Abstract

Results suggest that there is an intersite communication between the glutaminase, transamidase, and ATP hydrolysis activities of amidotransferase enzyme. Data point out that the reactions are kinetically and stoichiometrically coupled and ATP hydrolysis and may be involved in the enzyme activation.

Abstract

A 15 amino acid peptide that can be used as an inhibitor of MAP kinase ER2 phosphorylation by the upstream kinase MEK has been synthesized. The peptide does not have much secondary structure in aqueous buffer but can assume an alpha-helical structure in aprotic solvent. It can also be used as an inhibitor of p38 and ERK phosphorylation by MKK3 but is 3-fold weaker as an inhibitor of SEK activation by JNK1.

Abstract

Conformational changes in the tropomyosin molecule initiate distinct excimer changes in the smooth muscle tropomyosin isoforms beta and gamma in the presence of caldesmon (CaD). In the absence of actin, the excimer fluorescence from pyrene at Cys-190 of gamma-tropomyosin homodimer decreases by adding CaD, while the excimer from the Cys-36 of beta-tropomyosin homodimer indicates a biphasic change.
