Sepsis, a life-threatening condition, involves complex interactions among metabolic alterations, inflammatory mediators, and host responses. This study utilized a bidirectional Mendelian randomization approach to investigate the causal relationships between 1400 metabolites and sepsis, and the mediating role of inflammatory factors. We identified 36 metabolites significantly associated with sepsis (p < 0.05), with AXIN1, FGF-19, FGF-23, IL-4, and OSM showing an inverse association, suggesting a protective role, while IL-2 exhibited a positive correlation, indicating a potential risk factor. Among these metabolites, Piperine and 9-Hydroxystearate demonstrated particularly interesting protective effects against sepsis. Piperine’s protective effect was mediated through its interaction with AXIN1, contributing to a 16.296% reduction in sepsis risk. This suggests a potential pathway where Piperine influences sepsis outcomes by modulating AXIN1 levels. 9-Hydroxystearate also exhibited a protective role against sepsis, mediated through its positive association with FGF-19 and negative association with IL-2, contributing 9.436% and 12.565%, respectively, to its protective effect. Experimental validation confirmed significantly elevated IL-2 levels and reduced FGF-19, AXIN1, piperine, and 9-hydroxyoctadecanoic acid levels in sepsis patients compared to healthy controls. Piperine levels positively correlated with AXIN1, while 9-hydroxyoctadecanoic acid levels negatively correlated with IL-2 and positively correlated with FGF-19, supporting the Mendelian randomization findings. Our findings provide insights into the molecular mechanisms of sepsis, highlighting the unique roles and contributions of specific metabolites and their interactions with inflammatory mediators. This study enhances our understanding of sepsis pathophysiology and opens avenues for targeted therapeutic interventions and biomarker development for sepsis management. However, further research is essential to validate these pathways across diverse populations and fully explore the roles of these metabolites in sepsis.

BackgroundSepsis is a life-threatening organ dysfunction resulting from a dysregulated host response to infection, yet the potential causal relationship between the immunophenotype and sepsis remains unclear.MethodsGenetic variants associated with the immunophenotype served as instrumental variables (IVs) in Mendelian randomization (MR) to elucidate the causal impact of the immunophenotype on three sepsis outcomes. Additionally, a two-step MR analysis was conducted to identify significant potential mediators between the immunophenotype and three sepsis outcomes.ResultsOur MR analysis demonstrated a significant association between the immunophenotype and sepsis outcome, with 36, 36, and 45 the immunophenotype associated with the susceptibility, severity, and mortality of sepsis, respectively. Specifically, our analysis highlighted the CD14+ CD16+ monocyte phenotype as a significant factor across all three sepsis outcomes, with odds ratios (ORs) and corresponding confidence intervals (CIs) indicating its impact on sepsis (OR = 1.047, CI: 1.001-1.096), sepsis in Critical Care Units (OR = 1.139, CI: 1.014-1.279), and sepsis-related 28-day mortality (OR = 1.218, CI: 1.104-1.334). Mediation analyses identified seven cytokines as significant mediators among 91 potential cytokines, including interleukin-5 (IL-5), S100A12, TNF-related apoptosis-inducing ligand (TRAIL), T-cell surface glycoprotein CD6 isoform, cystatin D, interleukin-18 (IL-18), and urokinase-type plasminogen activator (uPA). Furthermore, reverse MR analysis revealed no causal effect of sepsis outcomes on the immunophenotype.ConclusionOur MR study suggests that the immunophenotype is significantly associated with the susceptibility, severity, and mortality of patient with sepsis, providing, for the first time, robust evidence of significant associations between immune traits and their potential risks. This information is invaluable for clinicians and patients in making informed decisions and merits further attention.

In response to the critical implications of single tail rotor failures in unmanned helicopters, this study proposes a distributed electric tail rotor yaw channel controller that employs Active Disturbance Rejection Control (ADRC) techniques. A dynamics model was created for an unmanned helicopter with a distributed electric tail rotor. This model was then employed to verify the yaw channel tracking performance of the helicopter through simulation. The verification process involved the use of a cascaded dual closed-loop active disturbance rejection control strategy. The ADRC framework was enhanced by developing an improved fal function to replace the conventional fal function. This addressed the challenges of excessive error gain and overshoot oscillation in the Extended State Observer (ESO). A test setup for the distributed electric tail rotor of an unmanned helicopter was devised and constructed for empirical testing purposes. This setup was used as a reference for the 700-class electric model helicopter. The findings of the simulation demonstrate that the control strategy for the unmanned helicopter’s distributed electric tail rotor, enhanced by the refined cascaded ADRC method, achieves superior response speed and minimized overshoot during yaw channel tracking. Furthermore, it displays a degree of enhanced robustness and improved control proficiency in comparison to the conventional ADRC approach. The outcomes of the test stand validate the efficacy of this control strategy.

Cognitive impairment is an important component of non motor symptoms in Parkinson's disease (PD), and if not addressed in a timely manner, it can easily progress to dementia. However, no effective method currently exists to completely prevent or reverse cognitive impairment associated with PD. We therefore aimed to investigate the therapeutic effect of near-infrared region II light (NIR-II) region illumination on cognitive impairment in PD through behavioral experiments (water maze and rotary rod) and multiple fluorescence immunohistochemistry techniques. The 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP)-induced group was compared with the MPTP- untreated rat group, showing a significant reduction in escape latency and significant increase in the fall latency in the MPTP-treated group. The horizontal analysis results indicated that NIR-II phototherapy improved the learning and cognitive abilities as well as coordination and balance abilities of rats. Post-treatment, the MPTP rats showed significantly shortened, escape latency, prolonged target quadrant residence time, and prolonged fall latency compared with pre-treatment. The longitudinal analysis results reaffirmed that NIR-II phototherapy improved the learning and cognitive abilities as well as coordination and balance abilities of rats. The multiple fluorescence immunohistochemistry analysis trend plot showed that the activated microglia and astrocytes in the hippocampus were highest in MPTP-induced PD untreated group, moderate in MPTP-induced PD treatment group, and lowest in the control group. Our data indicates that NIR-II illumination improves learning and cognitive impairment as well as coordination and balance abilities in PD rats by downregulating the activation of microglia and astrocytes in the hippocampus.

BackgroundAcute pancreatitis (AP) is a severe digestive system disorder with a significant risk of progressing to sepsis, a major cause of mortality. Unraveling the immunological pathways in AP is essential for developing effective treatments, particularly understanding the role of specific immune cell traits in this progression.MethodsEmploying a bidirectional two-sample Mendelian Randomization (MR) approach, this study first examined the causal relationship between AP and 731 immune cell traits to identify those significantly associated with AP. Subsequently, we explored the causal associations between 731 immune cell traits and sepsis. The analysis utilized extensive genome-wide association studies (GWAS) summary datasets, with a focus on identifying common immune cell traits with statistically significant causal associations between AP and sepsis.ResultsOur investigation identified 44 immune cell traits unidirectionally associated with AP and 36 traits unidirectionally associated with sepsis. Among these, CD127 on CD28+ CD45RA- CD8+ T cells emerged as a common mediator, accounting for 5.296% of the increased risk of sepsis in AP patients. This finding highlights the significant role of specific memory CD8+ T cells in the pathophysiology of AP and its progression to sepsis.ConclusionThis study elucidates the critical role of specific immune cell traits, particularly CD127hi memory CD8+ T cells, in the progression of AP to sepsis. Our findings provide a foundation for future research into targeted immune-modulatory therapies, potentially improving patient outcomes in AP-related sepsis and offering new insights into the complex immunological dynamics of this condition.

Large volumes of Early Mesozoic intermediate to basic igneous rocks related to the evolution of the Paleo-Tethys Ocean are exposed in the East Kunlun Orogenic Belt (EKOB). The petrography, geochemistry, and results of zircon U-Pb dating of Defusheng intermediate to basic rocks from the eastern segment of the EKOB are presented in this report. Zircon U–Pb dating of the intermediate to basic rocks yields ages of 239–245 Ma (Middle Triassic). Defusheng intermediate to basic rocks have low TiO2 contents (0.80–1.47 wt.%) and widely varying MgO (3.14–6.08 wt.%), and are enriched in large ion lithophile elements and light rare earth elements, having a geochemical composition similar to that of island arc basalts. The variation diagrams of major elements indicate that the Defusheng intermediate to basic rocks underwent fractional clinopyroxene and olivine crystallization. Depletion of the high-field-strength elements Nb, Ta, and Ti may have been caused by the mantle wedge having been infiltrated by fluids derived from the subducted slab. The Defusheng intermediate to basic rocks represent magmatic records of the Early Mesozoic oceanic crust subduction in Eastern Kunlun. This indicates that the final closure of the Paleo-Tethyan Ocean and the beginning of collisional orogeny occurred after the Middle Triassic.

To ensure the accuracy of state of charge (SOC) and state of health (SOH) estimation for battery packs while minimizing the amount of pre-experiments required for aging modeling and the scales of computation for online management, a decisive-cell-based estimation method with training-free characteristic parameters and a dynamic-weighted estimation strategy is proposed in this paper. Firstly, to reduce the computational complexity, the state estimation of battery packs is summed up to that of two decisive cells, and a new selection approach for the decisive cells is adopted based on the detection of steep voltage changes. Secondly, two novel ideas are implemented for the state estimation of the selected cells. On the one hand, a set of characteristic parameters that only exhibit local curve shrinkage with aging is chosen, which keeps the corresponding estimation approaches away from training. On the other hand, multiple basic estimation approaches are effectively combined by their respective dynamic weights, which ensures the estimation can maintain a good estimation accuracy under various load profiles. Finally, the experimental results show that the new method can quickly correct the initial setting deviations and have a high estimation accuracy for both the SOC and SOH within 2% for a series battery pack consisting of cells with obvious inconsistency.

Drought and salt stresses, the major environmental abiotic stresses in agriculture worldwide, affect plant growth, crop productivity, and quality. Therefore, developing crops with higher drought and salt tolerance is highly desirable. This study reported the isolation, biological function, and molecular characterization of a novel maspardin gene, OsMas1, from rice. The OsMas1 protein was localized to the cytoplasm. The expression levels of OsMas1 were up-regulated under mannitol, PEG6000, NaCl, and abscisic acid (ABA) treatments in rice. The OsMas1 gene was introduced into the rice cultivar Zhonghua 11 (wild type, WT). OsMas1-overexpression (OsMas1-OE) plants exhibited significantly enhanced salt and drought tolerance; in contrast, OsMas1-interference (OsMas1-RNAi) plants exhibited decreased tolerance to salt and drought stresses, compared with WT. OsMas1-OE plants exhibited enhanced hypersensitivity, while OsMas1-RNAi plants showed less sensitivity to exogenous ABA treatment at both germination and post-germination stages. ABA, proline and K+ contents and superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and photosynthesis activities were significantly increased. In contrast, malonaldehyde (MDA), hydrogen peroxide (H2O2), superoxide anion radical (O2-·), and Na+ contents were significantly decreased in OsMas1-OE plants compared with OsMas1-RNAi and WT plants. Overexpression of OsMas1 up-regulated the genes involved in ABA signaling, proline biosynthesis, reactive oxygen species (ROS)-scavenging system, photosynthesis, and ion transport under salt and drought stresses. Our results indicate that the OsMas1 gene improves salt and drought tolerance in rice, which may serve as a candidate gene for enhancing crop resistance to abiotic stresses.

Background: As the omicron variant of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) surges amid the coronavirus disease 2019 (COVID-19) pandemic, there is limited comorbidities data associated with viral shedding time (VST). We aimed to investigate the effect of comorbidities on VST in asymptomatic and mild patients with omicron. Methods: A multi-center, retrospective, observational study was conducted from March 12, 2022 to May 24, 2022 in Shanghai. The analysis was adjusted for patients’ baseline demographic, using log-rank test and logistic regression model. Results: The study enrolled 198,262 subjects. The median duration of viral shedding time (VST) was 8.29 days. The number of cumulative viral shedding events was significantly lower in the chronic obstructive pulmonary disease (COPD), hyperlipidemia, diabetes, urinary system disease, and cardiocerebrovascular disease than in the no corresponding comorbidities group. Patients with comorbidities had a lower incidence of viral shedding, and the most significant independent risk factor is COPD (aOR 1.78, 95% CI: 1.53–2.08, p

The simultaneous high-capacity and fast removal of radioactive barium ion (Ba2+) from nuclear wastewater is necessary but still faces large challenge. In this study, a novel adsorbent containing sulfonic group, MOF-808-SO3H, was prepared based on the oxidation of –SH group. Characterizations confirm the successful introduction of –SO3H, good porosity, and stability of the material. The adsorbent exhibits excellent capture ability for Ba2+ due to strong hard acid-base and electrostatic interactions, with the adsorption capacity of 152.0 mg g−1 and fast equilibrium adsorption time, superior to other reported materials. Optimal pH was found to be almost neutral and temperature cannot affect the adsorption largely. Besides, the irreversible adsorption was demonstrated and thereby the secondary pollution may be avoided. Therefore, our work provides an efficient adsorbent for capturing Ba2+ irreversibly, and proposes a feasible strategy for constructing adsorbents with simultaneous high adsorption capacity and fast kinetics.

Crop growth stages are integral components of plant phenology and are of significant ecological and agricultural importance. While the use of remote sensing methods for phenology identification in cropland ecosystems has been extensively explored in previous studies, the focus has often been on land surface phenology, primarily related to the start and end of the growing season. In contrast, the monitoring of crop growth within an agronomic framework has been limited, particularly in the context of recently developed solar-induced chlorophyll fluorescence (SIF) data. Additionally, some critical growth stages have not received adequate attention or evaluation. This study aims to assess the utility of SIF data, collected from both ground and satellite measurements, for identifying critical crop growth stages within the realm of remote sensing phenological estimation. A comparative analysis was conducted using enhanced vegetation index (EVI) data at the Shangqiu site in the North China Plain from 2018 to 2022. Both SIF and EVI time-series data, obtained from ground and satellite sources, undergo a comprehensive phenological estimation framework encompassing pre-processing, modeling, and transition characterization. This approach involves reconciling time-series phenological patterns with crop growth stages, revealing the necessity of redefining the mapping relationship between these two fundamental concepts. After preprocessing the time-series data, the framework incorporates the phenological modeling process employing two double logistic models and a spline model for comparison. Additionally, it includes phenological transition characterization using four different methods. Consequently, each input dataset undergoes an assessment, resulting in 12 sets of estimations, which are compared to select the ideal estimation portfolio for identifying the growth stages of maize and winter wheat. Our findings highlight the efficacy of SIF data in accurately identifying the growth stages of maize and winter wheat, achieving remarkable results with an R-square exceeding 0.9 and an RMSE of less than 1 week for key growth stages (KGSs). Notably, SIF data demonstrate superior accuracy, robustness, and sensitivity to phenological events when compared to EVI data. This study establishes an estimation portfolio utilizing SIF data, involving the Gu model, a double logistic model, as the preferred phenological modelling method together with various compositing methods and transition characterization methods, suitable for most KGSs. These findings create opportunities for future research aimed at enhancing and standardizing crop growth stage identification using remote sensing data for a wide range of KGSs.

The lack of innovative standards for biosafety in synthetic biology is an unresolved policy gap that limits many potential applications in synthetic biology. We argue that a massive support for standardization in biosafety is required for synthetic biology to flourish.

Background: Evidence indicates that chronic stress promotes progression of colorectal liver metastases (CLM). Mangiferin is the active chemical constituent of the rhizomes of Anemarrhena asphodeloides Bunge. Mangiferin (MGF) exerts anti-inflammatory, anti-proliferative, anti-angiogenic, anti-fibrotic and antioxidant effects in a variety of cancers. Its mechanism in chronic stress and tumor growth is still poorly understood. Methods: To investigate the effects of MGF on the CLM and tumor-associated depression, activated hepatic stellate cells (a-HSCs), HT-29 CRC cells, were used in chronic unpredictable mild stress (CUMS) of tumor-bearing models. Potential antidepressant activity was determined by FST, TST, SIT and serum cytokine (IL-6, IL-18 and TNF-α) examination. Downstream signaling molecules were detected by Western blot, immunohistochemistry and fluorescence microscopy. Results: CUMS induced depression behavior and depression-related cytokines and promoted tumor growth in CLM. MGF-treated mice significantly improved chronic stress behaviors by reducing depression-related cytokines. In addition, MGF treatment inhibits WAVE2 signaling pathway, leading to TGF-β1 induced HSC inhibition, thereby reducing depressive behavior and tumor growth in CLM. Conclusion: MGF can alleviate CUMS induced tumor growth and the treatment of CLM patients with MGF may be beneficial.

Abstract The third isoform of the Na+–Ca2+ exchanger (NCX3) is crucial for a physiological fine-tuning of the Ca2+ fluxes in excitable tissues. In this view, the NCX3 accounts for the aberrant Ca2+ influx seen during neuronal excitotoxicity, such as in Alzheimer’s disease (AD). However, little is known about NCX3 regulation and functional properties. Withania somnifera (L.) Dunal (W. somnifera), a traditional indigenous plant widely recognized for having numerous medicinal values, was undertaken to determine its potential therapeutic benefit against aggregated Aβ1–42-induced NCX3 dysregulation and the thereof cognition impairment in 5xFAD mice. The undertaken sourced dried roots of authenticated W. somnifera physicochemical compositional tests satisfied standards of pharmacognostic quality, and further phytochemical analysis of the roots methanol extract revealed the roots constitute several antioxidants. Following an intra-gastric gavage administration of synthesized W. somnifera roots methanolic extract from postnatal day 30 (P30) to P75, in vivo cognitional studies and then neurochemical examinations of the NCX3 expression level, Aβ plaque deposition, and antioxidant activities in the AD-associated brain regions of 4-month-old 5xFAD mice suggests that the oxidative stress normalizing effects of W. somnifera constituents, operating on the NCX3, may have a therapeutic role in the improvement of cognition in AD.

The study of the changes in stress and deformation of frozen walls during excavation has always been a hot topic in underground freezing engineering, and the size of the plastic zone is an important basis for evaluating the stability of frozen walls. In response to the shortcomings in the current design of horizontal frozen walls, based on the internal excavation unloading conditions of the frozen wall in artificial ground freezing, an elastoplastic mechanical model for the interaction between a circular horizontal freezing wall and unfrozen soil mass is established under nonuniform loads to obtain the corresponding solutions for stress and displacement in the system. In this study, considering the shear stress of the plastic zone, the method for solving the traditional plastic zone contour equation is modified; consequently, the modified solution of the contour equation of the plastic zone for the frozen wall is obtained. Using this theoretical solution, the influence of the external load p0 and the lateral pressure coefficient λ on the contour line of plastic zone and tensile stress zone are analyzed by combining the project case, the calculation results show that: the λ=0.485 is the critical point where the inner edge of the frozen wall just happens to have tensile stress. When λ<0.485, the tensile stress zone is inevitable in the inner edge of the frozen wall vertical direction, and its range is only related to λ and increases with the decrease of λ. The p0 only affects the magnitude of tensile stress in the region, but does not affect its range. At this time, the frozen wall compression plastic zone contour evolves from crescent shaped to ear shaped with the increase of p0. When 0.485<λ<0.61, there will be no tensile stress zone, the frozen wall compression plastic zone contour also evolves from crescent shaped to ear shaped with the increase of p0. When λ>0.61, there will be also no tensile stress zone, with the increase of p0, the compression plastic zone contour evolves from the crescent shaped in the horizontal direction to the elliptical shaped, and there is no ear-shaped plastic zone in the whole evolution process. Based on our results, we show that our method can be used to provide a theoretical basis for the stability evaluation and parameter (thickness) design calculation of horizontal frozen walls under nonuniform loads.

A series of four-terminal V7(Bz)8-WGNR devices were established with wrinkled graphene nanoribbon (WGNR) and vanadium-benzene nanowire (V7(Bz)8). The spin-polarized V7(Bz)8 as the gate channel was placed crossing the plane, the concave (endo-positioned) and the convex (endo-positioned) surface of WGNR with different curvatures via Van der Waals interaction. The density functional theory (DFT) and nonequilibrium Green’s function (NEGF) methods were adopted to calculate the transport properties of these devices at various bias voltages (VS) and gate voltages (VG), such as the conductance, spin-polarized currents, transmission spectra (TS), local density of states (LDOS), and scattering states. The results indicate that the position of V7(Bz)8 and the bending curvature of WGNR play important roles in tuning the transport properties of these four-terminal devices. A spin-polarized transport property is induced for these four-terminal devices by the spin-polarized nature of V7(Bz)8. Particularly, the down-spin channel disturbs strongly on the source-to-drain conductance of WGNR when V7(Bz)8 is endo-positioned crossing the WGNR. Our findings on the novel property of four-terminal V7(Bz)8-WGNR devices provide useful guidelines for achieving flexible graphene-based electronic nanodevices by attaching other similar multidecker metal-arene nanowires.

Abstract Background Annexin A1 (ANXA1) exerts anti-nociceptive effect through ANXA1 receptor formyl peptide receptor 2 (FPR2/ALX (receptor for lipoxin A4), FPR2) at the dorsal root ganglia (DRG) level. However, the mechanisms remain elucidated. By using radiant heat, hot/cold plate, tail flick, von Frey, and Randall-Selitto tests to detect nociception in intact and chemical (capsaicin, menthol, mustard oil, formalin or CFA) injected AnxA1 conditional knockout (AnxA1 −/−) mice, applying calcium imaging and patch clamp recordings in cultured DRG neurons to measure neuronal excitability, conducting immunofluorescence and western blotting to detect the protein levels of TRPV1, FPR2 and its downstream molecules, and performing double immunofluorescence and co-immunoprecipitation to investigate the interaction between Calmodulin (CaM) and TRPV1; we aim to uncover the molecular and cellular mechanisms of ANXA1’s role in antinociception. In this study, the effect of annealing atmosphere on the electrical properties, optical properties, microstructure and defects of ZnO∶Al films was investigated in detail. The crystallization, surface and fracture morphologies, electrical properties, resistivity, transmittance, Raman vibration and photoluminescence of ZnO∶Al films were characterized by SEM, XRD, Hall effect test instruments, ultraviolet-visible spectrophotometer, Raman spectra and PL spectra. The results show that after annealing in air atmosphere, the carrier concentration of AZO films decreases from 1.63 × 1020 cm−3 to 7.1 × 1018 cm−3, the optical band gap varies from 3.51 eV to 3.37 eV, the transmittance in NIR increased and the band edge emission in PL spectrum shifted from 3.49 eV to 3.34 eV. On the other hand, while the carrier concentration of AZO films annealing in H2 atmosphere was 5.3 × 1020 cm-3, the optical band gap increases to 3.78 eV, the transmittance in NIR decreases remarkably, the emission intensity of the band edge increases by more than 10 times and the band edge emission in PL spectrum shifts to 3.57 eV. Furthermore, the 4 order LO phonons are observed in ZnO:Al films annealed in air.

High-fat diet (HFD)-induced insulin resistance (IR) in skeletal muscle is often accompanied by mitochondrial dysfunction and oxidative stress. Boosting nicotinamide adenine dinucleotide (NAD) using nicotinamide riboside (NR) can effectively decrease oxidative stress and increase mitochondrial function. However, whether NR can ameliorate IR in skeletal muscle is still inconclusive. We fed male C57BL/6J mice with an HFD (60% fat) ± 400 mg/kg·bw NR for 24 weeks. C2C12 myotube cells were treated with 0.25 mM palmitic acid (PA) ± 0.5 mM NR for 24 h. Indicators for IR and mitochondrial dysfunction were analyzed. NR treatment alleviated IR in HFD-fed mice with regard to improved glucose tolerance and a remarkable decrease in the levels of fasting blood glucose, fasting insulin and HOMA-IR index. NR-treated HFD-fed mice also showed improved metabolic status regarding a significant reduction in body weight and lipid contents in serum and the liver. NR activated AMPK in the skeletal muscle of HFD-fed mice and PA-treated C2C12 myotube cells and upregulated the expression of mitochondria-related transcriptional factors and coactivators, thereby improving mitochondrial function and alleviating oxidative stress. Upon inhibiting AMPK using Compound C, NR lost its ability in enhancing mitochondrial function and protection against IR induced by PA. In summary, improving mitochondrial function through the activation of AMPK pathway in skeletal muscle may play an important role in the amelioration of IR using NR.

Late Paleozoic–early Mesozoic intrusive rocks are distributed widely along the northern margin of the Qaidam Basin in the northern Tibetan Plateau. To constrain the tectonic evolution, we carried out petrological, chronological, and geochemical studies of the Chahannuo gabbros. LA-ICP-MS Zircon U–Pb dating yields an age of 255.0 ± 0.9 Ma for the gabbros, which confirms the existence of Indosinian tectono-magmatic activity on the northern margin of the Qaidam Basin. The Chahannuo gabbros have low whole-rock SiO2, Fe2O3 contents, and high Al2O3 contents, which suggests a calc-alkaline affinity. In addition, the gabbros have high MgO, Cr, and Ni contents and Mg#, similar to those predicted of the regional basaltic melts, and indicating that they were affected mainly by fluid from the subducted slab. The Chahannuo gabbros are characterized by arc-like trace element patterns, with enrichment in LREE and LILE, and depletion in HREE and HFSE. No obvious negative Eu anomalies also indicate that no significant magmatic differentiation has occurred. The low Nb/La ratio and Ti content in gabbros samples suggests that the Chahannuo gabbros were partially contaminated by the crust during their formation. The Chahannuo gabbros have high incompatible element ratios (Rb/Sr, Th/Nd, and Th/La), which are closer to the category of enriched mantle. Combing our data with previous data from contemporaneous magmatism in the region, we suggest that the Chahannuo gabbros formed in a continental arc environment related to the northward subduction of the Paleo-Tethyan oceanic plate.

Yamusi granodiorite in the eastern part of the Qilian Orogen consists mainly of gneissic granodiorite. Researchers have studied other nearby rock masses, and many studies, such as those focusing on the zircon U-Pb age chronology and geochemistry of Yamusi granodiorite, still need to be completed. We obtained a new LA–ICP–MS zircon U-Pb age of 480.3 ± 1.3 Ma for Yamusi granodiorite, which suggested that it was formed during the early Ordovician period. The whole-rock geochemical data show that this granodiorite is relatively rich in Na and poor in K (K2O/Na2O = 0.40–0.73). The granodiorite is metaluminous–weakly peraluminous and can be classified as medium-K calc-alkaline granite. It yields high Sr/Y ratios (35.17–53.78) and low Yb (N ratios of 1.77–9.03 (mean = 3.88). The petrogenesis research suggests that the granodiorite originated mainly from the partial melting of the mafic lower crust, with a minor mantle-derived component. Based on the regional geological setting, we suggest that the Yamusi granodiorite was formed during the northward subduction of the Proto-Tethyan oceanic crust to form an intracontinental arc.

The first impoundment of a super-high dam is a crucial period from dam construction to operation, in which the prediction of the dam deformation is vital for the continued safety of the dam. Therefore, a multi-head attention model based on Pearson K-means clustering is proposed, which is shortened to PKMA. The inputs of the PKMA include measurements of the displacements of plumb lines, water levels, air temperatures, dam body temperatures, water temperatures, and foundation temperatures. Among these inputs, variables related to displacements are regarded as the dominant explanatory factors. Hence, the K-means clustering based on the Pearson index is utilised to increase the weights of displacements in the PKMA. To involve the interactions between inputs, the MA mechanism of neural networks is used to simulate the relationship between inputs and deformation targets. The PKMA model had a maximum MSE of 1.2518 and a maximum MAE of 0.9017 for the model performance metrics at the study measurement points. Compared to the comparison models MA, HST, and LSTM, the performance metrics of the PKMA model are an improvement of an average of 87.02%, 72.42%, and 69.24%.

The transition from the Proto- to the Paleo-Tethys is still a controversial issue. This study reports a new petrology, zircon U–Pb geochronology, and whole-rock geochemistry of volcanic rocks from the Maoniushan Formation in the Nankeke area, northern Qaidam (NQ) of the Tibetan Plateau, to provide new evidence for the transition from the Proto- to the Paleo-Tethys oceans. The volcanic suite consists mainly of rhyolitic crystal lithic tuff lavas and minor basalts. Zircon U–Pb data indicate that the bimodal volcanic rocks were formed during the Early Devonian (ca. 410–409 Ma). Geochemically, the basalts have low contents of SiO2 (48.92 wt.%–51.19 wt.%) and relatively high contents of MgO (8.94 wt.%–9.99 wt.%), TiO2 (1.05 wt.%–1.29 wt.%), K2O (2.35 wt.%–4.17 wt.%), and K2O/Na2O ratios (1.04–2.56), showing the characteristics of calc-alkaline basalts. Their rare earth element (REE) patterns and trace element spider diagrams are characterized by enrichments in LREEs (LREE/HREE = 18.31–21.34) and large ion lithophile elements (LILEs; Rb, Th, and K) and depletion in high-field-strength elements (HFSEs; Nb, Ta, P, and Ti), with slight negative Eu anomalies (Eu/Eu* = 0.82–0.86), which are similar to Etendeka continental flood basalts (CFB). These features suggest that the basalts were most likely derived from low degree (1%–5%) partial melting of the asthenospheric mantle, contaminated by small volumes of continental crust. In contrast, the felsic volcanics have high SiO2 (68.41 wt.%–77.12 wt.%), variable Al2O3 (9.56 wt.%–12.62 wt.%), low MgO, and A/CNK ratios mostly between 1.08 and 1.15, defining their peraluminous and medium-K calc-alkaline signatures. Their trace element signatures show enrichments of LREEs and LILEs (e.g., Rb, Th, U, K, and Pb), depletion of HFSEs (e.g., Nb, Ti, Ta, and P), and negative Eu anomalies (Eu/Eu* = 0.22–0.66). These features suggest that the felsic volcanics were derived from partial melting of the middle crust, without interaction with mantle melts. Considering all the previous data and geochemical features, the Maoniushan Formation volcanic rocks in NQ formed in a post-collisional extensional setting associated with asthenospheric mantle upwelling and delamination in the Early Devonian. Together with the regional data, this study proposed that the Proto-Tethys Ocean had closed and evolved to the continental subduction/collision orogeny stage during the Middle to Late Ordovician, evolved to the post-collisional extensional stage in the Early Devonian, and finally formed the Zongwulong Ocean (branches of the Paleo-Tethys Ocean) in the Late Carboniferous, forming the tectonic framework of the Paleo-Tethys Archipelagic Ocean in the northern margin of the Tibetan Plateau.

The North China Block and the South China Block collided in the Middle Triassic, but there is still a lack of consensus regarding the end of collisional orogeny and the closure time of the Paleo-Tethys. In this paper, we report zircon U–Pb ages and geochemistry for the Shimen pluton in the northern margin of the West Qinling Orogenic Belt to investigate its genesis and tectonic environment. The new findings allow to constrain the end time of the Triassic orogeny in the Qinling Orogenic Belt and the closure time of the Paleo-Tethys. The weighted average 206Pb/238U ages of the Shimen pluton are 218.6 ± 1.5 Ma and 221.0 ± 1.7 Ma. Thus, we suggest that the Shimen pluton crystallized at the 218.6 Ma and 221.0 Ma and was formed during the Late Triassic (Norian). The Shimen pluton is mainly syenogranite and has alkaline dark minerals aegirine–augite. It is composed of 73.45 to 77.80 wt.% SiO2, 8.28 to 9.76 wt.% alkali, and 11.35 to 13.58 wt.% Al2O3, with A/CNK ranging from 0.91 to 1.02 and 10,000 Ga/Al ranging from 2.39 to 3.15. These findings indicate that the Shimen pluton is typical A-type granite. The plutons have low rare earth element contents, ranging from 73.92 to 203.58 ppm, with a moderate negative Eu anomaly. All the samples are enriched in large-ion lithophile elements, such as Rb, Nd, Th and U, and light rare earth elements, and are depleted in high field strength elements, such as Nb, P, Zr, Ba, and Sr. The depletion of Ba, Sr, and Zr may be related to the fractionation and evolution of the granite. According to the petrological and geochemical characteristics, the Shimen pluton is an A1-type granite formed in an anorogenic extensional environment. Combined with its tectonic characteristics and petrogenesis, the Shimen pluton was probably formed by the partial melting of the crust under high temperature and low pressure in the intraplate environment after the subduction of the South China Block beneath the North China Block. This observation indicates that the Triassic orogeny in the Qinling Orogenic Belt had ended and the Paleo-Tethys-Mianlve Ocean had also closed by the Late Triassic (Norian).

With the increasing penetration of new-type loads such as electric vehicles and hydrogen fuel vehicles in urban power grids, the peak-to-valley load difference increases sharply, and a multi-energy coordination model is desirable. This article proposes a day-ahead operation model of an urban energy system considering traffic flows and peak shaving, which can positively contribute to multi-energy complement and low-carbon emission. The proposed model minimizes the total cost of electricity and gas by optimizing the charging and discharging strategies of energy storage, in which the output of the wind turbine and energy management of the energy hub are adaptively adjusted. The urban energy system is represented by a second-order cone (SOC) energy flow model, and hence, the optimization problem is modeled as a mixed integer SOC programming (MISOCP). Finally, test results on an integrated urban energy network indicate that the energy storage and multi-energy coordination can alleviate the peak load cutting and valley filling. The relationship between urban grid operation cost and peak-valley difference is also discussed. The maximum utilization of renewable energy sources using gasoline vehicles has been presented in this study to illustrate cost and emission reductions for a sustainable integrated electricity and transportation infrastructure.

Abstract Background Compelling evidence indicates that status epilepticus is a prevalent cause of rhabdomyolysis. However, cases of rhabdomyolysis induced by a single seizure accompanied by viral encephalitis are rarely reported. Herein, we present a case of adult Herpes Simplex Encephalitis complicated with rhabdomyolysis. Case presentation A 32-year-old male was patient presented with fever accompanied by episodes of convulsions, myalgia, and oliguria, which exacerbated the delirium. Routine blood examination showed impaired kidney function and elevated myoglobin (Mb) and creatine phosphokinase (CK) levels. MRI scanning revealed a damaged frontotemporal lobe and limbic system. In addition, herpes simplex virus (HSV) pathogen was identified in the cerebrospinal fluid thus indicating HSV infection. Therefore, a diagnosis of rhabdomyolysis triggered by HSV infection accompanied by epilepsy was made. Notably, the patient recovered well after early intervention and treatment. Conclusion The case presented here calls for careful analysis of rhabdomyolysis cases with unknown causes, minor seizures, and without status epilepticus. This case also indicates that HSV virus infection might contribute to the rhabdomyolysis.

Background: A panel of experts proposed a new definition of metabolic dysfunction-associated fatty liver disease (MAFLD) in 2020. To date, the associations between adipokines, such as adiponectin, adipsin, and visfatin and MAFLD remain unclear. Therefore, we aimed to evaluate the associations between each of these three adipokines and MAFLD using different diagnostic criteria. Methods: In total, 221 participants were included in our study based on medical examination. Detailed questionnaire information, physical examination, abdominal ultrasound, and blood-biochemical-test indexes were collected. The levels of adipokines were tested by using an enzyme immunoassay. Logistic regression models were used to assess the associations of the adipokines with MAFLD. Results: In total, 122 of the participants were diagnosed with MAFLD. Higher levels of adipsin and lower levels of adiponectin were found in the MAFLD group than in the non-MAFLD group (all p < 0.05). According to the logistic regression analysis, the ORs were 0.11 (95% CI: 0.05–0.23) for adiponectin, 4.46 (95% CI: 2.19–9.12) for adipsin, and 0.51 (95% CI: 0.27–0.99) for visfatin when comparing the highest tertile with the lowest tertile (all p-trend < 0.05). The inverse association between adiponectin and MAFLD was strongest when T2DM was used as the diagnostic criterion alone, and the positive association between adipsin and MAFLD was strongest when BMI was used as the diagnostic criterion alone. There was no significant association between visfatin and MAFLD, regardless of whether each of BMI, T2DM, or metabolic dysregulation (MD) was used as the diagnostic criterion for MAFLD alone. Conclusion: Adipsin levels were positively associated with MAFLD and adiponectin levels were inversely associated with MAFLD. The strength of these associations varied according to the different diagnostic criteria for MAFLD.

Targeting Nicotinamide adenine dinucleotide (NAD) metabolism has emerged as a promising anti-cancer strategy; we aimed to explore the health benefits of boosting NAD levels with nicotinamide riboside (NR) on hepatocellular carcinoma (HCC). We established three in vivo tumor models, including subcutaneous transplantation tumor model in both Balb/c nude mice (xenograft), C57BL/6J mice (allograft), and hematogenous metastatic neoplasm in nude mice. NR (400 mg/kg bw) was supplied daily in gavage. In-situ tumor growth or noninvasive bioluminescence were measured to evaluate the effect of NR on the HCC process. HepG2 cells were treated with transforming growth factor-β (TGF-β) in the absence/presence of NR in vitro. We found that NR supplementation alleviated malignancy-induced weight loss and metastasis to lung in nude mice in both subcutaneous xenograft and hematogenous metastasis models. NR supplementation decreased metastasis to the bone and liver in the hematogenous metastasis model. NR supplementation also significantly decreased the size of allografted tumors and extended the survival time in C57BL/6J mice. In vitro experiments showed that NR intervention inhibited the migration and invasion of HepG2 cells triggered by TGF-β. In summary, our results supply evidence that boosting NAD levels by supplementing NR alleviates HCC progression and metastasis, which may serve as an effective treatment for the suppression of HCC progression.

The formation of the East Kunlun Orogen (EKO) was related to the tectonic evolution of the Proto-Tethys and Paleo-Tethys Oceans. However, how the Paleo-Tethys Ocean transited from the Proto-Tethys Ocean, and whether the Paleo-Tethys Ocean subducted northward beneath the East Kunlun–Qaidam Terrane in Carboniferous to Permian times, is still highly debated. Early Carboniferous Halaguole and Late Carboniferous to Early Permian Haoteluowa formations are extensively outcropped in the EKO, north Tibetan Plateau, and have thus recorded key information about the tectonic processes of the Paleo-Tethys Ocean that have implications for the reconstruction of the Northern Paleo-Tethys Ocean (Buqingshan Ocean). Siliciclastic rocks within these formations are collected for petrogeological, geochemical, and detrital zircon U–Pb dating research. Our results show that sandstones from Halaguole and Haoteluowa formations have an average total quartz–feldspar–lithic fragment ratio of Q67F12L21 and Q50F20L30, respectively, indicating relatively high compositional maturity. The geochemical results suggest that the average values of the Chemical Index of Alteration (CIA) are 57.83 and 64.66; together with their angular to subangular morphology, this indicates that their source rocks suffered from weak weathering and the sandstones are the result of proximal deposition. Geochemical features such as the low La/Th, TiO2, and Ni values suggest that the parental rocks in the provenance area are mainly acidic igneous rocks with minor intermediate igneous and old sedimentary components. The detrital zircon U–Pb age spectrum of these samples is dominated by age peaks at ~405–503 Ma and ~781–999 Ma, with subordinate age peaks at ~1610–2997 Ma and ~1002–1529 Ma, which show tectono–thermal events similar to those of the North Qimatag Belt (NQB), North Kunlun Terrane (NKT), and South Kunlun Terrane (SKT). These features suggest a contribution from the Early Paleozoic magmatic arc and Proterozoic basements in the NQB, NKT, and SKT to the Halaguole and Haoteluowa formations in these areas. In addition, the youngest zircon age of ~440 Ma from these sandstones is greater than the depositional age of Halaguole and Haoteluowa formations, which is a typical basin depositional feature in a passive continental margin. Geochemical tectonic discrimination diagrams, based on a major and trace element Ti/Zr–La/Sc plot, in combination with a detrital zircon age distribution pattern, all suggest a passive continental margin setting. Considering this together with the previous data, we argue that the Paleo-Tethys Ocean did not begin to subduct northward and that there was no oceanic subduction zone in the south EKO during Carboniferous to Early Permian times. Combining this information with that from previous studies suggests that the initial opening of the Paleo-Tethyan Ocean may have occurred before the Early Carboniferous time, and all the branches of the Paleo-Tethys Ocean constituted a complex ocean–continent configuration across parts of what is now Asia during the Early Carboniferous to Early Permian.

The rhyolites which are widely exposed to the northern margin of the East Kunlun orogenic belt were chosen as a research object to discern the post-orogenic tectonic evolution of the East Kunlun orogenic belt and reconstruct the post-collision orogenic processes of the Buqingshan- A’nyemaqen Ocean. We researched zircon U-Pb ages and geochemistry characteristics of the Late Triassic rhyolites in the eastern segment of the East Kunlun Orogenic Belt in the northern Tibetan Plateau. Zircon U-Pb dating yields coeval ages of 200.4 ± 1.4 Ma and 202.8 ± 1.2 Ma for the Keri rhyolites of the East Kunlun Orogenic Belt, indicating that the volcanic rocks were formed in the Late Triassic Rhaetian–Early Jurassic Hettangian. The Keri rhyolite is a product of the late magmatism of the Elashan Formation volcanic rocks. The rhyolites include rhyolitic brecciated tuff lavas and rhyolitic tuff lavas. The rhyolites are peraluminous and are high-K calc-alkaline, with high contents of SiO2, K2O, TFe2O3, and low P2O5 contents. The A/CNK ratios range from 0.97 to 1.09, indicating that the rhyolites are metaluminous to weakly peraluminous. The chondrite-normalized rare earth element (REE) distribution shows a significant negative Eu anomaly and low total REE concentrations. All samples are depleted in high field strength elements (HFSEs, e.g., Eu, Sr, Ti, and P), heavy rare earth elements (HREEs), and enriched in large ion lithophile elements (LILEs, e.g., Rb, Zr, Nd, Th, and U) and light rare earth elements (LREEs). The Keri rhyolite has the characteristics of A1-type magmatic rock, formed in an anorogenic environment after the closure of the Paleo-Tethys Ocean, and was the product of late magmatism in the Elashan Formation volcanic rocks.

Soil remediation agents (SRAs) such as biochar and hydroxyapatite (HAP) have shown a promising prospect in in situ soil remediation programs and safe crop production. However, the effects of SRAs on soil microbial communities still remain unclear, particularly under field conditions. Here, a field case study was conducted to compare the effects of biochar and HAP on soil bacterial communities in a slightly Cd-contaminated farmland grown with sweet sorghum of different planting densities. We found that both biochar and HAP decreased the diversity and richness of soil bacteria, but they differently altered bacterial community structure. Biochar decreased Chao1 (−7.3%), Observed_species (−8.6%), and Shannon indexes (−1.3%), and HAP caused Shannon (−2.0%) and Simpson indexes (−0.1%) to decline. The relative abundance (RA) of some specific taxa and marker species was differently changed by biochar and HAP. Overall, sweet sorghum cultivation did not significantly alter soil bacterial diversity and richness but caused changes in the RA of some taxa. Some significant correlations were observed between soil properties and bacterial abundance. In conclusion, soil remediation with biochar and HAP caused alterations in soil bacterial communities. Our findings help to understand the ecological impacts of SRAs in soil remediation programs.

At present, the digital image watermarking algorithm has not embedded the synchronization signal in the image, resulting in the poor performance of the embedded image in terms of, for example, imperceptibility, anti-attack ability, and robustness. Therefore, a digital image watermarking algorithm based on scrambling and singular value decomposition is proposed. The digital watermark is preprocessed by dimensionality reduction and encryption; the digital image is processed in sections and embedded in the synchronization signal. The digital watermark image is embedded in the digital image watermark by using the low-frequency energy ratio technology of sound channel. The watermark image extraction step is designed to extract the watermark image. The transmittance of the degraded image is calculated, the transmittance is refined by relying on the soft matting algorithm and guiding filter, the image contrast in the image frequency domain is enhanced, and the results are mapped to the appropriate visual range to optimize the visual brightness of the image. Finally, according to the uniqueness of singular value matrix, the distributed characteristics of image matrix data are described, the visual effect is enhanced, and the research of digital image watermarking algorithm is completed. The experimental results show that the algorithm can extract the watermark information completely, the image contrast and singular value have been significantly improved, and the algorithm has better anti-attack performance.

Backgrounds: Up to February 16, 2020, 355 cases have been confirmed as having COVID-19 infection on the Diamond Princess cruise ship. It is of crucial importance to estimate the reproductive number (R0) of the novel virus in the early stage of outbreak and make a prediction of daily new cases on the ship. Method: We fitted the reported serial interval (mean and standard deviation) with a gamma distribution and applied “earlyR” package in R to estimate the R0 in the early stage of COVID-19 outbreak. We applied “projections” package in R to simulate the plausible cumulative epidemic trajectories and future daily incidence by fitting the data of existing daily incidence, a serial interval distribution, and the estimated R0 into a model based on the assumption that daily incidence obeys approximately Poisson distribution determined by daily infectiousness. Results: The Maximum-Likelihood (ML) value of R0 was 2.28 for COVID-19 outbreak at the early stage on the ship. The median with 95% confidence interval (CI) of R0 values was 2.28 (2.06–2.52) estimated by the bootstrap resampling method. The probable number of new cases for the next ten days would gradually increase, and the estimated cumulative cases would reach 1514 (1384–1656) at the tenth day in the future. However, if R0 value was reduced by 25% and 50%, the estimated total number of cumulative cases would be reduced to 1081 (981–1177) and 758 (697–817), respectively. Conclusion: The median with 95% CI of R0 of COVID-19 was about 2.28 (2.06–2.52) during the early stage experienced on the Diamond Princess cruise ship. The future daily incidence and probable outbreak size is largely dependent on the change of R0. Unless strict infection management and control are taken, our findings indicate the potential of COVID-19 to cause greater outbreak on the ship. Autism spectrum disorder (ASD) is a serious neurodevelopmental disorder and characterized by early childhood-onset impairments in social interaction and communication, restricted and repetitive patterns of behavior or interests. So far there is no effective treatment for ASD, and the pathogenesis of ASD remains unclear. Genetic and epigenetic factors have been considered to be the main cause of ASD. It is known that endocannabinoid and its receptors are widely distributed in the central nervous system, and provide a positive and irreversible change toward a more physiological neurodevelopment. Recently, the endocannabinoid system (ECS) has been found to participate in the regulation of social reward behavior, which has attracted considerable attention from neuroscientists and neurologists. Both animal models and clinical studies have shown that the ECS is a potential target for the treatment of autism, but the mechanism is still unknown. In the brain, microglia express a complete ECS signaling system. Studies also have shown that modulating ECS signaling can regulate the functions of microglia. By comprehensively reviewing previous studies and combining with our recent work, this review addresses the effects of targeting ECS on microglia, and how this can contribute to maintain the positivity of the central nervous system, and thus improve the symptoms of autism. This will provide insights for revealing the mechanism and developing new treatment strategies for autism.

The East Kunlun Orogen on the northern margin of the Tethyan orogenic system records a history of Gondwana dispersal and Laurasian accretion. Uncertainties remain regarding the detailed histories of northern branches of the Paleo-Tethys Ocean in East Kunlun Orogen (Buqingshan Ocean). Based on a synthesis of sedimentary, structural, lithological, geochemical, and geochronological data from the East Kunlun Orogen and adjacent regions, this paper discusses the spreading and northward consumption of the Paleo-Tethys Ocean during Late Paleozoic–Early Mesozoic times. The main evolutionary stages are: (1) during Carboniferous to Middle Permian, the Paleo-Tethys Ocean (Buqingshan Ocean) was in an ocean spreading stage, as suggested by the occurrence of Carboniferous MORB-, and OIB-type oceanic units and Carboniferous to Middle Permian Passive continental margin deposits; (2) the Buqingshan Ocean subducted northward beneath the East Kunlun Terrane, leading to the development of a large continental magmatic arc (Burhan Budai arc) and forearc basin between ~270–240 Ma; (3) during the late Middle Triassic to early Late Triassic (ca. 240–230 Ma), the Qiangtang terrane collided with the East Kunlun–Qaidam terranes, leading to the final closure of the Buqingshan Ocean and occurrences of minor collision-type magmatism and potentially inception of the Bayan Har foreland basin; (4) finally, the East Kunlun Orogen evolved into a post-collisional stage and produced major magmatic flare-ups and polymetallic mineral deposits between Late Triassic to Early Jurassic (ca. 230–200 Ma), which is possibly related to asthenospheric mantle upwelling induced by delamination of thickened continental lithosphere and partial melting of the lower crust. In this paper, we propose that the Wilson cycle-like processes controlled the Late Paleozoic–Early Triassic tectonic evolution of East Kunlun, which provides significant implications for the evolution of the Paleo-Tethys Ocean.

Numerous Indosinian granitoids occur in the East Kunlun Orogen (EKO). The Indosinian was a key transitional period associated with the evolution of the Paleo-Tethys Ocean. Here, we study the relationship between the petrogenesis of the granitoids and the regional tectonic setting based on a comprehensive analysis of the petrology, geochronology, and geochemistry of typical granitoids in the eastern part of the EKO. The Indosinian granitoid compositions are dominated by quartz diorites, granodiorites, monzogranites, porphyritic monzogranites, and syenogranites. Early Indosinian granitoids are large, granitic batholiths, while the middle and late Indosinian granitoids are smaller in size. From the early Indosinian to late Indosinian, the granitoids show a transition from a medium-K calc-alkaline to high-K calc-alkaline composition. They are enriched in light rare earth elements (LREEs) and large-ion lithophile elements (LILEs) and depleted in high-field-strength elements (HFSEs), especially for the Helegangxilikete and the Kekeealong plutons. The late Indosinian granitoids have relatively low Y and Yb contents, high Sr contents, and high La/Yb and Sr/Y ratios, which suggests adakitic affinity. The zircon saturation temperatures of the early Indosinian syenogranite and the Keri syenogranite are above 800 °C. The zircon saturation temperatures of other Indosinian granites (average 749 °C) are lower than those of the biotite and amphibole partial melting experiment. In the early Indosinian (255–240 Ma), numerous granitoids were the products of the partial melting of the juvenile lower crust by mafic magma underplating. This underplating is geodynamically related to the continuous subduction of a branch of Paleo-Tethys Ocean, with slab break-off, rapid upwelling, and mantle decompression. In the middle Indosinian (240–230 Ma), the compression that accompanied the continent–continent collision was not conducive to fluid activity, and hence, the formation of magma could be attributed to dehydration partial melting of muscovite, biotite, or amphibole. In the late Indosinian (230–200 Ma), the delamination of thickened crust would provide heat and channels for fluid migration, leading to a flare-up of the magmas. The composition and petrogenesis of the Indosinian granitoids in the eastern EKO are the result of processes associated with the subduction, collisional, and post-collisional stages, during the evolution of the Paleo-Tethys Ocean.

Background: The purpose of this study was to investigate the relationship between folic acid and iron nutrition during pregnancy and congenital heart disease (CHD) in the offspring. Methods: Conditional logistic regression models and nonlinear mixed-effects models were used to analyze the effects of folic acid and iron nutrition during pregnancy on CHD in offspring. Results: After adjusting for confounders, folic acid or iron supplementation during pregnancy reduced the risk for fetal CHD (OR = 0.60 (0.45, 0.82) or 0.36 (0.27, 0.48)). Similarly, dietary iron intake during pregnancy (≥29 mg/d) was associated with a reduced risk of fetal CHD (OR = 0.64 (0.46, 0.88)). Additionally, compared with women who only supplemented folic acid (OR = 0.59 (0.41, 0.84)) or iron (OR = 0.32 (0.16, 0.60)), women who supplemented both folic acid and iron had lower risk for newborns with CHD (OR = 0.22 (0.15, 0.34)). Similarly, compared with women who only supplemented folic acid (OR = 0.59 (0.41, 0.84)) or higher dietary iron intake (≥29 mg/d) (OR = 0.60 (0.33, 1.09)), women who supplemented both folic acid and higher dietary iron intake (≥29 mg/d) had lower risk for the newborn with CHD (OR = 0.41 (0.28, 0.62)). The combined effects were significant in the multiplication model (OR = 0.35 (0.26, 0.48) or 0.66 (0.50, 0.85)) but not in the additive model. Conclusions: Our study found that folic acid and iron nutrition during pregnancy were associated with a reduced risk of CHD in the offspring and confirmed a statistically significant multiplicative interaction between folic acid and iron nutrition on the reduced risk of CHD in offspring.

Spinal stenosis is a common disease affecting the elderly that is present in a various forms. Its high incidence forces researchers to pay more attention and offer countermeasures. We used the Web of Science Core collection and PubMed database to obtain 5,606 scientific studies concerning spinal stenosis, and the number of publications maintained a roughly increasing trend from 108 in 2000 to 512 in 2018, only declining in 2011. Bibliometric analysis was conducted using the online analysis software CiteSpace and Bibliographic Item Co-Occurrence Matrix Builder (BICOMB). The United States maintains academic leadership in this field. The journal SPINE was the most authoritative, with 695 articles and an average of 12.73 citations. The exported major MeSH terms were further biclustered with gCLUTO according to co-word analysis to reveal research hotspots, including etiology, pathogenesis, clinical manifestation, conservative treatment, operative indication, internal implantation, and postoperative complications. After combination, the main topics focused on pathogenesis and surgical treatment. Narrowing causes flavum ligamentum hypertrophy, and posterior longitudinal ligament ossification is widely accepted. Additionally, minimally invasive surgery and internal implantation fixation are more valid in the clinic. Refining pathological classification and optimizing surgical methods and instrument properties will be important future research directions for spinal stenosis.

Summary: Flexible viscoelastic sensors have gained significant attention in wearable devices owing to their exceptional strain-dependent electrical resistance. Most of the strain sensors are elastic composites, thus the internal stress is often preserved during the deformation when they are attached to the uneven target. Therefore, there is a pressing need for viscoelastic composites with highly self-adapted electromechanical properties sensitive to multiexternal circumstances. This work reports a liquid-metal-filled magnetorheological plastomer (LMMRP) that shows a high response behavior to the external stimulus such as magnetic field, temperature, and force. The shape-deformable LMMRP can transform from an insulator to a conductor under applying a magnetic field, thus the further viscoelastic sensor possesses a magnetic field “on-off” switch effect. The microstructure-dependent magnetic/thermal/mechanical-electrical coupling characteristics are investigated, and several proof-of-concept sensor applications, such as magnetic control, environment recognition, and motion monitoring, are demonstrated. These LMMRP composites show a broad potential in flexible sensors and soft electronics.

Abstract Background Chemotherapy is the primary established systemic treatment for patients with breast cancer, especially those with the triple-negative subtype. Simultaneously, the resistance of triple-negative breast cancer (TNBC) to chemotherapy remains a major clinical problem. Our previous study demonstrated that the expression levels of PTN and its receptor PTPRZ1 were upregulated in recurrent TNBC tissue after chemotherapy, and this increase was closely related to poor prognosis in those patients. However, the mechanism and function of chemotherapy-driven increases in PTN/PTPRZ1 expression are still unclear. Methods We compared the expression of PTN and PTPRZ1 between normal breast and cancer tissues as well as before and after chemotherapy in cancer tissue using the microarray analysis data from the GEPIA database and GEO database. The role of chemotherapy-driven increases in PTN/PTPRZ1 expression was examined with a CCK-8 assay, colony formation efficiency assay and apoptosis analysis with TNBC cells. The potential upstream pathways involved in the chemotherapy-driven increases in PTN/PTPRZ1 expression in TNBC cells were explored using microarray analysis, and the downstream mechanism was dissected with siRNA. Results We demonstrated that the expression of PTN and PTPRZ1 was upregulated by chemotherapy, and this change in expression decreased chemosensitivity by promoting tumour proliferation and inhibiting apoptosis. CDKN1A was the critical switch that regulated the expression of PTN/PTPRZ1 in TNBC cells receiving chemotherapy. We further demonstrated that the mechanism of chemoresistance by chemotherapy-driven increases in the CDKN1A/PTN/PTPRZ1 axis depended on the NF-κB pathway. Conclusions Our studies indicated that chemotherapy-driven increases in the CDKN1A/PTN/PTPRZ1 axis play a critical role in chemoresistance, which suggests a novel strategy to enhance chemosensitivity in breast cancer cells, especially in those of the triple-negative subtype.

Due to the characteristics of high strength, high chemical activity and low heat conduction, titanium alloy materials are generally difficult to machine. As a typical titanium alloy with higher strength and lower heat conductivity, the machinability of titanium alloy TC21 is very poor and its cutting process is companied with larger cutting force and rapid tool wear. Straight-tooth milling tool is often used to cut the groove and side surface in the titanium alloy parts. And the milling method can be used to investigate the cutting mechanism because the cutting force has only two components and the better chip morphology is obtained. To investigate the straight-tooth milling process of TC21 alloy, a series of milling force experiments have been done. In addition, a 3D finite element model (FEM) for the straight-tooth milling process of TC21 alloy is presented to simulate the milling process. In the model, the constitutive material model, the failure model, the friction model and the heat transfer model were adopted. Through the simulation, chip formation, stress distribution, cutting force and milling temperature were obtained. The cutting force reaches its maximum when the spindle speed reaches about 13000 rpm, and then decreases as the speed continues to increase. The results confirmed that the similar "Salomon" phenomenon existed in the cutting process of TC21 alloy.

Background: Nicotinamide riboside (NR) is a nicotinamide adenine dinucleotide (NAD+) precursor which is present in foods such as milk and beer. It was reported that NR can prevent obesity, increase longevity, and promote liver regeneration. However, whether NR can prevent ethanol-induced liver injuries is not known. This study aimed to explore the effect of NR on ethanol induced liver injuries and the underlying mechanisms. Methods: We fed C57BL/6 J mice with Lieber-DeCarli ethanol liquid diet with or without 400 mg/kg·bw NR for 16 days. Liver injuries and SirT1-PGC-1α-mitochondrial function were analyzed. In in vitro experiments, HepG2 cells (CYP2E1 over-expressing cells) were incubated with ethanol ± 0.5 mmol/L NR. Lipid accumulation and mitochondrial function were compared. SirT1 knockdown in HepG2 cells were further applied to confirm the role of SirT1 in the protection of NR on lipid accumulation. Results: We found that ethanol significantly decreased the expression and activity of hepatic SirT1 and induced abnormal expression of enzymes of lipid metabolism in mice. Both in vivo and in vitro experiments showed that NR activated SirT1 through increasing NAD+ levels, decreased oxidative stress, increased deacetylation of PGC-1α and mitochondrial function. In SirT1 knockdown HepG2 cells, NR lost its ability in enhancing mitochondrial function, and its protection against lipid accumulation induced by ethanol. Conclusions: NR can protect against ethanol induced liver injuries via replenishing NAD+, reducing oxidative stress, and activating SirT1-PGC-1α-mitochondrial biosynthesis. Our data indicate that SirT1 plays an important role in the protection of NR against lipid accumulation and mitochondrial dysfunctions induced by ethanol. Cholinergic neurons in the diagonal band of Broca degenerate early in Alzheimer’s disease. Here the authors show that in healthy mice, these cholinergic inputs innervate newborn neurons in the hippocampus, and that loss of this innervation in an Alzheimer’s disease model leads to impairments in spatial memory.

Abstract An abnormal neuronal activity in the amygdala is involved in the pathogenesis of anxiety disorders. However, little is known about the mechanisms. High-anxiety mice and low-anxiety mice, representing the innate extremes of anxiety-related behaviors, were first grouped according to their anxiety levels in the elevated plus maze test. We found that the mRNA for endothelin-1 (ET1) and ET1 B-type receptors (ETBRs) in the amygdala was down-regulated in high-anxiety mice compared with low-anxiety mice. Knocking down basolateral amygdala (BLA) ET1 expression enhanced anxiety-like behaviors, whereas over-expressing ETBRs, but not A-type receptors (ETARs), had an anxiolytic effect. The combined down-regulation of ETBR and ET1 had no additional anxiogenic effect compared to knocking down the ETBR gene alone, suggesting that BLA ET1 acts through ETBRs to regulate anxiety-like behaviors. To explore the mechanism underlying this phenomenon further, we verified that most of the ET1 and the ET1 receptors in the BLA were expressed in pyramidal neurons. The ET1–ETBR signaling pathway decreased the firing frequencies and threshold currents for the action potentials of BLA pyramidal neurons but did not alter BLA synaptic neurotransmission. Together, these results indicate that amygdalar ET1-ETBR signaling could attenuate anxiety-like behaviors by directly decreasing the excitability of glutamatergic neurons.

Ischemic stroke, which accounts for 87% of all strokes, constitutes the leading cause of morbidity and mortality in China. Although the genetics and epigenetics of stroke have been extensively investigated, few studies have examined their relationships at different stages of stroke. This study assessed the characteristics of transcriptome changes at different stages of ischemic stroke using a mouse model of transient middle cerebral artery occlusion (tMCAO) and bioinformatics analyses. Cerebral cortex tissues from tMCAO mice at days 1, 3, 7, 14, and 28 were removed for RNA-Seq and small RNA-Seq library construction, sequencing, and bioinformatics analysis. We identified differentially expressed (DE) genes and miRNAs and revealed an association of the up-regulated or down-regulated DEmiRNAs with the correspondingly altered DEgene targets at each time point. In addition, different biological pathways were activated at different time points; thus, three groups of miRNAs were verified that may represent potential clinical biomarkers corresponding to days 1, 3, and 7 after ischemic stroke. Notably, this represents the first functional association of some of these miRNAs with stroke, e.g., miR-2137, miR-874-5p, and miR-5099. Together, our findings lay the foundation for the transition from a single-point, single-drug stroke treatment approach to multiple-time-point multi-drug combination therapies.

Background/Aims: Poor viability of transplanted mesenchymal stem cells (MSCs) within the ischemic heart limits their therapeutic potential for cardiac repair. Globular adiponectin (gAPN) exerts anti-apoptotic effects on several types of stem cells. Herein, we investigated the effect of gAPN on the MSCs against apoptosis induced by hypoxia and serum deprivation (H/SD). Methods: MSCs exposed to H/SD conditions were treated with different concentrations of gAPN. To identify the main type of receptor, MSCs were transfected with siRNA targeting adiponectin receptor 1 or 2 (AdipoR1 or AdipoR2). To elucidate the downstream pathway, MSCs were pre-incubated with AMPK inhibitor Compound C. Apoptosis, caspase-3 activity and mitochondrial membrane potential were evaluated. Results: H/SD-induced MSCs apoptosis and caspase-3 activation were attenuated by gAPN in a concentration-dependent manner. gAPN increased Bcl-2 and decreased Bax expressions. The loss of mitochondrial membrane potential induced by H/SD was also abolished by gAPN. The protective effect of gAPN was significantly attenuated after the knockdown of AdipoR1 rather than AdipoR2. Moreover, Compound C partly suppressed the anti-apoptotic effect of gAPN. Conclusions: gAPN inhibits H/SD-induced apoptosis in MSCs via AdipoR1-mediated pathway, possibly linked to the activation of AMPK. gAPN may be a novel survival factor for MSCs in the ischemic engraftment environment.

As one form in power-by-wire actuation system, the Electro backup hydraulic actuation system (EBHAs) has broad application prospects in the aviation field. In this paper, a numerical simulation model of the system performance for EBHAs is established. Based on the detailed description of its working principle, the mathematical models of EBHAs and its subsystems, as well as numerical model in the Matlab / Simulink environment are established, and a larger number of numerical simulations are carried out, which contain the normal working mode and the backup working mode. Furthermore, numerical simulation results are compared and analyzed with those results of experimental, which further verified the system simulation model established in this paper. It can be concluded from the results that the dynamic response of the system can meet the requirements of the design, no matter the normal or backup working modes. And the simulation results of the system sweep frequency are basically consistent with the test results, and the error is less than 10%. It is clear that the simulation model presented in this paper can accurately describe the system characteristics of EBHAs, which is useful to the design of EBHAs system.

Sepsis is a life-threatening state of organ dysfunction caused by infection and which can induce severe neurological disorders that lead to neuroinflammation and cognitive impairment. Inflammation has been reported to cause neuronal apoptosis in sepsis, which can finally lead to cognitive impairment. Previous studies have suggested that BML-111 can exhibit anti-inflammatory and proresolution activities. Additionally, silent information regulator 1 (SIRT1) can inhibit the NF-κB signaling pathway in an inflammation state. However, the role of the SIRT1/NF-κB signaling pathway in the protective effects of BML-111 against sepsis-induced neuroinflammation and cognitive impairment remains unclear. This study aimed to determine the effects of BML-111 on neuroinflammation and cognitive impairment induced by sepsis. Male C57BL/6J mice were subjected to cecal ligation and puncture (CLP) or a sham operation. BML-111 was administered via intracerebroventricular injection (0.1 mg/kg) immediately after CLP. Boc-2 (50 μg/kg) was administered intracerebroventricularly 30 min before CLP, and EX527 (10 μg) was administered every 2 days for a total of three times before CLP, also intracerebroventricularly. Some of the surviving mice underwent open-field, novel-object-recognition, and fear-conditioning behavioral tests at 7 days after surgery. Some of the other surviving mice were killed at 24 h after surgery to assess synaptic damage (PSD95 and Synapsin1), markers of inflammation [tumor necrosis factor alpha (TNF-α) and interleukin (IL)-1β], cytoplasmic p65, nuclear p65, Ac- NF-κB and SIRT1. At 48 h after CLP, TUNEL and glia-activation by immunofluorescence investigations were performed on a separate cohort of surviving animals. The results suggested that sepsis resulted in cognitive impairment, which was accompanied by the decreased the expression of PSD95 and Synapsin1, increased amount of TUNEL-positive cells and the activation of glias, increased production of TNF-α and IL-1β, increased expression of nuclear p65, Ac- NF-κB, and decreased expression of SIRT1 and cytoplasmic p65. It is especially notable that these abnormalities could be reduced by BML-111 treatment. EX527, an SIRT1 inhibitor, abolished the effects of BML-111. These results demonstrate that BML-111 can reduce the neuroinflammation and cognitive impairment induced by sepsis via SIRT/NF-κB signaling pathway.

Research on fast-growing microbes holds promise for many industrial applications, including shortening test and trial times in research and development stages and reducing the operation costs for production. Such microbes can be obtained either by selecting naturally occurring variants or via metabolic engineering approaches, either eliminating ‘unnecessary’ or adding necessary pathways affecting growth speed in the cell. Here, we review recent research and development of engineered fast-growing strains in industrial biotechology, with a special focus on vaccine production using (synthetic biology) engineered pathogenic strains. We will discuss whether this represents a security concern and whether the industrial biotech sector needs to pay more attention to issues of Gain-of-Function (GoF) while developing and harnessing these fast-growing microbes. We will also shed a light on the use of in-built biosafety circuits as a way to control the propagation of fast-growing strains, including their capacity to survive in the environment. Other possible GoF concerns raised by the publication of research results in this field will be also addressed. In conclusion, judging from the current development from the field, assessing the potential GoF risks on engineered fast-growing microbes does not lead to a clear generalized outcome. We argue that fast growing strains need to be evaluated in combination with their wild type and engineered characteristics, and require always a case-by-case assessment. Monitoring the progress of the field and proactively raising awareness on the GoF issues among the scientists are important for the further development of the field.
