Due to the hydrophilicity and black-phase instability of FA perovskites, ambient humidity is an unavoidable issue in the processing of perovskite solar cells (PSCs). MACl is among the most popular additives for improving perovskite films, but our experiments confirm that the direct addition of MACl into the precursor solution deteriorates the stability of the final α-FAPbI 3 films in humid air, which is attributed to the unwanted pinholes induced by MACl volatilization. To solve this problem, a novel confined-space annealing strategy (CSA) is intentionally developed to control the amount of MACl at a low level. Through retarding the volatilization of MACl and blocking moisture ingress, dense and δ-phase-free FAPbI 3 films with excellent crystallinity and stability are achieved at 100 °C under high humidity (RH: 60 ± 10%). We further compare the same amounts of MAI and FACl additives with MACl, discovering that only when MA and Cl work together can pure α-FAPbI 3 films be obtained; therefore, a mechanism of MA-assisted nucleation and Cl-induced diffusion recrystallization is inferred. As a result, the PSCs employing optimal films yield a champion power conversion efficiency (PCE) of 17.27% and retain over 90% of the initial PCE after exposure to high humidity for 480 h. Our results offer deep insights into the thermodynamic and kinetic behaviors of MA and Cl in film growth and are beneficial for air-processed FA-based PSCs for commercial application.

Non-flammable and oxidatively stable sulfones are promising electrolyte solvents for thermally stable high-voltage Li batteries. In addition, sulfolane-based high-concentration electrolytes (HCEs) show high Li + ion transference numbers. However, LiPF 6 has not yet been investigated as the main salt in sulfone-based HCEs for Li batteries. In this study, we investigated the phase behaviors, solvate structures, and transport properties of binary and ternary mixtures of LiPF 6 and the following sulfone solvents: sulfolane (SL), dimethyl sulfone (DMS), ethyl methyl sulfone (EMS), and 3-methyl sulfolane (MSL). The stable crystalline solvates Li(SL) 4 PF 6 and Li(DMS) 2.5 PF 6 with high melting points were formed in the LiPF 6 /SL and LiPF 6 /DMS mixtures, respectively. In contrast, LiPF 6 /EMS, LiPF 6 /MSL, and LiPF 6 /SL/another sulfone mixtures remained liquids over a wide temperature range. Raman spectroscopy revealed that SL and another sulfone are competitively coordinated to Li + ions to dissociate LiPF 6 in the ternary mixtures. Although the ionic conductivity decreased with increasing LiPF 6 concentration due to an increase in viscosity, Li + ions diffused faster than PF 6 - via exchanging ligands in the HCE [LiPF 6 ]/[SL]/[DMS] = 1/2/2, resulting in a higher Li ion transference number than that in conventional Li battery electrolytes.

Two polypyridyl ruthenium(II) complexes [Ru(DIP) 2 (BIP)](PF 6 ) 2 (DIP = 4,7-diphenyl-1,10-phenanthrolie, BIP = 2-(1,1'-biphenyl-4-yl)-1H-imidazo[4,5-f][1,10]phenanthroline, Ru1) and [Ru(DIP) 2 (CBIP)](PF 6 ) 2 (CBIP = 2-(4'-chloro-1,1'-biphenyl-4-yl)-1H-imidazo[4,5-f][1,10]phenanthroline, Ru2) were synthesized. The cytotoxic activities in vitro of Ru1, Ru2 toward B16, A549, HepG2, SGC-7901, HeLa, BEL-7402, non-cancer LO2 were investigated using MTT method (3-(4,5-dimethylthiazole)-2,5-diphenltetraazolium bromide). Unexpectedly, Ru1, Ru2 can't prevent these cancer cells proliferation. To improve the anti-cancer effect, we used liposomes to entrap the complexes Ru1, Ru2 to form Ru1lipo, Ru2lipo. As expectation, Ru1lipo and Ru2lipo exhibit high anti-cancer efficacy, especially, Ru1lipo (IC 50 3.4 ± 0.1 μM), Ru2lipo (IC 50 3.5 ± 0.1 μM) display strong ability to block the cell proliferation in SGC-7901. The cell colony, wound healing, and cell cycle distribution show that the complexes can validly inhibit the cell growth at G2/M phase. Apoptotic studied with Annex V/PI doubling method showed that Ru1lipo and Ru2lipo can effectively induce apoptosis. Reactive oxygen species (ROS), malondialdehyde, glutathione and GPX4 demonstrate that Ru1lipo and Ru2lipo improve ROS and malondialdehyde levels, inhibit generation of glutathione, and finally result in a ferroptosis. Ru1lipo and Ru2lipo interact on the lysosomes and mitochondria and damage mitochondrial dysfunction. Additionally, Ru1lipo and Ru2lipo increase intracellular Ca 2+ concentration and induce autophagy. The RNA-sequence and molecular docking were performed, the expression of Bcl-2 family was investigated by Western blot analysis. Antitumor in vivo experiments confirm that 1.23 mg/kg, 2.46 mg/kg of Ru1lipo possesses a high inhibitory rate of 53.53% and 72.90% to prevent tumor growth, hematoxylin-eosin (H&E) results show that Ru1lipo doesn't cause chronic organ damage and strongly promotes the necrosis of solid tumor. Accumulation of vimentin is the core event in epithelial-mesenchymal transition (EMT). Post-translational modifications have been widely reported to play crucial roles in imparting different properties and functions to vimentin. Here, a novel modification of vimentin, acetylated at Lys 104 (vimentin-K104Ac) is identified, which is stable in lung adenocarcinoma (LUAD) cells. Mechanistically, NACHT, LRR, and PYD domain-containing protein 11 (NLRP11), a regulator of the inflammatory response, bind to vimentin and promote vimentin-K104Ac expression, which is highly expressed in the early stages of LUAD and frequently appears in vimentin-positive LUAD tissues. In addition, it is observed that an acetyltransferase, lysine acetyltransferase 7 (KAT7), which binds to NLRP11 and vimentin, directly mediates the acetylation of vimentin at Lys 104 and that the cytoplasmic localization of KAT7 can be induced by NLRP11. Malignant promotion mediated by transfection with vimentin-K104Q is noticeably greater than that mediated by transfection with vimentin-WT. Further, suppressing the effects of NLRP11 and KAT7 on vimentin noticeably inhibited the malignant behavior of vimentin-positive LUAD in vivo and in vitro. In summary, these findings have established a relationship between inflammation and EMT, which is reflected via KAT7-mediated acetylation of vimentin at Lys 104 dependent on NLRP11., (© 2023 The Authors. Background: This study aimed to investigate the underlying molecular mechanisms of TRIM58 in the development of colorectal cancer (CRC). CRC is one of the most common cancers of the digestive tract worldwide. The ubiquitin-proteasome system regulates many oncogenic or tumor-suppressive proteins. TRIM58, an E3 ubiquitin ligase and a member of the tripartite motif protein family, is a potential prognostic marker that indicates poor prognosis in cancer. Currently, the precise molecular mechanisms for the TRIM58-mediated CRC progression remain unclear., Methods: To examine the effects of TRIM58 on cell viability, cell cycle progression, and apoptosis in CRC, Cell Counting Kit-8 and flow cytometry assays were employed. The AKT inhibitor LY294002 was used to examine the effects of AKT signaling on TRIM58-mediated cell viability, cell cycle progression, and apoptosis in CRC. Additionally, Co-IP and ubiquitination assays were used to examine the correlation between TRIM58 and RECQL4., Results: TRIM58 overexpression inhibited CRC cell viability and promoted cell cycle arrest and apoptosis, in which the TRIM58 knockdown demonstrated inversed effects via the AKT signaling pathway. TRIM58 inhibited RECQL4 protein levels through its ubiquitin ligase activity, and RECQL4 overexpression inhibited TRIM58 overexpression-mediated CRC cell viability, cell cycle progression, and apoptosis. The downregulation of TRIM58 and upregulation of RECOL4 were observed in human CRC tissue, and TRIM58 demonstrated antitumor effects in CRC-induced tumor growth in a mouse model., Conclusions: TRIM58 acts as a tumor suppressor in CRC through the promotion of RECQL4 ubiquitination and inhibition of the AKT signaling pathway and may be investigated for the successful treatment of CRC., (© 2023. Plexcitonics is a rapidly developing interdisciplinary field that holds immense potential for the creation of innovative optical technologies and devices. This field focuses on investigating the interactions between plasmons and excitons in hybrid systems. In this review, we provide an overview of the fundamental principles of plasmonics and plexcitonics and discuss the latest advancements in plexcitonics. Specifically, we highlight the ability to manipulate plasmon-exciton interactions, the emerging field of tip-enhanced spectroscopy, and advancements in optical chirality and nonlinearity. These recent developments have spurred further research in the field of plexcitonics and offer inspiration for the design of advanced materials and devices with enhanced optical properties and functionalities.

In this article, four new Ru(II) complexes [Ru(dmbpy) 2 (TFBIP)](PF 6 ) 2 (dmbpy = 4,4'-dimethyl-2,2'-bipyridine, TFPIP = 2-(4'-trifluoromethyl)-[1,1'-biphenyl]-4-yl)-1H-imidazo[4,5-f][1,10]phenanthroline) (Ru1), [Ru(bpy) 2 (TFBIP)](PF 6 ) 2 (bpy = 2,2'-bipyridine) (Ru2), [Ru(phen) 2 (TFBIP)](PF 6 ) 2 (phen = 1,10-phenanthroline) (Ru3) and [Ru(dmp) 2 (TFBIP)](PF 6 ) 2 (dmp = 2,9-dimethyl-1,10-phenanthroline) (Ru4) were synthesized and characterized by elemental analysis, HRMS, IR, 1 H NMR, 13 C NMR and 19 F NMR. The in vitro anticancer effect of the complexes on HepG2, A549, B16, HeLa, BEL-7402 and non-cancer LO2 cells was screened using 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) method. The results illustrate that the complexes display moderate anticancer activity. Apoptotic assay with Annexin V/PI double staining method indicated that complexes induce apoptosis in HepG2 cells. Also, the complexes interfere with the mitochondrial functions, accompanied by the production of intracellular ROS as well as a reduction of mitochondrial membrane potential. The results obtained from the western blot demonstrated that the complexes upregulate pro-apoptotic Bax and downregulate anti-apoptotic Bcl-2, which further activates caspase 3 and promotes the cleavage of PARP. RNA-sequence showed that the complexes upregulate the expression of 40 genes and downregulate 66 genes. Antitumour in vivo demonstrated that Ru1 inhibits the tumor growth with a high inhibitory rate of 51.19%. Nuclease, such as RNase H and DNase I, plays key roles in plenty of cellular processes and could be potential therapeutic target for drug development. It is necessary to establish rapid and simple-to-use methods to detect nuclease activity. Herein, we develop a Cas12a-based fluorescence assay without any nucleic acid amplification steps for ultrasensitive detection of RNase H or DNase I activity. By our design, the pre-assembled crRNA/ssDNA duplex triggered the cleavage of fluorescent probes in the presence of Cas12a enzymes. However, the crRNA/ssDNA duplex was selectively digested with the addition of RNase H or DNase I, which leaded to fluorescence intensity changes. Under optimized conditions, the method exhibited good analytical performance, achieving a limit of detection (LOD) as low as 0.0082 U/mL for RNase H and 0.13 U/mL for DNase I, respectively. The method was feasible for analysis of RNase H in human serum and cell lysates, as well as for screening of enzyme inhibitors. Moreover, it can be adopted to image RNase H activity in living cells. In this study, two new iridium(III) polypyridyl complexes [Ir(bzq) 2 (DIPH)](PF 6 ) (bzq = deprotonated benzo[h]quinoline, DIPH = 4-(2,5-dibromo-4-(1H-imidazo[4,5-f][1,10]phenanthrolim-2-yl)-4-hydroxybutan-2-one) (Ir1) and [Ir(piq) 2 (DIPH)](PF 6 ) (piq = deprotonated 1-phenylisoquinoline) (Ir2) were synthesized and characterized by elemental analysis, HRMS, 1 H and 13 C NMR. The cytotoxic activity of Ir1, Ir2, Ir1lipo and Ir2lipo against cancer cells SGC-7901, HepG2, A549, HeLa, B16 and normal NIH3T3 cells in vitro was evaluated using 3-(4,5-dimethylthiazole-2-yl)-2,5-biphenyl tetrazolium bromide (MTT) method. Ir1 and Ir2 showed no cytotoxic activity, but their liposome-entrapped Ir1 (Ir1lipo) and Ir2 (Ir2lipo) showed significant cellular activity, especially sensitive to SGC-7901 with IC 50 values of 4.7 ± 0.2 and 12.4 ± 0.5 μM, respectively. The cellular uptake, endoplasmic reticulum (ER) localization, autophagy, tubulin polymerization, glutathione (GSH), malondialdehyde (MDA) and release of cytochrome c were investigated to explore the mechanisms of apoptosis. The calreticulin (CRT), heat shock protein 70 (HSP70), high mobility group box 1 (HMGB1) were also explored. Western blotting showed that Ir1lipo and Ir2lipo inhibited PI3K (phosphoinositide-3 kinase), AKT (protein kinase B), p-AKT and activated Bcl-2 (B-cell lymphoma-2) protein and apoptosis-regulated factor caspase 3 (cysteinyl aspartate specific proteinase-3) and cleaving PARP (poly ADP-ribose polymerase). Iridium(III) complexes are largely studied as anti-cancer complexes due to their excellent anti-cancer activity. In this article, two new iridium(III) complexes [Ir(piq) 2 (THPIP)]PF 6 (THPIP = 2,4-di-tert-butyl-6-(1H-imidazo[4,5-f][1,10]phenanthrolin-2-yl)phenol, piq = deprotonated 1-phenylisoquinoline) (Ir1) and [Ir(bzq) 2 (THPIP)]PF 6 (bzq = deprotonated benzo[h]quinolone) (Ir2) were synthesized. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays showed that complex Ir1 exhibits moderate activity (IC 50  = 29.9 ± 4.6 μM) and Ir2 shows high cytotoxicity (IC 50  = 9.8 ± 1.8 μM) against BEL-7402 cells. Further studies on the mechanism showed that Ir1 and Ir2 induced apoptosis by changing the mitochondrial membrane potential, Ca 2+ release, ROS accumulation, and cell cycle arrest at the S phase. The complexes can effectively inhibit cell colony formation and migration. The expression of B-cell lymphoma-2 (Bcl-2) family proteins, PI3K (phosphatidylinositol 3-kinase), AKT (protein kinase B), mTOR (mammalian target of rapamycin), and p-mTOR was studied by immunoblotting. Complexes Ir1 and Ir2 downregulated the expression of anti-apoptotic protein Bcl-2 and increased the expression of autophagy-related proteins of Beclin-1 and LC3-II. Further experiments showed that the complexes inhibited the production of glutathione (GSH) and increased the amounts of malondialdehyde (MDA). Fluorescence of HMGB1 was significantly increased. We also investigated the effect of the complexes on the expression of genes using RNA-sequence analysis, we further calculated the lowest binding energies between the complexes and proteins using molecular docking. Organic solar cells (OSC) based on organic semiconductor materials that convert solar energy into electric energy have been constantly developing at present, and also an effective way to solve the energy crisis and reduce carbon emissions. In the past several decades, efforts have been made to improve the power conversion efficiency (PCE) of OSCs. During this period, a variety of structural and material forms of OSCs have evolved. Commercializing OSCs, extending their service life and exploring their future development are promising but challenging. In this review, we first briefly introduce the development of OSCs and then summarize and analyze the working principle, performance parameters, and structural features of OSCs. The development of industry has triggered an increasingly severe demand for new functional materials. In recent years, researches on high-entropy oxides (HEOs) are more comprehensive and in-depth, and their fascinating properties are gradually known to the public. The unique elemental synergistic effect and lattice distortion endow the high-entropy family with various untapped potential, and wide application fields and outstanding performance of HEOs make them candidates for future materials. In this review, the concept, structure, and synthesis of HEOs are firstly highlighted. Secondly, a variety of excellent properties and applications in the fields of mechanics, electrics, thermotics, optics and magnetics are summarized. Atomic chains and organic conjugated molecules are of great important research value in molecular optoelectronics, due to their special optoelectronic properties. The fully conjugated nature of ladder phenylenes (LPs) provide some unique properties that have potential applications in the fabrication of molecular electronics devices. Our results reveal optoelectronic properties apply density function theory and non-equilibrium green's function theory, including unit-dependent light absorption, Raman scattering, phonon energy band structure, the chemical potential dependent density of states, electrical conductivity, I-V curve, transmission spectrum, etc. Our research provides theoretical guidance for the regulation of light-harvesting regions based on LPs structures, and theoretical support for the design of nano-scale optoelectronic devices., (Copyright © 2022 Elsevier B.V. The nonlinear optical properties of a D-A (donor-acceptor) conjugated organic molecule with polythiophene (PT) as the donor and indene-C 60 bisadduct (IC 60 BA) as the acceptor are theoretically investigated, which exhibits a large two-photon absorption (TPA) cross-section up to 8000 GM at the wavelength of 780 nm. Combining surface plasmon resonances (SPRs) with nonlinear optics, nonlinear properties can be strongly enhanced. In this paper, an appropriate nonlinear plexciton method by the coupling of Au@Ag nanorods and an Ag film is designed, in which the TPA properties of the PT:IC 60 BA complex can be increased by 10 6 times. The angle dependence on polarization and incidence is investigated to obtain the maximum of plasmonic enhancement. Our results emphasize the physical mechanism of nonlinear plexcitons and provide a feasible method to improve the nonlinear properties of organic solar cell materials.

Background: Total arch replacement (TAR) with Frozen elephant trunk (FET) treatment of acute DeBakey type I aortic dissection (ADIAD) is complicated, carries a high complication/mortality risk and remains controversial on the optimal hypothermic level, cerebral perfusion and visceral organ protection strategy. We developed a new strategy named "Brain-Heart-first" in which the surgical procedures and the management of cardiac perfusion/cerebral protection during Cardiopulmonary bypass (CPB) were redesigned, and TAR with FET technique can be performed under mild hypothermia with satisfactory outcomes., Objective: Our aims were to describe a new surgical strategy under mild hypothermia (≥30°C) for the treatment of ADIAD and to report the operative outcomes of 215 patients., Methods: We conducted a retrospective analysis of 215 consecutive cases of ADIAD treated with our new strategy., Results: The durations of CPB, aortic cross-clamping, antegrade cerebral perfusion, operation, mechanical ventilation support, and Intensive Care Unit stay were 139.7 ± 52.3 min, 55.6 ± 27.4 min, 14.1 ± 3.1 min, 6.0 ± 1.7 h, 40.0 h and 4.0 d, respectively. The 30-day mortality was 9.8%, with cerebral stroke occurring in nine patients (4.2%), paraplegia in one patient (0.5%) and postoperative renal injury requiring dialysis in 21 patients (9.8%). When the power conversion efficiency (PCE) of perovskite solar cells (PSCs) rapidly approaches that of commercial solar cells, the stability becomes the most important obstacle for the commercialization of PSCs. Aside from the widely studied slow PCE degradation, the PSCs also show a unique rapid PCE degradation. Although the degradation due to oxygen and humidity can be avoided by encapsulation, that due to bias voltage, light and heat could not be effective suppressed and will lead to considerable degradation. Usually, the rapid PCE degradation is believed to be from ion migration. However, a systematic investigation is yet to be carried out. This work quantitatively and systematically investigated the relationships between external fields (bias voltage, light or heat), ion migration and device performance. By comparing the performance of reference PSCs after 90 min degradation under these fields, we conclude that (1) the electric field affects the spatial distribution of mobile ions; (2) the light field changes the mobile ion densities and drives the ion migration; (3) the heat field results in perovskite decomposition as well as changing the mobile ion densities. Objectives: Long non-coding RNA (LncRNA) is an important transcriptional and post-transcriptional regulatory molecule in the body. In recent years, relationship between LncRNA and malignant phenotype of tumor cells has been revealed gradually. This study aims to investigate the expression characteristics of pit-oct-unc class 3 homeobox 3 related long non-coding RNA (Linc-POU3F3) in esophageal cancer and its relationship with radiation resistance (IR) as well as the expressions of cancer stem cell (CSC) markers in esophageal cancer cells., Methods: The expression characteristics and potential interaction molecules of Linc-POU3F3 in esophageal cancer were collected from the public database via bioinformatics retrieval. Forty-two pair samples of esophageal cancer tissues and corresponding adjacent tissues were collected. Human normal esophageal epithelial cells (HEEC) and human esophageal cancer cell lines (ECA109, TE-1, TE-2, TE-13) were cultured. Real-time quantitative PCR (qPCR) was used to detect the expression level of Linc-POU3F3 in clinical tissues and cells. The formation of TE-13 IR cell line induced by different doses of radiation served as IR group cells, and the same condition treated with 0 Gy dose was set as control group (control) cells. Meanwhile, we used cell transfection technology to construct random interference sequence (siControl) cells and interference (siLinc-POU3F3) cells. In ECA109 cells, we transfected blank and over expressed Linc-POU3F3 plasmids as vector and over-expressed group (oeLinc-POU3F3). The mRNA and protein expressions of CD44, CD133 and CD90 were detected by qPCR and Western blotting, respectively. MTS [3-(4,5-dimenthylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt] was used to detect the cell viability under different radiation doses, and the resistance of IR cells was verified by clone formation experiment., Results: The expression of Linc-POU3F3 was correlated with the tumor progression and poor prognosis of esophageal cancer. The level of Linc-POU3F3 mRNA expression was significantly higher in esophageal cancer tissues and cell lines than that in normal adjacent tissues and cell lines (all P <0.01). The expressions of Linc-POU3F3 mRNA and protein expressions of CD44, CD133, and CD90 in IR cells were significantly higher than those in control cells (all P <0.01). The expression of Linc-POU3F3 in siLinc-POU3F3 cell was significantly lower than that in the siControl cells ( P <0.01), and the inhibition rate was 87.21%. The mRNA and protein expressions of CD44, CD133, and CD90 in the siLinc-POU3F3 cells were significantly lower than those in the siControl cells (all P <0.05). The expressions of linc-POU3F3, CD44, CD133, and CD90 mRNA and protein in the oeLinc-POU3F3 cells were significantly higher than those in the vector cells. The relative activity and clone formation ability in the IR cells were significantly higher than those in the control cells at 2, 4, and 8 Gy doses (all P <0.01). The relative activity in the siLinc-POU3F3 cells was significantly lower than that in the siControl cells at 4 and 8 Gy doses ( P <0.01). The relative activity in the oeLinc-POU3F3 cells was significantly higher than that in the vector cells at 4 and 8 Gy doses ( P <0.01)., Conclusions: Linc-POU3F3 is up-regulated in esophageal cancer and can promote IR and the expression of CSC markers in esophageal cancer cells.

The successful fabrication of WS 2 /MoS 2 heterostructures provides more possibilities for optoelectronic and thermoelectric applications than graphene because of their direct bandgap characteristics; therefore, scientific investigations on WS 2 /MoS 2 heterostructures are more significant and thriving. In this paper, we review the latest research progress in WS 2 /MoS 2 heterostructures, and look forward to their properties and applications. Firstly, we analyze the crystal structure and electronic structure of WS 2 , MoS 2 , and their heterostructures. Secondly, we comprehensively present the widely used methods for preparing heterostructures. Finally, based on the unique physical characteristics of WS 2 /MoS 2 heterostructures, we focus on their properties and applications in mechanics, electronics, optoelectronics, and thermoelectronics.

Organic-inorganic perovskite single crystals are promising in the field of optoelectronics due to their excellent optoelectronic properties. However, the ion transport of perovskite precursor is poor in confined spaces, which results in difficulty in the preparation of perovskite single-crystal films. Herein, MAPbBr 3 films consisting of square grains were fabricated by the surface-confined process using the organic molecule PEAI (phenethylammonium iodide). Under the effect of oversaturation gradient, PEA + is combined with the surface of perovskite grain from top to side, which constrains the lateral growth of grains and induces a downward growth of perovskite, leading to the formation of square grains. With the improvement of concentration PEAI, the perovskite film exhibits a decreased side length of grains (from 0.98 to 12.96 μm) and increased grain number and coverage, as well as crystallinity. The perovskite single crystalline grain films with PEAI showed double photoluminescence (PL) emission peaks due to the existence of iodine-rich perovskite. This work may provide a practical way to fabricate high-quality perovskite films for perovskite photoelectronic devices.

Objectives: To explore the relationship between long non-coding RNA (lncRNA) long stress-induced noncoding transcript 5 (LSINCT5) and erotinib resistance to lung cancer cells and the potential mechanisms., Methods: Human lung cancer cell line A549, H520, H358, H1299, SPCA1, and PC9 were collected and cultured. Epidermal growth factor receptor (EGFR) mutant lung cancer cell line PC9 was divided into a control group, a resistance group, a interference group I and II. The control group was treated with dimethylsulfoxide (DMSO) for 10 weeks and then was transfected with control target sequence expression vector. The resistant group was treated with erlotinib at gradient concentration (0.1, 0.2, 0.4, 0.8, and 1.6 μmol/L, respectively) for 2 weeks and then transfected with control target sequence expression vector. Interference group I and II were treated with erlotinib at gradient concentration (0.1, 0.2, 0.4, 0.8, and 1.6 μmol/L, respectively) for 2 weeks and then transfected with the shRNA targeting expression vectors 1 and 2. 50% inhibitory concentration (IC 50 ) of erlotinib was detected by cell counting kit-8 (CCK-8) assay. The mRNA expressions of LSINCT5, phosphatidylinositol 3-kinase (PI3K) and protein kinase B (Akt) were measured by real-time PCR. The protein levels of PI3K, Akt, and phospho-Akt (p-Akt) were detected by Western blotting. The divergences of Akt and IgG binding to LSINCT5 were detected by RNA immunoprecipitation (RNA-IP) experiment., Results: The expression of LSINCT5 in PC9 cells was significantly higher than that in other lung cancer cell lines (all P <0.05). Compared with the control group, the IC 50 of erotinib and the expression of LSINCT5, PI3K, and Akt mRNA and protein in the resistance group were significantly higher (all P <0.05), and the IC 50 of erotinib and the expression of LSINCT5, Akt, and p-Akt in the interference group I and II were significantly lower (all P <0.05). Compared with IgG, LSINCT5 binding to Akt was increased significantly ( P <0.05)., Conclusions: The expression of LSINCT5 is high in the erlotinib-resistant cells. Interference with LSINCT5 may inhibit the expression and activity of Akt and promote the cell sensitivity to erlotinib.

Background: Calpain 1 (CAPN1) has been found to be a promoter of cancer progression. PTPN1 as a physiological target molecule of CAPN1 plays a dephosphorylated role on multiple receptor tyrosine kinases. This study aimed to reveal the effects of CAPN1/PTPN1 on malignant phenotype and EGFR-TKI resistance of lung adenocarcinoma (LUAD) cells., Methods: A total of 84 primary LUAD tissues and paired paracancerous normal tissues were collected. Quantitative real-time PCR (qRT-PCR) and immunohistochemical (IHC) methods were used to measure the expression of CAPN1 and PTPN1 in tissues. qRT-PCR and western blot were used to detect the expressions of CAPN1, PTPN1, c-Met and PIK3R2 in cell lines. Cell counting kit-8 (CCK-8), colony formation and transwell assay were carried out to evaluate cell erlotinib resistance, proliferation, migration and invasion. Co-IP assay was used to verify the interaction between proteins. Cycloheximide (CHX) was applied to block protein synthesis., Results: CAPN1, c-Met and PIK3R2 were significantly upregulated and the correlation was positive in LUAD, while PTPN1 was decreased. EGFR-sensitive mutation was related to CAPN1/PTPN1. in vitro studies showed that PTPN1 can mediate dephosphorylation of c-Met and PIK3R2 by binding with both, thereby weakening cell proliferation, metastasis and erlotinib resistance, while CAPN1 could enhance the degradation of PTPN1 protein as a cancer promoter., Conclusions: CAPN1 enhances the malignant behavior and erlotinib resistance of LUAD cells via degrading PTPN1 and then activating c-Met/PIK3R2, which suggests CAPN1/PTPN1 may serve as tumor markers or potential targets for diagnosis and treatment of LUAD., Key Points: Significant findings of the study Superior CAPN1 and inferior PTPN1 were related to activation of c-Met/PIK3R2 in lung adenocarcinoma. Moreover, regulations of CAPN1 and PTPN1 induced the changes of malignant behavior and erlotinib resistance. Perovskite solar cells (PSCs) have gained tremendous research interest because of their tolerance of defects, low cost, and facile processing. In PSC devices, PbI 2 has been utilized to passivate defects at perovskite film surfaces and GBs; however, a systematic mechanism of PbI 2 in situ passivation for enhancing the solar cells efficiency has not been fully explored. Here, this work, we systematically studies the effect of the precise PbI 2 ratio and the PbI 2 in situ passivation mechanism based on trap density, carrier lifetime, Fermi level, and so forth. This study finds the appropriate ratio of I/Pb to be around 2.57:1 using energy-dispersive spectroscopy. After the moderate excess PbI 2 in situ passivation, the trap density is reduced from 6.12 × 10 16 to 3.38 × 10 16 cm -3 , and the carrier lifetime is extended from 168.35 to 368.77 ps by using fs-TA spectroscopy. This result indicates that the moderate excess PbI 2 in situ passivation can reduce the trap density and suppress the nonradiative recombination. The efficiency of solar cell has shown a nearly 11.3% improvement of 19.55% for an I/Pb ratio of 2.57:1 compared with 2.69:1. It also demonstrates that the efficiency of PSCs can be enhanced effectively by PbI 2 in situ passivation.

In recent years, perovskite solar cells (PSCs) have attracted great attention in the photovoltaic research field, because of their high-efficiency (certified 22.1%) and low-cost. In this review paper, we briefly introduce the history of efficiency development for PSCs, and discuss some of the major problems for large-area (≥1 cm 2 ) PSC devices. In addition, we summarize the recent progress in the aspects of fabrication methods for large-area perovskite films, and improving the efficiency and stability of the large-area PSC devices. Finally, we give a short summary and outlook of large-area PSC devices. This article is mainly organized into three parts. The first part focuses on the main fabricating technologies for large-area perovskite films. The second section discusses some methods that are used to improve the efficiency of PSCs.
