Abstract

The variable regions of the alpha- and beta-chains of the T-cell receptor (TCR) of the cytotoxic T-lymphocyte clone 2C was expressed in Escherichia coli. The single chain subunit was overexpressed as a 27-kDa protein. Serological examination showed that the 27-kDa protein is in the native conformation, being reactive to anti-antibodies to TCR. Therefore, this expression system may be used to produce receptor domains in quantities amenable to nuclear magnetic resonance analyses. Furthermore, the proteins may be used as modulators of the immune response.
