Abstract

Research has been conducted on the cytotoxic marine natural products bengamides B and E. The preparation of these bengamides has been carried out via the amide coupling of a protected polyhydroxylated lactone intermediate with the substituted aminocaprolactam intermediate and the results are presented.

Abstract

A reaction between 1-acetyl-1-(diazoacetyl) cyclopropane and 4-bromo-5, 5-dimethyl-2-cyclopentenone proceeds six steps to synthesize (plus or minus)-illudin M, a sesquiterpene produced by the fungus Omphalotus illudens. A carbonyl ylide 1,3-dipolar cycloaddition reaction catalyzed by a rhodium(II) carbenoid formed from the cyclopropane and catalytic rhodium(II) diacetate is the primary step of the synthesis. Alteration of the dipole or dipolarophile in the cycloaddition step leads to illudin M synthesis.
