The monospecific family Mysteriomorphidae was recently described based on two fossil specimens from the Late Cretaceous Kachin amber of northern Myanmar. The family was placed in Elateriformia incertae sedis without a clear list of characters that define it either in Elateroidea or in Byrrhoidea. We report here four additional adult specimens of the same lineage, one of which was described using a successful reconstruction from a CT-scan analysis to better observe some characters. The new specimens enabled us to considerably improve the diagnosis of Mysteriomorphidae. The family is definitively placed in Elateroidea, and we hypothesize its close relationship with Elateridae. Similarly, there are other fossil families of beetles that are exclusively described from Cretaceous ambers. These lineages may have been evolutionarily replaced by the ecological revolution launched by angiosperms that introduced new co-associations with taxa. These data indicate a macroevolutionary pattern of replacement that could be extended to other insect groups.

New Zealand kauri is an ancient, iconic, gymnosperm tree species that is under threat from a lethal dieback disease caused by the oomycete Phytophthora agathidicida. To gain insight into this pathogen, we determined whether proteinaceous effectors of P. agathidicida interact with the immune system of a model angiosperm, Nicotiana, as previously shown for Phytophthora pathogens of angiosperms. From the P. agathidicida genome, we defined and analysed a set of RXLR effectors, a class of proteins that typically have important roles in suppressing or activating the plant immune system. RXLRs were screened for their ability to activate or suppress the Nicotiana plant immune system using Agrobacterium tumefaciens transient transformation assays. Nine P. agathidicida RXLRs triggered cell death or suppressed plant immunity in Nicotiana, of which three were expressed in kauri. For the most highly expressed, P. agathidicida (Pa) RXLR24, candidate cognate immune receptors associated with cell death were identified in Nicotiana benthamiana using RNA silencing-based approaches. Our results show that RXLRs of a pathogen of gymnosperms can interact with the immune system of an angiosperm species. Within modern gymnosperms, conifers and Ginkgo are exclusively wind pollinated whereas many gnetaleans and cycads are insect pollinated. For cycads, thrips are specialized pollinators. We report such a specialized pollination mode from Early Cretaceous amber of Spain, wherein four female thrips representing a genus and two species in the family Melanthripidae were covered by abundant Cycadopites pollen grains. These females bear unique ring setae interpreted as specialized structures for pollen grain collection, functionally equivalent to the hook-tipped sensilla and plumose setae on the bodies of bees. The most parsimonious explanation for this structure is parental food provisioning for larvae, indicating subsociality. This association provides direct evidence of specialized collection and transportation of pollen grains and likely gymnosperm pollination by 110-105 million years ago, possibly considerably earlier.

The majority of generalisations concerning plant responses to herbivory are based on studies of natural or simulated defoliation. However, effects caused by insects feeding on plant sap are likely to differ from the effects of folivory. We assessed the general patterns and sources of variation in the effects of sap feeding on growth, photosynthesis, and reproduction of woody plants through a meta-analysis of 272 effect sizes calculated from 52 papers. Sap-feeders significantly reduced growth (-29%), reproduction (-17%), and photosynthesis (-27%); seedlings suffered more than saplings and mature trees. Deciduous and evergreen woody plants did not differ in their abilities to tolerate damage imposed by sap-feeders. Different plant parts, in particular below- and above-ground organs, responded similarly to damage, indicating that sap-feeders did not change the resource allocation in plants. The strongest effects were caused by mesophyll and phloem feeders, and the weakest by xylem feeders. Generalist sap-feeders reduced plant performance to a greater extent than did specialists. Methodology substantially influenced the outcomes of the primary studies; experiments conducted in greenhouses yielded stronger negative effects than field experiments; shorter (<12 months) experiments showed bigger growth reduction in response to sap feeding than longer experiments; natural levels of herbivory caused weaker effects than infestation of experimental plants by sap-feeders. Studies conducted at higher temperatures yielded stronger detrimental effects of sap-feeders on their hosts. We conclude that sap-feeders impose a more severe overall negative impact on plant performance than do defoliators, mostly due to the lower abilities of woody plants to compensate for sap-feeders' damage in terms of both growth and photosynthesis.

In Oct. 2000 and Nov. 2003 samples for isolation of protostelids were collected from throughout Great Smoky Mountains National Park. Collecting sites included examples of all major forest types found in the park along with a number of non-forest vegetation types at 460-1920 m above sea level. Most samples consisted of aerial litter (dead but still attached plant parts) and ground litter, but some samples of the bark from living trees and coarse woody debris also were collected. Twenty-six of the 32 described species of protostelids were recovered, along with five species that appear to be new to science. Of the 205 cultures analyzed, 177 (86.5%) yielded protostelid fruiting bodies with an average of 3.22 species per sample. Protostelium mycophaga was the most frequently observed species followed by Soliformovum irregularis and Schizoplasmodiopsis pseudoendospora. Differences in protostelid community diversity and species richness were found between the aerial litter and ground litter microhabitats of primary plant tissues, between primary tissues and bark of living trees, between bark from angiosperms and bark from gymnosperms, and between primary tissues and coarse woody debris.

Integration of cultural practices, such as planting date with insecticide-based strategies, was investigated to determine best management strategy for flea beetles (Phyllotreta spp.) (Coleoptera: Chrysomelidae) in canola (Brassica napus L.). We studied the effect of two spring planting dates of B. napus and different insecticide-based management strategies on the feeding injury caused by fleabeetles in North Dakota during 2002-2003. Adult beetle peak emergence usually coincided with the emergence of the early planted canola, and this resulted in greater feeding injury in the early planted canola than later planted canola. Use of late-planted canola may have limited potential for cultural control of flea beetle, because late-planted canola is at risk for yield loss due to heat stress during flowering. Flea beetle injury ratings declined when 1) the high rate of insecticide seed treatment plus a foliar insecticide applied 21 d after planting was used, 2) the high rate of insecticide seed treatment only was used, or 3) two foliar insecticide sprays were applied. These insecticide strategies provided better protection than the low rates of insecticide seed treatments or a single foliar spray, especially in areas with moderate-to-high flea beetle populations. The foliar spray on top of the seed treatment controlled later-emerging flea beetles as the seed treatment residual was diminishing and the crop became vulnerable to feeding injury. The best insecticide strategy for management of flea beetle was the high rate of insecticide seed treatment plus a foliar insecticide applied at 21 d after planting, regardless of planting date.

Almost 2000 samples of the dried plants and animals used in Chinese traditional medicine were collected from a storehouse in Huainan city, Anhui province, China, where they had been kept for at least 6 months. When examined, every sample was found to contain acaroid mites. The mites included representatives of 44 species belonging to 22 genera and seven families. It seems that samples of Chinese traditional medicine frequently become severely infested with mites while in storage. More importance should be attached to protecting the medicines against mite infestation and to protecting against acariasis those who ingest or simply handle the medicines.

The pine processionary caterpillar Thaumetopoea pityocampa (Lepidoptera: Notodontidae) is considered to be a serious pest of medical importance. The hair on the dorsum of the last instar larvae of the moth may cause urticarial reactions (erucism) as well as eye problems and temporary blindness. In Israel, the pest occurs in all pine plantations as well as on ornamental pine trees in urban areas. The biology, ecology and management of the moth population are discussed as well as the mechanism of action of the urticarial hairs and their medical significance. Awareness of the life cycle and ecology of the pest may reduce the contact of the population with the urticarial hairs and prevent the morbidity caused by it.

The likelihood of sympatric speciation is enhanced when assortative mating is a by-product of adaptation to different habitats. Pleiotropy of this kind is recognized as important in parasites that use their hosts as a long-range cue for finding mates, but is generally assumed to have limited applicability for most other organisms. In the larch budmoth, Zeiraphera diniana (Lepidoptera: Tortricidae), sympatric host races feed on larch or pine. Zeiraphera diniana females attract males (call) by releasing host-independent long-range pheromones. Pheromone composition differs strongly between host races, but we show in an experimental field study that cross-attraction can occur at a rate of 0.03-0.38. Cross-attraction to larch females increases when they call from neighborhoods (8-m radius) rich in pine or from pine trees. Cross-attraction to pine females similarly increases when calling from neighborhoods rich in larch, but there is no significant effect of calling substrate. Males, as well as females, of this species preferentially alight on their own host, and in neighborhoods where their own host is common. This effect of tree species and host neighborhood on assortative mating is therefore due, at least in part, to the numbers of males of each host race present within approximately 200 m2 surrounding the female. This proximity effect is enhanced by the clumped distributions of the hosts themselves. Host chemistry might also affect pheromone production and/or response directly, but we have evidence neither for nor against this. This work provides empirical evidence that host adaptation has a pleiotropic effect on assortative mating in a species with host-independent long-range mating signals. Sympatric speciation via pleiotropy between ecological traits and assortative mating may thus be more common than generally supposed: Clumped resource distributions and habitat choice by adults are widespread.

Beetles in the weevil subfamilies Scolytinae and Platypodinae are unusual in that they burrow as adults inside trees for feeding and oviposition. Some of these beetles are known as ambrosia beetles for their obligate mutualisms with asexual fungi--known as ambrosia fungi--that are derived from plant pathogens in the ascomycete group known as the ophiostomatoid fungi. Other beetles in these subfamilies are known as bark beetles and are associated with free-living, pathogenic ophiostomatoid fungi that facilitate beetle attack of phloem of trees with resin defenses. Using DNA sequences from six genes, including both copies of the nuclear gene encoding enolase, we performed a molecular phylogenetic study of bark and ambrosia beetles across these two subfamilies to establish the rate and direction of changes in life histories and their consequences for diversification. The ambrosia beetle habits have evolved repeatedly and are unreversed. The subfamily Platypodinae is derived from within the Scolytinae, near the tribe Scolytini. Comparison of the molecular branch lengths of ambrosia beetles and ambrosia fungi reveals a strong correlation, which a fungal molecular clock suggests spans 60 to 21 million years. Bark beetles have shifted from ancestral association with conifers to angiosperms and back again several times. Each shift to angiosperms is associated with elevated diversity, whereas the reverse shifts to conifers are associated with lowered diversity. The unusual habit of adult burrowing likely facilitated the diversification of these beetle-fungus associations, enabling them to use the biomass-rich resource that trees represent and set the stage for at least one origin of eusociality.

During our studies on the components in Japanese cedar, Cryptomeria japonica, we found that the crude methanol extract of C. japonica showed intense antifeeding activity against one snail species, Acusta despesta, which is well-known as a pest of many vegetables and crops. The active components in the extract were separated into the hexane and ethyl acetate soluble fractions. From the active ethyl acetate soluble fraction, two norlignans, sequirin-C and agatharesinol, were isolated and identified as the active compounds. Both compounds inhibited feeding behavior of A. despesta at 30 microg/cm2 and 40 microg/cm2 concentrations, respectively, when applied by an eggplant leaf or filter paper containing 20 microl of 5% sucrose solution.

Coeloides bostrichorum Giraud parasitoids (Hymenoptera: Braconidae) attack late larval stages of various bark beetle species breeding in spruce. Volatile compounds collected from Norway spruce (Picea abies) infested by Ips typographus L. (Coleoptera: Scolytidae) were analysed by coupled gas chromatography-mass spectrometry (GC-MS) and GC-electroantennographic detection (GC-EAD). Monoterpene hydrocarbons are the predominant volatile compounds of fresh Norway spruce, while the presence of oxygenated monoterpenes indicates damaged trees. Between one and eight EAD-active oxygenated monoterpenes were used, in amounts reflecting their natural abundance in spruce trees containing bark beetle larvae, to prepare five synthetic baits which were tested in wind tunnel bioassays. Odour samples collected from spruce logs containing the preferred host stage were attractive, while similar samples from uninfested logs failed to elicit any flight activity. However, when a four- or an eight-component synthetic bait was added to volatiles collected from uninfested spruce logs, this combination was as attractive as volatiles collected from infested spruce logs.

Several shifts from ancestral conifer feeding to angiosperm feeding have been implicated in the unparalleled diversification of beetle species. The single largest angiosperm-feeding beetle clade occurs in the weevils, and comprises the family Curculionidae and relatives. Most authorities confidently place the bark beetles (Scolytidae) within this radiation of angiosperm feeders. However, some clues indicate that the association between conifers and some scolytids, particularly in the tribe Tomicini, is a very ancient one. For instance, several fragments of Gondwanaland (South America, New Caledonia, Australia and New Guinea) harbour endemic Tomicini specialized on members of the formerly widespread and abundant conifer family Araucariaceae. As a first step towards resolving this seeming paradox, we present a phylogenetic analysis of the beetle family Scolytidae with particularly intensive sampling of conifer-feeding Tomicini and allies. We sequenced and analysed elongation factor 1alpha and nuclear rDNAs 18S and 28S for 45 taxa, using members of the weevil family Cossoninae as an out-group. Our results indicate that conifer feeding is the ancestral host association of scolytids, and that the most basal lineages of scolytids feed on Aramucaria. If scolytids are indeed nested within a great angiosperm-feeding clade, as many authorities have held, then a reversion to conifer feeding in ancestral scolytids appears to have occurred in the Mesozoic, when Araucaria still formed a major component of the woody flora.

Nuclear ribosomal DNA (nrDNA) ITS sequences and partial sequences of three non-coding chloroplast DNA (cpDNA) introns and spacers were used to assess genetic variation within and among three presumed host races of the hemi-parasite Viscum album L. Currently, identification of host races occurs via the host trees, and morphological differences are minute at best. cpDNA and nrDNA ITS sequences revealed little sequence variation, but the variation found consistently supported the distinction of three host races. cpDNA and ITS sequences were not incongruent, as assessed by the incongruence length difference test. A combined analysis supported the sister group relationship between mistletoes from deciduous trees and fir.
