This Assessment Update by the Environmental Effects Assessment Panel (EEAP) of the United Nations Environment Programme (UNEP) considers the interactive effects of solar UV radiation, global warming, and other weathering factors on plastics. The Assessment illustrates the significance of solar UV radiation in decreasing the durability of plastic materials, degradation of plastic debris, formation of micro- and nanoplastic particles and accompanying leaching of potential toxic compounds. Micro- and nanoplastics have been found in all ecosystems, the atmosphere, and in humans. While the potential biological risks are not yet well-established, the widespread and increasing occurrence of plastic pollution is reason for continuing research and monitoring. Plastic debris persists after its intended life in soils, water bodies and the atmosphere as well as in living organisms. To counteract accumulation of plastics in the environment, the lifetime of novel plastics or plastic alternatives should better match the functional life of products, with eventual breakdown releasing harmless substances to the environment., (© 2024. Forcibly displaced populations are among the most vulnerable groups in disasters. They experience poorer health conditions compared with nondisplaced individuals. However, a clear picture is lacking regarding the overall health problems encountered by disaster-induced mid- to long-term displaced people. This study investigated these disorders prevalence and identified their correlates among long-settled displaced populations worldwide. The current scoping review follows the PRISMA-ScR guidelines; a systematic search was conducted on PubMed, Web of Science, and CINAHL and included original peer-reviewed studies, commentary, reviews, and grey literature published in English between January 1990 to June 2022. In the thematic and content analysis, the authors applied the narrative review approach to identify themes and sub-themes. Forty-eight documents were identified as fully relevant to this study. The largest number of published papers were from Asia, followed by the Middle East, the United States, and Europe. IDPs in developed countries were the most researched populations. Human-made disasters were addressed by 89% of the included studies. The four main thematic categories included were "physical health," "mental health," "inadequate facilities," and "lack of healthy behaviour." The worsening of noncommunicable diseases had the highest prevalence, followed by communicable diseases. Due to their condition, forcibly displaced migrants face a triple burden of communicable diseases and noncommunicable diseases such as mental health issues. Health-related research and policy need to consider the links among disasters, health problems, and forced migration as a determinant of health in the new era of climate change-driven displacements.

Attempts to link human development and biodiversity conservation goals remain a constant feature of policy and practice related to protected areas (PAs). Underlying these approaches are narratives that simplify assumptions, shaping how interventions are designed and implemented. We examine evidence for five key narratives: 1) conservation is pro-poor; 2) poverty reduction benefits conservation; 3) compensation neutralises costs of conservation; 4) local participation is good for conservation; 5) secure tenure rights for local communities support effective conservation. Through a mixed-method synthesis combining a review of 100 peer-reviewed papers and 25 expert interviews, we examined if and how each narrative is supported or countered by the evidence. The first three narratives are particularly problematic. PAs can reduce material poverty, but exclusion brings substantial local costs to wellbeing, often felt by the poorest. Poverty reduction will not inevitably deliver on conservation goals and trade-offs are common. Compensation (for damage due to human wildlife conflict, or for opportunity costs), is rarely sufficient or commensurate with costs to wellbeing and experienced injustices. There is more support for narratives 4 and 5 on participation and secure tenure rights, highlighting the importance of redistributing power towards Indigenous Peoples and Local Communities in successful conservation. The accumulation of soluble and cell-wall bound UV-absorbing compounds (i.e., flavonoids) in the epidermis and the mesophyll of leaves is a response of plants to UV exposure. These compounds are known to function in UV screening, but they are also of potential value for food quality. One way to non-destructively monitor UV screening in leaves is by optical methods, from which UVA-PAM and Dualex instruments stand out. The degree and rapidity to which plants can modulate UV screening in response to fluctuating solar UV conditions is poorly understood. In this study, okra plants were exposed to two solar radiation treatments (near-ambient UV [+UV] and attenuated UV [-UV]) and the epidermal UV transmittance (T UV ; UVA-PAM) and flavonoid index (Dualex) were measured in the youngest and second youngest mature leaves over three consecutive days and within an individual day. The day-to-day (measured near solar noon) and diurnal (over the course of a day) measurements of leaf optical properties indicated that T UV decreased and flavonoid index increased in the adaxial epidermis ~50% until 15:00 CDT then returned close to morning values later in the day. Correlations between UV-B radiation and T UV and flavonoid index revealed highest values 30 min to 1 h prior to the measurements. These findings indicate that plants can respond quickly to fluctuating solar UV conditions and underlines the importance of the harvest-time point for health-promoting compounds in fruit and vegetables. Flavonoids are favored compounds in plant responses to UV exposure and act in UV absorption and antioxidant activity. Here, it was investigated, with okra as a model species, how fast plants can react to changing UV conditions and to what extent these reactions take place. Okra ( Abelmoschus esculentus ) plants were exposed to either full or nearly no UV radiation. The diurnal rhythm of the plants was driven by the UV radiation and showed up to a 50% increase of the flavonoid content (measured optically in the +UV plants). This was reflected only in the trends in UV-absorption and antioxidant activity of the extracts but not in the soluble flavonoid glycosides and hydroxycinnamic acid derivatives. In a second experiment, a transfer from a -UV to a +UV condition at 9:00 CDT showed the immediate start of the diurnal rhythm, while this did not occur if the transfer occurred later in the day; these plants only started a diurnal rhythm the following day. After an adaptation period of seven days, clear differences between the +UV and -UV plants could be found in all parameters, whereas plants transferred to the opposite UV condition settle between the +UV and -UV plants in all parameters. Broadly, it can be seen that the flavonoid contents and associated functions in the plant are subject to considerable changes within one day and within several days due to the UV conditions and that this can have a considerable impact on the quality of plant foods.

Cyclic imine toxins exhibit fast acting neurotoxicity and lethality by respiratory arrest in mice explained by their potent antagonistic activity against muscular nicotinic acetylcholine receptors. We performed a survey of gymnodimine-A, 13-desmethyl spirolide-C, 13,19-didesmethyl spirolide-C, 20-methyl spirolide-G, pinnatoxin-A, pinnatoxin-G, portimine-A and 28-O-palmitoyl ester of pinnatoxin-G in 36 shellfish samples collected in coastal areas of 8 European countries using a microplate receptor binding assay and UPLC-MS/MS for toxin identification and quantification. The major toxins found in these samples were pinnatoxin-G, 20-methyl spirolide-G, 13-desmethyl spirolide-C, gymnodimine-A and portimine-A. Traces of 13,19-didesmethyl spirolide-C, pinnatoxin-A and 28-O-palmitoyl ester of pinnatoxin-G were also detected. The rapid death of mice was correlated with higher pinnatoxin-G concentrations in mussel digestive gland extracts injected intraperitoneally. Our survey included nontoxic control samples that were found to contain moderate to trace amounts of several cyclic imine toxins. Shellfish may bioaccumulate not only cyclic imine toxins but also a large number of acyl derivatives as a product of metabolic transformation of these neurotoxins. This is the first report in which portimine-A and 28-O-palmitoyl ester of pinnatoxin-G were detected in shellfish extracts from digestive glands of mussels collected in Ingril lagoon. The bioaccumulation of portimine-A is particularly of concern because it is cytotoxic and is able to induce apotosis. The mode of action of 28-O-palmitoyl ester of pinnatoxin-G was studied by receptor binding-assay and by two-electrode voltage clamp electrophysiology. The antagonistic behavior of the acylated pinnatoxin-G towards nicotinic acetylcholine receptor of muscle type is shown here for the first time. Since cyclic imine toxins are not regulated further monitoring of these emerging toxins is needed to improve evidence gathering of their occurrence in shellfish commercialized for human consumption in Europe given their potent antagonism against muscle and neuronal nicotinic acetylcholine receptors., (Copyright © 2020. Aquatic and terrestrial organisms are being exposed to a number of anthropogenically-induced environmental stresses as a consequence of climate change. In addition, climate change is altering various linkages that exist between ecosystems on land and in water. Here we compare and contrast how climate change is altering aquatic and terrestrial environments and address some of the ways that the organisms in these ecosystems, especially the primary producers, are being affected by climate change factors, including changes in temperature, moisture, atmospheric carbon dioxide and solar UV radiation. Whereas there are some responses to climate change in common between terrestrial and aquatic ecosystems (e.g., changes in species composition and shifting geographic ranges and distributions), there are also responses that fundamentally differ between these two (e.g., responses to UV radiation). Climate change is also disrupting land-water connections in ways that influence biogeochemical and hydrologic cycles, and biosphere-atmosphere interactions in ways that can modify how aquatic and terrestrial ecosystems are affected by climate change and can influence climate change. The effects of climate change on these ecosystems are having wide-ranging effects on ecosystem biodiversity, structure and function and the abilities of these systems to provide essential services., (Copyright © 2019 Elsevier B.V. The UVR8 photoreceptor in Arabidopsis thaliana is specific for ultraviolet-B (UV-B; 280-315 nm) radiation and its activation leads to a number of UV-B acclimation responses, including the accumulation of flavonoids. UVR8 participates in a signaling cascade involving COP1 and HY5 so that the absence of any of these components results in a reduction in the ability of a plant to accumulate flavonoids in response to UV; Cop1 mutants show high dropouts and hy5-ks50 hyh double mutants show very low levels of flavonoids. The predominant phenolics in Arabidopsis thaliana are sinapic acid derivatives as well as non-aclyated quercetin and kaempferol di- and triglycosides containing glucose and rhamnose as glycosylated sugar moieties. How this flavonoid profile in Arabidopsis thaliana is affected by UV radiation, how rapidly these changes occur in changing UV conditions, and which components of the UV-B signalling pathway are involved in rapid UV acclimatization reactions is poorly understood. In the present study, we examined these questions by characterizing the flavonoid profiles of Arabidopsis thaliana signalling mutants and wild types grown under different UV levels of constant UV-B+PAR ratios and then transferring a subset of plants to alternate UV conditions. Results indicate that flavonoid accumulation in Arabidopsis thaliana is triggered by UV and this response is amplified by higher levels of UV but not by all compounds to the same extent. The catechol structure in quercetin seems to be less important than the glycosylation pattern, e.g. having 2 rhamnose moieties in determining responsivity. At low UV+PAR intensities the introduction of UV leads to an initial tendency of increase of flavonoids in the wild types that was detected after 3 days. It took 7 days for these changes to be detected in plants grown under high UV+PAR intensities suggesting a priming of PAR. Thus, the flavonoid profile in Arabidopsis thaliana is altered over time following exposure to UV and PAR, but the functional significance of these changes is currently unclear.

Plants perceive ultraviolet-B (UV-B) radiation through the UV-B photoreceptor UV RESISTANCE LOCUS 8 (UVR8), and initiate regulatory responses via associated signalling networks, gene expression and metabolic pathways. Various regulatory adaptations to UV-B radiation enable plants to harvest information about fluctuations in UV-B irradiance and spectral composition in natural environments, and to defend themselves against UV-B exposure. Given that UVR8 is present across plant organs and tissues, knowledge of the systemic signalling involved in its activation and function throughout the plant is important for understanding the context of specific responses. Fine-scale understanding of both UV-B irradiance and perception within tissues and cells requires improved application of knowledge about UV-attenuation in leaves and canopies, warranting greater consideration when designing experiments. In this context, reciprocal crosstalk among photoreceptor-induced pathways also needs to be considered, as this appears to produce particularly complex patterns of physiological and morphological response. Through crosstalk, plant responses to UV-B radiation go beyond simply UV-protection or amelioration of damage, but may give cross-protection over a suite of environmental stressors. Overall, there is emerging knowledge showing how information captured by UVR8 is used to regulate molecular and physiological processes, although understanding of upscaling to higher levels of organisation, i.e. organisms, canopies and communities remains poor. Achieving this will require further studies using model plant species beyond Arabidopsis, and that represent a broad range of functional types. More attention should also be given to plants in natural environments in all their complexity, as such studies are needed to acquire an improved understanding of the impact of climate change in the context of plant-UV responses. Furthermore, broadening the scope of experiments into the regulation of plant-UV responses will facilitate the application of UV radiation in commercial plant production. By considering the progress made in plant-UV research, this perspective highlights prescient topics in plant-UV photobiology where future research efforts can profitably be focussed. This perspective also emphasises burgeoning interdisciplinary links that will assist in understanding of UV-B effects across organisational scales and gaps in knowledge that need to be filled so as to achieve an integrated vision of plant responses to UV-radiation.

Exposure of plants and animals to ultraviolet-B radiation (UV-B; 280-315 nm) is modified by stratospheric ozone dynamics and climate change. Even though stabilisation and projected recovery of stratospheric ozone is expected to curtail future increases in UV-B radiation at the Earth's surface, on-going changes in climate are increasingly exposing plants and animals to novel combinations of UV-B radiation and other climate change factors (e.g., ultraviolet-A and visible radiation, water availability, temperature and elevated carbon dioxide). Climate change is also shifting vegetation cover, geographic ranges of species, and seasonal timing of development, which further modifies exposure to UV-B radiation. Since our last assessment, there has been increased understanding of the underlying mechanisms by which plants perceive UV-B radiation, eliciting changes in growth, development and tolerances of abiotic and biotic factors. However, major questions remain on how UV-B radiation is interacting with other climate change factors to modify the production and quality of crops, as well as important ecosystem processes such as plant and animal competition, pest-pathogen interactions, and the decomposition of dead plant matter (litter). In addition, stratospheric ozone depletion is directly contributing to climate change in the southern hemisphere, such that terrestrial ecosystems in this region are being exposed to altered patterns of precipitation, temperature and fire regimes as well as UV-B radiation. These ozone-driven changes in climate have been implicated in both increases and reductions in the growth, survival and reproduction of plants and animals in Antarctica, South America and New Zealand. In this assessment, we summarise advances in our knowledge of these and other linkages and effects, and identify uncertainties and knowledge gaps that limit our ability to fully evaluate the ecological consequences of these environmental changes on terrestrial ecosystems.

Dryland ecosystems cover nearly 45% of the Earth's land area and account for large proportions of terrestrial net primary production and carbon pools. However, predicting rates of plant litter decomposition in these vast ecosystems has proven challenging due to their distinctly dry and often hot climate regimes, and potentially unique physical drivers of decomposition. In this study, we elucidated the role of photopriming, i.e. exposure of standing dead leaf litter to solar radiation prior to litter drop that would chemically change litter and enhance biotic decay of fallen litter. We exposed litter substrates to three different UV radiation treatments simulating three-months of UV radiation exposure in southern New Mexico: no light, UVA+UVB+Visible, and UVA+Visible. There were three litter types: mesquite leaflets (Prosopis glandulosa, litter with high nitrogen (N) concentration), filter paper (pure cellulose), and basswood (Tilia spp, high lignin concentration). We deployed the photoprimed litter in the field within a large scale precipitation manipulation experiment: ∼50% precipitation reduction, ∼150% precipitation addition, and ambient control. Our results revealed the importance of litter substrate, particularly N content, for overall decomposition in drylands, as neither filter paper nor basswood exhibited measurable mass loss over the course of the year-long study, while high N-containing mesquite litter exhibited potential mass loss. We saw no effect of photopriming on subsequent microbial decay. We did observe a precipitation effect on mesquite where the rate of decay was more rapid in ambient and precipitation addition treatments than in the drought treatment. Overall, we found that precipitation and N played a critical role in litter mass loss. In contrast, photopriming had no detected effects on mass loss over the course of our year-long study. These results underpin the importance of biotic-driven decomposition, even in the presence of photopriming, for understanding litter decomposition and biogeochemical cycles in drylands., Competing Interests: The authors have declared that no competing interests exist.

Ongoing changes in Earth's climate are shifting the elevation ranges of many plant species with non-native species often experiencing greater expansion into higher elevations than native species. These climate change-induced shifts in distributions inevitably expose plants to novel biotic and abiotic environments, including altered solar ultraviolet (UV)-B (280-315 nm) radiation regimes. Do the greater migration potentials of non-native species into higher elevations imply that they have more effective UV-protective mechanisms than native species? In this study, we surveyed leaf epidermal UV-A transmittance (T UV A ) in a diversity of plant species representing different growth forms to test whether native and non-native species growing above 2800 m elevation on Mauna Kea, Hawaii differed in their UV screening capabilities. We further compared the degree to which T UV A varied along an elevation gradient in the native shrub Vaccinium reticulatum and the introduced forb Verbascum thapsus to evaluate whether these species differed in their abilities to adjust their levels of UV screening in response to elevation changes in UV-B. For plants growing in the Mauna Kea alpine/upper subalpine, we found that adaxial T UV A , measured with a UVA-PAM fluorometer, varied significantly among species but did not differ between native (mean = 6.0%; n = 8) and non-native (mean = 5.8%; n = 11) species. When data were pooled across native and non-native taxa, we also found no significant effect of growth form on T UV A , though woody plants (shrubs and trees) were represented solely by native species whereas herbaceous growth forms (grasses and forbs) were dominated by non-native species. Along an elevation gradient spanning 2600-3800 m, T UV A was variable (mean range = 6.0-11.2%) and strongly correlated with elevation and relative biologically effective UV-B in the exotic V. thapsus ; however, T UV A was consistently low (3%) and did not vary with elevation in the native V. reticulatum . Results indicate that high levels of UV protection occur in both native and non-native species in this high UV-B tropical alpine environment, and that flexibility in UV screening is a mechanism employed by some, but not all species to cope with varying solar UV-B exposures along elevation gradients.

Phenylbutazone is a non-steroidal anti-inflammatory drug licensed for use in horses to treat musculoskeletal disorders. It is not permitted in the European Union for use in animals destined for the food chain. Official statistics provided by the European Food Safety Authority (EFSA) show that 0.18% of bovines tested in the European Union between 2008 and 2014 for non-steroidal anti-inflammatory drugs were non-compliant, with phenylbutazone representing over 28% of these. Anecdotal evidence suggests animals that have not been treated with the drug may have produced non-compliant samples, possibly through some form of contamination. In this study, ultra-high-performance liquid chromatography coupled with mass-spectrometric detection was applied to bovine plasma samples to determine if detectable residues (CCα = 0.28 ng ml -1 ) may occur in untreated animals as a result of environmental contamination through normal farming practice. The study demonstrates that waste from animals treated with phenylbutazone, and spread on an area of pasture, can contaminate untreated bovines grazing the pasture many weeks later. It was determined that this contamination, which can persist over a significant period, may be due to the ingestion of as little as 30 μg phenylbutazone by a 500 kg bullock.

Tetrodotoxin (TTX), often referred to as the 'puffer fish' poison, is a marine toxin and it has been identified as the agent responsible for many food poisoning incidents around the world. It is a neurotoxin that blocks voltage-gated sodium channels, resulting in respiratory paralysis and even death in severe cases. It is known to occur in many different species of fish and other organisms. The toxin is mainly found in the Southeast Asia region. Worryingly, TTX is starting to appear in European waters. It is suspected that this is a consequence of Lessepsian migration, also known as the Erythrean invasion. Therefore, straightforward and reliable extraction and analytical methods are now urgently required to monitor seafood of European origin for TTX. This paper provides a versatile, dependable and robust method for the analysis of TTX in puffer fish and trumpet shellfish using LC-MS/MS. A three-stage approach was implemented involving: (1) the screening of samples using fast multiple reaction monitoring (MRM) mass spectral analysis to identify quickly positive samples on a triple quadrupole mass spectrometer (QqQMS/MS), the API 3000; (2) a Fourier-transform (FT)-MS full-scan analysis of positive samples to collect qualitative data; and (3) a method with a longer chromatography run to identify and quantitate the positive samples using the QqQMS. The quantitative LC-QqQMS method delivered excellent linearity for solvent-based standards (0.01-7.5 µg ml -1 ; R 2  ≥ 0.9968) as well as for matrix-matched standards (0.05-37.50 µg g -1 ; R 2  ≥ 0.9869). Good inter-day repeatability was achieved for all the relevant analytes with %RSD values (n = 9) ranging from 1.11% to 4.97% over a concentration range of 0.01-7.5 µg ml -1 . A sample clean-up procedure for the puffer fish and trumpet shellfish was developed to ensure acceptable and reproducible recoveries to enable accurate and precise determination of TTX in a myriad of tissues types. Blank mackerel matrix was used for the TTX standard spiking studies in order to calculate the recoveries of the toxin during the extraction procedure. The recovery was 61.17% ± 5.42% for the extraction protocol. MS/MS studies were performed on a linear-trap quadruple-Orbitrap mass spectrometer (LTQ-Orbitrap) to obtain high-mass-accuracy data of the target analytes and their characteristic fragment ions in the puffer fish and trumpet shellfish samples. This facilitated identification of TTX and its associated analogues. These high-mass-accuracy studies facilitated the development of a rapid MRM-based quantitative method for TTX determination on the LC-QqQMS.

Tetrodotoxin (TTX) is an emerging toxin in the European marine environment. It has various known structural analogues. It acts as a sodium channel blocker; the ability of each analogue to bind to the sodium channel varies with the particular structure of each analogue. Thus, each analogue will vary in its toxic potential. TTX analogues co-occur in food samples at variable concentrations. An LC-MS method was developed for the identification and quantitation of several analogues of TTX using an LTQ-Orbitrap XL mass spectrometer. The LTQ-Orbitrap XL mass spectrometer facilitates high mass accuracy measurement up to 100,000 full width at half maximum (FWHM). Using high resolution at 100,000 FWHM allows for the identification of TTX and its analogues in various matrices, including puffer fish and molluscan shellfish samples (Δ ppm = 0.28-3.38). The confirmation of characteristic fragment ions of TTX and its analogues was achieved by determining their elemental formulae via high mass accuracy. A quantitative method was then developed and optimised using these characteristic fragment ions. The limit of quantitation (LOQ) of the method was 0.136 µg g(-1) (S/N = 10) and the limit of detection (LOD) was 0.041 µg g(-1) (S/N = 3) spiking TTX standard into TTX-free mackerel fish extracts. The method was applied to naturally contaminated puffer fish and molluscan shellfish samples to confirm the presence of TTX and its analogues.

The accumulation of ultraviolet (UV)-absorbing compounds (flavonoids and related phenylpropanoids) in the epidermis of higher plants reduces the penetration of solar UV radiation to underlying tissues and is a primary mechanism of acclimation to changing UV conditions resulting from ozone depletion and climate change. Previously we reported that several herbaceous plant species were capable of rapid, diurnal adjustments in epidermal UV transmittance (T UV), but how widespread this phenomenon is among plants has been unknown. In the present study, we tested the generality of this response by screening 37 species of various cultivated and wild plants growing in four locations spanning a gradient of ambient solar UV and climate (Hawaii, Utah, Idaho and Louisiana). Non-destructive measurements of adaxial T UV indicated that statistically significant midday decreases in T UV occurred in 49 % of the species tested, including both herbaceous and woody growth forms, and there was substantial interspecific variation in the magnitude of these changes. In general, plants in Louisiana exhibited larger diurnal changes in T UV than those in the other locations. Moreover, across all taxa, the magnitude of these changes was positively correlated with minimum daily air temperatures but not daily UV irradiances. Results indicate that diurnal changes in UV shielding are widespread among higher plants, vary both within and among species and tend to be greatest in herbaceous plants growing in warm environments. These findings suggest that plant species differ in their UV protection "strategies" though the functional and ecological significance of this variation in UV sunscreen protection remains unclear at present.

The accumulation of ultraviolet (UV)-absorbing compounds (flavonoids and related phenylpropanoids) and the resultant decrease in epidermal UV transmittance (TUV ) are primary protective mechanisms employed by plants against potentially damaging solar UV radiation and are critical components of the overall acclimation response of plants to changing solar UV environments. Whether plants can adjust this UV sunscreen protection in response to rapid changes in UV, as occurs on a diurnal basis, is largely unexplored. Here, we use a combination of approaches to demonstrate that plants can modulate their UV-screening properties within minutes to hours, and these changes are driven, in part, by UV radiation. For the cultivated species Abelmoschus esculentus, large (30-50%) and reversible changes in TUV occurred on a diurnal basis, and these adjustments were associated with changes in the concentrations of whole-leaf UV-absorbing compounds and several quercetin glycosides. Similar results were found for two other species (Vicia faba and Solanum lycopersicum), but no such changes were detected in Zea mays. These findings reveal a much more dynamic UV-protection mechanism than previously recognized, raise important questions concerning the costs and benefits of UV-protection strategies in plants and have practical implications for employing UV to enhance crop vigor and quality in controlled environments., (© 2015 John Wiley & Sons Ltd.)

We report the dramatic increase of the Seebeck coefficient S and thermoelectric performance of calcium cobaltite Ca3Co4O9+δ ceramics through non-stoichiometric addition of minute amount of Ba. The nominal chemistry of polycrystal pellets are Ca3BaxCo4O9+δ (x = 0, 0.01, 0.05, and 0.1). At 323 K, S of Ca3Co4O9+δ is 135 μV K(-1), whereas S of Ba incorporated Ca3Ba0.05Co4O9+δ is 162.5 μV·K(-1), which is the highest S value near room temperature regime reported for calcium cobaltite. The increase of S for Ca3Ba0.05Co4O9+δ sample is accompanied by the decrease of the electrical resistivity ρ, resulting in high power factor S(2)/ρ of 843 μW·m(-1) K(-2) at 1007 K. Moreover, the thermal conductivities κ of Ca3BaxCo4O9+δ decrease with the increase of the Ba addition. The figure-of-merit ZT for Ca3Ba0.05Co4O9+δ reaches 0.52 at 1073 K and a factor of 2.5 increment in comparison with undoped Ca3Co4O9+δ. Nanostructure examinations show that the added Ba segregated at the Ca3Co4O9+δ grain boundaries, while the Ca3Co4O9+δ grain interior is free of Ba. Performance enhancement is attributed to the carrier filtering effect caused by the Ba segregation. In addition, Ba segregation promotes the better crystal alignment and the development of crystal texture.

The accumulation of UV-absorbing compounds (flavonoids and other phenylpropanoid derivatives) and resultant decrease in the UV transmittance of the epidermis in leaves (TUV), is a primary protective mechanism against the potentially deleterious effects of UV radiation and is a critical component of the overall acclimation response of plants to changing UV environments. Traditional measurements of TUV were laborious, time-consuming and destructive or invasive, thus limiting their ability to efficiently make multiple measurements of the optical properties of plants in the field. The development of rapid, nondestructive optical methods of determining TUV has permitted the examination of UV optical properties of leaves with increased replication, on a finer time scale, and enabled repeated sampling of the same leaf over time. This technology has therefore allowed for studies examining acclimation responses to UV in plants in ways not previously possible. Here we provide a brief review of these earlier studies examining leaf UV optical properties and some of their important contributions, describe the principles by which the newer non-invasive measurements of epidermal UV transmittance are made, and highlight several case studies that reveal how this technique is providing new insights into this UV acclimation response in plants, which is far more plastic and dynamic than previously thought., (Copyright © 2014 Elsevier Masson SAS. Epidermal UV transmittance (TUV ) and UV-absorbing compounds were measured in sun and shade leaves of Populus tremuloides and Vicia faba exposed to contrasting light environments under field conditions to evaluate UV acclimation potentials and regulatory roles of photosynthetically active radiation (PAR) and UV in UV-shielding. Within a natural canopy of P. tremuloides, TUV ranged from 4 to 98% and showed a strong nonlinear relationship with mid-day horizontal fluxes of PAR [photon flux density (PFD) = 6-1830 µmol m⁻²  s⁻¹]; similar patterns were found for V. faba leaves that developed under a comparable PFD range. A series of field transfer experiments using neutral-density shade cloth and UV blocking/transmitting films indicated that PAR influenced TUV during leaf development to a greater degree than UV, and shade leaves of both species increased their UV-shielding when exposed to full sun; however, this required the presence of UV, with both UV-A and UV-B required for full acclimation. TUV of sun leaves of both species was largely unresponsive to shade either with or without UV. In most, but not all cases, changes in TUV were associated with alterations in the concentration of whole-leaf UV-absorbing compounds. These results suggest that, (1) moderate-to-high levels of PAR alone during leaf development can induce substantial UV-protection in field-grown plants, (2) mature shade leaves have the potential to adjust their UV-shielding which may reduce the detrimental effects of UV that could occur following sudden exposures to high light and (3) under field conditions, PAR and UV play different roles in regulating UV-shielding during and after leaf development., (© 2013 Scandinavian Plant Physiology Society.)

Tetrodotoxin (TTX) is a potent neurotoxin emerging in European waters due to increasing ocean temperatures. Its detection in seafood is currently performed as a consequence of using the Association of Analytical Communities (AOAC) mouse bioassay (MBA) for paralytic shellfish poisoning (PSP) toxins, but TTX is not monitored routinely in Europe. Due to ethical and performance-related issues associated with this bioassay, the European Commission has recently published directives extending procedures that may be used for official PSP control. An AOAC-accredited high-performance liquid chromatography (HPLC) method has now been accepted by the European Union as a first action screening method for PSP toxins to replace the MBA. However, this AOAC HPLC method is not capable of detecting TTX, so this potent toxin would be undetected; thereby, a separate method of analysis is required. Surface plasmon resonance (SPR) optical biosensor technology has been proven as a potential alternative screening method to detect PSP toxins in seafood. The addition of a similar SPR inhibition assay for TTX would complement the PSP assay in removing the MBA. The present report describes the development and single laboratory validation in accordance with AOAC and IUPAC guidelines of an SPR method to be used as a rapid screening tool to detect TTX in the sea snail Charonia lampas lampas, a species which has been implicated in 2008 in the first case of human TTX poisoning in Europe. As no current regulatory limits are set for TTX in Europe, single laboratory validation was undertaken using those for PSP toxins at 800 μg/kg. The decision limit (CCα) was 100 μg/kg, with the detection capability (CCβ) found to be ≤200 μg/kg. Repeatability and reproducibility were assessed at 200, 400, and 800 μg/kg and showed relative standard deviations of 8.3, 3.8, and 5.4% and 7.8, 8.3, and 3.7% for both parameters at each level, respectively. At these three respective levels, the recovery of the assay was 112, 98, and 99%.

Tetrodotoxin (TTX) is one of the most potent marine neurotoxins reported. The global distribution of this toxin is spreading with the European Atlantic coastline now being affected. Climate change and increasing pollution have been suggested as underlying causes for this. In the present study, two different sample preparation techniques were used to extract TTX from Trumpet shells and pufferfish samples. Both extraction procedures (accelerated solvent extraction (ASE) and a simple solvent extraction) were shown to provide good recoveries (80-92%). A UPLC-MS/MS method was developed for the analysis of TTX and validated following the guidelines contained in the Commission Decision 2002/657/EC for chemical contaminant analysis. The performance of this procedure was demonstrated to be fit for purpose. This study is the first report on the use of ASE as a mean for TTX extraction, the use of UPLC-MS/MS for TTX analysis, and the validation of this method for TTX in gastropods., (Copyright © 2012 Elsevier Ltd. A new data collection strategy for performing synchrotron energy-dispersive X-ray diffraction computed tomography has been devised. This method is analogous to angle-dispersive X-ray diffraction whose diffraction signal originates from a line formed by intersection of the incident X-ray beam and the sample. Energy resolution is preserved by using a collimator which defines a small sampling voxel. This voxel is translated in a series of parallel straight lines covering the whole sample and the operation is repeated at different rotation angles, thus generating one diffraction pattern per translation and rotation step. The method has been tested by imaging a specially designed phantom object, devised to be a demanding validator for X-ray diffraction imaging. The relative strengths and weaknesses of the method have been analysed with respect to the classic angle-dispersive technique. The reconstruction accuracy of the method is good, although an absorption correction is required for lower energy diffraction because of the large path lengths involved. The spatial resolution is only limited to the width of the scanning beam owing to the novel collection strategy. The current temporal resolution is poor, with a scan taking several hours. The method is best suited to studying large objects (e.g. for engineering and materials science applications) because it does not suffer from diffraction peak broadening effects irrespective of the sample size, in contrast to the angle-dispersive case.

Hydrogen storage properties of Ti-doped nanosized (~20 nm) NaAlH(4) supported on carbon nanofibers were affected by the stage at which Ti was introduced. When Ti was deposited first followed by NaAlH(4), sorption properties were superior to the case where NaAlH(4) was deposited first followed by NaAlH(4). This was the result of both a smaller NaAlH(4) particle size and the more extensive catalytic action of Ti in the former material.

High-throughput continuous hydrothermal flow synthesis has been used as a rapid and efficient synthetic route to produce a range of crystalline nanopowders in the Ce-Zn oxide binary system. High-resolution powder X-ray diffraction data were obtained for both as-prepared and heat-treated (850 degrees C for 10 h in air) samples using the new robotic beamline I11, located at Diamond Light Source. The influence of the sample composition on the crystal structure and on the optical and physical properties was studied. All the nanomaterials were characterized using Raman spectroscopy, UV-visible spectrophotometry, Brunauer-Emmett-Teller surface area and elemental analysis (via energy-dispersive X-ray spectroscopy). Initially, for 'as-prepared' Ce(1-x)Zn(x)O(y), a phase-pure cerium oxide (fluorite) structure was obtained for nominal values of x=0.1 and 0.2. Biphasic mixtures were obtained for nominal values of x in the range of 0.3-0.9 (inclusive). High-resolution transmission electron microscopy images revealed that the phase-pure nano-CeO(2) (x=0) consisted of ca 3.7 nm well-defined nanoparticles. The nanomaterials produced herein generally had high surface areas (greater than 150 m(2) g(-1)) and possessed combinations of particle properties (e.g. bandgap, crystallinity, size, etc.) that were unobtainable or difficult to achieve by other more conventional synthetic methods.

Brief (1-100 min) irradiations with three different ultraviolet-B (UV-B) and ultraviolet-C (UV-C) wave bands induced increases the UV-absorbing pigments extracted from cucumber (Cucumis sativus L.) and Arabidopsis. Spectra of methanol/1% HCl extracts from cucumber hypocotyl segments spanning 250-400 nm showed a single defined peak at 317 nm. When seedlings were irradiated with 5 kJ m(-2) UV-B radiation containing proportionally greater short wavelength UV-B (37% of UV-B between 280 and 300 nm; full-spectrum UV-B, FS-UVB), tissue extracts taken 24 h after irradiation showed an overall increase in absorption (91% increase at 317 nm) with a second defined peak at 263 nm. Irradiation with 1.1 kJ m(-2) UV-C (254 nm) caused similar changes. In contrast, seedlings irradiated with 5 kJ m(-2) UV-B including only wavelengths longer than 290 nm (8% of UV-B between 290 and 300 nm; long-wavelength UV-B, LW-UVB) resulted only in a general increase in absorption (80% at 317 nm). The increases in absorption were detectable as early as 3 h after irradiation with FS-UVB and UV-C, while the response to LW-UVB was first detectable at 6 h after irradiation. In extracts from whole Arabidopsis seedlings, 5 kJ m(-2) LW-UVB caused only a 20% increase in total absorption. Irradiation with 5 kJ m(-2) FS-UVB caused the appearance of a new peak at 270 nm and a concomitant increase in absorption of 72%. The induction of this new peak was observed in seedlings carrying the fah1 mutation which disrupts the pathway for sinapate synthesis. The results are in agreement with previously published data on stem elongation indicating the existence of two response pathways within the UV-B, one operating at longer wavelengths (>300 nm) and another specifically activated by short wavelength UV-B (<300 nm and also by UV-C).

Tomographic energy dispersive diffraction imaging (TEDDI) is a recently developed synchrotron-based characterization technique used to obtain spatially resolved X-ray diffraction and fluorescence information in a noninvasive manner. With the use of a synchrotron beam, three-dimensional (3D) information can be conveniently obtained on the elemental composition and related crystalline phases of the interior of a material. In this work, we show for the first time its application to characterize the structure of a heterogeneous catalyst body in situ during thermal treatment. Ni/gamma-Al(2)O(3) hydrogenation catalyst bodies have been chosen as the system of study. As a first example, the heat treatment in N(2) of a [Ni(en)(3)](NO(3))(2)/gamma-Al(2)O(3) catalyst body has been studied. In this case, the crystalline [Ni(en)(3)](NO(3))(2) precursor was detected in an egg-shell distribution, and its decomposition to form metallic Ni crystallites of around 5 nm was imaged. In the second example, the heat treatment in N(2) of a [Ni(en)(H(2)O)(4)]Cl(2)/gamma-Al(2)O(3) catalyst body was followed. The initial [Ni(en)(H(2)O)(4)]Cl(2) precursor was uniformly distributed within the catalyst body as an amorphous material and was decomposed to form metallic Ni crystallites of around 30 nm with a uniform distribution. TEDDI also revealed that the decomposition of [Ni(en)(H(2)O)(4)]Cl(2) takes place via two intermediate crystalline structures. The first one, which appears at around 180 degrees C, is related to the restructuring of the Ni precursor on the alumina surface; the second one, assigned to the formation of a limited amount of Ni(3)C, is observed at 290 degrees C.

Tomographic Energy-Dispersive Diffraction Imaging (TEDDI) enables a unique non-destructive mapping of the interior of bulk objects, exploiting the full range of X-ray signals (diffraction, fluorescence, scattering, background) recorded. By analogy to optical imaging, a wide variety of features (structure, composition, orientation, strain) dispersed in X-ray wavelengths can be extracted and colour-coded to aid interpretation. The ultimate aim of this approach is to realise real-time high-definition colour X-ray diffraction imaging, on the timescales of seconds, so that one will be able to 'look inside' optically opaque apparatus and unravel the space/time-evolution of the materials chemistry taking place. This will impact strongly on many fields of science but there are currently two barriers to this goal: speed of data acquisition (a 2D scan currently takes minutes to hours) and loss of image definition through spatial distortion of the X-ray sampling volume. Here we present a data-collection scenario and reconstruction routine which overcomes the latter barrier and which has been successfully applied to a phantom test object and to real materials systems such as a carbonating cement block. These procedures are immediately transferable to the promising technology of multi-energy-dispersive-detector-arrays which are planned to deliver the other breakthrough, that of one-two orders of magnitude improvement in data acquisition rates, that will be needed to realise real-time high-definition colour X-ray diffraction imaging.

Aldehydes substituted with a quaternised pyridinium or quinolinium ring have been investigated for the development of latent fingerprints. Two routes were developed to a novel in situ formed azacyanine dye. This dye might form in the fingerprint where reagents are concentrated but does not form appreciably in solution experiments as evidenced by the lack of an absorption band at 600 nm. N-Alkyl and N-aryl substituted benzimidazole-2-carboxaldehydes give stable fluorescent fingerprints.

Studies were conducted on three herbaceous plant species growing in naturally high solar UV environments in the subalpine of Mauna Kea, Hawaii, USA, to determine if diurnal changes in epidermal UV transmittance (T(UV)) occur in these species, and to test whether manipulation of the solar radiation regime could alter these diurnal patterns. Additional field studies were conducted at Logan, Utah, USA, to determine if solar UV was causing diurnal T(UV) changes and to evaluate the relationship between diurnal changes in T(UV) and UV-absorbing pigments. Under clear skies, T(UV), as measured with a UV-A-pulse amplitude modulation fluorometer for leaves of Verbascum thapsus and Oenothera stricta growing in native soils and Vicia faba growing in pots, was highest at predawn and sunset and lowest at midday. These patterns in T(UV) closely tracked diurnal changes in solar radiation and were the result of correlated changes in fluorescence induced by UV-A and blue radiation but not photochemical efficiency (F(v)/F(m)) or initial fluorescence yield (F(o)). The magnitude of the midday reduction in T(UV) was greater for young leaves than for older leaves of Verbascum. Imposition of artificial shade eliminated the diurnal changes in T(UV) in Verbascum, but reduction in solar UV had no effect on diurnal T(UV) changes in Vicia. In Vicia, the diurnal changes in T(UV) occurred without detectable changes in the concentration of whole-leaf UV-absorbing compounds. Results suggest that plants actively control diurnal changes in UV shielding, and these changes occur in response to signals other than solar UV; however, the underlying mechanisms responsible for rapid changes in T(UV) remain unclear.

Elevated concentrations of sulfide in groundwater (approximately 63 mg S(2-)/L in water and 500 mg dissolved H2S/L dissipating from the wellhead) at a field site near South Lovedale (OK, USA) were inhibiting the activity of sulfate-reducing bacteria (SRB) that are known to degrade contaminants, including benzene, toluene, ethylbenzene, and xylenes. Elevated concentrations of these contaminants, except for toluene, also were present in this groundwater. Microcosms were established in the laboratory using groundwater and sediment collected from the field site and amended with various nutrient, substrate, and inhibitor treatments. All microcosms initially were amended with FeCl2 to induce FeS precipitation and, thereby, to reduce aqueous sulfide concentrations. Complete removal of benzene, ethylbenzene, and m+p-xylenes (BEX; o-xylene not detected) was observed within 39 d in treatments with various combinations of nutrient and substrate amendments, including treatments with no amendments (other than FeCl2). This indicates that the elevated concentration of sulfide is the only limiting factor to BEX biodegradation at this site under anaerobic conditions and that treating the groundwater with FeCl2 may be a simple remedy to both facilitate and enhance BEX degradation by the indigenous SRB population.

We examined the influence of short-term exposures of different UV wavebands on the elongation and phototropic curvature of hypocotyls of cucumbers (Cucumis sativus L.) grown in white light (WL) and dim red light (DRL). We evaluated (1) whether different wavebands within the ultraviolet B (UV-B) region elicit different responses; (2) the hypocotyl elongation response elicited by ultraviolet C (UV-C); (3) whether irradiation with blue light-enriched white light (B/WL) given simultaneous with UV-B treatments reversed the effect of UV in a manner indicative of photoreactivation; and (4) whether responses in WL-grown plants were similar to those grown in DRL. Responses to brief (1-100 min) irradiations with three different UV wavebands all induced inhibition of elongation measured after 24 h. When WL-grown seedlings were irradiated with light containing proportionally greater short wavelength UV-B (37% of UV-B between 280 and 300 nm), inhibition of hypocotyl elongation was induced at a threshold of 0.5 kJ m(-2), whereas exposure to UV-B including only wavelengths longer than 290 nm (and only 8% of UV-B between 290 and 300 nm) induced inhibition of hypocotyl elongation at a threshold of 1.6 kJ m(-2). The UV-C treatment induced reduction in elongation at a threshold of <0.01 kJ m(-2) for DRL-grown plants and <0.03 kJ m(-2) for WL-grown plants. B/WL caused 50% reversal of the short-wavelength UV-B-induced inhibition of elongation in DRL-grown seedlings but did not reverse the effect of long-wavelength UV-B. B/WL caused 30% reversal of the UV-C-induced inhibition of elongation in WL-grown seedlings but did not affect the response to short-wavelength UV-B. Short-wavelength UV-B also induced positive phototropic curvature in both types of seedlings, and this was reversed 60% or completely in DRL-grown and WL-grown seedlings, respectively. The similarity of responses between the etiolated (DRL-grown) and de-etiolated (WL-grown) seedlings indicates that the short-wavelength specific response may be relevant to natural light environments, and the apparent photoreactivation implicates DNA damage as the sensory mechanism for the response.

We examined the influence of short-term exposure of different UV wavebands on the fine-scale kinetics of hypocotyl growth of dim red light-grown cucumbers (Cucumis sativus L.) and other selected dicotyledonous seedlings to evaluate: (1) whether responses induced by UV-B radiation (280-320 nm) are qualitatively different from those induced by UV-A (320-400 nm) radiation, and (2) whether different wavebands within the UV-B elicit different responses. Responses to brief (30 min) irradiations with 3 different UV wavebands all included transient inhibition of elongation during irradiation followed by wavelength specific responses. Irradiations with proportionally greater short wavelength UV-B (37% of UV-B between 280 and 300 nm) induced inhibition of hypocotyl elongation within 20 min of onset of irradiation, while UV-B including only wavelengths longer than 290 nm (and only 8% of UV-B between 290 and 300 nm) induced inhibition of hypocotyl elongation with a lag of 1-2 h. The response to short wavelength UV-B was persistent for at least 24 h, while the response to long wavelength UV-B lasted only 2-3 h. The UV-A treatment induced reductions in elongation rates of approximately 6-9 h following exposure followed by a continued decline in rates for the following 15-18 h. Short wavelength UV-B also induced positive phototropic curvature in both cucumber and Arabidopsis seedlings, and this response was present in nph-1 mutant Arabidopsis seedlings defective in normal blue light phototropism. Reciprocity was not found for the response to short wavelength UV-B. The short wavelength and long wavelength UV-B responses differed in dose-response relationships and both short wavelength responses (phototropic curvature and elongation inhibition) increased sharply at wavelengths below 300 nm. These results indicate that different photosensory processes are involved in mediating growth and morphological responses to short wavelength UV-B (280-300 nm), long wavelength UV-B (essentially 300-320 nm) and UV-A. The existence of two separate types of hypocotyl inhibition responses to UV-B, with one that depends on the intensity of the light source, provides alternate interpretations to findings in other studies of UV-B induced photomorphogenesis and may explain inconsistencies between action spectra for inhibition of stem growth.

Regulation of cystic fibrosis transmembrane regulator (CFTR) and epithelial sodium channel (ENaC) in airway epithelia strongly influences the rate of mucociliary clearance (MCC) by determining the volume of airway surface liquid. MCC increases in response to stimuli originating on the airway surface, and CFTR and ENaC in airway epithelia appear to be regulated by local rather than systemic signaling. Although all signals that regulate CFTR and ENaC in airways have not been identified, the release of nucleotides from airway epithelial cells exposed to physical stimuli initiates a series of events that coordinately favor increased MCC. These events include activation of adenosine A2B receptors that stimulate CFTR and P2Y2 receptors that inhibit ENaC. Together these actions result in an increased volume of airway surface liquid and increased MCC rates. Stimulation of CFTR by A(2B)AR uses protein kinase (PK) A signaling elements that are localized within the apical/subapical compartment, including G proteins, adenylyl cyclase, PKA-II, A-kinase anchoring proteins, and phosphodiesterases. Inhibition of ENaC by P2Y2 receptors appears to be mediated by phospholipase C-beta3, possibly through an effect on the levels of phosphatidylinositol 4,5-bisphosphonate in the apical membrane.

The arborescent legume, honey mesquite (Prosopis glandulosa), appears to play a central role in patch dynamics of southern Texas savannas by modifying soils and microclimate and by facilitating the ingress, establishment and/or growth of shrubs in its understorey. As an indirect test for the occurrence and persistence of facilitation in mature shrub clusters (patches), we examined the gas exchange, water relations and production of associated shrubs growing in patches where a Prosopis overstorey was present and in patches where Prosopis had succumbed to natural mortality. Surface (0-10 cm) soils associated with shrub patches were enriched in total [N] and [C] compared to soils of neighboring herbaceous zones. However, there were no detectable differences in soil [N] or [C] in patches with and without Prosopis. Foliar [N] and biomass of various shrub species were also statistically comparable for patches with and without Prosopis. These results are in accordance with other studies that indicate the nutrient legacy associated with Prosopis occupation of a patch may persist for decades after its demise. In comparison to plants growing in the absence of Prosopis, leaf water potentials (predawn and midday), and net photosynthesis and water vapor conductance (morning and midday) of outer-canopy sunlit leaves over an annual growth cycle were comparable for two common evergreen shrubs, Zanthoxylum fagara and Berberis trifoliolata, growing in patches with a live Prosopis. These findings indicate that the presence of Prosopis was not enhancing the growth or activity of mature understorey shrubs; facilitation may, therefore, be important only during early stages of cluster development. In addition, we found no indication that the loss of Prosopis has initiated a downward phase in a cyclic succession of patch initiation, growth and death. Rather, the understorey shrubs appear to be able to maintain growth and productivity in the absence of a Prosopis overstorey, and may, therefore, represent persistent components of woody patches on these savanna landscapes.

Plant and soil water relationships in a typical nebraska Sandhills prairie were examined to 1) explain the observed distribution patterns of several dominant grasses along a topographic gradient, and 2) show how spatial and temporal variations in soil moisture are critical to community organization on a sandy substrate. An experimental transect encompassing the major community and soil types along a steep, west-facing vegetated dune was established. Maximum available water was shown to be significantly higher in the fine textured surface soils of the lowland sites than the coarse textured sands of the dune sites. Seasonal (1979) patterns of available soil moisture of the sampling sites on the transect showed that in the upper elevation dune sands, moisture was available in the entire profile with surface depletions not occurring until mid to late summer. In contrast, moisture in the surface 60-80 cm in the fine textured lowland soils was exhausted by early to mid-summer with the entire profile nearly dry by late summer. Deep-rooted, C 4 species, Andropogon hallii and Calamovilfa longifolia which are common on upper, coarser sandy soils showed significantly greater water stress on fine textured soils than on dune sands. C 3 , shallowrooted species, Agropyron smithii, Stipa comata, and Koeleria cristata always experienced lower mid-day and predawn leaf water potentials than the C 4 species. The C 3 species, with the exception of Koeleria are most abundant on finer textured soils that provide substantial moisture during their peak activity in the spring. It appears that the C 4 species show more conservative water use patterns than the C 3 species as significantly lower leaf conductances in the C 4 's were measured when soil water was abundant. The C 3 species appear to be opportunistic with available water and rapidly deplete surface soil moisture as a result of high transpiration rates. These data suggest that the temporal and spatial distribution of available water along this gradient controls species distribution according to rooting morphology, photosynthetic physiology, and water deficits, incurred by transpirational losses. Competitive interactions between species that utilize soil moisture differently may be an important factor in community organization.
