Lichens are symbiotic associations consisting of a photobiont (algae or cyanobacteria) and a mycobiont (fungus). They are known to produce a variety of unique secondary metabolites. To access this biosynthetic potential for biotechnological applications, deeper insights into the biosynthetic pathways and corresponding gene clusters are necessary. Here we provide a comprehensive view of the biosynthetic gene clusters of all organisms comprising a lichen thallus: fungi, green algae, and bacteria. We present two high-quality PacBio metagenomes, in which we identified a total of 460 biosynthetic gene clusters. Lichen mycobionts yielded 73-114 clusters, other lichen associated ascomycetes 8-40, green algae of the genus Trebouxia 14-19, and lichen-associated bacteria 101-105 clusters. The mycobionts contained mainly T1PKSs, followed by NRPSs, and terpenes; Trebouxia reads harbored mainly clusters linked to terpenes, followed by NRPSs and T3PKSs. Other lichen-associated ascomycetes and bacteria contained a mix of diverse biosynthetic gene clusters. In this study, we identified for the first time the biosynthetic gene clusters of entire lichen holobionts. Lichen secondary metabolites are characterized by huge pharmacological potential. Our research focused on assessing the anticancer and neuroprotective activity of Hypogymnia physodes acetone extract (HP extract) and physodic acid, its major component. The antitumor properties were evaluated by cytotoxicity analysis using A-172, T98G, and U-138 MG glioblastoma cell lines and by hyaluronidase and cyclooxygenase-2 (COX-2) inhibition. The neuroprotective potential was examined using COX-2, tyrosinase, acetylcholinesterase (AChE), and butyrylcholinesterase (BChE) activity tests. Moreover, the antioxidant potential of the tested substances was examined, and the chemical composition of the extract was analyzed. For physodic acid, the permeability through the blood-brain barrier using Parallel Artificial Membrane Permeability Assay for the Blood-Brain Barrier assay (PAMPA-BBB) was assessed. Our study shows that the tested substances strongly inhibited glioblastoma cell proliferation and hyaluronidase activity. Besides, HP extract diminished COX-2 and tyrosinase activity. However, the AChE and BChE inhibitory activity of HP extract and physodic acid were mild. The examined substances exhibited strong antioxidant activity. Importantly, we proved that physodic acid crosses the blood-brain barrier. We conclude that physodic acid and H. physodes should be regarded as promising agents with anticancer, chemopreventive, and neuroprotective activities, especially regarding the central nervous system diseases.

Temporal and spatial atmospheric deposition trends of elements to the boreal forest surrounding bitumen production operations in the Athabasca Oil Sands Region (AOSR), Alberta, Canada were investigated as part of a long-term lichen bioindicator study. The study focused on eight elements (sulfur, nitrogen, aluminum, calcium, iron, nickel, strontium, vanadium) that were previously identified as tracers for the major oil sand production sources. Samples of the in situ epiphytic lichen Hypogymnia physodes were collected in 2002, 2004, 2008, 2011, 2014, and 2017 within a ~150 km radius from the center of surface oil sand production operations in the AOSR. Site-specific time series analysis conducted at eight jack pine upland sites that were repeatedly sampled generally showed significant trends of increasing lichen concentrations for fugitive dust linked elements, particularly at near-field (<25 km from a major oil sands production operation) sample locations. Multiple regional scale geostatistical models were developed and evaluated to characterize broad-scale changes in atmospheric deposition based on changes in H. physodes elemental concentrations between 2008 and 2014. Empirical Bayesian kriging and cokriging lichen element concentrations with oil sands mining, bitumen upgrading, coke materials handling, and limestone quarry/crushing influence variables produced spatial interpolation estimates with the lowest validation errors. Gridded zonal mean lichen element concentrations were calculated for the two comprehensive sampling years (2008, 2014) and evaluated for spatial and temporal change. Lichen sulfur concentrations significantly increased in every grid cell within the domain with the largest increases (44-88%) in the central valley in close proximity to the major surface oil sand production operations, while a minor nitrogen concentration decrease (-20%) in a single grid cell was observed. The areal extent of fugitive dust element deposition generally increased with significantly higher deposition to lichens restricted to the outer grids of the enhanced deposition field, reflecting new and expanding surface mining activity., (Copyright © 2019 The Authors. The aim of the study was to assess heavy metals (Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn) air pollution in Bosnia and Herzegovina by using a lichen, Hypogymnia physodes. Metal content was determined by flame atomic absorption spectrometry (FAAS) and was between very high naturality or alteration to middle naturality or alteration. Strong correlations between Cr and Ni confirmed mainly anthropogenic sources. The scanning electron microscopy and energy dispersive X-ray spectroscopy (SEM/EDS) analysis of C, O, Na, Mg, Al, Si, P, S, Cl, K, Ca, V, Co, As, Sn, Sb, Hg and Bi were performed on the lichen surface and hyphae of the transplanted samples. Despite significant damage to tissue and cell integrity, the recurrent presence of particulate matter in lichen indicates the considerable presence of dust in the urban atmosphere which, according to chemical composition, may be due to anthropogenic and natural sources such as soil.

The sources and spatial distribution of polycyclic aromatic hydrocarbons (PAHs) atmospheric deposition in the boreal forests surrounding bitumen production operations in the Athabasca Oil Sands Region (AOSR), Alberta, Canada were investigated as part of a 2014 passive in-situ bioindicator source apportionment study. Epiphytic lichen species Hypogymnia physodes samples (n = 127) were collected within a 150 km radius of the main surface oil sand production operations and analyzed for total sulfur, total nitrogen, forty-three elements, twenty-two PAHs, ten groups of C1-C2-alkyl PAHs and dibenzothiophenes (polycyclic aromatic compounds; PACs), five C1- and C2-alkyldibenzothiophenes, and retene. The ΣPAH + PAC in H. physodes ranged from 54 to 2778 ng g -1 with a median concentration of 317 ng g -1 . Source apportionment modeling found an eight-factor solution that explained 99% of the measured ΣPAH + PAC lichen concentrations from four anthropogenic oil sands production sources (Petroleum Coke, Haul Road Dust, Stack Emissions, Raw Oil Sand), two local/regional sources (Biomass Combustion, Mobile Source), and two lichen biogeochemical factors. Petroleum Coke and Raw Oil Sand dust were identified as the major contributing sources of ΣPAH + PAC in the AOSR. These two sources accounted for 63% (43.2 μg g -1 ) of ΣPAH + PAC deposition to the entire study domain. Of this overall 43.2 μg g -1 contribution, approximately 90% (39.9 μg g -1 ) ΣPAH + PAC was deposited within 25 km of the closest oil sand production facility. Regional sources (Biomass Combustion and Mobile Sources) accounted for 19% of ΣPAH + PAC deposition to the entire study domain, of which 46% was deposited near-field to oil sand production operations. The aim of the study was to assess the metal pollution in the vicinity of the Bukowno smelter near Olkusz in southern Poland. Birch and oak leaves, pine needles and a lichen Hypogymnia physodes, overgrowing pine bark were collected at stands at different distances from the smelter and analysed for cadmium (Cd), copper (Cu), lead (Pb) and zinc (Zn) content. Concentrations of metals in the lichen were usually higher than in the tree leaves/needles and decreased with distance from the smelter, apart from the Cu content. The strongest correlation was noticed between Cd and Pb concentrations, which indicates a common pollution source (the smelter). Our results show that birch leaves can be potentially useful as a bioindicator of Zn air pollution since this species was shown to accumulate high amounts of zinc, related to environmental pollution with that metal, in their leaves.

Lichen species typically have a characteristic profile of secondary metabolites. Dense populations of Hypogymnia physodes growing frequently as epiphytes on tree branches have harmful effects on the host, likely due to their secondary compounds, which were undetected in tree tissues until now. The aim of the present study was to re-characterise the suite of secondary metabolites of H. physodes thalli and to estimate their translocation into spruce (Picea abies) bark. Thallus and bark extracts were compared using ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). The compounds were identified based on their UV, MS and MS/MS spectra as well as retention factors of their TLC analysis. In addition to the previously described secondary metabolites (protocetraric, physodalic, 3-hydroxyphysodic, physodic, and 2'-O-methylphysodic acids, atranorin and chloroatranorin) of H. physodes, further three were identified in its thalli: conphysodalic, 4-O-methylphysodic and α-alectoronic acids. Fragmentation patterns from the negative ionisation of each compound were proposed, some of which were described for the first time. Among all of the detected lichen substances, a few, e.g., physodalic, 3-hydroxyphysodic, physodic acids and atranorin, were present in the bark of spruce branches that were abundantly colonised by lichen. The newly identified compounds of H. physodes thalli may belong to its constant or accessory secondary metabolites. These compounds may be useful in the chemotaxonomic classification of this species. The presence of some lichen substances in spruce bark confirmed their ability to penetrate host tissues. The aim of this study is to determine the chemical composition, and evaluate the genotoxic, and anti-growth potency of the methanol extracts of lichen species Hypogymnia physodes (L.) Nyl. (HPE). Anti-growth effect was tested in two different human breast cancer cell lines (MCF-7 and MDA-MB-231) by the MTT and ATP viability assays and apoptosis was assayed by the caspase-cleaved cytokeratin 18 (M30-antigen). Genotoxic activity of HPE was studied using chromosome aberration and micronuclei tests in human lymphocytes culture in vitro. The chemical composition of H. physodes was analyzed by using direct thermal desorption method coupled with comprehensive gas chromatography-time of flight mass spectrometry (GCXGC-TOF/MS). Our results indicate that HPE has an anti-growth effect at relatively lower concentrations, while relatively higher concentrations are required for genotoxic activity. Several lichen species have been used for medicinal purposes throughout the ages, and they were reported to be effective in the treatment of different disorders including tuberculosis, hemorrhoids, ulcer, dysentery and cancer. It is revealed that they may be easily accessible sources of natural drugs that could be used as a possible food supplement or in pharmaceutical industry after their safety evaluations. However, so far, the nature and/or biological roles of plenty of lichenes have not been elucidated exactly. The aim of this study was to investigate the genetic and oxidative effects of water extracts of three different lichen species; Hypogymnia physodes, Ramalina polymorpha and Usnea florida in cultured human blood cells (n = 5) for the first time. All lichen species were collected from the Erzurum and Artvin provinces (in Turkey) during August 2010. The lichen extracts were added into culture tubes at various concentrations (0 to 2000 mg/L). Chromosome aberrations (CA) and micronucleus (MN) tests were used for genotoxic influences estimation. In addition, biochemical parameters (total antioxidant capacity (TAC) and total oxidative stress (TOS)) were examined to determine oxidative effects. In our in-vitro test systems, it was observed that all tested lichen extracts had no mutagenic effects on human lymphocytes. Furthermore, these extracts exhibited antioxidant properties due to the type of lichen species added to the cultures. In conclusion, these lichens can be a new resource of therapeutics as recognized in this study with their non-mutagenic and antioxidant features.
