Early cancer diagnosis through characterizing light propagation and nanotechnology increases the survival rate. The present research is aimed at evaluating the consequence of using natural nanoparticles in cancer therapy and diagnosis. Colon cancer cells were differentiated from the normal cells via investigating light diffusion combined with the fluorescence effect of the Ashwagandha chitosan nanoparticles (Ash C NPs). Ionic gelation technique synthesized the Ash C NPs. High-resolution transmission electron microscope, dynamic light scattering, and zeta potential characterized Ash C NPs. Fourier transform infrared spectroscopy analyzed Ash C NPs, chitosan, and Ashwagandha root water extract. Moreover, the MTT assay evaluated the cytotoxicity of Ash C NPs under the action of near-infrared light (NIR) irradiation. The MTT assay outcomes were statistically analyzed by Bonferroni post hoc multiple two-group comparisons using one-way variance analysis (ANOVA). Based on the Monte-Carlo simulation technique, the spatially resolved steady-state diffusely reflected light from the cancerous and healthy cells is acquired. The diffuse equation reconstructed the optical fluence rate using the finite element technique. The fluorescent effect of the nanoparticles was observed when the cells were irradiated with NIR. The MTT assay revealed a decrease in the cell viability under the action of Ash C NPs with and without laser irradiation. Colon cancer and normal cells were differentiated based on the optical characterization after laser irradiation. The light diffusion equation was successfully resolved for the fluence rate on cells' surfaces showing different normal and cancer cells values. Nanomedicine and fluorescent optical imaging are effective in early cancer detection. The current study synthesized biocompatible nanocomposites from natural biomaterials towards inexpensive and safe cancer theragnostic. Two forms of nanocomposites were synthesized using the ionic gelation method: 1. Chitosan/ Withania Somnifera /tripolyphosphate nanocomposites, 2. Withania Somnifera/Chitosan nanocomposites. The nanocomposites were characterized by dynamic light scattering, zeta potential, and the transmission electron microscope. Fourier transform infrared spectroscopy analyzed the Withania Somnifera root water extract, Chitosan, and the synthesized nanocomposites. The cytotoxicity of the nanocomposites was investigated against the colon cancer cells (Caco2 cells) in the absence and the presence of laser (665 nm, 5 mW) irradiation. MTT assay evaluated the cytotoxicity, and Trypan blue assay assessed the cell viability. Cancerous cells were photographed under the inverted microscope in the presence and the absence of laser irradiation. Results were analyzed statistically using one-way variance (ANOVA) analysis with Bonferroni post-Hoc multiple two-group comparisons. The characterization results ensured the successful synthesis of Withania Somnifera/Chitosan nanocomposites. The results showed an increase in the cytotoxicity against colon carcinoma and a decrease in cell viability in the presence and absence of Near-infrared laser irradiation under the action of nanocomposites. The cytotoxicity of the synthesized nanocomposites increased by exposing the cells to the laser. The shining light of the nanocomposites appeared on the cells photographed under the inverted microscope. The synthesized natural nanocomposites promise systemic cytotoxicity will be efficient in molecular imaging in vivo applications., (© 2022. We propose a new optical method based on comparative holographic projection for visual comparison between two abnormal follow-up magnetic resonance (MR) exams of glioblastoma patients to effectively visualize and assess tumor progression. First, the brain tissue and tumor areas are segmented from the MR exams using the fast marching method (FMM). The FMM approach is implemented on a computed pixel weight matrix based on an automated selection of a set of initialized target points. Thereafter, the associated phase holograms are calculated for the segmented structures based on an adaptive iterative Fourier transform algorithm (AIFTA). Within this approach, a spatial multiplexing is applied to reduce the speckle noise. Furthermore, hologram modulation is performed to represent two different reconstruction schemes. In both schemes, all calculated holograms are superimposed into a single two-dimensional (2D) hologram which is then displayed on a reflective phase-only spatial light modulator (SLM) for optical reconstruction. The optical reconstruction of the first scheme displays a 3D map of the tumor allowing to visualize the volume of the tumor after treatment and at the progression. Whereas, the second scheme displays the follow-up exams in a side-by-side mode highlighting tumor areas, so the assessment of each case can be fast achieved. The proposed system can be used as a valuable tool for interpretation and assessment of the tumor progression with respect to the treatment method providing an improvement in diagnosis and treatment planning., Competing Interests: The authors have declared that no competing interests exist.

Chitosan-tripolyphosphate nanoparticles (C-TPP NPs) were synthesized to investigate their cytotoxicity against colon cancer cells (Caco2 cells) in the absence and the presence of a near-infrared (NIR) laser to evaluate their influence in cancer detection using the NIR laser and to evaluate the NIR laser on cancer treatment. The synthesized NPs were characterized by Fourier transform infrared (FT-IR) spectroscopy, dynamic light scattering (DLS), zeta potential (ZP), and transmission electronic microscope (TEM). The cytotoxicity was analyzed by the MTT test and the cell viability was assessed using the Trypan blue method. C-TPP NPs showed increased cytotoxicity and decreased cell viability against Caco2 cells. Upon laser exposure only, the cell viability decreased. The C-TPP NPs appeared to have a shining light on the cancerous cells which were photographed under the inverted microscope.

Optical differentiation is a promising tool in biomedical diagnosis mainly because of its safety. The optical parameters' values of biological tissues differ according to the histopathology of the tissue and hence could be used for differentiation. The optical fluence rate distribution on tissue boundaries depends on the optical parameters. So, providing image displays of such distributions can provide a visual means of biomedical diagnosis. In this work, an experimental setup was implemented to measure the spatially-resolved steady state diffuse reflectance and transmittance of native and coagulated chicken liver and native and boiled breast chicken skin at 635 and 808 nm wavelengths laser irradiation. With the measured values, the optical parameters of the samples were calculated in vitro using a combination of modified Kubelka-Munk model and Bouguer-Beer-Lambert law. The estimated optical parameters values were substituted in the diffusion equation to simulate the fluence rate at the tissue surface using the finite element method. Results were verified with Monte-Carlo simulation. The results obtained showed that the diffuse reflectance curves and fluence rate distribution images can provide discrimination tools between different tissue types and hence can be used for biomedical diagnosis., Competing Interests: The authors declare no conflict of interest.

The application of He-Ne laser technologies for description of articular cartilage degeneration, one of the most common diseases worldwide, is an innovative usage of these technologies used primarily in material engineering. Plain radiography and magnetic resonance imaging are insufficient to allow the early assessment of the disease. As surface roughness of articular cartilage is an important indicator of articular cartilage degeneration progress, a safe and noncontact technique based on laser speckle image to estimate the surface roughness is provided. This speckle image from the articular cartilage surface, when illuminated by laser beam, gives very important information about the physical properties of the surface. An experimental setup using a low power He-Ne laser and a high-resolution digital camera was implemented to obtain speckle images of ten bovine articular cartilage specimens prepared for different average roughness values. Texture analysis method based on gray-level co-occurrence matrix (GLCM) analyzed on the captured speckle images is used to characterize the surface roughness of the specimens depending on the computation of Haralick's texture features. In conclusion, this promising method can accurately estimate the surface roughness of articular cartilage even for early signs of degeneration. The method is effective for estimation of average surface roughness values ranging from 0.09 µm to 2.51 µm with an accuracy of 0.03 µm., Competing Interests: The authors declare no conflict of interest.
