Microplastics (MPs) and heavy metals (HMs) are important pollutants in terrestrial ecosystems. In particular, the "island" landscape's weak resistance makes it vulnerable to pollution. However, there is a lack of research on MPs and HMs in island landscapes. Therefore, we used Helan Mountain as the research area. Assess the concentrations, spatial distribution, ecological risks, sources, and transport of MPs and HMs in the soil and blue sheep (Pseudois nayaur) feces. Variations in geographical distribution showed a connection between human activity and pollutants. Risk assessment indicated soil and wildlife were influenced by long-term pollutant polarization and multi-element inclusion (I geo , Class I; PHI, Class V; RI (MPs), 33 % Class II, and 17 % Class IV; HI = 452.08). Source apportionment showed that tourism and coal combustion were the primary sources of pollutants. Meanwhile, a new coupling model of PMF/Risk was applied to quantify the source contribution of various risk types indicated transportation roads and tourism sources were the main sources of ecological and health risks, respectively. Improve the traceability of pollution source risks. Furthermore, also developed a novel tracing model for pollutant transportation, revealing a unique "source-sink-source" cycle in pollutant transportation, which provides a new methodological framework for the division of pollution risk areas in nature reserves and the evaluation of spatial transport between sources and sinks. Wildlife activity patterns, which reveal the daily allocation of time and energy, are crucial for understanding survival pressures, adaptive strategies, and behavioral characteristics in different environments. Among ungulates, grouping behavior is a prevalent adaptive trait that reflects the population structure, mating systems, and life history strategies formed over long-term evolutionary processes. This study aimed to elucidate the daily activity patterns and grouping characteristics of the rare goitered gazelle ( Gazella subgutturosa ) in the Helan Mountains of western China from 2022 to 2023 using camera trap monitoring. With a total of 3869 camera days of effective trapping, we recorded 442 independent detections of goitered gazelles. The results revealed the following: (1) Goitered gazelle is primarily active during the day, showing an activity pattern similar to crepuscular animals, with two activity peaks occurring after dawn and before dusk. (2) Daily activity patterns showed both seasonal and sex differences. In the warm season, morning activity peaks occurred earlier, and afternoon peaks occurred later compared to the cold season. The overlap in daily activity patterns between females and males in the warm season was lower than that in the cold season, and this trend persisted throughout the year. (3) The number of times different types of groups were observed varied significantly, with single males and single females accounting for a larger proportion of all observed groups. There was no significant difference in group size across seasons, with groups typically consisting of 1-2 individuals. Our study provides detailed insights into the temporal ecology and population structure of goitered gazelles in arid and semi-arid ecosystems. This information will guide the identification of future conservation priorities and the development of management plans for the reserve.

Background & Aims: Non-invasive models stratifying clinically significant portal hypertension (CSPH) are limited. Herein, we developed a new non-invasive model for predicting CSPH in patients with compensated cirrhosis and investigated whether carvedilol can prevent hepatic decompensation in patients with high-risk CSPH stratified using the new model., Methods: Non-invasive risk factors of CSPH were identified via systematic review and meta-analysis of studies involving patients with hepatic venous pressure gradient (HVPG). A new non-invasive model was validated for various performance aspects in three cohorts, i.e., a multicenter HVPG cohort, a follow-up cohort, and a carvedilol-treating cohort., Results: In the meta-analysis with six studies (n = 819), liver stiffness measurement and platelet count were identified as independent risk factors for CSPH and were used to develop the new "CSPH risk" model. In the HVPG cohort (n = 151), the new model accurately predicted CSPH with cutoff values of 0 and -0.68 for ruling in and out CSPH, respectively. In the follow-up cohort (n = 1,102), the cumulative incidences of decompensation events significantly differed using the cutoff values of <-0.68 (low-risk), -0.68 to 0 (medium-risk), and >0 (high-risk). In the carvedilol-treated cohort, patients with high-risk CSPH treated with carvedilol (n = 81) had lower rates of decompensation events than non-selective beta-blockers untreated patients with high-risk CSPH (n = 613 before propensity score matching [PSM], n = 162 after PSM)., Conclusions: Treatment with carvedilol significantly reduces the risk of hepatic decompensation in patients with high-risk CSPH stratified by the new model.

This study investigates the adaptive strategies of the Alashan Ground Squirrel (Spermophilus alashanicus) in response to habitat changes, as rodents are sensitive indicators of ecosystem changes. Despite its ecological importance, the genome and microbiome of this species have not been thoroughly studied. This research fills this gap by presenting the first comprehensive metagenomic and transcriptomic datasets of the species. Transcriptomic data was collected from five tissue types, including heart, liver, cecum, muscle, and blood, resulting in the assembly of 72,156 unigenes. Metagenomic sequencing identified predominant bacterial groups such as Firmicutes, Bacteroidetes, Verrucomicrobia, Urovircota, and Proteobacteria. Our workflow involved RNA and DNA extraction, library preparation, assembly, and annotation, yielding valuable insights into gene discovery, microbial composition, and further genome and microbial function studies. Background: This study aimed to establish and validate a nomogram model for predicting 90-day mortality in patients with acute basilar artery occlusion receiving endovascular thrombectomy., Methods and Results: A total of 242 patients with basilar artery occlusion undergoing endovascular thrombectomy were enrolled in our study, in which 172 patients from 3 stroke centers were assigned to the training cohort, and 70 patients from another center were assigned to the validation cohort. Univariate and multivariate logistic regression analyses were adopted to screen prognostic predictors, and those with significance were subjected to establish a nomogram model in the training cohort. The discriminative accuracy, calibration, and clinical usefulness of the nomogram model was verified in the internal and external cohorts. Six variables, including age, baseline National Institutes of Health Stroke Scale score, Posterior Circulation-Alberta Stroke Program Early CT (Computed Tomography) score, Basilar Artery on Computed Tomography Angiography score, recanalization failure, and symptomatic intracranial hemorrhage, were identified as independent predictors of 90-day mortality of patients with basilar artery occlusion and were subjected to develop a nomogram model. The nomogram model exhibited good discrimination, calibration, and clinical usefulness in both the internal and the external cohorts. Additionally, patients were divided into low-, moderate-, and high-risk groups based on the risk-stratified nomogram model., Conclusions: Our study proposed a novel nomogram model that could effectively predict 90-day mortality of patients with basilar artery occlusion after endovascular thrombectomy and stratify patients with high, moderate, or low risk, which has a potential to facilitate prognostic judgment and clinical management of stroke.

Global climate change, habitat fragmentation, and human interference have resulted in a significant, ongoing decline in the population of goitered gazelles. Effective conservation strategies require an understanding of resource requirements of threatened species, such as dietary needs. Therefore, we aimed to elucidate the food composition and seasonal dietary changes of goitered gazelles through microhistological analyses of fresh feces. Fabaceae (11.5%), Gramineae (9.4%), Chenopodiaceae (20.2%), Asteraceae (10.1%), and Rosaceae (19.5%) formed the primary dietary components of goitered gazelle. Additionally, Krascheninnikovia arborescens (13.4%) and Prunus sibirica (16.3%) were identified as the key forage plants. Forbs (50.4%) were the predominant plants for grazing throughout the year, particularly in the spring (72.9%). The proportion of trees in the diet was highest in the autumn (36.7%) and comparatively lower in other seasons. Furthermore, the proportions of shrubs (22.0%) and graminoids (14.8%) both reached their peaks in the winter. Our findings indicate that goitered gazelles strategically forage seasonally to cope with resource bottlenecks, enhancing their adaptability to arid and semi-arid habitats. Our study provides essential ecological information for the conservation of goitered gazelles and emphasizes the importance of dietary studies of species of ecological significance in environmentally sensitive areas.

The Alashan ground squirrel ( Spermophilus alashanicus ) is primarily distributed in the regions of Inner Mongolia and Ningxia, China. In this study, we present the first complete mitochondrial genome of S. alashanicus . The genome spans 16,464 base pairs and comprises 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a single control region with a marked AT bias. The overall GC content is 35.4%. Phylogenetic analyses indicate that S. alashanicus clusters are closely associated with S. dauricus . Objective: We sought to describe the resolution time of chronic subdural hematoma (CSDH) after middle meningeal artery embolization (MMAE) and potential variables that may affect hematoma resolution., Methods: A retrospective analysis was performed on CSDH patients between December 2018 and December 2021. Patient characteristics, radiologic manifestations, and data of hematoma resolution were recorded. Univariate and multivariate analyses were conducted to identify predictors of CSDH resolution time., Results: A total of 53 patients were enrolled including 53 hematomas. Only 1 participant relapsed and did not require surgical evacuation. Hematoma resolution was observed in 27 (50.9%) at 4 months and 48 (90.6%) cases at the last radiologic follow-up. The median MMAE-to-resolution time was 19 weeks (interquartile range: 8-24). The burr-hole irrigation + MMAE group showed faster hematoma resolution than MMAE alone during early follow-up periods, but no significant difference was found at 6 months. Increased thickness of residual hematoma, excessive postoperative midline shift, high-density hematoma, mixed-density hematoma, separated hematoma, and anticoagulant or antiplatelet agents used were predictive of nonresolution at 4 months as determined by univariate analysis, whereas anticoagulant or antiplatelet agents used and high-density hematoma were not significant on multivariate analysis. No significant association was noted between hematoma resolution and comorbidities or other hematoma radiologic features., Conclusions: MMAE is an effective and minimally invasive treatment for CSDH with a lower recurrence rate. The median resolution time of CSDH following MMAE was 19 weeks (interquartile range: 8-24). Burr-hole irrigation contributed to early hematoma resolution but had no significant effect at 6 months. Climate warming poses a significant threat to species worldwide, particularly those inhabiting arid and semi-arid regions where extreme temperatures are increasingly prevalent. However, empirical studies investigating how moderate heat events affect the physiological processes of arid and semi-arid animals are largely scarce. To address this knowledge gap, we used an arid and semi-arid lizard species (Phrynocephalus przewalskii) as a study system. We manipulated thermal environments to simulate moderate heat events (43.5 ± 0.3°C during the heating period) for lizards and examined physiological and biochemical traits related to survival, metabolism, locomotion, oxidative stress, and telomere length. We found that the body condition and survival of the lizards were not significantly affected by moderate heat events, despite an increase in body temperature and a decrease in locomotion at high test temperatures were detected. Mechanistically, we found that the lizards exhibited down-regulated metabolic rates and enhanced activities of antioxidative enzymes, resulting in reduced oxidative damage and stable telomere length under moderate heat events. Based on these findings, which indicated a beneficial regulation of fitness by physiological and biochemical processes, we inferred that moderate heat events did not have a detrimental effect on the toad-headed agama, P. przewalskii. Purpose: Circulating tumor DNA (ctDNA) is more representative and accurate than biopsy and is also conducive to dynamic monitoring, facilitating accurate diagnosis and prognosis of glioma. Therefore, the present study aimed to establish and validate a novel amplified method for the detection of IDH1 R132H and BRAF V600E, which were associated with the genetic diagnosis of glioma., Patients and Methods: A dual-signal amplification method based on magnetic aggregation and catalytic hairpin assembly (CHA) was constructed for the simultaneous detection of ctDNAs. When target ctDNAs are present, the CHA reaction is initiated and leads to the assembly of Au-Ag nanoshuttles (Au-Ag NSs) onto magnetic beads (MBs). Further enrichment of MBs under an external magnetic field facilitated the dual-signal amplification of SERS., Results: The limit of detection (LOD) for IDH1 R132H and BRAF V600E in serum was as low as 6.01 aM and 5.48 aM. The reproducibility and selectivity of the proposed SERS analysis platform was satisfactory. Human interventions have a great potential of spatially cornering and limiting species, therefore investigating the species distribution is one of the most crucial issues for managing wildlife populations and suggesting robust conservation strategies. Water deer (Hydropotes inermis) are widespread in China throughout history and are endemic to East Asia. However, they disappeared from Northeast China for years. We rediscovered the water deer in a previous study in Jilin Province, China. Then, we conducted further research in Northeast China to determine their distribution status, supplying fundamental data for the recovery and expansion of their population. An interview survey, line transect survey and infrared camera monitoring were carried out in some counties/cities in Northeast China from June to December 2021. The results showed that the water deer were distributed in Wuchang city of Heilongjiang Province, Changbai Korean Autonomous County, Baishan Municipal District, Ji'an city, Hunchun city, Huadian city, Antu County and Helong County of Jilin Province, Benxi Manchu Autonomous County, Huanren Manchu Autonomous County, Kuandian Manchu Autonomous County, Fengcheng city and Donggang city of Liaoning Province. The ensemble species distribution model constructed by sdm within the TSS of various models that were set as weight revealed that the potential distribution area of the water deer in the study area was 8764.66 km 2 (28.77% of the study area). Constant adaptation to environmental changes is required by ruminants to allow them to adapt to different ecological niches and feeding habits. In addition, the morphology and function of ruminant digestive systems reveal some adaptive evolutionary characteristics. Blue sheep ( Pseudois nayaur ) display a variety of morpho-physiological adaptations that are typical of grazers. In this study, we collected 64 adult blue sheep samples (whole animal carcasses) from the Helan Mountains, China, during different seasons. The external morphological parameters, digestive system morphological indexes, and rumen surface enlargement factors were determined. Our results reveal that the rumen and reticulum weights were positively correlated with the body weight ( p = 0.004), while the food channel aperture, intestinal length, and weight of the blue sheep digestive tract presented no significant differences between different seasons ( p > 0.05) and sexes ( p > 0.05). There were significant differences ( p < 0.001) in the density, length, and width of mastoids, and the rumen surface enlargement factor was 2.85 ± 1.37, which is typical of roughage feeders. The nutritional and ecological characteristics of blue sheep represent obvious morphological and physiological adaptations to an herbivorous diet. Adopting a quick foraging strategy allows blue sheep to rapidly consume and excrete large amounts of feed, thus obtaining the required energy for their activities and facilitating better adaption to environmental changes.

Blue sheep and red deer, second-class key protected animals in China, are sympatric species with a high degree of overlap of food resources in the Helan Mountains, China. Previous studies with blue sheep and red deer in nature have shown that their physiology is closely related to their gut microbiota. However, growth stages and changes occurring in these species in captivity are still unknown. Thus, 16S rRNA gene sequencing was used to explore diversity, composition and function of the gut microbiota in these two animal species. The diversity and structure of the gut microbiota in captive blue sheep and red deer changed at different growth stages, but the dominant microbiota phyla in the gut microbiota remained stable, which was composed of the phyla Firmicutes, Bacteroidetes and Verrucomicrobia. Moreover, gut microbiota diversity in juvenile blue sheep and red deer was low, with the potential for further colonization. Functional predictions showed differences such as red deer transcription being enriched in adults, and blue sheep adults having a higher cell wall/membrane/envelope biogenesis than juveniles. Microbial changes between blue sheep and red deer at different growth stages and between species mainly depend on the abundance of the microbiota, rather than the increase and absence of the bacterial taxa.

Substantial variation in the environment directly causes remodeling of the colonized gut microbiota, controlling community diversity, and functions in the host to tune-up their adaptive states. However, the mechanisms of microbial community assembly in response to environmental changes remain unclear, especially in endangered ruminants. In this study, we analyzed the microbial communities of 37 fecal samples collected from captive and wild Alpine musk deer ( Moschus chrysogaster ) to characterize the complexity and assembly processes using 16S rRNA gene sequencing. We found significantly different diversities and compositions of gut microbiota among both groups associated with different living environments. Heterogeneous selection was the predominant factor regulating the gut microbiota community under similar climatic conditions, indicating that microbial community assembly was largely driven by deterministic mechanisms. The species co-occurrence network showed complex and tight connections with a higher positive correlation in the wild environment. Moreover, the captive group exhibited significant differences in chemoheterotrophy and fermentation compared with the wild group, but the opposite was observed in animal parasites or symbionts, which might be closely related to diet, energy supply, and healthcare of animals. The Siberian Rubythroat ( Calliope calliope ), a shy and cautious passerine bird, is widely distributed throughout Siberia, Asia, and Europe. In this study, the complete mitogenome of the Siberian Rubythroat samples collected in Heilongjiang, China, was sequenced. The whole length of the complete mitochondrial genome is 16,840 bp, consisting of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one control region. Only one overlapping gene was found among the 13 protein-coding genes (ND4L/ND4). The length of the control region is 1068 bp. The nucleotide composition is 29.32% A, 23.05% T, 14.80% G, and 32.82% C. The Brambling ( Fringilla montifringilla ) (Linnaeus 1758) is a member of the Passeriformes family of birds and primarily feeds on grass seeds and grains. Muscle tissue was collected from birds sampled from Moar Mountain, China, and the complete mitochondrial genome was sequenced. Its mitochondrial genome consists of 13 protein-coding genes (PCGs), 2 rRNA genes (12S rRNA and 16S rRNA), 22 tRNA genes, and 1 control region (CR). The genome comprises 30.30% A, 23.32% T, 14.31% G, and 32.07% C bases. Conservation goals can only be best achieved when there is firm support and cooperation from locals, especially in emerging economies where poor communities often bear most of the cost of human-wildlife conflicts (HWCs). In this study, we explored the economic losses caused by wild boars in two districts, i.e., the Peshawar district and the Nowshera district, in north-western Pakistan. Between May and June 2022, 589 respondents from 53 villages were interviewed. The results revealed that the wild boar was chiefly involved in crop raiding, causing an annual economic loss of USD 12,030 (USD 20.42/household). The highly raided crops included maize (40.24%), followed by wheat (24.95%), vegetables (22.65%), and sugarcane (6.29%). Wild boars were also held accountable for orchard damages. Most people consider the wild boar a common species in the area and want it completely eliminated. We believe that the increasing wild boar population is alarming and should be noticed. The government should collaborate with the local communities to use innovative methods to deter wild boars. Compensation schemes for crop damages should be launched. Moreover, the regular investigation of the wild boar population size and their carrying capacities should be enlisted as integral parts of wildlife management in the area.

The Yellow-rumped Flycatcher ( Ficedula zanthopygia ) is a secondary cavity nesting bird and widely distributed in China, Mongolia, Russia and Korea. In this study firstly sequenced the mitotic genome of the Yellow-rumped Flycatcher ( Ficedula zanthopygia ) gathered at Maorshan, China, using Illumina high-throughput sequencing technology, and then annotated the assembly. The total length of the complete mitochondrial genome is 16,730 bp and it consists of 13 protein-coding, 22 tRNA, 2 rRNA genes, and 1 control region (CR). The CR is 1148 bp in length. The nucleotide composition is 29.59% A, 14.75% T, 32.13% G, 23.54% C. In this study, we sequenced and assembled the complete mitochondrial genome of Dryobates minor by next-generation sequencing. The mitochondrial genome of Dryobates minor is 16,847 bp in length and consists of 13 protein-coding genes (PCGS), two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes and 1 control region (CR). The CG content of the mitochondrial genome is 47.46%. Only one overlap among the 13 protein-coding genes was found: ND4L/ND4. The Pale-legged Leaf-Warbler ( Phylloscopus tenellipes ) (Swinhoe, 1860) is an olive-brown warbler that is extensively dispersed in Asia. We sequenced the whole mitochondrial genome of a Pale-legged Leaf-Warbler collected on Maor Mountain, Heilongjiang Province, China. The mitochondrial genome is 16,972 bp and contains 13 protein-coding genes, two rRNA genes, 22 tRNA genes, and one control region (CR). The CR is 1098 bp. The nucleotide sequence is composed of 29.11% A, 22.98% T, 14.64% G, and 33.27% C. Environmental changes along an altitudinal gradient can facilitate the differentiation of life-history features in ectothermic species, but little attention has been devoted to the reciprocal influence of altitude and alpine slope directionality on life-history variation. According to life-history theory, increased environmental stress causes a change in reproductive allocation from number to quality of offspring, as well as a stronger trade-off between size and number of offspring. To clarify the influence of environmental pressures on the life-history features of the Qinghai toad-headed lizard Phrynocephalus vlangalii along an altitudinal cline, we surveyed late pregnant females from 3 populations of low (2,600 m), middle (3,400 m), and high (3,600 m) elevations in the Dangjin Mountain of Gansu, China from July to October 2019, and compared their inter-population differences in maternal body size, reproductive characteristics, offspring growth, and locomotor performance. Because of lower temperatures, higher humidity, and lower light intensity caused by slope aspect and altitude, the middle-altitude region experienced stronger environmental stress than the high- and low-altitude regions. Our results showed that females were larger at middle- and high-altitude sites and smaller at the low-altitude site, following Bergmann's rule. We also found that females from low-altitude population gave birth earlier than those from the middle and high altitudes. Our results showed a shift in the offspring size-number trade-off of P. vlangalii in response to colder and harsher environments, with lizards from the alpine steppe (i.e. the middle- and high-altitude habitats) producing fewer but larger offspring than those from the warm steppe (i.e. the low-altitude habitat). Low-altitude juveniles grew faster than high-altitude ones, but at the same rates as middle-altitude juveniles. This result demonstrates that the growth of P. vlangalii was associated with temperature and light intensity. Background: Head and neck cancer (HNC) comprises a heterogeneous group of cancers. In view of the distinct biological characteristics and treatment strategies, clinical physicians require high-quality clinical practice guidelines (CPGs) which could provide reliable recommendations on medical practices. We aimed to evaluate the reporting quality of CPGs in the field of HNC., Methods: We developed rigorous search strategies before searching the domestic and international literature databases (n=568) including Medline (via PubMed), Chinese National Knowledge Infrastructure (CNKI) and Wanfang as well as websites of guideline organizations (n=8) published between January 1, 2018 to July 1, 2021 for appropriate guidelines on HNC. We included all evidence-based guidelines about HNC in English or Chinese. We excluded translations, summaries and interpretations of guidelines, as well as older versions of guidelines if an updated edition was available. Data were extracted and the reporting quality was evaluated by two investigators independently guided by the Reporting Items for Practice Guidelines in Healthcare (RIGHT) checklist., Results: A total of 21 guidelines complied with the inclusion criteria. Items show distinctions with reporting proportions among seven RIGHT domains. The proportions of reported items in each RIGHT domain were 75.4% for basic information, 63.1% for background, 42.9% for evidence, 55.1% for recommendations, 42.9% for review and quality assurance, 26.2% for funding and declaration and management of interests, and 50.8% for other information., Discussion: The average reporting quality of the recently published guidelines for HNC was moderate. Objective: To study the relationship between DNA double-strand break (DSB) repair gene mutations and the risk of papillary thyroid microcarcinoma (PTMC)., Methods: One hundred patients with PTMC or benign thyroid nodules (BTNs) at Henan Cancer Hospital were retrospectively analyzed. The DSB repair capacity of peripheral blood T lymphocytes in the two groups was assessed by flow cytometry. Data were compared using Student's t-test to evaluate the relationship between DSB repair capacity and the risk of PTMC. Factors influencing DSB repair capacity were analyzed by multivariate logistic regression analysis. The relationship between PTMC and DSB repair capacity was analyzed by univariate analysis. Targeted next-generation DNA sequencing was applied to screen and analyze DSB repair genes related to PTMC., Results: The DSB repair capacity was 31.30% in the PTMC group and 44.40% in the BTN group, with that of the former being significantly lower (P < 0.05). Multivariate logistic regression analysis of age, sex, obesity status, radiation and other factors showed that radiation exposure was positively correlated with reduced DSB repair capacity(OR = 3.642; 95% CI 1.484-8.935, P = 0.020). Moreover, univariate analysis showed that a reduction in DSB repair capacity was a risk factor for PTMC(OR = 2.333; 95% CI 1.027-5.300, P = 0.043).Targeted next-generation DNA sequencing was performed on the DSB repair genes discovered, and those that were mutated in association with PTMC were Rad50 and FANCA; Rad51 mutations were related to BTN., Conclusion: Radiation exposure is positively associated with induced DSB repair gene mutations, which may cause a reduced capacity for DSB repair and eventually lead to PTMC.

The gonadotropin-releasing hormone (GnRH) signaling pathway controls reproductive functions and cancer growth and progression. However, few studies investigated roles of genetic variants of GnRH pathway genes in survival of patients with non-small cell lung cancer (NSCLC). Therefore, we first evaluated associations between 22,528 single-nucleotide polymorphisms (SNPs) in 101 GnRH pathway genes and survival of 1185 NSCLC patients using a dataset from Prostate, Lung, Colorectal and Ovarian Cancer Screening Trial. We found 572 SNPs to be significantly associated with overall survival (OS) of NSCLC (P ≤ 0.05, Bayesian false discovery probability ≤0.80). We then validated these SNPs in another dataset with 984 NSCLC patients from the Harvard Lung Cancer Susceptibility Study. Finally, two independent SNPs (HBEGF rs4150236G>A and ITPR3 rs116454384C>T) remained significantly associated with NSCLC OS in the combined analysis with hazards ratios of 0.84 (95% confidence interval = 0.76-0.92, P = 0.0003) and 0.85 (0.78-0.94, 0.0012), respectively; their genetic score (the number of protective genotypes) was associated with a better OS and disease-specific survival (P trend  = 0.0002 and 0.0001, respectively). Further expression quantitative trail loci analysis showed a significant correlation between ITPR3 rs116454384 T allele and an increased mRNA expression level in both whole blood and normal lung tissue, and high ITPR3 mRNA expression levels in tumors were associated with a better survival of NSCLC patients. Because ITPR3 mutations were rare in tumors, ITPR3 rs116454384C>T likely had an effect on cancer progression by regulating the gene expression. Therefore, genetic variants of HBEGF rs4150236G>A and ITPR3 rs116454384C>T may be predictors for NSCLC survival, but HBEGF rs4150236G>A functional relevance remains to be determined., (Copyright © 2020 Elsevier Inc. Four adult female worms of Trichuris were isolated from an individual of the wild blue sheep (Pseudois nayaur) inhabiting the Helan Mountains, China, during an epidemiological survey of this wild ruminant. Although there were some differences among the worms in posterior end (rectum) morphology and egg shape, little information regarding species status could be inferred from their morphology. Phylogenetic trees were constructed based on sequences of the ITS1 segment of ribosomal RNA (rRNA), and the sequences of the four Trichuris specimens from wild blue sheep were divided into two distinct lineages (Clade A and Clade B). The two specimens in Clade A were named Genotype I, and had the closest relationship with Trichuris skrjabini; the two specimens in Clade B were named Genotype II and had the closest genetic relationship with a previously described Trichuris sp. In the two Trichuris genotypes identified in the present study, the 18S fragments (261 to 262 bp) of the newly obtained sequences were found to be highly conserved, with merely one insert mutation of a single nucleotide present. The genetic distance based on ITS1 between members of Clade A, composed of two T. skrjabini individuals and two Genotype I individuals, ranged from 0 to 0.0034. These distances are within the intraspecies variation of Trichuris (0-0.0272), suggesting that the Genotype I individuals infesting the wild blue sheep were T. skrjabini. In Clade B, the newly obtained sequences clustered with Trichuris sp. specimens isolated from ruminants (sheep and black goat) with strong support, and the genetic distance ranged from 0.0068 to 0.017, which is also within the intraspecies variation of Trichuris (0-0.0272). However, the genetic distances between the Clade A and Clade B were 0.0442 to 0.0578, which are higher than the intraspecies distances in Trichuris but lower than the interspecies distances (0.102-0.5078). These results implied that Clade A and Clade B most likely represent two subpopulations of T. skrjabini; however, the possibility that Clade A is T. skrjabini and Clade B is a Candidatus Trichuris could not be excluded.

Natural wildlife habitats are regularly subjected to anthropogenic pressures for different purposes, which are heading the biodiversity towards drastic decline. Several endangered wild species are raised in captivity with the aim of re-introduction. In some instances, mixed herds' rearing approach in captivity is adopted for providing social enrichment to captive stocks; however, the impacts of species on each other are least documented. We tested our prediction that keeping mixed herds of captive wild sheep and antelopes provides adequate social enrichment to the captive stocks: if interspecific interactions are balanced. In the current study, we studied the interspecific competition between mixed herds of captive Punjab urial ( Ovis vignei punjabiensis ) and Indian gazelle ( Gazella bennettii ) at Manglot Wildlife Park, Nowshera District, Khyber Pakhtunkhwa Province, Pakistan. We documented the negative effects of behavioural interference by Punjab urial on the feeding behaviour of Indian gazelle. The outcome of the current study revealed that Punjab urial are highly intolerant towards Indian gazelle, with high interference during feeding. Out of the total aggressive events, 77% ( N = 1259) of events ended up with win/loss, in which Punjab urial dominated the Indian gazelle 3.5 times. Moreover, lopsided dominance by Punjab urial resulted in increased intraspecific competition among Indian gazelle ( p <&nbsp;0.001). Current study divulged Indian gazelle to be the subordinate species, with less intake of food. Instead of providing social enrichment by heterospecifics, the Punjab urial is negatively affecting the Indian gazelle, therefore, the results of our study discourage the practice of admix captive breeding for wild sheep and antelopes.

Background and Purpose: This study aimed to analyze the impact of baseline posterior circulation Acute Stroke Prognosis Early Computed Tomography Score (pc-ASPECTS) on the efficacy and safety of endovascular therapy (EVT) for patients with acute basilar artery occlusion., Methods: The BASILAR was a nationwide prospective registry of consecutive patients with a symptomatic and radiologically confirmed acute basilar artery occlusion within 24 hours of symptom onset. We estimated the effect of standard medical therapy alone (SMT group) versus SMT plus EVT (EVT group) for patients with documented pc-ASPECTS on noncontrast CT, both as a categorical (0-4 versus 5-7 versus 8-10) and as a continuous variable. The primary outcomes included favorable functional outcomes (modified Rankin Scale ≤3) at 90 days and mortality within 90 days., Results: In total, 823 cases were included: 468 with pc-ASPECTS 8 to 10 (SMT: 71; EVT: 397), 317 with pc-ASPECTS 5 to 7 (SMT: 85; EVT: 232), and 38 with pc-ASPECTS 0 to 4 (SMT: 13; EVT: 25). EVT was associated with higher rate of favorable outcomes (adjusted relative risk with 95% CI, 4.35 [1.30-14.48] and 3.20 [1.68-6.09]; respectively) and lower mortality (60.8% versus 77.6%, P =0.005 and 35.0% versus 66.2%, P< 0.001; respectively) than SMT in the pc-ASPECTS 5 to 7 and 8 to 10 subgroups. Continuous benefit curves also showed the superior efficacy and safety of EVT over SMT in patients with pc-ASPECTS ≥5. Furthermore, the prognostic effect of onset to puncture time on favorable outcome with EVT was not significant after adjustment for pc-ASPECTS (adjusted odds ratio, 0.98 [95% CI, 0.94-1.02])., Conclusions: Patients of basilar artery occlusion with pc-ASPECTS ≥5 could benefit from EVT. The Ferruginous Duck ( Aythya nyroca ) is a diving duck that is widely distributed in Asia, Africa, and Europe. We determined the complete mitogenome of the Ferruginous Duck gathered at Ningxia, China. The total length of the complete mitochondrial genome is 16,623 bp and it consists of 13 protein-coding, 22 tRNA, 2 rRNA genes, and 1 control region (CR). Only one overlap among the 13 protein-coding genes was found: ND4L/ND4. The CR is 1068 bp in length. The nucleotide composition is 29.66% A, 22.28% T, 15.35% G, 32.71% C. Background: The present study was conducted in Jul 2019 and Jan 2020 in two wildlife parks of the Nowshera district, Khyber Pakhtunkhwa, Pakistan, where the endangered Punjab urial ( Ovis vignei punjabiensis ) is successfully bred in captivity. We determined diversity of internal and external parasites that take advantage of the situation of congestion, resulting in massive mortalities of wild animals in captivity., Methods: Internal parasites of living urial were determined by direct wet smear and flotation methods, while dead urial was necropsied for any pertaining observation., Results: All examined fecal samples were found infected with gastrointestinal parasites, which had significant difference in the total abundance in winter and summer. S. papillosus and H.contortus, and a single protozoan, Eimeria spp. were the dominant parasites in fecal samples. Ticks collected from urial enclosures and dead animals were of single species H. anatolicum . Theileria spp. was observed in blood, while hydatid cysts were found in lungs and liver of necropsied urial., Conclusion: The study indicates that internal parasites such as Haemonchus contortus and Strongyloides papillosus , while external parasites as Hyalomma anatolicum ticks played major role in the population decline. Meiofauna particularly marine nematodes around the Caiwei Guyot in the northwest Pacific Ocean and a Polymetallic Nodule Field in the northeast Pacific Ocean were studied. Due to the geographic structure, the Caiwei Guyot and the Polymetallic Nodule Field had different environmental characteristics. Meiofaunal abundances around the Guyot area ranged from 9.18 to 25.59 ind./10 cm 2 , which were much lower than those in the Polymetallic Nodule Field. Marine nematode was the most dominant group. A total of 123 species, belonging to 74 genera and 29 families were found. Xyalidae (21.43%), Cyatholaimidae (9.82%), Linhomoeidae (8.03%) were the dominant families. The values of species number, Margalef's species richness and Shannon-Wiener diversity index ranged from 15 to 62, 4.75 to 12.84 and 2.58 to 3.93, respectively. The combination of water depth, silt-clay content and chlorophyll-a concentration can best explain the differences of nematode community. Microbiota of the wild blue sheep ( Pseudois nayaur ) presents a seasonal variation due to different dietary selection and feeding strategies from different ecological niches chosen by different sex in summer. To address those issues, we analyzed the variation of gut microbiota based on the material from the feces, with 16S rRNA and meta-genome aimed to explore seasonal and gender differences. The results indicate that seasonal dietary changes and gender differentiation, as expected, cause the variation in sheep's gut microbiota structure. The variation of the former is more significant than the latter. Dominant Firmicutes exists a significantly higher abundance in summer than that in winter. Subordinate Bacteroides expresses no seasonal difference between the two seasons. Compared with the winter group, the summer group is featured by abundant enzymes digesting cellulose and generating short-chain fatty acids (SCFAs), such as beta-glucosidase (EC: 3.2.1.21) for cellulose digestion, and butyrate kinase (EC:2.7.2.7) in butyrate metabolism, implying that the changes of the composition in intestinal flora allow the sheep to adapt to the seasonalized dietary selection through alternated microbial functions to reach the goal of facilitating the efficiency of energy harvesting. The results also show that the blue sheep expresses a prominent sexual dimorphism in the components of gut microbiota, indicating that the two sexes have different adaptations to the dietary selection, and demands for physical and psychological purposes. To identify genetic variants for risk of squamous cell carcinoma of the head and neck (SCCHN), we conducted a two-phase genome-wide association study consisting of 7,858,089 SNPs in 2,171 cases and 4,493 controls of non-Hispanic white, of which, 434,839 typed and 7,423,250 imputed SNPs were used as the discovery. SNPs with P < 1 × 10 -3 were further validated in the OncoArray study of oral and pharynx cancer (5,205 cases and 3,232 controls of European ancestry) from databases of Genotypes and Phenotypes. Meta-analysis of the discovery and replication studies identified one novel locus 6p22.1 ( P = 2.96 × 10 -9 for the leading rs259919) and two cancer susceptibility loci 6p21.32 (rs3135001, HLA-DQB1 ) and 6p21.33 (rs1265081, CCHCR1 ) associated with SCCHN risk. Further stratification by tumor site revealed four known cancer loci (5p15.33, 6p21.32, 6p21.33, and 2p23.1) associated with oral cavity cancer risk and oropharyngeal cancer risk, respectively. In addition, one novel locus 18q22.2 ( P = 2.54 × 10 -9 for the leading SNP rs142021700) was identified for hypopharynx and larynx cancer risk. For SNPs in those reported or novel loci, we also performed functional annotations by bioinformatics prediction and expression quantitative trait loci analysis. Collectively, our identification of four reported loci (2p23.1, 5p15.33, 6p21.32, and 6p21.33) and two novel loci (6p22.1 and 18q22.2) for SCCHN risk highlight the importance of human leukocyte antigen loci for oropharyngeal cancer risk, suggesting that immunologic mechanisms are implicated in the etiology of this subset of SCCHN. It is actually a circular molecular of 16,816 bp in length and consists 13 protein-coding genes, 22 transfer-RNA genes, 2 ribosomal-RNA genes, and 1 control region (D-loop, 1,243 bp in length). The nucleotide composition is 30.3% A, 30.1% C, 14.5% G, 25.1% T. Importance: Several randomized clinical trials have recently established the safety and efficacy of endovascular treatment (EVT) of acute ischemic stroke in the anterior circulation. However, it remains uncertain whether patients with acute basilar artery occlusion (BAO) benefit from EVT., Objective: To evaluate the association between EVT and clinical outcomes of patients with acute BAO., Design, Setting, and Participants: This nonrandomized cohort study, the EVT for Acute Basilar Artery Occlusion Study (BASILAR) study, was a nationwide prospective registry of consecutive patients presenting with an acute, symptomatic, radiologically confirmed BAO to 47 comprehensive stroke centers across 15 provinces in China between January 2014 and May 2019. Patients with acute BAO within 24 hours of estimated occlusion time were divided into groups receiving standard medical treatment plus EVT or standard medical treatment alone., Main Outcomes and Measures: The primary outcome was the improvement in modified Rankin Scale scores (range, 0 to 6 points, with higher scores indicating greater disability) at 90 days across the 2 groups assessed as a common odds ratio using ordinal logistic regression shift analysis, adjusted for prespecified prognostic factors. The secondary efficacy outcome was the rate of favorable functional outcomes defined as modified Rankin Scale scores of 3 or less (indicating an ability to walk unassisted) at 90 days. Safety outcomes included symptomatic intracerebral hemorrhage and 90-day mortality., Results: A total of 1254 patients were assessed, and 829 patients (of whom 612 were men [73.8%]; median [interquartile] age, 65 [57-74] years) were recruited into the study. Of these, 647 were treated with standard medical treatment plus EVT and 182 with standard medical treatment alone. Ninety-day functional outcomes were substantially improved by EVT (adjusted common odds ratio, 3.08 [95% CI, 2.09-4.55]; P < .001). Moreover, EVT was associated with a significantly higher rate of 90-day modified Rankin Scale scores of 3 or less (adjusted odds ratio, 4.70 [95% CI, 2.53-8.75]; P < .001) and a lower rate of 90-day mortality (adjusted odds ratio, 2.93 [95% CI, 1.95-4.40]; P < .001) despite an increase in symptomatic intracerebral hemorrhage (45 of 636 patients [7.1%] vs 1 of 182 patients [0.5%]; P < .001)., Conclusions and Relevance: Among patients with acute BAO, EVT administered within 24 hours of estimated occlusion time is associated with better functional outcomes and reduced mortality.

The blue sheep is an endemic species to the Qinghai-Tibet Plateau and surrounding regions. It has been regarded as having 2 subspecies: Pseudois nayaur nayaur and P. n. szechuanensis. However, such a classification remains controversial. Herein, we analyze 10 microsatellite loci and part of the mitochondrial control region for clarification in such taxonomic debates. We use samples from 168 individuals from 6 geographic populations covering almost all the distribution areas of the species in China to carry out comparisons. Phylogenetic trees derived from both the microsatellite and mitochondrial markers combined with the discriminant analysis of principal components (DAPC) and the STRUCTURE analysis reveal that the individuals in the Helan Mountains are well grouped with a distinct evolutionary lineage and are significantly different from the other populations of P. n. szechuanensis according to Fst values, implying that this isolated population should be categorized as a valid subspecies; namely, Pseudois nayaur alashanicus. Reptiles are especially vulnerable to climate warming because their behavior, physiology, and life history are highly dependent on environmental temperature. In this study, we envisaged new probable mechanisms underlying the high vulnerability of lizards, wherein heat exposure induces oxidative stress and leads to immunosuppression. To test this hypothesis, we conducted a warming experiment on a lizard (Eremias multiocellata) from a desert steppe in Inner Mongolia from May to September using open-top chambers set up in their natural habitat and compared the components of oxidative stress (antioxidant ability [Superoxide dismutase (SOD) activity], extent of oxidative damage [malondialdehyde (MDA) content]), and immunocompetence (white blood cells [WBC] counts and immunoglobulin M [IgM] expression) between the warming and control groups. At the end of the experiment, the warming treatment did not affect the survival rate of the lizards. However, MDA content, but not SOD activity, was significantly higher in the warming group than in the control group. The WBC counts and IgM expression were significantly lower in the warming group than in the control group. Five novel H5N6 influenza viruses, including four highly pathogenic avian influenza viruses and one low pathogenic avian influenza virus, were isolated from migratory birds in Ningxia, China, in November 2017. To understand the genetic origination of the novel H5N6 virus, and the infectivity and pathogenicity of the four highly pathogenic avian influenza viruses in mammals, phylogeographic analyses and infection studies in mice were performed. The phylogenetic and phylogeographic analyses showed that the H5N6 isolates, which are closely related to the viruses from Korea, Japan and the Netherlands, originated from reassortant virus between H5N8 and HxN6 viruses from western Russia. We determined the whole mtDNA genome of the water deer ( Hydropotes inermis) in Benxi, Liaoning. The total length of the complete mitochondrial genome is 16,355 bp and it consists of 13 protein-coding, 22 tRNA, rRNA genes, and 1 control region (CR). Two overlaps among the 13 protein-coding genes were found: ND4L/ND4 and ND5/ND6. The CR is 928 bp in length. As for the wild animals, their diet components are always changed, so that we have to monitor such changes by analyzing the modification of intestinal microbial community. Such effort allows us to amend their conservation strategies and tactics accordingly so that they are able to appropriately adapt to the new environment and dietary selection. In this study we focus on the gut flora of two groups of an endangered species, Alpine musk deer ( Moschus chrysogaster ), wild group (WG) which is compared with that of the individuals of the same species but kept in the captivities (CG), a control group. Such a project is aimed to work out whether the composition of the gut microbes has significantly been changed due to captive feedings. To do so, we used 16S rRNA amplicon sequencing to characterize gut bacteria of the musk deer from the two groups. The results show that there is a significant difference in community structure of the bacteria: WG shows significant enrichment of Firmicutes and depletion of Bacteroidetes, while CG has a significant abundance of Proteobacteria and Euryarchaeota. Metagenomics was used to analyze the differences in functional enzymes between the two groups. The related results indicate that genes in WG are mostly related to the enzymes digesting cellulose and generating short-chain fatty acids (SCFAs) for signaling pathways, but CG shows enrichment in methanogenesis, including the CO 2 /H 2 pathway and the methylotrophic pathway. Thus, this study indicates that the Firmicutes-rich gut microbiota in the WG enables individuals to maximize their energy intake from the cellulose, and has significant abundance of Euryarchaeota and methanogenesis pathways that allow them to reduce redundant energy consumption in methane metabolism, ensuring them to adapt to the wild environments., (Copyright © 2020 Sun, Sun, Shi, Liu, Zhao, Lu, Gao, Zhu, Chen, Zhang, Pan, Li, Teng and Guo.)

We determined the whole mtNDA genome of the Plateau brown frog ( Rana kukunoris ) in Helan Mountains, Inner Mongolia. The complete mitochondrial genome consists of 13 protein-coding, 22 tRNA, 2 rRNA genes, and 1 control region (CR), and its total length is 16,644 bp. Three overlaps among the 13 protein-coding genes were found: ATP8/ATP6, ND4L/ND4, and ND5/ND6. The CR is 837 bp in length. The nucleotide composition is 27.49% A, 29.06% T, 14.77% G, 28.68% C. Background and Objective: The Punjab urial (Ovis vignei punjabiensis) is an endangered wild sheep of Pakistan, raised in captivity with the aim of re-introduction. To date, no information is available about population trends of this species in captivity. The current study was conducted with the aim to evaluate the population trend to better guide captive breeding for improved productivity and conservation value., Materials and Methods: Annual population data recorded and maintained by the Wildlife Department Khyber Pakhtunkhwa, Pakistan, were used. The data were compiled and analyzed in Microsoft Excel 2010 for determining growth rates and package Growthcurver in R-version 3.5.1 was used to produce a graphical representation of the population trend., Results: The overall average annual population growth rate was rN = 0.22. Results revealed a fast initial growth rate with an average value of rN = 0.4 per year. Birth rates of bN = 0.45 for the first nine years were considerably higher than the death rates dN = 0.22 and the population increased with exponential growth. In the subsequent year, very high mortalities rates (dN = 1.2), likely attributed to the clumping of the population, resulted in the collapse of the population, leaving it in a state of unstable equilibrium., Conclusion: Results support the evaluation of management data to reveal carrying capacity in captive populations, to guide and inform appropriate release of surplus animals into natural habitats.

To ensure that captive breeding and other associated programs are more robust and sustainable, it is of utmost importance to ensure optimum welfare. Although it is well known that standard welfare is crucial for successful captive breeding, there is still a lack of welfare assessment protocols for wild species. The current study aimed to develop a leading baseline welfare assessment protocol for assessing welfare in captive Punjab urial. This protocol is based on the welfare protocol for domestic sheep from the Welfare Quality ® project, coupled with all the information obtained from the published literature on the species' biology and ecology. This protocol consists of 4 principles, 12 criteria, and 31 animal- and resource-based indicators. The protocol was tested and applied to three different herds of Punjab urial at two different facilities. Glutamine dependence is a unique metabolic defect seen in cutaneous melanoma (CM), directly influencing the treatment and prognosis. Here, we investigated the associations between 6025 common single-nucleotide polymorphisms (SNPs) in 77 glutamine metabolic pathway genes with CM-specific survival (CMSS) using genotyping datasets from two published genome-wide association studies (GWASs). In the single-locus analysis, 76 SNPs were found to be significantly associated with CMSS (P < .050, false-positive report probability < 0.2 and Bayesian false discovery probability < 0.8) in the discovery dataset, of which seven SNPs were replicated in the validation dataset and three SNPs (HAL rs17676826T > C, LGSN rs12663017T > A, and NOXRED1 rs8012548A > G) independently predicted CMSS, with an effect-allele attributed adjusted hazards ratio of 1.52 (95% confidence interval = 1.19-1.93) and P < .001, 0.68 (0.54-0.87) and P = .002 and 0.62 (0.46-0.83) and P = .002, respectively. The model including the number of unfavorable genotypes (NUGs) of these three SNPs and covariates improved the five-year CMSS prediction (P = .012) than the one with other covariates only. Further expression quantitative trait loci (eQTL) analysis found that the LGSN rs12663017 A allele was significantly associated with increased messenger RNA (mRNA) expression levels (P = 8.89 × 10 -11 ) in lymphoblastoid cell lines of the 1000 Genomes Project database. In the analysis of the genotype tissue expression (GTEx) project datasets, HAL rs17676826 C and NOXRED1 rs8012548 G alleles were significantly associated with their mRNA expression levels in sun-exposed skin of the lower leg (P = 6.62 × 10 -6 and 1.37 × 10 -7 , respectively) and in sun-not-exposed suprapubic skin (P < .001 and 1.43 × 10 -8 , respectively). Abnormal methionine dependence in cancer cells has led to methionine restriction as a potential therapeutic strategy. We hypothesized that genetic variants involved in methionine-metabolic genes are associated with survival in nonsmall cell lung cancer (NSCLC) patients. Therefore, we investigated associations of 16,378 common single-nucleotide polymorphisms (SNPs) in 97 methionine-metabolic pathway genes with overall survival (OS) in NSCLC patients using genotyping data from two published genome-wide association study (GWAS) datasets. In the single-locus analysis, 1,005 SNPs were significantly associated with NSCLC OS (p < 0.05 and false-positive report probability < 0.2) in the discovery dataset. Three SNPs (RUNX3 rs7553295 G > T, AMD1 rs1279590 G > A and MSRA rs73534533 C > A) were replicated in the validation dataset, and their meta-analysis showed an adjusted hazards ratio [HR] of 0.82 [95% confidence interval (CI) =0.75-0.89] and p meta  = 2.86 × 10 -6 , 0.81 (0.73-0.91) and p meta  = 4.63 × 10 -4 , and 0.77 (0.68-0.89) and p meta  = 2.07 × 10 -4 , respectively). A genetic score of protective genotypes of these three SNPs revealed an increased OS in a dose-response manner (p trend  < 0.0001). Further expression quantitative trait loci (eQTL) analysis showed significant associations between these genotypes and mRNA expression levels. Moreover, differential expression analysis further supported a tumor-suppressive effect of MSRA, with lower mRNA levels in both lung squamous carcinoma and adenocarcinoma (p < 0.0001 and < 0.0001, respectively) than in adjacent normal tissues. Additionally, low mutation rates of these three genes indicated the critical roles of these functional SNPs in cancer progression. The complete mitochondrial genome of Helan Mountain chukar ( Alectoris chukar potanini ) was first determined. The genome was 16,685 bp in length, comprising 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a putative control region. The phylogenetic analysis based on the complete mitochondrial genome sequences revealed a close relationship between A. chukar potanini and Tetraogallus tibetanus .
