The thrombin-bound structure of fibrinopeptide A was determined by nuclear magneticresonance imaging analysis coupled with template aligniment using bovine trypsin inhibitor bound to trypsin as a template. The results showed that a previously described model having unsatisfactory binding possibilities could beimproved by reorienting two residues. The analysis led to the design and synthesis of hybrid peptide mimetics and inhibitors which could successfully bind to thrombin. Similar approaches may be used for studying secondary structure specificity in other systems, like the cleavage of prohormones and zymogens.
