In terms of infectious diseases caused by a variety of microorganisms, the ability to promptly and accurately identify the causative agents is the first step on the path to all types of effective management of such infections. Among the various factors that are affecting global bee health, viruses have often been linked to honey bee colony losses and they pose a serious threat to the fraction of agriculture that depends on the service of pollinators. Over the past few decades, PCR-based molecular methods have provided powerful tools for rapid, specific, and sensitive detection and the quantification of difficult-to-grow pathogenic microorganisms such as viruses in honey bees. However, PCR-based methods require nucleic acid extraction and purification, which can be quite laborious and time-consuming and they involve the use of organic solvents and chaotropic agents like phenol and chloroform which are volatile and highly toxic. In response, we developed a novel and non-sacrificial method for detecting viral infections in honey bees. As little as 1 μl of hemolymph was collected from adult workers, larvae, and queens of bee colonies by puncturing the soft inter-tergal integument between the second and third dorsal tergum with a fine glass capillary. The hemolymph was then diluted and subjected to RT-PCR analysis directly. The puncture wound caused by the glass capillary was found to heal automatically and rapidly without any trouble and the lifespan of the experimental workers remained unaffected. Using this method, we detected multiple viruses including Deformed wing virus (DWV), Black queen cell virus (BQCV), and Sacbrood virus (SBV) in infected bees. Furthermore, expressed transcripts that indicate the induction of innate immune response to the virus infections were also detected in the hemolymph of infected bees. The simplicity and cost-effectiveness of this innovative approach will allow it to be a valuable, time-saving, safer, and more environmentally friendly contribution to bee disease management programs., (Copyright © 2021. Honey bees Apis mellifera forage in a wide radius around their colony, bringing back contaminated food resources that can function as terrestrial bioindicators of environmental pesticide exposure. Evaluating pesticide exposure risk to pollinators is an ongoing problem. Here we apply five metrics for pesticide exposure risk (prevalence, diversity, concentration, significant pesticide prevalence, and hazard quotient (HQ)) to a nation-wide field study of honey bees, Apis mellifera in the United States. We examined samples from 1055 apiaries over seven years for 218 different pesticide residues and metabolites, determining that bees were exposed to 120 different pesticide products with a mean of 2.78 per sample. Pesticides in pollen were highly prevalent and variable across states. While pesticide diversity increased over time, most detections occurred at levels predicted to be of low risk to colonies. Varroacides contributed most to concentration, followed by fungicides, while insecticides contributed most to diversity above a toxicity threshold. High risk samples contained one of 12 different insecticides or varroacides. Nutrition is involved in regulating multiple aspects of honey bee biology such as caste, immunity, lifespan, growth and behavioral development. Deformed wing virus (DWV) is a major pathogenic factor which threatens honey bee populations, and its replication is regulated by the nutrition status and immune response of honey bees. The alimentary canal of the honey bee is home to a diverse microbial community that provides essential nutrients and serves to bolster immune responses. However, to what extent gut bacteria affect honey bee nutrition metabolism and immunity with respect to DWV has not been investigated fully. In this study, newly emerged worker bees were subjected to four diets that contained (1) pollen, (2) pollen and antibiotics, (3) neither pollen nor antibiotics or (4) antibiotics alone. The expression level of two nutrition genes target of rapamycin ( tor ) and insulin like peptide ( ilp1 ), one nutritional marker gene vitellogenin ( vg ), five major royal jelly protein genes ( mrjp1 - 5 ), one antimicrobial peptide regulating gene relish ( rel ), and DWV virus titer and its replication intermediate, negative RNA strand, were determined by qRT-PCR from the honey bees at 7 days post-antibiotic treatment. Additionally, honey bee head mass and survival rate were measured. We observed that antibiotics decreased the expression of tor and rel , and increased DWV titer and its replication activity. Expression of ilp1 , mrjp1 - 5 and vg , and honey bee head mass were also reduced compared with bees on a pollen diet. Antibiotics also caused a significant drop in survivorship, which could be rescued by addition of pollen to the diet. Of importance, pollen could partially rescue the loss of vg and mrjp2 while also increasing the head mass of antibiotic-treated bees. RNA viruses that contain single-stranded RNA genomes of positive sense make up the largest group of pathogens infecting honey bees. Sacbrood virus (SBV) is one of the most widely distributed honey bee viruses and infects the larvae of honey bees, resulting in failure to pupate and death. Among all of the viruses infecting honey bees, SBV has the greatest number of complete genomes isolated from both European honey bees Apis mellifera and Asian honey bees A. cerana worldwide. To enhance our understanding of the evolution and pathogenicity of SBV, in this study, we present the first report of whole genome sequences of two U.S. strains of SBV. The complete genome sequences of the two U.S. SBV strains were deposited in GenBank under accession numbers: MG545286.1 and MG545287.1. Both SBV strains show the typical genomic features of the Iflaviridae family. The phylogenetic analysis of the single polyprotein coding region of the U.S. strains, and other GenBank SBV submissions revealed that SBV strains split into two distinct lineages, possibly reflecting host affiliation. The phylogenetic analysis based on the 5'UTR revealed a monophyletic clade with the deep parts of the tree occupied by SBV strains from both A. cerane and A. mellifera , and the tips of branches of the tree occupied by SBV strains from A. mellifera . The study of the cold stress on the pathogenesis of the SBV infection showed that cold stress could have profound effects on sacbrood disease severity manifested by increased mortality of infected larvae. This result suggests that the high prevalence of sacbrood disease in early spring may be due to the fluctuating temperatures during the season. This study will contribute to a better understanding of the evolution and pathogenesis of SBV infection in honey bees, and have important epidemiological relevance.

The synergistic interactions between the ectoparasitic mite Varroa destructor and Deformed wing virus (DWV) lead to the reduction in lifespan of the European honey bee Apis mellifera and often have been implicated in colony losses worldwide. However, to date, the underlying processes and mechanisms that form the multipartite interaction between the bee, mite, and virus have not been fully explained. To gain a better understanding of honey bees' defense response to Varroa mite infestation and DWV infection, the DWV titers and transcription profiles of genes originating from RNAi, immunity, wound response, and homeostatic signaling pathways were monitored over a period of eight days. With respect to DWV, we observed low viral titers at early timepoints that coincided with high levels of Toll pathway transcription factor Dorsal, and its downstream immune effector molecules Hymenoptaecin , Apidaecin , Abaecin , and Defensin 1 . However, we observed a striking increase in viral titers beginning after two days that coincided with a decrease in Dorsal levels and its corresponding immune effector molecules, and the small ubiquitin-like modifier (SUMO) ligase repressor of Dorsal, PIAS3. We observed a similar expression pattern for genes expressing transcripts for the RNA interference (Dicer/Argonaute), wound/homeostatic (Janus Kinase), and tissue growth (Map kinase/Wnt) pathways. Our results demonstrate that on a whole, honey bees are able to mount an immediate, albeit, temporally limited, immune and homeostatic response to Varroa and DWV infections, after which downregulation of these pathways leaves the bee vulnerable to expansive viral replication. The critical insights into the defense response upon Varroa and DWV challenges generated in this study may serve as a solid base for future research on the development of effective and efficient disease management strategies in honey bees.

At least two parasitic mites have moved from Asian species of honeybees to infest Apis mellifera. Of these two, Varroa destructor is more widespread globally while Tropilaelaps mercedesae has remained largely in Asia. Tropilaelaps mites are most problematic when A. mellifera is managed outside its native range in contact with Asian species of Apis. In areas where this occurs, beekeepers of A. mellifera treat aggressively for Tropilaelaps and Varroa is either outcompeted or is controlled as a result of the aggressive treatment regime used against Tropilaelaps. Many mite control products used worldwide may in fact control both mites but environmental conditions differ globally and thus a control product that works well in one area may be less or ineffective in other areas. This is especially true of volatile compounds. In the current research we tested several commercial products known to control Varroa and powdered sulfur for efficacy against Tropilaelaps. Additionally, we tested the cultural control method of making a hive division to reduce Tropilaelaps growth in both the parent and offspring colony. Making a split or nucleus colony significantly reduced mite population in both the parent and nucleus colony when compared to un-manipulated control colonies. The formic acid product, Mite-Away Quick Strips®, was the only commercial product that significantly reduced mite population 8 weeks after initiation of treatment without side effects. Sulfur also reduced mite populations but both sulfur and Hopguard® significantly impacted colony growth by reducing adult bee populations. Apivar® (amitraz) strips had no effect on mite or adult bee populations under the conditions tested.

Parasitic Tropilaelaps (Delfinado and Baker) mites are a damaging pest of European honey bees (Apis mellifera L.) in Asia. These mites represent a significant threat if introduced to other regions of the world, warranting implementation of Tropilaelaps mite surveillance in uninfested regions. Current Tropilaelaps mite-detection methods are unsuitable for efficient large scale screening. We developed and tested a new bump technique that consists of firmly rapping a honey bee brood frame over a collecting pan. Our method was easier to implement than current detection tests, reduced time spent in each apiary, and minimized brood destruction. This feasibility increase overcomes the test's decreased rate of detecting infested colonies (sensitivity; 36.3% for the bump test, 54.2% and 56.7% for the two most sensitive methods currently used in Asia). Considering this sensitivity, we suggest that screening programs sample seven colonies per apiary (independent of apiary size) and 312 randomly selected apiaries in a region to be 95% sure of detecting an incipient Tropilaelaps mite invasion. Further analyses counter the currently held view that Tropilaelaps mites prefer drone bee brood cells. Tropilaelaps mite infestation rate was 3.5 +/- 0.9% in drone brood and 5.7 +/- 0.6% in worker brood. We propose the bump test as a standard tool for monitoring of Tropilaelaps mite presence in regions thought to be free from infestation. However, regulators may favor the sensitivity of the Drop test (collecting mites that fall to the bottom of a hive on sticky boards) over the less time-intensive Bump test.

Recent declines in honey bee populations and increasing demand for insect-pollinated crops raise concerns about pollinator shortages. Pesticide exposure and pathogens may interact to have strong negative effects on managed honey bee colonies. Such findings are of great concern given the large numbers and high levels of pesticides found in honey bee colonies. Thus it is crucial to determine how field-relevant combinations and loads of pesticides affect bee health. We collected pollen from bee hives in seven major crops to determine 1) what types of pesticides bees are exposed to when rented for pollination of various crops and 2) how field-relevant pesticide blends affect bees' susceptibility to the gut parasite Nosema ceranae. Our samples represent pollen collected by foragers for use by the colony, and do not necessarily indicate foragers' roles as pollinators. In blueberry, cranberry, cucumber, pumpkin and watermelon bees collected pollen almost exclusively from weeds and wildflowers during our sampling. Thus more attention must be paid to how honey bees are exposed to pesticides outside of the field in which they are placed. We detected 35 different pesticides in the sampled pollen, and found high fungicide loads. The insecticides esfenvalerate and phosmet were at a concentration higher than their median lethal dose in at least one pollen sample. While fungicides are typically seen as fairly safe for honey bees, we found an increased probability of Nosema infection in bees that consumed pollen with a higher fungicide load. Our results highlight a need for research on sub-lethal effects of fungicides and other chemicals that bees placed in an agricultural setting are exposed to.

An international initiative is developing a scientifically rigorous approach to evaluate the potential risks to nontarget arthropods (NTAs) posed by insect-resistant, genetically modified (IRGM) crops. It adapts the tiered approach to risk assessment that is used internationally within regulatory toxicology and environmental sciences. The approach focuses on the formulation and testing of clearly stated risk hypotheses, making maximum use of available data and using formal decision guidelines to progress between testing stages (or tiers). It is intended to provide guidance to regulatory agencies that are currently developing their own NTA risk assessment guidelines for IRGM crops and to help harmonize regulatory requirements between different countries and different regions of the world.

Work-related upper limb disorders (WRULDs) are difficult to diagnose due to the limited availability of recognized objective assessment methods. This case series report demonstrates the use of the "typing capacity cycle" test and standardized clinical tests as outcome measures to assess work capacity in four high-intensity keyboard and mouse users who remain at work before and after the implementation of a six-month treatment program. Pain intensity, the duration of pain before treatment, the duration of treatment, type of work, and the location of the pain were recorded for each patient. Function was assessed before and after treatment using the Functional Grading Scale. The assessment results were analyzed to determine any improvements made after rehabilitation. The outcomes indicate that all patients improved their resting pain and work capacity with the three-phase rehabilitation program.

A field study was conducted over 2 yr to determine the effects of transgenic sweet corn containing a gene from the bacterium Bacillus thuringiensis (Bt) on the diversity and abundance of nontarget arthropods. The Bt hybrid (expressing Cry1Ab endotoxin for lepidopteran control) was compared with near-isogenic non-Bt and Bt hybrids treated with a foliar insecticide and with a near-isogenic non-Bt hybrid without insecticides. Plant inspections, sticky cards, and pitfall traps were used to sample a total of 573,672 arthropods, representing 128 taxonomic groups in 95 families and 18 orders. Overall biodiversity and community-level responses were not significantly affected by the transgenic hybrid. The Bt hybrid also had no significant adverse effects on population densities of specific nontarget herbivores, decomposers, and natural enemies enumerated at the family level during the crop cycle. As expected, the insecticide lambda-cyhalothrin had broad negative impacts on the abundance of many nontarget arthropods. One insecticide application in the Bt plots reduced the overall abundance of the natural enemy community by 21-48%. Five applications in the non-Bt plots reduced natural enemy communities by 33-70%. Nontarget communities affected in the insecticide-treated Bt plots exhibited some recovery, but communities exposed to five applications showed no trends toward recovery during the crop cycle. This study clearly showed that the nontarget effects of Bt transgenic sweet corn on natural enemies and other arthropods were minimal and far less than the community-level disruptions caused by lambda-cyhalothrin.
