Researchers describe a system that can distinguish viable bacterial cells from damaged bacterial cells by using permeant and impermeant fluorescent probes. A full or partial quenching of the permeant probe by the impermeant probe will occur if the cell is damaged and no quenching will occur if it is intact and viable.

Abstract

An assessment is presented on a technique combining the LIVE/DEAD BacLight viability stain with confocal scanning laser microscopy, using the probiotic strains Bifidobacterium sp. strain UCC35612 and Lactobacillus paracasei NFBC 338. Findings indicate that the BacLight stain technique needs to be optimized, as it yielded bacterial counts higher than those obtained by conventional nutrient-algar-based plate counting.

The fluorescent protein stain NanoOrange can be used to detect bacterial flagella and is as accurate as transmission electron microscopy. Its advantage over TEM is that it stains rapidly, does not require fixation or dehydration, and consequently can be used on live samples.

Abstract

Staining Cryptosporidium parvum oocysts with a fluorescent dye can distinguish between those that are viable and those that are not. A study using the fluorogenic dyes SYTO-9 and SYTO-59 showed that oocysts that fluoresced strongly were not infective whereas those that did not fluoresce or only fluoresced weakly were infective.

Abstract

Eight blue nucleic acid dyes were compared on their utility in staining living and dead bacteria from freshwater and seawater samples undergoing flow cytometric analysis for bacterial load. Results reveal that the SYBR-II and SYTO-9 are efficient in staining living and dead microorganisms from freshwater samples. Bacterial enumeration from fixed and unfixed seawater samples are better carried out with the SYBR-II dye.

A Gram stain of cervical secretions can accurately detect most women who do not have gonorrhea or chlamydia. This technique is much cheaper than other assays such as enzyme immunoassay (ELISA), DNA probe hybridization and PCR. For this reason, it should be used as the initial test in population screening.

Abstract

The Pap smear appears to be moderately useful in identifying patients with bacterial vaginosis. Gram stain tests to confirm bacterial vaginosis were performed on 210 patients with suspicious Pap smear tests. Fifty-five percent of the patients positive for bacterial vaginosis also had evidence of the infection on their Pap smear. Nearly all of the patients negative for bacterial vaginosis had no evidence of the infection on their Pap smear.
