This study aims to evaluate differences in gut microbiota structures between infertile women undergoing frozen embryo transfer (FET) with gestational diabetes mellitus (GDM) and healthy controls (HCs), and to identify potential markers. We comprehensively enrolled 193 infertile women undergoing FET (discovery cohort: 38 HCs and 31 GDM; validation cohort: 85 HCs and 39 GDM). Gut microbial profiles of the discovery cohort were investigated during the pre-pregnancy (Pre), first trimester (T1), and second trimester (T2). The microbial community in the HCs group remained relatively stable throughout the pregnancy, while the microbial structure alteration occurred in the GDM group during T2. A model based on ten bacteria and ten metabolites simultaneously was used to predict the risk of GDM developing in the pre-pregnancy state with the ROC value of 0.712. Catechins, the main active components of tea polyphenols, boast remarkable antioxidant activities because of their unique structures. This translates to a range of potential health benefits, including fighting antibacterial, inflammation, and even cancers. However, extracting these beneficial compounds can be tricky as they're prone to degradation. Thankfully, recent advancements have yielded successful methods for isolating and purifying catechins, allowing us to obtain them in their purest form. The power of catechins isn't just theoretical. In vitro and in vivo studies have demonstrated promising results in treating various conditions like inflammation, cancer, neurodegenerative diseases, cardiovascular diseases, diabetes, and more. This review dives deep into the methods used to extract, isolate, and purify catechins. Key Message: NnNAC100-NnSBEII modules enhance starch content of the rhizome in Nelumbo nucifera Gaertn. Nelumbo nucifera Gaertn. is a popular aquatic vegetable and traditional Chinese medicine whose quality and taste are mainly determined by the starch. Although starch-related genes have been functionally characterized, the regulated mechanism of enzyme (SBE) remains unclear. In this study, we identified and functionally elucidated the functions of NnSBEII and NnNAC100 using transient overexpression of NnSBEII and NnNAC100 in rhizomes of lotus, and it significantly increased the amylopectin content and total starch content. Accordingly, functional complementation assay in defective Arabidopsis also showed that NnSBEII compensated for the low content of starch in the mutant sbe2.2. In addition, overexpression of NnSBEII and NnNAC100 significantly increased the content of starch in transgenic lines. Consistently, opposite results were observed under the background of repressed NnSBEII and NnNAC100 in rhizomes of lotus. Furthermore, yeast one-hybrid and dual-luciferase assays revealed that NnNAC100 could directly bind to the NnSBEII promoter and promote the expression of NnSBEII. Transient overexpression of NnNAC100 upregulated NnSBEII expression significantly, while the expression level of AtSBE2.2 in transgenic Arabidopsis overexpressing NnNAC100 was higher than that of WT, which indicated that NnNAC100 promoted the synthesis of amylopectin by enhancing the expression of NnSBEII. In addition, we found that NnSBEII could form a complex protein by interacting with soluble starch synthase (NnSS2) to increase the activity of the SBEII enzyme. These results reveal a novel mechanism that the NnNAC100-NnSBEII-NnSBEII/NnSS2 module regulates amylopectin biosynthesis and these will provide insights into the broader implications of the regulation mechanism of starch biosynthesis., Competing Interests: Declarations. Postoperative cognitive dysfunction (POCD) is a recognized clinical phenomenon characterized by cognitive impairment in patients following anesthesia and surgery, especially in the elderly. However, the pathogenesis of POCD remains unclear. In the last decades, lactate's neuroprotective properties have been increasingly mentioned. The study tested the hypothesis that lactate may attenuate the cognitive impairment induced by anesthesia and surgery in aged mice through SIRT1-dependent antioxidant and anti-inflammatory effects. We used 18-month-old C57BL/6 mice to establish the POCD animal model by exploratory laparotomy with isoflurane anesthesia. For the interventional study, mice were administered lactate, with or without the potent and selective SIRT1 inhibitor EX-527. Behavioral tests including open field (OF), Y maze and fear conditioning (FC) tests were performed from 4 to 7 days after anesthesia and surgery. Immunofluorescence staining and Western blot were employed to assess oxidative damage, activation of microglia and astrocytes, levels of proinflammatory cytokines, and the expression of plasticity-related proteins. Lactate treatment can ameliorate oxidative stress, neuroinflammation, and the decreased levels of plasticity-related proteins induced by anesthesia and surgery, ultimately improving cognitive impairment in aged mice. However, co-treatment with lactate and EX-527 diminished the beneficial effects. During acute respiratory distress syndrome (ARDS), delayed apoptosis of neutrophils and impaired efferocytosis of macrophages constitute two critical limiting steps, leading to secondary inflammatory storm and posing a significant threat to human health. However, due to the failure of previous single target-centric treatments to effectively address these two limiting steps in controlling the inflammatory storm, no available therapies are approved for ARDS treatment. Herein, inspired by spontaneous inflammation resolution, two kinds of Apoptosis and Efferocytosis Restored Nanoparticles (AER NPs) are proposed to overcome these two limiting steps for counteracting severe inflammatory storm. For the first limiting step, neutrophil-targeted apoptosis-restored nanoparticles (AR NPs) accelerated the programmed apoptosis of inflammatory neutrophils. The resolution of the first limiting step facilitated the accumulation of macrophage-targeted and efferocytosis-restored nanoparticles (ER NPs), thereby restoring macrophage efferocytosis and alleviating the second limiting step. The results indicated that after sequential treatment with AER NPs, recruited neutrophils decreased to 13.86%, and macrophage efferocytosis increased to 563.24%. AER NPs promoted inflammation resolution and established a self-healing virtuous loop by addressing the two limiting steps, ultimately effectively treating ARDS. Vitamin D binding protein (VDBP) is a potential biomarker of major depressive disorder (MDD). This study demonstrates for the first time that VDBP is highly expressed in core emotion-related brain regions of mice susceptible to chronic unpredictable mild stress (CUMS). Specifically, the overexpression of microglia (MG)-derived VDBP in the prelimbic leads to depression-like behavior and aggravates CUMS-induced depressive phenotypes in mice, whereas conditional knockout of MG-derived VDBP can reverse both neuronal damage and depression-like behaviors. Mechanistically, the binding of MG-derived VDBP with the neuronal receptor megalin mediates the downstream SRC signaling pathway, leading to neuronal and synaptic damage and depression-like behaviors. These events may be caused by biased activation of inhibitory neurons and excitatory-inhibitory imbalance. Importantly, this study has effectively identified MG-derived VDBP as a pivotal mediator in the interplay between microglia and neurons via its interaction with the neuronal receptor megalin and intricate downstream impacts on neuronal functions, thus offering a promising therapeutic target for MDD., (© 2024 The Author(s). Background: Coronavirus disease 2019 (COVID-19) is a global pandemic with high mortality, and the treatment options for the severe patients remain limited. Previous studies reported the altered gut microbiota in severe COVID-19. But there are no comprehensive data on the role of microbial metabolites in COVID-19 patients., Methods: We identified 153 serum microbial metabolites and assessed the changes in 72 COVID-19 patients upon admission and one-month after their discharge, comparing these changes to those in 133 healthy control individuals from the outpatient department during the same period., Results: Our study revealed that microbial metabolites varied across different stages and severity of COVID-19 patients. These altered microbial metabolites included tryptophan, bile acids, fatty acids, amino acids, vitamins and those containing benzene. A total of 13 distinct microbial metabolites were identified in COVID-19 patients compared to healthy controls. Notably, correlations were found among these disrupted metabolites and organ injury and inflammatory responses related to COVID-19. Furthermore, these metabolites did not restore to the normal levels one month after discharge. Importantly, two microbial metabolites were the core microbial metabolites related to the severity of COVID-19 patients., Conclusions: The microbial metabolites were altered in the acute and recovery stage, correlating with disease severity of COVID-19. Background: The frequency of salt added at the table (i.e., to food after it has been prepared, during consumption) could reflect an individual's long-term dietary preference and habitual intake in Western countries. However, little is known about the association between the frequency of salt added at the table and incident dementia. This study investigates the association of the frequency of salt added at the table with the risk of dementia and explores differences in the associations among people with apolipoprotein E ε4 genotypes., Methods: Individuals indicated whether they added salt to their food at the table (salt used for cooking was not included) "never/rarely", "sometimes", "usually", or "always". Cox proportional hazards models were used to compute hazard ratios with 95 % confidence intervals and evaluate the association between the frequency of salt added at the table and incident dementia., Results: We observed a graded relation between the frequency of salt added at the table and spot urine sodium concentrations and estimated 24-h sodium excretion. Over a median follow-up of 13.7 years, there were 9373 cases of all-cause dementia, including 4119 of Alzheimer's disease and 2052 of vascular dementia. With a higher frequency of salt added at the table, the risk of all-cause dementia, Alzheimer's disease and vascular dementia increased in a monotonic manner after adjustment for potential confounding factors (all P-trend <0.001). These associations of the frequency of salt added at the table with the risks of all-cause dementia, Alzheimer's disease and vascular dementia were greater in apolipoprotein E ε4 heterozygotes and homozygotes., Conclusion: Our study showed that a higher frequency of salt added at the table was associated with a higher risk of incident dementia. Objective: To use routine demographic and clinical data to develop an interpretable individual-level machine learning (ML) model to diagnose knee osteoarthritis (KOA) and to identify highly ranked features., Methods: In this retrospective, population-based cohort study, anonymized questionnaire data was retrieved from the Wu Chuan KOA Study, Inner Mongolia, China. After feature selections, participants were divided in a 7:3 ratio into training and test sets. Class balancing was applied to the training set for data augmentation. Four ML classifiers were compared by cross-validation within the training set and their performance was further analyzed with an unseen test set. Classifications were evaluated using sensitivity, specificity, positive predictive value, negative predictive value, accuracy, area under the curve(AUC), G-means, and F1 scores. The best model was explained using Shapley values to extract highly ranked features., Results: A total of 1188 participants were investigated in this study, among whom 26.3% were diagnosed with KOA. Comparatively, XGBoost with Boruta exhibited the highest classification performance among the four models, with an AUC of 0.758, G-means of 0.800, and F1 scores of 0.703. The SHAP method reveals the top 17 features of KOA according to the importance ranking, and the average of the experience of joint pain was recognized as the most important features., Conclusions: Our study highlights the usefulness of machine learning in unveiling important factors that influence the diagnosis of KOA to guide new prevention strategies. Recently, the research on developing lead-free double-B-cation halide perovskites has attracted attention. However, Cs 2 InBiCl 6 , the most promising one, was shown to be thermodynamically unstable. To improve the stability, organic dication DiMA 2+ (i. e., (CH 2 NH 3 ) + (CH 2 ) 3 (CH 2 NH 3 ) + ) is introduced to design the new double perovskite DiMAInBiX 6 (X=Cl, Br, and I) based on Cs 2 InBiX 6 by replacing two Cs + with a DiMA 2+ . Density functional theory calculations were performed to study the geometric and photoelectric properties of the newly designed materials. Unlike Cs 2 InBiX 6 , both DiMAInBiCl 6 and DiMAInBiBr 6 can exist stably against decompositions, indicating that the replacement of Cs + by DiMA 2+ will improve stability of double perovskites due to the staple effect. In addition, the replacement causes an increase in the band gaps of DiMAInBiX 6 . Particularly, the calculated band gap of DiMAInBiBr 6 (1.31 eV) is close to the optimal value of single junction perovskite solar cell (PSC). Under ideal conditions, the PSC model constructed with DiMAInBiBr 6 performs the best in theoretical simulation. This work proposes DiMAInBiBr 6 as a promising light absorber of PSC, which has high stability and photoelectric performance. Background: The activation of hepatic stellate cells play a pivotal role in the pathogenesis of hepatic fibrosis. However, the current lack of specifically identified targets on these cells poses a significant challenge in developing targeted delivery tools for effective anti-hepatic fibrosis therapeutics in clinical practice., Methods: Cell-systematic evolution of ligands by exponential enrichment method was conducted on HSC-T6 cell line to screen out activated hepatic stellate cell-specific aptamers. The specificity of the selected aptamers in targeting hepatic stellate cells was confirmed after truncation optimization. Furthermore, the optimal aptamer was conjugated with miR-23b-5p via C6 linkage to evaluate the targeting specificity of this complex and assess its potential in downregulating liver fibrosis-related proteins and slowing down the progression of liver fibrosis., Results: The present study successful identified 11 highly enriched single-stranded DNA sequences (APT1-11) that specifically target activated hepatic stellate cells. Subsequent affinity detection and optimization truncation led to the selection of APT8(16-34), which effectively targeted activated hepatic stellate cells both in vivo and in vitro. Moreover, when conjugated with miR-23b-5p, APT8(16-34) also exhibited internalization ability into activated hepatic stellate cells. The delivered cargo miR-23b-5p by APT8 (16-34) effectively targeted to mRNA, leading to translational inhibition and subsequent downregulation of related proteins., Conclusions: We have identified APT8 (16- 34), which exhibits specific targeting and internalization capabilities into activated hepatic stellate cells. Background: Limited research has been conducted on self-harm among preadolescents in China. This study investigated the influence of multidimensional stress on high levels of self-harm behavior in preadolescents., Method: This large-scale cross-sectional study of 7-14-year-old primary school students in grades 3-6 was conducted in Southwest China between September and December 2020. Data on sociodemographic characteristics and multidimensional stressors were collected and analyzed., Results: The self-harm prevalence among the 48,117 preadolescents was 13.6 % (n = 6561), with respective prevalence rates of 13.2 % in males and 14.1 % in females. Chi-square and binary logistic regression analyses were employed. The most important factors for self-harm were ranked as follows: high academic pressure (OR = 2.00, 95 % CI 1.90-2.09), poor relationship with parents (OR = 1.89, 95 % CI 1.78-2.00), frequently being bullied (OR = 1.53, 95 % CI 1.47-1.60), early-onset menstruation (OR = 1.33, 95 % CI 1.22-1.46), frequent smartphone use (OR = 1.31, 95 % CI 1.26-1.37), poor classmate relationships (OR = 1.31, 95 % CI 1.24-1.38), poor relationship between parents (OR = 1.11, 95 % CI 1.08-1.14), poor family financial situation (OR = 1.11, 95 % CI 1.05-1.17), and being bullied within the past 1 month (OR = 1.05, 95 % CI 1.02-1.07)., Limitations: The limitations of this study include its cross-sectional design, the use of a non-structured questionnaire, the subjectivity of some items, the reliance on a single question about self-harm, and the possibility that participants may have concealed the true nature of their behaviors., Conclusion: Preadolescents in China exhibited high rates of self-harm behaviors. Background: The inflammatory response is associated with cardiac repair and ventricular remodeling after myocardial infarction (MI). The key inflammation regulatory factor thymic stromal lymphopoietin (TSLP) plays a critical role in various diseases. However, its role in cardiac repair after MI remains uncertain. In this study, we elucidated the biological function and mechanism of action of TSLP in cardiac repair and ventricular remodeling following MI., Method and Result: Wild-type and TSLP receptor (TSLPR)-knockout ( Crlf2 -/- ) mice underwent MI induction via ligation of the left anterior descending artery. TSLP expression was upregulated in the infarcted heart, with a peak observed on day 7 post-MI. TSLP expression was enriched in the cardiomyocytes of infarcted hearts and the highest expression of TSLPR was observed in dendritic cells. Crlf2 -/- mice exhibited reduced survival and worsened cardiac function, increased interstitial fibrosis and cardiomyocyte cross-sectional area, and reduced CD31 + staining, with no change in the proportion of apoptotic cardiomyocytes within the border zone. Mechanistically, reduced Treg cell counts but increased myeloid cell infiltration and an increased ratio of Ly6C high /Ly6C low monocyte were observed in the ani hearts of Crlf2 -/- mice. Further, TSLP regulated CD4 + T cell activation and proliferation at baseline and after MI, with a greater impact on Treg cells than on conventional T cells. Eutrophication and phosphorus scarcity pose significant environmental and resource management challenges, necessitating the development of efficient phosphorus removal and recovery technologies. In this study, lanthanum hydroxide-coated sludge biochar adsorbent was prepared via in-situ liquid precipitation and its phosphorus adsorption performance was evaluated. The adsorbent obtained under the optimized conditions, i.e., 5:1 ratio of La to sludge and 550 °C of annealing temperature (designated as La-C-550), exhibited a maximum phosphorus adsorption capacity of 76.4 mg-P/g at 25 °C. Mechanistic analysis revealed a multilayer adsorption process involving both physical and chemical interactions, with chemical adsorption primarily driven by the complex substitution of hydroxyl by phosphate. La-C-550 demonstrated stable performance across a pH range of 3-7, and maintained resilience against interference from common anions and humic acid. In continuous flow tests using actual secondary effluent, the adsorbent stably reduced phosphorus concentrations to below 0.1 mg-P/L, complying with the Chinese Surface Water Environmental Quality Class II standards. Objective: To investigate the correlation of the clinical characteristics, fever characteristics, serum biomarkers with cytokine release syndrome (CRS) in patients with relapsed/refractory multiple myeloma (R/R MM) treated with chimeric antigen receptor T cell (CAR-T) immunotherapy., Methods: 104 R/R MM patients who received CAR-T cell therapy at the Affiliated Hospital of Xuzhou Medical University from June 2017 to November 2021 were included, and the correlations of their clinical characteristics, fever characteristics, serum biomarkers with the severity of CRS were analyzed., Results: Among 104 R/R MM patients receiving CAR-T treatment, no CRS was observed in 8 cases (7.7%), and 96 cases (92.3%) developed CRS. Patients with high-risk cytogenetics had a higher risk of developing CRS ( P =0.040), while patients who had previously received autologous hematopoietic stem cell transplantation (ASCT) had a lower risk of developing CRS ( P =0.004). There was a significant difference in the duration of fever between patients with grade 1-2 and grade 3-5 CRS ( P =0.006). The highest body temperature varied among patients with different treatment regimens ( P =0.001). The decrease in total protein in patients with CRS was more significant than in patients without CRS ( P =0.002). Within one month after CAR-T cell infusion, the degree of albumin recovery in patients with grade 3-5 CRS was lower than that in patients with grade 0-2 CRS ( P =0.037). Compared to patients with grade 1-2 CRS, patients with grade 3-5 CRS showed a significant increase in heart rate after CAR-T cell infusion ( P =0.013), while IL-6, C-reactive protein (CRP), and serum ferritin (SF) also showed significant increases ( P =0.007, P < 0.001, P =0.003)., Conclusion: High-risk cytogenetics is a risk factor for severe CRS. Long duration of fever is a clinical characteristic of severe CRS. CRP can better reflect the severity of CRS.

Aims: To investigate the effect of optimal diastolic blood pressure (DBP) level on cardiovascular disease (CVD) events, considering different age groups. And the nonlinear relationship between DBP and CVD events by age were evaluated., Methods: 358,734 participants of the UK biobank were included. Multivariable Cox regressions were performed to evaluate the relation between DBP and cardiovascular risk, and the natural cubic spline was applied to estimate the nonlinear relationship between the DBP measures and the hazard., Results: In the UK Biobank, a total of 358,734 participants (mean [SD] age, 55.3 [8.1] years; 205,215 [57.2 %] female) were included in this analysis. The rate of CVD cases was 6.05 per 1000 person-years. For each SD increase, DBP was associated with elevated CVD events for both <50 year (HR, 1.19; 95 % CI, 1.12 to 1.26) and 50-59 year-old group (HR, 1.04; 95 % CI, 1.01 to 1.08). However, for every SD increment, a marginal protective effect against CVD events was observed between DBP and the CVD events in elder people (≥60 years old) and lower DBP (<70 mm Hg) may confer an increased CVD risks (HR, 1.10; 95 % CI, 1.03 to 1.17)., Conclusions: For adults younger than 60 years old, the study seems to support the ACC/AHA blood pressure guidelines lowering the DBP threshold to 80 mm Hg. The ongoing research on the role of immunotherapy in advanced ovarian cancer (OC) and current clinical trials indicate that patients shown limited response to immune checkpoint inhibitor (ICI) monotherapy. When combined with other treatments or drugs, the efficacy of immunotherapy will be significantly improved. Biomarkers can be used to identify patients with better responses, thereby improving the precision and efficacy of immunotherapy. Key biomarkers for advanced OC include homologous repair deficiency, programmed death-ligand (PD-L) 1 expression, chemokines, and tumor infiltrating lymphocytes. These biomarkers could be applied in the future to select the most suitable patient populations. Background: Z-Ligustilide (ZL) is an essential phthalide found in Ligusticum chuanxiong Hort, a commonly used traditional Chinese medicine for treating atherosclerosis (AS) clinically. ZL has been shown to be effective in treating AS. However, the underlying mechanism of ZL against AS and its potential targets remain elusive., Purpose: The purpose of this research was to assess the influence of ZL on AS and explore the role of the gut microbiome in mediating this effect., Methods: A well-established AS mouse model, apolipoprotein E deficient (ApoE -/- ) mice was used to examine the effects of ZL on AS, inflammation, and the intestinal barrier. To analyze the changes in gut microbial community, we employed the 16S rRNA gene sequencing. Antibiotic cocktail and fecal microbiota transplantation (FMT) were employed to clarify the contribution of the gut microbiota to the anti-AS effects of ZL. The mechanism through which ZL provided protective effects on AS and the intestinal barrier was explored by untargeted metabolomics, as well as by validating the involvement of cannabinoid receptor 2 (CB2R) in mice and Caco-2 cells., Results: Oral administration of ZL inhibited the development of atherosclerotic lesions, improved plaque stability, inhibited the increase in serum and atherosclerotic inflammation, and improved intestinal barrier function. Fecal bacteria from ZL-treated mice induced similar beneficial effects on AS and the intestinal barrier. We used 16S RNA gene sequencing to reveal a significant increase in Rikenella abundance in both ZL-treated mice and ZL-FMT mice, which was associated with the beneficial effects of ZL. Further function prediction analysis of the gut microbiota and CB2R antagonist intervention experiment in mice and Caco-2 cells showed that the activation of CB2R resulted in the enhancement of the intestinal barrier by ZL. Furthermore, the analysis of metabolomic profiling revealed the enrichment of capsaicin upon ZL treatment, which induced the activation of CB2R in human colon epithelial cells., Conclusion: Our study is the first to demonstrate that oral treatment with ZL has the potential to alleviate AS by reducing inflammation levels and enhancing the intestinal barrier function. Objectives: The risk of new-onset hypertension is influenced by habitual fish oil supplementation, but whether the association is modified by genetic predisposition is unknown., Methods: A total of 213,604 participants without hypertension were identified at baseline from the UK Biobank between 2006 and 2010. The weighted polygenetic risk score (PRS) comprising 118 identified single-nucleotide polymorphisms (SNPs) was used to quantify genetic susceptibility. Cox regression models were applied to determine the association between fish oil supplementation, PRS, and hypertension and evaluate the effect modification of genetic susceptibility., Results: During a median follow-up of 13.8 years, 18,498 new-onset hypertension cases were identified. Approximately 30.6 % (65,452) of participants were habitual fish oil users. The hazard ratio (HR) of habitual fish oil users for hypertension was 0.94 (95 % confidence interval [CI], 0.91-0.98). Fish oil nonusers with a high genetic risk had an increased risk of hypertension (HR, 1.52; 95 % CI, 1.41-1.64) compared to fish oil users with a low genetic risk. In addition, an interaction on the additive scale between the fish oil use and intermediate or high levels of genetic susceptibility was observed. Background: Considering the finite time within a 24-h day, the distribution of time spent on movement behaviours has been found to be associated with health outcomes., Objectives: This systematic review and meta-analysis aimed to summarise and evaluate the overflow effects of interventions targeting a single behaviour (physical activity, sedentary behaviour/screen time, or sleep) on other non-targeted behaviours among children and adolescents., Methods: Six databases (MEDLINE [Ovid], PsycINFO [ProQuest], EMBASE [Ovid], PubMed, Web of Science and SPORTDiscus [EBSCO]) were searched for relevant studies published before 13 May, 2024. Randomised controlled trials and clustered randomised controlled trials that targeted a single behaviour and also assessed the effects on non-targeted behaviours, comprised of healthy children under the age of 18 years, were included. Movement behaviours can be measured either objectively or subjectively. The revised Cochrane risk-of-bias tool for randomised trials was adopted to evaluate the risk of bias., Results: A total of 102 studies with 45,998 participants from 21 countries were identified, and 60 of them with 26,183 participants were incorporated into the meta-analysis. The meta-analysis demonstrated that physical activity interventions led to a reduction in the proportion of each day spent in sedentary behaviour (mean difference =  - 0.95% of wear time, 95% confidence interval - 1.44, - 0.45, I 2  = 39%). Sedentary behaviour interventions resulted in increased standing time (mean difference = 3.87%, 95% confidence interval 1.99, 5.75, I 2  = 0%). Interventions targeting screen time did not yield changes in physical activity or sleep. The findings on the effectiveness of sleep interventions on non-targeted behaviours and of physical activity interventions on sleep were inconclusive., Conclusions: Overall, the findings suggested that interventions aimed at increasing physical activity or reducing sedentary behaviour had overflow effects on non-targeted behaviours, but the effect sizes were small. Cordyceps cicadae (Hypocreales: Cordycipitaceae) is a renowned entomopathogenic fungus used as herbal medicine in China. However, wild C. cicadae resources have been threatened by heavy harvesting. We hypothesised that Bombyx mori L. (Lepidoptera: Bombycidae) could be a new alternative to cultivate C. cicadae due to the low cost of rearing. Bacterial communities are crucial for the formation of Cordyceps and for promoting the production of metabolites. To better understand the bacterial community structure associated with Cordyceps, three Claviciptaceae fungi were used to explore the pathogenicity of the silkworms. Here, fifth-instar silkworms were infected with C. cicadae, Cordyceps cateniannulata (Hypocreales: Cordycipitaceae) and Beauveria bassiana (Hypocreales: Cordycipitaceae). Subsequently, we applied high-throughput sequencing to explore the composition of bacterial communities in silkworms. Our results showed that all three fungi were highly pathogenic to silkworms, which suggests that silkworms have the potential to cultivate Cordyceps. After fungal infection, the diversity of bacterial communities in silkworms decreased significantly, and the abundance of Staphylococcus increased in mummified larvae, which may play a role in the death process when the host suffers infection by entomopathogenic fungi. Furthermore, there were high similarities in the bacterial community composition and function in the C. cicadae and C. cateniannulata infected samples, and the phylogenetic analysis suggested that these similarities may be related to the fungal phylogenetic relationship. The intricate hierarchical structure of musculoskeletal tissues, including bone and interface tissues, necessitates the use of complex scaffold designs and material structures to serve as tissue-engineered substitutes. This has led to growing interest in the development of gradient bone scaffolds with hierarchical structures mimicking the extracellular matrix of native tissues to achieve improved therapeutic outcomes. Building on the anatomical characteristics of bone and interfacial tissues, this review provides a summary of current strategies used to design and fabricate biomimetic gradient scaffolds for repairing musculoskeletal tissues, specifically focusing on methods used to construct compositional and structural gradients within the scaffolds. The latest applications of gradient scaffolds for the regeneration of bone, osteochondral, and tendon-to-bone interfaces are presented. Furthermore, the current progress of testing gradient scaffolds in physiologically relevant animal models of skeletal repair is discussed, as well as the challenges and prospects of moving these scaffolds into clinical application for treating musculoskeletal injuries., Competing Interests: Declarations. Astrocytes, which actively participate in cognitive processes, have a complex spongiform morphology, highlighted by extensive ramified fine processes that closely enwrap the pre- and post-synaptic compartments, forming tripartite synapses. However, the role of astrocyte morphology in cognitive processes remains incompletely understood and even controversial. The actin-binding protein Ezrin is highly expressed in astrocytes and is a key structural determinant of astrocyte morphology. Here, we found that Ezrin expression and astrocyte fine process volume in the hippocampus of male mice increased after learning but decreased after lipopolysaccharide injection and in a mouse model of postoperative cognitive dysfunction, both of which involved models with impaired cognitive function. Additionally, astrocytic Ezrin knock-out led to significantly decreased astrocytic fine process volumes, decreased astrocyte-neuron proximity, and induced anxiety-like behaviors and cognitive dysfunction. Astrocytic Ezrin deficiency in the hippocampus was achieved by using a microRNA silencing technique delivered by adeno-associated viruses. Down-regulation of Ezrin in hippocampal astrocytes led to disrupted astrocyte-synapse interactions and impaired synaptic functions, including synaptic transmission and synaptic plasticity, which could be rescued by exogenous administration of D-serine. Remarkably, decreased Ezrin expression and reduced astrocyte fine processes volumes were also observed in aged mice with decreased cognitive function. Moreover, overexpression of astrocytic Ezrin increased astrocyte fine process volumes and improved cognitive function in aged mice. Background: Parkinson's disease (PD) is an age-related neurodegenerative disease characterized by the death of dopamine neurons in the substantia nigra. A large number of studies have focused on dopamine neurons themselves, but so far, the pathogenesis of PD has not been fully elucidated., Results: Here, we explored the significance of oligodendrocyte precursor cells (OPCs)/oligodendrocytes in the pathogenesis of PD using a bioinformatic approach. WGCNA analysis suggested that abnormal development of oligodendrocytes may play a key role in early PD. To verify the transcriptional dynamics of OPCs/oligodendrocytes, we performed differential analysis, cell trajectory construction, cell communication analysis and hdWGCNA analysis using single-cell data from PD patients. Interestingly, the results indicated that there was overlap between hub genes and differentially expressed genes (DEGs) in OPCs not in oligodendrocytes, suggesting that OPCs may be more sensitive to PD drivers. Colchicine is a widely prescribed anti-inflammatory drug for the treatment of gout, familial Mediterranean fever and pericarditis, but its narrow therapeutic window presents a significant risk of severe toxicity. Despite its clinical relevance, the molecular mechanisms underlying colchicine's pharmacological effects and associated toxicity and explored potential therapeutic interventions to mitigate its adverse effects. We showed the colchicine's impact on cellular morphology in human umbilical vein endothelial cells (HUVEC) and HeLa cells including cell rounding and detachment following 24 h of exposure that revealed pronounced cytotoxic effects. We then established a large-scale screening model to identify small molecules capable of reversing colchicine-induced cellular toxicity, and identified MLN4924, an inhibitor of the Cullin-RING E3 ligase (CRL) system, as a promising candidate for mitigating colchicine-induced cellular injury. Through a comprehensive multi-omics approach including transcriptomics, proteomics, phosphoproteomics and ubiquitinomics, we systematically characterized the molecular perturbations caused by colchicine and delineated the protective mechanisms of MLN4924. We found that MLN4924 exerted its protective effects by modulating critical cellular pathways, specifically preventing the dysregulation of cell cycle progression, mitotic disruption and microtubule destabilization triggered by colchicine. Furthermore, proteomic and phosphoproteomic analyses revealed significant alterations in kinase signaling networks, with combined inhibition of CDK1 and PAK1 emerging as an effective strategy to counteract colchicine-induced cellular dysfunction. CO as a typical σ-donor is one of the most important ligands in chemistry, while planar B 13 + is experimentally known as the most prominent magic-number boron cluster analogous to benzene. Joint gas-phase mass spectroscopy, collision-induced dissociation, and first-principles theory investigations performed herein indicate that B 13 + reacts with CO successively under ambient conditions to form a series of boron carbonyl complexes B 13 (CO) n + up to n = 7, presenting the largest boron carbonyl complexes observed to date with a quasi-planar B 13 + core at the center coordinated by n CO ligands around it. Extensive theoretical analyses unveil both the chemisorption pathways and bonding patterns of these aromatic B 13 (CO) n + monocations which, with three delocalized π bonds well-retained over the slightly wrinkled B 13 + moiety, all prove to be boron carbonyl analogs of benzene tentatively named as boron carbonyl aromatics (BCAs). Their π-isovalent B 12 (CO) n ( n = 1-6) complexes with a quasi-planar B 12 coordination center are predicted to be stable neutral BCAs.

Background: Unplanned reoperation postspinal surgery (URPS) leads to prolonged hospital stays, higher costs, decreased patient satisfaction, and adversely affects postoperative rehabilitation. This study aimed to develop and validate prediction models (nomograms) for early URPS risk factors using machine learning methods, aiding spine surgeons in designing prevention strategies, promoting early recovery, reducing complications, and improving patient satisfaction., Methods: Medical records of 639 patients who underwent reoperation postspinal surgery from the First Affiliated Hospital of Air Force Medical University (2018-2022) were collected, including baseline indicators, perioperative indicators, and laboratory indicators. After applying inclusion and exclusion criteria, 122 URPS and 155 non-URPS patients were identified and randomly divided into training (82 URPS and 111 non-URPS) and validation (40 URPS and 44 non-URPS) cohorts. Three machine learning methods (least absolute shrinkage and selection operator regression, Random Forest, and Support Vector Machine Recursive Feature Elimination) were used to select feature variables, and their intersection was used to develop the prediction model, tested on the validation cohort., Results: Six factors-implant, postoperative suction drainage, gelatin sponge, anticoagulants, antibiotics, and disease type-were identified to construct a nomogram diagnostic model. The area under the curve of this nomogram was 0.829 (95% confidence interval 0.771-0.886) in the training cohort and 0.854 (95% confidence interval 0.775-0.933) in the validation cohort. Calibration curves demonstrated satisfactory agreement between predictions and actual probabilities. Background:  The occurrence and development of ST-segment elevation myocardial infarction (STEMI) are accompanied by coronary atherothrombosis and occlusion, and immune responses play prominent roles in their pathogeneses. However, the causes of atherothrombosis remain elusive, and a comprehensive study of T cell-mediated immune responses in coronary thrombi from STEMI patients is lacking., Objectives:  The aim of this study was to determine the heterogeneity and clonality of CD3 + T lymphocytes in STEMI patients at the single-cell level., Methods:  Paired single-cell RNA and T cell receptor (TCR) sequencing was performed on CD3 + T lymphocytes in the coronary thrombi and peripheral blood of STEMI patients, as well as the blood from control subjects without coronary artery disease (CAD)., Results:  Compared with those in the peripheral blood of STEMI patients, the activation, cytotoxicity, proinflammatory, and prothrombotic characteristics of CD3 + T lymphocytes in coronary thrombi were decreased, and the clonality of CD3 + T cells was increased. Compared with those from non-CAD controls, T lymphocytes from STEMI patients exhibited an upregulation of genes related to recent TCR engagement, suggesting antigen-specific stimulation in STEMI. Surgical site infection (SSI) following lumbar disc herniation (LDH) surgery leads to prolonged hospital stays, increased costs and reoperations. Therefore, we aim to develop and validate a nomogram to predict the risk of SSI following LDH surgery, thereby helping spine surgeons design personalized prevention strategies and promote early recovery. Data from 647 patients with SSI who underwent LDH surgery at the First Affiliated Hospital of Air Force Medical University (AFMU) from 2020 to 2023 were collected. Ultimately, 241 patients with SSI were selected based on inclusion and exclusion criteria. Patients were randomly divided into training and validation sets with a ratio of 7:3. LASSO regression, univariate, and multivariate logistic regression were utilized to identify target variables and establish the prediction model, which was subsequently validated. Six factors-Age, Body Mass Index (BMI), Postoperative Suction Drainage (PSD), Gelatin Sponge (GS), None-Preoperative Antibiotic (NPTA), and Thrombin Time (TT)-were selected to construct the nomogram model. In the training set, the area under the curve (AUC) for the nomogram was 0.818 (95% CI 0.779-0.857). In the validation set, the AUC was 0.782 (95% CI 0.717-0.846). Calibration curves for both sets showed satisfactory agreement between predicted and actual SSI probabilities. Decision curve analysis indicated that the nomogram is clinically useful with a threshold range of 1-90%. The Clinical Impact Curve (CIC) demonstrated an acceptable cost-benefit ratio. Based on the important feature of sulfur with excellent selectivity toward selenite in the presence of selenate, a simple and low-cost adsorbent of solid phase extraction known as sulfur loading activated carbon (SAC-6) was successfully prepared and applied for selenite (Se(IV)) analysis in water. Microstructure and morphological characteristics of SAC-6 had been identified by XRD, TEM, BET and FT-IR. In the static adsorption experiments, Se(IV) could be separated in a wide range of pH values (pH=3-11). The retention process of Se(IV) onto SAC-6 was characterized as spontaneous exothermic reaction. An obvious change of adsorption mechanism occurred in static and dynamic adsorption processes shown that the behaviors followed monolayer and hybrid adsorption. The theoretical maximum adsorption capacity of SAC-6 calculated by Langmuir-Freundlich was 13.48 mg/g. The microcolumn filled with SAC-6 was applied to extract Se(IV) in water solution. The detection limit of Se(IV) analytical procedure was confirmed as 0.27 μg/L within a linear range of 10-1000 μg/L. A good precision with relative standard deviation of 1.34 % (100 μg/L, n = 6) was achieved. The high adaptability and accuracy of SAC-6 microcolumn was validated by analyzing natural water samples and certified reference materials. Sporopollenin is a natural biological macromolecule consisting of highly cross-linked carbon, hydrogen, and oxygen atoms, with a highly porous structure and multifunctional groups. In this work, a novel surface molecularly imprinted polymer based on magnetically aminated cattail sporopollenin (MACSp-SMIP) was prepared for the specific and efficient adsorption of resveratrol, with the aim of purifying resveratrol from Polygonum cuspidatum extracts. MACSp-SMIP was found to have a porous structure covered with the multi-layered sponge-like imprinted polymers. MACSp-SMIP had a high adsorption capacity for resveratrol (65.77 mg·g -1 ) and excellent selectivity (imprinting factor 5.64). The adsorption of resveratrol by MACSp-SMIP was a homogeneous diffusion dominated by chemical adsorption with three stages of external diffusion, internal diffusion, and micropore diffusion. MACSp-SMIP was used as an adsorbent in molecularly imprinted solid-phase extraction for the purification of resveratrol from P. cuspidatum extracts, achieving a resveratrol recovery of 94.33 % and a purity of 76.67 % in the final products. MACSp-SMIP maintained a satisfactory recovery of resveratrol (88.18 %) after six cycles. Minimally processed fruits are increasingly demanded in modern society, but the management of perishable waste pomaces (WPs) and the products' short shelf-life are still big issues. Here, a facile approach of reconstruing apple pomace (AP) into edible bio-nanocomposite coatings of fresh-cutting apple slices was successfully developed through alkaline demethylation followed by high-pressure homogenization. The fibrillation of AP fibers is largely improved by -COO - at a concentration of 1.23 mmol g -1 , which is released through alkaline demethylation of pectin, instead of relying on intricated or costly cellulose modifications. The average width of AP nanofibers (AP-NFs) downsizes to 18 nm. By casting, AP-NFs fabricate homogeneous films with comparable transparency (56 % at 600 nm), superior mechanical strength (6.4 GPa of Young modulus and 81.7 MPa of strength) and oxygen barrier properties (79 mL μm m -2  day -1  bar -1 ), and non-toxicity. Objective: Although menopause is considered a risk factor for multimorbidity, few studies have explored the association between reproductive lifespan and multimorbidity. This study aimed to explore the association between reproductive lifespan and multimorbidity in postmenopausal Chinese women., Methods: This cross-sectional study selected postmenopausal women as study participants. The reproductive lifespan refers to the interval between menarche and menopause. Multimorbidity refers to having two or more self-reported chronic diseases. We used a logistic regression model to explore potential associations based on the adjustment of a set of covariates., Results: In total, 1,310 postmenopausal women with an average reproductive lifespan of 34 years were included in this study. The prevalence of multimorbidity was 22.2% (291/1,310) in postmenopausal women. Our findings showed that compared with postmenopausal women with the Q1 of reproductive lifespan (≤32 reproductive years), those with Q3 (35-37 reproductive years) and Q4 (≥38 reproductive years) were less likely to have multimorbidity (OR Q3 = 0.529, 95% CI Q3 = 0.347-0.805, OR Q4 = 0.510, 95% CI Q4 = 0.308-0.842), whereas those with Q2 (33-34 reproductive years) were not (OR = 0.700, 95% CI = 0.446-1.098). This study also revealed a linear trend in the association between the reproductive lifespan and multimorbidity; that is, the longer the reproductive lifespan, the lower the risk of multimorbidity., Conclusions: In postmenopausal Chinese women, a longer reproductive lifespan was associated with a lower prevalence of multimorbidity. This paper reports a case of a WDR45 variant inherited from an asymptomatic low-percentage mosaic mother. The proband boy was found to have significant psychomotor developmental delay, epilepsy, and abnormal liver function at four months of age, and a hemizygous variant WDR45 c.867&#95;869dupGTA (p.Y290*) was detected by high throughput sequencing, which has an ACMG rating of likely pathogenic variant. The same variant was detected by high-throughput sequencing of the amniotic fluid of the fetus at his mother's next pregnancy. Background: Acute mesenteric vein thrombosis (MVT) accounts for only 2%-10% of all cases of acute mesenteric ischaemia, with an incidence rate of ~0.1% in Europe and the United States. It represents < 10% of mesenteric infarction cases and is seen predominantly in older adults. In younger individuals, MVT is uncommon, with 36% of cases having unidentified mechanisms and causes., Case Summary: A 27-year-old man presented to the emergency department on February 29, 2024, with a chief complaint of intermittent abdominal pain for 3 day. He was previously in good health. As the abdominal pain was not alleviated by conventional treatment, an abdominal computed tomography (CT) scan was performed, which showed increased density in the portal and mesenteric veins. Further imaging, including portal vein ultrasound, mesenteric CT angiography, and enhanced abdominal CT, revealed widespread thrombosis of the portal vein system (including the main portal vein, left and right branches, proximal mesenteric vein, and splenic vein). After 10 day of thrombectomy and anticoagulation therapy, the patient's abdominal pain had improved significantly. Follow-up assessments indicated that portal venous blood flow had largely returned to normal. He was discharged on March 9, 2024. Endophytic fungi can effectively regulate the biosynthesis of health-beneficial metabolites in plants. However, few studies have revealed how the accumulation of host metabolites varies during interactions with endophytic fungi. Here, pigeon pea hairy root cultures (PPHRCs) were cocultured with an endophytic fungus Penicillium rubens to explore the impact on the biosynthesis and accumulation of cajaninstilbene acid (CSA). The results showed that CSA accumulation in PPHRCs increased significantly (15.29-fold) during the early stages of P. rubens colonization (fungal attachment and invasion phases). Once P. rubens successfully colonized the intercellular gap of hairy roots to form a symbiotic relationship, the CSA levels in PPHRCs decreased drastically. Moreover, P. rubens could be recognized by plant pattern recognition receptors that regulate immunity/symbiosis, triggering the expression of genes related to pathogenesis, CSA biosynthesis, and ABC transporter. Overall, P. rubens could enhance the accumulation of health-promoting CSA in PPHRCs during the early stages of colonization.

Exploring the different spin states of central metals in the complex to regulate the anti-tumor activity of cancer cells is of great significance in drug design and clinical use. However, it is a challenge to build a strong coupling between spin states and anti-tumor activities in one system. Herein, we present two complexes {Fe II 2 L 2 [Pd II (CN) 4 ] 2 }·2H 2 O (L = Bztpen (1), Bztppn (2); Bztpen = N -benzyl- N , N ', N '-tris(2-pyridylmethyl)ethylenediamine, Bztppn = N -benzyl- N , N ', N '-tris(2-pyridylmethyl)propylenediamine) showing different cytotoxic activities actuated by fine-tuning the structure with different spin states of Fe(II). Magnetic susceptibility measurements and X-ray diffraction revealed that the Fe(II) ion in complexes 1 and 2 remains in the LS and HS state, respectively, at room temperature. Cytotoxicity tests indicate that complex 1 is more biologically effective than complex 2. In complex 2, however, the high-spin Fe(II) played a key role in regulating its in vitro antitumor effects and seems to be associated with ROS-mediated apoptosis. These findings offer a new avenue for developing anti-cancer drugs by designing complexes with different spin states.

Currently, scientifically and reasonably specifying carbon emission reduction measures in the context of "double carbon" has become a common concern worldwide. China's administrative divisions have a notable impact on the formulation and implementation of relevant policies. Therefore the carbon emissions must be calculated accurately under China's administrative divisions at different scales. The spatiotemporal change characteristics of absorption and carbon emissions can provide scientific basis for the formulation of reasonable and differentiated carbon emission reduction policies in different administrative regions in China. To this end, this study used multi-source data such as remote sensing and statistics and integrated ecological models, statistics, and GIS space analysis and other methods to analyze the spatiotemporal dynamic change characteristics of carbon emissions and carbon absorption at different administrative scales （provinces, cities, and counties） in China. The results showed that： ① The total carbon absorption of vegetation in China continued to increase from 2000 to 2021 and the average value gradually increased. Differences were observed in spatiotemporal changes in carbon emissions at different administrative scales. The spatiotemporal changes at smaller scales were more evident. Carbon emissions showed obvious spatial differences of "high in the north and low in the south, high in the east and low in the west." ② The spatiotemporal distribution of CPI at the administrative scale was similar to that of carbon emissions and the overall trend was increasing annually. The pressure of carbon emissions on carbon absorption gradually weakened from the east to the central and western regions. ③ Spatiotemporal hotspot analysis showed that the overall spatial distribution of cold and hot spots in China's carbon absorption was as follows： In the spatial pattern of "hot in the east and cold in the west," the spatial distribution of cold and hot spots of carbon emissions showed agglomeration characteristics. The provincial scale was primarily oscillating hotspot whereas municipal and county scales were majorly continuous hot spots. Further results revealed that： ① Carbon absorption in different regions and periods in China showed significant variability, especially in the central and eastern regions. The possibility of offsetting carbon emissions by increasing carbon absorption remains. ② At the same scale, administrative regions （such as different provinces） and lower-level administrative regions at another scale （such as different cities in the same province） showed varying degrees of variability in carbon absorption and carbon emissions. Therefore, taking provincial administrative regions as an example for subsequent formulation considering carbon trading, emission reduction, and other policies, we should first consider the coordination of emissions between different cities in the province and then consider the coordination between provinces, which is expected to better promote the implementation of relevant policies.

PCOS is one of the most common endocrine disorders among women of reproductive age. While the mechanism involved is not yet fully characterized. Our study aims to examine the pregnancy outcomes of embryo transfers in women with PCOS after pretreatment, and to explore the possible effect of high androgen levels on endometrial receptivity. Retrospective cohort study was conducted to analyze pregnancy outcomes among 2714 infertile women with tubal factor and 452 PCOS women. Endometrium samples were collected from 6 controls and 6 PCOS patients to detect the expression of endometrial receptivity marks. The implantation rate, clinical and ongoing pregnancy rates and live birth rate in women with PCOS followed fresh embryo transfers were obviously decreased even after the pretreatment. Similar pregnancy outcomes were found in frozen-thawed embryo transfer cycles between women with or without PCOS. Strikingly, serum total testosterone (TT) levels on trigger day were significantly higher in PCOS women. Women with high TT levels presented significantly lower clinical and ongoing pregnancy rates, and the expression of insulin-like growth factor binding protein 1 (IGFBP-1), and leukemia inhibitory factor (LIF) in the endometria decreased significantly as well. High doses of testosterone significantly down-regulated the expression of IGFBP-1 and LIF in Ishikawa cells. Although endocrine abnormalities had been improved before the controlled ovarian stimulation (COS) cycle started, higher serum TT levels were detected on the trigger day of the COS cycle in PCOS patients, which may contribute to the decreased fresh embryo implantation by impairing endometrial receptivity., (© 2024. Background: The obesity paradox has been reported among older adults. However, whether the favorable effect of obesity is dependent on metabolic status remains largely unknown. We aimed to explore the association of metabolic obesity phenotypes and their changes with all-cause mortality among the Chinese oldest-old population., Methods: This prospective cohort study included 1207 Chinese oldest old (mean age: 91.8 years). Metabolic obesity phenotypes were determined by central obesity and metabolic status, and participants were classified into metabolically healthy obesity (MHO), metabolically unhealthy obesity (MUO), metabolically healthy non-obesity (MHN), and metabolically unhealthy non-obesity (MUN). The hazard ratios (HRs) and 95% confidence intervals (95% CIs) were estimated by Cox regression models., Results: During 5.3 years of follow-up, 640 deaths were documented. Compared with non-obesity, obesity was associated with a decreased mortality risk among participants with metabolically healthy (HR, 0.75; 95% CI, 0.63-0.91) while this association was insignificant among metabolically unhealthy. Compared to MHO, MHN (HR, 1.27; 95% CI, 1.06-1.53) and MUN (HR, 1.49; 95% CI, 1.10-2.02) were significantly associated with an increased mortality risk. Compared to those with stable MHO, those transited from MHO to MUO demonstrated a higher mortality risk (HR, 1.81; 95% CI, 1.06-3.11)., Conclusions: MHO predicts better survival among the Chinese oldest-old population. These findings suggest that ensuring optimal management of metabolic health is beneficial and taking caution in weight loss based on the individual body weight for the metabolically healthy oldest-old adults., (© 2024. Granular microporous hydrogels are emerging as effective biomaterial scaffolds for tissue engineering due to their improved characteristics compared to traditional nanoporous hydrogels, which better promote cell viability, cell migration, cellular/tissue infiltration, and tissue regeneration. Recent advances have resulted in the development of granular hydrogels made of non-spherical microgels, which compared to those made of spherical microgels have higher macroporosity, more stable mechanical properties, and better ability to guide the alignment and differentiation of cells in anisotropic tissue. The development of these hydrogels as an emerging research area is attracting increasing interest in regenerative medicine. This review first summarizes the fabrication techniques available for non-spherical microgels with different aspect-ratios. Then, it introduces the development of granular microporous hydrogels made of non-spherical microgels, their physicochemical characteristics, and their applications in tissue regeneration. Asthma is a chronic respiratory disease characterized by airway inflammation and remodeling. Epithelial-mesenchymal transition (EMT) of bronchial epithelial cells is considered to be a crucial player in asthma. Methyltransferase-like 14 (METTL14), an RNA methyltransferase, is implicated in multiple pathological processes, including EMT, cell proliferation and migration. However, the role of METTL14 in asthma remains uncertain. This research aimed to explore the biological functions of METTL14 in asthma and its underlying upstream mechanisms. METTL14 expression was down-regulated in asthmatic from three GEO datasets (GSE104468, GSE165934, and GSE74986). Consistent with this trend, METTL14 was decreased in the lung tissues of OVA-induced asthmatic mice and transforming growth factor-β1 (TGF-β1)-stimulated human bronchial epithelial cells (Beas-2B) in this study. Overexpression of METTL14 caused reduction in mesenchymal markers (FN1, N-cad, Col-1 and α-SMA) in TGF-β1-treated cells, but caused increase in epithelial markers (E-cad), thus inhibiting EMT. Also, METTL14 suppressed the proliferation and migration ability of TGF-β1-treated Beas-2B cells. Two transcription factors, ETS1 and RBPJ, could both bind to the promoter region of METTL14 and drive its expression. Elevating METTL14 expression could reversed EMT, cell proliferation and migration promoted by ETS1 or RBPJ deficiency. Litter decomposition significantly influences the carbon (C) dynamics of terrestrial ecosystems. Solar radiation is not only essential for photosynthetic C fixation and primary productivity, but also can directly or indirectly promote litter decomposition through photodegradation. Recently, photodegradation has been identified as a key factor driving litter decomposition and potentially impacts terrestrial C cycle. To enrich and develop the theory of litter decomposition, we summarized the mechanisms and main driving factors of photodegradation, and compared the responses of photodegradation to environment and climate changes at different scales. Photodegradation primarily includes photomineralization, photoinhibition, and photofaciliation, each affecting litter decomposition differently under various environmental conditions. Photodegradation is closely related to factors such as solar radiation, litter traits, temperature, moisture, microorganisms, and vegetation cover. The interactions among these factors complicate the patterns of photodegradation. Finally, we identified the main issues in litter photodegradation research and prospected future research directions. We emphasized the needs for in-depth exploration of photodegradation pathways and intrinsic mechanisms, quantification of its interactive effects with environmental factors, and optimization of traditional carbon turnover models.

Photodegradation driven by solar radiation has been confirmed as an important driving factor for litter decomposition. However, previous single-site studies could not quantify the relative contribution of variation in solar radiation to litter decomposition. To address it, we conducted a field experiment in Heshan National Field Research Station of Forest Ecosystem, Guangdong (Heshan Station, south subtropical climate), Jigongshan Ecological Research Station, Xinyang, Henan (Jigongshan Station, north subtropical climate) and Daqinggou Ecological Research Station, Institute of Applied Ecology, Chinese Academy of Sciences (Daqinggou Station, temperate climate) at intervals of 10 degrees. We examined litter decomposition of Populus davidiana and Larix olgensis , two species with significant differences in initial litter quality through an in-situ spectral-attenuation experiment. Treatments included full-spectrum, No-UV-B (attenuating UV-B radiation <315 nm) and No-UV & Blue (attenuating all UV and blue wavelengths <500 nm). After nearly 1-year decomposition, litter dry mass remaining of P. davidiana and L. olgensis under full-spectrum treatment was lowest at Heshan (30.2% and 36.3%), and highest at Jigongshan (37.3% and 45.8%). Among all sites, litter dry mass remaining was lowest under the full-spectrum, and lower than that of No-UV-B and No-UV & blue. UV and blue light significantly increased litter mass loss of P. davidiana and L. olgensis , with contributions of 59.7% and 57.0% (Heshan), 46.4% and 42.1% (Jigongshan), and 39.0% and 45.9% (Daqinggou), respectively. The contribution of UV-A and blue light (315-500 nm) was greater than UV-B (280-315 nm); the cumulative irradiance, soil temperature and moisture were the main driving factors for litter photodegradation.

Objective: Rehabilitation and recovery duration following anterior cruciate ligament reconstructive surgery play a pivotal role in restoring optimal knee functionality in athletes. This study aimed to explore the impact of a 3-month functional training programme aligned with enhanced recovery after surgery on recuperation subsequent to anterior cruciate ligament reconstructive surgery., Design: A quasi-experimental study., Subjects: A cohort of 34 patients aged 14 to 24, who underwent anterior cruciate ligament reconstructive surgery and adhered to enhanced recovery after surgery protocols during the perioperative period, were allocated to an experimental group and a control group according to their eligibility, capacity, and willingness to engage in the functional training programme., Methods: The participants in the experimental group underwent a 3-month regimen of functional training following anterior cruciate ligament reconstructive surgery, whereas the control group followed a conventional recovery approach. Evaluations were conducted both prior to and following the 3-month recovery interval, utilizing the Y-Balance Test, Functional Movement Screening, and Isokinetic Knee Test., Results: Assessment outcomes of the Y-Balance Test, Isokinetic Knee Test, and Functional Movement Screening exhibited significant enhancement (p < 0.05) within the experimental group, as opposed to the control group. These findings underscore that those athletes who undertook the 3-month functional training regimen within the experimental group exhibited heightened dynamic balance capabilities, increased knee joint mobility, and enhanced stability compared with their counterparts in the control group., Conclusion: Consequently, this underscores the efficacy of the 3-month functional training protocol aligned with enhanced recovery after surgery, as a means to effectively facilitate recuperation subsequent to anterior cruciate ligament reconstructive surgery.

Estimation of drug ingestion time (event time) and distinguishing between drug ingestion and external contamination are important for interpreting hair analysis results in forensics practice. Here, we present a matrix-assisted laser desorption/ionization-mass spectrometry imaging (MALDI-MSI) method for in situ analysis of intact hair. We applied a longitudinal cutting method for a single hair to analysis authentic hair samples from a victim of a drug-facilitated sexual assault (DFSA) case and zolpidem-soaked hair. MALDI-MSI showed that zolpidem-positive segments distributed at 4-6 mm or 6-8 mm from the root in three single hairs of a DFSA victim collected 25 days after the event, at concentrations ranging from 0.1 to 5.7 pg mm -1 , in agreement with the results from segmental analysis using liquid chromatography tandem mass spectrometry (LC-MS/MS). The estimation of drug intake time was about 20-30 days before sampling, which was consistent with the known time of drug intake. This MALDI-MS method allows imaging analysis of trace substances in a single hair and can realize the intuitive reflection of drug taking time. In addition, zolpidem applied by soaking was mainly distributed on both sides of the longitudinal hair shaft, whereas ingested zolpidem was found only in the middle of the hair shaft of the DFSA victim. The MALDI-MS images of unwashed and washed hair suggested that the amount of externally applied drug was decreased by washing, it was still present on surface layer (cuticle) sides although.
