Coelastrum sp.

Genetically modified (GM) crops tolerant to glyphosate have delivered significant economic benefits in farm management. However, the evolution of glyphosate resistance in weeds due to prolonged intensive use of glyphosate poses a serious threat to this weed management system. It is highly desirable in China to deploy dual herbicide-tolerant corn at the very beginning of GM corn release to delay the development of weed resistance to herbicides. Here, we report the creation and characterization of a herbicide-tolerant corn event SCB-29 that expresses both cp4 epsps and bar genes. This transgenic maize is tolerant to glyphosate up to 3600 g a.e. ha−1 and glufosinate up to 3600 g a.i. ha−1, which are quadruple the recommended rates for the two herbicides, respectively. SCB-29 is an event with only a single copy of T-DNA inserted into chromosome 10 of the maize genome. An event-specific PCR detection method was established and three generations of SCB-29 were detected by event-specific PCR suggesting that the transgenes are stably integrated into the maize genome. Analysis of the expression levels of the transgenes among plants of multiple generations by enzyme-linked immunosorbent assays suggested that the expressions are stable over different generations. Moreover, the major agronomic performances of SCB-29 appear to be similar to those of non-transgenic maize, suggesting that SCB-29 is not likely to have yield drag. Therefore, SCB-29 is an excellent herbicide-tolerant candidate to be developed into a commercial herbicide tolerance transgenic event.

Castor (Ricinus communis L.) is one of the world's top ten oil crops. China is the second largest producer of castor beans, but China's castor bean acreage has been declining. A principal reason for this decline is the prevalence of weeds and the absence of herbicide resistant varieties of castor plants. In this study, we used Agrobacterium-mediated gene transfer to introduce the EPSPS (5-enolpyruvylshikimate-3-phosphate synthase) gene, which confers resistance to glyphosate, and the Bar (phosphinothricin N-acetyltransferase) gene, encoding resistance to glufosinate, into castor 2129 (high stem excellent recovery line) and CSR 181 (dwarf stem excellent recovery line). The transgenic plants exhibited significantly increased expression of the EPSPS and Bar genes and significantly increased resistance to glyphosate and glufosinate. Proteomic analysis showed differences in the amounts of 20 proteins in the dwarf stem transgenic plants. Nine of these proteins are involved in photosynthesis and indirectly related to the mechanism of action of the Bar gene. Eight proteins are involved in defense, repair, stress response, oxidative phosphorylation, and metabolic pathways and are indirectly related to the mechanism of action of the EPSPS gene. These results provide a foundation for the preparation of high herbicide resistant castor varieties.

Wild oat (Avena fatua L.) and feather fingergrass (Chloris virgata Sw.) are among the most problematic weed species in Australian winter and summer cropping systems, respectively. Pot trials were conducted in respective seasons to evaluate the integrated effect of simulated grazing and foliar-applied herbicides on the control of these weed species. Different herbicides were applied 1, 5, and 12 d after grazing on A. fatua plants and 1, 3, 7, 10, and 14 d after grazing on C. virgata. In general, the efficacy of herbicides was better on A. fatua when applied 5 or 12 d after grazing (i.e., 7–20 cm tall plants) and 7 to 14 d (i.e., 10–22 cm tall plants) after grazing on C. virgata. Clethodim 90 g a.i.·ha−1, and haloxyfop 52 and 78 g a.i.·ha−1 resulted in 100% mortality of A. fatua seedlings, irrespective of their application timings. Delayed application (5 or 12 d after grazing) of clethodim 60 g a.i.·ha−1 and pinoxaden 20 and 30 g a.i.·ha−1 resulted in 100% mortality of A. fatua. Glyphosate at the field rate (370 g a.e. ha−1) was the least effective herbicide to control A. fatua plants after grazing. Glufosinate application after grazing resulted in the greatest mortality (69 to 81%) of C. virgata. Depending on application timing, only glufosinate was able to provide complete control of C. virgata seed production. Butroxydim, clethodim, and haloxyfop were found to be promising herbicides to manage C. virgata after grazing but their applications had to be delayed by 7 to 14 d after grazing. This study identified several successful herbicide treatments that could be applied after grazing or mowing for integrated control of A. fatua and C. virgata. However, to achieve complete control of C. virgata, the application of these herbicides needs to be followed by other tools, including additional herbicide applications.

Glutamine synthetase (GlnS) is a key enzyme in nitrogen metabolism. We investigated the effect of the GlnS inhibitor glufosinate on the infection of H. lacustris by the blastocladialean fungus P. sedebokerense, assuming that interfering with the host nitrogen metabolism will affect the success of the parasite. Complete inhibition of infection, which could be bypassed by the GlnS product glutamine, was observed at millimolar concentrations of glufosinate. However, this effect of glufosinate was attributed to its direct interaction with the blastoclad and not the host, which results in development and growth inhibition of the blastoclad. In our P. sedebokerense draft genome, we found that the sequence of GlnS is related to another fungal GlnS, type III, found in many poor known phyla of fungi, including Blastocladiomycota and Chytridiomycota, and absent in the main subkingdom of fungi, the Dikarya. We further tested the ability of the blastoclad to utilize nitrate and ammonia as inorganic nitrogen sources and glutamine for growth. We found that P. sedebokerense equally use ammonia and glutamine and use also nitrate, but with less efficiency. Altogether, our results show that GlnS type III is mandatory for the development and growth of P. sedebokerense and could be an efficient target to develop strategies for the control of the fungal parasite of H. lacustris.

Together, Conyza bonariensis, C. canadensis and C. sumatrensis show 105 reported cases of biotypes resistant to herbicides like glyphosate, paraquat, and acetolactate synthase (ALS) inhibitors. The application of pre-emergent herbicides combined with burndown herbicides is believed to be effective in controlling Conyza spp. during soybean pre-sowing management. The objective of this study was to evaluate the efficacy of sulfentrazone/diuron, imazethapyr/flumioxazin, and diclosulam in mixtures with diquat, paraquat, or glufosinate on the control of Conyza spp. Two field experiments were conducted in a randomized block design with four replicates. Treatments consisted of the application of pre-emergent plus burndown herbicides, besides the weedy control treatment (without application), for a total of 10 treatments. The control of Conyza spp. was evaluated at 7, 14, 21, 28, and 35 d after herbicide application, and symptoms of injury in soybean plants were evaluated at 14, 21, 28, and 35 d after herbicide application. The herbicides sulfentrazone/diuron, imazethapyr/flumioxazin, and diclosulam in combination with burndown herbicides diquat, paraquat, or glufosinate were effective in controlling Conyza spp. in the pre-sowing management of soybean, highlighting good options for pre- and post-emergent herbicide rotations. Mixtures with diclosulam showed a higher potential for injury to soybean plants than sulfentrazone/diuron and imazethapyr/flumioxazin.

The adoption of auxin-tolerant crops has increased awareness regarding herbicide off-target movement. Deposition aids are promoted as a possible solution to off-target movement, although their effect on spray canopy deposition are not well understood. Studies were conducted to determine the impact of deposition aids tank-mixed with herbicides on spray droplet size and canopy deposition. Commonly used herbicides were applied on soybean and cotton in combination with deposition aids (oil, polymer, and guargum). Interactions between herbicide solution and deposition aid influenced droplet size parameters for both cotton and soybean herbicides tested herein (p ≤ 0.0001). Generally, the addition of polymer and guargum deposition aids increased spray droplet size, whereas the addition of oil deposition aid decreased droplet size for some treatments. When herbicides were combined, the inclusion of deposition aids did not influence overall spray deposition on cotton (p = 0.82) and soybean (p = 0.72). When herbicide solutions were evaluated individually, the advent of deposition aids had inconsistent results with cotton and soybean spray deposition being unaffected, increased, or even decreased depending on the herbicide solution tested. For example, the polymer-based deposition aid increased spray deposition on cotton for applications of glyphosate + dicamba + S-metolachlor resulting in 1640.6 RFU (relative fluorescence units). However, the same deposition aid decreased spray deposition on cotton for applications of glyphosate + dicamba + acetochlor (1179.3 RFU). Although deposition aids influenced spray deposition on cotton and soybean for some herbicide combinations, their use should be determined on a case-by-case scenario.

Glutamine synthetase (GS) is a key enzyme involved in the nitrogen metabolism of higher plants. Abiotic stresses have adverse effects on crop production and pose a serious threat to global food security. GS activity and expression is known to be significantly modulated by various abiotic stresses. However, very few transgenic overexpression studies of GS have studied its impact on abiotic stress tolerance. GS is also the target enzyme of the broad spectrum herbicide Glufosinate (active ingredient: phosphinothricin). In this study, we investigated the effect of concurrent overexpression of the rice cytosolic GS1 (OsGS1;1) and chloroplastic GS2 (OsGS2) genes in transgenic rice on its tolerance to abiotic stresses and the herbicide Glufosinate. Our results demonstrate that the co-overexpression of OsGS1;1 and OsGS2 isoforms in transgenic rice plants enhanced its tolerance to osmotic and salinity stress at the seedling stage. The transgenic lines maintained significantly higher fresh weight, chlorophyll content, and relative water content than wild type (wt) and null segregant (ns) controls, under both osmotic and salinity stress. The OsGS1;1/OsGS2 co-overexpressing transgenic plants accumulated higher levels of proline but showed lower electrolyte leakage and had lower malondialdehyde (MDA) content under the stress treatments. The transgenic lines showed considerably enhanced photosynthetic and agronomic performance under drought and salinity stress imposed during the reproductive stage, as compared to wt and ns control plants. The grain filling rates of the transgenic rice plants under reproductive stage drought stress (64.6 ± 4.7%) and salinity stress (58.2 ± 4.5%) were significantly higher than control plants, thereby leading to higher yields under these abiotic stress conditions. Preliminary analysis also revealed that the transgenic lines had improved tolerance to methyl viologen induced photo-oxidative stress. Taken together, our results demonstrate that the concurrent overexpression of OsGS1;1 and OsGS2 isoforms in rice enhanced physiological tolerance and agronomic performance under adverse abiotic stress conditions, apparently acting through multiple mechanistic routes. The transgenic rice plants also showed limited tolerance to the herbicide Glufosinate. The advantages and limitations of glutamine synthetase overexpression in crop plants, along with future strategies to overcome these limitations for utilization in crop improvement have also been discussed briefly.

Since weed management is such a critical component of agronomic crop production systems, herbicides are widely used to provide weed control to ensure that yields are maximized. In the last few years, herbicide-resistant (HR) crops, particularly those that are glyphosate-resistant, and more recently, those with dicamba (3,6-dichloro-2-methoxybenzoic acid) and 2,4-D (2,4-dichlorophenoxyacetic acid) resistance are changing the way many growers manage weeds. However, past reliance on glyphosate and mistakes made in stewardship of the glyphosate-resistant cropping system have directly led to the current weed resistance problems that now occur in many agronomic cropping systems, and new technologies must be well-stewarded. New herbicide-resistant trait technologies in soybean, such as dicamba-, 2,4-D-, and isoxaflutole- ((5-cyclopropyl-4-isoxazolyl)[2-(methylsulfonyl)-4-(trifluoromethyl)phenyl]methanone) resistance, are being combined with glyphosate- and glufosinate-resistance traits to manage herbicide-resistant weed populations. In cropping systems with glyphosate-resistant weed species, these new trait options may provide effective weed management tools, although there may be increased risk of off-target movement and susceptible plant damage with the use of some of these technologies. The use of diverse weed management practices to reduce the selection pressure for herbicide-resistant weed evolution is essential to preserve the utility of new traits. The use of herbicides with differing sites of action (SOAs), ideally in combination as mixtures, but also in rotation as part of a weed management program may slow the evolution of resistance in some cases. Increased selection pressure from the effects of some herbicide mixtures may lead to more cases of metabolic herbicide resistance. The most effective long-term approach for weed resistance management is the use of Integrated Weed Management (IWM) which may build the ecological complexity of the cropping system. Given the challenges in management of herbicide-resistant weeds, IWM will likely play a critical role in enhancing future food security for a growing global population.

According to Article 12 of Regulation (EC) No 396/2005, the European Food Safety Authority (EFSA) has reviewed the Maximum Residue Levels (MRLs) currently established at European level for the pesticide active substance glufosinate. In order to assess the occurrence of glufosinateresidues in plants, processed commodities, rotational crops and livestock, EFSA considered the conclusions derived in the framework of Directive 91/414/EEC, the MRLs established by the Codex Alimentarius Commission as well as European authorisations reported by Member States (incl. the supporting residues data). Based on the assessment of the available data, MRL proposals were derived and a consumer risk assessment was carried out. Some information required by the regulatory framework was found to be missing and a possible acute risk to consumers was identified. Hence, the consumer risk assessment is considered indicative only, some MRL proposals derived by EFSA still require further consideration by risk managers and measures for reduction of the consumer exposure should also be considered.
